Solanum lycocarpum (Solanaceae) is a woody species found in the Brazilian Cerrado. The flowers are pollinated by Xylocopa spp bees, and seeds are dispersed by mammals with distinct home range sizes. As a consequence, relative contributions of pollen and seeds to overall gene flow can vary according to different spatial scales. We studied the genetic structure of four natural populations of S. lycocarpum separated by 19 to 128 km, including individuals located along dirt roads that interlink three of the populations. A total of 294 individuals were genotyped with five nuclear and six chloroplast microsatellite markers. Significant spatial genetic structure was found in the total set of individuals; the Sp statistic was 0.0086. Population differentiation based on the six chloroplast microsatellite markers (θ(pC) = 0.042) was small and similar to that based on the five nuclear microsatellite markers (θ(p) = 0.054). For this set of populations, pollen and seed flow did not differ significantly from one another (pollen-to-seed flow ratio = 1.22). Capability for long distance seed dispersion and colonization of anthropogenic sites contributes to the ability of S. lycocarpum to maintain genetic diversity. Seed dispersion along dirt roads may be critical in preserving S. lycocarpum genetic diversity in fragmented landscapes. 相似文献

Restricted gene dispersion – resulting from both self-pollination and limited capability of pollen migration, as well as seed dispersion at short distances – has been considered the main reason for spatial genetic structuring in plant populations. This study evaluated the intrapopulation genetic structure and the mating system in four populations of Solanum lycocarpum, a woody bush occurring in Brazilian Cerrado vegetation. Two hundred and twenty-four individuals were genotyped through five nuclear SSR loci (30 alleles) and six cpSSR loci (82 haplotypes). The study evidenced that the species mates predominantly by outcrossings , that biparental inbreeding is not common, and that there are almost 10 trees participating as pollen donors per mother-tree. The populations were formed by many mother lineages, indicating efficient seed dispersion by the fauna and the occurrence of multiple foundation events. Spatial genetic structure was observed in three populations (average Sp=0.0184 ± 0.0030) and it resulted from both restricted seed dispersion and from vegetative reproduction. During the collection of seeds for ex situ conservation, seeds must be gathered from 150 to 200 mother trees, so that the effective size of 500 individuals is retained. The sampling must comprise the biggest possible number of populations in a wide area to enable the maintenance of the biggest possible haplotypic diversity. 相似文献

The design and feasibility of whole-genome-association studies are critically dependent on the extent of linkage disequilibrium (LD) between markers. Although there has been extensive theoretical discussion of this, few empirical data exist. The authors have determined the extent of LD among 38 biallelic markers with minor allele frequencies >.1, since these are most comparable to the common disease-susceptibility polymorphisms that association studies aim to detect. The markers come from three chromosomal regions-1,335 kb on chromosome 13q12-13, 380 kb on chromosome 19q13.2, and 120 kb on chromosome 22q13.3-which have been extensively mapped. These markers were examined in approximately 1,600 individuals from four populations, all of European origin but with different demographic histories; Afrikaners, Ashkenazim, Finns, and East Anglian British. There are few differences, either in allele frequencies or in LD, among the populations studied. A similar inverse relationship was found between LD and distance in each genomic region and in each population. Mean D' is.68 for marker pairs <5 kb apart and is.24 for pairs separated by 10-20 kb, and the level of LD is not different from that seen in unlinked marker pairs separated by >500 kb. However, only 50% of marker pairs at distances <5 kb display sufficient LD (delta>.3) to be useful in association studies. Results of the present study, if representative of the whole genome, suggest that a whole-genome scan searching for common disease-susceptibility alleles would require markers spaced < or = 5 kb apart. 相似文献

We genotyped 15 microsatellite loci in order to evaluate the effects of habitat fragmentation, caused by flooding of the Tucuruí reservoir, on the genetic structure of Alouatta belzebul in eastern Amazonia. The analysis included two populations sampled in 1984, representing both margins of the Tocantins river, and three populations sampled 18 years later. Minimal differences in the diversity levels between present-day (Ho = 0.62-0.69 and A(R) = 6.07-7.21) and pre-flooding (Ho = 0.60-0.62 and A (R) = 6.27-6.77) populations indicated there was no significant loss of genetic variability, possibly because of successful management strategies applied during the flooding. The changes observed were limited to shifts in the composition of alleles, which presumably reflect the admixture of subpopulations during flooding. Given this, there were significant differences in the Rst values (p = 0.05) in all but one between-site comparison. Both present-day and original populations showed a deficit of heterozygotes, which suggests that this may be typical of the species, at least at a local level, perhaps because of specific ecological characteristics. The relatively large number of private alleles recorded in all populations may be a consequence of the Wahlund effect resulting from population admixture or a process of expansion rather than the loss of rare alleles through genetic drift. Additionally, the levels of genetic variability observed in this study were higher than those reported for other species of Neotropical primates, suggesting good fitness levels in these A. belzebul populations. Regular genetic monitoring of remnant populations, especially on islands, should nevertheless be an integral component of long-term management strategies. 相似文献

Hippophae rhamnoides ssp. sinensis is an ecologically and economically important species that has been widely used as a pioneer plant in China. In this study, we employed both nuclear ISSR and maternal cpSSR markers to survey the genetic diversity and structure of populations of ssp. sinensis representing three different landscapes, the northwestern desert and grassland region, the alpine vegetation region of the Qinghai-Tibetan Plateau, and the northeastern humid forest region. In all, 12 natural populations with a scattered distribution in the area were studied. The genetic diversities of populations were found to be uneven, and the total genetic diversity was low on the basis of both types of marker. Mantel tests based on both individual Euclidean distance matrices and population genetic distance (measured by ??pt) matrices showed that the two marker systems detected similar trends with respect to genetic distances between populations. The analysis of molecular variance (AMOVA) revealed significant differentiation among populations and among regions for both types of marker. Although the detected pattern of isolation-by-distance among all sampled populations confirmed the earlier colonization pathway, the low level of gene flow and the lack of isolation by distance within each region suggested the presence of an additional dispersal barrier. UPGMA dendrograms and PCA plots also revealed clear clustering and significant regional differentiation. Our results indicate that the genetic structure of ssp. sinensis has been affected by habitat fragmentation and restricted population sizes. We propose that the biology of reproduction and ecology have played determinant roles in the development of the regional structure of populations. The genetic information obtained will help to establish conservation strategies and programs for sustainable management of H. rhamnoides ssp. sinensis. 相似文献

We analyzed the genetic structure and diversity of Qualea grandiflora Mart., the most abundant woody species in the Brazilian Cerrado. Eight microsatellite loci were used to analyze samples from four populations subjected to different types of anthropic pressure, distributed throughout the state of São Paulo in the regions of Assis, Brotas, Itirapina and Pedregulho. Results indicated a mean number of 12 alleles per locus, but only six effective alleles. Alleles private to particular populations and rare alleles were also detected. An excess of homozygotes and moderate levels of inbreeding were observed. No clones were identified. All populations departed from Hardy–Weinberg equilibrium (p < 0.05). Spatial structure was observed in the distribution of specimens in distance classes ranging from 30 to 40 km and three genetic clusters were identified, with genotypes in the Pedregulho population differing from the others by up to 90 %. The influence of the Wahlund effect on the studied populations lies between 8.5 and 53.3 %. Estimates of effective population size were low (<10), and the minimum viable area for conservation in the short-, medium- and long-term was estimated to be between 4 and 184 ha. Gene flow was high enough to counter the effects of genetic drift. The genetic diversity and divergence between the studied populations indicated that the Pedregulho population should be considered an Evolutionary Significant Unit and a Management Unit. 相似文献

BACKGROUD AND AIMS: Berchemiella wilsonii var. pubipetiolata (Rhamnaceae) is distributed in fragmented habitat patches in eastern China. It is highly endangered because of severe disturbance by anthropogenic activities. Information on genetic variation and structure is critical for developing successful conservation strategies for this species. METHODS: Allozyme variation of population genetic diversity and structure was investigated for a total of 98 individuals sampled from four extant populations using isoelectric focusing in thin-layer polyacrylamide slab gels. KEY RESULTS: Based on 20 loci scored from the nine enzymes examined, a high genetic diversity was detected at both the species and population level, while there was a loss of low frequency alleles (<0.1) in all populations. Most loci showed deviation from Hardy-Weinberg equilibrium due to excess of heterozygotes in all populations, suggesting that selection for heterozygotes has occurred in this species. The genetic diversity was mainly found within populations with a moderate genetic differentiation (F(ST) = 0.13), but the two geographically discontinuous population groups showed significant differences, with F-statistic values of 0.078 for the Zhejiang populations and 0.014 for the Anhui populations, respectively. CONCLUSIONS: It appears most likely that this species has experienced a recent decrease in population size, and genetic drift in small populations has resulted in a loss of alleles occurring at low frequency. The differentiation into two population groups reflects a population genetic consequence that has been influenced by the different land-use in the two regions. Some conservation concerns are discussed together with possible strategies for implementing in situ and ex situ conservation. 相似文献

When a forest is fragmented, this increases the amount of forest edge relative to the interior. Edge effects can lead to loss of animal and plant species and decreased plant biomass near forest edges. We examined the influence of an anthropogenic forest edge comprising cattle pasture, coconut plantations, and human settlement on the mantled howler (Alouatta palliata), white-faced capuchin (Cebus capucinus), Central American spider monkey (Ateles geoffroyi), and plant populations at La Suerte Biological Research Station (LSBRS), Costa Rica. We predicted that there would be lower monkey encounter rate, mean tree species richness, and diameter at breast height (DBH) in forest edge versus interior, and that monkeys would show species-specific responses to edge based on diet, body size, and canopy height preferences. Specifically, we predicted that howler monkeys would show positive or neutral edge effects due to their flexible folivorous diet, large body size, and preference for high canopy, capuchins would show positive edge effects due to their diverse diet, small body size, and preference for low to middle canopy, and spider monkeys would show negative edge effects due their reliance on ripe fruit, large body size, and preference for high upper canopy. We conducted population and vegetation surveys along edge and interior transects at LSBRS. Contrary to predictions, total monkey encounter rate did not vary between the forest edge and forest interior. Furthermore, all three species showed neutral edge effects with no significant differences in encounter rate between forest edge and interior. Interior transects had significantly higher mean tree species richness than edge transects, and interior trees had greater DBH than edge trees, although this difference was not significant. These results suggest that forest edges negatively impact plant populations at La Suerte but that the monkeys are able to withstand these differences in vegetation. 相似文献

Cerrado (Brazilian savanna) areas present a high number of endemic and unknown species and consequently have been included among the world’s biodiversity hotspots. After a survey of zygomycetes from Cerrado areas of the “Reserva Biológica de Mogi Guaçu” (RBMG) in São Paulo State, we have found a Mucor-like fungus that produces very large sporangiospores and clusters of giant cells visible to the naked-eye. This isolate was characterized morphologically, based on partial LSU (28S) and complete ITS rDNA sequences. An extraordinary diversity of epiphytic lichens is found in the boreal rainforest of central Norway, the highest-latitude rainforest in the world. These rainforest relicts are located in ravine systems, and clear cutting has increased the distance between remaining patches. We hypothesized that the relatively small lichen populations in the remaining forest stands have suffered a depletion of genetic diversity through bottlenecks and founder events. To test this hypothesis, we assessed genetic diversity and structure in the populations of the tripartite lichen Lobaria pulmonaria using eight SSR loci. We sampled thalli growing on Picea abies branches and propagules deposited in snow at three localities. Contrary to expectations, we found high genetic diversity in lichen and snow samples, and high effective sizes of the studied populations. Also, limited genetic differentiation between populations, high historical migration rates, and a high proportion of first generation immigrants were estimated, implying high connectivity across distances <30km. Almost all genetic variation was attributed to variation within sites; spatial genetic structures within populations were absent or appeared on small scales (5-10m). The high genetic diversity in the remaining old boreal rainforests shows that even relict forest patches might be suitable for conservation of genetic diversity. 相似文献

Macaúba (Acrocomia aculeata) is a palm of economic importance, widely distributed in natural forests from Mexico to Uruguay. We analyzed the genetic diversity of populations of macaúba (A. aculeata) in the northern region of the State of Minas Gerais, Brazil. Young leaves from 10 macaúba individuals encompassing 49 genotypes of macaúba were collected from Montes Claros, Itacambira, Brasília de Minas, Mirabela, and Gr?o Mogol. After extraction and amplification of samples, the amplified fragments were separated by electrophoresis. We found high levels of genetic diversity within the populations. Genetic diversity indices were high, except in the Itacambira and Mirabela populations. Results show that Mirabela and Itacambira populations can require conservation strategies because they present lower values of genetic diversity. 相似文献

Human lymphocytes in the G0 stage were irradiated with UV light and X-rays. A 2-fold increase in the yield of dicentrics was observed in comparison with the yield for X-rays alone. This synergistic effect was constant irrespective of the variation in the UV dose between 50 and 100 erg/mm2.The individual chromosomes participated in interchange aberrations as expected from a random distribution per mitotic chromosome length unit. This observation is in contrast with the recent finding that X-ray-induced chromosome-type breakage is preferentially located on chromosomes with relatively large amounts of R-bands. Thus, the present data indicate that the additional breakage points, due to the synergism, had a different distribution between chromosomes from those induced by X-irradiation alone. The population genetic structure of the freshwater snail, Biomphalariaglabrata, was assessed in the municipality of Virgem das Graças(Minas Gerais, Brazil), a schistosomiasis-endemic area. Theseven microsatellite loci that were used to genotype 326 individualsfrom 17 collection sites within a 3-km radius revealed highgenetic variability. However, we found a deficit of heterozygotesrelative to HardyWeinberg expectations for most of theloci and in many collection sites, indicating a high level ofinbreeding. This may be the result of founder effects and sib-mating.A high level of overall population genetic differentiation amongB. glabrata collection sites suggested low gene flow at thisrelatively small spatial scale in this region. However, subsequentanalyses also indicated that these patterns may also resultfrom founder events and that an exchange of individuals betweenparticular collection sites does exist. The isolation-by-distancecorrelation between collection sites of B. glabrata was foundto be significant. However, only approximately 6% of the variancein FST was explained by geographic distances suggesting thatthere are other factors affecting genetic differentiation amongthese collection sites. It is likely that the current populationgenetic structure reflects patterns of both migration and founderevents. Results from this study are important for understandingthe potential for local adaptation between schistosomes andtheir snail hosts at this epidemiologically relevant scale. 相似文献

Disentangling the effects of natural environmental features and anthropogenic factors on the genetic structure of endangered populations is an important challenge for conservation biology. Here, we investigated the combined influences of major environmental features and stocking with non‐native fish on the genetic structure and local adaptation of Atlantic salmon (Salmo salar) populations. We used 17 microsatellite loci to genotype 975 individuals originating from 34 French rivers. Bayesian analyses revealed a hierarchical genetic structure into five geographically distinct clusters. Coastal distance, geological substrate and river length were strong predictors of population structure. Gene flow was higher among rivers with similar geologies, suggesting local adaptation to geological substrate. The effect of river length was mainly owing to one highly differentiated population that has the farthest spawning grounds off the river mouth (up to 900 km) and the largest fish, suggesting local adaptation to river length. We detected high levels of admixture in stocked populations but also in neighbouring ones, implying large‐scale impacts of stocking through dispersal of non‐native individuals. However, we found relatively few admixed individuals suggesting a lower fitness of stocked fish and/or some reproductive isolation between wild and stocked individuals. When excluding stocked populations, genetic structure increased as did its correlation with environmental factors. This study overall indicates that geological substrate and river length are major environmental factors influencing gene flow and potential local adaptation among Atlantic salmon populations but that stocking with non‐native individuals may ultimately disrupt these natural patterns of gene flow among locally adapted populations. 相似文献

Habitat fragmentation and climate change are two major threats on biodiversity. Fragmentation limits the number of patches and their decreased connectivity cannot always maintain populations at dynamic equilibrium. The natural extreme fragmentation of marine cave habitats represents an opportunity to understand how these processes interact. The hypothesis of a low gene flow among populations due to a high level of fragmentation was tested by analysing sequence variation in a fragment of the mitochondrial gene of the cytochrome oxidase subunit I in 170 individuals (23 localities across the NW Mediterranean) of two marine cave-dwelling mysids of the genus Hemimysis. The species Hemimysis margalefi recently replaced its congener Hemimysis speluncola, a species shift that could be related to the warming of the Mediterranean Sea and to differences in their thermal tolerances. There were too few H. speluncola samples to further discuss their genetic structuring, but for H. margalefi, the present study revealed high levels of genetic diversity and genetic structuring, as shown by the eight genetically distinct groups identified. The Croatian group might constitute a sibling species due to a strong divergence (15%). Nevertheless, these groups present reduced but orientated gene flow according to the general circulation in the Mediterranean, and fit a stepping-stone model. At local scale (Marseille area, France), gene flow among caves is dependent on unexpected local hydrodynamic barriers, that determine metapopulation sizes. Through the example of mysid species inhabiting marine caves, the present work confirms the strong influence of habitat disjunction (natural fragmentation) on population structure, and stresses the importance of coastal geomorphological features in inducing complex interactions between the circulation of water masses and the circulation of genes. 相似文献

Background and Aims
At least seven species of Agave, including A. parryi, were cultivated prehistorically in Arizona, serving as important sources of food and fibre. Many relict populations from ancient cultivation remain in the modern landscape, offering a unique opportunity to study pre-Columbian plant manipulation practices. This study examined genetic and morphological variation in six A. p. var. huachucensis populations of unknown origin to compare them with previous work on A. parryi populations of known origin, to infer their cultivation history and to determine whether artificial selection is evident in populations potentially managed by early agriculturalists.Methods
Six A. p. var. huachucensis and 17 A. parryi populations were sampled, and morphometric, allozyme and microsatellite data were used to compare morphology and genetic structure in purportedly anthropogenic and wild populations, as well as in the two taxa. Analysis of molecular variance and Bayesian clustering were performed to partition variation associated with taxonomic identity and hypothesized evolutionary history, to highlight patterns of similarity among populations and to identify potential wild sources for the planting stock.Key Results A
p. var. huachucensis and A. parryi populations differed significantly both morphologically and genetically. Like A. parryi, wild A. p. var. huachucensis populations were more genetically diverse than the inferred anthropogenic populations, with greater expected heterozygosity, percentage of polymorphic loci and number of alleles. Inferred anthropogenic populations exhibited many traits indicative of past active cultivation: greater morphological uniformity, fixed heterozygosity for several loci (non-existent in wild populations), fewer multilocus genotypes and strong differentiation among populations.Conclusions
Where archaeological information is lacking, the genetic signature of many Agave populations in Arizona can be used to infer their evolutionary history and to identify potentially fruitful sites for archaeological investigation of ancient settlements and cultivation practices. The aim of this work was to describe the yeast populations present during the manufacturing of Minas cheese of the region of Serra da Canastra, Minas Gerais state, Brazil. Canastra cheese is produced from raw cow’s milk at the farmhouse level using artisanal procedures and natural whey cultures as starters. Samples from 10 farms were studied, and they included: raw milk, natural starter, cheese curd before salting and cheese after 5 days of ripening. The most frequent yeasts in whey, curd and cheese were Debaryomyces hansenii, Kluyveromyces lactis, Kodamaea ohmeri and Torulaspora delbrueckii. Many yeast isolates were able to produce proteases, lipases and β-galactosidades. Production of these enzymes by yeasts in the cheese would contribute to the development of the characteristic flavor and smell during the ripening process. 相似文献
