Experiments are described which attempt to clarify the quantitative relationship between sieve-tube loading of sucrose, and ATP-turnover rates in the phloem of willow (Salix viminalis L.). Two experimental approaches have been made towards the solution of this problem. In the first of these the respiratory breakdown of 14C-sugars was measured in segments of willow stem when no sieve-tube transport was taking place, and also under conditions where transport was occurring in response to the feeding of individuals of the aphid Tuberolachnus salignus (Gmelin). An increase in respiratory activity, measured by the output of 14CO2, was found to occur as a consequence of transport. Since the rate of sieve-tube sugar loading could be measured by the production of honeydew from the aphids, and by making assumptions concerning the production of ATP in respiration, it was concluded that the stoichiometry of sucrose loading was 1.9 mol ATP · (mol sucrose)-1. A somewhat higher value of 2.5 mol ATP · (mol sucrose)-1 was found using the second approach. In this, attempts were made to measure ATP turnover rates using [32P]orthophosphate supplied to strips of willow bark which bore exuding aphid stylets.  相似文献

Polyamine content and enzyme activities in the biosynthetic and degradative pathways of polyamine metabolism were investigated in sieve-tube sap, xylem sap and tissues of seedlings and adult plants of Ricinus communis L. Polyamines were present in tissues and translocation fluids of both seedlings and adult plants in relatively high amounts. Only free polyamines were translocated through the plant, as indicated by the finding that only the free form was detected in the phloem and the xylem sap. Removal of the endosperm increased the polyamine content in the sieve-tube exudate of seedlings. The level and pattern of polyamines in tissue of adult leaves changed during leaf age, but not, however, in the sieve-tube sap. Xylem sap was relatively poor in polyamines. Polyamine loading in the phloem was demonstrated by incubating cotyledons with [14O]putrescine and several unlabelled polyamines. Feeding cotyledons with cadaverine and spermidine led to a decrease in the level of putrescine in sieve-tube sap, indicating a competitive effect. Comparison of polyamine content in the tissue and export rate showed that the export would deplete the leaves of polyamines within 1–3 d, if they were not replenished by biosynthesis. Polyamine biosynthesis in Ricinus proceeds mostly via arginine decarboxylase, which in vitro is 100-fold more active than ornithine decarboxylase. The highest arginine decarboxylase, ornithine decarboxylase and diamine oxidase activities were detected in cotyledons, while in sieve-tube sap only a slight arginine decarboxylase activity was found.   1. The giant willow aphid Tuberolachnus salignus is an invasive pest in New Zealand, attacking over 50 species and hybrids of willow. The aphids produce copious amounts of honeydew, which is used by other insects as a food source.
  2. When foraged by honeybees, T. salignus honeydew causes honey to crystallize in the comb and affects bee health; these effects are associated with the elevated melezitose content in the honeydew. The impact of host plant-related factors on T. salignus honeydew melezitose content remains unknown.
  3. This study investigated the effect of willow cultivar and plant age on the melezitose content (and that of other sugars) of T. salignus honeydew. To do so, we conducted high-performance liquid chromatography analyses of honeydew samples from 13 willow clones collected in the same season (autumn) from 1- and 2-year old plants under field conditions.
  4. Melezitose was the most abundant of the measured sugars in most samples, but its content did not vary significantly with willow cultivar or plant age. By contrast, sucrose was significantly affected by both factors. Fructose and glucose were significantly impacted by willow plant age and cultivar, respectively. A significant cultivar*age interaction was observed for all sugars.
  5. We recommend the selection of resistant willow cultivars and further research on potential biocontrol agents to lessen melezitose-related problems in apiculture industries.

The leaves of willow cuttings were allowed to assimilate 14CO2,and the rate of increase of activity in the sieve-tube sap,collected as aphid honeydew, was compared with the output ofrespiratory 14CO2 from the stem at 25 °C and at 0 °C. In the case of 3–5-week-old young shoots, the relativerate of breakdown of the 14C-labelled sugars was reduced at0 °C as compared with 25 °C. With 2–4-year-oldmature stems, however, the rate of breakdown of labelled sugarsin transit through the phloem was increased at 0 °C relativeto that at 25 °C. When the labelled sugars were being respiredin isolated stem segments where no transport was taking place,then low temperatures markedly reduced the rate of breakdownin both young shoots and mature stems. These results are discussed in relation to the results of otherworkers who have studied temperature effects on sieve-tube transport.  相似文献

Variation in phloem sap composition is important in determining aphid performance and is known to occur at both diurnal timescales and in response to plant age. For field grown potato plants, Solanum tuberosum L. (Solanaceae), we determined diurnal variation in components of phloem sap, measured by ethylene diamine tetra‐acetate exudation, and tested for impacts of plant age. The effects of plant age and diurnal cycles on honeydew production by Macrosiphum euphorbiae (Thomas) and Myzus persicae (Sulzer) (both Hemiptera: Aphididae) were also quantified. Both the ratio of sucrose to amino acids and the composition of amino acids in phloem sap varied significantly with time of day. Dietary essential amino acids contributed a smaller proportion of amino acids in the phloem sap of older plants and during early phases of the diurnal cycle. The only significant effect on aphid honeydew production was of the diurnal cycle for Ma. euphorbiae, although increased honeydew production during the day when compared with the production at night, was consistent across the two species. In contrast with studies carried out at seasonal scales, we found limited evidence for variation in phloem sap composition in response to plant age, consistent with our results for honeydew production. These data highlight the need for improved understanding of how seasonal and diurnal physiology of plants influence performance in phloem sap feeding insects.  相似文献

Summary (2-Chloroethyl)trimethylammonium chloride (Cycocel or CCC), an inhibitor of gibberellin biosynthesis, when repeatedly supplied to the root systems of balsam (Impatiens glandulifera Royle) plants reduces growth in height and the level of gibberellin-like substances in the bleeding sap that exudes from the stumps of detopped plants. Within twelve hours after a single application of the inhibitor to decapitated field peas (Pisum arvense), there are quantitative and qualitative changes in the gibberellins of the sap compared with those in sap collected over the same period of time from untreated plants. These changes are interpreted in terms of the possible blockage by CCC of normal gibberellin production and diversion of precursors into synthesis of abnormal gibberellins.  相似文献

Abstract.The effects of water stress on phloem sap quality of the melon, Cucumis melo, and how this, in turn, has an impact on the sweet potato whitefly, Bemisia tabaci were studied. Melon plants were grown under watering regimes that produced plants with or without water stress. Plants showed strong developmental responses to the treatments; water-stressed plants were shorter, with fewer, smaller leaves than those without stress. There was, however, no effect of plant water stress on the development period of whiteflies feeding on these plants, or on the weights of male or female adults. Honeydew production was used as an indirect measure to test whether the absence of insect developmental or behavioural effects was due to differential phloem sap ingestion. Feeding rates on the stressed plants were almost half those on unstressed plants, and there was also variation in the daily pattern of honeydew production. Phloem sap and honeydew were analysed to determine why the feeding behaviours differed. Amino acid composition of the phloem sap was similar in both groups of plants, but carbohydrate concentrations were greater in water-stressed plants, indicating that lower feeding rates may have been due either to the physical or nutritional quality of the phloem sap. The honeydew of insects that were feeding on water-stressed plants contained a greater concentration of carbohydrate than those on unstressed plants, and was composed of a significantly greater proportion of glucose and the disaccharide, trehalulose. This isomerization of more complex sugars from those in the diet suggests that B. tabaci uses a mechanism of osmoregulation to actively maintain its internal water status. It is concluded that transient conditions of water stress in this host plant do not affect the development of B. tabaci, due to physiological and behavioural changes in response to diets with different nutritional and physical properties. The implications of this finding for the feeding biology of B. tabaci on desert-grown crops are discussed.  相似文献

The mature, functional sieve-tube system in higher plants is dependent upon protein import from the companion cells to maintain a functional long-distance transport system. Soluble proteins present within the sieve-tube lumen were investigated by analysis of sieve-tube exudates which revealed the presence of distinct sets of polypeptides in seven monocotyledonous and dicotyledonous plant species. Antibodies directed against sieve-tube exudate proteins from Ricinus communis L. demonstrated the presence of shared antigens in the phloem sap collected from Triticum aestivum L., Oryza sativa L., Yucca filamentosa L., Cucurbita maxima Duch., Robinia pseudoacacia L. and Tilia platyphyllos L. Specific antibodies were employed to identify major polypeptides. Molecular chaperones related to Rubisco-subunit-binding protein and cyclophilin, as well as ubiquitin and the redox proteins, thioredoxin h and glutaredoxin, were detected in the sieve-tube exudate of all species examined. Actin and profilin, a modulator of actin polymerization, were also present in all analyzed phloem exudates. However, some proteins were highly species-specific, e.g. cystatin, a protease-inhibitor was present in R. communis but was not detected in exudates from other species, and orthologs of the well-known squash phloem lectin, phloem protein 2, were only identified in the sieve-tube exudate of R. communis and R. pseudoacacia. These findings are discussed in terms of the likely roles played by phloem proteins in the maintenance and function of the enucleate sieve-tube system of higher plants. Summary Auxin activity was detected in honeydew obtained from the aphid Tuberolachnus salignus (Gmelin) feeding on willow (Salix viminalis). Active uptake of 14C-indolyl-3-acetic acid (IAA) into the sieve tubes was demonstrated by irrigating the cambial surface of willow bark with 14C-IAA solution and assaying aphid stylet exudate. When, however, 14C-IAA was applied to the peridermal tissues of the bark or to a mature leaf most of the radioactivity (collected in honeydew or stylet exudate) co-chromatographed with indolyl-3-acetyl-aspartic acid (IAAsp). The presence of IAAsp in honeydew was not affected by extraction procedure or by aphid metabolism. Honeydew obtained from willow treated with 14C-tryptophan contained only 14C-tryptophan. When 14C-IAA was applied in agar to the cut end of willow segments the radioactivity was found to move in a basipetally polar manner. The direction of movement of radioactivity in the sieve tubes, however, was found to be influenced by the proximity of the roots. Nevertheless, there was evidence that endogenous auxin in the sieve tubes does move in a predominantly basipetal direction.  相似文献

Rare unifloral willow (Salix spp.) honeys obtained from nectar or honeydew were investigated by direct RP‐HPLC‐DAD method in order to identify and quantify compounds that can be used as possible markers of their origin. Antioxidant and antiradical activities of willow honeys were evaluated using FRAP (=ferric reducing antioxidant assay) and DPPH (=1,1‐diphenyl‐2‐picrylhydrazyl radical) tests, respectively. Also HMF (=5‐(hydroxymethyl)furfural), diastase activity, and CIE L*a*b*C*h* chromatic coordinates were evaluated. Abscisic acids (ABA) are typical of willow nectar honey, with a predominance of (Z,E)ABA on (E,E)ABA (98.2 and 31.7 mg/kg, resp.). Kinurenic acid and salicylic acid are useful to mark willow honeydew honey. The proposed HPLC‐DAD method proved to be easy and reliable to identify the two different Salix spp. honeys, being not affected from any sample preparation artifact. Total antioxidant activity measured with the FRAP assay ranged from 3.2 to 12.6 mmol Fe2+/kg, and the antiradical activity measured with the DPPH assay ranged from 0.6 to 3.0 mmol TEAC (=Trolox equivalent antioxidant capacity)/kg in nectar and honeydew honeys, respectively. Salix spp. nectar and honeydew honeys proved to be two completely different honeys, because, besides color attributes, they show different antioxidant properties and specific compounds.  相似文献

The level of gibberellin-like substances was determined in the cotyledons and axis of developing seedlings of dwarf (Little Marvel) and normal (Tall Telephone) cultivars of pea (Pisum sativum L.). The effect of cotyledon removal with GA3 application on growth was also examined. Greater levels of gibberellin-like substances were observed in the cotyledons of the normal cultivar than the dwarf. This was particularly evident in the cotyledons during the early stages of seedling growth. Subsequently there was a decline in GA levels in the cotyledons. This was coincidental with a rise in GA content in the axis with markedly greater levels in the normal than the dwarf cultivar. Decotyledonated dwarf and normal plants supplied with GA were much taller than the decotyledonated controls. This observation along with those of the gibberellin levels in the cotyledons and axis, provided circumstantial evidence that there may be translocation of gibberellins from the cotyledons to the axis.  相似文献

Certain aspects of the secretion of solutes into, and removalfrom, the sieve tubes of isolated stem segments and rooted cuttingsof Salix viminalis have been studied. Sieve-tube sap was obtainedeither as honeydew from whole individuals or via the severedstylets of the aphid Tuberolachnus salignus (Gmelin). It was shown that interference occurred between the chemicallyunrelated solutes, sucrose and the cations potassium and rubidium.On raising the potassium concentration in the sieve-tube sapby passing a solution of this ion through the xylem, the sucroseconcentration declined. When the sucrose concentration fellover a period of days due to respiratory loss of carbohydratesfrom an isolated stem segment, a concomitant rise in eitherthe potassium or rubidium level in the sap occurred. When a solution of sodium was passed through the xylem, theconcentration of this ion in the sieve-tube sap rose, whilstthat of potassium fell at first, but later rose higher thanits initial value, indicating that both antagonism and synergycan occur between these ions. On introducing both these cationsinto the xylem simultaneously, more sodium than potassium wastaken up by the segment, though the increase in the sodium concentrationin the sieve-tube sap was less than that of the potassium. Perfusingthe xylem with a calcium solution had no effect upon the concentrationof potassium in the sieve tube. It has been shown that the rate of translocation of a solutealong the sieve tube, as measured by the two colony technique,depends upon the rate of removal of this solute from the sievetube. The amount of such lateral loss from the sieve tube isrelated to the potential gradient for a solute between the sievetube and surrounding cells.  相似文献

The Asian citrus psyllid (ACP), Diaphorina citri Kuwayama (Hemiptera: Liviidae), transmits the citrus greening pathogen ‘Candidatus Liberibacter asiaticus’ (CLas) by feeding on citrus phloem sap. Because phloem sap is rich in sugars but low in amino acids, ACP sucks large quantities and excretes most of it as honeydew. We studied the chemical composition of ACP honeydew on various host plants. Honeydew samples were analyzed with gas chromatography–mass spectrometry. Fourteen sugars, 13 amino acids, and six organic acids were detected in the honeydew of ACP. Sugars composed about 95% of the total compounds. Sucrose and trehalose were the predominant sugars, composing about 58 and 23% of the total sugars, respectively. Proline, asparagine, aspartic acid, and glutamic acid were the most abundant amino acids in ACP honeydew. The host plant and its infection with CLas had some effect on the honeydew composition. Glucose, chiro‐inositol, myo‐inositol, inositol, maltose, and turanose were lower in honeydew collected from CLas‐infected citrus compared to that collected from non‐infected trees. In CLas‐infected citrus (pineapple sweet orange, Citrus sinensis L. Osbeck) and Bergera koenigii (L.) Spreng. [curry leaf tree (both Rutaceae)] honeydews, valine, alanine, serine, glutamine, glycine, and the organic acids were lower than in honeydew from healthy citrus. Mannose, galactose, inositol, mannitol, an unknown disaccharide, and proline were higher in the honeydew collected from B. koenigii than in honeydew collected from healthy citrus (pineapple sweet orange), whereas fructose, chiro‐inositol, myo‐inositol, trehalose, and lactic acid were lower. The findings of this study help us understand the metabolism and the nutrient needs of ACP that transmits CLas, the pathogen of huanglongbing in citrus.  相似文献

We analysed interactions in the system of two Barley Yellow Dwarf Virus (BYDV) strains (MAV and PAV), and wheat (cv. Tinos) as host plant for the virus, and the cereal aphid Sitobion avenae (F.) as vector, in particular whether or not infection by the virus might alter host plant suitability in favour of vector development. By measuring the amino acid and sugar content in the phloem sap of infected and non‐infected wheat plants we found a significant reduction in the concentration of the total amount of amino acids on BYDV‐infected plants. Qualitative and quantitative analysis of honeydew and honeydew excretion indicated a lower efficiency of phloem sap utilisation by S. avenae on infected plants. In addition, S. avenae excreted less honeydew on infected plants. Both BYDV strains significantly affected aphid development by a reduction in the intrinsic rate of natural increase. Hence, infection by the virus reduced the host suitability in terms of aphid population growth potential on BYDV‐infected plants. However, more alate morphs developed on virus‐infected plants. These findings are discussed in relation to the population dynamics of S. avenae, and, as a consequence, the spread of BYDV.  相似文献

We studied the dynamics of phytohormone levels (indole-3-acetic acid, abscisic acid, cytokinins, and gibberellin-like substances) in red beet (Beta vulgaris L.) roots at different developmental stages in comparison with the data on the dynamics of sugar accumulation in order to test possible hormonal regulation of sugar accumulation. The obtained data suggest the involvement of cytokinins and gibberellin-like substances in the control of sugar accumulation in the roots, while indole-3-acetic acid, abscisic acid, and gibberellin-like substances can control the outflow. Summary It is shown that there is one growth inhibitor in the phloem sap and two in the xylem sap of willow (S. viminalis L.). The concentration of the inhibitor in the phloem sap, (+)-abscisic acid, increases as the plants enter dormancy. This is also shown for (+)-abscisic acid in the xylem sap, but the concentration of the second inhibitor decreases in a reciprocal manner.  相似文献

It has been widely accepted that herbivory induces morphological, phenological, and chemical changes in a wide variety of terrestrial plants. There is an increasing appreciation that herbivore‐induced plant responses affect the performance and abundance of other arthropods. However, we still have a poor understanding of the effects of induced plant responses on community structures of arthropods. We examined the community‐level effects of willow regrowth in response to damage by larvae of swift moth Endoclita excrescence (Lepidoptera: Hepialidae) on herbivorous and predaceous arthropods on three willow species, Salix gilgiana, S. eriocarpa and S. serissaefolia. The leaves of sprouting lateral shoots induced by moth‐boring had a low C:N ratio. The overall abundance and species richness of herbivorous insects on the lateral shoots were increased on all three willow species. Densities of specialist chewers and sap‐feeders, and leaf miners increased on the newly emerged lateral shoots. In contrast, the densities of generalist chewers and sap‐feeders, and gall makers did not increase. Furthermore, ant and spider densities, and the overall abundance and species richness of predaceous arthropods increased on the lateral shoots on S. gilgiana and S. eriocarpa, but not S. serissaefolia. In addition to finding that effects of moth‐boring on arthropod abundance and species richness varied among willow species, we also found that moth‐boring, willow species, and their interaction differentially affected community composition. Our findings suggest that moth‐boring has community‐wide impacts on arthropod assemblages across three trophic levels via induced shoot regrowth and increase arthropod species diversity in this three willow species system.  相似文献

Structurally complex metabolites such as carbohydrates, organic acids, and amino acids are leached from plants by rain and dew. Applied GA3 (1 mg/ml) absorbed by a 5-cm stem segment of Chrysanthemum morifolium and translocated to a single attached leaf was leached from the leaf with distilled water. To detect endogenous gibberellins in plant leachates, Chrysanthemum morifolium ‘Princess Anne’ plants were leached under 8-hr and 18-hr photoperiods. The leachates were collected and extracted at pH 8.2 with methylene chloride to*** remove inhibitors and at pH 3.0 to remove gibberellins. The extract containing the gibberellins was fractionated on a silica gel column by an eluting phase of 0 to 100% ethyl acetate-n-hexane solutions. The presence of gibberellin-like substances was confirmed by their elution pattern from the silica gel column and by two bioassays, the lettuce hypocotyl and the Rumex bioassays. The results suggest that substances similar to GA1–3 and GA5 were leached from plants growing under an 18-hr photoperiod. The leachate from plants growing under an 8-hr photoperiod contained gibberellin-like substances similar to GA1–3 and GA6.  相似文献

Gradients of tritiated water, 35S-sulphate and 32P-phosphate, were established in isolated segments of willow stems. Sieve tube exudate was collected as honeydew from the high activity end of the segment. After girdling the stem a few centimetres from the site of sieve tube puncture, the specific activity of 35S and 32P in the honeydew rose, whilst the specific activity of tritium remained constant. These findings indicate that prior to girdling, unlabelled sulphates and phosphates were contributing to the honeydew, whilst there had been no detectable contribution by unlabelled water from the low activity end of the segment. The data support the conclusions drawn from previous experiments by Peel et al. (1969), that water is relatively immobile in sieve tubes of willow when compared to solutes.  相似文献

本文通过对白背飞虱Sogatella furcifeta Horvath在不同苗龄抗虫品种Rathu Hee nati(简称RHT)和感虫品种Taichung Native 1(简称TNl)稻株上的取食反应,对摄入食物中糖类的利用和体内葡糖苷酶的活性变化,以及对稻株含糖量的分析,初步探讨了白背飞虱对稻株糖类物质的利用。白背飞虱在抗虫、感虫品种上的取食均随稻株苗龄的增长而降低,沮不管品种的抗虫性如何,稻株内的总糖量却随稻株苗龄的增加而上升。高效液相色谱分析飞虱分泌的蜜露结果表明,在RHT上分泌的蜜露里,蔗糖和果糖的含量明显地低于在TNI上分泌的。由此推测白背飞虱从RHT稻株韧皮部中吸食的汁液较从TNI中的少,但利用率高。取食RHT后的白背飞虱,体内的葡糖苷酶活性明显地较取食TNl后的低,并与飞虱体重的变化呈正相关。在白背飞虱体内可能存在着两种独立的、可控制食物的摄入及摄入食物的消化和吸收的机制。  相似文献
