A low fluence of ultraviolet radiation (UV) causes cultured cells of Rosa damascena Mill cv. Gloire de Guilan to lose intracellular K+. This effect required the presence of Ca2+ in the medium. A reduction in the concentration of free Ca2+ to 10−5 M with ethyleneglycol-bis-(β-aminoethyl-ether)-N.N.N',N'-tetraacetic acid (EGTA) buffer inhibited the UV-stimulated efflux; this was correlated with a discharge of the membrane potential and a stimulation of the leakage of K+ from unirradiated cells. All the same effects were seen with La3+ at 0.2 m M. At 0.02 m M La3+, the UV-stimulated efflux of K+ was blocked without concomitant effects on the membrane potential or K+ efflux from control cells. It is suggested that removal of Ca2+ blocks or masks the UV-induced leakage of K+ by destabilizing the plasma membrane. In addition, La3+ may specifically inhibit the UV-stimulated opening of K+ or anion channels.  相似文献

Irradiation of cultured rose ( Rosa damascena Mill. cv. Gloire de Guilan) cells with ultraviolet light caused of loss of K+, which occurred with sigmoid kinetics. The kinetics of loss of K+ were not changed when the extracellular concentration of K+ was held constant during the period of efflux. Furthermore, the rate of loss of K+ was approximately the same even though the K+ concentration in the medium was increased from 0.1 to 10 m M . The kinetics of uptake of the lipophilic methyltriphenylphosphonium cation, an indicator of the plasma membrane potential, were linear throughout the period of K+ efflux, suggesting that the starting and stopping of K+ efflux do not reflect a passive response to changes in the membrane potential of the cells. The results are interpreted in terms of activation and inactivation of an efflux channel or pump for K+.  相似文献

Broad-band radiation from a high-pressure Hg-vapor lamp, including ultraviolet wavelengths from 290 to 400 nm, blue, green and red wavelengths, did not induce the synthesis of H2O2 in cultured rose cells. This was in contrast to the effects of shortwave (254 nm) ultraviolet radiation, even though, like shortwave ultraviolet radiation, the UV-B component of the broadband radiation induced a striking K+ efflux from the cells, and this efflux has been associated with H2O2 synthesis in a previous report. The UV-A and visible wavelengths were shown to inhibit the synthesis of H2O2. This effect was associated with inhibition of peroxidase, an enzyme reported to be involved in the synthesis of H2O2 in cell walls. UV-B radiation inhibited the alternate pathway for mitochondrial electron transport, but there was no evidence that this effect contributed to the inhibition of H2O2 synthesis in cells treated with broad-band radiation.  相似文献

Clones of Rosa damascena Mill. cv. Gloire de Guillan, selected for growth in solid medium containing 56 m M NaClO3, were studied to determine the reason for their resistance to this toxic salt. The cells grew on medium containing nitrate as the only nitrogen source, and they synthesized nitrate reductase (EC 1.6.6.2) in the presence of nitrate. The cells were resistant in the presence of nitrate. However, their resistance was greatly increased by the presence of glutamate in the medium. The cells took up [36Cl]-ClO3- and reduced it to ClO2, but the fraction of ClO3 that they reduced under our experimental conditions was less than that reduced by wild type. The slower production of ClO2 apparently accounted for the resistance of the cells to ClO3. We suggest several possible reasons for the low rate of reduction of ClO3.  相似文献

The effects of cadmium and lead on the internal concentrations of Ca2+ and K+, as well as on the uptake and translocation of K(86Rb+) were studied in winter wheat (Triticum aestivum L. a. MV-8) grown hydroponically at 2 levels of K+ (100 uM and 10 mM). Cd2+ and Pb2+ were applied in the nutrient solution in the range of 0.3 to 1000 u.M. Growth was more severely inhibited by Cd2+ and in the high-K+ plants as compared to Pbz+ and low-K+ plants. Ions of both heavy metals accumulated in the roots and shoots, but the K+ status influenced their levels. Ca2+ accumulation was increased by low concentrations of Cd2+ mainly in low-K+ shoots, whereas it was less influenced by Pb2+. The distribution of Cd2+ and Ca2+ in the plant and in the growth media indicated high selectivity for Cd2+ in the root uptake, while Ca2+ was preferred in the radial and/or xylem transport. Cd2+ strongly inhibited net K+ accumulation in high-K+ plants but caused stimulation at low K+ supply. In contrast, the metabolis-dependent influx of K+(86Rb+) was inhibited in low-K+ plants, while the passive influx in high-K+ plants was stimulated. Translocation of K+ from the roots to the shoots was inhibited by Cd2+ but less influenced in Pb2+-treated plants. It is concluded that the effects of heavy metals depend upon the K+-status of the plants.  相似文献

Suspension-cultured rose ( Rosa damascena Mill. cv. Gloire de Guilan) cells irradiated with UV-C (254 nm. 558 J m−2) showed a transient production of H2O2 as measured by chemiluminescence of luminol in the presence of peroxidase (EC 1.1 1.1.7). The peak concentration of H2O2, which occurred at about 60–90 min after irradiation, was 8–9 μ M . The time course for the appearance of H2O2 matched that for UV–induced K+ efflux. Treatments that inhibited the UV-induced efflux of K+, including heat and overnight incubation with cycloheximide and diethylmaleate, also inhibited the appearance of H2O2. The converse was not always true, since catalase (EC 1.11.1.6. and salicylhydroxamic acid, which inhibited luminescence, did not stop K+ efflux. We conclude that H2O2 synthesis depends on K+ efflux. Because H2.O2 in the extracellular space is required for lignin synthesis in many plant tissues, we suggest that the UV–stimulated production of H2O2 is an integral part of a defensive lignin synthesis.  相似文献

Passive fluxes of K+ (86Rb) into roots of sunflower ( Helianthus annuus L. cv. Uniflorus) were determined at low K+ concentration (0.1 and 1.0 mM K+) in the ambient solution. Metabolic uptake of K+ was inhibited by 10−4M 2,4-dinitrophenol (DNP). K+ (86Rb) fluxes were studied both continuously and by time differentiation of uptake. In high K+ roots passive uptake was directly proportional to the K+ concentration of the uptake solution, indicating free diffusion. This assumption was supported by the fact that passive Rb+ uptake was not affected by high K+ concentrations. In low K+ roots the passive uptake of K+ was higher than in high K+ roots. The increase was possibly due to carrier-mediated K+ transport. As K+ effluxes were quantitatively similar to influxes, it is suggested that passive K+ fluxes represent exchange diffusion without relation to net K+ transport.  相似文献

The effect of phytochrome on K+ transport in guard cells of Commelina communis L. was studied following stomatal movement and using the K+−channel blockers tetraethylammonium (TEA), Cs+ and quinidine. TEA and quinidine prevented stomatal opening and closure in red light, but not when it was supplemented with far-red. This indicates that channels that can be blocked by TEA and quinidine are regulated by phytochrome. Evidence for a phytochrome effect on K+ leakage through other membranal compartments was also found. These phytochrome effects are modified by temperature. Elevated temperature decreases the involvement of channels and increases K+ transport through other membrane compartments, while low temperature causes channel opening and diminishes K+ leakage. The interaction between phytochrome effects and those of temperature is discussed.  相似文献

Betula papyrifera Marsh, seedlings adapted very poorly to flooding for up to 60 days. Responses to flooding included increased ethylene production; stomatal closure; leaf senescence; drastic inhibition of shoot growth, cambial growth, and root growth; decay of roots, and death of many seedlings. Flooding inhibited growth of leaves that formed prior to flooding, inhibited formation of new leaves, and induced abscission of old leaves. As a result of extensive leaf abscission, fewer leaves were present after flooding than before flooding was initiated. The drastic reduction in leaf area was associated with greatly decreased growth of the lower stem and roots. No evidence was found of adaptive morphological changes to flooding. The data indicate that intolerance of B. papyrifera seedlings to flooding is an important barrier to regeneration of the species on sites subject to periodic inundation.  相似文献

The uptake of Na+ and the loss of Ca2+ and K+ by seeds of Acacia tortilis (Forsk.) Hayne (salt tolerant) and A. coriacea DC. (salt sensitive) were determined after 24 h soaking in 250 mol m-1,3 NaCl or in distilled water. Na+ uptake was higher by the seed coat than by the embryo of both species and higher by A. coriacea than by A. tortilis. The greater Na+ uptake by A. coriacea was associated with greater Ca and K+ leakage. The Na+ concentration of solution imbibed by the embryo of both species was lower than the Na+ concentration in the external solution, indicating an exclusion of Na+. When A. tortilis and A. coriacea seeds were treated with a series of NaCl concentration (0–400 mol m-1,3), the exclusion mechanism was particularly clear with A. tortilis at lower concentrations (50 and 150 mol m-1,3) of NaCl. In contrast, the seed coat of both species accumulated Na+. Thus the seed coat may play an important role in ion exchange. These results show that it is important to consider the seed coat and embryo separately rather than the whole seed when considering ion exchange in relation to salinity tolerance.  相似文献

Plants of barley ( Hordeum vulgare L. cv. Salve) were grown with 6.5–35% relative increase of K+ supply per day (RKR) using a special computer-controlled culture unit. After a few days on the culture solution the plants adapted their relative growth rate (RGR) to the rate of nutrient supply. The roots of the plants remained in a low salt status irrespective of the rate of nutrient supply, whereas the concentration of K+ in shoots increased with RKR. Both Vmax and Km for K+(86Rb) influx increased with RKR. It is concluded that with a continuous and stable K+ stress, the K+ uptake system is adjusted to provide an effective K+ uptake at each given RKR. Allosteric regulation of K+ influx does not occur and efflux of K+ is very small.  相似文献

More substances leaked from a higher-vigor seed sample than from a lower-vigor sample. This indicates that, in some cases, electric conductivity does not represent seed vigor level very well, especially for high-vigor seeds. Results from germination, germination index, leachate conductivity, and the ratio of K^+/Na^+ from three-seed lots of Chinese cabbage (Brassica pekinensis (Louv.) Rupr) showed that K^+/Na^+ correlated well with germination and germination index. The ability of K^+/Na^+ to indicate well changes in vigor was further supported by investigation in soybean (Glycine max (L.) Merr.) seeds and another cultivar of Chinese cabbage seeds. Thus, seed leakage of K^+/Na^+ can accurately indicate seed vigor, whereas the conductivity test failed to do so. Furthermore, K^+/Na^+ showed up bigger quantitative differences in vigor level than did the conductivity test. This findings provide a more sensitive and accurate index for the assessment of seed vigor. The mechanisms of Na^+ and K^+ ion transport are also discussed.  相似文献

The stimulation of dicotyledonous leaf growth by light depends on increased H+ efflux, to acidify and loosen the cell walls, and is enhanced by K+ uptake. The role of K+ is generally considered to be osmotic for turgor maintenance. In coleoptiles, auxin‐induced cell elongation and wall acidification depend on K+ uptake through tetraethylammonium (TEA)‐sensitive channels (Claussen et al., Planta 201, 227–234, 1997), and auxin stimulates the expression of inward‐rectifying K+ channels ( Philippar et al. 1999) . The role of K+ in growing, leaf mesophyll cells has been investigated in the present study by measuring the consequences of blocking K+ uptake on several growth‐related processes, including solute accumulation, apoplast acidification, and membrane polarization. The results show that light‐stimulated growth and wall acidification of young tobacco leaves is dependent on K+ uptake. Light‐stimulated growth is enhanced three‐fold over dark levels with increasing external K+, and this effect is blocked by the K+ channel blockers, TEA, Ba++ and Cs+. Incubation in 10 mm TEA reduced light‐stimulated growth and K+ uptake by 85%, and completely inhibited light‐stimulated wall acidification and membrane polarization. Although K+ uptake is significantly reduced in the presence of TEA, solute accumulation is increased. We suggest that the primary role of K+ in light‐stimulated leaf growth is to provide electrical counterbalance to H+ efflux, rather than to contribute to solute accumulation and turgor maintenance.  相似文献

The classic compartment analysis of ion efflux from roots is often applied with the assumption that there is a system of 3 compartments in series. However, complex ion transport across the root tissues, as well as influences from the shoot, may complicate the picture. The present experiments were performed to study the immediate effects that excision of the shoot before the experiment exerts on the efflux of Rb+(86Rb+) and of K+(86Rb+) from 9-day-old roots of plants of barley (Hordeum vulgare L. cv. Salve). The efflux from high K+ and low K+ roots of intact and detopped plants were compared. After excision of the shoot of high K+ plants, a marked increase in efflux was observed after 2.5 h with a maximum at about 7 h. The increase in efflux was seen as a peak in plots of efflux versus time. Excision of the shoot from low K+ roots did not give rise to a consistent increase in efflux. Regular K+ ion efflux curves were observed from roots of intact plants of high or low K+ status. Furthermore, after a pulse treatment of 9-day-old roots of intact plants of high or low K+ status with a solution containing Rb+(86Rb+), the Rb+(86Rb+) transport to the shoots was not reduced during the following 3 h in unlabelled solution. It is suggested that both the peak appearing in the efflux plots and the maintained tracer transport to the shoots after transfer of the roots to an unlabelled solution indicate the existence of a K+/Rb+ transport system in the symplasm of the roots that has only a slow exchange with the bulk cytoplasm and vacuoles.  相似文献

The effects of external K+, H+ and Ca2+ concentrations on the intracellular K+ concentration, [K+]i, and the K+-ATPase activity in 2-day-old mung bean roots [ Vigna mungo (L.) Hepper] were investigated. [K+]i, in mung bean roots was markedly decreased by external K+ or H+ stress and did not recover the initial value even after the stress was removed. This decrease in [K+]i, gradually disappeared with the addition of (Ca2+. Ca2+ may offset the harmful effects of ion stress. Ca2+ seems to have two effects on K+ transport; control of K+ permeability and activation of K+ uptake, although K+-ATPase activity was inhibited by Ca2+ concentrations higher than 10–4 M. We suggest that Ca2+ activates K+ uptake indirectly through the acidification of the cytoplasm.  相似文献

Abstract Three calmodulin antagonists, trifluoperazin, compound 48/ 80 and calmidazolium provoke K+ efflux from metabolizing yeast cells. This K+ efflux is accompanied by a shrinkage of the cells. Part of the cells show a gradual shrinkage whilst the remainder of the cells shrink via an all-or-none process. Cells shrunken via an all-or-none process have the same size as cells which have been completely permeabilized by a cationic detergent with loss of all their K+.  相似文献

In this study, amiodarone, at very low concentrations, produced a clear efflux of K+. Increasing concentrations also produced an influx of protons, resulting in an increase of the external pH and a decrease of the internal pH. The K+ efflux resulted in an increased plasma membrane potential difference, responsible for the entrance of Ca2+ and H+, the efflux of anions and the subsequent changes resulting from the increased cytoplasmic Ca2+ concentration, as well as the decreased internal pH. The Δ tok1 and Δ nha1 mutations resulted in a smaller effect of amiodarone, and Δ trk1 and Δ trk2 showed a higher increase of the plasma membrane potential. Higher concentrations of amiodarone also produced full inhibition of respiration, insensitive to uncouplers and a partial inhibition of fermentation. This phenomenon appears to be common to a large series of cationic molecules that can produce the efflux of K+, through the reduction of the negative surface charge of the cell membrane, and the concentration of this cation directly available to the monovalent cation carriers, and/or producing a disorganization of the membrane and altering the functioning of the carriers, probably not only in yeast.  相似文献

Four-week-old sunflower plants ( Helianthus annuus L. cv. Halcón), grown in different nutrient solutions, were used to study the effects of gibberellic acid (GA3) on K+ (Rb+) uptake by roots or transport to the shoot. Gibberellic acid application to the nutrient solution did not affect the exudation process of excised roots. When GA3 was sprayed on leaves 2 to 6 days before excising the roots, the rate of exudation and the K+ flux increased. When the exudation study was done keeping the roots in a nutrient solution in which Rb+ replaced K+, the GA3 effects were evident also on Rb+ uptake and transport. In intact plants, GA3 increased the Rb+ transported to the shoot but did not affect Rb+ accumulation in the root. It is suggested that these GA3 effects can be explained if it is assumed that GA3 acts on the transport of ions to the xylem vessels.  相似文献
