Fungi were isolated monthly from collections of first year needles, second year needles, and both surface sterilized and non-sterilized abscised brown needles in the canopy and one-year-old litter on the forest floor. The vertical and phenological distribution of the 8 commonest fungi are reported. After the second growing season in the litterMortierella isabellina, Trichoderma spp., andPenicillium spp. overgrow these fungi on the primary isolation plates using the procedure adopted in this phase of the needle study.Presented at the Eighth International Congress of Biometeorology, 9–14 September 1979, Shefayim, Israel

Morphological changes in the decomposing litter ofAbies spp. andBetula spp. in a mor humus form were studied by a rapid thin section method. According to the morphological characteristics, the epidermis, mesophyll and vascular bundleof Abies needle litter were classified into four types: (i) newly fallen; (ii) slightly decomposed; (iii) moderately decomposed; and (iv) greatly decomposed. The distribution of these tissue types along the profile of the forest floor was then investigated. The morphological changes in other litter types, such as branches, scales andBetula leaves during decomposition were observed directly with microscope and electron microscope. Five vertical thin sections and 80 horizontal thin sections were used for these observations and investigations. the decomposition ofAbies litter was slower than that ofBetula litter. The relative decomposition rate of the tissues was in the order of: mesophyll>vascular bundle >epidermis inAbies needles; mesophyll≥epidermis>vascular bundles inBetula leaves; and inner bark >xylem>outer bark in bothAbies andBetula branches. The last remains of the litter were usually stomata, segments of seminiferous scale and outer bark ofAbies. The decomposition of plant litter occurred mainly within the L and F layers of the soil (0–5 cm in depth).Abies needles andBetula leaves completely disappeared at depths of 0–6 cm and 0–4 cm, respectively. Branches disappeared within the top of 5 cm and 6–8 cm forBetula and forAbies, respectively. The scales ofAbies were most slowly decomposed in the soil layers.

Fungi, especially basidiomycetous litter decomposers, are pivotal to the turnover of soil organic matter in forest soils. Many litter decomposing fungi have a well-developed capacity to translocate resources in their mycelia, a feature that may significantly affect carbon (C) and nitrogen (N) dynamics in decomposing litter. In an eight-month long laboratory study we investigated how the external availability of N affected the decomposition of Scots pine needles, fungal biomass production, N retention and N-mineralization by two litter decomposing fungi – Marasmius androsaceus and Mycena epipterygia. Glycine additions had a general, positive effect on fungal biomass production and increased accumulated needle mass loss after 8 months, suggesting that low N availability may limit fungal growth and activity in decomposing pine litter. Changes in the needle N pool reflected the dynamics of the fungal mycelium. During late decomposition stages, redistribution of mycelium and N out from the decomposed needles was observed for M. epipterygia, suggesting autophagous self degradation.

Decomposition of Picea abies needles and production of extracellular enzymes involved in decomposition of lignin, cellulose, hemicelluloses and other organic compounds were studied in fungal strains of interior needle colonizers isolated from needles in different stages of decomposition (attached to trees, and early and late decomposition stages in the litter horizon). In total, 12 strains of ascomycetes (members of Helotiales, Hypocreales, Dothideales, Diaporthales and Eurotiales) and four basidiomycetes (Polyporales, Agaricales and Russulales) were tested. Significant decomposition of needles was recorded for all fungal isolates. All isolates produced cellobiohydrolase, β-glucosidase, β-xylosidase, N-acetylglucosaminidase, α-glucosidase, phosphatase and arylsulfatase and most fungi also produced endocellulase, endoxylanase and laccase in needle litter. In addition, other hemicellulases were produced by all strains. Mn-peroxidase was only produced by two basidiomycetes. Although enzyme activities varied, fungi associated with needles on fallen trees exhibited enzyme production comparable with later litter colonizers, and there was no significant difference in enzyme production between ascomycete and basidiomycete strains.

The fungal species involved in the decomposition of needle litter and their response to intraspecific genetic variation of trees are poorly known. First, we compared the needle decomposition and fungal decomposers underneath eight different Norway spruce clones in situ. This experiment revealed 60-70% loss of needle mass in two years. Although spruce clones differed considerably in growth (twofold height difference) and their needles differed in chemical composition, no significant difference was found for loss of needle mass under the spruce clones. Furthermore, the spruce clones did not affect the community structure of the fungal decomposers. Fungi inhabiting needle litter were identified by extracting ribosomal RNA (rRNA) and sequencing complementary DNA (cDNA) of internal trascribed spacer 1 (ITS1) region. The most frequent identifications were Lophodermium, Pezizales, Mycena, and Marasmius, suggesting that endophytic fungi were involved in the decomposition process. Second, we evaluated the potential of endophytes to decompose needle material in a microcosm experiment in which all other fungi than endophytes were excluded. Within 2 years, the endophytes had decomposed 35-45% of the needle mass. Sequences of Mollisia, Lophodermium, Lachnum, and Phialocephala were most frequently found in rRNA and rDNA extracted from the needles at the end of the microcosm experiment. The dominant needle endophyte in fresh, green needles was Lophodermium piceae, and this species was also found frequently in the needle material after 2 years of decay both in the field and laboratory experiments. Moreover, the relative abundance of Lophodermium-derived denaturing gradient gel electrophoresis (DGGE) bands correlated positively with the decomposition in the microcosm experiment. Hence, our results suggest a significant role of endophytic fungi, and particularly L. piceae, in the process of needle decomposition in boreal forests.

We investigated microfungal assemblages on leaf litter within a subalpine forest in central Japan and their variation with season, litter depth, and litter species. Microfungal assemblages were compared for Abies needles and Betula leaf litter collected from litter and fermentation layers of the forest floor during the growing season in spring, summer, and autumn. A total of 35 and 42 species were isolated from Abies needles and Betula leaf litter, respectively. The observed variation in microfungal assemblages was primarily attributable to seasonal differences. The frequencies of Trichoderma viride, Volutella ciliata, Mucor sp., and Umbelopsis ramanniana increased in summer, leading to a high degree of similarity of microfungal assemblages in different litter depths and litter species. Microfungal assemblages on Abies needles in spring and autumn and those on Betula leaves in spring were characterized by Trichoderma viride, V. ciliata, Thysanophora penicillioides, Trichoderma polysporum, and (or) Mortierella alpina. Microfungal assemblages on Betula leaves in autumn were characterized by the absence of these species and the occurrence of Cladosporium cladosporioides. The results were discussed with an emphasis on the role of microfungi in decomposition processes and the impact on fungi of predicted future increases in global temperature.

Spruce needles of different age, litter materials and soil samples from the L-, O-and A-horizons of a mor profile were analysed by temperature-programmed pyrolysis (Py) in combination with field ionization mass spectrometry (FIMS). The integrated Py-FI mass spectra give characteristic fingerprints of the biomaterials investigated. The application of principal component analysis to the mass spectral data results in a clear discrimination and classification of the samples reflecting the chemical modifications and transformations of organic matter by biochemical and biogeochemical processes. The chemical compositions are determined by processes such as enrichment and/or translocation of plant constituents (e.g. carbohydrates, lignin, lipids, suberin, and aliphatic polymers) during maturation and senescence of needles; amendment of new components; decomposition; selective preservation and humification processes in the soil environment.During needle maturation, major chemical changes include the decrease of carbohydrate content, condensation of lignin, and crosslinking of waxes. Senescent needles are characterized by lower contents of carbohydrates and lower yields of monomeric pyrolysis products from lignin. The contribution of different litter materials to the humus layer can be estimated by differences in chemical composition. During litter decomposition and humification on the forest floor, carbohydrate content decreases rapidly. The lignin content remains almost constant but some subunits are continuously oxidized. Wax material accumulates until the mechnical disintegration of the needle occurs. In the O-horizons polymeric aliphatic materials are enriched in humified plant remains. A constant increase of aryl-alkyl esters from suberin in the O-horizons is due to both root input and selective preservation. In general, mainly aliphatic polymers and aryl-alkyl esters accumulate during the genesis of mor profiles under conifers.

Elevated atmospheric carbon dioxide (CO2) and ozone (O3) concentrations have both been shown to affect plant tissue quality, which in turn could affect litter decomposition and carbon (C) and nutrient cycling. In order to evaluate effects of climate change on litter chemistry, needle litter was collected from Scots pine (Pinus sylvestris L.) saplings exposed to elevated CO2 or O3 concentration and their combination over three growing seasons in open‐top chambers. The decomposition of needle litter was followed for 19 months in a pine forest. During decomposition, needle samples for secondary compound analysis were collected and the mass loss of needles was followed. Main nutrients and total phenolics were analysed from litter in the beginning and at the end of the experiment. After 19‐month decomposition, the accumulated mass loss was about 34%; however, no significant differences were found in decomposition rates of needle litter between various treatments. Concentrations of total monoterpenes were about 4%, total resin acids 21% and total phenolics 14% of the initial concentrations in litter after 19‐month decomposition. In the beginning of litter decomposition, concentrations of individual monoterpenes –α‐pinene and β‐pinene – were significantly higher in needle litter grown under elevated CO2. However, concentrations of total monoterpenes during the whole decomposition period were not significantly affected by CO2 or O3 treatments. Concentrations of some individual and total resin acids were higher in needle litter grown under elevated CO2 or O3 than under ambient air. There were no significant differences in concentrations of total phenolics as well as nitrogen (N) and the main nutrient concentrations between treatments during decomposition. High concentrations of monoterpenes and resin acids in needles might slightly delay C recycling in forest soils. It is concluded that elevated CO2 and O3 concentrations do not have remarkable impacts on litter decomposition processes in Scots pine forests.

Fungi play an important role in leaf litter decomposition due to their ability to break down the lignocellulose matrix, which other organisms are unable to digest. However, little is known regarding the factors affecting components of fungal diversity. Here, we quantified richness of internal fungi in relation to litter nutrient and phenolic concentrations, sampling season (spring or fall), and premature leaf shedding due to low precipitation and infestation of bark beetles (mainly Ips typographus and Ips duplicatus). The study was conducted in 37-year-old Norway spruce [Picea abies (L.) Karst.] stands, with three plots each in mixed forest (MF) and coniferous forest (CF) site conditions in south-central Poland. Fifty-four species of sporulating fungi were identified in 2,330 freshly fallen needles sampled during 2003-2005, including 45 species in MF and 31 in CF. The significantly higher number of species in MF was likely related to moister conditions at that site. Among isolated fungi, 22% (12 species) were identified as endophytes of Norway spruce in prior studies. During spring of 2005, we found less than half the number of isolates and fungal species at each forest site as compared to fall for the two prior years. This pattern was observed in typical soil fungi (e.g., Penicillium daleae, Penicillium purpurogenum) and endophytes/epiphytes (e.g., Aureobasidium pullulans, Alternaria alternata, Cladosporium spp., and Lophodermium piceae). Premature shedding of needles was the most likely cause of this decline because it shortened the time period for fungi to infect green needles while on the tree. For all sites and sampling periods, richness of internal fungi was strongly and positively related to the age of freshly fallen litter (assessed using needle Ca concentration as a needle age tracer) and was also negatively related to litter phenolic concentration. Richness of internal fungi in freshly fallen litter may be adversely affected by low soil moisture status, natural inhibitors slowing fungal colonization (e.g., phenolics) and biotic (e.g., insect infestation) and abiotic (e.g., drought) factors that shorten leaf life span.

Fungi play key roles in forest ecosystems and help to shape the forest’s diverse functions. However, little is known about the diversity of phyllospheric fungi or their possible relationships with fungal communities residing in different micro-environments of Pinus massoniana forests. We investigated seven different sample types: mature needles (NM), dead needles (ND), needles falling as litter (L), fermenting needles (F), humus (H), top soil (0–20 cm) (TS), and secondary soil (20–40 cm) (SS). These seven fungal communities were examined and compared with ITS amplicons using a high-throughput sequencing technique. A total of 1213 fungal operational taxonomic units (OTUs) were obtained at a 97% sequence similarity level. Distinct fungal communities were associated with different sample types. A greater number of OTUs were present in both NM and F samples than those shared by both NM and TS samples, indicating that phyllospheric fungi may play crucial roles in litter decomposition. Sixty OTUs (the core microbiome) were found in all sample types, and they may probably play different ecological roles in different sample types. These findings extend our knowledge of the fungal diversity of the phyllosphere and its possible interactions with fungal communities found in distinct forest micro-habitats.
