Organelles morphologically similar to microbodies have been found in several tissues of atmospheric species of Tillandsia from different habitats. The presence of catalase was demonstrated by the DAB reaction thus confirming the microbody nature of these organelles. They are a feature of the Tillandsia species with normal photosynthetic carbon fixation and with CAM. Their size is consistently small. The nucleoid observed in the microbodies shows a characteristic morphology which has not been reported before within other plant microbodies. This nucleoid is composed of minute tubular structures, for which the authors here propose a three-dimensional arrangement.  相似文献

Plication of staminal filaments is an important diagnostic character for Tillandsia subgenus Anoplophytum (ca. 45 species). The monophyletic integrity of subgenus Anoplophytum has recently been questioned, and we conducted an anatomical investigation of plicate staminal filaments to better characterize this putative synapomorphy. Developmental studies show that the filament plications, or folds, become visible during or soon after anthesis. Serial sections of preplication filaments and filaments in sequential stages of plication were prepared and observed with light microscopy. A uniform sequence of parenchyma cell collapse begins three to four cell layers out from the vascular bundle and proceeds centrifugally to the epidermis. Eventually the epidermal cells collapse, leaving only the vascular bundle and a few surrounding parenchyma cells intact. Above and below the zone of plication, all parenchyma and epidermal cells in the filament remain intact. Species traditionally placed in subgenera Tillandsia and Allardtia have been found with plicate staminal filaments that are anatomically and develop-mentally identical to those studied from subgenus Anoplophytum. Alone, staminal filament plication does not appear to be a good diagnostic character for subgenus Anoplophytum, and doubts concerning the monophylesis of this subgenus are reinforced. The functional significance of stamen filament plication remains unknown.  相似文献

The genetic structure of two epiphytic species, Tillandsia ionantha and T. recurvata, was investigated using enzyme electrophoresis. Electrophoretic data suggest that T. ionantha and T. recurvata differ in breeding system, in agreement with predictions based on their strikingly different floral morphologies. Electrophoretic data suggest extremely high levels of inbreeding for T. recurvata, whereas Tillandsia ionantha exhibits characteristics of an outcrossing species. Values of P, H, and mean number of alleles per locus are much higher in T. ionantha than in T. recurvata. The mean value of FIS for T. ionantha is low (0.056), closely approaching expectations at Hardy-Weinberg equilibrium. In contrast, the mean value of FIS in T. recurvata (1.000) indicates a complete absence of heterozygotes. The two species also differ in genetic structure. Low FST values for T. ionantha indicate little variation in allele frequencies among populations. In contrast, FST values are high for T. recurvata, suggesting substantial genetic heterogeneity among populations. In addition, the mean value of I is higher in T. ionantha (0.995) than in T. recurvata (0.931). Population genetic data are in agreement with the suggestion of Benzing (1978), who proposed that extreme epiphytes such as T. recurvata, would be characterized by increased autogamy ensuring high seed set. Due to high chromosome numbers in Bromeliaceae (most taxa have x = 25), the family has been considered polyploid. However, with the exception of an additional isozyme for PGM in T. recurvata, the two species are isozymically diploid.  相似文献

The fine structure of the pollen grain of Tillandsia caput-medusae Morr. (Bromeliaceae) prior to germination has been studied. The development, after the first mitosis, is here schematized in three stages which are in accordance with the main steps described in angiosperms. The ultrastructural modifications occurring in the generative and vegetative cells are discussed in view of their different destiny. The results obtained are compared with the data known about tropical orchids and epiphyteic plants like Tillandsia. The following differences have been observed: a large vacuole in the vegetative cell; rapid thinning of the wall between the generative and vegetative cells; great quantity of ribosomes and rough endoplasmic reticulum surrounding the vacuoles in the generative cell. The above-listed ultrastructural features may have a meaning, considering the peculiar environmental conditions in which the epiphytism of Tillandsia is achieved.  相似文献

Structural features of the foliar epidermal surfaces of selected epiphytic members of subfamily Tillandsioideae of Bromeliaceae were examined to determine why xeromorphic nonimpounding (atmospheric) species with a dense layer of foliar trichomes are seldom found in deeply shaded humid portions of the forest profile, whereas tank-forming members of the same subfamily often abound in such sites. Two features of the peltate trichome which appear to play a major role in enforcing habitat preferences among members of Tillandsioideae are (1) anatomical features of the walls of certain trichome cells which affect the mobility of the shield and. (2) variations in juxtaposition of trichomes and stomates on the leaf surface. Both features are involved in inhibition of gas exchange in wet leaves; trichome shield flexibility is also associated with high leaf reflectivity when the leaf surface is dry.  相似文献

Two distinct morphological forms characterize the ontogenetic development of the epiphytic bromeliad Tillandsia deppeana. Juveniles are characterized by a non-impounding rosette of small, linear leaves covered with elaborate trichomes possessing a 4 + 8 + 16 + 64 shield cell pattern. The broader transitional leaves, which form an impounding rosette prior to the initiation of true adult leaves, also possess trichomes with the 4 + 8 + 16 + 64 cell pattern. Adult individuals have large, broad leaves with overlapping, sheathing bases which impound water and debris. These leaves have trichomes with shields exhibiting a 4 + 8 + 32 cell pattern. Trichome density is fairly uniform in the juvenile leaves with trichomes covering 100% of leaf surfaces, whereas in the adult leaves density is high at the base and diminishes significantly toward the apex. Stomatal density of both juvenile and adult leaves increases from the base to the apex, although this is most pronounced in the adults. Stomata in the adults are also arranged in longitudinal series parallel and abaxial to parallel rows of mesophyll tissue. The results of this study indicate that juveniles of T. deppeana are more similar morphologically to adult atmospheric-type tillandsioid species than to the tank-forming adults into which they eventually develop.  相似文献

Analyses of Tillandsia circinnata Schlecht specimens from nutrient-stressed and more fertile habitats in southern Florida confirmed earlier findings that its vegetative and reproductive vigor are enhanced as shoot tissue concentrations of P, K and Mg increase. Demands by this perennial herb for N, P and K are more pronounced than for B, Ca, Cu, Fe, Mg, Mn, Mo and Zn, primarily because N, P and K must be maintained at relatively high concentrations in vegetative tissues and are heavily expended during seed production. When most or all of this epiphyte's self-fertile flowers set fruit— a fairly common occurrence in most of the populations examined—asexual propagation by offshoot is subordinated to the sexual effort, especially if the specimen is growing in an unusually sterile habitat and has very limited mineral resources. Predictions implicit in r-K selection and related life history theory are consistent with the notion presented here that T. circinnata is following a pattern of nutrient use for reproductive purposes that represents a highly adaptive way to expend slowly accumulated mineral resources in stressful habitats represented by small, scattered, short-lived microsites. Specifically, it expends scarce mineral resources during the reproductive effort in a way that greatly enhances fecundity and vagility while minimizing the rate at which new microsites and reproducing individuals must be recruited. Reproduction and mineral use by T. circinnata is compared with that of T. utriculata, a semelparous epiphyte that shares many of the same forest habitats in southern Florida.  相似文献

Tillandsia L. Subg. Phytarrhiza (Visiani) Baker (Bromeliaceae) is a distinctive group of about 35 epiphytic species. These exhibit a range of habits from xeric to mesic. The evolutionary relationships of the contrasting adaptations need to be established here as well as in the subfamily as a whole. Relations between the subgenus and other tillandsioids are problematical and phylogenetic reconstruction of its member-species would be facilitated by identification of Phytarrhiza's relative (sister taxon) sharing the same most recent common ancestor with Phytarrhiza. This paper examines the two most likely sister taxa, Subg. Pseudo-Catopsis Baker and Subg. Diaphoranthema (Beer) Baker. Diaphoranthema is rejected as sister taxon. The accepted evolutionary tree, rooted by Pseudo-Catopsis, indicates that most habital evolutionary changes in Phytarrhiza have been between mesic and semi-mesic forms and from mesic to xeric forms. Methods developed for testing specific evolutionary hypotheses are broadly applicable.  相似文献

双歧杆菌活菌制剂对烧伤鼠肠源性感染的预防作用   总被引:3,自引:0,他引:3  
本文以30%烫伤同时口服抗生素,喂饲双歧杆菌制剂的大鼠为模型,将Wistar大鼠分为烧伤治疗组,烧伤对照组及正常对照组。并于伤后24、48、72、96小时分别观察盲肠内容物菌群,粘蛋白,分泌型免疫球蛋白A(SIgA);肠道细菌移位率;腔静脉内毒素含量。结果发现烧伤后肠道菌群有不同程度的变化,念珠菌迅速过度生长、厌氧菌下降、肠内需厌氧菌比例严重失调,治疗组中粘蛋白含量,肠道细菌移位率均明显小于烧伤对照组;而SIgA含量则高于对照组,菌群改变较对照组轻。故认为双歧杆菌制剂有助于预防烧伤肠源性感染的发生。  相似文献

为了解共生菌对黑豆蚜蛋白质、氨基酸代谢的影响 ,用利福平处理黑豆蚜以除去其细胞内共生细菌 ,产生脱共生蚜虫。结果表明 ,被脱去共生菌的蚜虫与未经抗生素处理的正常蚜虫相比 ,7日龄时 ,脱共生蚜虫每毫克鲜重的总蛋白含量降低了 2 9% ,每毫克鲜重的游离氨基酸含量提高了 17%。对黑豆蚜取食的蚕豆苗韧皮部组织中必需氨基酸所占的比例进行分析后发现 ,蚕豆苗韧皮部组织中的必需氨基酸含量仅占 2 0 % ,而有共生菌的黑豆蚜组织中必需氨基酸已达到 4 4% ,脱共生后降低到 37% ,这些结果证明了黑豆蚜的胞内共生菌为其寄主提供了部分必需氨基酸。通过对游离氨基酸组成的分析发现 ,在测定的 17种氨基酸中 ,必需氨基酸中的苏氨酸在共生蚜虫中所占的比例为 2 1 6 % ,在脱共生蚜虫中仅为 16 7%。同样 ,非必需氨基酸中的酪氨酸和丝氨酸 ,在共生蚜虫中分别占总游离氨基酸的 8 9%和 5 6 % ,而在脱共生蚜虫中却分别升高到 2 1 1%和 13 6 %。这些结果表明 ,各种氨基酸比例的失调 ,造成了脱共生蚜虫蛋白质合成受阻和部分游离氨基酸的积累 ,并因此导致蚜虫发育和繁殖的失调。  相似文献

To evaluate the role of bacterial symbionts (Buchnera spp.) in the black bean aphids (Aphis craccivora Koch), the aphids were treated with the antibiotic, rifampicin, to eliminate their intracellular symbiotic bacteria. Analysis of protein and amino acid concentration in 7‐day‐old of aposymbiotic aphids showed that the total protein content per mg fresh weight was significantly reduced by 29%, but free amino acid titers were increased by 17%. The ratio of the essential amino acids was in general only around 20% essential amino acids in phloem sap of broad bean, whereas it was 44% and 37% in symbiotic and aposymbiotic aphids, respectively, suggesting that the composition of the free amino acids was unbalanced. For example, the essential amino acid, threonine represented 21.6% of essential amino acids in symbiotic aphids, but it was only 16.7% in aposymbiotic aphids. Likewise, two nonessential amino acids, tyrosine and serine, represented 8.9% and 5.6% of total amino acids in symbiontic aphids, respectively, but they enhanced to 21.1% and 13.6% in aposymbiotic aphids. It seems likely that the elevated free amino acid concentration in aposymbiotic aphids was caused by the limited protein anabolism as the result of the unbalanced amino acid composition.  相似文献

The evolution of key innovations, novel traits that promote diversification, is often seen as major driver for the unequal distribution of species richness within the tree of life. In this study, we aim to determine the factors underlying the extraordinary radiation of the subfamily Bromelioideae, one of the most diverse clades among the neotropical plant family Bromeliaceae. Based on an extended molecular phylogenetic data set, we examine the effect of two putative key innovations, that is, the Crassulacean acid metabolism (CAM) and the water‐impounding tank, on speciation and extinction rates. To this aim, we develop a novel Bayesian implementation of the phylogenetic comparative method, binary state speciation and extinction, which enables hypotheses testing by Bayes factors and accommodates the uncertainty on model selection by Bayesian model averaging. Both CAM and tank habit were found to correlate with increased net diversification, thus fulfilling the criteria for key innovations. Our analyses further revealed that CAM photosynthesis is correlated with a twofold increase in speciation rate, whereas the evolution of the tank had primarily an effect on extinction rates that were found five times lower in tank‐forming lineages compared to tank‐less clades. These differences are discussed in the light of biogeography, ecology, and past climate change.  相似文献

P accumulation and metabolic pathway in N2-fixing Anabaena flos-aquae (Lyngb.) Bréb were investigated in P-sufficient (20 μMP) and P-limited (2 μMP) turbidostats in combined N-free medium. The cyanobacterium grew at its maximum rate (μmax, 1.13 d?1) at the high P concentration and at 65% of μmax under P limitation, with total cell P concentrations (QP) at steady states of 12.0 and 5.2 fmol·cell?1, respectively. At steady state, polyphosphates (PPi) accounted for only 3% of QP (0.4 fmol·cell?1) in P-rich cells. Its concentration in P-limited cells was 5.8% (0.3 fmol·cell?1). On the other hand, sugar P was very high at 22% of QP in P-rich cells and was undetectable in P-limited cells. Pulse chase experiments with 32P showed that P-rich cells initially incorporated the labeled P into the acid-soluble PPi fraction within the first few minutes and to a lesser extent into nucleotide P. Radioactivity in the PPi then declined rapidly with concomitant increases in sugar P and nucleotide P fractions. In contrast, in P-limited cells, no radiolabel was detected in acid-soluble PPi, and 32P was initially incorporated into nucleotide P, sugar P, and ortho P fractions. The latter two fractions then subsequently declined. Therefore, under N2-fixing conditions the cyanobacteria appeared to store P as sugar P and also utilize P through different pathways under P-rich and -limited conditions. When nitrate was supplied as the N source under P-sufficient conditions, PPi accounted for about 15% of steady-state QP, but no sugar P was detected. Therefore, the same organism stored P in different cell P fractions depending on its N sources.  相似文献

The roles of ethylene in corolla growth and senescence have been extensively studied; light, temperature, and abscisic acid (ABA) have already been implicated in rapid corolla opening (unfolding) in morning glory. In the present study, a possible interaction between ABA and ethylene production was examined. While applied ABA promoted corolla unfolding, it also promoted ethylene production. Furthermore, the effect of ABA could be eliminated by the ethylene biosynthesis inhibitors, aminoethoxyvinyl glycine (AVG) and cobalt ions. Simultaneously applied aminocyclopropane carboxylic acid (ACC) augmented the ABA response, but ACC applied alone promoted corolla unfolding as effectively as ABA alone. Measurements of ethylene production during eight successive stages of flower opening showed that the ethylene burst widely reported to occur in opened flowers prior to corolla senescence actually begins before the corolla unfolds. Ethylene production seems to be a later part of the sequence of biochemical events that leads to both corolla unfolding and synchronized inrolling and senescence.  相似文献

The UTEX 2193 strain of Scenedesmus armatus (Chod.) Chod, when cultured in any of several media (whether natural or artificial, concentrated or dilute) produced a variety of colonial morphologies as well as a unicell population. Morphological expression was related to culture ape. When the initial cell density was just a feu1 hundred cells per mL. the culture first produced a unicell population, then spiny colonies, and as stationary phase was approached, spine-less colonies. Two classes of spiny colonies were detected. Type I colonies had elongate cells with the terminal cells shorter than median cells. Spines were longer than cell length. The wider, oval, grainy cells of Type II colonies were uniform m length. Spines were shorter and thicker than those on Type I colonies. Only Type I colonies produced unicells: the latter appeared as two morphs. The smaller unicell was obovoid with four delicate spines: the larger had ovate cells bearing four thicker spines. Control of unicell development in all media was achieved by carefully monitoring colony type and cell number used for the inoculum. A unicellular population developed in batch culture in defined media, both concentrated and dilute, when the initial cell density (either Type I or Type II colonies) was low (below 1000 cells-mL?1), as well as in synchronous cultures. With higher initial cell densities, e.g. 2 × 104 cells·mL?1, the inoculum had to contain Type I colonies to produce unicells. Unicells were also produced in water from Agronomy Pond, where the strain originated. We discuss the role of unicell populations in the distribution of Scenedesmus.  相似文献

The Km for ammonia for glutamine synthetase and glutamate dehydrogenase was measured in enzyme extracts from Skeletonema costatum (Grev.) Cleve. At similar physiological pH and temperature the half-saturation constant for glutamine synthetase was 29 μM, whereas for GDH it was 28mM. On the basis of relative enzymic activity, as well as substrate affinity, it is suggested that glutamine synthetase is the enzyme primarily responsible for the incorporation of ammonium into the amino acid pool, when extracellular nitrogen is at ecological concentrations.  相似文献

Diatoms and related algae have plastids that are surrounded by four membranes. The outer two membranes are continuous with the endoplasmic reticulum and the inner two membranes are analogous to the plastid envelope membranes of higher plants and green algae. Thus the plastids are completely compartmentalized within the ER membranes. The targeting presequences for nuclear-encoded plastid proteins have two recognizable domains. The first domain is a classic signal sequence, which presumably targets the proteins to the endoplasmic reticulum. The second domain has characteristics of a transit peptide, which targets proteins to the plastids of higher plants. To characterize these targeting domains, the presequence from the nuclear-encoded plastid protein AtpC was utilized. A series of deletions of this presequence were fused to Green Fluorescent Protein (GFP) and transformed into cells of the diatom, Phaeodactylum tricornutum. The intracelluar localization of GFP was visualized by fluorescence microscopy. This work demonstrates that the first domain of the presequence is responsible for targeting proteins to the ER lumen and is the essential first step in the plastid protein import process. The second domain is responsible to directing proteins from the ER and through the plastid envelope and only a short portion of the transit peptide-like domain is necessary to complete this second processing step. In vivo data generated from this study in a fully homologous transformation system has confirmed Gibbs' hypothesis regarding a multistep import process for plastid proteins in chromophytic algae.  相似文献
