To discriminate between the roles of differential pollen tube growth and inbreeding depression in causing reduced seed production following self-pollination of distylous Amsinckia grandiflora (Boraginaceae), callose plug and pollen tube production were compared following cross- and self-pollinations of A. grandiflora. Pin (long-styled morph) × thrum (short-styled morph) crosses yielded more pollen tubes than pin × pin crosses or pin self-pollinations; no difference could be detected for the pin × pin or pin self-pollination categories. Using thrums as the female parent, no significant differences were detected in any pollination category. These results indicate that reduced seed production following self-fertilization of A. grandiflora is likely to be the result of inbreeding depression rather than inhibition of self pollen tubes.  相似文献

Summary Intraspecific and reciprocal interspecific crosses involving Zinnia angustifolia clones and Z. elegans lines showed that in both species, sporophytic self-incompatibility (SI) systems were present. Intensity of SI varied among clones and lines, and high self seed set was associated with a concomitant decrease in callose fluorescence in papillae and pollen tubes. Incomplete stigmatic inhibition of pollen germination and tube growth was observed in reciprocal interspecific crosses and associated with callose synthesis, suggesting S-gene activity. Seed set and progeny obtained following Z. angustifolia×Z. elegans matings was comparable to intraspecific compatible matings of Z. angustifolia although the rate of pollen tube growth through the style was slower. In Z. elegans × Z. angustifolia matings, additional prezygotic barriers were present and acted between pollen tube penetration of the stigma and syngamy. SI X SI interspecific incompatibility was essentially unilateral, with no embryos or progeny obtained when Z. elegans was the pistillate parent. Long-, mid-, and short-styled clonal accessions of oca (Oxalis tuberosa) were intercrossed in a complete diallelic design. Pollen tube growth in styles was monitored in all diallelic crosses. Pollen fertility was estimated by two tests: staining of pollen grains with aceto-carmine and detection of β-galactosidase activity by the substrate X-Gal. The two methods of pollen fertility estimation were equally useful to detect fertility levels. Pollen originating from short stamens had the highest fertility (85%) and pollen from long stamens had the lowest fertility (70%). Pollen fertility was high throughout, but its degree varied with the stylar morph on which the pollen was formed. Long-styled accessions had the highest rates of fertile pollen. Differences in pollen fertility at different anther levels in the same style morph were also apparent. Pollen grain diameter of the six morph-anther level combinations was inversely correlated with pollen fertility. Pollen grains from long stamens were the largest and pollen grains from short stamens were the smallest. Neither pollen fertility nor pollen grain size had an influence on pollen tube growth in the style or on fruit and seed set. Pollen tubes growing within the styles were inhibited at a different level for each of the 18 cross combinations in the diallel. Although legitimate crosses had greatest pollen tube growth, some of the illegitimate inter- and intramorph crosses had equally high scores. Of all illegitimate crosses, mid-styled seed parents had the lowest level of stylar incompatibility. Fruit and seed set were highly correlated with the extent of pollen tube growth in the style. The number of pollen tubes entering ovules in a flower was in good agreement with the number of seeds produced per fruit. Therefore, it is concluded that stylar incompatibility is the major determinant of limited seed formation in oca even in the most successful legitimate cross combinations. Hand pollinations experiments with distylous Amsinckia douglasiana A. DC. (Boraginaceae) revealed that legitimate (intermorph) crosses produce more seed than self-pollinations and that self-pollinations yield more seed than illegitimate (intramorph) crosses. We tested the occurrence of cryptic incompatibility in both style-length morphs by pollinating them with a mixture of legitimate and illegitimate pollen, each homozygous for a different electrophoretically distinguishable allele of phosphoglucose isomerase. The success of intramorph pollen was greatly reduced in mixture; only about 4% of the offspring of both pins and thrums were sired by illegitimate pollen. One pin and one thrum had sufficiently high seed set that the results cannot be attributed to selective abortion of embryos. In those individuals, discrimination against illegitimate pollen must have occurred on the stigma or in the style. Based on our findings, it appears that illegitimate matings are rare in natural populations. The progeny of legitimate crosses was biased toward pins, which suggests some regulation of morph ratios independent of the style-length locus. Prezygotic discrimination against the thrum genotype or selective abortion of embryos with the thrum genotype are both possible, but selective mortality of seedlings with the thrum genotype cannot be ruled out for some families.  相似文献

In Brassica oleracea, sporophytic self-incompatibility prevents germination of self pollen, or normal growth of self pollen tubes. After self-pollination, the papillae of stigmas synthesize callose. The role of Ca++ in the formation of stigmatic callose was tested by adding compounds that interact with Ca++ to suspensions of pollen that were known to induce callose formation in self stigmas. The calcium channel antagonist, lanthanum, and the calcium chelating agent, EGTA, reduced or abolished the callose response to self-pollen suspensions. In the presence of Ca++, the calcium ionophore, A23187, induced callose in stigmatic papillae when added to pollen suspensions, or alone. Therefore, callose deposition in response to incompatible pollinations appears to be a calcium-dependent process. Pretreatment of pistils with 100 μm 2-deoxy-D-glucose abolished the callose response to self-pollination, while self pollen remained inhibited and cross pollen grew normally in treated pistils. Thus, callose formation in the stigma is not an essential part of the self-incompatibility mechanism preventing the growth of self pollen in Brassica.  相似文献

Outcross, self- and mixed pollinations were performed inCrocus vernus subsp.vernus, a species with bicellular pollen, dry stigmas and hollow style. No differences were noted among the above pollinations concerning the germination of pollen and the growth of pollen tubes until the top of ovary. Within 45 min after pollinations 62% of pollen grains germinated. Pollen tubes penetrated the papilla cuticle extending along the papilla wall; on entry into stigmatic lobes they continued growth in the stylar secretion to ovarian locules. Here, however, self-pollen tubes failed to reach or to enter the ovule micropyle; while pollen tubes from either outcross- or mixed pollinations grew until fertilizing ovules. These observations gave evidence of a self-incompatibility system inCrocus, which appeared to be neutralized by mentor effect. The ovary as site of incompatibility response is discussed.  相似文献

Distyly, floral polymorphism frequently associated with reciprocal herkogamy, self‐ and intramorph incompatibility and secondary dimorphism, constitutes an important sexual system in the Rubiaceae. Here we report an unusual kind of distyly associated with self‐ and/or intramorph compatibility in a perennial herb, Hedyotis acutangula. Floral morphology, ancillary dimorphisms and compatibility of the two morphs were studied. H. acutangula did not exhibit precise reciprocal herkogamy, but this did not affect the equality of floral morphs in the population, as usually found in distylous plants. Both pin and thrum pollen retained relatively high viability for 8 h. The pollen to ovule ratio was 72.5 in pin flowers and 54.4 in thrum flowers. Pistils of pin flowers remained receptive for longer than those of thrum flowers. No apparent difference in the germination rate of pin and thrum pollen grains was observed when cultured in vitro, although growth of thrum pollen tubes was much faster than that of pin pollen tubes. Artificial pollination revealed that pollen tube growth in legitimate intermorph crosses was faster than in either intramorph crosses or self‐pollination, suggesting the occurrence of cryptic self‐incompatibility in this species. Cryptic self‐incompatibility functioned differently in the two morphs, with pollen tube growth rates after legitimate and illegitimate pollination much more highly differentiated in pin flowers than in thrum flowers. No fruit was produced in emasculated netted flowers, suggesting the absence of apomixis. Our results indicate that H. acutangula is distylous, with a cryptic self‐incompatibility breeding system.  相似文献

Summary Visible light and UV epifluorescence microscopy were used to assess self-incompatibility (SI) in Zinnia angustifolia clones. Pistils were fixed 24 h after pollination and stained either with aniline blue in lactophenol (visible light microscopy) or decolorized aniline blue (fluorescence microscopy). Percentage of florets with embryos 21 days following pollination (% embryo set) was used as a control. Embryo set following self- or incompatible cross-pollinations ranged from 0% to 9.9%, whereas compatible crosses yielded 55.5%–87.1% embryo set. Observations using visible light microscopy indicated pollen load and number of germinated grains were significantly higher for compatible compared to incompatible crosses, and both variables were positively correlated (r = 0.89–0.96) to % embryo set. Examinations with UV epifluorescence microscopy revealed pollen load was higher and little or no callose accumulated in stigmatic papillae following compatible crosses, whereas for incompatible crosses, pollen load was low and callose lenticules were deposited in stigmatic papillae; the correlation between pollen load and % embryo set was 0.89. The intensity of callose fluorescence of the pollen tube-papillae attachment sites was quantitatively measured via micro spectrofluorometry. Callose fluorescence intensity ranged from 47.9% to 62.6% for incompatible and from 6.4% to 9.9% for compatible crosses, and was negatively correlated (r= — 0.95) with % embryo set. The growth of pollen tubes of a sampling of Lilium species in styles of L. longiflorum incubated at 24 C for 48 hr after pollination indicated two types of interspecific incompatibility. Pollen tubes of two self-compatible species of section Leucolirion, the section including L. longiflorum, stopped growth abruptly upon reaching the stylar canal, were of abnormal morphology, and were incapable of continued growth with longer incubation. Pollen tubes of self-compatible or self-incompatible species of sections Sinomartagon, Pseudolirium, Liriotypus, and Daurolirion approached but did not exceed the length of intraspecific incompatible pollen tubes in styles of L. longiflorum. Pollen tube morphology was normal and tubes were capable of continued growth with additional incubation. Unilateral interspecific incompatibility occurred in reciprocal crosses between self-incompatible L. longiflorum and self-compatible L. regale and L. formosanum, but exceptions occurred in Aurelian hybrids. Incubation of interspecifically pollinated L. longiflorum styles at 39 C, which removes the self-incompatibility reaction, had no effect on interspecific incompatibility.  相似文献

For species with bicellular pollen, the attrition of pollen tubes is often greatest where the style narrows at the transition between stigmatic tissue and the transmitting tissue of the style. In this region, the tubes switch from predominantly autotrophic to predominantly heterotrophic growth, the generative cell divides, the first callose plugs are produced, and, in species with RNase-type self-incompatibility (SI), incompatible tubes are arrested. We review the literature and present new findings concerning the genetic, environmental and stylar influences on the performance of pollen before and during the autotrophic-heterotrophic transition of pollen tube growth. We found that the ability of the paternal sporophyte to provision its pollen during development significantly influences pollen performance during the autotrophic growth phase. Consequently, under conditions of pollen competition, pollen selection during the autotrophic phase is acting on the phenotype of the paternal sporophyte. In a field experiment, using Cucurbita pepo, we found broad-sense heritable variation for herbivore-pathogen resistance, and that the most resistant families produced larger and better performing pollen when the paternal sporophytes were not protected by insecticides, indicating that selection during the autotrophic phase can act on traits that are not expressed by the microgametophyte. In a study of a weedy SI species, Solanum carolinense, we found that the ability of the styles to arrest self-pollen tubes at the autotrophic-heterotrophic transition changes with floral age and the presence of developing fruits. These findings have important implications for selection at the level of the microgametophyte and the evolution of mating systems of plants.  相似文献

Various combinations of intermorph, selfing and intramorph pollinations were carried out in Averrhoa carambola and the pollinated pistils were observed under fluorescence and scanning electron microscopes in time-course experiments. In both compatible and incompatible pollinations, similar behavior of pollen germination and penetration was observed in the first 4 h after pollination. In compatible intermorph pollination, pollen tubes were found at the base of the transmitting tract of the style at 8 h and 24 h after pollination in both the pin and thrum morphs. With thrum flowers, selfing resulted in pollen tubes being uniformly arrested at the junction between the stigmatic and stylar tissues. Penetration of pollen tubes into the upper portion of style was observed in thrum intramorph pollination and, when the second member was treated as the gynoecial tissue under the same pollination, penetration of tubes was further enhanced. Pin flowers, on selfing, resulted in pollen tube penetration farther down the style than was the case with thrum selfing. Intramorph pollination of pin morph behaved in a similar manner to selfing and was not affected by genotypes. Beside the stamen-style dimorphism, the receptive surface of the cob stigma was larger in pin than thrum flowers. While pin pollen was round, thrum pollen was oblong in shape with pin to thrum ratio on the polar axis being 1.2 and on the equatorial axis 0.8. The stigma of pin morph belonged to the dry type, while that of the thrum resembled the wet type.  相似文献

Efforts to introduce Dutch Elm Disease resistance into the American elm (Ulmus americana L.) through breeding with Asian elms has been hampered by sexual incompatibility. Controlled pollinations of Ulmus americana and the Siberian elm (Ulmus pumila L.) were studied in detail to gain insight into the nature of this incompatibility. Microscopic observations revealed that germination and early pollen tube growth were inhibited on the stigmatic surface following both intra- and interspecific incompatible pollinations. Both qualitative and quantitative differences in pollen inhibition on the stigmatic surface indicated that the inhibition may involve the action of an inhibitory substance. Detailed observations on callose deposition indicated that this β-1,3 glucose polymer may implement the inhibition.  相似文献

异型花柱是一种受遗传因素控制的花型多态性现象,包括二型花柱和三型花柱两种类型.本文以茜草科艳丽耳草(Hedyotis pulcherrima)为实验材料,通过对其野外居群的花型、花部形态及花粉特征等观察,发现艳丽耳草野外居群同时存在长花柱型花和短花柱型花,长/短花柱型花的数量比例为1∶1.两型花具有精确的交互式雌雄异位特征,并且该特征与花冠长度相关性显著.长/短花柱型花的柱头裂片长度、花粉大小及淀粉含量等具有二型性.花粉体外培养时花粉萌发率及花粉管生长速率无显著性差异.人工授粉后,艳丽耳草长/短花柱型花型间异交花粉管生长形态正常,授粉24 h后花粉管均已进入子房.而长/短花柱型花在自交及型内异交下均表现为不亲和,花粉管生长停止于柱头,花粉管顶端累积胼胝质并膨大.艳丽耳草没有无融合生殖现象,型间人工辅助异交授粉结实率为100%,显著高于自然结实率.本研究结果表明,艳丽耳草是典型的二型花柱植物,并具有异型自交不亲和系统.  相似文献

BACKGROUND AND AIMS: Distyly has been hypothesized to promote cross-pollination by reducing intrafloral and geitonogamous self-pollination, and enhancing intermorph pollination. Distylous plants typically display both reciprocal herkogamy and a heteromorphic incompatibility system, which allows mating only between morphs. Distyly and its pollination consequences were examined in two Pentanisia species with long-tubed flowers which are pollinated almost exclusively by butterflies. METHODS: Anther and stigma heights were measured to quantify reciprocal herkogamy. The type of incompatibility system was determined by observing pollen tubes and seed production following controlled hand pollination. Pollen loads on pollinators and stigmas were also examined to assess the efficiency of intermorph pollen flow. KEY RESULTS: Pentanisia prunelloides and P. angustifolia exhibit reciprocal herkogamy and a host of ancillary dimorphisms, including pollen colour, exine sculpturing, stigmatic papilla shape and floral-tube pubescence. Controlled hand-pollinations revealed the presence of a strong heteromorphic incompatibility system in both species. The site of incompatibility differed between the morphs; intramorph pollen tubes were blocked in the style of the short-styled morph and on the stigmatic surface of the long-styled morph. Butterflies carried pollen from the short- and long-styled morphs primarily on their head and proboscis, respectively. Natural pollination resulted in a higher proportion of pollen transfer from long- to short-styled plants than vice versa. Nevertheless, fruit set did not differ between morphs. CONCLUSIONS: Both Pentanisia species are fully distylous. Reciprocal herkogamy results in pollen from the two morphs being carried on different locations on pollinators' bodies, which in turn promotes intermorph pollination. Intramorph pollination does not result in fertilization, because of an effective heteromorphic incompatibility system.  相似文献

Most species of Dalechampia have expanded stigmatic surfaces that extend from the stylar tip part way down the sides of the elongated styles. Pollen grains landing at the stylar tip grow tubes directly down the style to the ovary. Pollen grains landing on the lateral stigmatic surfaces, however, exhibit indirect pollen-tube growth: the tubes grow first to the stylar tip, bend 180°, and then grow to the ovary. Indirect pollen-tube growth appears to be reproductively important: more than half the arriving pollen lands on the lateral stigmatic surfaces, and virtually all of these grains produce tubes that grow indirectly to the ovary; pollen on the lateral surface fertilizes seeds about as effectively as pollen at the tip (with direct tube growth). Indirect pollen-tube growth may be the result of a correlated response to selection for expanded stigmatic surfaces in Dalechampia, or it may be an adaptation to increase the intensity of pollen competition.  相似文献

A program of experimental field pollinations was carried out on 14 species of woody Rubiaceae in a Costa Rican wet forest in order to test for the presence of self-incompatibility systems. Species of Cephaelis, Coussarea, Faramea, Hamelia, Posoqueria, Psychotria, Rudgea, and Warszewiczia were investigated in the study. Ten of the species are distylous, and of these, nine were found to be self-incompatible. The site of the incompatibility barrier varied between and within species. Short style plants always had stigmatic inhibition of pollen tubes following self and intraform pollinations, but the site of the rejection response in long style plants was quite variable. In several species, pollen tubes, resulting from incompatible pollinations of the long style flowers, penetrated to the base of the style. Fruit set was followed in two of these species, and despite deep penetration of self and intraform pollen tubes in the long style morph, only interform pollinations resulted in fruit set. Four of the species tested are florally monomorphic and each was found to be self-incompatible on the basis of fruit set patterns. Pollen tubes in the styles of self-pollinated flowers of two of the monomorphic species, penetrated to the ovary, but no fruits resulted from selfpollinations.  相似文献

Heteromorphic characters and structural features of the pollen tube pathway are described in tristylous Pontederia sagittata to assess their influence on the pollination process and in mediating self-incompatibility behavior. Heteromorphic characters that distinguish the floral morphs include style length, stigma depth, stigmatic papillae length, stylar parenchyma cell length, area of the stylar canal, stamen height, anther size, and pollen size. Unlike several distylous species that have been investigated, the exine of pollen in P. sagittata was not strongly differentiated among the pollen types, and no differences in stigma cytochemistry were apparent. Features common to the floral morphs were a wet stigma, a hollow trilobed stylar canal separating into two sterile and one fertile canal, and a single anatropous ovule with a highly elaborated integumentary obturator. The similarity in structural features of the pollen tube pathway of P. sagittata to those found in monocotyledonous taxa with gametophytic self-incompatibility suggests that phylogenetic constraints may have influenced the evolution of trimorphic incompatibility in the Pontederiaceae.  相似文献

The rates of germ tube elongation in vitro of pollen from cherry leaf roll virus (CLRV)-infected birch (Betula pendula) did not differ significantly from those of pollen from virus-free trees. Differences in percentage germination of pollen collected from trees at different sites were significant but percentages of germination of pollen from virus-infected and virus-free trees did not differ greatly from one another. In vivo, pollen from infected trees germinated on healthy and CLRV-infected stigmas and callose plugs formed in both types of tissues. However, the extent of callose plug formation was greater in the pollen tubes of virus-free grains germinating in infected stigmas than in reciprocal crosses. CLRV coat antigen was detected by ELISA in stigmatic tissue, from healthy trees, on which virus-carrying pollen grains had germinated. Using fluorescein isothiocyanate conjugated to CLRV-specific γ-globulin, viral coat antigen was detected throughout germ tubes from virus-carrying but not virus-free pollen germinating in vitro. Protoplasts released following Driselase digestion of pollen germ tubes from virus-infected trees contained CLRV antigen detectable by ELISA. During germination of virus-infected pollen there was little synthesis of viral coat protein or nucleic acid but, following inoculation with purified virus particles, protoplasts made from healthy germinating pollen produced increasing amounts of CLRV-specific antigen implying that CLRV replicated in this system.  相似文献

 Style squashes and stylar grafts were used to examine the growth of Nicotiana alata pollen tubes in self-compatible and self-incompatible styles. Compatible tubes typically showed a uniform layer of callose deposition in the walls and in small plugs spaced at regular intervals within the tube. Incompatible tubes were characterised by the variability of callose deposition in the walls and by larger, closer and more irregularly spaced plugs. There was no difference in the growth rate of compatible and incompatible tubes during growth through the stigma, but within the style most compatible tubes grew 20–25 mm day-1 (maximum 30 mm day–1), whereas incompatible tubes grew 1.0–1.5 mm day-1 (maximum 5 mm day–1). Many incompatible tubes continued to grow until flowers senesced, and only a small proportion died as a consequence of tip bursting. Grafting compatibly pollinated styles onto incompatible styles showed that the incompatible reaction could occur in pollen tubes between 2 and 50 mm long, and that inhibition of pollen tube growth occurred in both the upper and lower parts of the transmitting tract. Grafting incompatibly pollinated styles onto compatible styles showed that the incompatible reaction was fully reversible in at least a proportion of the pollen tubes. The findings are not consistent with the cytotoxic model of inhibition of self-pollen tubes in solanaceous plants, which assumes that the incompatible response results from the degradation of a finite amount of rRNA present in the pollen tube.
