Carapa guianensis is a timber species found in Central America and the north of South America. We have developed microsatellite primers which will allow analysis of gene flow and population genetic structure in natural populations of this tree species. Polymorphism of five microsatellite loci was evaluated using a total of 12 adult trees from a natural population. An average of 4.2 alleles per locus was detected, and expected heterozygosity ranging from 0.397 to 0.806. These loci are being used for genetic population analysis in a managed forest in the state of Pará, in Amazonian Brazil, as part of the Dendrogene project.  相似文献

In order to evaluate the consequences of forest fragmentation on populations of Magnolia obovata, we compared genetic diversity and reproductive characteristics at two nearby sites, one conserved and one fragmented. The genetic diversity between adults trees of the different sites was not significantly different. However, saplings in the conserved site showed a significantly higher genetic diversity than both adult trees in the conserved site and saplings in the fragmented sites; this was found to be the result of the larger gene flow into the conserved site. The density of the adult trees was significantly related to all of the reproductive traits analyzed (fertilization of ovules, insect attack to seeds, ovules that developed into seeds and outcrossing at the stage of seeds) at both sites. At both sites, fertilization of ovules and insect attack on seeds were positively correlated to adult tree density while outcrossing rate was negatively correlated to adult tree density. The fertilization of ovules and outcrossing were more dependent on adult tree density in the fragmented site than in the conserved site. The probability of ovules developing into outcrossed seeds showed a negative correlation with adult tree density at both sites, indicating the advantage of low density for this species and possibly implying a resilience to habitat fragmentation. A two-generation-analysis did not identify significant differences between sites in terms of the structure of the pollen pool and the number of pollen donors. Although fragmentation affected reproductive characteristics, the effect on seedling establishment and subsequent survival remains to be determined. Proposals for future studies that will assist in the development of management strategies for forests suffering fragmentation are made.  相似文献

Deforestation and selective logging in the tropics may have serious consequences on genetic processes in tropical tree populations, affecting long-term survival of a given species as well as tropical forest communities. Because understanding the effects of human-induced changes on genetic processes is of utmost importance in formulating sound conservation and management plans for tropical forest communities, we developed microsatellite or simple sequence repeat (SSR) markers for the tropical tree Carapa guianensis (Meliaceae) and assessed the polymorphism of SSRs in adult and sapling populations in a large contiguous forest and in selectively logged and fragmented forests. The number of alleles in polymorphic loci ranged between 4 and 28. No inbreeding was detected in saplings or adult cohorts, but the allelic richness was lower in the sapling cohort of the isolated fragment. Genetic distances, Nei's D and (delta&mgr;)2, and RST values among saplings were greater than among adult cohorts, suggesting restriction of gene flow due to deforestation and habitat fragmentation. These SSR loci may be used to address many related questions regarding the population and conservation genetics of tropical trees.  相似文献

Eremanthus erythropappus, commonly known as "candeia", is an abundant pioneer tree species, forming dense populations known as "candeial", but it is also found in forests at middle stages of succession. Trees from forests are bigger and occur in lower density than in the "candeial". The objectives of the present study were to investigate if the decrease in population density during successional process is accompanied by 1) changes in within-population genetic diversity, and 2) differentiation of populations. Eight populations, four of early successional stage ("candeial") and four of middle successional stages (forest), were analyzed with RAPD markers. The genetic diversity found was high compared to other tree species analyzed with RAPD markers. AMOVA revealed that most of the genetic variations of E. erythropappus were found within populations (85.7%), suggesting that this species is predominantly outcrossing. The relatively low differentiation among the populations can be attributed to small distances among the populations analyzed (0.2 to 10.8 km). No indication that populations from middle successional habitats show lower genetic variation than populations from early successional stages was found. The percentage of polymorphic fragments (82.8 and 84.8%) and the Shannon indexes (0.442 and 0.455) were similar in "candeial" and forest, respectively. These results suggest that if an increase in selection intensity occurred during succession, it did not result in a decrease in genetic diversity or that the selection effect was balanced by other factors, such as gene flow. Higher significant differentiation among E. erythropappus populations from "candeial" in relation to that among populations from forest was also not detected.  相似文献

Summary This study demonstrates the impact of natural factors and human activities on biodiversity at gene level on a keystone Mediterranean forest ecosystem species. We monitored the within and among population gene diversity of Cedrus libani, a forest tree species of the Eastern Mediterranean mountains. We used paternally inherited chloroplast microsatellites (57 haplotypes) and bi-parentally inherited isozymes (12 loci) to estimate allelic richness, heterozygosity, and differentiation in 18 natural and 1 planted populations from Turkey and Lebanon. We showed that there is a phylogeographic structure in C. libani, and that forests from Lebanon and Turkey constitute two genetically isolated groups which probably arose from distinct refugia after the last Quaternary glacial cycle. We found extensive gene flow and relatively low differentiation in Turkey, as well as little evidence of genetic drift within populations. However, one population we analyzed, which was planted more than 20 centuries ago, and is isolated from core populations in Turkey, demonstrated extremely low genetic diversity and deserves high conservation priority. In contrast, we found low gene flow, high differentiation and severe cases of genetic drift in Lebanon. As forests there are the remnants of millennia-long extensive deforestation, all deserve high conservation priority.  相似文献

Comprehending the size distribution within a population and grasping its connection to tree maturation (onset of reproductive phase) is pivotal for formulating sustainable management strategies and conserving numerous tree species. The objective of this study was to compare the structure and probability of fruit production of Carapa guianensis in two types of forest (varzea, or floodplain, vs. terra firme, or upland) in the northeastern Amazon, Brazil. The study was conducted in four permanent plots of 300 × 300 m (two for each type of forest). All individuals ≥10 cm in diameter at breast height (DBH) were mapped and measured for DBH, height, and canopy characteristics (shape and position), and liana infestation. Sixty-seven trees were inventoried in the upland forest and 297 in the floodplain forest. The trees were larger in the upland (33.36 ± 1.84 cm) than in the floodplain (21.92 ± 1.5 cm) and more than half (56%) of the upland trees and more than two thirds (67%) of floodplain trees were reproductive. The minimum diameter for fruit production in the floodplain (10.2 cm of DBH) was more than two times smaller than in the upland (25.5 cm of DBH). The probability of fruit production of a tree of the same size was twice as high in the floodplain compared with the upland, and trees with rounded canopies had a higher probability of fruit production in the upland. Our results showed that the probability of fruit production can be modeled with variables of tree size and canopy shape, and that intrinsic factors of the forests influence the reproductive maturity of C. guianensis which presents greater precocity and productive potential of seeds in the estuarine floodplain forest of the Amazon River.  相似文献

Carapa guianensis Aubl. (Meliaceae), a hard wood tree from the Brazilian Amazon, has large recalcitrant seeds that can germinate and establish in both flood-free (terra-firme) and flood-prone (várzea) forests. These seeds, although large, can float. This study was designed to experimentally examine seed longevity under floating conditions ex-situ and its effects on subsequent germination and seedling growth. Many seeds germinated while floating, and radicle protrusion occurred from 3 to 42 d after the start of the floating treatment (tap water, room temperature 20–30 °C). Shoots of newly germinated floating seedlings may elongate up to 37.0 cm in 20 d without loss of viability. Epicotyl and first leaf emergence were delayed by floating. Seeds that did not germinate while floating were then placed on vermiculite and watered daily, where many seeds resumed germination. Germination during and after floating was affected by the length of the floating treatment: 88% germinated after 1 mo, 82% germinated after 2 mo and 70% germinated after 2.5 mo. These results indicate that Carapa guianensis has physiological variation regarding dormancy in response to seed floatation. The fact that floatation induces dormancy in recalcitrant seeds of this economically important species can be relevant to initiatives of ex situ storage of seeds.  相似文献

Recent studies suggest that tropical tree species exhibit low inbreeding and high gene dispersal levels despite the typically low density of conspecifics in tropical forests. To examine this, we undertook a study of pollen gene dispersal and mating system of two Amazonian tree species. We analyzed 341 seeds from 33 trees at four microsatellite loci in a Carapa guianensis population from Brazil, and 212 seeds from 22 trees at four microsatellite loci in a Sextonia rubra population from French Guiana. Differentiation of allele frequencies among the pollen pool of individual trees was ΦFT= 0.053 (95% CI: 0.027–0.074) for C. guianensis and ΦFT= 0.064 (95% CI: 0.017–0.088) for S. rubra. The mean pollen dispersal distances were estimated at 69–355 m for C. guianensis , and 86–303 m for S. rubra , depending on the pollen dispersal model and the estimate of reproductive tree density used. The multi-locus outcrossing rate was estimated at 0.918 and 0.945, and the correlation of paternity at 0.089 and 0.096, for C. guianensis and S. rubra , respectively, while no significant levels of biparental inbreeding were detected. Comparing trees with high and low local density of conspecifics, we found no evidence for differences in inbreeding levels. The results are discussed within the framework of the emerging picture of the reproductive biology of tropical forest trees.  相似文献

We compared the seed fate of two animal‐dispersed, large‐seeded timber species (Dipteryx panamensis [Fabaceae] and Carapa guianensis [Meliaceae]) in logged and fragmented forests with that for continuous forest in northeastern Costa Rica. For both species, we quantified rates of seed removal (an index of vertebrate predation) and the fate of dispersed seeds (those carried away from their original location that either germinated or were not subsequently removed within three months). We predicted that (1) fewer seeds would be dispersed by vertebrates in fragmented forest than in continuous forest due to low population abundances after hunting and/or loss of suitable habitat, and (2) seed predation rates would be higher in forest fragments than in continuous forest due to high abundance of small‐bodied seed consumers. We compared three forest fragments currently managed for timber (140–350 ha) and a large reserve of continuous forest (La Selva, 1500 ha and connected to a national park). An exclusion experiment was performed (seeds placed in the open vs. seeds within semipermeable wire cages; 5 cm mesh size) to evaluate the relative roles of large and small animals on seed removal. Seed germination capacity did not differ among all four sites for both species. Removal of Dipteryx seeds was higher in forest fragments (50% removal within 10 days and related to the activity of small rodents) compared to La Selva (50% removal after 50 days). Also, more Dipteryx seeds were dispersed at La Selva than in fragmented forests. Contrary to our predictions, removal of Carapa seeds was equally high among all four sites, and there was a trend for more seeds of Carapa to be dispersed in fragments than in La Selva. Our results suggest that fragmentation effects on tree seed fate may be specific to species in question and contingent on the animal biota involved, and that management strategies for timber production based on regeneration from seed may differ between forest patches and extensive forests.  相似文献

Selective logging may impact patterns of genetic diversity within populations of harvested forest tree species by increasing distances separating conspecific trees, and modifying physical and biotic features of the forest habitat. We measured levels of gene diversity, inbreeding, pollen dispersal and spatial genetic structure (SGS) of an Amazonian insect-pollinated Carapa guianensis population before and after commercial selective logging. Similar levels of gene diversity and allelic richness were found before and after logging in both the adult and the seed generations. Pre- and post-harvest outcrossing rates were high, and not significantly different from one another. We found no significant levels of biparental inbreeding either before or after logging. Low levels of pollen pool differentiation were found, and the pre- vs. post-harvest difference was not significant. Pollen dispersal distance estimates averaged between 75 m and 265 m before logging, and between 76 m and 268 m after logging, depending on the value of tree density and the dispersal model used. There were weak and similar levels of differentiation of allele frequencies in the adults and in the pollen pool, before and after logging occurred, as well as weak and similar pre- and post-harvest levels of SGS among adult trees. The large neighbourhood sizes estimated suggest high historical levels of gene flow. Overall our results indicate that there is no clear short-term genetic impact of selective logging on this population of C. guianensis.  相似文献

The level and distribution of genetic diversity can be influenced by species life history traits and demographic factors, including perturbations that might produce population bottlenecks. Deforestation and forest fragmentation are common sources of population disturbance in contemporary populations of forest ecosystems. Although the genetic effects of forest fragmentation and deforestation have been examined by assessing levels of genetic variation in forest fragments that remain after logging, few considerations have been made of the populations that re-colonize once-cleared areas. Here we examine the effects of human-mediated population bottlenecks on the level and distribution of genetic diversity in natural populations of the long-lived forest tree species, Acer saccharum (sugar maple). We compared genetic variation and structure for populations of sugar maple found within old-growth forested area and in area that has re-colonized since logging. In this study the percent polymorphic loci and allelic richness estimates were reduced in the logged populations compared to old-growth populations. Jackknifed estimates of population genetic differentiation showed significantly higher differentiation among logged populations, with this result being consistently seen when individuals within populations were grouped according to diameter at breast height. The result of decreased genetic variation and higher levels of genetic structure among logged populations suggests that even one extensive bout of logging can alter the level and distribution of genetic variation in this forest tree species.  相似文献

The effects of population density on male and female reproductive success of Betula maximowicziana were evaluated in two mixed and two post-fire stands, with various population densities, ranging from 1.9 to 300.0 trees per ha, in central Hokkaido, Japan. First, we investigated ecological determinants of reproductive success (seed set and germination) of both seeds collected from the trees (tree seeds) and dispersed seeds collected from seed traps (dispersed seeds). We then evaluated the effects of population density on seed set and germination of tree seeds and dispersed seeds using a generalized linear mixed model (GLMM). Subsequently, we genotyped 950 seeds collected from mother trees and 940 seeds trapped after dispersal derived from tree seeds and dispersed seeds, respectively, using eleven microsatellite loci. Using the acquired data, we then evaluated the outcrossing rate and effective number of pollen donors (N ep) of the tree seeds, and the genetic structure of both pollen pools and dispersed seed populations. The seed set and germination rate of dispersed seeds was significantly lower both in the lowest-density stand and in the highest-density stand. The GLMM revealed that seed set and germination rates of dispersed seeds may be maximal at approximately 120 trees per hectare (optimal density). Outcrossing rates were consistently high (t m = 0.995), regardless of the population density. In contrast, N ep was lower in the lowest-density stands. Significant genetic structure of the dispersed seed population was found in two low-density stands, probably due to the limitation of overlapping seed shadows.  相似文献

Canopy tree species are the dominant elements of the species-rich, fragile and endangered tropical rain forest ecosystems, yet little is known about the genetics of these species. We provide an estimate of the outcrossing rate in a population of Pithecellobium pedicellare, a large canopy tree in the tropical rain forests of Costa Rica. The outcrossing rate was high (t = 951 ± 0.021) and the pollen pool contributing to the progeny arrays used to measure outcrossing rate showed departure from homogeneity. The high outcrossing rate indicates that individuals scattered over a large area in this low density population could be bound with each other via outcrossing. However, despite a high outcrossing rate, the potential for inbreeding in this population is not negligible. We found a relatively large number of albino and chlorotic seedlings among the progeny arrays of several seed parents. Heterogeneity of pollen pool further indicates that the population, though widely outcrossed, may be structured. A more detailed study of mating systems in tropical rain forest trees may provide additional insights into the mating patterns of these trees. Such studies will be useful not only in understanding the dynamics of micro-evolutionary processes, but also in the conservation and management of tropical forest trees.  相似文献

Polymorphic allozyme loci were used to estimate outcrossing rates for three tree species from a disturbed dry forest in southern Costa Rica. Estimates of the multilocus outcrossing rates of Cedrela odorata and Jacaranda copaia were 0.969 and 0.982, respectively, and suggest that these species may be self-incompatible. The subcanopy tree Stemmadenia donnell-smithii also demonstrated little self-fertilization based on an estimated outcrossing rate of 0.896. Significant heterogeneity in pollen allele frequencies among maternal trees was detected for at least two enzyme loci for each species. A test of correlated mating between progeny of S. donnell-smithii revealed that all seeds within a fruit were singly sired. In addition, the low estimates of biparental inbreeding and significant differences in pollen and ovule allele frequencies for this species suggest that gene flow into the sampled forest fragment may occur. The implications of deforestation on the mating systems of these tropical tree taxa are discussed.  相似文献

Hippophae rhamnoides ssp. sinensis is endemic to China, and it is a dioecious, outcrossing plant. Although many studies have been undertaken mainly on its agricultural, nutritional, medical, and ornamental value, little is known about its population genetics. This study uses random amplified polymorphic DNA to investigate the genetic diversity and population genetic structure of 13 natural populations of the subspecies sinensis. Fifteen primers amplified 107 reproducible bands, with 95 (88.79%) being polymorphic. The gene diversity within population was 0.168, considerably lower than that of tree species and most perennial, outcrossing species, but higher than that of annual or short-lived, selfing species. The Gst value showed that 18.3% of the total genetic variation resided among populations, a little lower than that of outcrossing species. The present results are quite similar to those previously reported in another subspecies, H ssp. . rhamnoides rhamnoides. The low genetic differentiation among populations in ssp. sinensis may be attributed to the long-distance dispersal of seeds facilitated by birds, in addition to its characteristics of outcrossing, wind pollination, and widespread distribution. No association between genetic distance and geographical distribution was found. The population relationships revealed by the UPGMA dendrogram parallel this result, in that genetic distance did not increase with geographic separation. This pattern of population differentiation may imply the adaptation of ssp. s populations to the local environment, given that its habitats vary greatly across its distribution.  相似文献

人类活动严重干扰着自然生态系统,其中砍伐是对森林生态系统最常见的干扰之一,它导致森林退化,植物种群变小,甚至灭绝,遗传多样性也随之下降。当被破坏的森林未被转换性利用时,则会逐渐恢复,但由于瓶颈效应,恢复起来的生态系统中植物种群的遗传结构可能会改变。恢复种群遗传组成的改变一方面与干扰的强度、频度和持续时间有关,另一方面,也受植物生活史特点的深刻影响。然而,我国对于砍伐后恢复起来的森林生态系统中生物多样性的改变,尤其是遗传多样性的改变的研究并不多见。研究在浙江省宁波市天童国家森林公园及周边地区选择了5个苦槠种群,采用SSR微卫星标记来分析砍伐对苦槠种群遗传结构的影响。5对多态SSR引物共得到了29个等位基因。种群内维持了较高的遗传多样性,种群间遗传分化程度较低,基因流达8.68。恢复林和成熟林种群的遗传多样性相差不大,以阿育王寺地区恢复种群的最高;表明砍伐对于苦槠种群遗传多样性的影响不大,这与苦槠较强的萌条能力有关。尽管如此,在恢复种群中观察到近期的种群瓶颈,显示出砍伐对种群遗传组成的影响;而在一个成熟林中也观察到种群瓶颈,这是因片断化导致种群变小之故。植被保存最好的天童国家森林公园内苦槠种群的遗传多样性却较低,这可能与成熟林中苦槠优势度较低有关。  相似文献

The effects of reduced‐impact logging (RIL) on the regeneration of commercial tree species were investigated, as long‐term timber yields depend partly on the availability of seedlings in a managed forest. On four occasions during a 20‐month period in the Tapajós National Forest (Eastern Amazon, Brazil), seven commercial tree species were assessed as follows: the long‐lived pioneers Bagassa guianensis and Jacaranda copaia; the partially shade‐tolerant Hymenaea courbaril, Dipteryx odorata, and Carapa guianensis; and the totally shade‐tolerant Symphonia globulifera and Manilkara huberi. In 2439 10 × 10 m plots, all individuals < 20 cm diameter at breast height (dbh) were assessed over three intervals, before, during, and after the forest being logged. Before logging, the density of seedlings and saplings of the seven species did not change. Logged trees were spatially aggregated, with 9.2 percent of the plots being heavily impacted by logging. After logging, the recruitment rate increased more than the mortality rate, so that post‐harvesting densities of seedlings and saplings increased. The increase in density was concentrated in logged plots with more disturbances. It is concluded that post‐harvesting heterogeneity of micro‐environments created by RIL may be an important component to be taken into account for sustainable forest management and conservation of commercial species.  相似文献

Dipteryx alata is a native fruit tree species of the cerrado (Brazilian savanna) that has great economic potential because of its multiple uses. Knowledge of how the genetic variability of this species is organized within and among populations would be useful for genetic conservation and breeding programs. We used nine simple sequence repeat (SSR) primers developed for Dipteryx odorata to evaluate the genetic structure of three populations of D. alata located in central Brazil based on a leaf sample analysis from 101 adults. The outcrossing rate was evaluated using 300 open-pollinated offspring from 25 seed-trees. Pollen dispersal was measured by parentage analysis. We used spatial genetic structure (SGS) to test the minimal distance for harvesting seeds in conservation and breeding programs. Our data indicate that the populations studied had a high degree of genetic diversity and population structure, as suggested by the high level of divergence among populations . The estimated outcrossing rate suggested a mixed mating system, and the intrapopulation fixation index was influenced by SGS. We conclude that seed harvesting for genetic conservation and breeding programs requires a minimum distance between trees of 196 m to avoid collecting seeds from related seed-trees.  相似文献

Wind-pollinated trees such as conifers have been considered relatively resistant to the deleterious effects of fragmentation and low density by virtue of their abundant production of airborne pollen. However, some studies indicated that population density affects the mating system even in wind-pollinated trees. In the present study, we examined the mating system and genetic structure of an endangered population (Boso) of Pinus parviflora var. parviflora, whose size has fallen by 80 % in only the last quarter century. Genetic analyses using four nuclear microsatellite markers showed that the population was in Hardy–Weinberg equilibrium and harbored a high level of genetic variation comparable to that of a larger population. Additionally, only weak genetic structure was detected among local patches. On the other hand, the outcrossing rate of seeds in the Boso population was much lower (0.277 from MLTR and 0.297 from paternity analysis) than that found in a larger population of the species (0.778 from MLTR). The outcrossing rate for each mother tree was found to correlate positively with local density around the tree in the Boso population. This suggests that the extensive selfing would be a direct result of pollen limitation caused by the extremely low density of the population: an average of 0.201 trees per hectare for local patches. Because the adult population was in HWE, it is likely that the selfed seeds produced at present cannot contribute to population regeneration. The present study suggests that low population density would increase extinction risk even for wind-pollinated conifer populations.  相似文献

Abstract

Human-induced disturbances threaten the genetic variation of wild plant populations. The genetic diversity and spatial population structure of the moss Isothecium myosuroides, a late-successional forest species, was investigated in subtropical cloud forests (La Gomera, Canary Islands) using inter-simple sequence repeat (ISSR) markers. Inter- and intrapopulation genetic variability was assessed in two ancient and four disturbed forest stands, which were classified according to their vegetation, forest age and type of disturbance. ISSR analysis of 144 epiphyte colonies with eight primers resulted in 211 reliably amplified bands. Our findings show that in disturbed forest stands, the population structure is increased, and the genetic diversity decreased compared with the levels observed in ancient forests. Although ancient and disturbed stands were located relatively close to each other, the (re-) established epiphyte populations did not reach their original genetic condition, 40 years following disturbance. Strong differentiation among populations of I. myosuroides at several spatial scales and differences in genetic diversity are mainly related to the local environmental conditions and the availability of suitable microhabitats in anthropogenically disturbed forest stands.  相似文献
