We have found three inherited inversions in Chironomus riparius populations from the Borok fishpond, namely: (A3d-B1a) in the arm A (C5a-C6a) in the arm D and (B3b-4d/e) in the arm F. Increase of heterochromatin in some bands of chromosome F (B3h, B3h + B3c--C1a) and puffs appearance in the arms C, D and E have been observed. We saw also changes in functional activity of nucleolar organizer (N) and Balbiani rings (BRe/BRb). It has been found that some of inversion breakpoints coincide with the Alu and Hinf satellite DNA localization sites.  相似文献

Summary Acute toxicity tests using a static-with-replacement testing procedure were carried out with the four larval instars of the freshwater detritivore Chironomus riparius (Meigen). Median lethal concentrations (10, 24, 48 and 96 h LC50's) indicated great differences in sensitivity to cadmium between instars. Larvae became more tolerant with increasing age, the most resistant stage (fourth instar) having a 24 h LC50 of 2,400 mg Cd l-1, approximately 950 times greater than the corresponding value of 2.1 mg Cd l-1 recorded for the most sensitive (first instar) stage. These observations are discussed in relation to the establishment of water quality standards.  相似文献

Five variable microsatellite loci are reported for the nonbiting midge species Chironomus riparius and Chironomus piger. All loci show considerable intraspecific variation and species‐specific alleles, which allow to discriminate among the two closely related species and their interspecific hybrids, and to estimate genetic diversity within and between populations. Additionally, the loci were localized on C. riparius polytene chromosomes to verify their single copy status and investigate possible chromosomal linkage. The described markers are used in different studies with regard to population and ecological genetics and evolutionary ecotoxicology of Chironomus.  相似文献

A new technique for the establishment of a permanent laboratory culture of Chirono-mus riparius is described. Larvae and pupae were reared in plastic tanks containing tap water, filamentous algae and a commercially prepared fishfood. The tanks were aerated, maintained at a constant temperature of 24°C and subjected to a constant lighting regime of 13 h of light followed by 11 h of darkness. Adults flew freely in a modified handling box, mated, and oviposited on strips of filter paper placed in the plastic tanks containing the aquatic stages. It has proved unnecessary to change the water or medium, which has saved both time and effort. Moreover, larvae established themselves in ail parts ofthe alga, and therefore more could be kept in each tank than when, as in previous methods, they are provided only with a substratum on the floor of the container.  相似文献

Superoxide dismutase (SOD, EC 1.15.1.1) is an enzyme involved in the scavenging of reactive oxygen species (ROS) into molecular oxygen and hydrogen peroxide. In this study, a copper-zinc superoxide dismutase (Cu-ZnSOD) gene and a manganese superoxide dismutase (MnSOD) gene in aquatic midge, Chironomus riparius (CrSODs) was identified using an Expressed Sequence Tag (EST) database generated by 454 pyrosequencing. A multiple sequence alignment of C. riparius sequences revealed high homology with other insect sequences in terms of the amino acid level. Phylogenetic analysis of the CrSODs revealed that they were grouped with SODs of other organisms, such as Polypedilum vanderplanki, Drosophila melanogaster, Aedes aegypti, Anopheles gambiae, Culex quinquefasciatus and Bombyx mori. Expression of the corresponding CrSODs was analyzed during different developmental stages and following exposure to various environmental contaminants with different mode of actions i.e., paraquat, cadmium, benzo[a]pyrene, and chloropyrifos. CrSOD gene expression was significantly up or down regulated in response to exposure to the chemicals tested. The overall results suggested that SOD gene expression provided a platform for the understanding of oxidative stress responses caused by exposure to various environmental contaminants, and the SOD genes could be used as biomarkers for environmental disturbances such as oxidative stress initiated by xenobiotics.  相似文献

A population of Chironomus riparius from a Po river station near Moncalieri (a trace-metal polluted station) was studied. In this population was established a great variability of band structure of polytene chromosomes as well as paracentric heterozygous inversions, deletions, deficiencies, partial breaks, diploid chromosome fragments, and changes in functional activity and appearance of heterochromatin. In arms A through F, some bands had an increased size compared to the standard chromosomic map. Some bands appeared in a heterozygous or normal homozygous state or were amplified. In all arms, many condensed stable bands appeared in the decondensed state when compared to the standard map. Asynaptic zones in arms E and G as well as heterozygous Balbiani rings and NORs were established. Very often the 4th chromosome was almost completely heteropycnotic and looded like a pompon chromosome. For the first time in this species, a high frequency of ectopic pairings of different arms was observed. Telomeric regions involved in ectopic pairings had a granular appearance, as did some centromeres. The hypothesis is advanced that such a high frequency of structural rearrangements could be correlated with genomic distribution of specific mobile elements.  相似文献

SUMMARY. The effects of a sewage effluent in Moat Brook, Staffordshire, were determined from regular collections of water and biological samples from directly comparable stations above and below the sewage outfall.
The eutrophic conditions below the outfall result in the replacement of Polypedilum laetum by Chironomus riparius (except in the spring) as the dominant member of the Chironominae. Continuous recruitment of first instar larvae to the population of C. riparius in summer and autumn results in much overlap of generations, but emergence trap captures suggest at least five generations in the year.
The usefulness of C. riparius as an indicator of enriched conditions in lotic ecosystems should be considered in relation to oviposition behaviour and the occurrence and seasonal variation in density of the adults and aquatic stages (including the more conspicuous third and fourth instar larvae).  相似文献

Larvae of the midge, Chironomus riparius Mg., have four anal papillae arranged as two pairs, one dorsal to and the other ventral to the anus. Structural study with light and electron microscopes has revealed that their integument consists of a thin cuticle overlaying a thick, syncytial epidermis which is specialised to facilitate ion transport. There is a distinct neck or collar region at the junction of each papilla with the rest of the body wall. Although in many respects these structures resemble those in mosquitoes, there are both morphological and physiological features which suggest that differences exist in the means by which ion exchange with the environment is controlled and effected in the two families.  相似文献

In order to identify Chironomus hemoglobin (Hb) as a biomarker of ecotoxicity monitoring; herein, the effects of cadmium chloride (Cd) on Hb parameters were investigated in the 4th instar larvae of Chironomus riparius. The expressions of globin mRNA and hemolymph protein, using ecotoxicoproteomic approach, were investigated. Conventional ecotoxicity tests were also conducted to validate the ecotoxicological relevance of the response of Chironomus Hb as a biomarker. The proteomic analysis indicated that exposure to Cd lead alteration in the expression of hemolymph protein, with the total expressions of 12 hemolymph protein spots decreasing in response to treatment, with that of two increasing in response to Cd exposure. In addition, all of the spots differentially expressed in response to Cd treatment were identified as globin proteins. The decreased total Hb content observed in the hemolymph of larvae exposed to Cd suggested that the decreased expression of selected globin proteins in response to Cd exposure impacted on Hb synthesis. The overall results suggested that Hb could be a target molecule for exposure to Cd in C. riparius, with a proteomic approach appearing to be an ideal tool for the discovery of biomarkers in ecotoxicological research.  相似文献

Chironomus circumdatus is one of the most common and wide-spread species in India. Cytogenetic studies pertaining to the mitotic and polytene chromosomes, nucleolar organizer regions, C-banding and naturally occurring chromosomal polymorphism have been carried out for the first time in this Indian species. Altogether seven inversions comprising six paracentric and one pericentric are detected in the Indian populations. The distribution of inversions in relation to different environmental conditions is discussed.  相似文献

Phospholipid-hydroperoxide glutathione peroxidase (PHGPx) is an antioxidant enzyme in the glutathione peroxidases (GPx) family that reduces hydroperoxides of phospholipids and maintains the integrity of biomembranes. Here, we report the identification and characterization of a full length cDNA of PHGPx from the ecotoxicologically important aquatic midge Chironomus riparius (CrPHGPx1) from the Expressed Sequence Tags (ESTs) database generated through pyrosequencing. The 837 base pair (bp) cDNA contained an open reading frame of 597 bp, and a 75 bp 5' and a 159 bp 3'untranslated region. The theoretical molecular mass of the deduced amino acid (aa) sequence (197 aa) was 22.40 kDa with an estimated pI of 8.77. The Cys-codon was present at residue 74 and also the active site residues Gln(91) and Trp(164). The active-site motifs and GPx family signature motifs LAFPCNQF(101-108) and WNFTK(163-168) were also found. Phylogenetic analysis showed that CrPHGPx1 is grouped with PHGPx1 from other species and is more closely related to insects belonging to the dipteran order. The mRNA of CrPHGPx1 was detected in larvae, pupae and adults. The expression of CrPHGPx1 is induced by cadmium exposure indicating that the mRNA expression of CrPHGPx1 is differently regulated in response to oxidative stress caused by environmental stressors.  相似文献

Intra- and interspecific variation and divergence of multilocus markers for genomic DNA of the sibling species from the thimmi group,Chironomus riparius and C. piger, were studied by PCR with arbitrary primers (RAPD). A high level of RAPD polymorphism was determined in both laboratory and natural populations of these species. The genetic distances were estimated between the C. riparius populations and between the sibling species C. riparius and C. piger. The genetic distance between C. riparius andC. piger was 4 to 5 times higher than that between the C. riparius populations. A comparison of the variation and divergence for the RAPD markers with those for other genomic markers—enzyme-coding genes and chromosomes (gene linkage groups)—showed that different components of the genome differed in their contribution to the genome divergence.  相似文献

Summary The larval integument of the midge, Chironomus riparius Mg., is unusually thin although it conforms with the normal insect pattern. The cuticle of the post-cephalic segments is about 3 m thick and overlies an epidermis which has an irregular basal plasma membrane resulting in spaces occurring between it and the basement membrane. The ventral tubuli have a similar epidermis but the cuticle is somewhat thinner. The anal papillae have the thinnest cuticular covering with a uniquely folded epicuticle of variable thickness, and their epidermis has the characteristics of a transporting epithelium. No evidence of pore canals could be found in the cuticle of any part except the head capsule which has a remarkably smooth epicuticle and a distinct layer which may represent the exocuticle. There are no spaces between the basement membrane and basal plasma membrane of the epidermis in the head. Ultrastructural evidence would suggest that gaseous exchange can occur across most of the post-cephalic integument.The author is indebted to Mrs. L. Rolph and Mr. R.L. The Smoking Hills in the North West Territories, Canada have burnt for centuries causing long term acidification of ponds in the vicinity. Chironomus riparius , a red chironomid larva dominated the bottom fauna in the ponds. It was investigated if it was the buffering capacity of the hemolymph rather than the oxygen binding capacity of its hemoglobin that gave these larvae a selective advantage and explained their high abundance. Canadian C. riparius larvae were compared with larvae of the same species from Sweden and with larvae of Chironomus plumosus and C. anthracinus also from Sweden. The Canadian strain of C. riparius survived 7 d exposure to pH 3.5 much better than the others. Titration curves on the hemolymph showed that the Canadian larvae had the highest buffering capacity. Within the pH range 6.0–7.0 it was 17–18 meq pH−1 1−1 as compared with 13–15 meq pH−1 1−1 for the others. The hemoglobin content of the Canadian larvae was found to be twice as high as that of the same species from Sweden.  相似文献

The effects of various mercury chloride (HgCl2) concentrations associated with bottom sediments on mercury accumulation in chironomid larvae and the modifications of morphofunctional parameters (mortality, growth, and structure of the mouth apparatus) have been studied. It was shown that, upon exposure, the larvae accumulated mercury in amounts exceeding its sediment concentrations. The mortality rate and metamorphosis duration increased with an increase in metal concentrations, while the animal size remained unaffected. Among all investigated morphological structures, the antennae were the most heavily effected.  相似文献

2,4-Dichlorophenoxyacetic acid (2,4-D) is an agricultural contaminant found in rural ground water. It remains to be determined whether neither 2,4-D poses environmental risks, nor is the mechanism of toxicity known at the molecular level. To evaluate the potential ecological risk of 2,4-D, we assessed the biological parameters including the survival rate, adult sex ratio of emerged adults, and mouthpart deformities in Chironomus riparius after long-term exposure to 2,4-D. The larvae were treated with 0.1, 1 or, 10 μg L? 1 of 2,4-D for short- and long-term exposure periods. The sex ratio was changed in C. riparius exposed to only 10 μg L? 1 of 2,4-D, whereas mouthpart deformities were observed as significantly higher in C. riparius exposed to 0.1 μg L? 1 of 2,4-D. Survival rates were not significantly affected by 2,4-D. Furthermore, we evaluated the molecular and biochemical responses of biomarker genes such as gene expression of heat shock proteins (HSPs), ferritins and glutathione S-transferases (GSTs) in C. riparius exposed to 2,4-D for 24 h. The expressions of HSP70, HSP40, HSP90 and GST levels in C. riparius were significantly increased after exposure to a 10 μg L? 1 concentration of 2,4-D, whereas ferritin heavy and light chain gene expressions were significantly increased at all concentrations of 2,4-D exposure. Finally, these results may provide an important contribution to our understanding of the toxicology of 2,4-D herbicide in C. riparius. Moreover, the 2,4-D-mediated gene expressions may be generated by 2,4-D is the causative effects on most probable cause of the observed alterations. These biological, molecular and morphological parameters and the measured parameters can be used to monitor 2,4-D toxicity in an aquatic environment.  相似文献

Summary The legs of chironomid midges from a laboratory colony were examined in the region of the joint between the fifth tarsal segment and the pretarsus, especially the surface of the unguitractor and the manner in which the unguitractor fits into a ventromedian groove in the edge of the tarsus when the joint is flexed. The region was reconstructed from serial sections to clarify the spatial relations of the internal structures to one another and to the external structures. Ultrastructural characteristics of the cells and cuticle suggest a secretory function of the unguitractor. An amphinematic scoloparium is suspended between the point at which the unguitractor attaches to its tendon and the transverse diaphragm within the tarsus. This mechanoreceptor could serve as a proprioceptive sensor of the position of unguitractor and tarsus; it could also be an exteroceptor, sensing vibration in the substrate and/or the air. In the context of functional morphology, the clamping of the unguitractor in the ventral hollow in the tarsus could have the effect of resetting the sensitivity of the sensor. Deformities and fluctuating asymmetry in chironomid larvae have been proposed as sensitive indicators of biological stress and are commonly used to assess the ecological impact of human activities. In particular, they have been associated in Chironomus riparius, the most commonly used species, with heavy metal and pesticide river pollution. In this study, the effect of lead and 4-nonylphenol on mouthpart morphological variation of Chironomus riparius larvae was investigated by traditional and geometric morphometrics. For this purpose, first to fourth instar larvae were exposed to sediment spiked with lead (from 3.0 to 456.9 mg/kg dry weight) or 4-NP (from 0.1 to 198.8 mg/kg dry weight). Mentum phenotypic response to pollutants was assessed by four parameters: (1) the frequency of deformities, (2) fluctuating asymmetry of mentum length, (3) fluctuating asymmetry of mentum shape and (4) the mentum mean shape changes. Despite the bioaccumulation of pollutants in the chironomid’s body, no significant differences between control and stressed groups were found for mouthpart deformities and fluctuating asymmetry of mentum length. Slight effects on mentum shape fluctuating asymmetry were observed for two stressed groups. Significant mean shape changes, consisting of tooth size increase and tooth closing, were detected for lead and 4-NP exposure respectively. Those variations, however, were negligible in comparison to mentum shape changes due to genetic effects. These results suggest that the use of mentum variation as an indicator of toxic stress in Chironomus riparius should be considered cautiously.  相似文献
