A laboratory experiment was conducted to quantify the effects of infestation of maize by Sitotroga cerealella (Olivier) on progeny production by two common secondary colonizers of grain, Tribolium castaneum (Herbst) and Oryzaephilus surinamensis (L.). Adults of both secondary pest species were allowed to oviposit for 3 wk on intact kernels of 'DeKalb 689', mechanically split kernels, kernels that had been infested for 3 mo by S. cerealella, and kernels that had been infested for 6 mo. Progeny of both species reached highest numbers on 6-mo infested maize. Prior infestation for 6 mo by S. cerealella makes maize a more suitable medium for reproduction by T. castaneum and O. surinamensis, much more than can be accounted for by mere disruption of kernel integrity resulting from larval feeding. The results highlight the importance of limiting establishment by S. cerealella on maize in storage.  相似文献

Abstract

The ineffective control measures of pathogens is due to variability among their populations. Hence, the study of pathogenic variation of Fusarium verticillioides strains on maize genotypes. Six F. verticillioides infected maize ear were randomly obtained from three agro-ecological zones in Southwest Nigeria. Pathogenicity of F. verticillioides strains at 1.0?×?106 spores/mL were examined in vivo based on the rating scale of 1–7 on maize genotypes T2L COMP.1.STR SYN-W-1, PVA SYN 8F2, and T2L COMP.4. The pathogens were inoculated to the maize genotypes on 8th week for disease severity and were all moderately susceptible, however, genotype T2L COMP.4 was the most susceptible. It was observed that, 9.4% were classified as highly virulent, 12.5% as virulent, 37.5% as moderately virulent, 21.8% as slightly virulent, and 18.8% as non-virulent. In all, Fusarium verticillioides strains displayed different degrees of virulence, however, maize genotype T2L COMP.4 was the most susceptible to ear rot.  相似文献

Two corn borer species are the principal maize insect pests in Europe, the European corn borer, Ostrinia nubilalis (Hübner), and the pink stem borer, Sesamia nonagrioides (Lefebvre). Hence, it would be advisable to evaluate the European maize germplasm for corn borer resistance to generate European varieties resistant to corn borer attack. The creation of the European Union Maize Landrace Core Collection (EUMLCC) allowed the screening of most of the variability for European corn borer resistance present among European maize local populations from France, Germany, Greece, Italy, Portugal, and Spain, testing a representative sample. The objective of this study was the evaluation of stem and ear resistance of the EUMLCC to European corn borer and pink stem borer attack. Trials were made at two Spanish locations that represent two very different maize-growing areas. Populations that performed relatively well under corn borer infestation for stem and ear damage were 'PRT0010008' and'GRC0010085', among very early landraces; 'PRT00100120' and 'PRT00100186', among early landraces; 'GRC0010174', among midseason landraces; and 'ESP0070441', among late landraces. Either the selection that could have happen under high insect pressure or the singular origin of determined maize populations would be possible explanations for the higher corn borer resistance of some landraces. Landraces 'PRT0010008', 'FRA0410090', 'PRT00100186', and 'ESP0090214' would be selected to constitute a composite population resistant to corn borers and adapted to short season, whereas populations 'ESP0090033', 'PRT00100530', 'GRC0010174', and 'ITA0370005' would be used to make a resistant composite adapted to longer season.  相似文献

Infection of the maize (Zea mays L.) with aflatoxigenic fungus Aspergillus flavus and consequent contamination with carcinogenic aflatoxin is a persistent and serious agricultural problem causing disease and significant crop losses worldwide. The rachis (cob) is an important structure of maize ear that delivers essential nutrients to the developing kernels and A. flavus spreads through the rachis to infect kernels within the ear. Therefore, rachis plays an important role in fungal proliferation and subsequent kernel contamination. We used proteomic approaches and investigated the rachis tissue from aflatoxin accumulation resistant (Mp313E and Mp420) and susceptible (B73 and SC212m) maize inbred lines. First, we compared rachis proteins from resistant and susceptible inbred lines, which revealed that the young resistant rachis contains higher levels of abiotic stress-related proteins and proteins from phenylpropanoid metabolism, whereas susceptible young rachis contains pathogenesis-related proteins, which are generally inducible upon biotic stress. Second, we identified A. flavus-responsive proteins in rachis of both resistant and susceptible genotypes after 10- and 35-day infection. Differential expression of many stress/defense proteins during rachis juvenility, maturation and after A. flavus challenge demonstrates that resistant rachis relies on constitutive defenses, while susceptible rachis is more dependent on inducible defenses.  相似文献

The influence of donor plant genotype, ear maturity, explant size, and the ratio of ovule-to-cob tissue on kernel development from in vitro pollinated ovules was examined. All genotypes evaluated in this study were capable of in vitro pollination/fertilization, however, significant differences were observed for the responses measured. Genotype means for complete kernel formation ranged from 1.5% to 25.4% with B73 exhibiting the highest response. Averaged over all genotypes, ear maturity effects were not significant, however, the genotype x ear maturity mean square was significant for swelling percentage. Explant size had a profound effect on in vitro kernel development. Averaged over all genotypes and ear maturities, 30-ovule explants resulted in more than twice as many ovules classified as complete kernels when compared to 10-ovule explants. Ovule-to-cob tissue ratio was also found to have highly significant effects on all three variables measured. An ovule-to-cob tissue ratio of 4:24 resulted in the highest percentages of swelling, embryos with incomplete embryos, and complete kernels.  相似文献

The significance of the glycolytic and gluconeogenic conversion of fructose-6-phosphate and fructose-1,6-bisphosphate on sugar metabolism was investigated in maize (Zea mays L.) kernels. Maximum extractable activities of the pyrophosphate (PPi) dependent phosphofructokinase, fructose-1,6-bisphosphatase, and the ATP-dependent phosphofructokinase were measured in normal and four maize genotypes, which accumulate relatively more sugars and less starch, to determine how these enzymes are affected by the genetic lesions. Normal endosperm accumulated more dry matter than the high sugar/low starch genotypes, but protein contents did not differ greatly among the genotypes. Mutation of several starch biosynthetic enzymes had little impact on the activities of PPi-dependent phosphofructokinase, fructose-1,6-bisphosphatase, and ATP-dependent phosphofructokinase, despite the altered capacity of the cell to synthesize starch. The PPi-dependent phosphofructokinase appeared to be more active toward glycolysis in all genotypes studied. Activity of the PPi-dependent phosphofructokinase in shrunken (low sucrose synthase genotype) did not differ from the activity in other genotypes, suggesting that the gluconeogenic production of PPi may not be the primary role of the enzyme. As expected, shrunken kernels contained more sugars and less starch than normal kernels throughout kernel development except at the very early stages. Developmental profiles of normal kernels also showed marked changes in the PPi-dependent phosphofructokinase activity, whereas the level of ATP-dependent phosphofructokinase activity remained relatively steady during kernel development. In addition, the ATP-dependent phosphofructokinase, and not the PPi-dependent phosphofructokinase, appeared to correlate more closely with respiration rate. These findings suggest that glycolysis catalyzed by the ATP-dependent phosphofructokinase may serve primarily to support energy production, and glycolysis catalyzed by the PPi-dependent phosphofructokinase may contribute mainly to generation of biosynthetic intermediates.  相似文献

Popcorn adapted to Spanish conditions could be an interesting and profitable alternative to field corn. However, little is known about breeding popcorn germplasm for adaptation to Spain. Sesamia nonagrioides Lefèvbre is the main insect pest affecting popcorn quality and yield under Spanish growing conditions. The objectives of the study were the search for sources of resistance to S. nonagrioides among popcorn germplasm and to study the genetics of the resistance to S. nonagrioides attack. Eight breeding populations along with a five-inbred line diallel and two popcorn commercial checks were evaluated under S. nonagrioides infestation in 2 yr. Significant differences were found among general combining ability (GCA) effects for days to silking, S. nonagrioides tunnel length, general appearance of the ear, kernel moisture, and yield. Specific combining ability (SCA) effects were found to be significant for yield and ear damage. Therefore, heterotic patterns among popcorn materials should be taken into account to generate new popcorn hybrids that are not only more productive but also have higher kernel quality. Breeding popcorn populations BSP4APC0 and PSPW1C1 could be base germplasms in a breeding program for obtaining parental inbreds of healthy kernel popcorn hybrids. New inbred lines could be generated from the cross BP1 x BP2 that would have improved GCA and SCA effects for S. nonagrioides resistance when crossed to South American inbreds.  相似文献

2010—2011年以耐荫性较弱的玉米杂交种豫玉22和耐荫性较强的玉米杂交种郑单958为材料,在抽雄前3 d开始进行弱光胁迫处理,吐丝后10 d恢复自然光照,研究弱光胁迫及光恢复对不同耐荫型玉米果穗生长发育及其内源激素含量的影响。结果表明:弱光胁迫下,玉米果穗生长发育明显减缓,穗长、穗粗和果穗干物质积累显著减小,秃尖度变大;穗行数、穗粒数和籽粒库容显著降低;吐丝期果穗顶部小穗子房发育停滞,已有败育迹象的籽粒在恢复自然光照后无明显改善;豫玉22果穗和籽粒性状在处理间的差异程度均大于郑单958。弱光胁迫下,两玉米杂交种果穗的ABA和ZR含量均升高,而GA含量和GA/ABA比值均降低;IAA含量和IAA/ABA比值在郑单958果穗中表现为升高,而豫玉22则表现为下降。  相似文献

Two maize (Zea mays L.) breeding populations with very high concentrations of maysin, a silk-expressed flavone glycoside, were tested for their ability to resist ear damage by the corn earworm, Helicoverpa zea Boddie, under field conditions. Tests were conducted in 2000 and 2001 at multiple locations in Georgia. The high maysin populations, EPM6 and SIM6, as well as resistant and susceptible checks, were scored for silk-maysin content, H. zea damage, and husk characters. In 2000, there was a negative correlation between husk tightness and earworm damage at three of five locations, while there was no significant correlation between damage and maysin content at any location. In 2001, EPM6 and SIM6 had approximately ten times the maysin content of the low-maysin control genotypes; nevertheless, earworm damage to EPM6 and SIM6 was either greater than or not significantly different from the low-maysin genotypes at all locations. The resistant control genotype, Zapalote Chico, had significantly less earworm damage than EPM6 and SIM6 for both years at all locations. The results of this study highlight the importance of identifying and quantifying husk and ear traits that are essential to H. zea resistance in maize.  相似文献

Phytic acid (Myoinositol 1, 2, 3, 4, 5, 6 hexakisphosphate) is a ubiquitous compound present in plants. It is an important constituent in seed reducing the bioavailability of phosphorous and mineral nutrients when fed to monogastric animals like swine, poultry, fish etc. Hence, identification of maize germplasm with reduced phytic acid content is imperative to formulate the breeding programs to evolve low phytate lines. Towards this, three hundred and thirty-eight maize germplasm accessions available at Department of Millets, TNAU, were raised and screened for phytic acid content which varied from 2.77 to 16.70 mg/g of seed. Based on the variability present, a reference set with fifty-eight genotypes for phytic acid was formulated. The reference set was formed with random genotypes selected from the base population to follow a normal distribution (skewness; 0.17, kurtosis; 0.61 and K–S test for normality Dn = 0.70) for phytic acid. The non-significant difference between the means of the base and the reference ensured the entire representation of the base in the formulated reference for phytic acid. Among all the lines in the reference set, the lowest phytic acid content were observed in the lines UMI-113 (2.77 mg/g) followed by UMI-300-1 (3.17 mg/g), UMI-467 (5.50 mg/g) and UMI-158 (6.58 mg/g) could be used as donors for low phytic acid in breeding programs. The principal component analysis for studying the extent of variability in the reference, revealed six major principal components that exhibited 80.40% of variation with flowering traits, ear height and phytic acid as a major contributor for variability. The characters namely plant stand, germination percentage, kernel yield, ear length, ear diameter and number of kernels per row were found to be positively correlated with the phytic acid and this emphasizes the negative pleiotropic effects of low phytic acid lines in germination and seed set. Thus this formulated reference set enables the breeders to handle minimum population for further grouping the genotypes to analyse their heterotic potential combined with low phytic acid.  相似文献

A study was undertaken to determine the ramification of maize shank, cob and kernel tissues by Stenocarpella maydis. Trials consisting of inoculated and uninoculated treatments were planted at two localities. Shank, cob and kernels of each treatment were divided into segments and S. maydis colonization was determined. Infection of the pedicel portion of maize kernels was significantly higher than the apical portion. Preferential colonization of the embryo's of kernels was observed. Colonization of cobs occurred primarily at the attachment end of the cob, with sclerenchymatous tissues showing the greatest re-isolation frequency. Shank segments did not show significant differences in S. maydis re-isolation frequency, although a tendency for higher re-isolations was observed at the stalk-attachment end. It is concluded that S. maydis colonization occurs at the base of the ear with mycelial ramification toward the tip of the ear. The sclerenchyma and placentae were the primary colonized cob tissues. as were the embryos in the kernels.  相似文献

The maize weevil Sitophilus zeamais Motsch. is an important pest of maize that attacks the grain both in the field and during storage. The damage caused by the maize weevil S. zeamais on maize landraces, Amarelo Antigo, Asteca, Caiano, Carioca, and Ferrinho, was evaluated by no-choice tests under laboratory conditions. The commercial varieties Sol da Manh?, BR 106, BR 451, and the synthetics PC 0203 and PC 9903 were evaluated for comparisons with the maize landraces. The parameters evaluated were susceptibility index, number of weevil progeny, development time, weevil progeny dry weight, and grain dry weight loss. The landraces were more susceptible to the maize weevil as compared to the commercial varieties. Based on the cluster analysis, two groups of susceptibility to the maize weevil were observed: one of more susceptible populations formed by local landraces and BR 451, and another less susceptible, with commercial varieties, synthetics, and the landrace Amarelo.  相似文献

Late-season infestations of European corn borer, Ostrinia nubilalis (Hübner), and corn earworm, Helicoverpa zea (Boddie), were sampled to develop binomial sequential sampling plans for larval infestations and damaged kernels in sweet corn, Zea mays L., ears, near harvest. Fields were sampled to obtain a range of larval densities likely to be encountered over a range of infestation levels and field conditions. Binomial sampling plans were developed for O. nubilalis larvae, H. zea larvae, O. nubilalis, and H. zea larvae combined, and for damaged sweet corn kernels. Observed densities ranged from 0.01 to 4.40 larvae per ear for O. nubilalis, 0.005-1.62 larvae per ear for H. zea, and 0.004-36.12 damaged kernels per ear. Results of resampling analyses, based on the proportion of ears infested with one or more larvae, or damaged kernels, indicated an average sample size of 34-37 ears was necessary to classify whether larval infestations, or the incidence of damaged kernels, exceeded 5%. Two operating characteristic curves are presented for each of the four sampling plans. Initial results, with upper bounds of 0.10, and alpha (type I) and beta (type II) error rates at 0.10 and 0.05, respectively, resulted in a 90% probability of making the correct management decision at infestation levels >10%. To improve performance of the sampling plans, we modified the binomial plans by reducing the upper bound to 0.075, while maintaining the same error rates. This plan resulted in a higher probability (>95%) of making the correct management decision to reject a sweet corn load when infestation levels are >10%.  相似文献

Flint maize, Zea mays L., varieties provide some interesting agronomic characteristics and kernels that possess a better ability than other kernels for developing high-quality flour. The pink stem borer, Sesamia nonagrioides Lefebvre, is an important constraint for the maize crop in Mediterranean regions. The objective of this work was to identify a "flint x flint" heterotic pattern that would perform well under artificial infestation by S. nonagrioides. A 10-population diallel was evaluated under infestation by S. nonagrioides in 2 yr. Variety effects were the only significant effects involved in stem and ear resistance to S. nonagrioides attack. Variety effects and average heterosis effects were the only significant effects for grain yield under artificial infestation conditions. Considering variety effects and cross-performance, the heterotic pattern Basto/Enano levantixo x Longfellow (BA/EL x LO) would be recommended for obtaining flint maize hybrids tolerant to S. nonagrioides attack because BA/EL had the most favorable variety effects for stem resistance, LO exhibited the most positive variety effects for grain yield, and the cross BA/EL x LO yielded significantly more than the remaining crosses.  相似文献

Incidence of insect damage, and association of insect damage with mycotoxigenic corn ear molds and mycotoxins was examined in commercial fields of Bt and non-Bt hybrids of different backgrounds in Illinois in 1998 and 1999. Nearly 50% Helicoverpa zea (Boddie) infestation sometimes occurred in Bt hybrids that express high levels of the protein in silks and kernels. Damage by European corn borer, Ostrinia nubilalis Hübner, was uncommon, even in non-Bt ears. Levels of total fumonisins were generally less (15- to 1.8-fold) in Bt versus non-Bt hybrids at the same site, with some significant differences. There were several instances where there were no significant differences in fumonisin levels between low/no Bt kernel hybrids and Bt hybrids that produced high levels of the protein in the kernel and silk tissue. However, significant correlations were often noted between numbers of insect-damaged kernels and total fumonisin levels, especially in 1998, suggesting in these cases that reducing insect damage was still reducing fumonisin levels. There was variability between the correlation coefficient for numbers of insect damaged kernels and fumonisin levels at different sites for the same year, different hybrids at the same site, and the same hybrid for different years. Although reductions in fumonisins in Bt hybrids were more limited than reported in the past, planting the Bt hybrids still appears to be a useful method for indirectly reducing mycotoxins in corn ears.  相似文献

Accumulation of aflatoxin in Spanish peanut kernel samples from different geographical areas in Texas during 1966, as detected by the thin-layer chromatographic method, was relatively low. Analysis of samples obtained from growers using artificial drying equipment (forced air and supplemental heat), when windrow conditions were unfavorable for rapid drying, suggests that this practice reduces the possibility of aflatoxin accumulation. In general, peanuts harvested from land planted to peanuts the previous year were more highly infested with fungi and contained more aflatoxin than peanuts grown on land planted with rye, oats, melons, or potatoes the previous year. Aflatoxin incidence tended to decrease from south to north Texas. These findings verify previous research observations that moist tropical climates are conducive to fungal infestation and aflatoxin accumulation. Detection of aflatoxin in sound mature kernels (kernels screened for minimal size) indicates that the practice of screening for removal of small immature kernels and removal of obviously damaged kernels does not completely eliminate aflatoxin contamination.  相似文献

Deficiency of iron and zinc causes micronutrient malnutrition or hidden hunger, which severely affects ~25% of global population. Genetic biofortification of maize has emerged as cost effective and sustainable approach in addressing malnourishment of iron and zinc deficiency. Therefore, understanding the genetic variation and stability of kernel micronutrients and grain yield of the maize inbreds is a prerequisite in breeding micronutrient-rich high yielding hybrids to alleviate micronutrient malnutrition. We report here, the genetic variability and stability of the kernel micronutrients concentration and grain yield in a set of 50 maize inbred panel selected from the national and the international centres that were raised at six different maize growing regions of India. Phenotyping of kernels using inductively coupled plasma mass spectrometry (ICP-MS) revealed considerable variability for kernel minerals concentration (iron: 18.88 to 47.65 mg kg–1; zinc: 5.41 to 30.85 mg kg–1; manganese: 3.30 to17.73 mg kg–1; copper: 0.53 to 5.48 mg kg–1) and grain yield (826.6 to 5413 kg ha–1). Significant positive correlation was observed between kernel iron and zinc within (r = 0.37 to r = 0.52, p < 0.05) and across locations (r = 0.44, p < 0.01). Variance components of the additive main effects and multiplicative interactions (AMMI) model showed significant genotype and genotype × environment interaction for kernel minerals concentration and grain yield. Most of the variation was contributed by genotype main effect for kernel iron (39.6%), manganese (41.34%) and copper (41.12%), and environment main effects for both kernel zinc (40.5%) and grain yield (37.0%). Genotype main effect plus genotype-by-environment interaction (GGE) biplot identified several mega environments for kernel minerals and grain yield. Comparison of stability parameters revealed AMMI stability value (ASV) as the better representative of the AMMI stability parameters. Dynamic stability parameter GGE distance (GGED) showed strong and positive correlation with both mean kernel concentrations and grain yield. Inbreds (CM-501, SKV-775, HUZM-185) identified from the present investigation will be useful in developing micronutrient-rich as well as stable maize hybrids without compromising grain yield.  相似文献

Aims: To quantify and to compare the occurrence of Fusarium species in maize kernels and stalk pieces, to analyse mycotoxins in kernels and maize crop residues, to evaluate two approaches to obtain kernel samples and to compare two methods for mycotoxin analyses. Methods and Results: The occurrence of Fusarium species in maize kernels and stalk pieces from a three‐year maize hybrid trial and 12 kernel samples from grower’s fields was assessed. Nine to 16 different Fusarium species were detected in maize kernels and stalks. In kernels, F. graminearum, F. verticillioides and F. proliferatum were the most prevalent species whereas in stalks, they were F. equiseti, F. proliferatum and F. verticillioides. In 2006, 68% of the kernel samples exceeded the recommended limit for pig feed for deoxynivalenol (DON) and 42% for zearalenone (ZON), respectively. Similarly, 75% of the samples from grower’s fields exceeded the limits for DON and 50% for ZON. In maize crop residues, toxin concentrations ranged from 2·6 to 15·3 mg kg?1 for DON and from 0·7 to 7·4 mg kg?1 for ZON. Both approaches to obtain maize kernel samples were valid, and a strong correlation between mycotoxin analysis using ELISA and LC‐MS/MS was found. Conclusions: The contamination of maize kernels, stalk pieces and remaining crop residues with various mycotoxins could pose a risk not only to animal health but also to the environment. With the hand‐picked sample, the entire Fusarium complex can be estimated, whereas combine harvested samples are more representative for the mycotoxin contents in harvested goods. Significance and Impact of the Study: This is the first multi‐year study investigating mycotoxin contamination in maize kernels as well as in crop residues. The results indicate a high need to identify cropping factors influencing the infection of maize by Fusarium species to establish recommendations for growers.  相似文献

Grain yield of the maize plant depends on the sizes, shapes, and numbers of ears and the kernels they bear. An automated pipeline that can measure these components of yield from easily‐obtained digital images is needed to advance our understanding of this globally important crop. Here we present three custom algorithms designed to compute such yield components automatically from digital images acquired by a low‐cost platform. One algorithm determines the average space each kernel occupies along the cob axis using a sliding‐window Fourier transform analysis of image intensity features. A second counts individual kernels removed from ears, including those in clusters. A third measures each kernel's major and minor axis after a Bayesian analysis of contour points identifies the kernel tip. Dimensionless ear and kernel shape traits that may interrelate yield components are measured by principal components analysis of contour point sets. Increased objectivity and speed compared to typical manual methods are achieved without loss of accuracy as evidenced by high correlations with ground truth measurements and simulated data. Millimeter‐scale differences among ear, cob, and kernel traits that ranged more than 2.5‐fold across a diverse group of inbred maize lines were resolved. This system for measuring maize ear, cob, and kernel attributes is being used by multiple research groups as an automated Web service running on community high‐throughput computing and distributed data storage infrastructure. Users may create their own workflow using the source code that is staged for download on a public repository.  相似文献
