芽胞杆菌抗菌活性和纤维素酶活性的测定   总被引:1,自引:1,他引:0  
目的筛选鸡用益生菌。方法通过牛津杯法、混菌法和DNS分光光度法对从鸡肠道分离的10株芽胞杆菌进行抑菌效果及纤维素酶活性测定。结果编号为Y1、Y5、Y7的3株芽胞杆菌对测试菌具有较好抑菌效果,编号为Y2、Y4、Y5的3株芽胞杆菌具有较高的产酶活性。结论从分离的10株芽胞杆菌中筛选出1株(编号为Y5)兼具较好抑菌效果和较高产酶活性的地衣芽胞杆菌,具有作为饲用益生菌开发的潜能。  相似文献

筛选黄曲霉毒素生物防控菌,为黄曲霉毒素的生物防控提供支持。以花生原产地土壤为材料,采用牛津杯法筛选所需菌株。对筛选出的拮抗菌株进行抑制产毒曲霉菌株的生长、产孢、降解黄曲霉毒素实验。筛选出2株黄曲霉毒素生防细菌,编号21-1-2、17-3,经鉴定,拮抗菌21-1-2为枯草芽胞杆菌,拮抗菌17-3为地衣芽胞杆菌。分别对拮抗菌对曲霉孢子萌发的抑制、抑制黄曲霉的生长和菌丝延长以及减少黄曲霉毒素的产生、对黄曲霉毒素的分解作用等几个方面进行研究,结果表明,拮抗菌可以明显抑制产毒曲霉孢子的萌发、生长、菌丝的延长,减少黄曲霉毒素的产生以及分解黄曲霉毒素。  相似文献

鸡肠源芽胞杆菌的分离、鉴定和抗菌活性   总被引:1,自引:1,他引:0  
从人工饲养的健康三黄肉鸡的盲肠和大肠中分离出10株芽胞杆菌,对其进行菌落形态观察、革兰染色、芽胞染色和生理生化特性鉴定,确定为枯草芽胞杆菌、地衣芽胞杆菌、蜡状芽胞杆菌、巨大芽胞杆菌、球形芽胞杆菌、短小芽胞杆菌、凝结芽胞杆菌。同时测定了它们对动物病原性大肠埃希菌和链球菌的抑菌活性。其中有9株芽胞杆菌对大肠埃希菌有抑制作用,8株芽胞杆菌对链球菌有抑制作用。结果表明芽胞杆菌分离株Y2、Y3、Y4、Y5、Y6、Y7、Y8具有作为益生菌开发的潜力。  相似文献

为筛选耐热淀粉酶产生菌并揭示其在温泉中的分布情况,从腾冲县腊幸村一小型热泉不同部位采集样品进行微生物分离及鉴定。样品在65℃培养,挑选形态有差异的菌落点种淀粉-台盼蓝平板,共获得34株具有淀粉降解能力的细菌。分子鉴定结果显示,从温泉底部沙土中筛选得到细菌具有较高的生物多样性,9株具有淀粉降解能力的细菌分别属于土芽胞杆菌(Geobacillus)、厌氧芽胞杆菌(Anoxybacillus)和芽胞杆菌(Bacillus)3个属、7个种,菌株间16S rDNA序列均有一定的差异。对编号为201(Geobacillus thermoglucosidasius)和208(Anoxybacillus flavithermus)的菌株胞外淀粉酶进行初步分析,其胞外淀粉酶最适温度分别为60和70℃,均高于淀粉糊化温度,具有一定的应用潜力。  相似文献

从几种泡菜中分离出86株菌,对其在适温和低温下产酸速率及硝酸盐降解能力进行测定,筛选出5株产酸速率快、硝酸盐降解能力强的菌株。经形态学鉴定及生理生化反应试验,初步鉴定为:植物乳杆菌2株,短乳杆菌1株,戊糖乳杆菌1株,肠膜明串珠菌葡聚糖亚种1株,并对5株菌的发酵性能进行了测定。  相似文献

为了加快菠萝渣发酵,本试验通过利用多种选择性培养基,从自然发酵的菠萝渣中分离到多种淀粉分解菌,经过初筛和复筛,获得了降解菠萝渣的淀粉降解功能菌株s2b7-1和s2b7-4,筛选出其最适培养基为改良LB培养基及其优化组合。通过形态、生理生化和分子综合鉴定得出s2b7-4菌株为枯草芽胞杆菌(Bacillus subtilis)。  相似文献

从学校食堂排放的废水中筛选出1株油脂降解菌株,经形态特征、生理生化特征、16S rDNA同源性序列分析,鉴定为克雷伯氏菌属,暂命名为Klebsiella sp.X-1。并利用正交实验进一步考察了pH、培养温度、摇床转速对油脂降解率的影响。实验结果表明,在高含油废水培养基中,pH 7.0、转速为140 r/min、培养温度为30 ℃时,72 h内该菌的油脂降解率最高达68.2%。  相似文献

目的对随机采样和田字格定点采样两种取样方式筛菌效率进行比较分析。方法利用随机采样和定点采样两种不同方式,采集了长白山地区的土样,取来自不同植物根际的土样,制成菌悬液后煮沸,涂布无氮平板以分离固氮芽胞杆菌。对分离的菌株进行菌落16S rDNA PCR,从中筛选鉴别巨大芽胞杆菌。结果定点采集土样分离出无晶体芽胞菌和有晶体芽胞菌分别是261株和76株,随机采集土样分离出无晶体芽胞菌和有晶体芽胞菌分别是279株和94株。经16S rDNA法鉴定分离巨大芽胞杆菌共19株。结论随机采样的取土方式与定点采样的取土方式差异度小,两种采集土样的方式在筛菌效率上并未表现出明显差异。  相似文献

从含油土壤中,筛选出能以0#柴油为惟一碳源的2株柴油降解菌,经鉴定分别为假单胞菌(Pseudomonoussp.)和芽孢杆菌(Bacillussp.)。通过其降解能力的测试,假单胞杆菌属细菌A8的降解能力较强;在含油浓度ρ=10g/L的摇瓶试验中,培养7d后其除油率分别为18%和31%。在含油浓度1%的土壤中,2株菌均能正常生长和繁殖,接种30d除油率分别为35%和48.3%。  相似文献

The enzymic activity (amalyse, protease, lipase, pectolytic and cellulase) has been studied in 5 strains of aerobic spore-forming bacteria (Bacillus subtilis, B. licheniformis, B. coagulans, B. pumilis, B. badius) being of interest for creation of medical and prophylactic biopreparations. The above-mentioned enzymes were found in some studied strains. This may provide participation of bacilli in the degradation processes of a number of substrates in the digestive tract of a human being and animals and is an advantage of preparations from the genus Bacillus bacteria as compared with the available biopreparations of other microbial cultures for prophylaxis and treatment of gastrointestinal diseases.  相似文献

We investigated the use of bacterial cells isolated from paddy crab for the extraction of oil from Jatropha seed kernels in aqueous media while simultaneously preserving the protein structures of this protein-rich endosperm. A bacterial strain-which was marked as MB4 and identified by means of 16S rDNA sequencing and physiological characterization as either Bacillus pumilus or Bacillus altitudinis-enhanced the extraction yield of Jatropha oil. The incubation of an MB4 starter culture with preheated kernel slurry in aqueous media with the initial pH of 5.5 at 37?°C for 6?h liberated 73% w/w of the Jatropha oil. Since MB4 produces xylanases, it is suggested that strain MB4 facilitates oil liberation via degradation of hemicelluloses which form the oil-containing cell wall structure of the kernel. After MB4 assisted oil extraction, SDS-PAGE analysis showed that the majority of Jatropha proteins were preserved in the solid phase of the extraction residues. The advantages offered by this process are: protein in the residue can be further processed for other applications, no purified enzyme preparation is needed, and the resulting oil can be used for biodiesel production.  相似文献

高效降解石油细菌的分离鉴定及降解能力的研究   总被引:2,自引:0,他引:2  
目的:为获得高效降解原油的菌株,从石油污染严重的土壤中采样,富集分离得到原油降解菌,并初步考察它们降解原油的能力。方法:通过富集培养、多次筛选分离得到三株优势菌,编号为SWH-1、SWH-2和SWH-3。通过16S rDNA序列分析和NCBI数据库的Blast比对分析,对其鉴定到种。通过差量法测定它们在室内摇瓶中对原油的降解率。结果:经鉴定,这三株菌分别为枯草芽孢杆菌(Bacillus subtillus)、多食鞘氨醇杆菌(Sphingobacterium multivorum)和嗜温鞘氨醇杆菌(Sphingobacterium thalpophi-lum)。在0.5g/L的原油培养基内培养1w,SWH-1和SWH-2的降解率较高,分别为33.89%和46.31%。将这两株菌进行混合培养降解原油,降解率高达51.73%。结论:所筛选到的枯草芽孢杆菌和多食鞘氨醇杆菌在生物修复方面具有很好的应用潜力,而且多食鞘氨醇杆菌在石油降解方面的报道尚属首次。  相似文献

Summary Removal of cooking oil from the domestic wastewater was carried out by immobilized photosynthetic bacteria, Rhodobacter shaeroide S (S) and R. shaeroides NR-3 (NR-3). The microorganisms were immobilized in sodium alginate (2%) or agar (2%). We treated 50 g of cooking oil suspended in artificial sewage wastewater under anaerobic dark conditions in a 15 l acrylic vessel. Results show that after 6 days of batch treatment, 74.2, 58.2 and 15.8% of oil was removed with the alginate-immobilized S, NR-3, and control, respectively. Relatively larger accumulations of volatile fatty acids such as propionic and acetic acids were observed in the control experiment compared with that in the immobilized cell (alginate and agar) experiment. In addition, continuous treatment of oil-containing wastewater was carried out with agar-immobilized S at a fixed dilution rate of 0.4/day. These results indicate that 96% of the oil was removed from the wastewater, and the maximum removal rate of oil reached approximately 3.83 kg oil/m3/d.  相似文献

在研究天然水沤法脱胶的过程中,通过初筛、复筛,从沤麻主生物期的沤麻液中筛选出两株茵落周围产生透明圈较大、脱胶酶活较高的菌株。通过形态观察,并对其多项生理、生化指标进行了分析研究,初步鉴定并命名为枯草芽孢杆菌A1和B1。初步加茵脱胶实验表明:枯草芽孢杆菌A1产生果胶酶、木聚糖酶,而不产生纤维素酶,脱胶周期为72小时;枯草芽孢杆茵B1产生果胶酶、木聚糖酶和纤维素酶,脱胶周期为50小时。  相似文献

目的:根据目标菌株的特性,应用选择培养基筛选出提高石油三次采油采收率的菌株,并对其进行相关性质的鉴定。方法:从大庆原油样品中经过富集培养,平板分离。结果:获得2株细菌,1号菌株属于假单胞菌属,2号菌株属于芽孢杆菌属。对两株细菌进行了定性分析,结果表明两株细菌均能产酸、产气、产表面活性剂。证明其具有降低原油黏度的作用。  相似文献

Accumulation of petroleum hydrocarbon residual considered a major environmental problem in the kingdom of Saudi Arabia cause of intensive efforts for oil detecting. Until now, In situ biodegradation considered the most effective method for petroleum hydrocarbon residual biodegradation. The aim of this study is isolation and identification biodegradable capability bacteria from contaminated sites in Khurais oil field, Dhahran, Saud Arabia via Different morphological and biochemical and molecular methods. Furthermore, degradation level in contaminated liquid medium and soil were evaluated. Three bacterial strains were selected from petroleum-contaminated soils of Khurais oil field depending on their capacity to grow in the existence of hydrocarbon components and identified according to morphological, biochemical. Interestingly, 16S rDNA sequencing fingerprinting results confirmed our bacterial identification as Bacillus subtilis, Pseudomonas aeruginosa and Bacillus cereu. Phyllogenetic tree was constructed and genetic similarity was calculated according to alignments results. Biodegradation patterns for different three isolates were reflected varied degradation ability for three isolates regarding incubation time. Different features were studied for three biodegrading bacterial strains and identified as Bacillus subtilis, Pseudomonas aeruginosa and Bacillus cereus. Remarkable degradation rate % patterns for hydrocarbons residual were recorded for all three isolates with varied.  相似文献

The aim of this study was to verify the effectiveness of Achillea ligustica essential oils against several oral microorganisms in comparison with a commercial essential oil-containing mouthrinse (Listerine(?)) and clove oil (containing 89% eugenol). The inhibition efficacy of A. ligustica essential oils alone and in combination with Listerine(?) was evaluated by the micro-dilution method. The most susceptible microorganisms were Bacillus cereus, Streptococcus pyogenes, and Candida albicans. The efficacy was similar to that of the clove oil. The antiseptic mouthwash Listerine(?) did not exert a strong inhibition on microbial strains tested, whereas its effectiveness increased significantly when essential oil was added. The study provides additional evidence for the in vitro inhibitory activity of A. ligustica essential oils on several pathogens, suggesting their usefulness in mouthrinse formulations as an adjunct to mechanical oral hygiene regimens. Essential oil-containing mouthrinses can be beneficial, safe components of daily oral health routines, representing an efficient and without side effect alternative to prevent and control oral infections.  相似文献

为提高育苗基质中废弃物木质素降解速率,在废弃物堆腐生产育苗基质高温阶段取样,筛选耐高温木质素降解菌,并对菌种进行鉴定,同时测定其对秸秆木质素和菌糠木质素的降解效果。获得了1株较好的木质素高温降解菌HZ11,鉴定为解淀粉芽胞杆菌(Bacillus amyloliquefaciens),结果显示,该菌株对秸秆木质素和菌糠木质素降解效果较好,50 ℃条件下,20 d木质素降解率分别为46.7%和42.4%。菌株HZ11在降解秸秆和菌糠方面具有很好的应用潜力,为利用农业废弃物生产育苗基质提供更加丰富的菌种资源,具有重要的参考价值。  相似文献

Contamination of water by meat production is an important and extensive environmental problem and even threat to human health. Biodegradation is a major mechanism which removes the pollutants from the environment. Therefore, the present study aimed to isolate and characterize a COD degrading bacteria which can effectively degrade slaughter wastewater. Six COD degrading bacteria were isolated from slaughtering waste water and sludge in Hunan a meat product Co., Ltd. And the COD degradation rate of each strain was determined by potassium permanganate method. Through observing morphologically and analyzing sequence to 16S rDNA, the highest COD degradation strain was Bacillus velezensis by preliminarily identified and classified, reaching 11.80%. The suitable conditions of the growth of Bacillus velezensis strain were 37 °C, pH 7.0, the peptone concentration 1.5%, and the yeast extract concentration 0.8%.  相似文献
