A method of measuring CO2gas exchange (caused, for example, by microalgal photosynthesis on emersed tidal mudflats) using open flow IR gas analyzers is described. The analyzers are integrated in a conventional portable photosynthesis system (LI-6400, LI-COR, Nebraska, USA), which allows manipulation and automatic recording of environmental parameters at the field site. Special bottomless measuring chambers are placed directly on the surface sediment. Measurements are performed under natural light conditions and ambient CO2concentrations, as well as under different CO2concentrations in air, and various PAR radiation levels produced by a LED light source built into one of the measurement chambers. First results from tidal channel banks in a north Brazilian mangrove system at Bragança (Pará, Brazil) under controlled conditions show a marked response of CO2assimilation to CO2concentration and to irradiance. Photosynthesis at 100molmol–1CO2in air in one sample of a well-developed algal mat was saturated at 309mol photons m–2s–1, but increased with increasing ambient CO2concentrations (350 and 1000mol mol–1CO2) in the measuring chamber. Net CO2assimilation was 0.8mol CO2m–2s–1at 100mol mol–1CO2, 5.9mol CO2m–2s–1at 350mol mol–1CO2and 9.8mol CO2m–2s–1at 1000mol mol–1CO2. Compensation irradiance decreased and apparent photon yield increased with ambient CO2concentration. Measurements under natural conditions resulted in a quick response of CO2exchange rates when light conditions changed. We recommend the measuring system for rapid estimations of benthic primary production and as a valuable field research tool in connection with certain ecophysiological aspects under changing environmental conditions.  相似文献

Dry weight and Relative Growth Rate of Lemna gibba were significantly increased by CO2 enrichment up to 6000 l CO2 l–1. This high CO2 optimum for growth is probably due to the presence of nonfunctional stomata. The response to high CO2 was less or absent following four days growth in 2% O2. The Leaf Area Ratio decreased in response to CO2 enrichment as a result of an increase in dry weight per frond. Photosynthetic rate was increased by CO2 enrichment up to 1500 l CO2 l–1 during measurement, showing only small increases with further CO2 enrichment up to 5000 l CO2 l–1 at a photon flux density of 210 mol m–2 s–1 and small decreases at 2000 mol m–1 s–1. The actual rate of photosynthesis of those plants cultivated at high CO2 levels, however, was less than the air grown plants. The response of photosynthesis to O2 indicated that the enhancement of growth and photosynthesis by CO2 enrichment was a result of decreased photorespiration. Plants cultivated in low O2 produced abnormal morphological features and after a short time showed a reduction in growth.  相似文献

This study reports survival and physiological responses of micropropagated Ceratonia siliqua L. cvs. Galhosa and Mulata plants during ex vitro acclimatization under ambient (AC; 330 mol mol–1) or elevated (EC; 810 mol mol–1) CO2 concentration and a photosynthetic photon flux density of 125 mol m–2 s–1. CO2 enrichment during acclimatization did not improve survival rate that was around 80 % for both treatments. Eight weeks after ex vitro transplantation, photosynthetic capacity and apparent quantum yield in acclimatized leaves were higher in comparison with those in in vitro-grown leaves, without any significant difference between CO2 treatments. Chlorophyll content increased after acclimatization. However, EC led to a decrease in the total amount of chlorophyll in new leaves of both cultivars, compared to those grown at AC. Soluble sugars and starch contents were not markedly affected by growth EC, although starch had significantly increased after transfer to ex vitro conditions. EC induced an increase in the stem elongation and in the effective life of leaves, and a decrease in the number of new leaves.  相似文献

Ponderosa pine (Pinus ponderosa Dougl. ex Laws.) seedlings were exposed to near ambient or elevated CO2 (average concentrations during the last growing season 446 versus 699 mol mol–1), combined with low or elevated O3 for three seasons. Ozone exposure during the last growing season (accumulated dose above threshold 0.06 mol mol–1) was 0.05 versus 26.13 mol mol–1 h. Needles of the youngest age class were harvested after the dormancy period. Ozone exposure decreased needle contents of chlorophyll a, chlorophyll b, and ascorbate, and resulted in a more oxidized total ascorbate and a more de-epoxidized xanthophyll cycle pool irrespective of the CO2 level. Trees under elevated CO2 had a more oxidized glutathione pool and lower chlorophyll a content. Contents of glutathione, tocopherol, and carotenoids were not affected by the CO2 or O3 treatments. There were no interactive effects between elevated CO2 and elevated O3 on any of the parameters measured. The results suggest that elevated atmospheric CO2 concentration does not compensate for ozone stress by increasing antioxidative capacity in ponderosa pine.  相似文献

Short-term responses of four carrot (Daucus carota) cultivars: Cascade, Caro Choice (CC), Oranza, and Red Core Chantenay (RCC) to CO2 concentrations (C a) were studied in a controlled environment. Leaf net photosynthetic rate (P N), intercellular CO2 (C i), stomatal conductance (g s), and transpiration rate (E) were measured at C a from 50 to 1 050 mol mol–1. The cultivars responded similarly to C a and did not differ in all the variables measured. The P N increased with C a until saturation at 650 mol mol–1 (C i= 350–400 mol mol–1), thereafter P N increased slightly. On average, increasing C a from 350 to 650 and from 350 to 1 050 mol mol–1 increased P N by 43 and 52 %, respectively. The P N vs. C i curves were fitted to a non-rectangular hyperbola model. The cultivars did not differ in the parameters estimated from the model. Carboxylation efficiencies ranged from 68 to 91 mol m–2 s–1 and maximum P N were 15.50, 13.52, 13.31, and 14.96 mol m–2 s–1 for Cascade, CC, Oranza, and RCC, respectively. Dark respiration rate varied from 2.80 mol m–2 s–1 for Oranza to 3.96 mol m–2 s–1 for Cascade and the CO2 compensation concentration was between 42 and 46 mol mol–1. The g s and E increased to a peak at C a= 350 mol mol–1 and then decreased by 17 and 15 %, respectively when C a was increased to 650 mol mol–1. An increase from 350 to 1 050 mol mol–1 reduced g s and E by 53 and 47 %, respectively. Changes in g s and P N maintained the C i:C a ratio. The water use efficiency increased linearly with C a due to increases in P N in addition to the decline in E at high C a. Hence CO2 enrichment increases P N and decreases g s, and can improve carrot productivity and water conservation.  相似文献

Isolated embryos ofKarwinskia humboldtiana were cultured in vitro. The growth of embryos and development to plantlets on woody plant medium supplemented with indole-3-acetic acid 6.10-2 mol l–1, gibberellic acid (GA3) 3.10-2 mol l–1, and 6-benzylaminopurine (BA) 2 mol l–1 was obtained. Multiplication of shoots and rooting of excised shoots has been achieved. In order to maintain axenic seedstock cultures axenically of thecommercially important red seaweed, Porphyra yezoensis, aprocedure was developed for axenic isolation and culture of conchocelis andmonospores. For axenic isolation of the conchocelis, contaminated microalgaewere most effectively removed by filtering contaminated samples through a100-m mesh after sonication. Removal of bacteria and otheralgaewas accomplished using a mixture of 5 agents (0.02% chitosan, 100 gml–1 GeO2, 10 gml–1 ampicillin, 40 gml–1 kanamycin and 200 gml–1 streptomycin). Axenic single colonies wereisolatedfrom a semi-solid medium prepared from 1% transfer gel. After collectingmonospores from the 40–50% density layer on a percoll-gradient, removalofbacteria and fungi from the monospores was accomplished using a mixture of 5antibiotics (3.5 g ml–1 nystatin, 2 mgml–1 ampicillin, 400 gml–1 kanamycin, 50 gml–1 neomycin and 800 gml–1 streptomycin). The eddy correlation technique was employed to measure net ecosystem carbon dioxide (CO2) (NEE) and water vapor exchange (LE) over a C3/C4 co-occurring wet temperate Miscanthus-type grassland in the Kanto plain of Japan in the 1999 growing season. The maximal mean canopy height and maximal leaf area index were 1.0m and 5.5, respectively. The daily maximal LE was approximately 540Wm–2. The maximum value of daily accumulative LE was 16.3MJday–1. Daily variation of the decoupling factor () suggests that in the morning LE decoupled with the atmosphere, and the available energy was the major driving force for LE, whereas in the afternoon LE coupled strongly with the atmosphere, and the atmospheric evaporative demand played a critical role in LE. The decline in (from 0.8 to 0.5) with the growing season demonstrates that LE decoupled from the atmosphere in the later growth season. The peak NEE value was 57.4µmolCO2m–2s–1 (the positive value signifies the canopy carbon gain was from the air). The maximal daily integrated NEE was 1.06molCO2m–2day–1 observed during the peak growth stage. A rectangular hyperbolic model was used to describe the relation between daytime NEE and incident photosynthetic photon flux density (PPFD). The net ecosystem CO2 was not light-saturated up to a PPFD level of 2000µmol m–2s–1. The initial slope estimated with the NEE–PPFD response model was approximately 0.042molCO2mol–1photon on average. The canopy light compensation point ranged from 210 to 430µmolm–2s–1 with an average of approximately 310µmolm–2s–1. Both the initial slope and the canopy light compensation point decreased as the canopy senesced. The switch in dominance from C3 to C4 plants played an important role in the canopy fluxes.  相似文献

Physiological responses to elevated CO2 at the leaf and canopy-level were studied in an intact pine (Pinus taeda) forest ecosystem exposed to elevated CO2 using a free-air CO2 enrichment (FACE) technique. Normalized canopy water-use of trees exposed to elevated CO2 over an 8-day exposure period was similar to that of trees exposed to current ambient CO2 under sunny conditions. During a portion of the exposure period when sky conditions were cloudy, CO2-exposed trees showed minor (7%) but significant reductions in relative sap flux density compared to trees under ambient CO2 conditions. Short-term (minutes) direct stomatal responses to elevated CO2 were also relatively weak (5% reduction in stomatal aperture in response to high CO2 concentrations). We observed no evidence of adjustment in stomatal conductance in foliage grown under elevated CO2 for nearly 80 days compared to foliage grown under current ambient CO2, so intrinsic leaf water-use efficiency at elevated CO2 was enhanced primarily by direct responses of photosynthesis to CO2. We did not detect statistical differences in parameters from photosynthetic responses to intercellular CO2 (A net-C i curves) for Pinus taeda foliage grown under elevated CO2 (550 mol mol–1) for 50–80 days compared to those for foliage grown under current ambient CO2 from similar-sized reference trees nearby. In both cases, leaf net photosynthetic rate at 550 mol mol–1 CO2 was enhanced by approximately 65% compared to the rate at ambient CO2 (350 mol mol–1). A similar level of enhancement under elevated CO2 was observed for daily photosynthesis under field conditions on a sunny day. While enhancement of photosynthesis by elevated CO2 during the study period appears to be primarily attributable to direct photosynthetic responses to CO2 in the pine forest, longer-term CO2 responses and feedbacks remain to be evaluated.  相似文献

The effects of eight microelements (I, BO3 3–, MoO4 2–, Co2+, Cu2+, Mn2+, Fe2+, Zn2+) on the biosynthesis of camptothecin and the growth of suspension cultures of Camptotheca acuminata were studied. The increase of I to 25 M l–1, Cu2+ to 1 M l–1, Co2+ to 2 M l–1 and MoO4 2– to 10 M l–1 in Murashige and Skoog (MS) medium resulted in 1.66, 2.84, 2.53 and 2.04 times higher of camptothecin yield than that in standard MS medium respectively. Combined treatment of I (25 M l–1), Cu2+ (1 M l–1), Co2+ (2 M l–1) and MoO4 2– (10 M l–1) lead to improve cell dry weight, camptothecin content, and camptothecin yield to 30.56 g l–1, 0.0299%, and 9.15 mg l–1, respectively, which were 20.2, 208.9 and 273.8% increment respectively when compared with those of control.  相似文献

Explants excised from strawberry (Fragaria x ananassa Duch.) plantlets were cultured in vitro for 21 days on half-strength MS (Murashige & Skoog 1962) basal liquid medium with 20 g l-1 sucrose and without sugar in the vessels capped with gas permeable microporous polypropylene film. The experiments were conducted under CO2 nonenriched (350–450 mol mol-1 in the culture room) and CO2 enriched (2,000 mol mol-1 during the photoperiod in the culture room) conditions with a PPF (photosynthetic photon flux) of 200 mol m-2 s-1. The CO2 concentration in the vessels decreased to approximately 200 mol mol-1 during the photoperiod on day 21 under CO2 nonenriched conditions. The fresh and dry weight, net photosynthetic rate (NPR) per plantlet, NPR per g leaf fresh weight, NPR per g leaf dry weight, the number of unfolded leaves, and ion uptake of PO4 3-, NO3 -, Ca2+, Mg2+ and K+ on day 21 were the greatest under photoautotrophic (no sugar in the medium) and CO2 enriched conditions. The photoautotrophic micropropagation of potato cv. Russet Burbank was investigated. Single node microcuttings were grown for four weeks on Murashige and Skoog (MS) medium with or without sucrose (30 g l–1) in the growth room at 21/19 °C day/night temperature, with 16-h photoperiod at 150 mol m–2 s–1, with or without supplemental CO2 at 1500 l l–1. A 20% increase in the number of nodes per stem (from 7.5 to 9.4) and a 50% increase in stem dry weight were observed in cultures grown on media with sucrose and in CO2 enriched atmosphere comparing to the conventionally micropropagated cultures or the cultures grown photoautotrophically on media without sucrose but in air supplemented with 1500 l l–1CO2. Stems of these cultures (from media with sucrose in CO2 enriched air) almost doubled in length the stems of cultures from the other two treatments. No significant differences were observed between Control (MS medium supplemented with sucrose, 30 g l–1) and photoautotrophic cultures coming from MS medium with no sucrose grown under 1500 l l–1 of CO2. Photoautotrophic cultures produced stems averaging 43.3 mm, with 7 nodes and weighing 9.2 mg (dry weight), similar to conventionally grown in vitro cultures (47.9 mm with 7.5 nodes, 9.7 mg dry weight). Growers may consider photoautotrophic culturing of potato in areas where the high sterility levels are difficult to maintain. Supplementing air in the growth room with 1500 l l–1 of CO2 could be beneficial for potato plantlet production even on media containing sucrose since it significantly improved quality, size and biomass of produced plantlets, speeding up the multiplication.  相似文献

Leaf photosynthetic rates were measured on field-grown soybeans during the 1980 season. Comparisons were made between different cultivars and isolines representative of maturity groups I–IV. Mature, fully expanded leaves at different nodes on the plant were measured in high light to determine which had the highest potential photosynthetic rates at any one time. Successive leaves during the growing season had maximum rates which increased from about 22 mol CO2 m-2 s-1 on 25 June to a peak of 30–44 mol CO2 m-2 s-1 in early August.The persistency and eventual decline in the maximum rate was associated with the maturity group and related dates of flowering, pod fill and onset of senescence. Early maturing cultivars (groups I and II) had higher peak rates (38–44 mol CO2 m-2 s-1) than later maturing cultivars (30–35 mol CO2 m-2 s-1, groups III and IV). However, the photosynthetic rates of early maturing cultivars declined rapidly after attaining their peak, whereas the leaves of later maturing cultivars maintained their photosynthetic activity for much longer.  相似文献

Synopsis Arsenic persists in Chautauqua Lake, New York waters 13 years after cessation of herbicide (sodium arsenite) application and continues to cycle within the lake. Arsenic concentrations in lake water ranged from 22.4–114.81 g l–1, = 49.0 ag l–1. Well water samples generally contained less than 10 g l–1 arsenic. Arsenic concentrations in lake water exceeded U.S. Public Health Service recommended maximum concentrations (10 g l–1) and many samples exceeded the maximum permissible limit (50 g l–1). Fish accumulated arsenic from water but did not magnify it. Fish to water arsenic ratios ranged from 0.4–41.6. Black crappie (Pomoxis nigromaculatus) contained the highest arsenic concentrations (0.14–2.04 g g–1 ), X = 0.7 g g–1) while perch (Perca flavescens), muskellunge (Esox masquinongy) and largemouth bass (Micropterus salmoides) contained the lowest concentrations (0.02–0.13 g g–1). Arsenic concentrations in fish do not appear to pose a health hazard for human consumers.  相似文献

In Taxus cuspidata callus, vanadyl sulfate (10 mg l–1) induced a high (146 g g–1 dry wt) production of 10-deacetylbaccatin III in comparison to 7 g g–1 dry wt of the control. The content of paclitaxel in this species increased from 16 g g–1 to 74 g g–1 dry wt when 20 mg phenylalanine l–1 was used. In T. media, p-aminobenzoic acid induced the highest content of 10-deacetylbaccatin III (481 g g–1 dry wt) versus 181 g g–1 in the control. Paclitaxel increased from 89 to 139 g g–1 dry wt after adding chitosan (20 mg l–1) to the cultures.  相似文献

The effect of lead on the filtration rate of the zebra musselDreissena polymorpha was investigated, together with the accumulation of Pb in the soft tissues of the mussels. The NOEC-filtration was 116 g.l–1 (0,56 mol.l–1) and the EC50-filtration was 370 g.l–1 (1.79 mol.l–1). The NOEC-accumulation was the concentration found in the control water (1.4g.l–1). These experiments show that the EC50-filtration for Pb is similar to that for Cd, higher than that for Cu and lower than that for Zn. The water quality criteria for lead allow 25 g Pb.l–1 in surface water. This will not cause short-term effects. Long-term effects may, however, occur, since an accumulation of Pb as low as 16 g.l–1 was recorded in this study.  相似文献

In late March 1997, an open-top-chamber (OTC) CO2 enrichment study was begun in the Colorado shortgrass steppe. The main objectives of the study were to determine the effect of elevated CO2 (720 mol mol–1) on plant production, photosynthesis, and water use of this mixed C3/C4 plant community, soil nitrogen (N) and carbon (C) cycling and the impact of changes induced by CO2 on trace gas exchange. From this study, we report here our weekly measurements of CO2, CH4, NOx and N2O fluxes within control (unchambered), ambient CO2 and elevated CO2 OTCs. Soil water and temperature were measured at each flux measurement time from early April 1997, year round, through October 2000. Even though both C3 and C4 plant biomass increased under elevated CO2 and soil moisture content was typically higher than under ambient CO2 conditions, none of the trace gas fluxes were significantly altered by CO2 enrichment. Over the 43 month period of observation NOx and N2O flux averaged 4.3 and 1.7 in ambient and 4.1 and 1.7 g N m–2 hr –1 in elevated CO2 OTCs, respectively. NOx flux was negatively correlated to plant biomass production. Methane oxidation rates averaged –31 and –34 g C m–2 hr–1 and ecosystem respiration averaged 43 and 44 mg C m–2 hr–1 under ambient and elevated CO2, respectively, over the same time period.  相似文献

We studied the effects of atmospheric CO2 enrichment (280, 420 and 560 l CO2 l–1) and increased N deposition (0,30 and 90 kg ha–1 year–1) on the spruce-forest understory species Oxalis acetosella, Homogyne alpina and Rubus hirtus. Clones of these species formed the ground cover in nine 0.7 m2 model ecosystems with 5-year-old Picea abies trees (leaf area index of approx 2.2). Communities grew on natural forest soil in a simulated montane climate. Independently of N deposition, the rate of light-saturated net photosynthesis of leaves grown and measured at 420 l CO2 l–1 was higher in Oxalis and in Homogyne, but was not significantly different in Rubus compared to leaves grown and measured at the pre-industrial CO2 concentration of 280 l l–1. Remarkably, further CO2 enrichment to 560 l l–1 caused no additional increase of CO2 uptake. With increasing CO2 supply concentrations of non-structural carbohydrates in leaves increased and N concentrations decreased in all species, whereas N deposition had no significant effect on these traits. Above-ground biomass and leaf area production were not significantly affected by elevated CO2 in the more vigorously growing species O. acetosella and R. hirtus, but the slow growing H. alpina produced almost twice as much biomass and 50% more leaf area per plant under 420 l CO2 l–1 compared to 280 l l–1 (again no further stimulation at 560 l l–1). In contrast, increased N addition stimulated growth in Oxalis and Rubus but had no effect on Homogyne. In Oxalis (only) biomass per plant was positively correlated with microhabitat quantum flux density at low CO2, but not at high CO2 indicating carbon saturation. On the other hand, the less shade-tolerant Homogyne profited from CO2 enrichment at all understory light levels facilitating its spread into more shady micro-habitats under elevated CO2. These species-specific responses to CO2 and N deposition will affect community structure. The non-linear responses to elevated CO2 of several of the traits studied here suggest that the largest responses to rising atmospheric CO2 are under way now or have already occurred and possible future responses to further increases in CO2 concentration are likely to be much smaller in these understory species.  相似文献

Hawaii's dry forests are among the most endangered of all ecosystems in the archipelago. Invasion of alien plant species into these ecosystems is one of the most significant threats to on-going efforts to preserve and restore Hawaii's remaining dry forests. Comparing the physiological performance of alien and native species can offer causal explanations behind the relative success of alien plant invasions within Hawaiian dry forests and elsewhere. We compared maximum rates of net CO2 assimilation, water-use efficiency (WUE), daily carbon gain, and leaf morphology for three native and two alien shrubby species growing within 1-m2 plots under two natural light (sub-canopy shade relative to open full sunlight) treatments. Maximum rates of net CO2 assimilation were similar between alien and native species (8.15 vs. 7.12molm–2s–1, respectively), however, native plants exhibited lower stomatal conductance and higher instantaneous WUE than alien plants in all treatments (0.13molm–2s–1 and 72.36mol CO2mol H2O–1 against 0.23 and 58.78, respectively). Alien plants had approximately 65% more aboveground biomass than native plants. This result may reflect differential seed production and seed bank viability between native and alien species. We found an overall strong, positive correlation between species-specific physiological traits and final species aboveground biomass. Based on this type of information we can predict species-specific boundaries across light gradients, and focus restoration efforts accordingly.  相似文献
