Discharge of wastewater from electroplating and leather industries is a major concern for the environment due to the presence of toxic Cr6+ and other ions, such as sulfate, nitrate, phosphate, etc. This study evaluated the potential of Tradescantia pallida, a plant species known for its Cr bioaccumulation, for the simultaneous removal of Cr6+, SO42?, NO3?, and PO43?. The effect of different co-ions on Cr6+ removal by T. pallida was examined following the Plackett-Burman design of experiments carried out under batch hydroponics conditions. The results revealed a maximum removal of 84% Cr6+, 87% SO42?, 94% NO3? and 100% PO43? without any phytotoxic effect on the plant for an initial Cr6+ concentration in the range 5–20 mg L?1. SO42? and NO3? enhanced Cr uptake at a high initial Cr concentration (20 mg L?1), whereas PO43? did not affect Cr uptake both at high and low initial Cr concentrations. The Cr6+ removal kinetics in the presence of different ions was well described by the pseudo-second-order kinetic model which revealed that both biosorption and bioaccumulation of the metal played an important role in Cr6+ removal. Increase in the total carbohydrate and protein content of the plant following Cr6+ and co-ions exposure indicated a good tolerance of the plant toward Cr6+ toxicity. Furthermore, enhancement in the lipid peroxidation and catalase activity in T. pallida upon Cr6+ exposure revealed a maximum stress-induced condition in the plant. Overall, this study demonstrated a very good potential of the plant T. pallida for Cr6+ removal from wastewater even in the presence of co-ions.  相似文献

The longer the seeds of dwarf French bean (Phaseolus vulgaris)were soaked in water before sowing, the smaller were the seedlingsthey produced. Soaking for 24 hours removed only little drymatter, but detectable amounts of a gibberellin and a betaine.The amounts of these two removed increased with increased timeof soaking. Long soaking of seeds decreased the amounts of gibberelinand auxin in the cotyledons, and of gibberellin, auxin, andbetaine in the primary leaves of seedlings. Aerating, cooling,or increasing the volume of water in which the seeds were soakedalso retarded the growth of the seedlings, but did not affectthe amount of growth substance removed from the seeds or containedin the cotyledons. The deleterious effect of soaking seeds wasnot alleviated by treating the seeds or seedlings with gibberellicacid or glycine betaine. The growth yield of Chlorella vulgaris, Y kJ defined as g cells harvested per kJ of light energy absorbed by the cells, was assessed in a turbidostat culture by varying CO2 and O2 partial pressures ( and ). The value of Y kJ ranged from 3.1×10-3 to 5.0×10-3 g cells/kJ under light-limited conditions [ = 1.02.4%, = 065%; total pressure of gas (composed of CO2, O2 and N2)=1 atm]. In the light-limited environment, the algal specific growth rate deteriorated appreciably with the increase of . The deterioration accounts for the above range of Y kJ observed. The growth inhibition due to oxygen that was defined by subtracting from 1.0 the ratio of at given values of to that at = 0% extended from 0.07–0.30 (7–30%). However, glycolate could not be detected in the turbidostat culture. Isotopic experiments on the specific rate of 14CO2 uptake also revealed that the inhibition due to oxygen was from 22–38% when was varied from 0 to nearly 100%. CCC (2-chloroethyltrimethylammonium chloride) decreased growth of primary leaves and stem internodes above the hypocotyl of dwarf French bean, probably because it decreased the amount of auxin produced by interaction between tryptophan and phenolic esters in the primary leaves. Growth of leaves was less affected than that of stems, and the gibberellin content of the primary leaves, previously shown to be associated with their expansion, was unaffected by CCC. CCC delayed death of the primary leaves, the breakdown of chlorophyll, and the increase in auxin associated with death of bean leaves. CCC had less effect on the growth of leaf discs, probably because they are not sites of growth-substance production. Glycine betaine, a quaternary ammonium compound similar to CCC, did not affect growth of bean plants, their metabolism of growth substances or the longevity of their primary leaves, but inhibited growth of leaf discs, probably directly.  相似文献

Pod storage protein (PSP) accumulated in developing pods of French bean (Phaseolus vulgaris L.) plants, and increasing the PSP mRNA level by pod removal resulted in the enhancement of PSP accumulation in pods that formed later. Pod storage protein was detected in flowers, young leaves and young stem internodes in addition to pods. Accumulation of PSP and its mRNA was induced by sink-removal in an organ-specific manner. In addition, wounding induced PSP accumulation systemically in leaves. Methyl jasmonate did not induce PSP synthesis but enhanced the synthesis that was induced by wounding. In senescing pods, PSP was degraded, and degradation products with molecular masses of 20 and 17 kDa were detected in the pods. Summary Bean (Phaseolus vulgaris L. Cv. Burpees Stringless) seeds were subjected to two cycles of presowing soaking and drying treatments with sodium molybdate and cobalt nitrite at 1 and 5 ppm concentrations used separately and also in combination. Sodium molybdate 2 ppm and cobalt nitrite 1 ppm used singly proved better than the remaining treatments with respect to nodulation, dry matter, nitrogen and yield. Combined treatment with sodium molybdate and cobalt nitrite did not produce additive effect on any parameter studied compared to their usage alone.  相似文献

Conjugal plasmid transfer was examined on the phylloplane of bean (Phaseolus vulgaris) and related to the spatial distribution pattern and metabolic activity of the bacteria. The donor (Pseudomonas putida KT2442) harbored a derivative of the TOL plasmid, which conferred kanamycin resistance and had the gfp gene inserted downstream of a lac promoter. A chromosomal insertion of lacIq prevented expression of the gfp gene. The recipient (P. putida KT2440) had a chromosomal tetracycline resistance marker. Thus, transconjugants could be enumerated by plating and visualized in situ as green fluorescent cells. Sterile bean seedlings were inoculated with donors and recipients at densities of approximately 105 cells per cm2. To manipulate the density and metabolic activity (measured by incorporation of [3H]leucine) of the inoculated bacteria, plants were grown at various relative humidities (RH). At 100% RH, the transconjugants reached a density of 3 × 103 CFU/cm2, corresponding to about one-third of the recipient population. At 25% RH, numbers of transconjugants were below the detection limit. Immediately after inoculation onto the leaves, the per-cell metabolic activity of the inocula increased by up to eight times (100% RH), followed by a decrease to the initial level after 96 h. The metabolic activity of the bacteria was not rate limiting for conjugation, and no correlation between the two parameters was observed. Apparently, leaf exudates insured that the activity of the bacteria was above a threshold value for transfer to occur. Transconjugants were primarily observed in junctures between epidermal cells and in substomatal cavities. The distribution of the transconjugants was similar to the distribution of indigenous bacteria on nonsterile leaves. Compared to polycarbonate filters, with cell densities equal to the overall density on the leaves, transfer ratios on leaves were up to 30 times higher. Thus, aggregation of the bacteria into microhabitats on the phylloplane had a great stimulatory effect on transfer.  相似文献

食物的数量和质量是影响动物免疫功能的重要因素。野生动物在选择食物时是否会优先选择有利于其健康的食物?为解决该问题,我们以黑线仓鼠(Cricetulus barabensis)为研究对象,通过饲喂其喜食食物花生(Arachis hypogaea),然后检测反映健康状况的细胞免疫能力和血液指标。将30只非繁殖期成年雌性黑线仓鼠随机分为正常饲料组(normal fodder group,NF,n=10)、花生加饲料组(normal fodder plus peanuts group,NP,n=10)和花生组(Peanut,n=10),饲喂39 d后检测各项指标。结果发现Peanut组的体重增加,NP和Peanut组的摄食量降低。NP组注射植物血球凝集素(Phytohaemagglutinin,PHA)后12 h的PHA反应显著高于NF组。3组间白细胞数和淋巴细胞数在统计学上没有显著差异,但NP和Peanut组白细胞数比NF组分别高22.9%和25.0%,淋巴细胞数分别高15.0%和22.5%。NP和Peanut组红细胞数和红细胞压积显著高于NF组,而Peanut组血小板数显著高于NF和NP组。这些结果表明饲喂花生能增强黑线仓鼠的细胞免疫能力、输氧能力和凝血功能,这可能是其喜食花生的生理原因。  相似文献

Polyhydroxyalkanoate (PHA) synthase PhaC plays a very important role in biosynthesis of microbial polyesters PHA. Compared to the extensively analyzed C-terminus of PhaC, N-terminus of PhaC was less studied. In this paper, the N-terminus of two class I PHA synthases PhaCRe and PhaCAh from Ralstonia eutropha and Aeromonas hydrophila, respectively, and one class II synthase PhaC2Ps of Pseudomonas stutzeri strain 1317, were investigated for their effect on PHA synthesis. For PhaCRe, deletion of 2–65 amino acid residues on the N-terminus led to enhanced PHB production with high PHB molecular weight of 2.50 × 106 Da. For PhaCAh, the deletion of the N-terminal residues resulted in increasing molecular weights and widening polydispersity accompanied by a decreased PHA production. It was found that 3-hydroxybutyrate (3HB) monomer content in copolyesters of 3-hydroxybutyrate and 3-hydroxyhexanoate (3HHx) increased when the first 2–9 and 2–13 amino acid residues in the N-terminus of PhaC2Ps were deleted. However, deletion up to the 40th amino acid disrupted the PHA synthesis. This study confirmed that N-terminus in different types of PHA synthases showed significant roles in the PHA productivity and elongation activity. It was also indicated that N-terminal mutation was very effective for the location of functional regions at N-terminus.  相似文献

The ethylene releasing compound, 2-chloroethylphosphonic acid (ethephon) inhibited nodule development in common bean (Phaseolus vulgaris L.) plants. In contrast, inhibitors of ethylene synthesis or its physiological activity enhanced nodulation. In a co-culture of bean seeds and rhizobia, ethephon inhibited rhizobial growth while inhibitors of ethylene synthesis or action did not influence the growth and proliferation of rhizobia. These data emphasize the role of ethylene as a regulator of nodulation in determinate nodulators and indicate that the ethylene signaling pathway involved in the nodulation process is not limited to the plant host but also involves the bacterial symbiont.  相似文献

Membrane fractions from bean hypocotyl or callus incorporate arabinose from UDP--L-arabinose into arabinan and xylose from UDP--D-xylose into xylan. The control of these syntheses has been studied during xylogenesis in stele and in xylogenesis induced in callus tissue. Induction of arabinan synthetase activity occurs during division and extension growth while that of xylan synthetase occurs subsequently during the period of secondary thickening of the cell wall. In order to investigate the effect of N form on dry matter (DM) formation and water uptake rate, French bean (Phaseolus vulgaris L. `Sotaxa') plants were grown with a split-root system. Three treatments were compared: sole nitrate (NO 3) supply (NN), sole ammonium (NH+ 4) supply (AA) and spatially separated supply of NO 3 and NH+ 4 (NA). The pH of the nutrient solutions was kept constant at 6.3 using a pH-stat system. 9 days after onset of the treatments, NN plants had higher root (36%) and shoot dry matter (11%) than AA plants. N form drastically influenced partitioning of assimilates: in the NA treatment, the root half exposed to NO 3 revealed a 170% higher DM than the root half exposed to NH+ 4. N form affected stable carbon-isotope discrimination () of leaf tissue. In leaves of plants which were supplied with NH+ 4 (AA; NA) was significantly more negative (–29.4, –29.6) than in NN treatment (–28.2). We explain this effect by differences in stomatal conductance. We suppose that the significantly less negative of root tissue under NH+ 4 supply is most probably related to higher PEP-case activity. The water uptake rate was higher in NN than in AA grown plants. This effect was found in both, short- and long-term experiments. In case of NA plants, the water uptake in the root part being exposed to NO 3 was 104% higher than in those receiving NH+ 4. At least in the case of the NA treatment we can exclude shoot growth effects as being responsible for differences in water uptake. We therefore assume that differences in root hydraulic conductivity are responsible for the observed effects.  相似文献

BOA (2-benzoxazolinone) is a potent phytotoxin present in several graminaceous crops such as rye, maize and wheat. Due to its wide range of phytotoxicity, it is considered as a potential pesticide. A study was conducted to explore the impact of BOA on the radicle and plumule elongation of mung bean (Phaseolus aureus) and associated changes in the macromolecular content - proteins and carbohydrates - and activities of enzymes like amylases, proteases, polyphenol oxidases and peroxidases. BOA significantly reduced the radicle and plumule length of P. aureus, and the contents of proteins and carbohydrates in both root and leaf tissue. On the other hand, activities of hydrolytic enzymes - proteases, amylases, polyphenol oxidases and peroxidases - increased substantially in both root and leaf tissue of P. aureus upon BOA exposure. This indicated that BOA treatment induced stress in P. aureus and enhanced enzyme activities to counter the induced stress and continue the growth. In other words, BOA-induced stress altered the plant biochemical status and related enzyme activities resulting in increased metabolism that serves to provide protection against cellular injury. Such studies providing information about the biomolecular content and enzymatic activities in response to natural products serve as clues for furtherance of knowledge about the modes of action of natural compounds of commercial interest.  相似文献

The survival of Rhizobium leguminosarum biovar phaseoli on seeds of bean was tested, using the cultivar Carioca. The seeds were treated seven days before inoculation with Benlate, Vitavax, Banrot, Difolatan or Ridomil fungicides. The rhizobial strains used were: CIAT 899, CPAC 1135 and CIAT 652. Strain CIAT 899 showed greater survival on the seed with fungicide than the other strains. Two hours after the contact with fungicides strains CIAT 652 and CPAC 1135 had significantly lower numbers of rhizobia than the treatment without fungicide. The Benlate and Banrot fungicides had the greatest effect on survival of rhizobial strains. There was a drastic mortality of the two strains, CIAT 652 and CPAC 1135, on seeds treated with Benlate and Ridomil. Under field conditions, granular inoculation produced fewer nodules, but a similar total nodule weight as seed inoculation. Serological tests (ELISA) showed that seed treatment with Benlate in connection with seed inoculation reduced drastically the occurrence of inoculated strains in nodules, while the same fungicide treatment and inoculation applied in the seed furrow did not affect the survival of the inoculated strain.  相似文献

The plant growth regulators, gibberellic acid (GA3), ethephon and chlormequat chloride (CCC) were sprayed on young lettuce, cauliflower and bean (Phaseolus vulgaris) plants, which had either been given or not been given a mechanically-induced stress (MIS) treatment. MIS was applied by brushing the plants with paper for 1.5 minutes each day. GA3 increased extension growth of bean and leaf length of lettuce in unbrushed plants as much as in brushed ones. CCC and ethephon were less effective at reducing the height of brushed bean plants compared to unbrushed ones. The effects of CCC on the growth of cauliflower and lettuce plants was not influenced by brushing, whereas unbrushed plants responded more readily to ethephon than did brushed ones. The effects of CCC on growth were generally similar to those of MIS whereas the effects of ethephon were in many ways different to MIS.The results are discussed in relation to the use of PGR and MIS treatments for modifying plant growth.  相似文献

Brassinolide, as a plant hormone, promotes growth of a number of plant species. Similar effects are induced by its epimer 24-epibrassinolide. In this paper we discuss the effects of brassinosteroids on the growth and proton extrusion in the green alga Chlorella vulgaris (Chlorophyceae). At concentrations between 10–15 and 10–8 m, brassinolide and 24-epibrassinolide induce a significant stimulation of growth and H+ extrusion.
