Hematopoiesis entails the generation of stem cells, the proliferation and maintenance of multipotential progenitors, and lineage commitment and maturation. During the past year, critical components of these steps have been defined. Notable are gene-targeting experiments in mice in which one or more hematopoietic lineages have been shown to be ablated upon inactivation of several nuclear regulatory proteins (GATA-2, Tal-1/SCL, Rbtn2/LMO2, PU.1, Ikaros, E2A, and Pax-5), and experiments that establish GATA-1 as a factor capable of programming at least three lineages (erythroid, thrombocytic, and eosinophilic) from a transformed avian progenitor cell. 相似文献

Biochemistry and structural biology are undergoing a dramatic revolution. Until now, we have tried to study subtle and complex biological processes by crude in vitro techniques, looking at average behaviors of vast numbers of molecules under conditions usually remote from those existing in the cell. Researchers have realized the limitations of this approach, but none other has been available. Now, we can not only observe the nuances of the behaviors of individual molecules but prod and probe them as well. Perhaps most important is the emerging ability to carry out such observations and manipulations within the living cell. The long-awaited leap to an in vivo biochemistry is at last underway. 相似文献

Abstract 1. In solitary parasitoids, several species can exploit the same host patch and competition could potentially be a strong selective agent as only one individual can emerge from a host. In cereal crops, Aphidius rhopalosiphi and A. ervi share the grain aphid Sitobion avenae as host. 2. The present work studied foraging strategies of both species on patches already exploited by the other species. The study analysed larval competition in multi‐parasitised hosts and compared the foraging behaviour of females with and without previous experience. 3. It was found that A. ervi wins larval competition three times more often than A. rhopalosiphi. Both species spent less time on patches exploited by a heterospecific than on unexploited ones. When they foraged on heterospecifically exploited patches, experienced females induced less mortality in aphids than inexperienced ones. 4. Although A. rhopalosiphi is a specialist on cereal aphids and is the most abundant species due to its early appearance in the season, S. avenae is still a profitable host for A. ervi, because: (i) A. rhopalosiphi leaves patches partially exploited, (ii) A. ervi wins larval competition in three out of four multi‐parasitised hosts, and (iii) A. ervi is only slightly deterred by the cornicular secretions of the host and can thus easily parasitise hosts. 相似文献

The literature on apoptosis has grown tremendously in recent years, and the mechanisms that are involved in this programmed cell death pathway have been enlightened. It is now known that apoptosis takes place starting from early development to adult stage for the homeostasis of multicellular organisms, during disease development and in response to different stimuli in many different systems. In this review, we attempted to summarize the current knowledge on the circumstances and the mechanisms that lead to induction of apoptosis, while going over the molecular details of the modulator and mediators of apoptosis as well as drawing the lines between programmed and non-programmed cell death pathways. The review will particularly focus on Bcl-2 family proteins, the role of different caspases in the process of apoptosis, and their inhibitors as well as the importance of apoptosis during different disease states. Understanding the molecular mechanisms involved in apoptosis better will make a big impact on human diseases, particularly cancer, and its management in the clinics. 相似文献

Sulfur heterocycles are common constituents ofpetroleum and liquids derived from coal, and they arefound in some secondary metabolites of microorganismsand plants. They exist primarily as saturated ringsand thiophenes. There are two major objectives drivinginvestigations of the microbial metabolism oforganosulfur compounds. One is the quest to develop aprocess for biodesulfurization of fossil fuels, andthe other is to understand the fates of organosulfurcompounds in petroleum- or creosote-contaminatedenvironments which is important in assessingbioremediation processes. For these processes to besuccessful, cleavage of different types of sulfurheterocyclic rings is paramount. This paper reviewsthe evidence for microbial ring cleavage of a varietyof organosulfur compounds and discusses the fewwell-studied cases which have shown that the C–S bondis most susceptible to breakage leading to disruptionof the ring. In most cases, the introduction of one ormore oxygen atom(s) onto the adjacent C atom and/oronto the S atom weakens the C–S bond, facilitating itscleavage. Asymmetric development of the vertebrate embryo has fascinated embryologists for over a century. Much has been learned since the asymmetric Nodal signaling cascade in the left lateral plate mesoderm was detected, and began to be unraveled over the past decade or two. When and how symmetry is initially broken, however, has remained a matter of debate. Two essentially mutually exclusive models prevail. Cilia-driven leftward flow of extracellular fluids occurs in mammalian, fish and amphibian embryos. A great deal of experimental evidence indicates that this flow is indeed required for symmetry breaking. An alternative model has argued, however, that flow simply acts as an amplification step for early asymmetric cues generated by ion flux during the first cleavage divisions. In this review we critically evaluate the experimental basis of both models. Although a number of open questions persist, the available evidence is best compatible with flow-based symmetry breakage as the archetypical mode of symmetry breakage. 相似文献

B. AbdullGaffar Impact factor in cytopathology journals: what does it reflect and how much does it matter? Objective: To study the trends of impact factor (IF) in four cytopathology journals. To investigate the factors that might influence IF in cytopathology literature and whether IF has any impact on cytopathology practice. Methods: The IFs of four cytopathology journals were searched from 2005 to 2009. The IFs and their relationships with the types and number of publications, publishers, the official societies, readership, the quality of their contents, the topics covered and the levels of evidence were compared. Results: Cancer Cytopathology (CC) had the highest IF. Acta Cytologica (AC) had the lowest IF, which appeared to be in decline. Cytopathology (C) and Diagnostic Cytopathology (DC) had a slow but steady increase in their IF. Components that might influence these differences could include the category and the society of the journal, targeted readers and certain types of publications. Publishers, the number of publications, the types of topics covered and the levels of evidence probably have no major effect on IF. Conclusions: IF has its own benefits and original applications. IF is a quantitative measure that does not reflect the levels of evidence in cytopathology journals. IF should not be abandoned because it might encourage competition between cytopathology journals, but it should not dictate their contents. 相似文献

Conundrums of competitive ability in plants: what to measure? 总被引:8,自引:0,他引:8
A survey of recent literature indicates that competitive ability in plants has been measured, in most studies, only in terms of the relative intensity of size suppression experienced by competitors within one growing season. Far fewer studies have recorded relative success in terms of survival and even fewer studies have recorded fecundity under competition. Differences in size suppression are usually assumed to reflect differences in relative abilities to deny resources to competitors. However, most previous studies have failed to control or account for other sources of variation in the size suppression that plants experience under competition, i.e. variation between mixtures in the resource supply/demand ratio (approach to carrying capacity), or variation in the degree of niche overlap between competitors, or variation in the intensity of concurrent facilitative interactions between competitors. For future studies, much greater caution is required in recognizing these inherent limitations of traditional measures of competitive ability and, hence, guarding against unfounded conclusions or predictions about potential for competitive success that are based on these measures. There is also a significant challenge for future studies to adopt empirical approaches for minimizing these limitations. Some initial recommendations are considered here based on an emerging view of competitive ability measured in terms of traits associated with all three conventional components of Darwinian fitness, i.e. not just growth (plant size) but also survival and fecundity allocation (offspring production per unit plant size per unit time). According to this model, differences in competitive ability imply differences in the ability, despite intense competition (i.e. low resource supply/demand ratio), to recruit offspring into the next generation and thereby limit offspring recruitment by other plants. The important traits of competitive ability, therefore, are not only those that allow a plant to deny resources to competitors, suppress their sizes and hence, maximize the plant's own size, but also those traits that allow the plant to withstand suppression from competition enough to persist, both as an individual (through survival) and across generations (through descendants). 相似文献

In the current omics era, innovative high-throughput technologies allow measuring temporal and conditional changes at various cellular levels. Although individual analysis of each of these omics data undoubtedly results into interesting findings, it is only by integrating them that gaining a global insight into cellular behaviour can be aimed at. A systems approach thus is predicated on data integration. However, because of the complexity of biological systems and the specificities of the data-generating technologies (noisiness, heterogeneity, etc.), integrating omics data in an attempt to reconstruct signalling networks is not trivial. Developing its methodologies constitutes a major research challenge. Besides for their intrinsic value towards health care, environment and industry, prokaryotes are ideal model systems to further develop these methods because of their lower regulatory complexity compared with eukaryotes, and the ease with which they can be manipulated. Several successful examples outlined in this review already show the potential of the systems approach for both fundamental and industrial applications, which would be time-consuming or impossible to develop solely through traditional reductionist approaches. 相似文献

The RAD6 pathway of budding yeast, Saccharomyces cerevisiae, is responsible for a substantial fraction of this organism's resistance to DNA damage, and also for induced mutagenesis. The pathway appears to incorporate two different recovery processes, both regulated by RAD6. The error-prone recovery prcess accounts for only a small amount of RAD6-dependent resistance, but probably all induced mutagenesis. The underlying mechanism, for error-prone recovery is very likely to be translesion synthesis. The error-free recovery process accounts for most of RAD6-dependent resistace, but its mechanism is less clear; it may entail error-free bypass by template switching and/or DNA gap filling by recombination. RAD6 regulates these activities by ubiquitinateins, and the roles they play in error-free and error-prone recovery, have not yet been established. 相似文献

In spite of the existence of a vast body of information on the plant diversity of the Amazon, there remain significant obstacles to informed decision-making and management for conservation. Species distributions are poorly understood and the relationships between diversity and composition of vegetation, ecosystem services and resilience to climatic fluctuations are insufficiently clear. The geographic distribution of phylogenetic diversity in relation to current protected areas is unexplored and very little is known about intraspecific genetic variability and its practical significance. Interpretation of vegetation differentiation and distribution remains relatively simplistic; there are still large parts of the basin for which few or no botanical data exist, and many rare and sparsely distributed species remain undiscovered. Improved understanding of the ecological roles, dynamics and associations of the species of greatest importance for the maintenance of sustainable livelihoods and ecosystem services, habitat restoration and adaptation to climate change is a high priority. In many cases these are common and widespread species. Some of these issues are explored by looking at the Cristalino region in northern Mato Grosso as a case-study. Effective integration, quality assessment, synthesis and application of existing data on the plant diversity of the Amazon will help to address these issues. However, more targeted information is needed from the ground. Future prioritisation of research effort will require a careful and pragmatic balance between the ‘traditional’ focus on rare and endemic species and species-rich communities, and the growing need to understand the key ‘framework’ elements that will determine the future of the Amazon environment. Similar situations are faced elsewhere in the tropics: for botanical research institutes in the 21st century this demands an urgent re-evaluation of core activities and concerted engagement with the issues and challenges facing conservation in a context of rampant population growth, climate change and environmental destruction. 相似文献

OBJECTIVE: To calculate the cost of coronary artery bypass grafting (CABG) and to compare it with the costs determined in two previous Canadian studies. DESIGN: Retrospective cost-analysis study. SETTING: A tertiary care referral hospital. PATIENTS: Fifty patients who had undergone successful triple and quadruple CABG between Jan. 3 and 30, 1989. MAIN RESULTS: The cost of CABG per patient varied from $10,982 to $33,676 (mean $14,328) (in 1988 Canadian dollars). The cost tended to increase with age and number of vessels grafted. Compared with the patients in the two previous Canadian studies our patients were older, had more vessels grafted and cost more to treat, even after the total hospital costs were adjusted for inflation. CONCLUSIONS: The population undergoing CABG is changing: it is older and has more diseased vessels. These changes have had a significant impact on the cost of CABG. Further study is required to determine the outcome and benefit of CABG in this group of patients. 相似文献

Treatment with the chimerical monoclonal antibody rituximab results in CD20-directed B cell depletion. Although this depletion is almost complete in the peripheral blood of nearly all patients with rheumatoid arthritis, a proportion of patients does not exhibit a clinical response. The paper by Nakou and colleagues suggests that a decrease in CD19+CD27+ memory B cells in both peripheral blood and bone marrow precedes the clinical response to rituximab. This finding adds to the emerging evidence that lack of response to rituximab is associated with persistence of B lineage cells in specific body compartments. 相似文献

The evidence base for drug treatment of hypertension is strong. Early trials using thiazide diuretics suggested a shortfall in prevention of coronary heart disease. The superiority of newer drugs has been widely advocated but trial evidence does not support an advantage of beta-blockers, angiotensin converting enzyme inhibitors, calcium channel blockers or alpha-blockers for this outcome. Even meta-analyses have failed to clarify matters. If this issue is to be settled, bigger and better trials of longer duration in high-risk patients are needed. Meanwhile, the importance of rigorous blood pressure control using multiple drugs has been established. This should be the focus of our attention rather than agonising over differences in cause-specific outcomes that may not be generalisable to all patient populations. 相似文献

The study of developmental sequences of physiological traits could be an important way of placing comparative developmental physiology (CDP) within the research agenda being forged by work on developmental plasticity. Here we focus on the concept of heterokairy defined by Spicer & Burggren in 2003 as changes in the timing of physiological development in an individual. The role of this concept in the future of the CDP is discussed. First we provide an historical perspective of the ideas that have led to the investigation of sequences in CDP. This is followed by a re-examination and clarification of the definition of physiological heterokairy before empirical case studies that (explicitly or implicitly) demonstrate physiological heterokairy are reviewed. We suggest that physiological heterokairy can be demonstrated through a wide range of invertebrate and vertebrate examples. However, care must be taken when inferring that heterokairy as a pattern is always the result of heterokairic processes as there is evidence that physiological heterokairy could result from the altered timing of both homologous or analogous physiological mechanisms. We conclude by discussing the potential link between heterokairy and heterochrony and suggest that the investigation of this link should be a major goal for workers in both CDP and developmental plasticity. 相似文献
