In 1995, the eyeless (ey) gene was dubbed the "master-regulator" of eye development in Drosophila. Not only is ey required for eye development, but its misexpression can convert many other tissues into eye, including legs, wings and antennae.(1) ey is remarkable for its ability to drive coordinate differentiation of the multiple cell types that have to differentiate in a very precise pattern to construct the fly eye, and for its power to override the previous differentiation programs of many other diverse tissues. Even more remarkable, the ey homolog Pax6 and homologs of other eye determination genes from Drosophila are also required for eye development in vertebrates,(2,3) prompting reassessment of the evolution of vision throughout the animal kingdom.(4,5) Now Kumar and Moses have published a study that throws a new light on ey function in Drosophila.(6) According to their work, ey becomes a master regulator of eye development much later than previously thought, and is regulated by signalling through the Notch and EGFR signaling pathways.  相似文献

The presence of glycosyltransferases on surfaces of mammalian cells has been reported by many investigators and a biological role for these enzymes in cell adhesion and cell recognition has been postulated. Critical analysis, however, showed 2 major complications regarding the assay for cell surface glycosyltransferases: (1) hydrolysis of the nucleotide sugar by cell surface enzymes and subsequent intracellular use of the free sugar and (2) loss of cell integrity if trypsinized or EDTA-treated cells were used in suspension asays. We have assayed intact, viable cells in monolayer for cell surface glycosyltransferases using conditions under which intracellular utilization of free sugars generated by hydrolysis of the nucleotide sugar was prevented. Our data demonstrate that the presence of galactosyltransferases on the surface of a variety of cells, including established (normal and virally transformed) as well as nonestablished cells, is unlikely. No evidence for the existence of cell surface fucosyl-and sialyltransferases could be obtained, but our data do not exclude the possibility that low levels of these enzymes are present.  相似文献

While innovations in modern microscopy, spectroscopy, and nanoscopy techniques have made single molecule observation a standard in many laboratories, the actual design of meaningful fluorescence reporter systems now hinders major scientific breakthroughs. Even though the field of chemical biology is supercharging the fluorescence toolbox, surprisingly few strategies exist that make the transition from model systems to biologically relevant applications. At the same time, the number of microscopy techniques is growing dramatically. We explain our view on how the impact of modern technologies is influenced not only by further hard‐ and software developments, but also by the availability and suitability of protein‐engineering tools. We identify how the largely independent research fields of chemical biology and fluorescence nanoscopy can influence each other to synergistically drive future technology that can visualize the localization, structure, and dynamics of molecular function without constraints.  相似文献

The diamondback moth (DBM), Plutella xylostella (L.) (Lepidoptera: Plutellidae), is oligophagous on plants in the Brassicaceae, and is considered one of the most significant insect pests of canola (Brassica napus L.), a major oilseed crop grown in the Prairie Provinces of Canada. The bertha armyworm (BAW), Mamestra configurata Walker (Lepidoptera: Noctuidae), is a generalist herbivore that preferentially feeds on canola plants. In the canola growing season in the Prairie Provinces of Canada, DBM feeding occurs prior to BAW herbivory. In this study, we test the effect of DBM herbivory on subsequent host use by BAW. Oviposition by female BAW was not influenced by previous DBM‐herbivory or mechanical damage of canola plants. Bertha armyworm larvae were deterred from feeding on canola previously damaged by DBM and larvae developed into lighter pupae when reared on DBM‐damaged plants. Bertha armyworm pupae that developed from larvae fed on mechanically damaged plants had similar pupal weights to those fed undamaged plants. Adult BAW reared on canola with previous DBM feeding damage had marginally smaller wings than moths reared on canola treated differently. The combination of these results suggests that herbivory by the brassicaceous specialist, DBM, negatively influences host use and larval performance by the generalist, BAW, on canola.  相似文献
