Leaf structure in Doxantha unguis-cati is polymorphic. The usual mature compound leaf is composed of two lanceolate leaflets and a terminal tripartite spine-tendril. Leaf primordia are initiated simultaneously in pairs on opposite flanks of the shoot apical meristem by periclinal cell divisions in the third subsurface layer of the peripheral flank meristem. Two leaflet primordia are the first lateral appendages of the compound leaf. Initiation of these leaflet primordia occurs on the adaxial side of a compound leaf primordium 63–70 μm long. Lamina formation is initiated at the base of a leaflet primordium 70–90 μm long and continues acropetally. Mesophyll differentiation occurs in later stages of development of leaflets. The second pair of lateral appendages of the leaf primordium differentiate as prongs of the tendril. Initiation of the second pair of lateral appendages occurs on the adaxial side of a primordium approximately 168 μm long. Acropetal procambialization and vacuolation of cells extend to the apex of tendrils about 112 μm long, restricting the tendril meristem to the adaxial side of the primordium and resulting in curvature of the tendril. The tendril meristem is gradually limited to a more basipetal position as elongation of apical cells continues. Initiatory divisions and early ontogenetic stages of leaflets and tendrils are similar. Their ontogeny differs when the lateral primordia are approximately 70 μm long. Marginal and submarginal initials differentiate within leaflets but not in tendrils. Apical growth of tendrils ceases very early in ontogeny as compared with leaflets.  相似文献

Tendrils on long shoots of Parthenocissus inserta occur in a regular pattern opposite the alternate distichous leaves at two successive nodes of each three nodes. Ontogenetic study shows that the tendril is initiated at the flank of the shoot apex during the second plastochron in an essentially axillary position. It is carried upward with growth of the internode above the axillant leaf and ultimately is situated opposite the next younger leaf. In a rhythmic pattern a different group of appendages is produced by the shoot apex at each node in the sequence of three. In acropetal order these are: at the tendrilless node the leaf subtends an axillary bud complex which in turn subtends a tendril; the leaf at the lower tendril-bearing node directly subtends a tendril, and the leaf at the upper tendril-bearing node subtends an axillary bud. Tendril primordia were not induced to develop as foliaceous shoots when cultured in vitro or in decapitation experiments, indicating that the meristem which becomes a tendril is determined early in its inception. Although built on a shoot pattern, the tendril is regarded as an organ sui generis with a possible relationship to the inflorescence. The morphological nature of the tendril is discussed in the light of theories postulated in the literature.  相似文献

Leaves of flowering plants are produced from the shoot apical meristem at regular intervals, with the time that elapses between the formation of two successive leaf primordia defining the plastochron. We have identified two genetic axes affecting plastochron length in Arabidopsis thaliana. One involves microRNA156 (miR156), which targets a series of SQUAMOSA PROMOTER BINDING PROTEIN-LIKE (SPL) genes. In situ hybridization studies and misexpression experiments demonstrate that miR156 is a quantitative, rather than spatial, modulator of SPL expression in leaf primordia and that SPL activity nonautonomously inhibits initiation of new leaves at the shoot apical meristem. The second axis is exemplified by a redundantly acting pair of cytochrome P450 genes, CYP78A5/KLUH and CYP78A7, which are likely orthologs of PLASTOCHRON1 of rice (Oryza sativa). Inactivation of CYP78A5, which is expressed at the periphery of the shoot apical meristem, accelerates the leaf initiation rate, whereas cyp78a5 cyp78a7 double mutants often die as embryos with supernumerary cotyledon primordia. The effects of both miR156-targeted SPL genes and CYP78A5 on organ size are correlated with changes in plastochron length, suggesting a potential compensatory mechanism that links the rate at which leaves are produced to final leaf size.  相似文献

Expression of KNOX (KNOTTED1-like homeobox) genes in the shoot apical meristem of Arabidopsis is required for maintenance of a functional meristem, whereas exclusion of KNOX gene expression from leaf primordia is required for the elaboration of normal leaf morphology. We have constructed a steroid-inducible system to regulate both the amount and timing of KN1 (KNOTTED1) misexpression in Arabidopsis leaves. We demonstrate that lobed leaf morphology is produced in a dose-dependent manner, indicating that the amount of KN1 quantitatively affects the severity of lobing. The KN1-glucocorticoid receptor fusion protein is not detected in leaves in the absence of steroid induction, suggesting that it is only stable when associated with steroid in an active state. By using a second inducible fusion protein to mark exposure of leaf primordia to the steroid, we determined the stage of leaf development that produces lobed leaves in response to KN1. Primordia as old as plastochron 7 and as young as plastochron 2 were competent to respond to KN1.  相似文献

Passiflora foetida bears an unbranched tendril, one or two laterally situated flowers, and one accessory vegetative bud in the axil of each leaf. The vegetative shoot apex has a single-layered tunica and an inner corpus. The degree of stratification in the peripheral meristem, the discreteness of the central meristem, and its centric and acentric position in the shoot apex are important plastochronic features. The procambium of the lateral leaf trace is close to the site of stipule initiation. The main axillary bud differentiates at the second node below the shoot apex. Adaxial to the bud 1–3 layers of cells form a shell-zone delimiting the bud meristem from the surrounding cells. A group of cells of the bud meristem adjacent to the axis later differentiates as an accessory bud. A second accessory bud also develops from the main bud opposite the previous one. A bud complex then consists of two laterally placed accessory bud primordia and a centrally-situated tendril bud primordium. The two accessory bud primordia differentiate into floral branches. During this development the initiation of a third vegetative accessory bud occurs on the axis just above the insertion of the tendril. This accessory bud develops into a vegetative branch and does not arise from the tissue of the tendril and adjacent two floral buds. The trace of the tendril bud consists of two procambial strands. There is a single strand for the floral branch trace. The tendril primordium grows by marked meristematic activity of its apical region and general intercalary growth.  相似文献

Preliminary studies establishing relationships between leaf plastochron index and Epilobium hirsutum L. shoot growth provide a method for rigorous selection of plants utilized in experiments designed to test the working hypothesis that endogenous auxin gradient interactions are factors of phyllotactic control in this species. Application of N-1-naphthylphthalamic acid (NPA), an auxin transport inhibitor, to one of the youngest bijugate primordia on the shoot meristem results in increased growth of the treated primordium. Fasciation between the treated primordium and one of the next primordia to be initiated alters relative vertical spacing of primordia. Angular shifts between subsequent primordia result in spiral transformation of Epilobium bijugate phyllotaxy. Application of α-4-chlorophenoxyisobutyric acid (CPIB), an auxin antagonist, to one of the youngest bijugate primordia on the shoot meristem results in decreased growth of the treated primordium that alters both radial and vertical spacing of primordia. This is followed by angular shifts between subsequent primordia resulting in spiral transformation of the bijugate phyllotaxy. Changes in the growth parameters of NPA- and CPIB-treated shoots are similar. Relative plastochron rates of radial and vertical shoot growth of induced spiral shoots are about half those of lanolin paste control shoots, as are the plastochrons and relative plastochron rates of leaf elongation. Treated shoot meristems have eccentricities of 0.5 as compared to bijugate control meristem eccentricities of 0.7. No significant difference is apparent between basal transverse areas of treated and control shoot meristems. The relative chronological rates of growth of treated shoots are not significantly different from those rates of control shoots. Spiral transformation results from changes in relative positions of leaf primordia insertion on the shoot meristem, not from changes in growth of treated shoots. These changes are accompanied by an increased rate of leaf initiation on a more circular shoot meristem. Existing theoretical models of phyllotaxy are discussed in relation to these chemically induced changes of Epilobium leaf arrangement.  相似文献

In higher plants, leaves initiate in constant spatial and temporal patterns. Although the pattern of leaf initiation is a key element of plant shoot architecture, little is known about how the time interval between initiation events, termed plastochron, is regulated. Here, we present a detailed analysis of plastochron2 (pla2), a rice (Oryza sativa) mutant that exhibits shortened plastochron and precocious maturation of leaves during the vegetative phase and ectopic shoot formation during the reproductive phase. The corresponding PLA2 gene is revealed to be an orthologue of terminal ear1, a maize (Zea mays) gene that encodes a MEI2-like RNA binding protein. PLA2 is expressed predominantly in young leaf primordia. We show that PLA2 normally acts to retard the rate of leaf maturation but does so independently of PLA1, which encodes a member of the P450 family. Based on these analyses, we propose a model in which plastochron is determined by signals from immature leaves that act non-cell-autonomously in the shoot apical meristem to inhibit the initiation of new leaves.  相似文献

The ontogeny of Epilobium hirsutum grown under natural summer photoperiod in a glasshouse was divided into vegetative, early transitional, transitional, and floral stages. Bijugate phyllotaxy, common to both the vegetative and early transitional stages, is transformed into spiral phyllotaxy during the transitional stage by an initial change in the divergence angle of a single primordium inserted at a unique level on the shoot. Leaf primordia subsequently are inserted in a spiral arrangement in the indeterminate floral shoot apex. The early transitional shoot apical meristem is about 1.5 times the volume of the vegetative meristem but expands at about two-thirds the relative plastochron rate of volume increment of the vegetative meristem. There are progressive decreases in the plastochron and relative plastochron rates of radial and vertical shoot growth through ontogeny. Relative chronological rates of shoot growth, however, are not altered during ontogeny. Spiral transformation results from changes in the relative points of insertion of leaf primordia on the shoot meristem. These changes are accompanied by an increased rate of primordia initiation on a more circular shoot meristem. The change in phyllotaxy during ontogeny is similar to that which was artificially induced by chemical modification of auxin concentration gradients in the shoot apex, with the additional feature that there is an initial increase in the volume of the shoot meristem prior to the natural spiral transformation. Size of the shoot apical meristem, however, appears to have little influence on Epilobium phyllotaxy; but the geometric shape of the meristem is well correlated with bijugate to spiral transformations. This suggests that geometric parameters of the shoot meristem should be considered in theoretical models of phyllotaxy.  相似文献

Leaf development was studied in the heterophyllous aquatic plant Hippuris vulgaris in order to characterize the developmental events that lead to the formation of aerial- vs. submerged-type leaves. Recent evidence that abscisic acid regulates leaf development in this species provided a basis for using abscisic acid as a developmental tool to accurately control leaf development. We found that leaf primordia were fully competent to develop into either aerial- or submerged-type leaves until the 10th plastochron, when they were ca. 300 μm long. Also, leaves between about the 10th and 21st plastochron formed sectored transition leaves (i.e., the basipetal portion was composed of aerial-type tissue and the apical portion was composed of submerged-type tissue, or vice versa), indicating that tissue determination as one or the other leaf type occurred on a local, as opposed to whole-leaf, level. Finally, no significant difference was observed between the apical dimensions of aerial or submerged-type shoots. These results indicate that the final determination of Hippuris vulgaris leaves occurs a) relatively late in leaf development, and b) independently of the shoot apex, and provide a basis for using this plant in further studies concerning leaf determination and pattern formation (e.g., stomates, lateral venation) in plants.  相似文献

The aim of the work was to report morphological changes whichoccur in the shoot apex during the morphogenetic switch to floweringin the model long day (LD) plant, Sinapis alba. During the floraltransition induced by 1 LD the growth rate of all componentsof the shoot apex is modified profoundly. The earliest changes,detected at 24 h after start of LD, include a decrease in plastochronduration and an increase of growth of leaf primordia. One daylater, the meristem dome starts to increase in volume, apicalinternodes have an increased height and there is a precociousoutgrowth of axillary meristems. All these changes precede initiationof flower primordia, which starts at about 60 h after the startof LD. Later changes include meristem doming, a decrease inthe plastochron ratio and a shift to a more complex phyllotaxis.All the changes, except the decreased plastochron ratio, arecharacteristics of an apex with an increased tempo of growth.The stimulation of longitudinal growth (height of apical intemodes)is more marked and occurs earlier than the reduction of radialgrowth (plastochron ratio). The early ontogeny of the pinnately, palmately, and ternately compound leaves in the Lardizabalaceae was studied by SEM. The leaf primordium of each of the three leaf types emerges as an identical short protrusion on the shoot apex; the leaf primordium produces the first leaflet initials laterally on its margin. Successive acropetal growth of the leaf axis and the following inception of the leaflet primordia are responsible for the pinnately compound leaf, whereas short basipetal growth accompanied with initiation of two or more pairs of leaflet initials results in a palmately compound leaf. If no elongation of the leaf axis nor additional inception of leaflet primordia occur during early ontogeny, a ternate leaf ensues.  相似文献

The compound leaf primordium of pea represents a marginal blastozone that initiates organ primordia, in an acropetal manner, from its growing distal region. The UNIFOLIATA (UNI) gene is important in marginal blastozone maintenance because loss or reduction of its function results in uni mutant leaves of reduced complexity. In this study, we show that UNI is expressed in the leaf blastozone over the period in which organ primordia are initiated and is downregulated at the time of leaf primordium determination. Prolonged UNI expression was associated with increased blastozone activity in the complex leaves of afila (af), cochleata (coch), and afila tendril-less (af tl) mutant plants. Our analysis suggests that UNI expression is negatively regulated by COCH in stipule primordia, by AF in proximal leaflet primordia, and by AF and TL in distal and terminal tendril primordia. We propose that the control of UNI expression by AF, TL, and COCH is important in the regulation of blastozone activity and pattern formation in the compound leaf primordium of the pea.  相似文献

Leaf initiation rate, leaf primordium growth rates, and apical volume growth rates were determined for seedlings of Triticum aestivum cv. Ramona 50 under controlled environmental conditions. Three leaf primordia are present in the caryopsis, and three more leaves are initiated within the first two weeks after germination with a mean plastochron length of 95.5 hr. Volume growth rates of the apical region were determined on six apices which had six primordia each. The mean radial expansion rate was 0.467/plastochron, and the vertical expansion rate was 0.457/plastochron. The volume expansion rate was 1.393/plastochron. The mean volume doubling time was 0.498 plastochrons or 47.1 hr.  相似文献

Growth of Ranunculus shoots through ontogeny is quantified by techniques utilizing scanning electron microscopy and studies on living plant material. The order of the contact parastichy phyllotaxy in the apical system is related to the relative plastochron rates of growth of the shoot. There is a change in the (2, 3) contact parastichy pattern of vegetative phyllotaxy to a transitional (3, 5) contact pattern which is maintained through sepal production. Formation of a 5(1, 1) whorl of petal primordia establishes a (5, 8) contact pattern with the sepal primordia. Subsequent initiation of stamen primordia, in spiral sequence, results in (5, 8, 13) triple contacts between petal and stamen primordia. The stamen primordia and carpel primordia arrangement is characterized by a (8, 13) contact parastichy pattern of phyllotaxy. Through ontogeny the volume of the shoot apex progressively increases but the shape of the apex, described by a second degree polynomial, remains constant. The plastochron and the relative plastochron rates of radial and vertical displacement of primordia progressively decrease during transition but there is no alteration of the chronological rate of apical expansion. The change in contact parastichy phyllotaxy through ontogeny is interpreted as a change in the relative positions of primordia insertion on the apex resulting from an increase in apical volume and an increased rate of primordia initiation.  相似文献

The origin and development of the tendrils were studied in 16 species of the Vitaceae: Ampelopsis (7 sp.), Parthenocissus (4 sp.), Vitis (3 sp.), and Tetrastigma (1 sp.). Two types of arrangement of leaf and tendril occur: (a) two successive nodes have leaf-opposed tendrils alternating with each other, followed by a third node, with a leaf unopposed by the tendril; (b) all the nodes have leaf-opposed tendrils. The tendril, like a leaf, is a lateral product of the apical meristem of the shoot. A leaf opposite a tendril is initiated earlier than the tendril. Anticlinal and periclinal divisions in the second and/or third layer of the peripheral meristem of the shoot apex initiate the tendril. The procambium of the tendril first appears towards its abaxial side. Vascularization of the tendril is independent of the axillary bud of the next node below. The positional relationship of the nodal plate vis-à-vis the leaf-opposed tendril shows that the tendril and the leaf belong to the same node. Histological evidence does not show the uplifting of the tendril to the next node above during internodal differentiation. Ontogenetic and morphologic correlation and homology between the inflorescence and the tendril do not substantiate that the tendril in the Vitaceae is an organ sui generis. All available evidence indicates that the tendril is an extra-axillary lateral branch.  相似文献

Periclinal cell divisions in vegetative shoot apices of Pisumand Silene were recorded from serial thin sections by mappingall the periclinal cell walls formed less than one cell cyclepreviously. The distribution of periclinal divisions in theapical domes corresponded to the distributions subsequentlyoccurring in the apices when the young leaf primordia were forming.In Pisum, periclinal divisions were almost entirely absent fromthe I1 region of the apical dome for half a plastochron justafter the formation of a leaf primordium and appeared, simultaneouslyover the whole of the next potential leaf site, about half aplastochron before the primordium formed. In Silene periclinaldivisions seemed to always present in the apical dome at thepotential leaf sites and also round the sides of the dome wherethe ensheathing leaf bases were to form. Periclinal divisionstherefore anticipated the formation of leaf primordia by occuring,in Pisum about one cell cycle and in Silene two or more cellcycles, before the change in the direction of growth or deformationof the surface associated with primordial initiation. We describe two recessive alleles of a rice heterochronic gene, plastochron1-1 (pla1-1) and pla1-2, that reduce the length of the plastochron to approximately half that of the wild type. Because the onset of the reproductive phase in pla1 was not temporally affected, the number of leaves produced in the vegetative phase was nearly twice that produced in the wild type. Panicle development was severely disturbed in pla1 mutants. In pla1-1, many primordia of primary rachis branches were converted into vegetative shoots. These ectopic shoots repeated the initiation of panicle development and the conversion of primary rachis branches into shoots. In the weak allele pla1-2, however, only the basal one or two primordia developed as vegetative shoots, and the remaining primordia developed to produce a truncated panicle. These results indicate that both vegetative and reproductive programs are expressed simultaneously during the reproductive phase of pla1; however, the degree varied depending on the strength of the allele. Accordingly, pla1 is a heterochronic mutation that extends the vegetative period. The shoot apical meristem of pla1 was larger than that of the wild type, although the shape was not modified. An in situ hybridization experiment using the histone H4 gene as a probe revealed that cell divisions are accelerated in the pla1 meristem. The PLA1 gene is considered to regulate the duration of the vegetative phase by controlling the rate of leaf production in the meristem.  相似文献

The origin and development of anlagen (undifferentiated primordia),inflorescences, tendrils and flowers in the grape cv. Shirazhas been investigated by scanning electron microscopy. Anlagenarise terminally by bisection of the apex of the so-called latentbud. The axis of the latent bud is continued by the originalapex and anlagen are displaced laterally. Micrographs presentedhere favour the interpretation of the grape-vine shoot as amonopodium. Anlagen formed distal to the 10th node of container grown vinesformed inflorescence primordia when plants were grown at hightemperatures (33°C day-28°C night). At lower temperatures(21°C day, 16°C night or 18°Cday, 13°C night)anlagen formed distal to the 10th node grew into tendril primordia.At basal nodes anlagen gave rise to shoot primordia. Investigation of the development and organography of the shoot systems of Microgramma vacciniifolia and M. squamulosa was undertaken for the purpose of determining: (1) the features of shoot growth that are responsible for the distinctive vining character of these epiphytic ferns; and (2) the mode of origin of branches and their contrast with leaf initiation. Shoots of both species are dorsiventral and plagiotropic (i.e., parallel to the substrate) in habit. Since the shoot apical meristem is radial in transectional symmetry, shoot dorsiventrality in Microgramma is a postgenital or secondary developmental event, and its inception is related to the initiation of lateral appendages. Leaves and buds arise in a distichous phyllotaxis and occupy opposite and alternating positions on the dorsal surfaces and flanks of the rhizome. Endogenous roots are initiated in two rows from the ventral surface of the stem, in the vicinity of the rhizome meristem; however, they do not emerge from the rhizome until some distance behind the tip and do not elongate until the region of substrate contact. We conclude that the vining nature of this fern rhizome is a result of precocious internodal elongation and the concomitant delay of leaf and bud expansion in the region of stem elongation. In addition, observation of branch origin confirms previous suggestions that branching in Microgramma is strictly lateral and extra-axillary and not a dichotomous derivative as proposed by some workers. Leaf and bud primordia differ not only in the nature of their respective vascular supplies but also in their actual course of initiation. In the case of the leaf, the primordium is precociously emergent and exhibits a lenticular apical cell at its summit when it is only one plastochron removed from the flanks of the apical meristem. By contrast, initials of the bud primordium divide less actively and remain in a sunken position for at least 5–6 plastochrons; only when the bud apex becomes expanded and emergent does a tetrahedral apical cell become recognizable at the tip of the bud promeristem. Because of the distinctive pattern of branch and leaf origin, as well as the lack of adventitious and phyllogenous origin of branch primordia, we suggest that the shoot of Microgramma is a useful test organism for the re-examination of the problem of leaf and bud determination in the ferns.  相似文献
