Two experiments were conducted during mid-gestation to examine effects in ewes of propylthiouracil (PTU) treatment alone or with melatonin on serum thyroid hormones, postpartum reproduction, and lamb performance. In the first experiment, beginning on day 0 (first day of treatment when all animals were 72.2+/-0.9 days of gestation), ewes received daily treatments (gavage) consisting of either 0mg (n=6) or 40 mg (n=6) PTU/kg body weight/day for 15 days. After 15 days, the 40 mg dosage was decreased to 20mg/kg body weight for an additional 20 days (35 days of PTU). Serum thyroxine (T4) did not differ (P>0.10) between groups through day 4; but on day 5, control females had a serum value of 67 ng/ml compared with 46 (+/-5)ng/ml for PTU-treated ewes (P=0.02). On the last day that 40 mg of PTU was administered, serum T4 averaged 67 and 7 (+/-5)ng/ml (P<0.001) in the two respective groups. Serum T4 remained low and was 80 and 1 ng/ml (P<0.001) in control and treated ewes on day 34. Serum T4 rose gradually after PTU but remained different from that observed in control ewes through day 48. Lambs from control and treated ewes had similar (P=0.46) T4 values at birth but lambs from PTU-treated ewes had lower (P=0.03) birth weights than did those from control ewes. Serum progesterone (P4) after parturition indicated a lack of cyclicity in all ewes. In the second experiment, beginning on day 0 (76.8+/-4.7 days of gestation), ewes received PTU as in Experiment 1. In addition, after 15 days of PTU, melatonin was given (i.m. injections at 5mg/day) for 30 days. Propylthiouracil decreased (P0.60) for lambs born to control and treated ewes. Female offspring of PTU+melatonin-treated dams reached puberty, became anestrus, and returned to cyclicity at similar (P>0.10) times to contemporary ewe lambs. Results indicate that 40/20mg PTU alone or with melatonin does not induce cyclicity after lambing in spring lambing ewes and has little effect on offspring performance.  相似文献

An experiment was conducted to determine whether natural royal jelly (RJ) paste administered orally or intramuscularly (i.m.) in conjunction with exogenous progesterone is associated with improved reproductive responses in ewes. Thirty 3-6-year-old Awassi ewes were randomly allocated into three (RJ-capsule, RJC; RJ-injection, RJI and control, CON) groups of 10 ewes each. All ewes were treated with intravaginal progesterone sponges for 12 days. Ewes in the RJC and RJI were administered orally or i.m. with a total of 3g of RJ given in 12 equal doses of 250 mg per ewe per day starting at the time of sponge insertion. At the time of sponge withdrawal (day 0, 0 h), ewes were exposed to three rams and checked for breeding marks at 6-h intervals for 3 days. Blood samples were collected from all ewes for analysis of progesterone concentrations. Pretreatment progesterone levels were <0.5 ng x ml(-1) in 16/30 and >1.3 ng x ml(-1) in the remaining ewes indicating luteal function and cyclicity. Similar reproductive responses and progesterone levels occurred in ewes of the RJC and RJI; therefore, data of the two groups were pooled. Following sponge insertion, progesterone levels increased rapidly and reached maximum values of 5.8+/-0.2 ng x ml(-1) within 2 days among ewes of the three groups, and then declined gradually to day 0 values of 1.6+/-0.1 and 1.9+/-0.1 ng x ml(-1) for the RJ-treated and CON ewes, respectively. The rate of progesterone decline was greater (P<0.001) in RJ-treated than in CON. Mean progesterone levels during the 12-day period were lower (P<0.001) in RJ-treated than in CON (2.8+/-0.2 ng x ml(-1) versus 3.3+/-0.2 ng x ml(-1)). Treatment with RJ resulted in greater (P<0.05) incidence of oestrus and shorter (P<0.05) intervals to onset of oestrus than CON. Based upon progesterone levels, ovulation occurred following day 0 in all ewes. Progesterone increased on day 3 in RJ-treated and on day 4 in CON ewes. Progesterone remained elevated through day 18 in 8/20 RJ-treated and 1/10 CON ewes (P=0.09). All pregnant ewes exhibited oestrus 14 h earlier (P<0.02), ovulated approximately 1 day earlier and had higher (P<0.001) luteal phase progesterone levels than non-pregnant ewes. Non-pregnant had higher (P<0.04) body weights than pregnant ewes. In conclusion, results demonstrate that both RJ treatments in conjunction with exogenous progesterone were equally capable of improving oestrus response and pregnancy rate.  相似文献

In the ewe, high level feeding is associated with low levels of circulating progesterone and that progesterone withdrawal is a prerequisite for lactogenesis and the onset of copious milk secretion. A total of 108 ewes was allocated to a 3 x 2 factorial experiment to determine the effect of diet on serum progesterone (P4) concentration and colostrum yield. On D 96 of pregnancy, the ewes were offered either grass silage, molassed sugar beet pulp (beet pulp) silage or grass silage supplemented with beet pulp. All ewes were given ad libitum access to silage. From D 126 of pregnancy, half the diets were supplemented with soybean meal with the aim of achieving a total crude protein intake of 220 g/ewe/day. Blood samples were taken on D 142 of pregnancy and at 1, 10 and 18 h post lambing for P4 determination. Metabolizable energy intakes (Mega Joules/ewe/day) of 9.1, 11.2 and 12.1 (SEM 0.37) and crude protein intakes (g/ewe/day) of 142.8, 167.4 and 162.0 (SEM 3.46) were recorded for grass silage, beet pulp ensiled and beet pulp supplemented silages, respectively, from D 126 to D 147 of pregnancy. Soybean meal supplementation increased the crude protein intake from 97.1 to 217.0 g/ewe/day (SEM 2.86) and Metabolisable Energy intake from 9.0 to 12.6 Mega Joules/ewe/day (SEM 0.31). Supplementation with soybean meal decreased P4 concentration (ng/ml) on D 142 (P < 0.05) and at 1 h (P < 0.01), 10 h (P < 0.01) and 18 h post lambing (P < 0.01). Ewes offered grass silage had higher P4 concentrations on D 142 (P < 0.01) and at 1 h after lambing (P < 0.05) than the beet pulp-supplemented ewes. There were negative linear relationships between P4 concentration at D 142 and colostrum yield at 1 h (P < 0.01) and between P4 concentration at 1 h and colostrum yield at 1 h (P < 0.001) and total yield at 18 h (P < 0.001). In conclusion, ewes which were underfed had higher P4 concentrations at all times, and there were negative linear relationships between colostrum yield and P4 concentrations on D 142 of pregnancy and at 1 h post lambing.  相似文献

The effect of progesterone and prostaglandin administration on the timing of farrowing was studied in three groups of 25 sows each. Progesterone treatment (100 mg/day) on days 112, 113 and 114 of gestation (group I) significantly prolonged the gestation length to 116.4 +/- 0.4 (mean +/- s.e.) days compared to the control sows (group III; 115.5 +/- 0.2; P less than 0.05). Administration of prostaglandin (200 micrograms Cloprostanol intramuscularly) on day 115 of gestation following progesterone treatment (group II) resulted in a gestation length of 116.0 +/- 0.1 days, with the sows farrowing 25.4 +/- 1.0 h after the prostaglandin injection. 80% of the sows farrowed between 0800 and 1700 h of day 116 of gestation. Plasma progesterone levels were maintained by the exogenous progesterone during treatment. At farrowing, higher levels of progesterone were observed in groups I and II compared to controls. Prostaglandin treatment did not significantly alter withdrawal of progesterone in progesterone treated sows, suggesting that the actions of exogenous prostaglandin is primarily on the myometrium and the cervix. Hormonal treatment in late pregnancy did not have any adverse effects on piglet viability and growth rate, or subsequent reproductive performances of sows. Lactation was initiated normally, and the concentrations of lactose, protein, fat, IgG, Na+, Ca2+ and K+ in colostrum and milk were similar in all groups during the first 5 days of lactation.  相似文献

Effects of estradiol benzoate and prostaglandin F(2alpha) (PGF(2alpha)) on concentrations of progesterone and estrogen in serum and the percentage of live births were determined in 21 gilts treated with exogenous progesterone in late gestation. All gilts received progesterone (25mg s.c. four times daily) from Days 108 through 113 of gestation. Gilts receiving no other treatments (controls) had elevated levels of progesterone through 1800 h on Day 114 (29.2 +/- 11.4 ng/ml) and farrowed at 115.6 +/- 0.3 d of gestation with a relatively low percentage of live births (66.8 +/- 17.3). Gilts treated with PGF(2alpha) administered at 0600 h on Day 114 had less (P<0.01) progesterone by 1800 h (7.0 +/- 1.3 ng/ml) relative to that of the controls, but they had similar gestation lengths (115.5 +/- 0.3) and percentages of live births (50.0 +/- 16.2). Administration of estradiol benzoate (10 mg) to gilts at 0600 h on Day 114 did not reduce progesterone on Day 114 or the gestation length relative to that of the control gilts, but it did increase (P<0.05) the percentage of live births (100%).  相似文献

Using autumn-lambing ewes, this study investigated (i) the effects of diet on gonadotrophin secretion and responsiveness of the hypothalamic-pituitary-ovarian axis to exogenous GnRH during the early post-partum period; and (ii) whether ovulation prior to completion of uterine involution results in an increased incidence of aberrant ovarian cycles. Thirty-two ewes rearing 1.9+/-0.12 lambs were equally allocated to two dietary treatments at lambing (22 October +/-0.2 day). Diets comprised ad libitum hay and 1.5 kg per ewe per day of one of two concentrates (11.5 MJ ME, 195 g CP per kg) containing 300 g kg(-1) cracked maize grain (M) or 300 g kg(-1) sugar beet pellets (S). Half of the ewes on each diet (G) received 25 i.v. injections of 250 ng GnRH in 2 ml 0.9% saline at 2 h intervals from days 12-14 post-partum while remaining ewes (N) were monitored for the resumption of spontaneous ovarian cyclicity. Blood samples were obtained from all ewes throughout the study (lambing to 18 December) for measurement of circulating hormone concentrations and the uteri and ovaries of all ewes were examined via laparoscopy on day 21 post-partum. There were no effects of dietary treatment on ewe daily live weight loss, lamb daily live weight gain or the immediate post-partum increase in circulating FSH concentrations. Diet did not affect insulin concentrations or LH pulse frequency on day 12 post-partum but LH pulse amplitude was lower in ewes fed concentrate M compared to concentrate S (1.4+/-0.10 versus 1.7+/-0.12 ng ml(-1), respectively, P<0.05) and this was associated with an increased interval to the resumption of spontaneous ovarian cycles (35+/-3.1 versus 26+/-2.1 day, respectively, P<0.05). Administration of exogenous GnRH increased (P<0.05) the proportion of ewes on both diets that ovulated within 20 days of parturition and advanced the onset of ovarian cyclicity in ewes fed concentrate M by 9.5 days (significance of interaction, P<0.05). Four ewes, all of which ovulated before day 22 post-partum, had extended luteal activity while in remaining ewes, duration of the first luteal phase was inversely related to the time of first ovulation (r(2)=0.16, P<0.05). Results demonstrate that (i) the onset of ovarian cyclicity is influenced by diet and can be advanced by administration of exogenous GnRH; and (ii) ovulation during the early post-partum period is associated with an increased incidence of extended luteal activity.  相似文献

The aim of the present study was to examine the effect of steroid-free bovine follicular fluid (bFF) on both ovulation and lambing rates. For this purpose, 30 adult ewes of the Karaguniki breed were randomly allocated to three treatment groups (A,B and C; n=10 ewes each) during the breeding season of 1988. The ewes in Group A received bFF (6 ml iv) twice daily during their luteal phase, starting on Day 5 and lasting until Day 9. The ewes in Group B received a mixture of bFF/arachid oil (3 ml sc, 2:1) on Days 3, 4, 5, 10 and 12 of the estrous cycle. The ewes in Group C (Controls) were treated subcutanecusly with a mixture of steroid-free bovine plasma and arachid oil (2:1) on the same days as the ewes in Group B. Plasma concentrations of progesterone showed that the luteal function during the treatment cycle was normal in all treated and control ewes. The ovulation and lambing rates, however, were greater in Group A (2.5 +/- 0.2 and 1.9 +/- 0.3, respectively) and in Group B (2.1 +/- 0.2 and 1.6 +/- 0.1, respectively) than in Group C (1.5 +/- 0.2 and 1.2 +/- 0.3, respectively). This study was carried out to determine ovine pregnancy-associated glycoprotein (oPAG) and progesterone (P4) levels in the serum of Churra and Merino ewes throughout gestation and the first month post partum. The oPAG levels were determined with an heterologus RIA using bovine PAG as standard and tracer and rabbit antiserum against oPAG, sensitivity was 4.0 ng/ml. The P4 levels were measured with a radioimmunological procedure, including a specific extraction step with petroleum ether (bp 60-80 degrees C) with a sensitivity of less than 0.1 ng/ml. There were no differences (P<0.10) in the oPAG profile between breeds from Weeks 1 to 18. From Week 18 to lambing, oPAG concentrations increased rapidly in Churra ewes (on average, from 250 to 650 ng/ml) while remaining relatively constant in the Merino ewes (around 250 ng/ml). No significant differences (P>0.05) were observed for mean weekly P4 levels between the 2 breeds. In both breeds, P4 increased throughout the whole length of gestation, with the highest level measured at Weeks 19-20, then declined 2 wk before parturition. No correlation was found between P4 and oPAG concentrations during gestation in either of the breeds. After lambing, oPAG and P4 levels decreased rapidly in 4 wk to basal values. In both breeds the oPAG concentrations at Weeks 19, 20 and 21 of gestation in ewes carrying male fetuses were higher than in those carrying female fetuses. From the results, we conclude that the breed and sex of the fetus could influence the production of oPAG.  相似文献

Fifteen ovariectomized ewes were treated with implants (s.c.) creating circulating luteal progesterone concentrations of 1.6 +/- 0.1 ng ml-1 serum. Ten days later, progesterone implants were removed from five ewes which were then infused with saline for 64 h (0.154 mol NaCl l-1, 20 ml h-1, i.v.). Ewes with progesterone implants remaining were infused with saline (n = 5) or naloxone (0.5 mg kg-1 h-1, n = 5) in saline for 64 h. At 36 h of infusion, all ewes were injected with oestradiol (20 micrograms in 1 ml groundnut oil, i.m.). During the first 36 h of infusion, serum luteinizing hormone (LH) concentrations were similar in ewes infused with saline after progesterone withdrawal and ewes infused with naloxone, but with progesterone implants remaining (1.23 +/- 0.11 and 1.28 +/- 0.23 ng ml-1 serum, respectively, mean +/- SEM, P greater than 0.05). These values exceeded circulating LH concentrations during the first 36 h of saline infusion of ewes with progesterone implants remaining (0.59 +/- 0.09 ng ml-1 serum, P less than 0.05). The data suggested that progesterone suppression of tonic LH secretion, before oestradiol injection, was completely antagonized by naloxone. After oestradiol injection, circulating LH concentrations decreased for about 10 h in ewes of all groups. Reproductive performance and fetal growth was determined in GnRH (4 microg synthetic GnRH agonist, Receptal) administered (i.m.) to ewes on day 12 post-mating (n = 103) compared to control ewes (n = 97) during the breeding season. Plasma progesterone and LH concentrations were analyzed. A total of 13 ewes was slaughtered on day 45 of pregnancy (six from control, seven from GnRH treated groups). GnRH administration on day 12 post-mating increased plasma progesterone concentration (4.39+/-0.25 ng/ml) compared to control group (3.43+/-0.15 ng/ml) on days 13-15 post-mating (P < 0.01). GnRH administration also increased plasma LH concentration between 1 and 4 h after GnRH administration (P < 0.01). Pregnancy rate was higher in GnRH treated group (84%) than control (66%) group (P < 0.05). The ewes in GnRH administered group had more twins (P < 0.05) than those in control group. The ovarian weights (P < 0.05) and the number of corpora lutea (CL) (P < 0.01) were greater in ewes slaughtered on day 45 of pregnancy in GnRH treated group than those in control group. GnRH administration on day 12 post-mating did not have any effect on products of conception at day 45 of pregnancy except on crown-rump length (CRL) of fetuses and cotyledon weight. CRL of fetuses and cotyledon weight in GnRH treated group was higher than those in control group (P < 0.05). In conclusion GnRH administration improved reproductive performance of ewes when administered on day 12 post-mating probably through its beneficial effect on embryo survival by enhancing luteal function, but not through stimulating fetal growth.  相似文献

The objective was to compare the effects of royal jelly (RJ) and eCG treatments on reproductive performance of ewes synchronized using intravaginal progesterone-releasing devices. Forty-two cycling Awassi ewes were treated intramuscularly (i.m.) with 15 mg PGF2alpha. On the following day, all ewes were administered with CIDR-G for 12 days and were randomly allocated to three (RJ, eCG and control) groups of 14 ewes each. Ewes in the RJ-treated group received daily i.m. treatments of 400mg RJ during the period of CIDR-treatment. Each ewe in the eCG-treated group received an i.m. treatment of 500 IU eCG at the time of CIDR-G removal (day 0) and no further treatment was given to ewes in the control group. Ewes were exposed to four fertile rams for 72 h, from the time of CIDR-G removal, and checked for breeding marks at 6-h intervals. Blood samples were collected from day -13 until day 0 and thereafter until day 19 for progesterone analysis. Royal jelly treatment resulted in a greater rate of decline and lower (P<0.02) progesterone concentrations between days -10 and 0 than eCG-treated and control ewes. Expression of estrus was similar among the three groups and intervals to onset of estrus were shorter (P<0.01) in RJ-treated (31.3h) and eCG-treated (29.8h) than control (41.3h) ewes. First-cycle pregnancy and lambing rates were greater (P<0.05) in RJ-treated (71.4 and 71.4%) and eCG-treated (85.7 and 78.6%) than in control (42.9 and 35.7%) ewes, respectively. Results demonstrate that the treatments of RJ and eCG in conjunction with CIDR-G were similarly effective in induction of estrus and improvement of pregnancy and lambing rates.  相似文献

Two practical regimens designed to induce estrus and ovulation in ewes in late anestrus were compared. Forty ewes were given a soluble glass rumen bolus containing 150 mg melatonin on July 9 and were joined with two vasectomized rams on July 23 and with three fertile rams on August 6. A second group of 40 ewes was treated with an intravaginal progestagen pessary (60mg medroxy-progesterone acetate) on July 23. Following pessary removal after 12 d, ewes were given 750 IU of pregnant mare serum gonadotropin (PMSG). Five fertile rams were joined with these ewes 48 h after progestagen removal. Melatonin concentrations were determined in single blood samples collected in early afternoon of July 21. Mating dates, lambing dates and litter sizes were recorded. Date of mating was significantly later in ewes treated with melatonin compared with those treated with progestagen plus PMSG (P<0.0001). All ewes given melatonin were mated within 4 wk, and those on progestagen plus PMSG treatment within one day of fertile ram introduction. Thirty-four ewes (85%) allocated to melatonin treatment and 36 (90%) allocated to progestagen plus PMSG treatment lambed (P>0.05). Mean (+/-SEM) lambing date was later in melatonin-treated ewes (January 17+/-1.2 d) compared to those given progestagen plus PMSG (December 30+/-0.6 d; P<0.0001). Mean litter size was lower in melatonin-treated ewes (1.5+/-0.1) compared with those given progestagen plus PMSG (2.0+/-0.1; P<0.001). Plasma melatonin concentrations indicated that 9 of 40 ewes treated with melatonin had circulating melatonin concentrations of less than 16 pg/ml. It is concluded that under conditions that existed in this experiment, treatment with progestagen plus PMSG in late anestrus resulted in more synchronous mating and lambing and a higher litter size than that following administration of a soluble glass rumen-degradable bolus containing melatonin.  相似文献

Between June 1 and August 24 of two successive summers, Targhee ewes (n = 64) and Finn x Targhee ewes (n = 44) were subjected to different photoperiods. Treatments were natural day length (ND), eight hours light: 16 hours of dark (8L:16D), 16L:8D shifted to 8L:16D (16L:8D-8L:16D) and seven hours of light:nine hours of dark:one hour of light:seven hours dark (7L:9D:1L:7D). Days to the mean first breeding mark were shortest (P<0.05) for the 16L:8D-8L:16D group (32 days) followed by the 7L:9D:1L:7D group (39 days). The longest mean intervals to marking were 52 (ND) and 47 days (8L:16D). Time to mean first breeding mark was 18 days shorter (P<0.05) in 1981 than in 1982. Mean serum progesterone values were 0.05) among treatments through week 6. The 8L:16D group had the most rapid rise and the highest terminal progesterone value, while the ND group had the slowest rise and lowest terminal value. Mean serum prolactin values in general started high and decreased over time. This decrease in all treatments preceded the rapid rise in progesterone. The most rapid prolactin decline was for the 8L:16D group. The percentage of exposed ewes lambing was lowest (P<0.05) for ND ewes (37%) and highest for 8L:16D ewes (81%). The shortest mean interval from the start of treatment to lambing was 210 days (7L:9D:1L:7D) followed by 214 days (8L:16D), 217 days (ND) and 218 days (16L:8D-8L:16D). Considering percentage lambing in combination with interval to lambing, the 8L:16D treatment was the most effective treatment.  相似文献

To determine whether progesterone facilitates PGF(2)alpha-induced luteolysis prior to day 5 of the estrous cycle, 48 Holstein-Friestian heifers were assigned at random to four treatments: 1) 4 ml corn oil/day + 5 ml Tris-HCl buffer (control); 2) 25 mg prostaglandin F(2)alpha (PGF(2)alpha); 3) 100 mg progesterone/day (progesterone); 4) 100 mg progesterone/day + 25 mg PGF(2)alpha (combined treatment). Progesterone was injected subcutaneously daily from estrus (day 0) through day 3. The PGF(2)alpha was injected intramuscularly on day 3. Estrous cycle lengths were decreased by progesterone: 20.2 +/- 0.56, 19.2 +/- 0.31 (control and PGF(2)alpha); 13.2 +/- 1.40, and 11.7 +/- 1.27 (progesterone and combined). The combination of progesterone and PGF(2)alpha did not shorten the cycle any more than did progesterone alone (interaction, P>0.05). PGF(2)alpha treatment reduced progesterone concentrations on day 6 (P<0.05) and both progesterone and PGF(2)alpha reduced plasma progesterone on day 8 (P<0.01 and P<0.05, respectively). LH was measured in blood samples collected at 10- min intervals for 4 hr on day 4 from three heifers selected at random from each of the four treatment groups. Mean LH concentration for control heifers ranged from 0.35 to 0.63 ng/ml (overall mean, 0.49 ng/ml) and for progesterone-treated heifers ranged from 0.12 to 0.30 ng/ml (overall mean, 0.23 ng/ml). LH concentrations were greater in control heifers (P<0.01). The mean LH pulse rate for control heifers was 2.7 pulses/heifers/4 hr, while that for the progesterone-treated heifers was 1.7 pulses/heifer/4 hr. The mean pulse amplitude for control and progesterone treatments was 0.47 ng/ml and 0.36 ng/ml, respectively. Neither pulse amplitude nor frequency were different between treatment groups.  相似文献

Twenty days before a regular fall breeding season, 93 mature Rambouillet ewes were randomly allotted to one of two groups to examine the response of cycling females exposed to sterile rams. Six vasectomized rams were joined with 46 ewes during the 20-day period while 47 ewes remained separate from the rams. All ewes were judged to have been cycling either by paint marks from rams on treated animals or by the cyclic nature of progesterone (sampled at four-day intervals) profiles in control ewes. After removal of sterile males, fertile Debouillet rams remained with the ewes during a 34-day breeding season. Approximately six weeks before beginning the lambing season, one-half the females in each sterile ram treatment group were forced to walk 0.8 km per day to examine the influence of exercise on subsequent reproductive performance. Presence of sterile males did not alter (P>0.10) lambing rate, average lambing date or percentage of ewes lambing during thirds of the lambing season suggesting that prebreeding exposure of cycling ewes to vasectomized rams does not enhance lambing rate or shorten the lambing season. Forced exercise resulted in increased daily feed consumption which was reflected in heavier (P<0.05) lamb birth weights. Percentage of ewes experiencing either dystocia or pregnancy toxemia was similar (P>0.20) in exercised and unexercised animals. Fifty Corriedale ewes were used in this study to evaluate pregnancy diagnosis in sheep. Ewes were bred under a pen mating system and pregnancy diagnosis was initiated from day 15 post-mating, applying the diagnostic techniques of trans-abdominal real-time B-mode ultrasonography, Preg-alert (A-mode ultrasonography), the Doppler ultrasonic fetal pulse detector or the plasma progesterone concentration assay (EIA). These tests were repeated fortnightly on all the ewes until the onset of lambing. The accuracy of trans-abdominal real-time B-mode ultrasonography (68%) at days 15–30 of pregnancy increased to 100% by days 61–75 and remained constant until lambing. The accuracy of the Preg-alert (56%) diagnosis at days 31–45 increased to 94% by days 91–105 of gestation and then decreased to 82% from days 136 of gestation to lambing. The accuracy of both the Doppler ultrasound (56%) at days 31–45 and plasma progesterone assay (98%) at days 15–30 of gestation increased to 100% at days 76–90 and 46–60 of gestation, respectively and remained constant until parturition. The mean plasma progesterone concentration at days 0–6 (1.41 ± 0.21 ng/ml) increased to 4.0 ± 0.87 ng/ml at days 16–30 (days 18.23 ± 0.78) post-mating. Animals returning to estrus recorded less than 1 ng/ml at days 18.23 ± 0.78 post-mating. The accuracy of both the B-mode ultrasonic technique (78%) and plasma progesterone assay (98%) was significantly higher (P < 0.05) than the accuracy obtained with the A-mode and Doppler ultrasound (both 56%) at days 31–45 of gestation. The study concluded that real-time B-mode ultrasonography is the earliest, most accurate, safest, fastest and most economical method of pregnancy diagnosis in sheep at farm level. The A-mode and Doppler methods can also be used under field conditions, where sophisticated laboratory facilities are not available. Plasma progesterone assays (EIA) can be used as a means of early pregnancy diagnosis in organized sheep farms with fair accuracy.  相似文献

This experiment tested the hypothesis that a lift in the nutrition of ewes, before lambing, to increase colostrum production would enhance lamb survival. In all, 261 mature Corriedale ewes, each with a single fetus from a synchronised mating, grazed native pasture to day 130 after mating; at which point they were weighed, condition scored and allocated to graze either native pasture or a pasture dominant with Lotus uliginosus. Five days later (14 days before the expected start of lambing) the ewes were allocated to one of four treatments and fed: (i) native pasture alone, (ii) native pasture plus a commercial high-energy lick, (iii) L. uliginosus pasture alone or (iv) L. uliginosus pasture plus whole maize. The weight, viscosity and concentration of components and immunoglobulin G in the colostrum that had accumulated at parturition, were measured for 10 ewes in each treatment. The lambs that survived to 20 days of age from the 221 ewes that were not milked, were recorded. The ewes supplemented with the lick or maize grain and those that grazed the L. uliginosus pasture alone accumulated two to three times more colostrum at birth than the ewes that grazed native pasture alone (396, 635 and 662 g v. 206 g; P < 0.01). The colostrum from the ewes that grazed only native pasture was more viscous (lower score) than that from the ewes supplemented with the lick or maize grain or the ewes that grazed the L. uliginosus pasture alone (scores of 4.1 v. 6.2, 6.5 and 6.4, P < 0.001) and, not surprisingly, the concentration of lactose in the colostrum of the ewes fed only native pasture was also much lower (1.1% v. 3.0%, 2.8% and 2.6%; P < 0.001)he survival of lambs from the ewes fed only native pasture was less than that of the lambs from ewes fed native pasture plus the commercial lick (81.8% v. 95.5%; P < 0.05) or the L. uliginosus pasture alone (92.4%, P < 0.05), and also tended to be lower than that for lambs born to ewes fed L. uliginosus pasture plus maize (91.8%, P = 0.08). The concentration of glucose in the blood of the lambs from the ewes that grazed only native pasture was lower than that of the other lambs (42.1 v. 60.2 ng/ml, P = 0.012). We conclude that the marked increase in colostrum production associated with the lift in ewe nutrition, just prior to lambing, enhanced lamb survival.  相似文献

Bovine somatotropin (bST) enhances ovarian follicular and embryonic development in sheep and cattle. In the present study, the objective was to assess whether bST given 5 days before the end of progestin-based estrous synchronization improves prolificacy and lambing rate in sheep. Pelibuey ewes (n=92) exhibiting estrous cycles at regular intervals received an intravaginal sponge containing 45mg of FGA for 12 days. Five days before sponge withdrawal, ewes were treated with either 125mg of bST sc (bST group; n=47) or saline solution (control; n=45). After the sponge was removed, ewes were observed for estrus and subsequently mated twice. Lambing rate and prolificacy was determined at birth. Blood samples were taken from the time of treatment until day 15 after estrus in eight ewes from the bST group and nine from the control group. Concentrations of IGF-I were determined by immunoradiometric assay and progesterone by RIA. Treatment with bST increased (P<0.01) the proportion of ewes with more than one lamb (bST, 56% compared with control, 26%) and prolificacy (bST, 1.6 compared with control, 1.3). Treatment with bST increased (P<0.05) the lambing rate of multiparous (bST, 92% compared with control, 67%) but not in ewes at the first time they were mated (bST, 71% compared with control, 87%; P>0.05). IGF-I concentrations were greater (P<0.01) in ewes treated with bST than in control ewes from 2 days after treatment. Progesterone concentrations did not vary (P>0.05) between groups. It is concluded that a single dose of bST 5 days before progestin withdrawal increases lambing rate and prolificacy in sheep. These effects are associated with an increase in circulating concentrations of IGF-I.  相似文献

Peripheral plasma progesterone and oestrogen concentrations were measured during late pregnancy and the parturient period in 12 ewes producing live lambs and three others producing stillborn lambs. Progesterone values started declining by 10 days before lambing but at minus 24 hours were still 6.1 +/- 3 ng/ml in the ewes bearing live lambs; during the last 24 hours progesterone was significantly lower in the ewes producing stillborn lambs. Oestrogens reached a maximum level of 550 +/- 280 pg/ml at the time of delivery and declined rapidly to basal values shortly after lambing. Oestrogens did not rise at lambing in the ewes producing stillborn lambs. In seven of the 12 ewes bearing live lambs, uterine activity, as determined by intrauterine pressure changes, was recorded throughout parturition and compared with the plasma values for progesterone and oestrogens. It was found that there was a highly significant positive correlation between uterine activity and plasma oestrogen concentrations, and a highly significant negative correlation between uterine activty and both plasma progesterone concentrations and the progesterone: oestrogen ratio.  相似文献

These studies test the hypothesis that pregnancy alters the feedback effects of cortisol on stimulated ACTH secretion. Ewes were sham-operated (Sham), or adrenalectomized (ADX) at approximately 108 days gestation and replaced with aldosterone (3 microg x kg(-1) x day(-1)) and with cortisol at either of two doses (ADX + 0.6 and ADX + 1 mg x kg(-1) x day(-1)); ewes were studied during pregnancy and postpartum. Mean cortisol levels produced in ADX ewes were similar to normal pregnant ewes (ADX+1) or nonpregnant ewes (ADX+0.6), respectively. Plasma ACTH concentrations in response to infusion of nitroprusside were significantly increased in the pregnant ADX+0.6 ewes (1,159 +/- 258 pg/ml) relative to pregnant Sham ewes (461 +/- 117 pg/ml) or the ADX+1 ewes (442 +/- 215 pg/ml) or the same ewes postpartum (151 +/- 69 pg/ml). Plasma ACTH concentrations were not significantly different among the groups postpartum. Increasing plasma cortisol to 20-30 ng/ml for 24 h before hypotension produced similar inhibition of ACTH in all groups. Pregnancy appears to decrease the effectiveness of low concentrations of cortisol to inhibit ACTH responses to hypotension.  相似文献
