The parameters of transmembrane potentials of cardiomyocytes and dynamics of fibrillation process have been investigated when altering the level of sympathetic action on the heart by injection of rausedile, katecholamines and by stimulation of the nucleus of hypothalamus. It was shown that the injection of rausedile decreases rest potential and potential activity, meanwhile electrical irritation significantly increases the period of spontaneously reversible fibrillation.  相似文献

Sun ginseng (SG) is heat-processed Panax ginseng C.A. Meyer steamed at 120 °C, which has ginsenoside-Rg3, -Rk1, and -Rg5 as its main ginsenoside components. The effect of SG on lipopolysaccharide (LPS)-induced liver injury in rats was investigated in this study. Intravenous injection of LPS induced excessive nitric oxide (·NO) generation in serum and increased the hepatic mitochondrial thiobarbituric acid-reactive substance (TBA-RS) level. However, the elevated TBA-RS level was significantly lowered by 15 consecutive days of SG administrations. In addition, up-regulated hepatic inducible nitric oxide synthase and heme oxygenase 1 levels in LPS-treated control rats were significantly lowered and increased, respectively, by 100 mg/kg body weight/day of SG administration. These antioxidant effects were thought to be partially related to the deactivation of nuclear factor-κB by SG administration.  相似文献

Background

Sleeve gastrectomy (SG) has emerged recently as a stand-alone bariatric procedure to treat morbid obesity and enhance glucose homeostasis. The aim of the study was to evaluate its effects in neonatally streptozotocin (STZ)-induced diabetic rats (n-STZ diabetic rats).

Methodology and Principal Findings

To induce diabetes, STZ (90 mg/kg) was administered intraperitoneally to 2-day-old male pups. When 12 weeks old, diabetic rats were randomized into sleeve operation group (SLG, n = 6) and sham operation group (SOG, n = 6). Body weights were monitored weekly, and daily consumption of water and food were followed for eight consecutive weeks postoperatively. Serum glucose levels were measured periodically at the 4th and 8th week after surgery. Insulin, ghrelin, glucose-dependent insulinotropic polypeptide (GIP) and Glucagon-like peptide-1 (GLP-1) levels were assayed at the end of the study. Our data showed that SLG rats exhibited significantly lower body weight gain in addition to reduced food and water intakes postoperatively compared to their sham-operation counterparts. However, resolution of diabetes was not observed in our study. Correspondingly, there were no significant differences between SOG rats and SLG rats in glucose metabolism-associated hormones, including insulin, GIP and GLP-1. In contrast, ghrelin level significantly decreased (P<0.01) in SLG group (58.01±3.75 pg/ml) after SG surgery compared to SOG group (76.36±3.51 pg/ml).

Conclusions

These observations strongly suggest that SG is effective in controlling body weight. However, SG did not achieve resolution or improvement of diabetes in n-STZ diabetic rats.  相似文献

Introduction

Anti-Ro antibodies can be found in the serum of the majority of patients with Sjögren''s syndrome (SS). Immunization with a 60-kDa Ro peptide has been shown to induce SS-like symptoms in mice. The aim of this study was to investigate factors involved in salivary gland (SG) dysfunction after immunization and to test whether the induction of SS could be improved.

Methods

Ro60 peptide immunization was tested in Balb/c mice, multiple antigenic peptide (MAP)-Ro60 and Pertussis toxin (PTX) were tested in SJL/J mice. In addition, two injection sites were compared in these two strains: the abdominal area and the tailbase. Each group of mice was tested for a loss of SG function, SG lymphocytic infiltration, anti-Ro and anti-La antibody formation, and cytokine production in cultured cells or homogenized SG extracts.

Results

Ro60 peptide immunization in the abdominal area of female Balb/c mice led to impaired SG function, which corresponded with increased Th1 cytokines (IFN-γ and IL-12) systemically and locally in the SG. Moreover, changing the immunization conditions to MAP-Ro60 in the abdominal area, and to lesser extend in the tailbase, also led to impaired SG function in SJL/J mice. As was seen in the Balb/c mice, increased IFN-γ in the SG draining lymph nodes accompanied the SG dysfunction. However, no correlation was observed with anti-MAP-Ro60 antibody titers, and there was no additional effect on disease onset or severity.

Conclusions

Effective induction of salivary gland dysfunction after Ro60 peptide immunization depended on the site of injection. Disease induction was not affected by changing the immunization conditions. However, of interest is that the mechanism of action of Ro60 peptide immunization appears to involve an increase in Th1 cytokines, resulting in the induction of SG dysfunction.  相似文献

目的:建立海水淹溺肺损伤(SWD-ALl)的大鼠模型,为SWD—ALl的基础及救治研究提供平台。方法:128只健康SD大鼠随机分为4组,即对照(CG)组和海水灌注(SGI、SG2、SG4)组(分别气管内注入1、2、4mL/kg海水)。观察大鼠气管内海水灌注后症状以及各组呼吸频率、心率、动脉血气、肺湿/干重r~(W/D),留取肺组织病理标本观察病变。结果:与对照组比较,海水灌注组大鼠心率、呼吸频率明显加快,气道有白色泡沫渗出物,口唇发绀,肺部满布湿罗音。动脉血气分析氧分压(PaO2)和二氧化碳分压(Pa—CO2)显著降低(P〈0.01);SGl、SG2组30min测氧合指数(Pa02/FiO2)低于300mmHg,之后逐渐升高至接近正常,SG4组各时间点测PaOJFi02得均在300mmHg以下;各组间比较,SG4组肺组织湿/干重比(W/D)明显高于其他组(P〈0.01)。肺组织病理观察,光镜下肺间质水肿,肺泡隔断裂,大量的红细胞及炎性细胞浸润。结论:经气管内灌注海水4mL/kg,可成功建立海水淹溺急性肺损伤的动物模型。  相似文献

We investigated changes in thymic tissue of male rats exposed to a 900 megahertz (MHz) electromagnetic field (EMF) on postnatal days 22–59. Three groups of six 21-day-old male Sprague-Dawley rats were allocated as: control (CG), sham (SG) and EMF (EMFG) groups. No procedure was performed on the CG rats. SG rats were placed in a Plexiglas cage for 1 h every day between postnatal days 22 and 59 without exposure to EMF. EMFG rats were placed in the same cage for the same periods as the SG rats and were exposed to 900 MHz EMF. Rats were sacrificed on postnatal day 60. Sections of thymus were stained for histological assessment. Oxidant/antioxidant parameters were investigated biochemically. Malondialdehyde (MDA) levels in EMFG increased compared to the other groups. Extravascular erythrocytes were observed in the medullary/corticomedullary regions in EMFG sections. We found that 900 MHz EMF applied for 1 h/day on postnatal days 22–59 can increase tissue MDA and histopathological changes in male rat thymic tissue.  相似文献

Structural genomics (SG) initiatives are currently attempting to achieve the high-throughput determination of protein structures on a genome-wide scale. Here we analyze the SG target data that have been publicly released over a period of 16 months to assess the potential of the SG initiatives. We use statistical techniques most commonly applied in epidemiology to describe the dynamics of targets through the experimental SG pipeline. There is no clear bottleneck among the key stages of cloning, expression, purification and crystallization. An SG target will progress through each of these steps with a probability of approximately 45%. Around 80% of targets with diffraction data will yield a crystal structure, and 20% of targets with HSQC spectra will yield an NMR structure. We also find the overlaps among SG targets: 61% of SG protein sequences share at least 30% sequence identity with one or more other SG targets. There is no significant difference in average structure quality among SG structures and other structures in the PDB determined by "traditional" methods, but on average SG structures are deposited to the PDB twice as quickly after X-ray data collection.  相似文献

本文观察了运动性低铁状态大鼠骨髓细胞转铁蛋白 (Tf)结合铁和非Tf结合铁摄入的变化。大鼠随机分为 6个月的运动组 (EG)和对照组 (SG)。SG平均每个幼红细胞Tf受体数为 890 15 0± 16 4849个 ,而在EG为 2 17536 0± 46 2 737个 (P <0 0 5 ) ,但受体的解离常数不受运动影响。EG中Tf的内吞平台和胞内铁聚积速度显著高于SG ,胞浆和胞内膜性成分中Tf结合铁和Fe(Ⅱ )摄入增加。EG的胞浆内Fe(Ⅱ )摄入的米氏常数值降低 ;细胞膜性成分中Fe(Ⅱ )摄入的最大速度增加。上述结果表明 ,运动不仅通过增加Tf受体的表达促进Tf结合铁的内吞 ,而且增强非Tf结合铁的内吞途径。尽管这些变化的机制尚不清楚 ,但它们有利于运动时血红素的合成  相似文献

To compare the functional state of the superior cervical (SCG) and stellate sympathetic ganglia (SG) of spontaneously hypertensive rats (SHR) with those of age-matched normotensive Wistar Kyoto rats (WKY), ganglion cell volume and area occupied by ganglion cells relative to each whole ganglionic area were morphometrically examined using the Texture Analyse System (TAS) in rats at 0, 10 and 30 days of age. The weight of each ganglion relative to animal weight was also measured. The ganglion cell volume and the relative area of ganglionic cells in both ganglia of SHR were significantly larger (P less than 0.05) than those of age-matched WKY at ages 0 and 10 days after birth. The relative ganglionic weights of SHR were significantly larger (P less than 0.01) compared with those of WKY at all ages examined, except for SG at 0 days after birth. These results show that the relative volume of sympathetic ganglion cells is greater in both SCG and SG of SHR than that of WKY, suggesting that hyperfunction of sympathetic ganglia occurs at the prehypertensive stage as a primary factor in the development of hypertension in SHR.  相似文献

The haemodynamic effects of 6 weeks nandrolone decanoate treatment (total dose 30 mg.kg-1) (SG I, n = 12) and their reversibility were studied in anaesthetised, open-chest male rats exposed to 5 min isoproterenol (2.5 micrograms.kg-1) and CaCl2 (25.0 mg.kg-1) loads. In SG I, the heart weight and its ratio to body weight were greater than in the untreated rats (CG I, n = 13) (p less than 0.05 and p less than 0.01 respectively). The initial heart rate and the inotropic and chronotropic responses to isoproterenol were lower in SG I than in CG I (p less than 0.05 in all cases). Peripheral resistance decreased during both infusions in SG I but remained unaltered in CG I (p less than 0.05). 6 weeks after finishing anabolic steroid treatment (SG II, n = 11), in the CaCl2-test the ejection fraction (p less than 0.05) and stroke index were smaller than in control rats of the same age (CG II, n = 12). Mean aortic pressure was lower in SG II than in CG II. In the CaCl2-test peripheral resistance was initially higher, but decreased during the infusion in SG II while it increased in CG II (p less than 0.05 in both cases). In conclusion, anabolic steroid treatment reversibly reduces the left ventricular response to isoproterenol. It decreases peripheral vascular tone during inotropic loads. Six weeks after the cessation of treatment, the pumping efficiency of the heart is reduced.  相似文献

The number and segmental distribution of cell bodies of sensory afferents and sympathetic efferent innervating to the knee joint of the rabbit and the Formosan rock-monkey were investigated using retrograde transport with horseradish peroxidase (HRP). After injecting HRP into the articular knee joint capsule of the rabbits, labeled neurons were found in the ipsilateral L4-S2 dorsal root ganglia (DRG). However, following injection of HRP into the articular cavity of the knee joint in the rabbit and the monkey, labeled neurons were found in both the ipsilateral DRG (L5-S2 and L4-S1 of the rabbit and monkey, respectively) and in the ipsilateral sympathetic ganglia (SG) (L4-S3 (rabbit) and L3-S1 (monkey)). The majority of labeled neurons within the DRG and the SG were composed of medium and large neurons in the monkey and the rabbit, respectively. The present findings suggest that the sensory projections from and sympathetic projection to the knee joint in rabbits and monkeys are similar, but that both projections of monkeys were "shifted" one segment cranially compared to the rabbit on both projections.  相似文献

We produced three monoclonal antibodies, SG1, SG2 and SG3, specific for human seminal -glutamyltransferase when characterized by enzyme-linked immunosorbent assay and immunoblotting. Seminal -glutamyltransferase was localized, by immunostaining, to the epithelial cells of the ductus epididymidis, seminal vesicle and prostate gland with SG1, those of the prostate gland with SG2, and those of the seminal vesicle with SG3. Rabbit polyclonal anti-seminal -glutamyltransferase serum reacted with the proximal convolution of the kidney and the bile capillaries of the liver, and with the epithelial cells of the reproductive organs. However, immunoreactivity was not observed in the kidney or liver with the monoclonal antibodies. Thus, these monoclonal antibodies are probably all specific to seminal -glutamyltransferase but recognize different epitopes.  相似文献

Nerve growth factor (NGF) plays a critical role in the maintenance and survival of both sympathetic and sensory nerves. Also, NGF can regulate receptor expression and neuronal activity in the sympathetic and sensory neurons. Abnormalities in NGF regulation are observed in patients and animals with heart failure (HF). Nevertheless, the effects of chronic HF on the levels of NGF within the sympathetic and sensory nerves are not known. Thus, the ELISA method was used to assess the levels of NGF in the stellate ganglion (SG) and dorsal root ganglion (DRG) neurons of control rats and rats with chronic HF induced by myocardial infarction. Our data show for the first time that the levels of NGF were significantly decreased (P < 0.05) in the SG and DRG neurons 6–20 weeks after ligation of the coronary artery. In addition, a close relation was observed between the NGF levels and the left ventricular function. In conclusion, chronic HF impairs the expression of NGF in the sympathetic and sensory nerves. Given that sensory afferent nerves are engaged in the sympathetic nervous responses to somatic stimulation (i.e. muscle activity during exercise) via a reflex mechanism, our data indicate that NGF is likely responsible for the development of muscle reflex-mediated abnormal sympathetic responsiveness observed in chronic HF.  相似文献

1.  A prominent population of olfactory receptor cells from the lobsterHomarus americanus is narrowly tuned to trans-4-hydroxyl-L-proline (Hyp, Fig. 1) suggesting that Hyp may be an important chemical signal for this animal (Johnson and Atema 1983). However, Hyp is usually bound in connective tissue proteins of lobster prey and thus may be unavailable in sufficient quantities as a free amino acid to stimulate chemoreceptors. To determine other possible adequate stimuli for Hyp sensitive cells we further examined their tuning using a variety of substances including other amino acids also found in collagens, Hyp isomers (Fig. 1), synaptic receptor agonists, ecdysones, purified natural collagens and their gelatins (Fig. 2), and different molecular weight fractions of a commercial gelatin solution (Table 1).
2.  Of a first group of Hyp sensitive cells (N=29) tested with the compounds in Table 1 A, 21 responded best to Hyp. Only a commercial gelatin solution (SG1) and its one-tenth dilution consistently elicited responses from these cells (Fig. 3). The remaining 8 cells responded best to the SG1 solutions (5 cells) or to one of the other test substances (Table 2).
3.  Of a second group of Hyp sensitive cells (N=27) tested with the collagen and gelatin solutions (Table 1b), 19 responded best to Hyp. Again, the Hyp best cells rarely responded to any test substance other than Hyp and a commercial gelatin solution, SG2, and its greater than 12 kD fraction, SG2-12 (Fig. 4). SG2 and SG2-12 were equally effective for the Hyp best cells. The remaining 8 cells responded best to either SG2 (2 cells), SG2-12 (2 cells) or one of the purified gelatin or collagen solutions (Table 3).
4.  A third group of Hyp sensitive cells (N=21) was tested with SG2, SG2-12, and a greater than 1 kD fraction of SG2 (SG2-1; Table 1c). Based on the mean response, the most effective stimulus for these cells was SG2-12, followed by SG2-1, SG2 and Hyp. The high mean response for the SG2 solutions was mainly due to a few cells giving large responses to these stimuli (Fig. 6). Ten of these 21 cells responded best to Hyp; all but 2 responded to one or more of the SG2 solutions; the other 11 cells responded best to either SG2, SG2-12 or SG2-1, which were all best stimuli for different cells (Fig. 5).
5.  Overall, the 77 Hyp sensitive cells tested here can be divided into two main types; 65% Hyp best cells and 31% gelatin best cells. The Hyp best cells seem to be a distinct population of receptors: they have no spontaneous activity and give low responses (15 spikes in 5 s) even to their best stimulus, Hyp. In contrast, the gelatin best cells are not infrequently spontaneously active and can give high responses to their best stimulus (up to 150 spikes in 5 s). In addition, when tested specifically in the third group, the Hyp cells appear to have a tuning spectrum distinct from the gelatin best cells (Fig. 7).
6.  Stimulation of Hyp cells by gelatin solutions may be due to Hyp-containing peptides derived from the gelatin. Enzymatic tissue breakdown from the lobster's prey could produce chemical mixtures that stimulate prominent receptor populations which respond to both high (gelatin best cells) and low (Hyp best cells) molecular weight substances. This could create a central representation of food based on parallel receptor lines of somewhat overlapping sensitivity. Together, Hyp best and gelatin best receptor cell populations may give important information on the presence and state of decay of the lobster's food.

The bilateral patterns of forward and reversed scaphognathite (SG) pumping are described for the American lobster. During forward pumping the two SGs usually function synchronously, but may also function independently. The nine muscles of one SG are arranged into four functional groups which are sequentially active during forward pumping. During reversed beats, motor neurones to one group of muscles are inactive while bursts to another are either delayed or missing. Reversal beats do not appear to alter the phasing of the central oscillators that generate the basic SG rhythm. Phase analysis of bilateral SG beating demonstrates two types of relationship: phase coupling or phase drifting with a tendency to couple. An animal may remain in one state for long periods of time or may alternate between states. Coupling can occur at more than one phase indicating phase multistability. The coupled state may remain constant at markedly different frequencies of beating, indicating phase rather than latency coupling between SGs. During the drifting state each SG tends to assume its "intrinsic" rate of oscillation. The drift state reflects the inherent asymmetry of the two SG systems. The influence of several parameters of sensory stimulation on phase and frequency of SG beating are analyzed.  相似文献
