The effects of different spectral light distribution on in vitro induction and proliferation of Oncidium protocorm-like bodies (PLBs) and subsequent growth of plantlets were investigated. Shoot tips (5 mm in length) of proliferating shoots of Oncidium “Gower Ramsey” were vertically incubated on 1/2 Murashige and Skoog (MS) medium supplemented with 1.0 mg l−1 6-benzyladenine (BA), and grown under either monochromatic red light-emitting diodes (LEDs) (RR), blue LEDs (BB), yellow LEDs (YY) or green LEDs (GG). Cultures grown under fluorescent lamps (FL) were used as control. Selected FL-induced PLBs were cut into 3- to 4-mm sections and incubated on MS medium supplemented with 1.0 mg l−1 BA and 0.5 mg l−1 α-naphthaleneacetic acid (NAA), and grown under RR, BB, YY, GG, or FL. Moreover, FL-differented shoots (15 mm in length with two leaves) were incubated on 1/2 MS medium with 0.5 mg l−1 NAA, and grown under either FL, RR, 10% blue + 90% red LEDs (1BR), 20% blue + 80% red LEDs (2BR), 30% blue + 70% red LEDs (3BR), BB, 80% red + 10% blue + 10% far-red LEDs (RBFr), or 80% red + 10% blue + 10% green LEDs (RBG). Overall, the red light spectrum enhanced induction, proliferation, and the carbohydrate contents of PLBs, as well as subsequent plantlet lengths, while the blue spectrum promoted differentiation, protein accumulation, and enzyme activities in PLBs, as well as pigment content accumulation in PLBs and developing plantlets. The combination of red and blue LEDs resulted in higher energy efficiency as well as dry weight and enzyme activities in these plantlets.  相似文献

The new species Gymnopilus maritimus is described from coastal plant communities of Juncus maritimus, growing on sandy soil or on decaying plants, from northwestern Sardinia (Italy). The distinguishing features of G. maritimus are: (1) an unusual habitat, (2) robust basidiomata, (3) mild taste, and (4) big and strongly warted spores. The new species is compared with the micromorphologically similar species G. fulgens sensu auct. Brit. p.p. and the biogeographically and ecologically similar species G. arenophilus, as well as with other European species. A photograph of fresh material, drawings of the main micromorphological features, and FESEM and optical microscope microphotographs of basidiospores are added. Furthermore, some notes on micromorphological characters of G. arenophilus are presented and its distribution area enlarged with a record from France. A key for the European species of Gymnopilus morphologically, ecologically, and/or biogeographically related to G. maritimus is presented. The phylogeny inferred from ITS rDNA sequences revealed that G. maritimus represents an independent species and that it is not related to G. arenophilus or G. fulgens. It is the sister group of the clade containing G. imperialis and G. spectabilis, but with a bootstrap support below 50%. The characters shared by the species in this clade are: (1) robust basidiomata, (2) pileus fibrillose or scaly-fibrillose, and (3) spores longer than 8 μm, dextrinoid and strongly warted. Gymnopilus imperialis and G. spectabilis differ by the basidiomata with membranous ring in the stem, living on conifers or decaying wood, and having narrower or wider spores, respectively. Warburgia ugandensis Sprague is a woody species in the family Canellaceae and an important source of medicines in Africa. Natural propagation of W. ugandensis is problematic due to its recalcitrant seeds and lack of an efficient in vitro regeneration system for this species. This study describes an efficient regeneration protocol. Petiole bases and shoot tips were used as explants. Callus tissue developed when the explants were cultured on Murashige and Skoog medium containing 30 g L−1 sucrose and 7 g L−1 agar (MS30 medium), supplemented with 1.0 mg L−1 indole-3-butyric acid (IBA), 1.6 mg L−1 6-benzylaminopurine (BA), and 0.1 mg L−1 thidiazuron (TDZ). Adventitious buds were efficiently induced from the callus when the MS30 medium was supplemented with 0.8 mg L−1 BA and 0.2 mg L−1 IBA. Root induction occurred within 7–10 d on half-strength MS30 medium supplemented with 0.8–1.0 mg L−1 1-napthalene acetic acid (NAA), 0.2 mg L−1 IBA, and 0.03% (w/v) activated charcoal (AC). Roots were followed by root elongation on the same medium but lacking NAA and IBA. Approximately 50% of the plantlets cultured produced roots, while more than 80% of the plantlets survived and successfully grew to maturity.

Apurinic/apyrimidinic (AP) endonucleases are important DNA repair enzymes involved in two overlapping pathways: DNA glycosylase-initiated base excision (BER) and AP endonuclease-initiated nucleotide incision repair (NIR). In the BER pathway, AP endonucleases cleave DNA at AP sites and 3'-blocking moieties generated by DNA glycosylases, whereas in NIR, the same AP endonucleases incise DNA 5' to a wide variety of oxidized bases. The flowering plant Arabidopsis thaliana contains three genes encoding homologues of major human AP endonuclease 1 (APE1): Arp, Ape1L and Ape2. It has been shown that all three proteins contain AP site cleavage and 3'-repair phosphodiesterase activities; however, it was not known whether the plant AP endonucleases contain the NIR activity. Here, we report that ARP proteins from Arabidopsis and common wheat (Triticum aestivum) contain NIR and 3'  5' exonuclease activities in addition to their AP endonuclease and 3'-repair phosphodiesterase functions. The steady-state kinetic parameters of reactions indicate that Arabidopsis ARP cleaves oligonucleotide duplexes containing α-anomeric 2'-deoxyadenosine (αdA) and 5,6-dihydrouridine (DHU) with efficiencies (kcat/KM = 134 and 7.3 μM−1·min−1, respectively) comparable to those of the human counterpart. However, the ARP-catalyzed 3'-repair phosphodiesterase and 3'  5' exonuclease activities (kcat/KM = 314 and 34 μM−1·min−1, respectively) were about 10-fold less efficient as compared to those of APE1. Interestingly, homozygous A. thaliana arp–/– mutant exhibited high sensitivity to methyl methanesulfonate and tert-butyl hydroperoxide, but not to H2O2, suggesting that ARP is a major plant AP endonuclease that removes abasic sites and specific types of oxidative DNA base damage. Taken together, these data establish the presence of the NIR pathway in plants and suggest its possible role in the repair of DNA damage generated by oxidative stress.  相似文献

Coastal wetlands are key in regulating coastal carbon and nitrogen dynamics and contribute significantly to climate change mitigation and anthropogenic nutrient reduction. We investigated organic carbon (OC) and total nitrogen (TN) stocks and burial rates at four adjacent vegetated coastal habitats across the seascape elevation gradient of Cádiz Bay (South Spain), including one species of salt marsh, two of seagrasses, and a macroalgae. OC and TN stocks in the upper 1 m sediment layer were higher at the subtidal seagrass Cymodocea nodosa (72.3 Mg OC ha−1, 8.6 Mg TN ha−1) followed by the upper intertidal salt marsh Sporobolus maritimus (66.5 Mg OC ha−1, 5.9 Mg TN ha−1), the subtidal rhizophytic macroalgae Caulerpa prolifera (62.2 Mg OC ha−1, 7.2 Mg TN ha−1), and the lower intertidal seagrass Zostera noltei (52.8 Mg OC ha−1, 5.2 Mg TN ha−1). The sedimentation rates increased from lower to higher elevation, from the intertidal salt marsh (0.24 g cm−2 y−1) to the subtidal macroalgae (0.12 g cm−2 y−1). The organic carbon burial rate was highest at the intertidal salt marsh (91 ± 31 g OC m−2 y−1), followed by the intertidal seagrass, (44 ± 15 g OC m−2 y−1), the subtidal seagrass (39 ± 6 g OC m−2 y−1), and the subtidal macroalgae (28 ± 4 g OC m−2 y−1). Total nitrogen burial rates were similar among the three lower vegetation types, ranging from 5 ± 2 to 3 ± 1 g TN m−2 y−1, and peaked at S. maritimus salt marsh with 7 ± 1 g TN m−2 y−1. The contribution of allochthonous sources to the sedimentary organic matter decreased with elevation, from 72% in C. prolifera to 33% at S. maritimus. Our results highlight the need of using habitat-specific OC and TN stocks and burial rates to improve our ability to predict OC and TN sequestration capacity of vegetated coastal habitats at the seascape level. We also demonstrated that the stocks and burial rates in C. prolifera habitats were within the range of well-accepted blue carbon ecosystems such as seagrass meadows and salt marshes.

A total of 502 specimens were obtained during monthly samplings from July to December 2015, by fishermen using seine nets, gillnets and cast nets. Length–weight (LWRs) and length–length (LLRs) relationships were estimated for five fish species collected from the Atrai River in the northwestern region of Bangladesh. The values of slope (b) in LWRs (r2 > .913, p < .001) were determined as 2.899 for Aspidoparia jaya, 3.059 for Gagata cenia, 3.004 for Gagata youssoufi, 2.755 for Salmophasia bacaila, and 3.222 for Salmophasia phulo, respectively. The linear relationships in TL, FL and SL were highly correlated (for all cases: r2 > .897, p < .001). No previous information on LWRs (except for G. cenia) and LLRs (except for Sbacaila) of these selected species were found in FishBase.  相似文献

Little is known about how the growth of individual Gambierdiscus species responds to environmental factors. This study examined the effects of temperature (15–34 °C), salinity (15–41) and irradiance (2–664 μmol photons m−2 s−1) on growth of Gambierdiscus: G. australes, G. belizeanus, G. caribaeus, G. carolinianus, G. carpenteri, G. pacificus and G. ruetzleri and one putative new species, Gambierdiscus ribotype 2. Depending on species, temperatures where maximum growth occurred varied between 26.5 and 31.1 °C. The upper and lower thermal limits for all species were between 31–34 °C and 15–21 °C, respectively. The shapes of the temperature vs. growth curves indicated that even small differences of 1–2 °C notably affected growth potentials. Salinities where maximum growth occurred varied between 24.7 and 35, while the lowest salinities supporting growth ranged from <14 to 20.9. These data indicated that Gambierdiscus species are more tolerant of lower salinities than is generally appreciated. Growth of all species began to decline markedly as salinities exceed 35.1–39.4. The highest salinity tested in this study (41), however, was lethal to only one species, Gambierdiscus ribotype 2. The combined salinity data indicated that differences in salinity regimes may affect relative species abundances and distributions, particularly when salinities are <20 and >35. All eight Gambierdiscus species were adapted to relatively low light conditions, exhibiting growth maxima at 50–230 μmol photons m−2 s−1 and requiring only 6–17 μmol photons m−2 s−1 to maintain growth. These low light requirements indicate that Gambierdiscus growth can occur up to 150 m depth in tropical waters, with optimal light regimes often extending to 75 m. The combined temperature, salinity and light requirements of Gambierdiscus can be used to define latitudinal ranges and species-specific habitats, as well as to inform predictive models.  相似文献

Respiration of four freshwater species of the amphipod crustacean Gammarus: G. fossarum, G. lacustris (river and lake), G. pulex and G. roeseli were measured in a closed, stirred respirometric chamber with a micro-electrode. Oxygen consumption, expressed as weight-specific oxygen uptake (Rs) in relation to decreasing oxygen concentration, varied at air saturation from 0.86 (G. lacustris, lake) to 2.06 μl O2 mg−1 AFDW h−1 (G. pulex). Rs also differed intra-specifically among the two populations of G. lacustris. G. lacustris (river), G. pulex and G. roeseli expressed moderate ability to regulate their oxygen consumption at decreasing oxygen concentrations, whereas the regulation ability was higher in G. lacustris (lake) and in G. fossarum, which maintain high oxygen uptake at oxygen levels >2 mg O2 l−1. All four Gammarus species are partial regulators in response to variations in oxygen concentration. The differences between species are considered too small to account for their natural distributions. It appears that the tolerances of Gammarus species to organic pollution depend only in part on oxygen conditions.  相似文献

Leptocarpha rivularis, commonly known as “palo negro”, is a species of the Astereaceae family, endemic in the forests of Southern Chile. The extracts of this plant have medicinal properties attributable to its secondary metabolites, mainly identified as flavonoids and terpenes. Among them, leptocarpine has gained importance as a powerful anticarcinogen. The aim of this study was the in vitro establishment of L. rivularis to develop future alternatives to obtain biomass for medicinal purposes. Young shoots obtained from plants in growth chambers were treated with Captan™ (2.5% (w/v)), subjected to disinfection method with ethanol, commercial chlorine, and antioxidants and tested in different Murashige & Skoog based media to establish in vitro culture. The most effective growth was observed with half strength Murashige & Skoog medium supplemented with 20 g L−1 sucrose and 3.2 g L−1 gelrite (MGS medium) with 95% survival at 30-d and shooting reaching 82% of the explants when Plant Preservative Mixture™ is added to the medium. The addition of 1 mg L−1 of indole-3-butyric acid to the MSG medium was necessary for inducing roots and obtaining well-rooted plants that were acclimatized and successfully established in the greenhouse. Our study represents the first report describing the in vitro culture of L. rivularis, allowing the sustainable use of this plant.

About 5 per cent of follicular lymphoma (FL) cases are double‐hit (DH) lymphomas. Double‐hit follicular lymphoma (DHFL) cell lines can improve our understanding and drug development on FL. But there are only few DHFL cell lines. Here, we established a new MYC/BCL2 DHFL cell line, FL‐SJC. The cells were obtained from the hydrothorax of a patient with MYC/BCL2 DHFL and cultured for 140 passages in vitro. FL‐SJC cells demonstrated CD19++, CD20+, CD22++, HLA‐DR+, CD10+, CD38+, Lambda+ CD23, CD5 and Kappa. The chromosome karyotypic analysis confirmed the co‐existence of t(8;22)(q24;q11) and t(14;18)(q32;q21), as well as additional abnormalities involving chromosomes 2 and 3. Fluorescence in situ hybridization analysis (FISH) showed IGH/BCL2 fusion gene and the MYC rearrangement. In addition, the FL‐SJC cells displayed KMT2D/MLL2 and CREBBP gene mutations. After subcutaneous inoculation of FL‐SJC cells, the SCID mice developed solid tumour masses within 6‐8 weeks. FL‐SJC cells were proven to be free of Epstein‐Barr (EB) virus infection and be multidrug‐resistant. In a conclusion, the FL‐SJC cell line has been identified as a novel MYC/BCL2 double‐hit follicular lymphoma that can be used as a potentially available tool for the clinical and basic research, together with the drug development for MYC/BCL2 DHFL.  相似文献

The kinetics of the decomposition reaction of 4‐(4,5‐diphenyl‐1H‐imidazol‐2‐yl)phenyl acetate ( 1 ) in basic alcoholic media was investigated, using a simple fluorescence (FL) spectrophotometric procedure. The process was conveniently studied using FL, since the triphenylimidazole‐derived ester 1 and its reaction products (the corresponding phenol 2 and phenolate 2 ? ) are all highly fluorescent (ΦFL > 37%). By carefully selecting excitation and emission wavelengths, observed rate constants k1 in the order of 10?3 to 10?2 s?1 were obtained from either reactant consumption (λex = 300 nm, λem = 400 nm) or product formation (λex = 350 nm, λem = 475 nm); these were shown to be kinetically equivalent. Intensity‐decay time profiles also gave a residual FL intensity parameter, shown to be associated to the distribution of produced species 2 and 2 ? , according to the basicity of the medium. Studying the reaction in both methanol (MeOH) and isopropanol (iPrOH), upon addition of HO?, provided evidence that the solvent's conjugate base is the active nucleophilic species. When different bases were used (tBuO?, HO?, DBU and TEA), bimolecular rate constants kbim ranging from 4.5 to 6.5 L mol?1 s?1 were obtained, which proved to be non‐dependent on the base pKaH, suggesting specific base catalysis for the decomposition of 1 in alcoholic media.  相似文献

Randia echinocarpa, an endemic plant to Northwest Mexico, is used as food and in traditional medicine, and several of its biological activities have been demonstrated (antioxidant, antimutagenic, antidiabetic, and immunomodulatory). Plant tissue culture is a safe and scalable system for plant propagation and production of bioactive compounds. Therefore, this study aims to establish protocols for seed germination and callus culture of R. echinocarpa and to evaluate the antioxidant activity of methanol extracts (ME) of plantlets and calli via the 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) methods. Seeds were cultured in media with different concentrations of Murashige and Skoog (MS) salts and sucrose, and a higher germination rate and plantlet growth was observed in half-strength MS medium with 15 g L−1 of sucrose. Calli were obtained from cotyledon and hypocotyl explants cultured in MS media with different concentrations of benzyl aminopurine (BAP) and indole-3-acetic acid (IAA). All treatments induced callus formation in 100% of explants; however, the medium containing 1 mg L−1 BAP + 1 mg L−1 IAA was selected because it produced calli with higher biomass and friable texture. The ME of cotyledons showed the highest antioxidant activity values (μmol Trolox per 100 g dry weight) in DPPH (345.5) and ABTS (1166.4) assays, whereas the ME of calli from hypocotyls showed a higher antioxidant activity than the ME of calli from cotyledons in both antioxidant assays. The tissue culture protocols established here will be useful for R. echinocarpa germplasm conservation and propagation, as well as for the production of bioactive compounds.

The biomass production and biochemical properties of marine and freshwater species of green macroalgae (multicellular algae), cultivated in outdoor conditions, were evaluated to assess the potential conversion into high-energy liquid biofuels, specifically biocrude and biodiesel and the value of these products. Biomass productivities were typically two times higher for marine macroalgae (8.5–11.9 g m−2 d−1, dry weight) than for freshwater macroalgae (3.4–5.1 g m−2 d−1, dry weight). The biochemical compositions of the species were also distinct, with higher ash content (25.5–36.6%) in marine macroalgae and higher calorific value (15.8–16.4 MJ kg−1) in freshwater macroalgae. Lipid content was highest for freshwater Oedogonium and marine Derbesia. Lipids are a critical organic component for biocrude production by hydrothermal liquefaction (HTL) and the theoretical biocrude yield was therefore highest for Oedogonium (17.7%, dry weight) and Derbesia (16.2%, dry weight). Theoretical biocrude yields were also higher than biodiesel yields for all species due to the conversion of the whole organic component of biomass, including the predominant carbohydrate fraction. However, all marine species had higher biomass productivities and therefore had higher projected biocrude productivities than freshwater species, up to 7.1 t of biocrude ha−1 yr−1 for Derbesia. The projected value of the six macroalgae was increased by 45–77% (up to US$7700 ha−1 yr−1) through the extraction of protein prior to the conversion of the residual biomass to biocrude. This study highlights the importance of optimizing biomass productivities for high-energy fuels and targeting additional coproducts to increase value.  相似文献

Nodulation of Alnus rubra seedlings after inoculation with soil from under A. rubra, Betula papyrifera. Rubus lacianutus, R. spectabilis, and R.ursinus on 2 recently harvested sites was compared. Nodulation capacity was low compared to other published reports, ranging from 0 to 18.9 infective units cm-3 of soil and was significantly affected by the site and plant species. Nodulation capacity of soil under alder was significantly higher than under all other species except R. spectabilis, regardless of site. There has been an increasing drive toward better valorising raw biological materials in the context of the sustainability of bio-based industries and the circular economy. As such, microalgae hold the ability to biosynthesise valuable metabolites, which are sought after within the bioenergy, pharmaceuticals, cosmetics or nutrition sectors. Owing to their bioactivities, the xanthophyll pigment fucoxanthin and the omega-3 polyunsaturated fatty acid eicosapentaenoic acid (EPA) have fostered increasing interests in terms of sustainably refining them from natural sources, such as microalgae. Together with the suitability of individual species to industrial cultivation, a key challenge resides in optimizing the yields of these compounds within the microalgal biomass they are retrieved from. The marine diatom Stauroneis sp. LACW24 was batch cultivated into its stationary phase of growth prior to being subjected at high cell density (1 × 106 cells mL−1) to seven different regimes of light exposure in replenished medium and under nutritional limitation (silica and nitrate) for 12 days. The highest EPA proportions and yields were obtained under blue LED in f/2 medium (16.5% and 4.8 mg g−1, respectively), double the values obtained under red LED illumination. The fucoxanthin yield was the highest when cells were subjected to blue LEDs (5.9 mg g−1), a fourfold increase compared to the nitrogen-limited treatment under white LEDs. These results indicate that a two-stage approach to the batch cultivation of this diatom can be used for enhancing the production of the high-value metabolites fucoxanthin and EPA post-stationary phase.  相似文献

Determining the abundance of N isotope (δ15N) in natural environments is a simple but powerful method for providing integrated information on the N cycling dynamics and status in an ecosystem under exogenous N inputs. However, whether the input of different N compounds could differently impact plant growth and their 15N signatures remains unclear. Here, the response of 15N signatures and growth of three dominant plants (Leymus chinensis, Carex duriuscula, and Thermopsis lanceolata) to the addition of three N compounds (NH4HCO3, urea, and NH4NO3) at multiple N addition rates were assessed in a meadow steppe in Inner Mongolia. The three plants showed different initial foliar δ15N values because of differences in their N acquisition strategies. Particularly, T. lanceolata (N2-fixing species) showed significantly lower 15N signatures than L. chinensis (associated with arbuscular mycorrhizal fungi [AMF]) and C. duriuscula (associated with AMF). Moreover, the foliar δ15N of all three species increased with increasing N addition rates, with a sharp increase above an N addition rate of ~10 g N m−2 year−1. Foliar δ15N values were significantly higher when NH4HCO3 and urea were added than when NH4NO3 was added, suggesting that adding weakly acidifying N compounds could result in a more open N cycle. Overall, our results imply that assessing the N transformation processes in the context of increasing global N deposition necessitates the consideration of N deposition rates, forms of the deposited N compounds, and N utilization strategies of the co-existing plant species in the ecosystem.  相似文献

Pfaffia glomerata has potential pharmacological and medicinal properties due to the production of a secondary metabolite known as the phytoecdysteroid 20-hydroxyecdysone (20E). There have been increasing efforts for massive in vitro propagation of Pfaffia plants due to high extractivism and overharvesting of this species. Research on the species has shown that photoautotrophic cultivation can improve the production of 20E. In addition, other abiotic factors such as the formulations of culture media can influence the morphophysiological behavior of the plants in vitro. Therefore, the objective of this study was to analyze the morphological and physiological performances of P. glomerata plants in different formulations of culture media, under photoautotrophic and photomixotrophic propagation conditions. Six medium formulations, the Driver and Kuniyuki medium (DKW), Correia et al. medium (JADS), Murashige and Skoog medium (MS), Quoirin and Lepoivre medium (QL), Rugini medium (OM), and Woody Plant medium (WPM), all supplemented with DKW vitamins, 100 mg L−1 myo-inositol, 6.5 g L−1 agar, and with or without 3% (w/v) sucrose, were evaluated. Cultures were maintained at 25 ± 2°C, with a 16 h-photoperiod under 60 μmol m−2 s−1 of irradiance under a fluorescent lamp for 50 d. Results showed that the presence or absence of sucrose, and the different nutritional formulations influenced growth, photosynthetic pigment content, endogenous levels of sugars, leaf morphology, levels of 20E, and transport of water and minerals in P. glomerata. Notably, OM, DKW, QL, and WPM media promoted higher production of 20E under photomixotrophic growth conditions.

Marine toxic dinoflagellates of the genus Gambierdiscus are the causative agents of ciguatera fish poisoning (CFP), a form of seafood poisoning that is widespread in tropical, subtropical and temperate regions worldwide. The distributions of Gambierdiscus australes, Gambierdiscus scabrosus and two phylotypes of Gambierdiscus spp. type 2 and type 3 have been reported for the waters surrounding the main island of Japan. To explore the bloom dynamics and the vertical distribution of these Japanese species and phylotypes of Gambierdiscus, the effects of light intensity on their growth were tested, using a photoirradiation-culture system. The relationship between the observed growth rates and light intensity conditions for the four species/phylotypes were formulated at R > 0.92 (p < 0.01) using regression analysis and photosynthesis-light intensity (P-L) model. Based on this equation, the optimum light intensity (Lmax) and the semi-optimum light intensity range (Ls-opt) that resulted in the maximum growth rate (μmax) and ≥80% μ max values of the four species/phylotypes, respectively, were as follows: (1) the Lmax and Ls-opt of G. australes were 208 μmol photons m−2 s−1 and 91–422 μmol photons m−2 s−1, respectively; (2) those of G. scabrosus were 252 and 120–421 μmol photons m−2 s−1, respectively; (3) those of Gambierdiscus sp. type 2 were 192 and 75–430 μmol photons m−2 s−1, respectively; and (4) those of Gambierdiscus sp. type 3 were ≥427 and 73–427 μmol photons m−2 s−1, respectively. All four Gambierdiscus species/phylotypes required approximately 10 μmol photons m−2 s−1 to maintain growth. The light intensities in coastal waters at a site in Tosa Bay were measured vertically at 1 m intervals once per season. The relationships between the observed light intensity and depth were formulated using Beer’s Law. Based on these equations, the range of the attenuation coefficients at Tosa Bay site was determined to be 0.058–0.119 m−1. The values 1700 μmol photons m−2 s−1, 500 μmol photons m−2 s−1, and 200 μmol photons m−2 s−1 were substituted into the equations to estimate the vertical profiles of light intensity at sunny midday, cloudy midday and rainy midday, respectively. Based on the regression equations coupled with the empirically determined attenuation coefficients for each of the four seasons, the ranges of the projected depths of Lmax and Ls-opt for the four Gambierdiscus species/phylotypes under sunny midday conditions, cloudy midday conditions, and rainy midday conditions were 12–38 m and 12–54 m, 1–16 m and 1–33 m, and 0 m and 0–16 m, respectively. These results suggest that light intensity plays an important role in the bloom dynamics and vertical distribution of Gambierdiscus species/phylotypes in Japanese coastal waters.  相似文献

The age, growth and reproductive biology of Marcusenius pongolensis, Oreochromis mossambicus and Schilbe intermedius were investigated in the Mnjoli Dam, Swaziland. Otolith annulus formation occurred in winter for M. pongolensis, and in spring/summer for O. mossambicus and S. intermedius. Maximum ages of 8, 6 and 8 years were recorded for M. pongolensis, O. mossambicus and S. intermedius, respectively. Growth was described by the von Bertalanffy growth model as Lt = 238.73(1 − exp−0.27(t+2.27) mm fork length (FL) for M. pongolensis, Lt = 226.83(1 − exp−0.45(t+2.02)) mm total length (TL) for O. mossambicus, and Lt = 214.59(1 − exp−0.60(t+1.20)) mm FL for S. intermedius. Sexual maturity was estimated for male and female M. pongolensis at 134 mm FL and 119 mm FL, respectively. Marcusenius pongolensis matured within their first year. Female O. mossambicus and S. intermedius matured at 239 mm TL and 205 mm FL, corresponding to 2 and 4 years of age, respectively. Extended spawning periods, with two spawning peaks was observed for M. pongolensis, one in spring (September) and the second in autumn (March) and one peak over late‐summer for S. intermedius.  相似文献

An efficient in vitro propagation system has been developed for the rapid micropropagation of Thymus serpyllum L. (Banajwain), an aromatic medicinal herb from nodal explant on MS medium. Phenolic leaching and high rate of contamination was the most significant problem in establishing in vitro culture of Thymus serpyllum which was overcome by preparing explants in an antioxidant ascorbic acid (1000 ppm) at 6°C for 45 min and addition of the same antioxidant (50 mgl−1) to the MS medium. The frequency of shoot production was influenced by different cytokinins (Kn, BAP, and Kn + BAP) and 95.56% shoot induction was observed when MS medium was supplemented with 1.0 + 2.0 mgl−1 (Kn + BAP). The maximum average number of shoots 16.93 ± 2.15 and average length (3.98 ± 0.55) was recorded when MS medium have 0.5 + 2.0 mgl−1 (Kn + BAP). The in vitro regenerated microshoots were rooted on MS and half strength MS medium and there was significant difference in root induction on both media under the influence of auxins (IAA, IBA, and NAA). The maximum average number (11.67 ± 3.03) and average root length (3.88 ± 0.71) was reported in half MS medium having 1.0 mgl−1 IBA. The complete regenerated plantlets were acclimatized under growth chamber before transferring to the earthen pots and showed 90% survival.
