Characterization of 42 highly polymorphic bovine microsatellite markers   总被引:1,自引:0,他引:1  
We have isolated 42 highly polymorphic microsatellite (GT/CA)n markers from Japanese black cattle Wagyu ( Bos taurus ). Forty-one of the markers were assigned to bovine autosomes with lod scores > 6, through linkage analyses performed on the International Bovine Reference Family Panel (IBRP). The remaining marker showed X-linked inheritance. These markers exhibited an average heterozygosity value of 0.67 with between four and 17 alleles on the IBRP.  相似文献

Turkey microsatellite DNA loci amplified by chicken-specific primers   总被引:11,自引:0,他引:11  
Forty-eight primer-pairs complementary to unique DNA sequences flanking chicken (genus Gallus ) genomic (TG)n microsatellite repeats were previously designed. These primer-pairs were used in the polymerase chain reaction to amplify turkey (genus Meleagris ) genomic DNA loci. Results indicated that the majority (92%) of these primer-pairs generated amplification products in turkey genomic DNA. Hybridization using end-labelled (TG)8 as a probe showed that, out of 41 primer-pairs tested, only 14 generated an amplification product that also contained a detectable (TG)n microsatellite repeat when turkey DNA was the template. Among 18 primerpairs tested for polymorphism, using three commercial turkey lines, five were found to exhibit length polymorphism, three of which did not contain a detectable TG repeat. Therefore, a significant portion of chicken microsatellite markers can be useful for genomic mapping and linkage analysis in the turkey, reducing the costs involved in producing turkey-specific microsatellite markers.  相似文献

One of the most widely studied simple sequences in the mammalian genome is the (TG)n dinucleotide sequence. Because these microsatellites are highly polymorphic, we chose to study microsatellites from cosmids to provide genetic markers for the porcine genome. After screening a porcine cosmid library with a (CA)10 probe, 20 cosmids containing microsatellites were subcloned and 17 microsatellites identified by sequencing. Oligonucleotide primers flanking the repeat were designed for seven (TG)n microsatellites with n > 14. These seven microsatellites revealed polymorphism and were regionally assigned to chromosomes by fluorescent in situ hybridization of initial cosmids. These seven loci will be useful for both the construction of the genetic map and as landmark loci on the physical map of the porcine genome.  相似文献

An Atlantic herring major histocompatibility class II A ( Clha-DAA ) cDNA sequence has been characterized and was shown to encode a leader peptide, alpha-1 domain, alpha-2 domain, connecting peptide, transmembrane and cytoplasmic region. The Clha-DAA protein sequence has all the characteristics of a teleost class II A protein with conserved cysteines in both the alpha-1 and the alpha-2 domains and two potential N-linked glycosylation sites. Exon 2 sequences encoding the polymorphic alpha-1 domain from different individuals were analysed and revealed the presence of at least two loci. The Clha-DAA gene consists of four exons and three short introns. Four unique intron 3 sequences from multiple individuals were obtained and were shown to contain a (TG)n microsatellite sequence. Primers were optimized such that only a single microsatellite locus designated Clha-DAA-INTR3 was amplified. Four herring populations from the North Sea and the Baltic Sea were genotyped for Clha-DAA-INTR3 . In total, 16 Clha-DAA-INTR3 alleles were detected; the distribution of the alleles showed no deviation from Hardy–Weinberg expectation. Levels of genetic differentiation among samples were of similar magnitude as have been reported earlier for neutral microsatellite loci between northern North Sea and Baltic Sea herring populations.  相似文献

Evidence for the presence of the metal-binding protein metallothionein, MT, in higher plants is equivocal. Although a number of MT-like metal complexes have been isolated from plants, the chemical structures of most of these compounds have not been fully elucidated. Recently a novel class of plant peptides, poly (γ-glutamylcysteinyl) glycines, (γEC)nG, have been discovered. These peptides bind metal ions, and in the presence of such ions the amount of (γEC), G in plant cells increases. The presence of peptide bonds through the γ-carboxyl group of glutamate, rather than the α-carboxyl group, suggests that these peptides are not encoded by structural genes but are the products of biosynthetic pathways. Cells which are resistant to supra-optimal concentrations of certain metal ions over-produce (γEC)n G. (γEC)n G. may be functional analogues of MT. Whether or not some plants also produce MT is an important question which remains to be answered.  相似文献

Development and mapping of ten porcine microsatellite markers   总被引:7,自引:0,他引:7  
Thirty (TG)n microsatellite clones were isolated from a pig genomic library, sequenced, and tested for their suitability to detect polymorphism on a panel of animals by means of the polymerase chain reaction. Ten of these clones were developed into suitable markers and subsequently segregation of these markers was determined in the five PiGMaP reference pedigrees. A linkage analysis was performed on these 10 microsatellites together with 365 other loci that have been typed on these reference families. Eight of the microsatellites have been mapped to eight different linkage groups that have been previously assigned to different chromosomes (chromosomes 1, 6, 7, 9, 14, 15, 17 and 18). Of the remaining two markers, one is X-linked and the other shows no linkage. The number of alleles detected by these microsatellites, in the reference pedigrees, varied from six to sixteen and the heterozygosity varied from 42 to 85% in the 26 unrelated founder animals of these reference pedigrees.  相似文献

Needle water potential at noon and diurnal variation in needle water potential was measured with a pressure chamber during the growth seasons (1974, 1975 and 1976) in a 20-year-old stand of Scots pine (Pinus sylvestris L.), subjected to daily irrigation and nutrient treatments at Jädraås, in central Sweden (60°50'N).
In unstressed conditions there was little difference in water potential amongst the treatments. However, during a dry spell in 1976 the diurnal curves of water potential in the control and combined irrigated and fertilized treatment diverged by up to 6 bar in the middle of the day. Irrigation alone resulted in higher midday water potentials (Ψn) than in the controls only during this period. At other times in 1974, 1975 and early 1976, the values of Ψn in the irrigated treatment were similar to the controls. Fertilization alone resulted in higher Ψn than in the controls in both 1975 and 1976 during both wet and dry periods. However, the largest difference between treatment and control occurred in the combined, irrigated and fertilized treatment during the dry spell in 1976.
Several possible explanations for the effect of fertilization on leaf water potential are discussed, including changes in hydraulic conductivity and water storage. The most likely explanation is a reduced transpiration rate as a result of more effective stomatal control of water loss.  相似文献

A cosmid library has been constructed from a sheep x hamster cell hybrid containing sheep chromosome t1, rob (6;24). Clones containing sheep DNA were identified by hybridizing to a total sheep genomic DNA probe. Small fragments (<500 bp) containing (AC)n microsatellites were subcloned and sequenced. Ten microsatellite markers were characterized and six were mapped back to chromosomes 6 and 24. The remaining microsatellites mapped to chromosome 26, which was shown to be present in a small proportion of cells of the cell line.  相似文献

The effects of sodium fluoride (0.3, 5 and 10 m M NaF) on root hydraulic conductivity, and gas exchange processes were examined in aspen ( Populus tremuloides Michx.) seedlings grown in solution culture. A long-term exposure of roots to NaF significantly decreased root hydraulic conductivity ( L p) and stomatal conductance ( g s). Root absorbed NaF significantly affected electrolyte leakage in leaf tissues and substantially restricted leaf expansion. NaF did not significantly affect leaf chlorophyll contents but decreased net photosynthesis ( P n). A short-term exposure of excised roots to 5 m M NaF and KF significantly decreased root water flow ( Q v) with a concomitant decline in root respiration and reduced g s when applied through intact roots or excised stems. The same molar concentration of NaCl also decreased Q v and g s in intact seedlings, but to a lesser extent than NaF or KF, and did not significantly affect root respiration. The results suggest that fluoride metabolically inhibited Q v or L p, probably by affecting water channel activity. We suggest that the metabolic inhibition of L p by root-absorbed fluoride affected gas exchange and leaf expansion in aspen seedlings.  相似文献

Abstract— Glycine sensitive [3H]strychnine binding has been demonstrated in the pigeon CNS. The kinetic parameters in spinal cord, tectum, telencephalon and cerebellum in this order were: K n, (nM) 5 , 5, 2.3, 6 and B max (fmol/mg protein) 1000, 275, 92, 20. The effect of various amino acids and strychnine analogs on the binding was measured. Retinal ablation caused a 22% decrease in the glycine sensitive [3H]strychnine binding in the tectum while lesions destroying up to 80% of the nucleus Ipc had no effect.  相似文献

Using a laboratory-constructed system that can measure the gas exchange rates of two leaf surfaces separately, the light responses of the adaxial and abaxial stomata in intact leaves of sunflower ( Helianthus annuus L.) were investigated, keeping the intercellular CO2 concentration ( C i) at 300  µ L L−1. When evenly illuminating both sides of the leaf, the stomatal conductance ( g s) of the abaxial surface was higher than that of the adaxial surface at any light intensity. When each surface of the leaf was illuminated separately, both the adaxial and abaxial stomata were more sensitive to the light transmitted through the leaf (self-transmitted light) than to direct illumination. Relationships between the whole leaf photosynthetic rate ( A n) and the g s for each side highlighted a strong dependence of stomatal opening on mesophyll photosynthesis. Light transmitted through another leaf was more effective than the direct white light for the abaxial stomata, but not for the adaxial stomata. Moreover, green monochromatic light induced an opening of the abaxial stomata, but not of the adaxial stomata. As the proportion of blue light in the transmitted light is less than that in the white light, there may be some uncharacterized light responses, which are responsible for the opening of the abaxial stomata by the transmitted, green light.  相似文献

Summary. Two-dimensional agarose gel (pH 8.6)-horizontal polyacrylamide gel (pH 9.0) electrophoresis of horse serum samples, followed by general protein staining, revealed genetic polymorphism of an unidentified protein tentatively designated serum protein 3 (SP3). The SP3 fractions appeared distinctly when a 14% concentration of acrylamide was used in the separation gels. The 2-D mobilities of SP3 fractions were quite similar to that of albumin. Family data were consistent with the hypothesis that the observed SP3 phenotypes were controlled by four co-dominant, autosomal alleles ( D,F,I,S ). Evidence was provided that the F allele can be further divided into two alleles ( F 1 and F 2); the mobilities of F1 and F2 variants were very similar. Each of the SP3 alleles gave rise to one fraction and each of the heterozygous types showed two fractions. More than 600 horses representing five different breeds (Swedish Trotter, North-Swedish Trotter, Thoroughbred, Arab and Polish Tarpan) were typed for SP3, and allele frequency estimates were calculated. Artificial chalk grassland swards were exposed to either ambient air or air enriched to 600 μ mol mol–1 CO2, using free-air CO2 enrichment technology, and subjected to an 8 week simulated grazing regime. After 14 months of treatment, ribulose-1,5-bisphosphate carboxylase (Rubisco) activity ( V c,max) and electron transport mediated ribulose-1,5-bisphosphate (RuBP) regeneration capacity ( J max), estimated from leaf gas exchange, were significantly lower in fully expanded leaves of Anthyllis vulneraria L. (a legume) and Sanguisorba minor Scop. grown in elevated CO2. After a change in source:sink balance brought about by defoliation, photosynthetic capacity was fully restored in A. vulneraria and S. minor, but acclimation continued in the grass Bromopsis erecta (Hudson) Fourr. Changes in net photosynthesis ( P n) with growth at elevated CO2 ranged from a 1·6% reduction in precut leaves of A. vulneraria to a 47·1% stimulation in postcut leaves of S. minor . Stomatal acclimation was observed in leaves of A. vulneraria (reduced stomatal density) and B. erecta (reduced stomatal conductance). The results are discussed in terms of whole-plant resource-use optimization and chalk grassland community competitive interactions at elevated CO2.  相似文献

Cattle and water buffalo sera labelled with vitamin D3[14C] (300 and 480 individual samples respectively) were subjected to starch gel electrophoresis followed by autoradiography in an attempt to identify a possible polymorphism of the proteins capable of binding this vitamin.

The temporal and spatial population genetic structure of ayu Plecoglossus altivelis (Salmoniformes: Plecoglossidae), an amphidromous fish, was examined using analysis of variation at six microsatellite DNA loci. Intracohort genetic diversities, as measured by the number of alleles and heterozygosity, were similar among six cohorts (2001–2006) within a population (Nezugaseki River), with the mean number of alleles per cohort ranging from 11·0 to 12·5 and the expected heterozygosity ranging from 0·74 to 0·77. Intrapopulational genetic diversities were also similar across the three studied populations along the 50 km coast, with the mean number of alleles and the expected heterozygosity ranging from 11·33 to 11·67 and from 0·75 to 0·76, respectively. The authors observed only one significant difference in pair-wise population differentiation ( F ST-value) between the cohorts within a population and among three populations. Estimates of the effective population size ( N e) based on maximum-likelihood method yielded small values (ranging from 94·8 to 135·5), whereas census population size ranged from c. 4800 to 24 000. As a result, the ratio of annual effective population sizes to census population size ( N e/ N ) ranged from 0·004 to 0·023. These estimates of N e/ N agree more closely with estimates for marine fishes than that of the larger estimates for freshwater fishes. The present study suggests that ayu which is highly fecund and shows low survival during the early life stages is also characterized by having low value of N e/ N , similar to marine species with a pelagic life cycle.  相似文献

Supernatant malate dehydrogenase (MDH) isozymes (as visualized by starch gel electrophoresis) are encoded by two distinct gene loci in both the largemouth and smallmouth bass. When an interspecific F1 hybrid is formed between these two fish, a unique MDH isozyme is generated. The results of freeze-thaw molecular hybridization (which is the first application of this technique to MDH) indicate that this unique isozyme in the F1 hybrid is a heterodimer composed of one subunit of each parental type. The F1 hybrids produced F2 hybrids which in turn formed the F3 hybrid population. The inheritance of alleles at the MDH-B locus is consistent with a single Mendelian autosomal locus. Furthermore, there is no evidence of linkage between the lactate dehydrogenase-E locus and the MDH-B locus.  相似文献

A new allele at the Lh locus has been identified in Pisum sativum L. and named lhi . This allele results in reduced GA levels in young shoots, and a dwarf phenotype. Gas chromatography-selected ion monitoring (GC-SIM) with dideuterated internal standards has been used to demonstrate a quantitative relationship between the level of endogenous GA1 and internode length using the three alleles ( Lh, lh and lhi ) at the Lh locus. These results are consistent with previous findings in peas (for alleles at the Le locus) and other species possessing a predominant early 13-hydroxylation pathway for GA biosynthesis and support the role of GA1 as the major native GA in peas with biological activity in its own right. However, in contrast to alleles at the Le locus, GA20 levels are also reduced in lh and lhi plants. The lhi allele also has possible pleiotropic effects on seed abortion, leading to a reduction in seed yield compared to plants homozygous for the previously characterised Lh or lh alleles.  相似文献

Ozone effects on plant water relations have been reported to be similar to those of water-deficit. The objective was to identify ozone-inducible (OI) clones from Atriplex canescens (saltbush) and determine if they were also responsive to water-deficit as well as SO2. cDNA clones derived from four different polyA RNAs which accumulate in 8-month-old shrub leaves exposed to ozone (0.2 μl I−1, 6 h day−1, 7 days) were isolated by differential screening, analyzed by northern blots, sequenced, and gene product homologies with other plant genes were determined. Clone OI12A-3 has homology with wound-inducible proteinase inhibitors, whereas clone OI8–3 protein is homologous to thiol proteases. Clones OI2–2 and OI14–3 putatively code for glycine-rich proteins with repeated motifs (Gly-Gly-Gly-Tyr-Gly-His)n and putative cell-wall-targeting signal peptides. Clone OI2–2 and particularly clone OI14–3 were also induced by both SO2 and water-deficit. These data indicate that woody plant genes associated with cell wall protein production and whose expression is induced by several stress factors may be responding to common oxidative stress pathways.  相似文献
