This work aimed to discuss the effects of exogenous abscisic acid (ABA) on the root growth regulation of maize seedlings under chilling stress. The roots of the maize cultivar Zhengdan 958 were irrigated with ABA (10?7, 10?6, 10?5 and 10?4 M) at the third true leaf stage under chilling duration (0, 2, 4, 6, and 8 days). The biomass, the phenylalanine ammonia lyase (PAL), and polyphenol oxidase (PPO) enzyme activities, total phenolic and flavonoid contents, the ferric reducing ability of plasma (FRAP) antioxidant capacity, and 2,2-azinobis (3-ethlbenzothiazo-line-6-sulfonic acid) diammonium salt radical (ABTS·+) scavenging capacity of the roots of maize seedlings were measured after the treatment. The results showed that appropriate concentrations of exogenous ABA effectively enhanced root biomass, increased PAL and PPO enzyme activities, and significantly increased total phenolic contents and flavonoid contents. Moreover, the ABA markedly improved the FRAP antioxidant capacity and ABTS·+ scavenging capacity under low-temperature stress. These results indicate that ABA-treated maize seedlings are resistant to chilling stress and that the optimum concentration of ABA is 10?5 M. Exogenous applications of ABA have a concentration effect in alleviating chilling stress, in which low concentrations have a promoting effect and high concentrations have an inhibiting effect.  相似文献

To investigate the influence of exogenous polyamines (PAs) on the root morphogenesis and arbuscular mycorrhizal development of Citrus limonia Osbeck seedlings, experiments were carried out in pots or specially designed containers. Analysis using WinRHIZO® sofware indicated that exogenous PAs significantly reduced the average root diameter, which was due to the increased percentage of fine roots and the decreased percentage of coarse roots. Root length and the 1st lateral number were also slight increased. The increase in the mitotic index of root tip cells revealed using hydroponic culture indicated the involvement of the mitosis process in the PA-induced changes in root morphogenesis. These changes finally led to the increases in the relative chlorophyll contents (SPAD) and the biomass of seedlings. Moreover, exogenous PAs affected the mycorrhizal colonization in a concentration-dependent manner, with low concentration (0.2 × 10?4 M) increasing but high concentration (1.0 × and 5.0 × 10?4 M) decreasing it. These results suggest that exogenous PAs can affect positively the growth of C. limonia seedlings via the pathway of altered root morphogenesis and mycorrhizal development.  相似文献

A hydroponic experiment was carried out to study the physiological mechanisms of N-acetyl cysteine (NAC) in mitigating cadmium (Cd) toxicity in two barley (Hordeum vulgare L.) genotypes, Dong 17 (Cd-sensitive) and Weisuobuzhi (Cd-tolerant). Addition of 200 μM NAC to a culture medium containing 5 μM Cd (Cd + NAC) markedly alleviated Cd-induced growth inhibition and toxicity, maintained root cell viability, and dramatically depressed O 2 ·? and ·OH, and malondialdehyde accumulation, significantly reduced Cd concentration in leaves and roots, especially in the sensitive genotype Dong 17. External NAC counteracted Cd-induced alterations of certain antioxidant enzymes, e.g., brought root superoxide dismutase and glutathione reductase, leaf/root peroxidase and glutathione peroxidase activities of the both genotypes down towards the control level, but elevated Cd-stress-depressed leaf catalase in Dong 17 and root ascorbate peroxidase activities in both genotypes. NAC counteracted Cd-induced alterations in amino acids and microelement contents. Furthermore, NAC significantly reduced Cd-induced damage to leaf/root ultrastructure, e.g. the shape of chloroplasts in plants treated with Cd + NAC was relatively normal with well-structured thylakoid membranes and parallel pattern of lamellae but less osmiophilic plastoglobuli compared with Cd alone treatment; nuclei of root cells were better formed and chromatin distributed more uniformly in both genotypes. These results suggested that under Cd stress, NAC may protects barley seedlings against Cd-induced damage by directly and indirectly scavenging reactive oxygen species and by maintaining stability and integrity of the subcellular structure.  相似文献

The present study aimed to analyze the biological effects induced by bioaccumulation of metals in common bean (Phaseolus vulgaris L.). Effects of mineral nutrient imbalance, total antioxidants level and DNA damage induced by accumulation of heavy metals, were investigated in bean seedlings treated with two selected metal concentrations for 7 days. Metal content is analyzed by inductively coupled plasma – atomic emission spectrometer (ICP-AES), for total antioxidants level assessment the Ferric-Reducing Antioxidant Power (FRAP) assay is used and Random Amplified Polymorphic DNA (RAPD) method was applied for investigation of DNA damages. The increasing metal concentration in the treatment medium changed synchronously metal content in samples, and decreased total antioxidant activity in all samples with exception only for samples treated with Ni and Cd. The obtained “DNA fingerprints” demonstrated that the increasing metal concentrations induced changes in RAPD profiles (disappearance and/or appearance of bands in comparison with untreated control samples). The highest number of missing bands was observed in samples treated with zinc (total 4 bands) and nickel (total 4 bands) at both concentrations. These results suggested that mineral nutrient imbalance is involved in changes of antioxidant levels and DNA damages of the seedlings, which may help to understand the mechanism of metal toxicity in plants.  相似文献

The effect of B toxicity on antioxidant responses of soybean (Glycine max) cv. Athow was investigated by growing plants for 43 days at 0.2 (control), 2 and 12 mg B kg?1. At the end of the treatment period, shoot growth, lipid peroxidation level, the activities of antioxidant enzymes including superoxide dismutase (SOD), peroxidase (POX), catalase (CAT), ascorbate peroxidase (APX) and glutathione reductase (GR), and their isoenzymes in leaves were measured. Boron concentration in leaves was significantly increased by the increasing levels of B treatment from 43 to 522 mg kg?1, and shoot dry matter was depressed at 12 mg B kg?1. Significant increases in SOD, CAT, and APX activities were determined in leaves under 12 mg B kg?1; however, GR activities were decreased while POX activity was unchanged. Increased enzymic antioxidant activity arose from a combination of newly formed isoenzymes and activation of existing isoenzymes. By contrast, SOD and GR activities were decreased by 2 mg B kg?1 concentration as compared to the control groups while POX activity was increased and the activity of CAT did not change. Malondialdehyde content increased under 2 mg B kg?1 but decreased under 12 mg B kg?1. These results suggest that higher antioxidant activity observed under 12 than at 2 mg B kg?1 provided higher free radical-scavenging capacity, and thus a lower level of lipid peroxidation in Athow. While the induction of increased antioxidant activity was related to internal boron levels, the signaling and coordination of responses remain unclear.  相似文献

This study was undertaken to evaluate genotoxic potential of Thermopsis turcica aqueous extracts on the roots of onion bulb (Allium cepa L.) by comet assay and random amplified polymorphic DNA technique. The Allium root growth inhibition test indicated that the EC50 and 2×EC50 values were 8 and 16 mg/ml concentrations of T. turcica aqueous extracts, respectively. The negative control (distilled water), positive control (methyl methane sulfonate, 10 mg/l) and 8 and 16 mg/ml concentrations of T. turcica extracts were introduced to the roots of onion bulbs for 24 and 96 h. The root growth, DNA damage in root cells and randomly amplified polymorphic DNA (RAPD) profiles of root tissue were used as endpoints of the genotoxicity. The comet assay clearly indicated that dose-dependent single strand DNA breaks in the root nuclei of onions were determined for the treatment concentrations of T. turcica extracts. In comparison to RAPD profile of negative control group, RAPD polymorphisms became evident as disappearance and/or appearance of RAPD bands in treated roots. The diagnostic and phenetic numerical analyses of RAPD profiles obviously indicated dose-dependent genotoxicity induced by Thermopsis extracts. In conclusion, the results clearly indicated that water extract of T. turcica has genotoxic potential on the roots of onion bulbs as shown by comet assay and RAPD technique.  相似文献

The phytoremediation potential and responses of Sagittaria trifolia to diesel were investigated. In order to elucidate the biochemical and physiological responses of S. trifolia to diesel, the chlorophyll content, root vitality, soluble protein content and antioxidant enzymes activity (peroxidase (POD), catalase (CAT) and antioxidant enzymes superoxide dismutase (SOD)) were determined in the plant tissues after 50 d of diesel treatment. The results showed the presence of S. trifolia significantly improved the removal ratios of diesel, from 21~36% in the control soils to 54~85% in the planted soils. The chlorophyll content, root vitality and soluble protein content all increased at low diesel concentration, then decreased at high diesel concentration. The activities of CAT and POD exhibited peak values at 5 g·kg?1 diesel treatment and declined at higher diesel concentrations. However, the activity of SOD kept stable at lower diesel concentration (1 and 5 g·kg?1), and also declined at higher diesel concentration. Collectively, S. trifolia had the ability to tolerate certain amount of diesel, but when the concentration was up to 10 g·kg?1, the growth of S. trifolia would be restrained. The results also showed that variation of antioxidant enzyme activity was an important response in plants to diesel pollution.  相似文献

Bone is one of the main target organs for the lanthanides (Ln). Biodistribution studies of Tm-based compounds in vivo showed that bone had significant uptake. But the effect of Tm3+ on primary mouse bone marrow stromal cells (BMSCs) has not been reported. So we investigated the effect and underlying mechanisms of Tm3+ on BMSCs. Cell viability, cell apoptosis, reactive oxygen species (ROS) level, lactate dehydrogenase (LDH) activity and mitochondrial membrane potential (MMP) were studied. The results indicated that Tm3+ increased the viability of BMSCs at concentrations of 1?×?10?7, 1?×?10?6, 1?×?10?5, and 1?×?10?4 mol/L in a dose-dependent manner, turned to decrease the viability of BMSCs at the highest concentration of 1?×?10?3 mol/L for 24, 48, and 72 h. Tm3+ at 1?×?10?3 mol/L promoted apoptosis of BMSCs, increased the ROS and LDH levels, and decreased MMP in BMSCs. Taken together, we demonstrated that Tm3+ at 1?×?10?3 mol/L might induce cellular apoptosis through mitochondrial pathway. These results may be helpful for more rational application of Tm-based compounds in the future.  相似文献

Iris pseudacorus L. (yellow flag) is a wide-use wetland plant for constructed wetlands for removing metals from wastewater. This study aims to understand effects of root iron plaque on sequestration and translocation of Cr and Ni in yellow flag seedlings using a hydroponic experiment. Yellow flag seedlings (4-week-old seedlings with 4–6 leaves) with or without iron plaque induction (at 50 mg Fe2+ L?1 for 72 hours) were spiked for 6 days in the Hoagland solution with Cr or Ni at 0.5, 5, and 50 mg L?1, equivalent to 1, 10, 100 times of thresholds of surface water quality, respectively. Results indicated that root iron plaque significantly reduced translocation of Cr and Ni to root but increased from root to shoot. Root iron plaque formation counteracted Cr toxicity to yellow flag seedlings while the control showed Cr toxicity to root at 5 mg L?1and to shoot at 50 mg L?1 with significant biomass loss. Neither Ni exposures caused significant biomass loss nor root iron plaque formation significantly changed Ni distribution among plant parts. Our study suggests that root iron plaque effects on metal sequestration and translocation in yellow flag seedlings were metal-dependent.  相似文献

Plants grown at limited P supply can increase the activity of phosphatases in roots to hydrolyse organic-P compounds in the soil thus improving plant P acquisition, but little information is available about the role of these enzymes for internal plant metabolism at limited-P conditions. This work intended to measure the activities of acid phosphatases and phytases in nodules of common bean (Phaseolus vulgaris) genotypes at different levels of P supply. The experiment was carried out in a 5?×?5 factorial design with four replicates, comprising five bean genotypes and five P levels (20, 40, 80, 160 and 320 μmol P plant?1 week?1) in nutrient solution. Root seedlings were inoculated with Rhizobium tropici and plants were grown in 1-l bottles. Nodule samples were detached from 39-day-old plants and enzyme activities were determined in crude extracts. Plants were harvested at the stage of pod setting. Polynomial models fitted to data indicated maximal values at the level of 194 μmol P for shoot mass, at 206 μmol P for nodule mass and at 221 μmol P for shoot N. Whereas shoot mass was 1.7 times lower at 20 than at 160 μmol P, nodule mass was 7.5 times lower. Concentration of P in nodules increased from 40 to 320 μmol P but remained stable between 20 and 40 μmol P, suggesting a minimal threshold concentration of 3 mg P g?1 for nodule growth. Activities of phosphatases and phytases in nodules decreased strongly as P supply was raised from 20 to 80 μmol P, remaining almost stable at higher P levels. Phosphatase activity ranged from 1,154 to 406 nmol min?1 g?1 (nodule fresh mass) from 20 to 80 μmol P respectively, while the phytase activity ranged from 55 to 14 nmol min?1 g?1 from 20 to 80 μmol P. Bean genotypes differed in shoot and nodule mass at the levels of 80 and 160 μmol P, whilst they differed in nodule enzyme activities only at the lowest P level, the relationship between nodule enzyme activities and growth of different bean genotypes was not evident. It is concluded that bean plants at P-deficient conditions increase the activities of phosphatases and phytases in nodules. This may constitute an adaptive mechanism for N2-fixing legumes to tolerate P deficiency, by increasing the utilisation of the scarce P within the nodules.  相似文献

Crop yield is severely affected by soil salinity, as salt levels that are harmful to plant growth occur in large terrestrial areas of the world. The present investigation describes the studies of enzymatic activities, in-gel assays, gene expression of some of the major antioxidative enzymes, tocopherol accumulation, lipid peroxidation, ascorbate and dehydroascorbate contents in a salt-sensitive rice genotype PB1, and a relatively salt-tolerant cultivar CSR10 in response to 200 mM NaCl. Salt solution was added to the roots of hydroponically grown 5-day-old etiolated rice seedlings, 12 h prior to transfer to cool white fluorescent?+?incandescent light (100 μmol photons m?2 s?1). Total tocopherol and ascorbate contents declined in salt-stressed rice seedlings. Among antioxidative enzymes, an increase in the activities of superoxide dismutase (EC 1.15.1.1), catalase (EC 1.11.1.6), ascorbate peroxidase (EC 1.11.1.11), glutathione reductase (EC 1.6.4.2), and their gene expression was observed in both cultivars in response to salt stress. The salt-tolerant cultivar CSR10 resisted stress due to its early preparedness to combat oxidative stress via upregulation of gene expression and enzymatic activities of antioxidative enzymes and a higher redox status of the antioxidant ascorbate even in a non-stressed environment.  相似文献

The hazardous potential of the Metosulam herbicide, particularly the cytogenetic and physiological effects on Vicia faba cv Assuit 25 plants has been studied. The results showed that the mitotic index (MI) decreased and chromosomal aberrations frequency increased by increasing of the concentration of herbicide and prolonging the duration of treatment. In the roots treated with highest concentration used (1 × 10?5 %) for 24 h, complete inhibition of cell division was observed. The chromosomal anomalies include chromosomal bridges and breaks that are regarded were indicative of a mutagenic potential of the herbicide. Seedling growth (fresh and dry weight) adversely affected as the duration and concentration of Metosulam herbicide increased. Soluble sugars, soluble proteins, total free amino acids and photosynthetic pigment content decreased significantly in root, stem and leaves of Vicia faba with increasing both the herbicide concentration and treatment duration. In contrast, proline content was highly accumulated, especially at the highest concentration (10?4 %) and the longest duration used (24 h). The results of antioxidant enzymes reveal that while the peroxidase activity decreased by increasing the concentration of herbicide and duration, the activities of catalase and ascorbate peroxidase increased.  相似文献

Brassinosteroids (Brs) are a newly recognized group of active steroidal hormones that occur at low concentrations in all plant parts and one of the active and stable forms is 24-epibrassinolide (EBR). We investigated the effect of EBR on tomato (Lycopersicon esculentum Mill.) and its mechanism when seedlings were exposed to low temperature and poor light stress conditions. Leaves of stress-tolerant ‘Zhongza9’ and stress-sensitive ‘Zhongshu4’ cultivars were pre-treated with spray solutions containing either 0.1 μM EBR or no EBR (control). The plants were then transferred to chambers where they were exposed to low temperatures of 12 °C/6 °C (day/night) under a low light (LL) level of 80 μmol?·?m?2?·?s?1. Exogenous application of EBR significantly increased the antioxidant activity of superoxide dismutase, catalase and peroxidase, and decreased the rate of O2?·?? formation and H2O2 and malondialdehyde contents. Additionally, the ATP synthase β subunit content was increased by exogenous hormone application. Based on these results, we conclude that exogenous EBR can elicit synergism between the antioxidant enzyme systems and the ATP synthase β subunit so that scavenging of reactive oxygen species becomes more efficient. These activities enable plants to cope better under combined low temperature and poor light stresses.  相似文献

The present study is aimed at assessing the extent of arsenic (As) toxicity under three different light intensities—optimum (400 μmole photon m?2 s?1), sub-optimum (225 μmole photon m?2 s?1), and low (75 μmole photon m?2 s?1)—exposed to Helianthus annuus L. var. DRSF-113 seedlings by examining various physiological and biochemical parameters. Irrespective of the light intensities under which H. annuus L. seedlings were grown, there was an As dose (low, i.e., 6 mg kg?1 soil, As1; and high, i.e., 12 mg kg?1 soil, As2)-dependent decrease in all the growth parameters, viz., fresh mass, shoot length, and root length. Optimum light-grown seedlings exhibited better growth performance than the sub-optimum and low light-grown seedlings; however, low light-grown plants had maximum root and shoot lengths. Accumulation of As in the plant tissues depended upon its concentration used, proximity of the plant tissue, and intensity of the light. Greater intensity of light allowed greater assimilation of photosynthates accompanied by more uptake of nutrients along with As from the medium. The levels of chlorophyll a, b, and carotenoids declined with increasing concentrations of As. Seedlings acquired maximum Chl a and b under optimum light which were more compatible to face As1 and As2 doses of As, also evident from the overall status of enzymatic (SOD, POD, CAT, and GST) and non-enzymatic antioxidant (Pro).  相似文献

Maize colonization by the phytopathogenic fungi Fusarium verticillioides leads to economical and food quality losses and also implicates potential health risks. In order to control this fungal species different strategies are being considered. In the present work we investigated the in situ effects of the use of two in vitro proven bacterial biological control agents against Fusarium verticillioides, using maize seedlings grown in greenhouse conditions. The treatment of maize seeds with Fusarium verticillioides 107 spores ml?1 negatively affected the length of the stem and the weight of the root systems of resultant seedlings, and also reduced the numbers of non-rhizospheric organisms with ammonification and nitrification activities. The addition of Bacillus amyloliquefaciens or Microbacterium oleovorans, at a concentration of 109 colony forming units ml?1, to the seeds significantly reduced Fusarium verticillioides count at the root inner tissues of resultant seedlings. When testing the impact of bacterial treatments on soil populations, no alterations with respect to control numbers of organisms with nitrification, ammonification or cellulolytic potentials were observed. Culturable bacterial richness and diversity calculated at the rhizoplane and root inner tissues of maize seedlings neither changed in bacterized treatments when compared to control treatment. Our study showed that the Fusarium verticillioides in vitro proven antagonists, Bacillus amyloliquefaciens and Microbacterium oleovorans (at 109 colony forming units ml?1), were also effective at greenhouse conditions without causing major changes in culturable rhizospheric and endophytic microbial richness and diversity.  相似文献

Greenhouse-grown cut flower roses are often irrigated with moderately saline irrigation water. The salt/ballast ions are either present initially in poor quality raw water or reclaimed municipal water, or accumulated in greenhouse irrigation water that is captured and reused. Such ions can inhibit root absorption of essential nutrients. The objective of this work was to quantify the influence of NaCl concentration on the uptake of nitrate and potassium by roses and develop a predictive model of uptake inhibition based on NaCl, NO3 ?, and K+ concentration. One year-old rose plants (Rosa spp. ‘Kardinal’ on ‘Natal Briar’ rootstock) were moved into growth chambers where nitrogen and potassium depletion were monitored during 6 days. Eight different initial NaCl treatments varying from zero to 65 mol m?3 were used and within these there were two initial NO3 ? and K+ concentrations: high concentration (HC, 7.0 mol m?3 and 2.6 mol m?3 NO3 ? and K+ respectively) or low concentration (LC, 3.5 mol m?3 and 1.3 mol m?3 NO3 ? and K+ respectively). Plant NO3 ? uptake was negatively affected by NaCl concentration. NO3 ? maximum influx (Imax) declined from 5.1 µmol to 2.5 µmol per gram of plant dry weight per hour as NaCl concentration increased from zero to 65 mol m?3. A modified Michaelis–Menten (M–M) equation taking into account inhibition by NaCl provided the best fit for NO3 ? uptake in response to varying NaCl concentration. K+ uptake was unaffected by NaCl concentration. A M–M equation that did not include inhibition was suitable for describing K+ uptake at varying NaCl concentration. The resulting empirical models could assist with decision making, such as: adjustment of NO3 ? fertilization based on NaCl concentration, necessity of water desalinization, or determination of the desired leaching fraction.  相似文献

Nitraria tangutorum Bobr. is a typical halophyte with superior tolerance to salinity. However, little is known about its physiological adaptation mechanisms to the salt environment. In the present study, N. tangutorum seedlings were treated with different concentrations of NaCl (100, 200, 300 and 400 mmol L?1) combined with five levels of Ca2+ (0, 5, 10, 15 and 20 mmol L?1) to investigate the effects of salt stress and exogenous Ca2+ on Na+ compartmentalization and ion pump activities of tonoplast and plasma membrane (PM) in leaves. Na+ and Ca2+ treatments increased the fresh weight and dry weight of N. tangutorum seedlings. The absorption of Na+ in roots, stems and leaves was substantially increased with the increases of NaCl concentration, and Na+ was mainly accumulated in leaves. Exogenous Ca2+ reduced Na+ accumulation in roots but promoted Na+ accumulation in leaves. The absorption and transportation of Ca2+ in N. tangutorum seedlings were inhibited under NaCl treatments. Exogenous Ca2+ promoted Ca2+ accumulation in the plant. Na+ contents in apoplast and symplast of leaves were also significantly increased, and symplast was the main part of Na+ intracellular compartmentalization. The tonoplast H+-ATPase and H+-PPase activities were significantly promoted under salt stress (NaCl concentrations ≤300 mmol L?1). PM H+-ATPase activities gradually increased under salt stress (NaCl concentrations ≤200 mmol L?1) followed by decreases with NaCl concentration increasing. The tonoplast H+-ATPase, H+-PPase and PM H+-ATPase activities increased first with the increasing exogenous Ca2+ concentration, reached the maximums at 15 mmol L?1 Ca2+, and then decreased. The tonoplast and PM Ca2+-ATPase activities showed increasing trends with the increases of NaCl and Ca2+ concentration. These results suggested that certain concentrations of exogenous Ca2+ effectively enhanced ion pump activities of tonoplast and PM as well as promoted the intracellular Na+ compartmentalization to improve the salt tolerance of N. tangutorum.  相似文献

Tea (Camellia sinensis (L.) O. Kuntze) hyper-accumulates fluoride (F), mainly in the leaves. To understand how tea copes with the stress caused by F, we tracked photosynthesis, antioxidant defense, and cell ultrastructure under different F concentrations (0–50 mg L?1). High F (≥5 mg L?1) caused decreases in photosynthetic and chlorophyll fluorescence parameters. Activated oxygen metabolism was altered by F, as manifested in increasing lipid peroxidation, electrolyte leakage (EL), and accumulation of H2O2. The activities of ascorbate peroxidase (APX, EC 1.11.1.1) and catalase (CAT, EC 1.11.1.6) increased at 0–5 mg L?1 F, but sharply decreased less than 10–50 mg L?1 F. The activity of manganese superoxide dismutase (Mn-SOD, EC 1.15.1.1) decreased with increasing F concentration. Expression of genes encoding antioxidant enzymes were in accordance with their measured activities. The results suggest that the antioxidant enzymes in the tea plant can eliminate reactive oxygen species (ROS) at <5 mg L?1 F, but not at 20–50 mg L?1 F. High F increased the number of epidermal hairs on tea leaves and decreased the stomatal aperture, reducing water loss. The leaf cellular structure appeared normal under 1–50 mg L?1 F, although starch grains in chloroplast increased with increasing F. Proline and betaine play important roles in osmotic regulation in tea plant tolerating F stress. ROS scavenging and greater number of epidermal hairs are likely parts of the tea plant F-tolerance mechanism.  相似文献

The effect of exogenously applied hydrogen peroxide on salt stress tolerance was investigated in Panax ginseng. Pretreatment of ginseng seedlings with 100 μM H2O2 increased the physiological salt tolerance of the ginseng plant and was used as the optimum concentration to induce salt tolerance capacity. Treatment with exogenous H2O2 for 2 days significantly enhanced salt stress tolerance in ginseng seedlings by increasing the activities of ascorbate peroxidase, catalase and guaiacol peroxidase and by decreasing the concentrations of malondialdehyde (MDA) and endogenous H2O2 as well as the production rate of superoxide radical (O2 ?). There was a positive physiological effect on the growth and development of salt-stressed seedlings by exogenous H2O2 as measured by ginseng dry weight and both chlorophyll and carotenoid contents. Exogenous H2O2 induced changes in MDA, O2 ?, antioxidant enzymes and antioxidant compounds, which are responsible for increases in salt stress tolerance. Salt treatment caused drastic declines in ginseng growth and antioxidants levels; whereas, acclimation treatment with H2O2 allowed the ginseng seedlings to recover from salt stress by up-regulation of defense-related proteins such as antioxidant enzymes and antioxidant compounds.  相似文献

Metformin (MET), an anti-diabetic oral drug with antioxidant properties, has been proved to provide cardioprotective effects in patients with diabetic disease. However, the mechanism is unclear. This study aimd to investigate the effects of MET on the expressions of receptor for advanced glycation end products (RAGE) and high mobility group box 1 protein (HMGB1) in hyperglycemia-treated neonatal rat ventricular myocytes. Cardiocytes were prepared and cultured with high glucose and different concentrations of MET. The expressions of RAGE and HMGB1 were evaluated by Western blot analysis. The superoxide dismutase (SOD), malondialdehyde (MDA), tumor necrosis factor-α (TNF-α), lactate dehydrogenase (LDH) and creatine kinase (CK) were measured. After 12 h-incubation, MET significantly inhibited the increase of MDA, TNF-α, LDH and CK levels induced by high glucose, especially at the 5 × 10?5 to 10?4 mol/L concentrations while inhibiting the decrease of SOD level. Meanwhile, RAGE and HMGB1 expression were significantly increased induced by hyperglycaemia for 24 h (P < 0.05). MET inhibited the expressions of RAGE and HMGB1 in a dose-dependent manner, especially at the 5 × 10?5 to 10?4 mol/L concentrations (P < 0.05). In conclusion, our study suggested that MET could reduce hyperglycemia-induced cardiocytes injury by inhibiting the expressions of RAGE and HMGB1.  相似文献
