Wild-type strains of the interfertile species Chlamydomonas eugametos (UTEX 9 and 10) and Chlamydomonas moewusii (UTEX 96 and 97) male readily and reciprocally; however, considerable lethality occurs among F1 hybrid meiotic products. We prepared two hybrid backcross lineages using C. eugametos and C. moewusii. One lineage began with the cross C. eugametos mating-type-plus (mt+) × C. moewusii mating-type-minus (mt?). An F1 mt+ hybrid from this cross was back-crossed to C. moewusii mt?, and a B1 mt+ hybrid was recovered. The B1 hybrid was again backcrossed to C. moewusii mt?, and this process was repeated through the fifth backcross. The other backcross lineage began with the reciprocal cross C. moewusii mt+× C. eugametos mt? and employed C. eugametos as the recurring mt? parent. This lineage also was continued through the fifth backcross. Meiotic product survival in the reciprocal interspecific crosses was less than 10%. In successive back-cross generations associated with both lineages, this value increased progressively to a maximum of 85–90%, the level observed for the intraspecific crosses. These results are consistent with the hypothesis that multiple genetic differences exist between C. eugametos and C. moewusii and that these are the major source of meiotic product lethality associated with the interspecific crosses. The inheritance of chloroplast genetic markers for resistance to streptomycin (sr-2) and for resistance to erythromycin (er-nM1) was also scored w the interspecific crosses and in the backcrosses. Most hybrid zygospores transmitted the resistance markers of the mt+ parent only, or of both parents, with the former zygospore type being more common. Although the intraspecific C. eugametos and C. moewusii crosses differ conspicuously with respect to the fraction of zygospores which transmit chloroplast genetic markers of both parents, the inheritance of chloroplast genetic markers in the interspecific crosses and backcrosses at' scribed here failed to clarify the genetic basis for this difference.  相似文献

We have attempted interspecific hybridizations among six species of rhabditid nematodes: Caenorhabditis elegans, Caenorhabditis briggsae, Caenorhabditis remanei, Caenorhabditis sp. v, Rhabditis sp., and Pelodera teres. Copulation was observed in all crosses between Caenorhabditis species; however, none resulted in the generation of stable hybrid populations. No copulation was observed in crosses between Caenorhabditis males and Rhabditis or Pelodera females, even when congeneric females were present, suggesting that Caenorhabditis males are able to selectively recognize congeneric females by a short-range stimulus. All pairwise combinations of Caenorhabditis species were isolated to some degree by gametic mechanisms; 7 of 12 combinations were cross infertile and 5 of 12 were cross-fertile but had low brood sizes. In cross-fertile combinations, most hybrid embryos were inviable and arrested prior to gastrulation. Only in crosses of C. briggsae males to C. sp. v females did any hybrids survive embryogenesis. Most of these C. briggsae/C. sp. v hybrids arrested during larval development, and the few that reached adulthood invariably were female. These results are consistent with the presence of at least two lethal factors in the C. briggsae-C. sp. v combination: a maternal lethal factor in the cytoplasm of C. briggsae and a recessive lethal factor on the X chromosome of C. sp. v.  相似文献

Summary A demographic study was carried out on two closely related species of the isopods Jaera (albifrons) ischiosetosa and J. (a.) albifrons and their F 1 hybrids. The results from the 16 possible combinations of crosses have permitted an analysis of the nature of the mechanisms assuring the isolation of the species studied. Although intraspecific crosses yield an immediate success, interspecific crosses in the absence of choice of mates progress only slowly during the course of weeks. The results of both crosses between hybrids and back-crosses turn out to be intermediate between those of intra- and interspecific crosses. The hybrids of the first generation are perfectly viable and their survival curves are identical to those of the parents. The fertility of parents in intra- and interspecific crosses is comparable, with the exception of the fragility of female descendants (heterogametic sex) in one direction of crossing. The fertility of the F 1 hybrids, however, crossed either among themselves or with their parents, is quite noticeably decreased: the time needed to double the size of the population is 2.5 times longer for the hybrids than for the parents. This hybrid breakdown completes the pre-fertilization isolating mechanisms: partial ecological isolation, and especially ethological isolation, is practically total when a choice of mates exists. The two species studied, for which demographic parameters are quite close, were raised together for ten generations and yielded only exceptional hybrids with a frequency which does not exceed that found under natural conditions.  相似文献

The method of polymerase chain reaction with random primers (RAPD PCR) was used to identify the progeny of the crosses between three sturgeon species, Amur sturgeon (Acipenser schrenckii Brandt, 1869), Siberian sturgeon (A. baerii Brandt, 1869), and sterlet (A. ruthenus Linnaeus, 1758). Using ten primers, genetic variation in 70 yearlings, produced in seven individual crosses: Acipenser schrenckii × A. schrenckii, A. baerii × A. baerii, A. ruthenus × A. ruthenus, A. schrenckii × A. baerii, A. baerii × A. schrenckii, A. schrenckii × A. ruthenus, and A. ruthenus × A. schrenckii was described and evaluated. It was demonstrated that the samples composed of hybrids from individual crosses were more variable than the samples of parental species. On the other hand, pooled samples of hybrids from two cross directions were genetically less variable than the pooled samples of their parents. The three main features of the hybrid RAPD profiles identified included: (1) preservation of marker DNA fragments of both parents in one genome; (2) presence of specific DNA fragments, absent from both parents; and (3) dependence of the frequency of some DNA fragments from the cross direction. Multidimensional scaling clearly distinguishes in the space of three coordinates the individuals of original species and the hybrid progeny with differentiation in the groups of direct and backcross hybrids. Analysis of relationships (UPGMA and NJ) pointed to substantial differentiation between the species, as well as between the species and hybrid progeny. Close genetic relationships between direct and backcross hybrids were demonstrated. Multilocus RAPD markers in association with statistical methods are considered to be the useful tool for discrimination of interspecific hybrids of sturgeon. Crosses were made in all combinations of the six diploid species of the Solarium nigrum complex that occur in North America. Some interspecific pollinations failed to yield viable seed; successful crosses gave rise to moderately to highly sterile F1 hybrids. Results of interspecific crosses suggested phylogenetic relationships that were not completely in accord with those suggested by morphology. Interspecific crosses also gave varied results. All interpopulational crosses within S. interius and S. sarachoides produced fully fertile hybrids. In contrast, hybrids within S. americanum and S. douglasii varied from fully fertile to almost completely sterile. Populations of S. pseudogracile could be divided into two groups which are geographically separated but not morphologically differentiated. Fully fertile hybrids resulted from crosses within a group, whereas crosses between groups gave hybrids with reduced fertility. Four crossing groups were observed within 5. nodiflorum; three of the parental groups are sympatric and are morphologically differentiated. Although hybrid sterility in interspecific crosses is sometimes used to support delimitation of species, the presence of sterility in intraspecific crosses suggests that such an interpretation is unwarranted for the S. nigrum complex. In most eudicot and monocot species, interspecific and interploidy crosses generally display abnormalities in the endosperm that are the major cause of a post‐zygotic hybridization barrier. In some eudicot species, however, this type of hybridization barrier can be overcome by the manipulation of ploidy levels of one parental species, suggesting that the molecular mechanisms underlying the species hybridization barrier can be circumvented by genome dosage. We previously demonstrated that endosperm barriers in interspecific and interploidy crosses in the genus Oryza involve overlapping but different mechanisms. This result contrasts with those in the genus Arabidopsis, which shows similar outcomes in both interploidy and interspecific crosses. Therefore, we postulated that an exploration of pathways for overcoming the species hybridization barrier in Oryza endosperm, by manipulating the ploidy levels in one parental species, might provide novel insights into molecular mechanisms. We showed that fertile hybrid seeds could be produced by an interspecific cross of female tetraploid Oryza sativa and male diploid Oryza longistaminata. Although the rate of nuclear divisions did not return to normal levels in the hybrid endosperm, the timing of cellularization, nucellus degeneration and the accumulation of storage products were close to normal levels. In addition, the expression patterns of the imprinted gene MADS87 and YUCCA11 were changed when the species barrier was overcome. These results suggest that the regulatory machinery for developmental transitions and imprinted gene expression are likely to play a central role in overcoming species hybridization barriers by genome dosage in the genus Oryza.  相似文献

以报春花属报春花组的小报春(Primula forbesii)和指叶报春组的岩生报春(Primula saxatilis)为亲本,对种间杂交的结实性及花粉管行为进行了观察。结果表明:小报春与岩生报春种间杂交表现为不亲和,正反交组合正常结实率为0,但正交、反交组合花粉在柱头表面的萌发和花粉管伸长过程有明显差异,小报春花粉授粉4h后可以在岩生报春柱头表面萌发,但花粉管伸长的速度明显比对照组[岩生报春(P)×岩生报春(T)、岩生报春(T)×岩生报春(P)]慢,并且花粉管生长弯曲,授粉192h后花粉管仍未到达子房;岩生报春的花粉可以在小报春柱头上正常萌发,授粉48h后花粉管到达子房;4个对照授粉组合均分别于24h(短花柱为母本)、48h(长花柱为母本)时完成受精过程。研究表明,岩生报春×小报春杂交存在受精前障碍,小报春×岩生报春杂交亲和性较好,并可通过幼胚拯救的方法获得组间杂种后代;花柱长度可能是影响种间杂交结实能力的因素之一。  相似文献

Summary Genetic relationships between 7 annual species of the genus Cicer, including the cultivated chickpea, have been studied. These species were assigned to 3 crossability groups. In each group interspecific hybrids could be obtained but their fertility differed considerably in the various cross combinations. Crosses between members of different groups yielded no viable seeds. The possibility of gene transfer from the wild species to the cultivated chickpea C. arietinum was also assessed. Only two species could be considered for this purpose, C. reticulatum, which is the wild progenitor of the cultivated species, and C. echinospermum, which is in the secondary gene pool of C. arietinum. A unique postzygotic reproductive barrier mechanism was found between the members of Group II, C. judaicum, C. pinnatifidum and C. bijugum. It is based on a disharmony in the growth rate of the stigma and the anthers at the time of anthesis of the F1 interspecific hybrid so that selfpollination is avoided. It is proposed that this kind of mechanism has been involved only when an effective spatial isolation between the three species had been obtained.  相似文献

探究不同松树种间杂交可育性,为松树种间杂交育种亲本选配提供依据,进而为松树的杂种优势研究积累材料。该研究利用随机区组设计,通过92个松树种间杂交组合与20个半同胞对照,分析9种松树种间杂交类型子代的平均球果产种数、平均球果产种量、百粒重、发芽率与成苗率5项育性指标,综合球果期、种期与苗期3个阶段的表现,利用模糊数学隶属函数法对不同松树种间杂交类型进行可育性综合评价。结果表明:加勒比松×湿地松(C×E)、湿地松×加勒比松(E×C)与湿地松×火炬松(E×T) 3种杂交类型育性表现优于或接近相应的半同胞对照,马尾松×湿地松(M×E)、马尾松×火炬松(M×T)、马尾松×加勒比松(M×C)、火炬松×加勒比松(T×C)、火炬松×马尾松(T×M)与火炬松×湿地松(T×E)杂交类型育性表现远低于半同胞对照,表现出低育性甚至不育。综合5项育性指标,亚组内杂交育性的整体表现明显优于亚组间杂交;湿地松与加勒比松为母本的3种杂交组合育性表现总体水平明显优于马尾松与火炬松为母本的另外6种杂交组合。9种杂交类型中除了火炬松×湿地松(T×E)外,均获得了具有生活力的杂交子代,尤其马尾松与3种国外松的杂交子代在相关研究中是首次获得,对松树种间杂交育种的研究具有重要意义。  相似文献

Artificial hybridizations were conducted with 37 species of four genera of the subtribe Peritylanae (Compositae). Crossability, pollen stainability, and meiotic behavior were examined for most of the resulting 61 F1 hybrid combinations. Intergeneric hybrids involving Amauria, Eutetras, Pericome, and Perityle, and infrageneric hybrids involving Perityle were readily obtained. Lower crossability and hybrid fertility were displayed by intergeneric crosses. The infrageneric crosses of Perityle were organized into two categories, intersectional and infrasectional, based upon the taxonomic sections that are recognized for the genus; sect. Pappothrix, sect. Laphamia, and sect. Perityle. In general, the crossability and hybrid fertility of intersectional crosses was markedly higher than that of intergeneric crosses. The results suggest a closer relationship between the three taxonomic sections of Perityle than between Perityle and other genera of the subtribe.  相似文献

The following Lolium species were shown to have 14 chromosomes: L. canariensis Steud. (= L. gracile Parl.), L. loliaceum (Bory and Chaub.) Hand.-Mazz., L. multiflorum Lam., L. perenne L., and L. temulentum L. B-chromosomes in addition to the normal complement were observed during metaphase I and anaphase I for L. persicum Boiss. and Hohen., L. remotum Schrank, L. rigidum Gaud., L. strictum Presl. (= L. rigidum?), and for several synthesized interspecific hybrids. General absence of B-chromosomes in diakinesis suggested that they originated by misdivision of A-chromosomes during prometaphase I. The B-chromosomes reached a maximum of eight but the number varied for microsporocytes of the same plant. B-chromosomes appeared spontaneously in progenies from sibbing of hybrid plants without supernumeraries but were also eliminated in the F2 from other sibbings. Degree of chromosome pairing during meiosis was unrelated to the subsequent presence of B-chromosomes. Normal pairing of seven bivalents was typical for L. perenne × L. multiflorum, L. perenne × L. rigidum, L. multiflorum × L. loliaceum, and L. rigidum × L. loliaceum. Between five and seven bivalents were recorded for L. multiflorum × L. persicum, L. multflorum × L. remotum, L. multiflorum × L. strictum, L. rigidum × L. persicum, L. rigidum × L. remotum, L. rigidum × L. strictum, and L. rigidum × L. temulentum. The results indicated that these Lolium species have a common and generally undifferentiated genome suggesting relatively recent speciation.  相似文献

A hybridization study was made utilizing 37 species of 10 sections in 41 intra- and intersectional combinations. A total of 721 pollinations was made with 138 fruits harvested; 89 produced viable crosses. Five intrasectional and 20 intersectional combinations resulted in successful hybrids. Among the intrasectional crosses, Phalaenanthe × Phalaenanthe exhibited the highest percentage of fruits and successful crosses while Callista × Callista produced no successful crosses. Eugenanthe × Eugenanthe exhibited low crossability. Among the intersectional crosses, Ceratobium × Phalaenanthe gave the highest percentage of successful crosses, indicating a close relationship between the two groups. Also, there appears to be a fairly close relationship between Latourea and Ceratobium based on crossability. Although the sectional classification of the genus by Schlechter on the basis of vegetative and floral morphology appears to be valid, crossability studies suggest that the Ceratobium and Phalaenanthe sections might be integrated and the Eugenanthe section separated into sections or subsections.  相似文献

Summary The inheritance of chloroplast (cp) DNA was examined in F1 hybrid progenies of two Populus deltoides intraspecific controlled crosses and three P. deltoides × P. nigra and two P. deltoides × P. maximowiczii interspecific controlled crosses by restriction fragment analysis. Southern blots of restriction digests of parental and progeny DNAs were hybridized to cloned cpDNA fragments of Petunia hybrida. Sixteen enzymes and five heterologous cpDNA probes were used to screen restriction fragment polymorphisms among the parents. The mode of cpDNA inheritance was demonstrated in progenies of P. deltoides × P. nigra crosses with 26 restriction fragment polymorphisms of cpDNA differentiating P. deltoides from P. nigra, as revealed by 12 enzyme-probe combinations, and in progenies of P. deltoides × P. maximowiczii crosses with 12 restriction fragment polymorphisms separating P. deltoides from P. maximowiczii, as revealed by 7 restriction enzyme-probe combinations. In all cases, F1 offspring of P. deltoides × P. nigra and P. deltoides × P. maximowiczii crosses had cpDNA restriction fragments of only their maternal P. deltoides parent. The results clearly demonstrated uniparental-maternal inheritance of the chloroplast genome in interspecific hybrids of P. deltoides with P. nigra and P. maximowiczii. Intraspecific P. deltoides hybrids also had the same cpDNA restriction fragments as their maternal parent. Maternal inheritance of the chloroplast genome in Populus is in agreement with what has been observed for most other angiosperms.  相似文献

To investigate the cause of variation in the interspecific crossability of Brassica napus, three different genotypes were studied in respect of their reproductive behavior after pollination with B. juncea. There were great differences among maternal genotypes in allowing foreign pollen to germinate on and penetrate into their stigmas, leading to a wide diversity of interspecific fertilization. The division of the hybrid primary endosperm nucleus and zygote appeared normal in all combinations of crosses. While the abundant free nuclei of the endosperm developed properly and never became cellular, the embryos degenerated as early as 10 days after pollination when the cultivar Rucabo, which had the highest fertilization record with species of B. juncea, was involved. When 81007 was used as female parent, the endosperm grew a little but the embryo halted at the heart-torpedo stage. Lack of nourishment might be responsible for the observed embryo abortion. Among the sic hybrid combinations, the cross 84014A x Changyang hunagjie was the only one where endosperm tissue was observable and an abnormal embryo occurred prior to cellular endosperm formation. Apart from the three typical embryological features, significant variation was also demonstrated among each of the cross combinations. Genetic diversity appears to exist not only between varieties, but also within cultivars. In addition, methods for developing interspecific crossable lines are discussed.  相似文献

The data on polymorphism and inheritance of the seed storage protein helianthinin are presented. The results of hybrid analysis indicate that in the annual sunflower Helianthus annuus, helianthinin synthesis is controlled by at least three loci: HelA, HelB, and HelC. Codominant alleles controlling different electrophoretic variants of polypeptides were identified at each of the loci. The HelA locus was inherited independently of HelB and HelC in a series of dihybrid crosses. The frequencies of recombination between loci HelB and HelC estimated in F2 and BC of two crossing combinations were respectively 21.8 and 19.0%. Segregation of the HelC-controlled variants in the progenies from the crosses of cultured sunflower with annual wild species and forms corresponded to that theoretically expected for Mendelian inheritance. The maternal type of helianthinin inheritance was observed in the progenies from the crosses of inbred H. annuus L. lines with perennial diploid and polyploid HelianthusL. species. Altered expression of the HelClocus was detected in some hybrid combinations. These alterations appeared in early (F1, F2) hybrid generations and were similar in different hybrid combinations. They did not depend on the perennial paternal species being more influenced by the maternal genotype and by the mode of obtaining hybrids (in an embryo culture or in the field). These results are explained by genomic shock generated by hybridization of genetically incompatible species.  相似文献

Ath6 is a novel quantitative trait locus associated with differences in susceptibility to atherosclerosis between C57BL/6J (B6) and C57BLKS/J (BKS) inbred mouse strains. Combining data from an intercross and a backcross (1593 meioses) between mice from B6 and BKS strains and from The Jackson Laboratory interspecific backcross panels, (C57BL/6J ×Mus spretus) F1× C57BL/6J and (C57BL/6J × SPRET/Ei) F1× SPRET/Ei, we constructed a consensus genetic map and narrowed Ath6 to a 1.07 ± 0.26 cM interval between the anonymous DNA marker D12Pgn4 and the gene Nmyc1. This region is near the proximal end of murine Chromosome (Chr) 12, which is homologous to the human chromosomal region 2p24-p25. Marker order in the Ath6 region was concordant among the two crosses and The Jackson Laboratory interspecific backcross panels. This high resolution map rules out candidate genes encoding apolipoprotein B, syndecan 1, and Adam17. The two Ath6 crosses have a combined potential resolution of 0.06 cM. The aim of this study is to investigate the evolution of intrinsic postzygotic isolation within and between populations of Mimulus guttatus and Mimulus nasutus. We made 17 intraspecific and interspecific crosses, across a wide geographical scale. We examined the seed germination success and pollen fertility of reciprocal F1 and F2 hybrids and their pure-species parents, and used biometrical genetic tests to distinguish among alternative models of inheritance. Hybrid seed inviability was sporadic in both interspecific and intraspecific crosses. For several crosses, Dobzhansky–Muller incompatibilities involving nuclear genes were implicated, while two interspecific crosses revealed evidence of cytonuclear interactions. Reduced hybrid pollen fertility was found to be greatly influenced by Dobzhansky–Muller incompatibilities in five out of six intraspecific crosses and nine out of 11 interspecific crosses. Cytonuclear incompatibilities reduced hybrid fitness in only one intraspecific and one interspecific cross. This study suggests that intrinsic postzygotic isolation is common in hybrids between these Mimulus species, yet the particular hybrid incompatibilities responsible for effecting this isolation differ among the populations tested. Hence, we conclude that they evolve and spread only at the local scale.  相似文献

Summary

Four species of sea urchins of the genus Echinometra, designated species A, B, C, and D, occur along the coast of Okinawa; they are distinguished by color pattern and other characteristics. The two most distinct species, Echinometra sp. A (Ea) with white-tipped spines and Echinometra sp. D (Ed) with nearly black spines (possibly E. oblonga), were examined for potential hybridization through a series of fertilization experiments and rearing of the resulting hybrids. Fertilization was reciprocally asymmetrical; Ed ova were readily fertilized by sperm of Ea, but less than 20% of the ova of Ea were fertilized by sperm of Ed. Nevertheless, hybrids resulting from crosses in both directions developed normally through larval and juvenile stages to produce sexually mature adults. Larvae of Ed♀×Ea♂ and Ed♂×Ed♀ were larger than those of Ea♂ × Ed♀ and Ea♂ × Ea♀, reflecting the larger eggs of Ed. However, Ea♂×Ea♀ juveniles and later stages, up to maturity, were consistently larger than those of equivalent ages of the other crosses; largest to smallest were Ea♂×Ea♀, Ed♂×Ea♀, Ea♂ × Ed♀, and Ed♂×Ed♀, respectively. Ova and sperm of the hybrids were viable and fertilizable in all combination of crosses, although fertilization of those of Ed♂×Ea♀ hybrids was consistently higher than those of Ea♂ × Ed♀ hybrids. These experiments represent the first reported successful production of hybrid sea urchins with viable gametes. They indicate that reproductive isolation is achieved by prezygotic isolating mechanisms in these two species. Of these mechanisms, gametic incompatibility is probably only partly involved, if at all, and differences in spawning times, habitat segregation, or other factors appear to be important for maintaining reproductive isolation in these closely related species.  相似文献

Shorea (Dipterocarpaceae) is a large genus in which many closely related species often grow together in Southeast Asian lowland tropical rain forests. Many Shorea species share common pollinators, and earlier studies suggested occurrence of interspecific hybridization and introgression. Here, we show morphological and molecular evidence of hybridization between Shorea species. In the census of all the trees of Shorea curtisii, Shorea leprosula, and Shorea parvifolia (>30 cm dbh) within the 164-ha area of Bukit Timah Nature Reserve in Singapore, we found 21 morphologically recognizable hybrid individuals. All of the putative hybrids could be distinguished obviously from the parental species on the basis of vegetative characters. Population genetic analysis of DNA sequences of two nuclear (GapC and PgiC) and chloroplast (trnL-trnF) regions demonstrated that each of the three species had several species-specific mutations. The nuclear sequences of the putative hybrids were heterozygote at all the species-specific sites between two parental species. Hybrid between S. curtisii and S. leprosula was found most, while S. curtisii × S. parvifolia and S. leprosula × S. parvifolia hybrids were also found. Almost no shared polymorphism between populations of the parental species suggests rarity of introgression. The study indicated that natural hybridization between sympatric Shorea species should not be uncommon, but all of the hybrid individuals were F1, and the post-F1 hybrids were considerably rare.  相似文献

In interspecific matings between the species Drosophila virilis and Drosophila texana, female sterility can be observed in F2 backcross females and in F2 hybrid females. The results presented in this report show that the female sterility, whenever it exists, is due to prevention of vitellogenin synthesis in the fat body, but other abnormalities such as defects with the hybrid ovaries are not excluded. The observation that sterility appears among females from backcrosses suggests that incompatibilities between interspecific genes may cause female sterility even in the presence of a complete habloid genome from one or the other species. Yet, the parallel observation that female sterility appears only in hybrid females with recombinant chromosomes indicates that sterility results when conspecific combinations of genes on the same chromosome are broken by interspecific recombination. © 1996 Wiley-Liss, Inc.  相似文献
