The Cajal body is an intriguing nuclear structure present in a great variety of plant, animal, and some fungal cells. Recent work on the ribonucleoprotein enzyme telomerase has indicated an unanticipated degree of intranuclear dynamics of both its RNA and protein subunits. The depolarisation-induced influx of 45Ca2+ into anterior pituitary tissue and GH3 cells through 'L'-type, nimodipine-sensitive channels was investigated. In anterior pituitary prisms, phorbol esters, activators of protein kinase C, caused an enhancement of K(+)-induced 45Ca2+ influx. However, in the GH3 anterior pituitary cell line, phorbol esters inhibited K(+)-induced 45Ca2+ influx. The modulation by phorbol esters in both tissues was stereo-specific and time- and concentration-dependent. The diacylglycerol analogue, 1,2-dioctanoyl sn-glycerol was able to mimic the phorbol ester-induced enhancement of calcium influx into anterior pituitary pieces, but was ineffective in GH3 cells. 1,2-Dioctanoyl sn-glycerol may selectively activate an isoform of protein kinase C which is responsible for enhanced 'L'-type Ca(2+)-channel activity.  相似文献

Selten (1980, J. theor. Biol. 84, 93(N)/01) has shown that mixed strategies cannot be evolutionarily stable in asymmetric games. Because every interaction features some asymmetry, this result apparently precludes mixed strategies in an evolutionary setting. In Maynard Smith's Hawk-Dove game (1982, Evolution and the theory of games (UP-Cambridge), for example, Selten's result restricts attention to pure-strategy evolutionarily stable outcomes in which the animals use the ability to condition their actions on asymmetries to coordinate, with one playing Hawk and one playing Dove, and with conflicts in which both animals play Hawk never arising. This result contrasts with the intuition that the mixed equilibrium of the Hawk-Dove game captures important aspects of many animal interactions, including the possibility of conflict. In this paper, we follow Eshel and Sansone (1995, J. theor. Biol. 177, 341-356) in enriching Selten's model to incorporate an important aspect of animal interactions, namely that payoffs and asymmetries may both be imperfectly observed. In the richer model, we find conditions under which effectively mixed strategies are stable in asymmetric games, as well as conditions under which they are not stable. Behavior will be conditioned on asymmetries, leading to pure-strategy equilibria in which conflict is avoided, when there are relatively large, observable asymmetries and small observable variations in payoffs. Under opposite conditions, evolutionarily stable equilibria will appear that are effectively mixed, including the potential for conflict.  相似文献

Inulin, EDTA, or maltose were given intragastrically to mice, either free or entrapped in mono-shell liposomes of egg phosphatidylcholine or the non-digestible diether and dialkyl analog of phosphatidylcholine. The method of reconstructing quantum bumps in photoreceptor cells from noise data by making use of shot noise theory is critically reviewed. The application of this method produces results irrespective of whether the conditions for reconstructing bumps by the method are satisfied or not and even irrespective of whether at high stimulus intensities quantum bumps exist or not. We argue that at high intensities the concept of quantum bumps indeed becomes physically meaningless and degenerates to a purely mathematical concept. In order to investigate the meaning of the results of the reconstruction method, we submit it to a test model for which bumps and single channel opening events can be evaluated analytically. By comparing the analytical results of the test model with that of the reconstruction method applied to the test model we find: (1) even at low intensities, the reconstructed bump values deviate from the analytical results by up to an order of magnitude due to the variability of the bumps, (2) at high intensities, the reconstruction method produces single chennel opening events rather than anything like a quantum bump. We also find, however, that there is no continuous transition from a bump at low intensities to a single channel event at high intensities.  相似文献

In the microcirculation, longitudinal conduction of vasomotor responses provides an essential means of coordinating flow distribution among vessels in a complex network. Spread of current along the vessel axis can display a regenerative component, which leads to propagation of vasomotor signals over many millimeters; the ionic basis for the regenerative response is unknown. We examined the responses to 10 s of focal electrical stimulation (30 Hz, 2 ms, 30 V) of mouse cremaster arterioles to test the hypothesis that voltage-dependent Na(+) (Na(v)) and Ca(2+) channels might be activated in long-distance signaling in microvessels. Electrical stimulation evoked a vasoconstriction at the site of stimulation and a spreading, nondecremental conducted dilation. Endothelial damage (air bubble) blocked conduction of the vasodilation, indicating an involvement of the endothelium. The Na(v) channel blocker bupivacaine also blocked conduction, and TTX attenuated it. The Na(v) channel activator veratridine induced an endothelium-dependent dilation. The Na(v) channel isoforms Na(v)1.2, Na(v)1.6, and Na(v)1.9 were detected in the endothelial cells of cremaster arterioles by immunocytochemistry. These findings are consistent with the involvement of Na(v) channels in the conducted response. BAPTA buffering of endothelial cell Ca(2+) delayed and reduced the conducted dilation, which was almost eliminated by Ni(2+), amiloride, or deletion of alpha(1H) T-type Ca(2+) (Ca(v)3.2) channels. Blockade of endothelial nitric oxide synthase or Ca(2+)-activated K(+) channels also inhibited the conducted vasodilation. Our findings indicate that an electrically induced signal can propagate along the vessel axis via the endothelium and can induce sequential activation of Na(v) and Ca(v)3.2 channels. The resultant Ca(2+) influx activates endothelial nitric oxide synthase and Ca(2+)-activated K(+) channels, triggering vasodilation.  相似文献

AMPA receptor ion channels are of paramount importance for postsynaptic excitation. Several reports demonstrate that AMPA receptors are present in the presynaptic compartment and point to a role of these receptors in the modulation of presynaptic function. We discuss here the possibility that not only ion influx through the receptor, but also biochemical cascades, activated by ligand binding and independent from ion flux, might contribute to AMPA mediated presynaptic modulation.  相似文献

Because the morphogenesis of biological systems is not fully understood, researches from various points of view are necessary. The present author has recently made computer simulations with his colleagues to construct branching systems of human organs, such as the lung airway and the liver blood vessels. In the simulations certain rules are assumed to govern bifurcating processes of the systems. These rules are expressed in terms of physical and geometrical concepts, such as minimum energy consumption and uniform filling of branches in the space of organs. Results of computer simulation are quite similar to real structures. However, actual mechanisms of morphogenesis, i.e. effects of genes or proteins, are not considered in these studies. In this article, the present work is discussed in relation to the concept of biological pattern formation by Meinhardt and a recent study by Miura and Shiota on lung growth.  相似文献

Epidermal growth factor receptor (EGFR) is highly expressed in certain cancer types and is involved in regulating the biological characteristics of cancer progression, including proliferation, metastasis, and drug resistance. Various medicines targeting EGFR have been developed and approved for several cancer types, such as lung and colon cancer. To date, however, EGFR inhibitors have not achieved satisfactory clinical results in breast cancer, which continues to be the most serious malignant tumor type in females. Therefore, clarifying the underlying mechanisms related to the ineffectiveness of EGFR inhibitors in breast cancer and developing new EGFR-targeted strategies (e.g., combination therapy) remain critical challenges. Various studies have demonstrated aberrant expression and maintenance of EGFR levels in breast cancer. In this review, we summarize the regulatory mechanisms underlying EGFR protein expression in breast cancer cells, including EGFR mutations, amplification, endocytic dysfunction, recycling acceleration, and degradation disorders. We also discuss potential therapeutic strategies that act directly or indirectly on EGFR, including reducing EGFR protein expression, treating the target protein to mediate precise clearance, and inhibiting non-EGFR signaling pathways. This review should provide new therapeutic perspectives for breast cancer patients with high EGFR expression.  相似文献

Conservation of species in agroecosystems has attracted attention. Irrigation channels can improve habitats and offer conditions for freshwater species conservation. Two questions from biodiversity conservation point of view are: 1) Can the irrigated channels maintain a rich diversity of macrophytes, macroinvertebrates and amphibians over the cultivation cycle? 2) Do richness, abundance and composition of aquatic species change over the rice cultivation cycle? For this, a set of four rice field channels was randomly selected in Southern Brazilian wetlands. In each channel, six sample collection events were carried out over the rice cultivation cycle (June 2005 to June 2006). A total of 160 taxa were identified in irrigated channels, including 59 macrophyte species, 91 taxa of macroinvertebrate and 10 amphibian species. The richness and abundance of macrophytes, macroinvertebrates and amphibians did not change significantly over the rice cultivation cycle. However, the species composition of these groups in the irrigation channels varied between uncultivated and cultivated periods. Our results showed that the species diversity found in the irrigation channels, together with the permanence of water enables these man-made aquatic networks to function as important systems that can contribute to the conservation of biodiversity in regions where the wetlands were converted into rice fields. The conservation of the species in agriculture, such as rice field channels, may be an important alternative for biodiversity conservation in Southern Brazil, where more than 90% of wetland systems have already been lost and the remaining ones are still at high risk due to the expansion of rice production.  相似文献
