北太平洋柔鱼微卫星标记的筛选及遗传多样性   总被引:2,自引:0,他引:2

采用(AC)12、(AG)12两种生物素探针,通过磁珠富集法构建了柔鱼部分基因组微卫星富集文库。68个阳性克隆中有60个含有微卫星序列,重复次数在10次以上的占86.84%,最高重复次数为33次。其中,完美型微卫星占60.53%,非完美型微卫星占36.84%,混合型微卫星占2.63%。除探针使用的AC/TG、AG/TC重复外,还得到ACAG、AGAC重复序列。利用筛选出的8个微卫星位点对北太平洋柔鱼6个群体的遗传多样性及遗传结构进行分析。结果表明,8个微卫星位点均为高度多态性位点(PIC=0.787—0.987),位点Bo103与位点Bo105极显著偏离Hardy-Weinberg平衡(P0.01)。6个地理位置的柔鱼群体显示出较高的遗传多样性水平(Ho=0.672—0.761,He=0.808—0.851)。两两群体间的Fst值以及AMOVA分析结果均表明,群体间遗传分化不显著(Fst=0.00559,P0.05),遗传差异主要来自于个体间。基于Nei's遗传距离的UPGMA聚类树显示,北太平洋东北部2个柔鱼群体(NE1、NE3)聚为一类,西北部3个群体(NW1、NW2、NW3)与东北部1个群体(NE2)另聚为一类,且群体NW1与群体NE2亲缘关系最近,遗传距离与地理距离线性相关分析没有呈现出正相关性(R=0.175,P0.05)。遗传结构分析结果推断北太平洋柔鱼存在1个理论群。柔鱼个体具有较强的游泳能力,在海流的作用下,群体之间存在较强的基因交流。建议今后在柔鱼资源开发利用过程中将北太平洋柔鱼看作1个管理单元。  相似文献

Escherichia coli single-strand (ss) DNA binding protein (SSB) is an essential protein that binds ssDNA intermediates formed during genome maintenance. SSB homotetramers bind ssDNA in two major modes, differing in occluded site size and cooperativity. The (SSB)35 mode in which ssDNA wraps, on average, around two subunits is favored at low [NaCl] and high SSB/DNA ratios and displays high unlimited, nearest-neighbor cooperativity forming long protein clusters. The (SSB)65 mode, in which ssDNA wraps completely around four subunits of the tetramer, is favored at higher [NaCl] (>200 mM) and displays limited low cooperativity. Crystal structures of E. coli SSB and Plasmodium falciparum SSB show ssDNA bound to the SSB subunits (OB folds) with opposite polarities of the sugar phosphate backbones. To investigate whether SSB subunits show a polarity preference for binding ssDNA, we examined EcSSB and PfSSB binding to a series of (dT)70 constructs in which the backbone polarity was switched in the middle of the DNA by incorporating a reverse-polarity (RP) phosphodiester linkage, either 3′-3′ or 5′-5′. We find only minor effects on the DNA binding properties for these RP constructs, although (dT)70 with a 3′-3′ polarity switch shows decreased affinity for EcSSB in the (SSB)65 mode and lower cooperativity in the (SSB)35 mode. However, (dT)70 in which every phosphodiester linkage is reversed does not form a completely wrapped (SSB)65 mode but, rather, binds EcSSB in the (SSB)35 mode with little cooperativity. In contrast, PfSSB, which binds ssDNA only in an (SSB)65 mode and with opposite backbone polarity and different topology, shows little effect of backbone polarity on its DNA binding properties. We present structural models suggesting that strict backbone polarity can be maintained for ssDNA binding to the individual OB folds if there is a change in ssDNA wrapping topology of the RP ssDNA.  相似文献

Atlantic herring (Clupea harengus), a vital ecosystem component and target of the largest Northwest Atlantic pelagic fishery, undergo seasonal spawning migrations that result in elusive sympatric population structure. Herring spawn mostly in fall or spring, and genomic differentiation was recently detected between these groups. Here we used a subset of this differentiation, 66 single nucleotide polymorphisms (SNPs) to analyze the temporal dynamics of this local adaptation and the applicability of SNP subsets in stock assessment. We showed remarkable temporal stability of genomic differentiation corresponding to spawning season, between samples taken a decade apart (2005 N = 90 vs. 2014 N = 71) in the Gulf of St. Lawrence, and new evidence of limited interbreeding between spawning components. We also examined an understudied and overexploited herring population in Bras d'Or lake (N = 97); using highly reduced SNP panels (NSNPs > 6), we verified little‐known sympatric spawning populations within this unique inland sea. These results describe consistent local adaptation, arising from asynchronous reproduction in a migratory and dynamic marine species. Our research demonstrates the efficiency and precision of SNP‐based assessments of sympatric subpopulations; and indeed, this temporally stable local adaptation underlines the importance of such fine‐scale management practices.  相似文献

Analyses of genetic variability and allelic composition in a species exhibiting reproductive fidelity to natal sites may provide important ecological indication of temporal population dynamics, facilitating understanding responses to past disturbances and future climate change. The walleye is an ecologically and economically valuable species, whose largest fishery centers in Lake Erie of the Laurentian Great Lakes; it exhibits reproductive site fidelity, despite otherwise wide-ranging dispersal. We tested whether genetic composition and diversity have remained temporally stable in Lake Erie’s Maumee River, which is the largest and most highly fished spawning run. This population has experienced over a century of exploitation, habitat alterations, and pollution, which may have affected genetic structure and might influence future sustainability. Fourteen nuclear DNA microsatellite loci were analyzed from 744 spawning run walleye to test genetic patterns across: (1) years (N = 12, spanning 1995–2013), (2) birth year cohorts, (3) the sexes, (4) those reproducing earlier (ages 2–6) versus later (7 or older) in life, and (5) the adults versus larvae. Results indicated stability in genetic diversity levels (mean H O = 0.76 ± 0.03) and allelic composition across years (F ST = 0.000–0.006, NS), cohorts (F ST = 0.000–0.013, NS), sexes (F ST = 0.000, NS), earlier versus later reproduction (F ST = 0.000, NS), and between the larvae and adults (F ST = 0.000–0.004, NS). Number of breeders and effective population size were substantial and consistent. This reproductive population thus has maintained genetic stability and high diversity, despite intensive anthropogenic pressures.  相似文献

The influence of quaternary structure on the low frequency molecular vibrations of the haem within deoxyhaemoglobin (deoxy Hb) and Oxyhaemoglobin (oxy Hb) was studied by resonance Raman scattering. The FeO2 stretching frequency was essentially identical between the high affinity (R) state (Hb A) and low affinity (T) state (Hb Kansas and Hb M Milwaukee with inositol hexaphosphate). However in deoxy Hb, only one of the polarized lines showed an appreciable frequency shift upon switch of quaternary structure, i.e. 215 to 218 cm?1 for the T state (Hb A, des-His(146β) Hb, and des-Arg(141α) Hb (pH 6.5)) and 220 to 221 cm?1 for the R state (des-Arg(141α) Hb (pH 9.0), des-His(146β)-Arg(141α) Hb and NES des-Arg(141α) Hb). Based on the observed 54Fe isotopic frequency shift of the corresponding Raman lines of deoxy Hb A (214 → 217 cm?1), of deoxy NES des-Arg Hb (220 → 223 cm?1), of the protoporphyrinato-Fe(II)-(2-methylimidazole) complex in the ferrous high spin state (207 → 211 cm?1) and of deoxymyoglobin (220 → 222 cm?1) (Kitagawa et al., 1979), and on substitution of perdeuterated for protonated 2-methylimidazole in the deoxygenated picket fence complex (TpivPP)Fe2+ (2-MeIm) (209 → 206 cm?1), and on the results of normal co-ordinates calculation carried out previously, we proposed that the 216 cm?1 line of deoxy Hb is associated primarily with the FeNε(HisF8) stretching mode and accordingly that the FeNε(HisF8) bond is stretched in the T state due to a strain exerted by globin.  相似文献

The sugar chains of microsomal and lysosomal β-glucuronidases of rat liver were studied by endo-β-N-acetylglucosaminidase H digestion and by hydrazinolysis. Only a part of the oligosaccharides released from microsomal β-glucuronidase was an acidic component. The acidic component was not hydrolyzed by sialidase and by calf intestinal and Escherichia coli alkaline phosphatases, but was converted to a neutral component by phosphatase digestion after mild acid treatment indicating the presence of a phosphodiester group. The neutral oligosaccharide portion of microsomal enzyme was a mixture of five high mannose-type sugar chains: (Manα1 → 2)0~4 [Manα1 → 6(Manα1 → 3)Manα1 → 6(Manα1 → 3)Manβ1 → 4GlcNAcβ1 → 4GlcNAc]. In contrast, lysosomal enzyme contains only Manα1 → 6 (Manα1 → 3) Manα1 → 6(Manα1 → 3) Manβ1 → 4GlcNAcβ1 → 4GlcNAc. The result indicates that removal of α1 → 2-linked mannosyl residues from (Manα1 → 2)4[Manα1 → 6(Manα1 → 3)Manα1 → 6(Manα1 → 3)Manβ1 → 4GlcNAcβ1 → 4GlcNAc → Asn] starts already in the endoplasmic reticulum of rat liver.  相似文献

Stochastic variability of biological processes and uncertainty of stock properties compel fisheries managers to look for tools to improve control over the stock. Inspired by animals exploiting hidden prey, we have taken a biomimetic approach combining catch and effort in a concept of Bayesian regulation (BR). The BR provides a real-time Bayesian stock estimate, and can operate without separate stock assessment. We compared the performance of BR with catch-only regulation (CR), alternatively operating with N-target (the stock size giving maximum sustainable yield, MSY) and F-target (the fishing mortality giving MSY) on a stock model of Baltic Sea herring. N-targeted BR gave 3% higher yields than F-targeted BR and CR, and 7% higher yields than N-targeted CR. The BRs reduced coefficient of variance (CV) in fishing mortality compared to CR by 99.6% (from 25.2 to 0.1) when operated with F-target, and by about 80% (from 158.4 to 68.4/70.1 depending on how the prior is set) in stock size when operated with N-target. Even though F-targeted fishery reduced CV in pre-harvest stock size by 19–22%, it increased the dominant period length of population fluctuations from 20 to 60–80 years. In contrast, N-targeted BR made the periodic variation more similar to white noise. We discuss the conditions when BRs can be suitable tools to achieve sustainable yields while minimizing undesirable fluctuations in stock size or fishing effort.  相似文献

It is well known that temporal fluctuations in small populations deeply influence evolutionary potential. Less well known is whether fluctuations can influence the evolutionary potentials of species with large census sizes. Here, we estimated genetic population parameters from as survey of polymorphic microsatellite DNA loci in archived otoliths from Adriatic European anchovy (Engraulis encrasicolus), a fish with large census sizes that supports numerous local fisheries. Stocks have fluctuated greatly over the past few decades, and the Adriatic fishery collapsed in 1987. Our results show a significant reduction of mean genetic parameters as a consequence of the population collapse. In addition, estimates of effective population size (Ne) are much smaller than those expected in a fishes with large population census sizes (Nc). Estimates of Ne indicate low effective population sizes, even before the population collapse. The ratio Ne/Ne ranged between 10−6 and 10−8, indicating a large discrepancy between the anchovy gene pool and population census size. Therefore, anchovy populations may be more vulnerable to fishery effort and environmental change than previously thought.  相似文献

Cathepsin D from porcine spleen contained mannose (3.3%), glucosamine (1.4%), and mannose 6-phosphate (0.08%). Essentially all of the oligosaccharides of cathepsin D could be released by endo-β-N-acetylglucosaminidase H, pointing to oligomajmoside types of structures. Three neutral oligosaccharide fractions, containing 5, 6, and 7 mannose residues, respectively, were isolated by gel permeation chromatography on Bio-Gel P-2. Studies using exoglycosidase digestions and 500-MHz 1H NMR spectroscopy revealed that their structures are [Manα1 → 2]0 or 1Manα1 → 6[Manα1 → 3]Manα1 → 6[(Manα1 → 2)0 or 1Manα1 → 3]Manβ1 → 4GlcNAcβ1 → 4 GlcNAc. These structures are identical to what have recently been proposed by Takahashi et al. for the major oligosaccharide units of cathepsin D from the same source (T. Takahashi P.G. Schimidt, and J. Tang (1983)J. Biol. Chem.258, 2819–2930), except for the occurrence of two isomeric oligosaccharides containing six mannoses. Only a part (3.4%) of the oligosaccharides were acidic, containing phosphates in monoester linkage. The phosphorylated oligosaccharides also consisted of oligomannoside-type chains which were analogous to, but more heterogeneous in size than the neutral oligosaccharides. Cathepsin D was bound to a mannose- and N-acetylglucosamine-specific lectin (mannan-binding protein) isolated from rabbit liver with the Ki value of 5.4 × 10?6m.  相似文献

Human antithrombin III contains four asparagine-linked sugar chains in one molecule. The sugar chains were quantitatively released as radioactive oligosaccharides from the polypeptide portion by hydrazinolysis followed by N-acetylation and NaB3H4 reduction. All of the oligosaccharides, thus obtained, contain N-acetylneuraminic acid. A same neutral nonaitol was released from all acidic oligosaccharides by sialidase treatment. N-Formyl, N-chloroacetyl, N-glycyl, N-isobutyryl, and N-pentanoyl derivatives of chitosan have been prepared. N-Acetylchitosan was the derivative most susceptible to chitinase from Streptomyces griseus and lysozyme from chicken egg-white, but the susceptibility was not restrictive. The relative rates of hydrolysis by chitinase with respect to R in the RCONH group were CH3 > CH3CH2 > H > CH3CH2CH2 > (CH3)2CH > NH2CH2 > ClCH2. Neither enzyme hydrolysed chitosan or its N-methylene, N-benzylidene, N-benzoyl, N-nicotinyl, and N-fatty acyl (C5C18) derivatives, and lysozyme did not hydrolyse N-butyrylchitosan. N-Acetylhexanoyl-chitosans, which had d.s. ratios of ~0.7: ~0.3 and ~0.3; ~0.7, were hydrolysed at ~0.75 and ~0.04 of the rate of N-acetylchitosan (powder) by chitinase. O-Acylation of N-acylchitosans caused a decrease in the rates of hydrolysis by chitinase. N-Acetylchitosan gels were hydrolysed at 8–13 times the rate for crab-shell chitin. These results indicate that not only N- and O-substituents but also the physical form of the substrates influence the rates of hydrolysis by these enzymes.  相似文献

The combining site of the Bauhinia purpurea alba lectin was studied by quantitative precipitin and precipitin inhibition assays. Of 45 blood group substances, glycoproteins, and polysaccharides tested, 35 precipitated over 75% of the lectin. Precursor blood group substances with I activity (Cyst OG 10% from 20% and Cyst OG 20% from 10%), desialized fetuin, and desialized ovine salivary glycoprotein, in which more than 75% of the carbohydrate side chains have dGalN Ac linked through α1 → to the OH group of Ser or Thr of a protein core, completely precipitated the lectin. The poorly reactive blood group substances after mild acid hydrolysis or Smith degradation, as well as sialic acid-containing glycoproteins after removal of sialic acid, had substantially increased activity so that more than 80% of the lectin was precipitated. Precipitability with various blood group substances and glycoproteins is ascribable to the terminal nonreducing dGalNAc, dGalβ1 → 3dGalNAc, dGalβ1 → 3 or 4dGlcNAc, and dGalβ1 → 3 or 4dGlcNAcβ1 → 3dGal determinants on the carbohydrate moiety. Of the monosaccharides tested for inhibition of precipitation, dGalNAc and its p-nitrophenyl and methyl α-glycosides were best. These compounds were four to five times better than the corresponding dGal compounds but methyl βDGalNAcp was only about 40% more active than methyl βdGalp. The α-anomers of p-nitrophenyl DGalNAcp and dGalp, were twice as active as the corresponding β-anomers. Methyl αDGalNAcp was four times as active as the β-anomer but the inhibitory power of the methyl α- and β-anomers of dGal were about equal. Among the oligosaccharides tested, dGalβ1 → 3dGalNAc and its tosyl derivatives were most active, the tosyl glycosides being about twice as active as dGalβ1 → 3dGalNAc, which was somewhat more active than dGalNAcα1 → 6dGal and dGalNAc, and 2.5 and 5 times as active as dGalNAcα1 → 3dGalβ1 → 3dGlcNAc and dGalNAcαl → 3dGa1, respectively (blood group A specific). These findings suggest that a subterminal dGalNAc β-linked and substituted on carbon 3 plays an important role in binding. Consistent with this inference are the findings that dGalβ1 → 3dGlcNAc and dGalβ1 → 6dGal were poorer inhibitors although dGalβ1 → 3dGlcNAc was two to three times as active as glycosides of dGal. Oligosaccharides with terminal nonreducing dGal and subterminal α-linked dGal were as active or less active than dGal. dGalβ1 → 3dGlcNAcβ1 → 3dGalβ1 → 4dGlc (lacto-N-tetraose) and dGalβ1 → 3dGlcNAcβ1 → 3dGal-β1-O-(CH2)8COOCH3 were equally active and 1.5 times as potent as dGalβ1 → 3dGlcNAc whereas dGalβ1 → 3dGlcNAcβ1 → 6dGal was only 40% as potent as dGalβ1 → 3dGlcNAc suggesting that a third sugar may be part of the determinant. Substitution of dGalβ1 → 3dGlcNAcβ1 → 3dGalβ1 → 4dGlc on the subterminal dGlcNAc by lFucα1 → 4 in lacto-N-fucopentaose II reduced activity fourfold; if the nonreducing dGal is substituted by lFucα1 → 3 as in lacto-N-fucopentaose I its activity is almost completely abolished. This suggests that a terminal nonreducing dGal as well as subterminal dGlcNAc are contributing to binding. The β → 3 linkage of the terminal dGal to the subterminal amino sugar is significant since dGalβ1 → 4dGlcNAc is a poorer inhibitor. Although the available data suggest that the combining site of the lectin Bauhinia purpurea alba may be most complementary to the structure dGalβ1 → 3dGalNAcβ1 → 3dGal, several other possibilities remain to be tested when suitable oligosaccharides become available.  相似文献

For the past several decades the Lake Mohave population of the federally endangered razorback sucker, Xyrauchen texanus, has had no natural recruitment because of intensive predation on larvae by non-native fishes. In response to impending extirpation, a repatriation program was implemented where larval razorbacks are collected from the wild following natural spawning, reared in protective custody, and then repatriated at a much larger size. In this study, we estimated annual (N bf) and generational female effective size (N ef) of the spawning stock by characterizing temporal genetic changes in mtDNA among larval cohorts, and then compared these estimates to the estimated number of reproductively capable (adult) females in Lake Mohave (N f). Razorback suckers have life history and mortality schedules that could yield values of N ef/N f as low as 10−5 due to match–mismatch recruitment (or the “Hedgecock effect”) that increases variance in reproductive success. Average N bf was estimated to be 160, N ef was 706, and the ratio N ef/N f was 0.29 and 0.38 for arithmetic and harmonic mean N f, respectively. Our findings indicate that (i) larval sampling in Lake Mohave sufficiently encompasses temporal and spatial variation within annual larval cohorts as to be representative of the spawning stock, (ii) roughly 3–16% (about 8% on average) of adult females contribute genetically to larval cohorts each year, and (iii) repatriated fishes appear to be contributing genetically to larval cohorts. Simultaneous genetic and demographic monitoring offers insights that neither approach can provide alone into effects of population decline and management practices in this species.  相似文献

Three series of complexes were studied: Co(acetylacetonate)n(dibenzoylmethanato)3-n, Co(acetylacetonate)n(benzoylacetonate)3-n and Co(dibenzoylmethanato)n(benzoylacetonate)3-n. The product mixtures were separated by thin-layer chromatography. Complete separation of all compounds and stereoisomers was obtained. No attempt was made to separate optical isomers which exist for each single stereoisomer. Characterization was by elemental analyses, NMR spectroscopy and the separation characteristics. Electronic absorption spectra were measured in acetonitrile in the 200–800 nm region. Interpretation of the spectral data is provided in terms of ∏ → ∏*, d → ∏*, d → d* transitions. Coilia dussumieri (Valenciennes, 1848) commonly called as golden anchovy, constitutes a considerable fishery in the northern part of both the west and east coasts of India. Despite its clear-cut geographic isolation, the species is treated as a unit stock for fishery management purposes. We evaluated 32 microsatellite primer pairs from three closely related species (resource species) belonging to the family Engraulidae through cross-species amplification in C. dussumieri. Successful cross-priming was obtained with 10 loci, which were sequenced for confirmation of repeats. Loci were tested for delineating the genetic stock structure of four populations of C. dussumieri from both the coasts of India. The number of alleles per locus ranged from 8 to 18, with a mean of 12.3. Results of pairwise F ST indicated genetic stock structuring between the east and west coast populations of India and also validated the utilization of identified microsatellite markers in population genetic structure analysis.  相似文献

High resolution nuclear magnetic resonance spectra were recorded in a chloroform solution of six Lewis-active or Lewis-like glycosphingolipids in permethylated and permethylated-reduced (LiAlH4) form. The samples were selected to cover the presently known structural variants of α-fucose linked to galactose and N-acetylglucosamine. Fucα1 → 2Gal, Fucα1 → 3GlcNAc, and Fucα1 → 4GlcNAc gave characteristic and well-separated anomeric resonances. Furthermore, upon reduction there was a strong deshielding effect on Fucα1 → 3GlcNAc and Galβ1 → 3GlcNAc (linkage vicinal to reduced amide), which makes it possible to differentiate type 1 (Galβ1 → 3GlcNAc) and type 2 (Galβ1 → 4GlcNAc) saccharide chains. This improved method of nuclear magnetic resonance spectroscopy is discussed in relation to sequence analysis by mass spectrometry, two microscale structural methods using the same type of derivatives and needing no degradations before analysis.  相似文献

S-Alkyl-N-alkylisothiourea compounds containing various cyclic amines were synthesized in the search for novel nonimidazole histamine H3 receptor (H3R) antagonists. Among them, four N-alkyl S-[3-(piperidin-1-yl)propyl]isothioureas 18, 19, 22, and 23 were found to exhibit potent and selective H3R antagonistic activities against in vitro human H3R, but were inactive against in vitro human H4R. Furthermore, three alkyl homologs 1820 showed inactivity for histamine release in in vivo rat brain microdialysis, suggesting differences in antagonist affinities between species. In addition, in silico docking studies of N-[4-(4-chlorophenyl)butyl]-S-[3-piperidin-1-yl)propyl]isothiourea 19 and a shorter homolog 17 with human/rat H3Rs revealed that structural differences between the antagonist-docking cavities of rat and human H3Rs were likely caused by the Ala122/Val122 mutation.  相似文献

Annual egg production in eastern Baltic cod Gadus morhua callarias in 1966–2000 has been assessed. A significant relationship between egg production and recruitment, progeny survival index, and 11psu-isohaline depth has been revealed. Comparison of egg production and spawning stock biomass indices has shown the time difference in their maxima and the absence of significant correlation between them. This difference is assumed to be due to an increase in the share of mature individuals that did not participate in spawning in the late 1970s–early 1980s. The determination of potential egg production, taking into account the share of individuals that spawn during the current year, yielded a significant positive relationship between its value and spawning stock biomass.  相似文献

The water quality of Feng-qing Lake, which is a landscape lake supplemented with reclaimed water, was surveyed to investigate the relationship between phytoplankton and environmental variables. A total of 29 water samples were collected to analyze temporal variations of phytoplankton and environmental factors from July 2013 to June 2014. Six phyla and 39 genera of phytoplankton were identified when the lake was supplied with reclaimed water. Among these, Cyanophyta and Chlorophyta account for 38.46% and 30.77% of phytoplankton, respectively. The dominant species in the lake are Pseudanabaena limnetica and Chlorella vulgaris, which are present the entire year. Other leading species include Cosmarium sp. and Raphidiopsis curvata. Principal component analysis (PCA) was conducted to analyze the relationship among environmental factors. Canonical correspondence analysis (CCA) was performed to investigate the relationship between environment factors and dominant species. The PCA result showed that temperature (T), total phosphorus (TP), total nitrogen (TN), transparency, and dissolved oxygen are the main factors that affect the eutrophication level of the lake. The CCA result revealed that TN, PO43−–P, chemical oxygen demand (COD), T, and chlorophyll a exhibit a close relation with dominant species. In particular, TN, salinity, and COD influence the growth of P. limnetica; T and COD influence the growth of R. curvata; and T, PO43−–P, NH3–N, and pH influence the growth of C. vulgaris and Cosmarium sp.  相似文献
