多孔板-MTF比色法测定植物和微生物代谢产物对真菌抑制活性的步骤为:在多孔板的每孔中依次加入浓度为105孢子/mL的供试真菌孢子悬液90μL,不同浓度的药液10 μL.25℃暗培养48 h,然后每孔中加入8mg/mL的MTT溶液10μL,继续培养10 h后,离心去上清,加入DMS0 150 μL,振荡30 min,离心后上清液在510nm测定吸光值.采用上述条件测定了白屈菜红碱对稻瘟病菌和西瓜枯萎病菌的MIC值分别为80和1.5μg/mL,IC50值分别为21.99和0.78 μg/mL;Diepoxinζ对稻瘟病菌的MIC和IC50值分别为200和96.21 μg/mL.多孔板-MTT比色法为快速有效地筛选和评价植物和微生物抗真菌活性成分创造了条件.  相似文献

采用活性追踪的方法从盾叶薯蓣内生芬芳镰刀菌Dzf2中分离到两个抗菌活性成分,通过物理化学性质和波谱学特征鉴定为镰刀菌酸(1)和9,10-脱氢镰刀菌酸(2)。采用多孔板-MTT-比色法和孢子萌发法测定了化合物的抗菌活性。镰刀菌酸和9,10-脱氢镰刀菌酸对供试细菌的半抑制浓度(IC50)值为35.35μg/mL至171.29μg/mL;对稻瘟菌孢子萌发的IC50值分别为28.83μg/mL和27.06μg/mL。  相似文献

1mg/mL KCl促进荷叶离褶伞菌丝生长;5或10mg/mL NaCl、5或10mg/mL MgSO4、5或10mg/mLKCl、10mg/mL H2PO4和1mg/mL CaSO4抑制菌丝生长;0.8mg/mL的MnSO4和CuSO4以及0.5mg/mL FeSO4、0.2或0.5mg/mL CoCl2和0.2、0.5或0.8mg/mL ZnSO4促进菌丝生长;0.5或0.2mg/mL CuSO4、0.2或0.5mg/mL MnSO4及0.8或0.2mg/mL FeSO4对菌丝生长的影响不显著;维生素B6、维生素C、维生素PP和维生素B1可促进菌丝生长,在含有10μg/L维生素B6的培养基上菌丝生长速度最快,但维生素C试用浓度较低(50μg/L)时对菌丝生长的影响不显著;吲哚丁酸、吲哚乙酸、奈乙酸对菌丝生长具有促进作用,但0.1、0.5或1.0μg/L赤霉素对菌丝生长的影响不显著。  相似文献

目的:验证重组人BD3-BPI(rhBD3-BPI)是否具有内毒素中和活性,研究其在高盐环境中是否能保持抗菌活性。方法:根据内毒素标准品绘制内毒素活性标准曲线,将100 μL梯度稀释的rhBD3-BPI-与100 μL 10EU/mL脂多糖(LPS)混匀,37℃水浴60min,同时设标准对照(只含10EU/mL LPS的标准品溶液),并以无热源水作为空白对照,采用基质显色法进行鲎试验测定LPS的活性;将6×10^8 CFU/mL的革兰阳性和阴性标准菌株及临床分离的多药耐药菌株接种于含1mg/mL rhBD3-BPI和0~250mmol/L不同浓度NaCl的液体细菌培养基中,37℃培养3h后用10mmol/L磷酸钠按1:1~1:1000的比例稀释,铺LB培养基平板,37℃过夜培养,观察各平板菌落生长情况,计数并计算杀菌率。结果:在5EU/mL的标准内毒素体系中,当rhBD3-BPI的浓度高于4μg/mL时即开始表现出一定的内毒素中和活性,当rhBD3-BPI的浓度分别为16、32 μg/mL时,其内毒素中和率分别为23%和88%,随后rhBD3-BPI对内毒素的中和活性趋于平稳,50 μg/mL的rhBD3-BPI对所有受检菌均表现出100%的杀伤效应。当NaCl浓度低于150mmol/L时,rhBD3-BPI对各受检菌的杀菌活性均未受明显影响;NaCI浓度升高至150-200mmol/L,rhBD3-BPI对各受检菌的杀菌活性有所下降,但其杀伤率仍在90%以上;当NaCl浓度高于200mmol/L时,盐浓度对rhBD3-BPI杀菌活性的影响才较为明显,但即使NaCl浓度达到250mmol/L,rhBD3-BPI的杀菌活性仍保持在85%以上。结论:rh-BD3-BPI具有内毒素中和活性,在高盐环境中具有良好的抗菌活性稳定性。  相似文献

几种药用植物精油的抗真菌活性研究   总被引:5,自引:0,他引:5  
研究了作用浓度为0:7-4.0μL/mL的南方药用植物香茅(Cymbopogon citratus)、广藿香(Pogostemon cablin(Blanco)Benth)、大叶桉(Eucalyptus robusta Smith)和蛇床(Cnidium monnier(L.)Cusson)的精油对6种植物病原真菌的抑制活性。结果表明,大叶桉、广藿香和香茅精油对串珠镰刀病菌(Fussarium moniliforme)、玉米弯孢霉菌(Curvjlaria lunata)对多种供试植物病原真菌的生长有明显抑制活性,其中,香茅精油在浓度为3.3μL/mL时,能完全抑制大部分供试真菌的生长。但不同精油对于各病原真菌的抑制率有一定差异。本研究为复合植物精油抗真菌农药的开发提供依据。  相似文献

考察壳聚糖(chitosan)、壳寡糖(chitosanoligosaccharides,COS)、茉莉酸甲酯(methyljasmonate,MJ)、水杨酸(salicylicacid,SA)和Cu2+等诱导子对藏红花悬浮培养细胞生长和藏红花色素合成的影响。结果表明:在实验考察浓度范围内,壳寡糖(1~500mg/L)和较低浓度壳聚糖(≤10mrdL)、MJ(≤10μmol/L)、SA(≤10μμmol/L)和Cu2+(≤1μmoL/L)对细胞生长无显著影响;较高浓度壳聚糖(≥100mg/L)、MJ(≥100μmol/L)、SA(≥100μmoL/L)和cu“(≥10μmoL/L)显著抑制细胞生长。5种诱导子对藏红花色素合成的诱导效果不同,并且与诱导子作用浓度和添加时间有关。MJ诱导效果最好,在细胞培养第0天添加终浓度100仙moL/LMJ,藏红花色素含量(以1克干细胞计)达到28.57mg,比对照提高177.9%。其次是cu“,在细胞培养第4天添加终浓度500μmoL/LCu2+,色素含量达到19.82mg,比对照提高108.2%。再次是壳聚糖和壳寡糖,在细胞培养第14天分别添加终质量浓度100mg/L壳聚糖和壳寡糖,色素含量分别达到18.33和17.39mg,比对照提高69.1%和69.0%。最后是SA,在细胞培养第14天添加终浓度10μmoL/LSA,色素含量达到14.65mg,比对照提高45.4%。  相似文献

目的:研究枳实提取物及其药效组分橙皮苷和新橙皮苷对氧化低密度脂蛋白(oxidized lowd ensity lipoprotein,Ox—LDL)损伤的人脐静脉内皮细胞(human umbilical vein endothelial cells line,HUVEC)细胞间黏附分子-1(intercellular adhesion molecule-1,ICAM-1)表达和一氧化氮(nitric oxide,NO)释放的影响。方法:体外培养HUVEC,50μg/mLOX—LDL制造HUVEC损伤模型。以MTS染色法检测细胞毒性确定用药浓度。细胞ELISA法测定细胞表面ICAM-1的含量,试剂盒测定细胞培养上清液中NO含量。结果:①枳实提取物小于等于2mg/mL时,橙皮苷浓度小于等于0.03125mg/mL时,新橙皮苷浓度小于等于0.25mg/mL时,HUVEC存活率分别大于80%。②2.0mg/mL和1.0mg/mL两个浓度的枳实提取物、15.625μg/mL的橙皮苷和0.2500mg/mL新橙皮苷对OX—LDL诱导的HUVEC的ICAM-1表达有显著抑制作用。③2.0mg/mL枳实提取物显著提高OX—LDL诱导的HUVEC和正常HUVEC培养液中的NO含量;7.813ixg/mL、15.625μg/mL和31.250μg/mL 3个浓度的橙皮苷能显著提高OX—LDL诱导的HUVEC培养液中的NO含量,31.250μg/mL的橙皮苷能促进正常HUVEC的NO释放;0.2500mg/mL和0.1250mg/mL 2个浓度的新橙皮苷能显著提高OX—LDL诱导的HUVEC培养液中的NO含量。结论:枳实提取物及其药效组分橙皮苷、新橙皮苷能抑制Ox-LDL诱导的HUVEC的ICAM-1表达,促进Ox-LDL诱导的HUVEC的NO释放。  相似文献

对喜树植株中的内生真菌进行分离纯化,共得到126株内生菌株。对126株内生菌株进行液体培养,并对其发酵产物进行抗菌活性测定。发现在供试质量浓度为5mg/mL时,126株菌株中有24株喜树内生真菌的发酵液对水稻纹枯病菌、稻瘟病菌以及辣椒疫霉具有不同程度的抑制菌丝生长的活性作用;其中,菌株XSY09的发酵液对此三种植物病原菌有明显的抑制作用,抑制率分别为74.97%、39.63%和58.49%。在0.1mg/mL的测试浓度下,菌株XSY09发酵液的乙酸乙酯相对三种植物病原菌均有较好的抑制作用。对该菌株的ITS序列进行测序分析,初步鉴定XSY09为炭疽菌属(Colletotrichum gloeosporioides)真菌。  相似文献

为达到鼻咽癌(nasopharyngeal carcinoma,NeC)的靶向化疗,该研究通过酰胺化反应和配位偶联技术制备叶酸(folicacid,FA)分子靶向载川页铂(cisplatin,CDDP)羧甲基-β-环糊精(carboxymethyl-β-cyclodextrin,CM—β—CD)纳米复合物(FA-CM—β—CD—CDDP),采用邻苯二胺(o-phenylenediamine,OPDA)比色法检测复合物中CDDP含量,紫外分光光谱检测FA含量,透射电镜观察复合物形态,激光粒度仪测定复合物粒径大小。荧光显微镜观察NPC叶酸受体(folatereceptor,FR)阳性HNE-1细胞及FR阴性CNE-2细胞对偶联FITC的复合物的吞噬及OPDA比色法检测细胞内CDDP的浓度。通过MTT法、集落形成实验和流式细胞术检测复合物对HNE-1细胞增殖能力和凋亡的影响。研究结果显示,复合物中偶联的FA和CDDP浓度分别为340gg/mL和2mg/mL,CDDP包封率达20.00%,复合物粒径均匀且大小为157.8nm。HNE-1细胞内见较多FITC,细胞内CDDP浓度为6.24ng/mL,而CNE-2细胞内FITC较少,细胞内CDDP浓度仅约2.01ng/mL。HNE-1生长抑制率在24h明显高于对照组(CM—β—CD—CDDP),其IC50(4.80μg/mL)明显低于对照组(6.97μg/mL),但当所载的CDDP终浓度达到16.00μg/mL时,两组抑制率均达到80%以上;作用48h两组抑制率无明显差异。在24h,当复合物的CDDP终浓度为1.00μg/mL时,HNE—1的集落形成率为33.21%,明显低于对照组(52.27%)。当复合物的CDDP终浓度为0.25μg/mL和1.00μg/mL时,HNE-1的凋亡率分别达12.65%和22.35%,明显高于对照组(6.91%和14.21%)。研究结果表明,成功构建的FA—CM—β—CD.cDDP纳米复合物能够靶向抑制FR阳性的NPC细胞增殖并促进其凋亡。  相似文献

采用水蒸气蒸馏法提取侧柏叶精油,通过GC和GC-MS分析精油的组成。从侧柏叶精油中鉴定出29种成分,主要为单萜和倍半萜,含量较高的化合物有:8-丙氧基-柏木烷(15.41%)、松油烯-4-醇(12.98%)、α-蒎烯(9.85%)、桧烯(9.92%)和3-蒈烯(6.77%)。侧柏叶精油具有广谱的抗细菌活性,对供试细菌的最低杀菌浓度(MBC)在0.1 mg/mL和1.0 mg/mL之间,最低抑菌浓度(MIC)在0.063 mg/mL和0.90 mg/mL之间,半抑制浓度(IC50)在0.044 mg/mL和0.763 mg/mL之间。侧柏叶精油能有效抑制菲啰嗪与Fe2+的螯合(IC50值为0.409mg/mL)和β-胡萝卜素-亚油酸的氧化(IC50值为0.526 mg/mL)。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
