Some applications of chimeric cellular models are presented to study the biological activities of steroid hormones. We have used several chimeric constructs encoding the DNA binding domain of Gal4 yeast protein fused to the hormone binding domain of various steroid receptors (MR, PR, GR and ER). Interactions of these chimeric receptors with a 17-mer DNA sequence, specific for Gal-4, control expression of the firefly luciferase as a reporter gene. Stable transfected cell lines expressing the firefly luciferase under the control of different steroids were established and an efficient and easy sub-cloning was allowed with the help of an imaging system using a single-photon-counting camera. In the cell lines obtained, the bioluminescent response can be easily measured and thus used to measure specific biological activities of steroid agonists or antagonists. We observed that the responses are effector-concentration-dependent and their biological activities will be compared to those of native receptors.  相似文献

The 6-His tagged firefly luciferase was highly expressed in E. coli and purified to homogeneity by affinity chromatography and gel filtration. After immunizing Balb/c mice with the antigen, 6 hybridomas clones were found to secrete monoelonal antibodies (mAbs) and the mAbs were also purified separately. The competitive binding experiments show that 2 mAbs can bind heat-denatured antigen or its proteolytic fragments but not the native lueiferase, suggesting that their epitopes might be accommodated in the internal segments of the protein. On the other hand, the other 4 mAbs are capable of binding both native and denatured antigens. It infers that their epitopes locate in the segments on the protein surface. The results also suggest that the six mAbs are all sequence-specific.  相似文献

A stably transfected CHO cell line (LUCLEAD) was used where the coding region of native Firefly luciferase was linked to the 3'-UTR of the bovine growth hormone, and the 5'-nucleotides coding for the albumin signal peptide were linked to the N-terminal end of the luciferase coding region. Incubation of cells with 1 or 2 mM sodium butyrate (SB) for 72 h had no effect on cell growth since cultures reached confluency at the same time as control cells. Although cell cultures incubated with SB at a concentration of 4 mM were only about 60% confluent the luciferase content was about 5-fold higher than that in control cells. Cells incubated with either 1 or 2 mM SB showed intermediate levels of luciferase content. The amount of the chaperone BiP in the cells was not affected by incubation with SB. The results indicate that SB can be used to effectively promote synthesis of recombinant luciferase.  相似文献

In order to clarify the mechanism of food-antigen recognition, the proliferative responses of peripheral blood mononuclear cells (PBMCs) to native, heat-denatured or pepsin-treated ovalbumin (OA) were investigated in 16 hen's egg-sensitive patients with atopic dermatitis (AD). Seven of them had hypersensitivity to boiled hen's egg and others had not. The responses of PBMCs to heat-denatured OA were lower than those to native OA in the patients without hypersensitivity to boiled hen's egg. However, there were no differences of the responses of PBMCs between heat-denatured OA and native OA in the patients with hypersensitivity to boiled hen's egg. Moreover, the reduction of the responses of PBMCs to pepsin-treated OA was recognized in six out of seven patients. The primary structure of OA did not change by heating or pepsin treatment according to sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). These results suggested that the secondary structure of OA changed in connection with the reduction of the responses of PBMCs to denatured OA. In addition, we demonstrated the suppressive effect of anti-HLA-DR and anti-HLA-DQ monoclonal antibodies on the proliferative response of PBMCs to OA. 植物对邻体根系的表型可塑性是指与无邻体对照相比, 即使个体平均可获取土壤资源相同, 在有邻体根系存在时植物也会改变根系生物量分配, 并影响其他功能性状和适合度。表型可塑性进化假说(evolution of plasticity hypothesis)认为外来植物在入侵地进化出了更强的表型可塑性。对该假说的验证多集中于外来植物对光照、水分、养分以及天敌等的可塑性进化, 但对邻体根系的可塑性在入侵植物中是否发生进化尚未见报道。我们采用同质园实验比较了喜旱莲子草(Alternanthera philoxeroides)入侵地(美国)和原产地(阿根廷)各5个基因型的适合度与功能性状对同基因型邻体根系的可塑性。结果表明: 喜旱莲子草的根冠比(P = 0.088)和比叶面积(P = 0.007)对同基因型邻体根系的可塑性在入侵地和原产地基因型间存在差异: 入侵地基因型在有邻体根系时根冠比和比叶面积增加, 而原产地基因型则相反。但是, 总生物量、贮藏根生物量、比茎长和分枝强度对邻体根系的可塑性在入侵地和原产地间没有显著差异。此外, 与分隔邻体根系相比, 同基因型邻体根系存在时总生物量(+9.9%)和贮藏根生物量(+13.9%)显著增加, 比茎长(-9.5%)显著降低。最后, 与原产地基因型相比, 总体上入侵地基因型的总生物量(+62.0%)和贮藏根生物量(+58.9%)增加, 比茎长(-28.5%)和分枝强度(-42.8%)降低。这些结果表明喜旱莲子草入侵地基因型与资源利用相关功能性状(如根冠比和比叶面积)对邻体根系的可塑性方向与原产地基因型相反; 但适合度和株型相关性状(如比茎长和分枝强度)对同基因型邻体根系的可塑性与原产地没有差异。  相似文献

Alien predators are widely considered to be more harmful to prey populations than native predators. To evaluate this expectation, we conducted a meta-analysis of the responses of vertebrate prey in 45 replicated and 35 unreplicated field experiments in which the population densities of mammalian and avian predators had been manipulated. Our results showed that predator origin (native versus alien) had a highly significant effect on prey responses, with alien predators having an impact double that of native predators. Also the interaction between location (mainland versus island) and predator origin was significant, revealing the strongest effects with alien predators in mainland areas. Although both these results were mainly influenced by the huge impact of alien predators on the Australian mainland compared with their impact elsewhere, the results demonstrate that introduced predators can impose more intense suppression on remnant populations of native species and hold them further from their predator-free densities than do native predators preying upon coexisting prey.  相似文献

Response to environmental heterogeneity caused by human activity was analyzed on Iris pumila reciprocal transplants between native steppe and anthropogenic (planted pine forest) habitats that were monitored during several growing seasons in a protected area of Deliblato Sand. Morphometric traits exhibited significant plastic responses to the environmental variability between native and anthropogenic habitats that differed in light quantity and quality, as well as in some other ecological indices. Significant differentiation between populations occupying those habitats was also detected. Plastic responses and population differences were substantially related to the variation in general size and had the same direction, plastic responses being larger in magnitude. Estimates of reproductive and vegetative performance of reciprocal transplants detected home site advantage in the native open but not in the secondary shade habitat created under anthropogenic influence.  相似文献

The present investigation determined that native angiotensins II and III (ANG II and III) were equipotent as pressor agents when ICV infused in alert rats, whereas native angiotensin IV (ANG IV) was less potent. An analogue of each of these angiotensins was prepared with a hydroxyethylamine (HEA) amide bond replacement at the N-terminus, yielding additional resistance to degradation. These three angiotensin analogues, HEA-ANG II, HEA-ANG III, and HEA-ANG IV, were equivalent with respect to maximum elevation in pressor responses when ICV infused; and each evidenced significantly extended durations of effect compared with their respective native angiotensin. Comparing analogues, HEA-ANG II had a significantly longer effect compared with HEA-ANG III, and HEA-ANG IV, whereas the latter were equivalent. Pretreatment with the AT1 receptor subtype antagonist, Losartan (DuP753), blocked subsequent pressor responses to each of these analogues, suggesting that these responses were mediated by the AT1 receptor subtype. Pretreatment with the specific AT4 receptor subtype antagonist, Divalinal (HED 1291), failed to influence pressor responses induced by the subsequent infusion of these analogues. These results suggest an important role for Ang III, and perhaps ANG IV, in brain angiotensin pressor responses mediated by the AT1 receptor subtype.  相似文献

During introduction, invasive plants can be released from specialist herbivores, but may retain generalist herbivores and encounter novel enemies. For fast-growing invasive plants, tolerance of herbivory via compensatory regrowth may be an important defense against generalist herbivory, but it is unclear whether tolerance responses are specifically induced by different herbivores and whether specificity differs among native and invasive plant populations. We conducted a greenhouse experiment to examine the variation among native and invasive populations of Chinese tallow tree, Triadica sebifera, in their specificity of tolerance responses to herbivores by exposing plants to herbivory from either one of two generalist caterpillars occurring in the introduced range of Triadica. Simultaneously, we measured the specificity of another defensive trait, extrafloral nectar (EFN) production, to detect potential tradeoffs between resistance and tolerance of herbivores. Invasive populations had higher aboveground biomass tolerance than native populations, and responded non-specifically to either herbivore, while native populations had significantly different and specific aboveground biomass responses to the two herbivores. Both caterpillar species similarly induced EFN in native and invasive populations. Plant tolerance and EFN were positively correlated or had no relationship and biomass in control and herbivore-damaged plants was positively correlated, suggesting little costs of tolerance. Relationships among these vegetative traits depended on herbivore type, suggesting that some defense traits may have positive associations with growth-related processes that are differently induced by herbivores. Importantly, loss of specificity in invasive populations indicates subtle evolutionary changes in defenses in invasive plants that may relate to and enhance their invasive success.  相似文献

The selection of a genetic reporter can be difficult because of the wide range of genes available. In order to reduce the selection, we compared the performance of different reporter genes: firefly luciferase (Photinus pyralis lucFF), bacterial luciferase operon (Photorhabdus luminescens luxCDABE), green fluorescent protein (Aequorea victoria gfp), and red fluorescent protein (Discosoma sp. dsred) in whole-cell bacterial sensors. Escherichia coli sensor bacteria were engineered to contain a reporter plasmid that carries the reporter gene under the control of mercury- (mer from Tn21) or arsenite- (ars from R773) responsive regulatory units. Characteristics of the strains were studied by using different arsenite or mercury concentrations and incubation times. The lowest detectable concentration of analytes and the fastest responses were achieved with lucFF or luxCDABE as reporter genes. The fluorescent proteins, GFP and DsRed, gave responses at higher analyte concentrations and after significantly longer incubation times. The results indicate that luciferases are better reporters in whole-cell sensor bacteria.  相似文献

Invasive nitrogen-fixing plants drive vegetation dynamics and may cause irreversible changes in nutrient-limited ecosystems through increased soil resources. We studied how soil conditioning by the invasive alien Lupinus nootkatensis affected the seedling growth of co-occurring native plant species in coastal dunes, and whether responses to lupin-conditioned soil could be explained by fertilisation effects interacting with specific ecological strategies of the native dune species. Seedling performance of dune species was compared in a greenhouse experiment using field-collected soil from within or outside coastal lupin stands. In associated experiments, we quantified the response to nutrient supply of each species and tested how addition of specific nutrients affected growth of the native grass Festuca arundinacea in control and lupin-conditioned soil. We found that lupin-conditioned soil increased seedling biomass in 30 out of 32 native species; the conditioned soil also had a positive effect on seedling biomass of the invasive lupin itself. Increased phosphorus mobilisation by lupins was the major factor driving these positive seedling responses, based both on growth responses to addition of specific elements and analyses of plant available soil nutrients. There were large differences in growth responses to lupin-conditioned soil among species, but they were unrelated to selected autecological indicators or plant strategies. We conclude that Lupinus nootkatensis removes the phosphorus limitation for growth of native plants in coastal dunes, and that it increases cycling of other nutrients, promoting the growth of its own seedlings and a wide range of dune species. Finally, our study indicates that there are no negative soil legacies that prevent re-establishment of native plant species after removal of lupins.  相似文献

Aims Elevated ozone and CO2 can differentially affect the performance of plant species. Variation among native, exotic and invader species in their growth and defense responses to CO2 and ozone may shape CO2 and ozone effects on invasions, perhaps in part also due to variation between native and invasive populations of invaders.Methods We manipulated ozone (control or 100 ppb) and CO2 (ambient or 800 ppm) in a factorial greenhouse experiment in replicated chambers. We investigated growth and defense (tannins) of seedlings of Triadica sebifera from invasive (USA) and native (China) populations and pairs of US and China tree species within three genera (Celtis, Liquidambar and Platanus).Important findings Overall, ozone reduced growth in ambient CO2 but elevated CO2 limited this effect. T. sebifera plants from invasive populations had higher growth than those from native populations in control conditions or the combination of elevated CO2 and ozone in which invasive populations had greater increases in growth. Their performances were similar in elevated CO2 because native populations were more responsive and their performances were similar with elevated ozone because invasive populations were more susceptible. Compared to other species, T. sebifera had high growth rates but low levels of tannin production that were insensitive to variation in CO2 or ozone. Both China and US Platanus plants reduced tannins with increased CO2 and/or ozone and US Liquidambar plants increased tannins with the combination of elevated CO2 and ozone. The growth results suggest that intraspecific variation in T. sebifera will reduce the effects of CO2 or ozone alone on invasions but increase their combined effects. The tannin results suggest that defense responses to CO2 and ozone will be variable across native and exotic species. The effects of CO2 and ozone on growth and defense of native and exotic species indicate that the benefit or harm to species from these global change drivers is an idiosyncratic combination of species origin and genus.  相似文献

Abstract The effects of pasture management options (sowing introduced legumes and grasses, timber treatment, applying fertilizer, cultivation before sowing, and stocking rate) on species diversity were measured at two experimental sites (Hillgrove and Cardigan) near Charters Towers, northeast Queensland. Species were divided into three groups (sown, native and exotic) and diversity was measured as species density (number of species recorded in each plot and number of species/quadrat) annually from 1982 to 1992. The responses of individual native and naturalized species to treatment were also determined. All management options affected diversity but the responses varied with site and season, and with the different measurement scales. The density of sown species either increased or was unaffected by all the management options; there were no significant decreases. The density of native species showed both positive and negative responses; it increased at high stocking rates and with tree killing at Hillgrove, and decreased with pasture sowing and cultivation. The density of exotic species increased as stocking rate was increased and decreased when pastures were sown (although not at the quadrat scale at Hillgrove). Overall the most diverse vegetation was on plots grazed at high stocking rates; at the plot scale these were native pastures but at the quadrat scale the sown pastures had more species. Among the native and naturalized species, only Portulaca spp. were more frequent on the oversown plots than the native pasture plots; 48% (Hillgrove) and 68% (Cardigan) of the species were less frequent on the oversown plots. Fertilizer application had little effect on species frequencies, while timber treatment resulted in both increases and decreases in frequency of a small number of species. The species were divided into four groups on the basis of their responses to stocking rate: a grazing-sensitive group (e.g. Themeda triandra), two grazing-tolerant groups which either slightly decreased (e.g. Chrysopogon fallax) or slightly increased (e.g. Sida spinosa) in frequency as stocking rate increased, and a fourth group of species which were frequent only at high stocking rates (e.g. Bothriochloa pertusa). There were no close relationships between herbage yield and species density.  相似文献

Five recombinant Toxoplasma gondii antigens, designated B427, C51, C55, V22, and MBP30 were assessed for their potential use in an enzyme-linked immunoassay (EIA) for detection of T. gondii infection in swine. The antigens were evaluated with sera from young pigs that had been fed 1-10,000 T. gondii oocysts of the VEG or GT-1 strains. Results were compared with an EIA using a native T. gondii antigen extract. All 5 recombinant antigens, as well as native antigen, detected antibody responses as soon as 3 wk after infection in pigs inoculated with 1 or 10 oocysts of the VEG strain. This antibody response persisted, at varying levels, for 14 wk when the experiment was terminated. All antigens also detected antibody responses in pigs 4 wk after inoculation with 10,000 oocysts of the GT-1 strain. The antibody response recognized by native antigen remained high through 51 wk after inoculation. However, there was considerable animal-to-animal variation in responses to the individual recombinant antigens. Only antigens C51 and MBP30 consistently detected a positive antibody response over the entire 51-wk course of the experiment. These results suggest that these antigens might be useful for the serological detection of T. gondii infection in pigs.  相似文献

The anti-predator behaviours of a New Zealand freshwater crayfish (Paranephrops zealandicus) to the native long-finned eel (Anguilla dieffenbachii) and the introduced brown trout (Salmo trutta) were investigated. Crayfish modified their behaviour in the presence of both trout and eels. However, a significantly greater number of defensive chela displays and swimming responses were made to eels than trout. Crayfish were able to use chemical cues from skin mucus to detect eels but not trout. Paranephrops zealandicus is able to make some appropriate defensive behavioural responses to the introduced brown trout as well as to its native predator, the long-finned eel. However, crayfish may be at greater risk from the introduced predator because of their apparent inability to detect trout using non-contact chemical cues. This may be a reflection of the different co-evolutionary histories crayfish have had with trout and eels.  相似文献

In NW Patagonia, South America, natural shrublands and mixed forests of short Nothofagus antarctica (G. Forst.) Oerst. trees are currently being replaced by plantations with Pseudotsuga menziesii (Mirb) Franco. This land use change is controversial because the region is prone to drought, and replacement of native vegetation by planted forests may increase vegetation water use. The goal of this study was to examine the physiological differences, especially the response of water flux and canopy conductance to microclimate, that lead to greater water use by exotic trees compared to native trees. Meteorological variables and sapflow density of P. menziesii and four native woody species were measured in the growing season 2005–2006. Canopy conductance (gc) was estimated for both the exotic (monoculture) and native (multi-species) systems, including the individual contributions of each species of the native forest. Sapflow density, stand-level transpiration and gc were related to leaf-to-air vapor pressure difference (VPD). All native species had different magnitudes and diurnal patterns of sapflow density compared to P. menziesii, which could be explained by the different gc responses to VPD. Stomatal sensitivity to VPD suggested that all native species have a stronger stomatal control of leaf water potential and transpiration due to hydraulic limitations compared to P. menziesii. In conclusion, differences in water use between a P. menziesii plantation and a contiguous native mixed forest of similar basal area could be explained by different gc responses to VPD between species (higher sensitivity in the native species), in addition to particular characteristics of the native forest structure.  相似文献

Differences among exotic species can be as large as differences between native and exotic species. Typically, however, only the distinction between native and exotic is made when predicting responses in a community. In this paper, I examine the response of plant species to experimental disturbance and exclusion of invasive European rabbits (Oryctolagus cuniculus) in a grassland community with exotic plants originating from five continents. I explore group responses based on native status, shared native range with rabbits, having a congener from the native range of rabbits, life-history (e.g., annual), and life-form (e.g., grass). Individual species responses to rabbits were idiosyncratic, but group responses were predicted by continent of origin, not native status. Native status did predict response to disturbance with almost uniform responses within groups. Exotic species, regardless of origin, were positively affected by disturbance. Native species, in contrast, were negatively affected by disturbance. These results suggest that grouping plant species by native status is valid for questions of disturbance, but when analyzing outcomes of interactions, factors other than native status, such as shared evolutionary history, should be considered.  相似文献

Spiders (Araneae) play key roles in ecosystems, not only as common and abundant generalist predators, but also as major contributors to biodiversity in many areas. In addition, due to their short generation times and high mobility, spiders respond rapidly to small changes in their environment, potentially making them useful indicators for restoration monitoring. However, few studies have focused on spider responses to grassland restoration in the United States. We compared degraded, native, and restored grassland sites to examine how spider communities and habitat respond to arid grassland restoration. We also examined how responses varied with the age of the restoration project. Spider communities in native sites differed from those in restored and degraded sites in several ways: native sites had fewer spiders and a different community composition than degraded and restored sites. However, native and restored sites had more species than degraded sites. Chronosequence data showed trends for lower abundance, higher species richness, and changing community composition as restoration projects mature. Several habitat variables were closely linked to variation in spider communities including cover of invasive annual grasses, litter, and biological soil crusts. Our data suggest that spider and vegetation responses to grassland restoration efforts can be successful in the long term—with resulting communities becoming more similar to native ones—and that spiders are useful indictors of grassland restoration. Our results also suggest that restoration may involve balancing trade‐offs between ecosystem services, with potential losses in predatory control offset by increases in biodiversity with restoration effort.  相似文献

The invasion of European perennial grasses represents a new threat to the native coastal prairie of northern California. Many coastal prairie sites also experience anthropogenic nitrogen (N) deposition or increased N availability as a result of invasion by N-fixing shrubs. We tested the hypothesis that greater seedling competitive ability and greater responsiveness to high N availability of exotic perennial grasses facilitates their invasion in coastal prairie. We evaluated pairwise competitive responses and effects, and the occurrence of asymmetrical competition, among three common native perennial grasses (Agrostis oregonensis, Festuca rubra, and Nassella pulchra) and three exotic perennial grasses (Holcus lanatus, Phalaris aquatica, and Festuca arundinacea), at two levels of soil N. We also compared the root and shoot biomass and response to fertilization of singly-grown plants, so we could evaluate how performance in competition related to innate plant traits. Competitive effects and responses were negatively correlated and in general varied continuously across native and exotic species. Two exceptions were the exotic species Holcus, which had large effects on neighbors and small responses to them, and competed asymmetrically with all other species in the experiment, and the native grass Nassella, which had strong responses to but little effect on neighbors, and was out-competed by all but one other species in the experiment. High allocation to roots and high early relative growth rate appear to explain Holcus’s competitive dominance, but its shoot biomass when grown alone was not significantly greater than those of the species it out-competed. Competitive dynamics were unaffected by fertilization. Therefore, we conclude that seedling competitive ability alone does not explain the increasing dominance of exotic perennial grasses in California coastal prairie. Furthermore, since native and exotic species responded individualistically, grouping species as ‘natives’ and ‘exotics’ obscured underlying variation within the two categories. Finally, elevated soil N does not appear to influence competition among the native and exotic perennial grasses studied, so reducing soil N pools may not be a critical step for the restoration of California coastal prairie.  相似文献

Prescribed fire is an important management tool for reducing the dominance of non‐native species in annual grasslands; both annual and perennial native species show strong vegetative responses in the subsequent growing season. However, although the post‐fire contribution of native species to the seed bank is assumed to be larger than in pretreatment years, the effects on seed quality, particularly viability and longevity, are not well understood. In this study, I germinated Nassella pulchra (purple needlegrass) seed that had been stored for 10 years after collection from target plants receiving treatment combinations of summer burning and grazing by sheep. Seeds from burned plants were larger and had higher germinability than seed from unburned plants. Seeds from plants that were both burned and grazed had the highest germination. The strong relationship between long‐term viability and seed size suggests greater maternal provisioning and increased seed quality subsequent to burning and grazing. I conclude that managing for seed quality may be a useful approach for conservation of native species in California's critically endangered grassland habitats.  相似文献
