Eu3+对山黧豆中的氨基酸及蛋白质代谢作用的研究   总被引:3,自引:1,他引:2  
对山黧豆(Lathyrus sativus L.)幼苗整株,喷施一定浓度的Eu3+溶液吸收后,对其相关的生理指标进行测试,结果表明与水喷比较,根中丝氨酸、甘氨酸、丙氨酸、蛋氨酸、亮氨酸、脯氨酸、胱氨酸与对照持平,缬氨酸、酪氨酸、苯丙氨酸、组氨酸略有升高外,其余均低于对照;茎中除精氨酸、脯氨酸、色氨酸略高于对照外,其余均低于对照;叶中除蛋氨酸、色氨酸略高于对照外,其余均低于对照.游离氨基酸总量增加时,茎>根>叶,减少时,根<茎<叶.蛋白质含量经Eu3+ 处理茎中减少,根和叶中增加;水解酶比活性根>茎>叶;Na+、K+-ATPase活力却表现根>茎>叶的趋势.表明Eu3+ 对山黧豆生理活性代谢起到一定的调节作用.  相似文献

本试验测定了两熟茭白(Zizania latifolia )品种“广益”茭秋茭膨大期间肉质茎中游离氨基酸含量的动态变化。从“孕茭”始期开始,每4天取20株膨大肉质茎混匀。共测定6次。游离氨基酸用4%磺基水杨酸提取,然后用Waters-244型高压液相色谱仪分析。主要结果如下:1.茭白膨大过程中,所测得的18种游离氨基酸以天门冬氨酸、丝氨酸、丙氨酸、酪氨酸和组氨酸含量较高,而蛋氨酸和甘氨酸的含量相对较低,其它氨基酸含量中等。天门冬氨酸、苏氨酸(包括天门冬酰胺和谷酰胺)一直呈增加趋势。而丝氨酸谷氨酸甘氨酸丙氨酸缬氨酸酪氨酸和精氢酸在前期提高.后期下降:蛋  相似文献

通过室内培养试验,研究了赤子爱胜蚓(Eisenia foetida)和威廉环毛蚓(Metaphire guillelmi)对土壤氨基酸组分及含量的影响,并探讨了两种不同生活型蚯蚓作用效果的异同。结果表明:蚯蚓活动可显著改变土壤氨基酸含量,爱胜蚓作用下土壤酸解氨基酸和游离氨基酸分别增加5.08 g/kg和7.72 mg/kg,环毛蚓作用下土壤酸解氨基酸和游离氨基酸分别增加3.86 g/kg和4.44mg/kg。各处理酸解氨基酸均以中性氨基酸所占比例为最大(平均51.9%),酸性氨基酸次之(平均23.3%),而含硫氨基酸(平均14.4%)及碱性氨基酸最少(平均10.4%)。各处理游离氨基酸同样以中性氨基酸为主,平均54.4%,而以碱性氨基酸含量最少,平均仅为7.2%。蚯蚓活动并未改变土壤氨基酸可检出种类,各处理分别检测出16种酸解氨基酸和14种游离氨基酸。土壤酸解氨基酸和游离氨基酸组分含量在蚯蚓作用下均有明显改变:加入爱胜蚓后土壤酸解氨基酸组分中天冬氨酸、精氨酸、甲硫氨酸、丙氨酸、赖氨酸和甘氨酸增幅较高,均在85.7%以上,缬氨酸、苏氨酸、丝氨酸、谷氨酸、亮氨酸、酪氨酸和组氨酸增幅较小在40.7%—62.7%间波动;加入环毛蚓后土壤酸解氨基酸组分中甲硫氨酸、赖氨酸、天冬氨酸、酪氨酸和丙氨酸增幅较大,均在71.9%以上,甘氨酸、精氨酸、异亮氨酸增幅适中,分别为56.8%、55.6%和54.9%;丝氨酸、亮氨酸、苏氨酸、谷氨酸、组氨酸和苯丙氨酸增幅最小,均在40%以下;游离氨基酸组分中组氨酸、精氨酸、甘氨酸、亮氨酸、异亮氨酸和丙氨酸在加入爱胜蚓后增加的幅度较大,增幅在150.0%以上,增幅较为缓和的氨基酸组分有天冬氨酸、苏氨酸、丝氨酸、缬氨酸、谷氨酸和苯丙氨酸,介于58.8%—92.1%之间;环毛蚓作用下,天冬氨酸、精氨酸、丝氨酸和异亮氨酸增幅最大,分别为184.2%、173.3%、163.0%和116.6%;苏氨酸、亮氨酸、缬氨酸和甘氨酸增幅较缓,介于52.3%—92.7%之间;谷氨酸、组氨酸、苯丙氨酸、丙氨酸和甲硫氨酸增幅较低,均在33.1%之下;而半胱氨酸在蚯蚓作用下显著降低,降幅为11.8%。对比两种生活型蚯蚓作用效果可知,土壤氨基酸总含量及各组分含量在爱胜蚓和环毛蚓作用下的增加或减少趋势相同(土壤酸解氨基酸组分缬氨酸除外),但改变幅度却存在明显差异,总体而言,爱胜蚓作用效果优于环毛蚓。  相似文献

作者研究了白蜡虫越冬前后体内游离氨基酸的组成及变化 ,并通过对虫体、蜜露及被寄生植物游离氨基酸含量及百分率组成进行对比分析 ,探讨了白蜡虫对游离氨基酸的利用问题。虫体含量较高的氨基酸有丙氨酸、谷氨酸、苏氨酸、丝氨酸、缬氨酸。大部分氨基酸及其总量在越冬期下降 ,越冬以后升高 ,逐渐超过越冬前水平。虫体大量吸收的氨基酸是丙氨酸 ,利用较少的氨基酸是天门冬氨酸 ,其它氨基酸或多或少地为白蜡虫所利用。  相似文献

分析比较了 33种不同来源的氨基酸产品红外图谱的差异 ,其中丝氨酸、门冬氨酸、醋酸赖氨酸、谷氨酸 (白色结晶性粉末 )、苏氨酸、缬氨酸、丙氨酸、亮氨酸、脯氨酸、盐酸组氨酸、盐酸精氨酸、酪氨酸、胱氨酸等 13种与标准图谱完全一致 ;甲硫氨酸、盐酸赖氨酸、甘氨酸、谷氨酸 (白色结晶 )等 4种与标准图谱不一致 ,其原因是 :甘氨酸和谷氨酸由晶型不同造成 ,甲硫氨酸因旋光性不同而造成 ,盐酸赖氨酸与相应的生化试剂图谱一致。  相似文献

水文研究了不同发育阶段蓖麻蚕组织中可溶性蛋白质在性质及含量上的变化。在各发育阶段可溶性蛋白质都包含有清蛋白类、球蛋白类及明胶。在化蛹初期, 组织提取液中还发现了初级(月示)和胨。除明胶以外的可溶性蛋白质总量及其中清蛋白类的含量因发育阶段而异:在蛹初期最高, 到羽化前后有显著下降, 球蛋白类的含量在各发育阶段比较稳定。雌雄两性的可溶性蛋白质含量不同, 在羽化前后这种两性差异最为显著, 雌性超过雄性约一倍之多。因此, 在蛹后期及成虫中, 蛋白质的两性差异在性质上比在数量上更为显著。 曾用纸上层析法分析了可溶性蛋白质的氨基酸组分。在各发育阶段, 氨基酸的种类相同, 包括胱氨酸(和半胱氨酸)、赖氨酸、组氨酸、精氨酸、丝氨酸、甘氨酸、天门冬氨酸、谷氨酸、苏氨酸、丙氨酸、脯氨酸、酪氨酸、缬氨酸(和蛋氨酸)、苯丙氨酸、以及亮氨酸(和异亮氨酸)。色氨酸在酸水解过程中被破坏。文中列出不同发育阶段可溶性蛋白质中各种氨基酸的百分含量。老熟幼虫吐丝结茧后, 甘氨酸、丙氨酸、酪氨酸和丝氨酸含量下降。  相似文献

以5种对枯萎病不同抗性黄瓜品种为试材,对其根系分泌物氨基酸组分进行测定,并对氨基酸组分与黄瓜品种枯萎病抗病性之间的相关性进行了分析。结果表明:在中抗品种根系分泌中检测到的16种氨基酸:半胱氨酸Cys、苏氨酸Thr、丙氨酸Ala、缬氨酸Val、异亮氨酸Lle、天冬氨酸Asp、亮氨酸Leu、苯丙氨酸Phe、甘氨酸Gly、甲硫氨酸Met、组氨酸His、谷氨酸Glu、酪氨酸Tyr、赖氨酸Lys、丝氨酸Ser和精氨酸Arg。其中的精氨酸在感病品种中没有被检出,组氨酸和精氨酸组分在抗病品种中没有被检出。根系分泌物中总氨基酸含量随品种抗性的增加而降低;精氨酸、丝氨酸和赖氨酸的含量与品种对枯萎病的病情指数呈负相关,其他13种氨基酸组分含量与品种对枯萎病的病情指数呈正相关,其中苯丙氨酸含量与病情指数呈显著正相关。丝氨酸与苯丙氨酸、天冬氨酸、丙氨酸、甘氨酸的比值均与品种对枯萎病的病情指数呈显著负相关,其中Ser/Phe与品种对枯萎病的病情指数呈极显著负相关。  相似文献

报道了1个世代中紫胶蚧Kerria laccaKerr寄生对寄主植物营养成分产生的影响。结果表明:紫胶蚧寄生对寄主植物营养成分有影响,被寄生枝条的氨基酸、无机盐和营养成分总量都出现变化。以久树Schleichera oleosa(Lour)为例,紫胶蚧寄生使久树枝条中氨基酸总量减少了35%。其中,天门冬氨酸、苏氨酸、丝氨酸、甘氨酸、丙氨酸、缬氨酸、异亮氨酸、亮氨酸、酪氨酸、苯丙氨酸的含量也都存在不同程度减少。但是,蛋氨酸、组氨酸、赖氨酸、精氨酸、色氨酸含量比对照枝条中高。被寄生枝条中氮、磷(P2O5)、铁等呈现下降趋势,钠元素含量几乎没有发生变化,而钾、钙、镁、铜、锌、锰等却有不同程度的增加,特别是钙元素含量,被寄生枝条中钙元素的含量是对照枝条的5.3倍左右。紫胶蚧寄生对枝条中总糖、蛋白质、脂肪、粗纤维、水溶性物质以及灰分含量都产生不同程度影响,被寄生枝条中蛋白质、粗纤维含量比对照枝条中含量低;而被寄生枝条中总糖、脂肪、水溶性物质、灰分含量比对照枝条中高。  相似文献

应用高效液相色谱(HPLC)技术,首次测定了湖北石首长江天鹅州白豚自然保护区野生长江江豚(Neophocaena phocaenoides asiaeorientalis)和中国科学院水生生物研究所白豚馆人工饲养的长江江豚血清中17种氨基酸的含量.结果表明,除了脯氨酸Pro、蛋氨酸Met和组氨酸His外,人工饲养江豚血清中其余14种氨基酸(天门冬氨酸Asp、谷氨酸Glu、丝氨酸Ser、精氨酸Arg、甘氨酸Gly、苏氨酸Thr、丙氨酸Ala、异亮氨酸Ile、亮氨酸Leu、苯丙氨酸Phe、缬氨酸Val、赖氨酸Lys、酪氨酸Tyr、胱氨酸Cys)的含量显著高于野生长江江豚血清中相应氨基酸的含量.野生江豚和人工饲养江豚的血清氨基酸含量均没有显著的性别差异.野生江豚性成熟个体与未成熟个体之间血清氨基酸含量也没有显著性的差异.在所检测的17种氨基酸中,豢养江豚Glu含量最高,其次为Asp和Lys.野生江豚同样是Glu最高,其次是Lys和Asp.豢养和野生江豚都是Met含量最低.野生和豢养江豚必需氨基酸(EAA)和非必需氨基酸(NEAA)之间的比率分别是0.83和0.92,具有极显著的差异(p <0.01).  相似文献

对尾叶桉60个无性系进行了生长量和桉树枝瘿姬小蜂的敏感性调查。结果表明, 生长性状在无性系单株间存在显著的相关性,而尾叶桉受姬小蜂的危害情况与生长性状间无显著相关性。对遗传参数的估算表明, 尾叶桉无性系抗桉树枝瘿姬小蜂水平有较高的重复力,可以通过无性系选择获得抗蜂、速生的尾叶桉无性系。最终通过独立淘汰法选择出6个优良的尾叶桉无性系。  相似文献

A common consequence of the predation or physical disturbance of sessile modular animals is the removal of part of the colony. This damage is often sub-lethal, and leaves the remainder of the colony to repair and regenerate lost tissue. Damage may, however, have lasting effects on the life history characteristics of a colony. We examined the effects of damage on growth and reproduction in the arborescent bryozoan Bugula neritina (Linnaeus), with a particular focus on the effects of the location of that damage. We predicted that a damaged colony would initially allocate more energy to damage repair and subsequent growth, increasing in size to decrease the risk of mortality, at the expense of its short-term reproductive potential. A short-term experiment assessed the immediate regenerative capacities of colonies, while a longer-term experiment measured overall colony responses to damage at different colony locations. In this experiment, zooids were removed in three colony regions; all on one colony branch, spread evenly across all growing tips, and a third, intermediate pattern. A third experiment tested the effects of predator-induced damage versus damage alone, by measuring the recovery of colonies subjected to either a predation event by the nudibranch Polycera hedgpethi, or to an imitation predation event, where an equivalent section of the colony was removed manually. Regeneration was rapid, with budding resuming a few days after damage occurred. Removing around 20% of the colony did not consistently alter colony growth rates in any experiment, but the onset of reproduction was delayed by 7 to 20 days, and subsequent colony fecundity reduced by as much as 70% in one damage treatment. The removal of the same amount of tissue by nudibranch predators, or manually removed in a treatment that mimicked this predation, had no detrimental effect on the onset of reproduction or the eventual reproductive output. Nudibranchs removed tissue by consuming whole colony branches, and the experimental treatment that removed one branch also had the least effect of three damage treatments. These results suggest the severity of long-term effects depends on the location of the injury, and that B. neritina does not modify its growth or reproductive patterns to minimise these effects.  相似文献

Selected interactions between the encrusting sponge Halichondria panicea and its primary predator, the dorid nudibranch Archidoris montereyensis, were investigated in a high-latitude rocky intertidal community spatially dominated by H. panicea. Feeding experiments were conducted in which A. montereyensis pairs were provided with sponge containing symbiotic zoochlorellae or sponge in which the zoochlorellae population had been reduced or removed by shading. Nudibranchs consuming H. panicea with symbiotic zoochlorellae had higher feeding, growth, and egg production rates than individuals eating aposymbiotic sponge. We simulated A. montereyensis predation on H. panicea by creating typically sized feeding grooves in the sponge. H. panicea's response was high linear growth rates into the experimental feeding grooves, generally recovering most of the groove area within 4 weeks. Overall, the sponge's rapid response to tissue damage minimizes grazing impacts and substrate loss and reduces susceptibility to wave removal.  相似文献

The importance of bacteria in the anaerobic bioremediation of groundwater polluted with organic and/or metal contaminants is well recognized and in some instances so well understood that modeling of the in situ metabolic activity of the relevant subsurface microorganisms in response to changes in subsurface geochemistry is feasible. However, a potentially significant factor influencing bacterial growth and activity in the subsurface that has not been adequately addressed is protozoan predation of the microorganisms responsible for bioremediation. In field experiments at a uranium-contaminated aquifer located in Rifle, CO, USA, acetate amendments initially promoted the growth of metal-reducing Geobacter species, followed by the growth of sulfate reducers, as observed previously. Analysis of 18S rRNA gene sequences revealed a broad diversity of sequences closely related to known bacteriovorous protozoa in the groundwater before the addition of acetate. The bloom of Geobacter species was accompanied by a specific enrichment of sequences most closely related to the ameboid flagellate, Breviata anathema, which at their peak accounted for over 80% of the sequences recovered. The abundance of Geobacter species declined following the rapid emergence of B. anathema. The subsequent growth of sulfate-reducing Peptococcaceae was accompanied by another specific enrichment of protozoa, but with sequences most similar to diplomonadid flagellates from the family Hexamitidae, which accounted for up to 100% of the sequences recovered during this phase of the bioremediation. These results suggest a prey–predator response with specific protozoa responding to increased availability of preferred prey bacteria. Thus, quantifying the influence of protozoan predation on the growth, activity and composition of the subsurface bacterial community is essential for predictive modeling of in situ uranium bioremediation strategies.  相似文献

Modulation of death is a pathogen strategy to establish residence and promote survival in host cells and tissues. Shigella spp. are human pathogens that invade colonic mucosa, where they provoke lesions caused by their ability to manipulate the host cell responses. Shigella spp. induce various types of cell death in different cell populations. However, they are equally able to protect host cells from death. Here, we have investigated on the molecular mechanisms and cell effectors governing the balance between survival and death in epithelial cells infected with Shigella. To explore these aspects, we have exploited both, the HeLa cell invasion assay and a novel ex vivo human colon organ culture model of infection that mimics natural conditions of shigellosis. Our results definitely show that Shigella induces a rapid intrinsic apoptosis of infected cells, via mitochondrial depolarization and the ensuing caspase-9 activation. Moreover, for the first time we identify the eukaryotic stress-response factor growth arrest and DNA damage 45α as a key player in the induction of the apoptotic process elicited by Shigella in epithelial cells, revealing an unexplored role of this molecule in the course of infections sustained by invasive pathogens.  相似文献

Many shallow water subtidal habitats in Massachusetts, USA have recently been invaded by five non-indigenous ascidian species: Ascidiella aspersa, Botrylloides violaceus, Didemnum sp., Diplosoma listerianum and Styela clava. This study examined the effects of seawater temperature, as a proxy for climate change, on B. violaceus and D. listerianum and the impact these ascidians have on native sessile fouling communities. Field experiments were conducted over a four month period at two locations (Lynn and Woods Hole, MA) to examine growth dynamics over regional thermal and geographic ranges. Invasive ascidians occupied as much as 80% of the primary substratum and accounted for the majority of species richness. B. violaceus and D. listerianum growth were similar at both study sites, but initial colony growth of D. listerianum was positively affected by temperature. B. violaceus and D. listerianum exhibited rapid two-week growth rates during the summer months with more rapid growth at the warmer Woods Hole site. Competition for space between B. violaceus and D. listerianum typically resulted in neutral borders between colonies. Overgrowth occurred if the colony of one species was disproportionably larger than the colony of the other species. Recruitment and growth of native species influenced the long-term composition of experimental communities more than the pre-seeding with B. violaceus or D. listerianum colonies. Elevated temperatures, however, increased initial growth of B. violaceus and D. listerianum and may have facilitated the species success to invade the communities during crucial periods of introduction. With projected global climate change, a rise in sea surface temperatures may exacerbate the cumulative impacts of invasions on benthic communities and facilitate the invasion of other non-native ascidian species.  相似文献

In the healthy adult brain microglia, the main immune-competent cells of the CNS, have a distinct (so-called resting or surveying) phenotype. Resting microglia can only be studied in vivo since any isolation of brain tissue inevitably triggers microglial activation. Here we used in vivo two-photon imaging to obtain a first insight into Ca2+ signaling in resting cortical microglia. The majority (80%) of microglial cells showed no spontaneous Ca2+ transients at rest and in conditions of strong neuronal activity. However, they reliably responded with large, generalized Ca2+ transients to damage of an individual neuron. These damage-induced responses had a short latency (0.4-4 s) and were localized to the immediate vicinity of the damaged neuron (< 50 μm cell body-to-cell body distance). They were occluded by the application of ATPγS as well as UDP and 2-MeSADP, the agonists of metabotropic P2Y receptors, and they required Ca2+ release from the intracellular Ca2+ stores. Thus, our in vivo data suggest that microglial Ca2+ signals occur mostly under pathological conditions and identify a Ca2+ store-operated signal, which represents a very sensitive, rapid, and highly localized response of microglial cells to brain damage. 对阿尔山落叶松主要蛀干害虫的种群生态位进行了研究,结果表明:在落叶松衰弱立木上,生态位宽度以落叶松八齿小蠹最高,在中龄和老龄树上分别达到0.5782、0.5498,在落叶松中龄和老龄枯死立木上,白带长角天牛的空间生态位宽度分别达到0.5143、0.6294,分布较衰弱木广,而云杉大黑天牛空间生态位宽度在衰弱和枯死立木上都最低。落叶松立木上的空间生态位重叠指数以落叶松八齿小蠹对其他3种优势天牛偏高,天牛之间的空间生态位相似性比例指数均较高且差距较小,说明4种害虫对空间生态位的占据有很大相似程度。4种害虫的空间种间竞争系数均很大,说明对空间和资源的竞争非常激烈,在这种情况下4种害虫能够共存,根本原因是由于取食部位的分化。  相似文献

Xanthone production in Hypericum perforatum (HP) suspension cultures in response to elicitation by Agrobacterium tumefaciens co-cultivation has been studied. RNA blot analyses of HP cells co-cultivated with A. tumefaciens have shown a rapid up-regulation of genes encoding important enzymes of the general phenylpropanoid pathway (PAL, phenylalanine ammonia lyase and 4CL, 4-coumarate:CoA ligase) and xanthone biosynthesis (BPS, benzophenone synthase). Analyses of HPLC chromatograms of methanolic extracts of control and elicited cells (HP cells that were co-cultivated for 24 h with A. tumefaciens) have revealed a 12-fold increase in total xanthone concentration and also the emergence of many xanthones after elicitation. Methanolic extract of elicited cells exhibited significantly higher antioxidant and antimicrobial competence than the equivalent extract of control HP cells indicating that these properties have been significantly increased in HP cells after elicitation. Four major de novo synthesized xanthones have been identified as 1,3,6,7-tetrahydroxy-8-prenyl xanthone, 1,3,6,7-tetrahydroxy-2-prenyl xanthone, 1,3,7-trihydroxy-6-methoxy-8-prenyl xanthone and paxanthone. Antioxidant and antimicrobial characterization of these de novo xanthones have revealed that xanthones play dual function in plant cells during biotic stress: (1) as antioxidants to protect the cells from oxidative damage and (2) as phytoalexins to impair the pathogen growth.  相似文献
