A comparison of the effects of a rapidly imposed water deficit with different leaf ages on chlorophyll a fluorescence and gas exchange was performed in maize (Zea mays L.) plants. The relationships between photosynthesis and leaf relative turgidity (RT) and ion leakage were further investigated. Leaf dehydration substantially decreased net photosynthetic rate (A) and stomatal conductance (G s), particularly for older leaves. With dehydration time, F v /F m maintained a relatively stable level for youngest leaves but significantly decreased for the older leaves. The electron transport rate (ETR) sharply decreased with intensifying dehydration and remained at lower levels during continuous dehydration. The photochemical quenching of variable chlorophyll fluorescence (q P) gradually decreased with dehydration intensity for the older leaves but increased for the youngest leaves, whereas dehydration did not affect the nonphotochemical chlorophyll fluorescence quenching (NPQ) for the youngest leaves but remarkably decreased it for the older leaves. The leaf RT was significantly and positively correlated with its F v /F m, ETR, and q P, and the leaf ion leakage was significantly and negatively correlated with F v /F m and NPQ. Our results suggest that the photosynthetic systems of young and old leaves decline at different rates when exposed to rapid dehydration.  相似文献

Estimates of thylakoid electron transport rates (Je) from chlorophyll fluorometry are often used in combination with leaf gas exchange measurements to provide detailed information about photosynthetic activity of leaves in situ. Estimating Je requires accurate determination of the quantum efficiency of Photosystem II (ΦP), which in turn requires momentary light saturation of the Photosystem II light harvesting complex to induce the maximum fluorescence signal (FM′). In practice, full saturation is often difficult to achieve, especially when incident photosynthetic photon flux density (Q) is high and energy is effectively dissipated by non-photochemical quenching. In the present work, a method for estimating the true FM′ under high Q was developed, using multiple light pulses of varying intensity (Q′). The form of the expected relationship between the apparent FM′ and Q′ was derived from theoretical considerations. This allowed the true FM′ at infinite Q′ to be estimated from linear regression. Using a commercially available leaf gas exchange/ chlorophyll fluorescence measurement system, Je was compared to gross photosynthetic CO2 assimilation (AG) under conditions where the relationship between Je and AG was expected to be linear. Both in C4 leaves (Zea mays) in ambient air and also in C3 leaves (Gossypium hirsutum) under non-photorespiratory conditions the apparent ratio between Je and AG declined at high Q when ΦP was calculated from FM′ measured simply using the highest available saturating pulse intensity. When FM′ was determined using the multiple pulse / linear regression technique, the expected relationship between Je and AG at high Q was restored, indicating that the ΦP estimate was improved. This method of determining FM′ should prove useful for verifying when saturating pulse intensities are sufficient, and for accurately determining ΦP when they are not. In order to evaluate the photosynthetic activity of a C3 leaf from the electron transport rate (ETR) of photosystem 2 (PS2), a new method was devised and examined using leaves of sweet potato. In this method, both surfaces of a leaf were sealed with transparent films to stop the gas exchange between the leaf and the atmosphere; hence the functions of both photosynthetic assimilation (CO2 uptake) and photorespiration (CO2 release) are restricted to the inside of the leaf. After both functional rates became equally balanced, ETR of the sealed leaf (ETRseal) was determined from the chlorophyll fluorescence. The measurements were conducted at different irradiances and leaf temperatures and by using leaves of different age. Under each measurement condition, ETRseal showed a close positive relationship with the photosynthetic potential, or the gross photosynthetic rate measured in the air of 2 % O2 (P G2%) before sealing. ETRseal may become an indicator to estimate or evaluate the photosynthetic activity of C3 leaves.  相似文献

Substantial lateral CO(2) diffusion rates into leaf areas where stomata were blocked by grease patches were quantified by gas exchange and chlorophyll a fluorescence imaging in different species across the full range of photosynthetic photon flux densities (PPFD). The lateral CO(2) flux rate over short distances was substantial and very similar in five dicotyledonous species with different vascular anatomies (two species with bundle sheath extensions, sunflower [Helianthus annuus] and dwarf bean [Phaseolus vulgaris]; and three species without bundle sheath extensions, faba bean [Vicia faba], petunia [Petunia hybrida], and tobacco [Nicotiana tabacum]). Only in the monocot maize (Zea mays) was there little or no evident lateral CO(2) flux. Lateral diffusion rates were low when PPFD <300 micromol m(-2) s(-1) but approached saturation in moderate PPFD (300 micromol m(-2) s(-1)) when lateral CO(2) diffusion represented 15% to 24% of the normal CO(2) assimilation rate. Smaller patches and higher ambient CO(2) concentration increased lateral CO(2) diffusion rates. Calculations with a two-dimensional diffusion model supported these observations that lateral CO(2) diffusion over short distances inside dicotyledonous leaves can be important to photosynthesis. The results emphasize that supply of CO(2) from nearby stomata usually dominates assimilation, but that lateral supply over distances up to approximately 1 mm can be important if stomata are blocked, particularly when assimilation rate is low.  相似文献

One broad-leaved pioneer tree, Alnus formosana, two broad-leaved understory shrubs, Ardisia crenata and Ardisia cornudentata, and four ferns with different light adaptation capabilities (ranked from high to low, Pyrrosia lingus, Asplenium antiquum, Diplazium donianum, Archangiopteris somai) were used to elucidate the light responses of photosynthetic rate and electron transport rate (ETR). Pot-grown materials received up to 3 levels of light intensity, i.e., 100%, 50% and 10% sunlight. Both gas exchange and chlorophyll (Chl) fluorescence were measured simultaneously by an equipment under constant temperature and 7 levels (0?C2,000 ??mol m?2 s?1) of photosynthetic photon flux density (PPFD). Plants adapted to-or acclimated to high light always had higher light-saturation point and maximal photosynthetic rate. Even materials had a broad range of photosynthetic capacity [maximal photosynthetic rate ranging from 2 to 23 ??mol(CO2) m?2 s?1], the ratio of ETR to gross photosynthetic rate (P G) was close for A. formosana and the 4 fern species when measured under constant temperature, but the PPFD varied. In addition, P. lingus and A. formosana grown under 100% sunlight and measured at different seasonal temperatures (15, 20, 25, and 30°C) showed increased ETR/P G ratio with increasing temperature and could be fitted by first- and second-order equations, respectively. With this equation, estimated and measured P G were closely correlated (r 2 = 0.916 and r 2 = 0.964 for P. lingus and A. formosana, respectively, p<0.001). These equations contain only the 2 easily obtained dynamic indicators, ETR and leaf temperature. Therefore, for some species with near ETR/P G ratio in differential levels of PPFD, these equations could be used to simulate dynamic variation of leaf scale photosynthetic rate under different temperature and PPFD conditions.  相似文献

We investigated the morphological and physiological acclimation of leaves grown within a canopy of Japanese oak tree (Quercus mongolica var. crispula) in terms of the susceptibility to photoinhibition under various growth light conditions. The maximum rates of photosynthesis (P(max) ) and electron transport (ETR(max) ) were higher in mature leaves grown under stronger light with higher area-based leaf nitrogen (N) content closely associated with higher leaf mass per area. The net photosynthetic (P(n) ) and electron transport (ETR) rates corresponding to the daily peak photosynthetic photon flux density (PPFD(max) ) during leaf maturation were almost comparable to P(max) and ETR(max) , respectively. Conversely, P(n) and ETR at the daily average PPFD (PPFD(avg) ) were substantially low in shade-grown leaves when compared with P(max) and ETR(max) . The susceptibility to photoinhibition at PPFD(max) , i.e. at sunflecks for the shade-grown leaves, was assessed by the rate of excess energy production. Although sun leaves showed higher rates of electron transport and thermal energy dissipation than shade leaves under PPFD(max) conditions, the rate of excess energy production was almost constant across shade to sun leaves. The shade leaves of the Japanese oak grown within a crown were suggested to adjust their N investment to maintain higher photosynthetic capacities compared with those required to maximize the net carbon gain, which may facilitate the dissipation of the excessive light energy of sunflecks to circumvent photoinhibition in cooperation with thermal energy dissipation.  相似文献

The impact of powdery (Uncinula necator) and downy mildew (Plasmopara viticola) on grapevine leaf gas exchange was analysed. Gas exchange measurements (assimilation A, transpiration E, stomatal conductance gs, intercellular concentration of CO2Ci) were made on three different leaf materials: (i) healthy tissue of diseased leaves, (ii) infected tissue of diseased leaves, (iii) healthy tissue of healthy leaves (control treatment). Using the same source of leaf tissue, photosynthetic pigment concentration (chlorophyll a, b) and fluorescence levels (minimal fluorescence F0, maximal fluorescence Fm and the optimal quantum yield [Fm ? F0]/Fm) were determined to explain the mechanism of action of the two diseases on leaf assimilation. The results indicated that powdery and downy mildew reduced the assimilation rates, not only through a reduction in green leaf area (visual lesions), but also through an influence on gas exchange of the remaining green leaf tissues, determining a ‘virtual lesion’. The ratios between virtual and visual lesions were higher in powdery mildewed leaves than in the downy mildewed leaves. The photosynthetic fluorescence level (Fv/Fm) was affected by neither of the two pathogens. The reduction in intercellular concentration of CO2 and photosynthetic pigment may explain the lower assimilation rates in the healthy tissues of powdery and downy mildewed leaves respectively.  相似文献

Two photosynthetic periods and photosynthetic photon flux densities (PPFD) were used to study the relationship between the rate of photosynthesis and starch accumulation in vegetative soybean leaves (Merr. cv Amsoy 71). Plants grown in short daily photosynthetic periods (7 hours) had higher rates of CO2 fixation per unit leaf dry weight and of leaf starch accumulation than plants grown in long daily photosynthetic periods (14 hours) irrespective of PPFD. CO2 fixation rates per unit leaf area were similar in 7-hour and 14-hour plants grown at low PPFD but were highest in 14-hour plants at the high PPFD. When single leaves of 14-hour plants were given 7-hour photosynthetic periods, their rates of starch accumulation remained unchanged. The programming of starch accumulation rate and possibly of photosynthetic rate by the length of the daily photosynthetic period is apparently a whole-plant, not an individual leaf, phenomenon. Programming of chloroplast starch accumulation rate by length of the daily photosynthetic and/or dark periods was independent of PPFD within the ranges used in this experiment.  相似文献

Gas exchange and fluorescence parameters were measured simultaneously in two Zea mays L. cultivars (Liri and 121C D8) to assess the relationship between the quantum yield of electron transport (PS2) and the quantum yield of CO2 assimilation (CO2) in response to photosynthetic photon flux density (PPFD). The cv. Liri was grown under controlled environmental conditions in a climate chamber (CC) while cv. 121C D8 was grown in CC as well as outdoors (OT). By exposing the two maize cultivars grown in CC to an increasing PPFD, higher photosynthetic and photochemical rates were evidenced in cv. Liri than in cv. 121C D8. In Liri plants the PS2/CO2 ratio increased progressively up to 27 with increasing PPFD. This suggests that the reductive power was more utilised in non-assimilatory processes than in CO2 assimilation at high PPFD. On the contrary, by exposing 121C D8 plants to increasing PPFD, PS2/CO2 was fairly constant (around 11–13), indicating that the electron transport rate was tightly down regulated by CO2 assimilation. Although no significant differences were found between PS2/CO2 of the 121C D8 maize grown under CC and OT by exposing them to high PPFD, the photosynthetic rate and photochemical rates were higher in OT maize plants.  相似文献

Increasing photosynthetic photon flux density (PPFD) received during development from 5.5 to 31.2 mol m-2 d-1 resulted in greater leaf and mesophyll cell surface areas in cotton (Gossypium hirsutum L.). The relationships between the amounts of these surface areas and potential CO2 assimilation by these leaves were evaluated. Leaf area (epidermal surface area of one side of a leaf), mesophyll cell surface area, and net rate of CO2 uptake (Pn) were measured from the time leaves first unfolded until P., was substantially reduced. At the higher PPFD, leaf and mesophyll surface areas increased more rapidly during expansion, and Pn per unit leaf area was greater than at the lower PPFD. Although leaves at the higher PPFD reached the maximum P., per unit mesophyll cell surface area 4 to 5 days earlier than leaves at the lower PPFD, the maxima for these P., were similar. Leaves grown at the higher PPFD had the potential to assimilate 2.2, 3.5, or 5.8 times the amount of CO2 as leaves from the lower PPFD when P., was expressed per unit mesophyll surface, per unit leaf surface, or per whole leaf, respectively. Greater and earlier development of both P., and mesophyll cell surface area at higher PPFD apparently had a compounding effect on the potential for carbon assimilation by a leaf.  相似文献

High temperature generally constrains plant growth and photosynthesis in many regions of the world; however, little is known about how photosynthesis responds to high temperature with regard to different leaf ages. The synchronous changes in gas exchange and chlorophyll fluorescence at three leaf age levels (just fully expanded, mature, and older leaves) of maize (Zea mays L.) were determined at three temperatures (30°C as a control and 36 and 42°C as the higher temperatures). High temperature significantly decreased the net CO2 assimilation rate (A), stomatal conductance (g s), maximal efficiency of photosystem II (PSII) photochemistry (F v/F m), efficiency of excitation energy capture by open PSII reaction centers ( F\textv /F\textm F^{\prime}_{\text{v}} /F^{\prime}_{\text{m}} ), photochemical quenching of variable chlorophyll fluorescence (q P), and the electron transport rate (ETR), whereas minimal fluorescence yield (F 0) and nonphotochemical quenching of variable chlorophyll fluorescence (q N) were increased. The youngest fully expanded leaves had higher A, ETR, and q P compared with older leaves. Higher temperature with old leaves led to significant malondialdehyde (MDA) accumulation, a proxy for lipid peroxidation damage from active oxygen species (AOS). MDA content was significantly negatively correlated with A, F v/F m, F\textv /F\textm F^{\prime}_{\text{v}} /F^{\prime}_{\text{m}} , and q P. Thus, the results suggest that photosynthetic potentials, including stomatal regulation and PSII activity, may be restricted at high temperature, together with increasing cell peroxidation, which may be closely associated with leaf age.  相似文献

Seagrass photosynthesis is usually measured in laboratory experiments, following oxygen evolution in closed chambers. Pulse amplitude modulated (PAM) fluorometry constitutes an alternative and non-intrusive method of measuring photosynthesis in the field. Validation of electron transport rate (ETR) measurements as reliable estimators of actual photosynthetic production requires that a significant linear relationship between oxygen production and ETR is demonstrated, and also that a 0.25 molar ratio between these two measures is verified. In this work, both parameters were measured simultaneously in laboratory experiments, over a range of light intensities, under well-defined and controlled conditions. A linear relationship was observed between the average rates of oxygen production and the electron transport rates for Zostera noltii (Hornemann) obtained at several irradiances. The molar ratio found between oxygen production and ETR was 0.15±0.02, lower than the theoretically expected value of 0.25. The use of PAM fluorescence as a valid proxy for photosynthetic production was validated for the range of 35-490 μmol photons m−2 s−1 (PAR), under the assumption that the electron sinks responsible for the molar ratio deviation remain constant in similar experimental conditions.  相似文献

The differential pigment composition and photosynthetic activity of sun and shade leaves of deciduous (Acer pseudoplatanus, Fagus sylvatica, Tilia cordata) and coniferous (Abies alba) trees was comparatively determined by studying the photosynthetic rates via CO(2) measurements and also by imaging the Chl fluorescence decrease ratio (R(Fd)), which is an in vivo indicator of the net CO(2) assimilation rates. The thicker sun leaves and needles in all tree species were characterized by a lower specific leaf area, lower water content, higher total chlorophyll (Chl) a+b and total carotenoid (Cars) content per leaf area unit, as well as higher values for the ratio Chl a/b compared to the much thinner shade leaves and needles that possess a higher Chl a+b and Cars content on a dry matter basis and higher values for the weight ratio Chls/Cars. Sun leaves and needles exhibited higher rates of maximum net photosynthetic CO(2) assimilation (P(Nmax)) measured at saturating irradiance associated with higher maximum stomatal conductance for water vapor efflux. The differences in photosynthetic activity between sun and shade leaves and needles could also be sensed via imaging the Chl fluorescence decrease ratio R(Fd), since it linearly correlated to the P(Nmax) rates at saturating irradiance. Chl fluorescence imaging not only provided the possibility to screen the differences in P(N) rates between sun and shade leaves, but in addition permitted detection and quantification of the large gradients in photosynthetic rates across the leaf area existing in sun and shade leaves.  相似文献

Diurnal changes in photosynthetic gas exchange and chlorophyll fluorescence were measured under full sunlight to reveal diffusional and non‐diffusional limitations to diurnal assimilation in leaves of Arisaema heterophyllum Blume plants grown either in a riparian forest understorey (shade leaves) or in an adjacent deforested open site (sun leaves). Midday depressions of assimilation rate (A) and leaf conductance of water vapour were remarkably deeper in shade leaves than in sun leaves. To evaluate the diffusional (i.e. stomatal and leaf internal) limitation to assimilation, we used an index [1–A/A350], in which A350 is A at a chloroplast CO2 concentration of 350 μ mol mol ? 1. A350 was estimated from the electron transport rate (JT), determined fluorometrically, and the specificity factor of Rubisco (S), determined by gas exchange techniques. In sun leaves under saturating light, the index obtained after the ‘peak’ of diurnal assimilation was 70% greater than that obtained before the ‘peak’, but in shade leaves, it was only 20% greater. The photochemical efficiency of photosystem II ( Δ F/Fm ′ ) and thus JT was considerably lower in shade leaves than in sun leaves, especially after the ‘peak’. In shade leaves but not in sun leaves, A at a photosynthetically active photon flux density (PPFD) > 500 μ mol m ? 2 s ? 1 depended positively on JT throughout the day. Electron flows used by the carboxylation and oxygenation (JO) of RuBP were estimated from A and JT. In sun leaves, the JO/JT ratio was significantly higher after the ‘peak’, but little difference was found in shade leaves. Photorespiratory CO2 efflux in the absence of atmospheric CO2 was about three times higher in sun leaves than in shade leaves. We attribute the midday depression of assimilation in sun leaves to the increased rate of photorespiration caused by stomatal closure, and that in shade leaves to severe photoinhibition. Thus, for sun leaves, increased capacities for photorespiration and non‐photochemical quenching are essential to avoid photoinhibitory damage and to tolerate high leaf temperatures and water stress under excess light. The increased Rubisco content in sun leaves, which has been recognized as raising photosynthetic assimilation capacity, also contributes to increase in the capacity for photorespiration.  相似文献

Background and Aims

Theory for optimal allocation of foliar nitrogen (ONA) predicts that both nitrogen concentration and photosynthetic capacity will scale linearly with gradients of insolation within plant canopies. ONA is expected to allow plants to efficiently use both light and nitrogen. However, empirical data generally do not exhibit perfect ONA, and light-use optimization per se is little explored. The aim was to examine to what degree partitioning of nitrogen or light is optimized in the crowns of three tropical canopy tree species.

Methods

Instantaneous photosynthetic photon flux density (PPFD) incident on the adaxial surface of individual leaves was measured along vertical PPFD gradients in tree canopies at a frequency of 0·5 Hz over 9–17 d, and summed to obtain the average daily integral of PPFD for each leaf to characterize its insolation regime. Also measured were leaf N per area (Narea), leaf mass per area (LMA), the cosine of leaf inclination and the parameters of the photosynthetic light response curve [photosynthetic capacity (Amax), dark respiration (Rd), apparent quantum yield (ϕ) and curvature (θ)]. The instantaneous PPFD measurements and light response curves were used to estimate leaf daily photosynthesis (Adaily) for each leaf.

Key Results

Leaf Narea and Amax changed as a hyperbolic asymptotic function of the PPFD regime, not the linear relationship predicted by ONA. Despite this suboptimal nitrogen partitioning among leaves, Adaily did increase linearly with PPFD regime through co-ordinated adjustments in both leaf angle and physiology along canopy gradients in insolation, exhibiting a strong convergence among the three species.

Conclusions

The results suggest that canopy tree leaves in this tropical forest optimize photosynthetic use of PPFD rather than N per se. Tropical tree canopies then can be considered simple ‘big-leaves’ in which all constituent ‘small leaves’ use PPFD with the same photosynthetic efficiency.Key words: Optimal resource allocation, nitrogen, photosynthetic capacity, leaf mass per area, tropical trees, radiation use efficiency, scaling, leaf angle, canopy architecture, big leaf model  相似文献

Seasonal variations in free putrescine, spermidine and spermine content, gas-exchange and chlorophyll fluorescence parameters were followed during winter and summer on leaves of a similar age from undisturbed holm oak trees (control, C) and resprouts (R) originated after fire. We observed a general trend of putrescine content decrease with increasing irradiance. Putrescine content decreased markedly from winter to summer, especially in R, which were located on a site with much higher irradiation. Daily summer variations in putrescine showed a decline at midday from morning values, and they were also more accentuated in R. Measurement of gas-exchange and chlorophyll fluorescence parameters showed marked differences between C and R under their respective light conditions. R showed higher values of PSII quantum yield (ΦPSII), photochemical quenching (qP) and intrinsic efficiency of open PSII centres () The ΦPSII/PPFD response curve showed that under the same irradiance, ΦPSII was enhanced in R and mainly under high light conditions. In spite of increasing irradiance from winter to summer, and especially in burned areas, the mentioned chlorophyll fluorescence parameters were maintained indicating the adaptation of the photosynthetic apparatus. Results derived from A/C i and A/PPFD response curves showed enhanced photosynthetic capacity and lower non-stomatal limitation of photosynthesis in R during summer stress. The contribution of putrescine decline in the photoadaptation of the photosynthetic apparatus of species growing in natural forest habitats is considered.  相似文献

薇甘菊(Mikania micranth)是世界性的入侵有害植物,对其入侵特性的理解将有助于我们更进一步揭示入侵机理和开展植物入侵的防治工作。叶片的光合作用是入侵植物薇甘菊入侵特性的研究内容之一,但到目前为止,仍没有开展对薇甘菊非同化器官茎的同化特性的研究。该文采取对比的研究方法,使用LICOR-6400气体交换系统和荧光系统对其幼嫩的绿色茎和成熟叶片的气体交换和叶绿素荧光特性进行测定,并对测定结果进行了对比分析,同时应用激光共聚焦显微镜对薇甘菊茎中叶绿体分布进行观察。使用叶绿素荧光系统测量瞬时叶绿素荧光特性表明,茎和叶的电子传输速率(Electron transport rate, ETR)和光系统Ⅱ实际光化学量子产量(ΦPSⅡ)存在较好的正比例关系,相关系数达到0.97,说明茎中存在和叶中类似的光合结构。但在应用LICOR-6400气体交换系统测量稳定状态下的CO2的气体交换速率时,观察到叶的气体交换速率稳定性较好,而茎的气体交换速率出现较大的波动,这可能是由于茎的气孔因素引起。综合来看,在相同面积和饱合光强下(光通量密度(Photosynthetic photo flux density, PPFD)=2 000 μmol·m -2·s-1),叶的ETR为42.44 μmol·m -2·s-1,茎的ETR为30.32 μmol·m -2·s-1。在相同面积和低光强(PPFD=10 μmol·m -2·s-1)下,叶的ΦPSⅡ为0.69;茎的ΦPSⅡ为0.57。在单位SPAD下,茎中ETR是每单位SPAD 4.24 μmol·m -2·s-1,是叶的2.3倍,实际光化学量子产量是每单位SPAD 0.08,是叶的3倍。在比较茎和叶ETR中观察到茎比叶有更强的强光适应能力。激光共聚焦图像观察到薇甘菊茎的叶绿体主要分布在两个区域:皮层区和维管束周围。对照以往关于茎中叶绿体功能的研究表明,可能分布在两个区域中的叶绿体功能上存在差别。如上,薇甘菊茎中存在一定的光合作用能力,且在叶绿体的光能利用瞬时效率上茎明显强于叶,但在茎中这些光合作用的具体作用仍不清楚。  相似文献

The effects of extreme phosphate (Pi) deficiency during growth on the contents of adenylates and pyridine nucleotides and the in vivo photochemical activity of photosystem II (PSII) were determined in leaves of Helianthus annuus and Zea mays grown under controlled environmental conditions. Phosphate deficiency decreased the amounts of ATP and ADP per unit leaf area and the adenylate energy charge of leaves. The amounts of oxidized pyridine nucleotides per unit leaf area decreased with Pi deficiency, but not those of reduced pyridine nucleotides. This resulted in an increase in the ratio of reduced to oxidized pyridine nucleotides in Pi-deficient leaves. Analysis of chlorophyll a fluorescence at room temperature showed that Pi deficiency decreased the efficiency of excitation capture by open PSII reaction centres (φe), the in vivo quantum yield of PSII photochemistry (φPSII) and the photochemical quenching co-efficient (qP), and increased the non-photochemical quenching co-efficient (qN) indicating possible photoinhibitory damage to PSII. Supplying Pi to Pi-deficient sunflower leaves reversed the long-term effects of Pi-deficiency on PSII photochemistry. Feeding Pi-sufficient sunflower leaves with mannose or FCCP rapidly produced effects on chlorophyll a fluorescence similar to long-term Pi-deficiency. Our results suggest a direct role of Pi and photophosphorylation on PSII photochemistry in both long-and short-term responses of photosynthetic machinery to Pi deficiency. The relationship between φPSII and the apparent quantum yield of CO2 assimilation determined at varying light intensity and 21 kPa O2 and 35 Pa CO2 partial pressures in the ambient air was linear in Pi-sufficient and Pi-deficient leaves of sunflower and maize. Calculations show that there was relatively more PSII activity per mole of CO2 assimilated by the Pi-deficient leaves. This indicates that in these leaves a greater proportion of photosynthetic electrons transported across PSII was used for processes other than CO2 reduction. Therefore, we conclude that in vivo photosynthetic electron transport through PSII did not limit photosynthesis in Pi-deficient leaves of sunflower and maize and that the decreased CO2 assimilation was a consequence of a smaller ATP content and lower energy charge which restricted production of ribulose, 1-5, bisphosphate, the acceptor for CO2.  相似文献

A method for measuring whole plant photosynthesis in Arabidopsis thaliana   总被引:5,自引:0,他引:5  
Measurement of photosynthesis of intact leaves of Arabidopsis thaliana has been prohibitive due to the small leaf size and prostrate growth habit. Because of the widespread use of Arabidopsis for plant science research it is important to have a procedure for accurate, nondestructive measurement of its photosynthesis. We developed and tested a method for analysis of photosynthesis in whole plants of Arabidopsis. Net carbon assimilation and stomatal conductance were measured with an open gas exchange system and photosynthetic oxygen evolution was determined from chlorophyll fluorescence parameters. Individual plants were grown in 50 cubic centimeter tubes that were attached with an air tight seal to an enclosed gas exchange chamber for measurement of carbon dioxide and water exchange by the whole plant. Chlorophyll fluorescence from intact leaves was simultaneously measured with a pulse modulated fluorometer. Photosynthetic CO2 assimilation and stomatal conductance rates were calculated with established gas exchange procedures and O2 evolution was determined from chlorophyll fluorescence measurement of Photosystem II yield. Carbon assimilation and oxygen evolution in response to light intensity and ambient CO2 concentration was measured and is presented here to demonstrate the potential use of this method for investigation of photosynthesis of Arabidopsis plants in controlled environment conditions.  相似文献
