The phylogeny of ligulate and sulfuric‐acid containing species of Desmarestia, occurring worldwide from polar to temperate regions, was revised using a multigenic and polyphasic approach. Sequence data, gametophyte characteristics, and sporophyte morphology support reducing a total of 16 taxa to four different species. (1) D. herbacea, containing broad‐bladed and highly branched forms, has dioecious gametophytes. The three other species have monoecious gametophytes: (2) D. ligulata which is profusely branched and, except for one subspecies, narrow‐bladed, (3) Japanese ligulate Desmarestia, here described as D. japonica sp. nov., which is morphologically similar to D. ligulata but genetically distant from all other ligulate taxa. This species may have conserved the morphology of original ligulate Desmarestia. (4) D. dudresnayi, including unbranched or little branched broad‐bladed taxa. A figure of the holotype of D. dudresnayi, which was lost for decades, was relocated. The taxonomy is complemented by a comparison of internal transcribed spacer and cytochrome c oxidase subunit I (cox1) as potential barcode loci, with cox1 offering good resolution, reflecting species delimitations within the genus Desmarestia.  相似文献

Phylogenetic relationships in the Desmarestiales (Phaeophyceae) were inferred among the monotypic Arthrocladia (Arthrocladiaceae) and 27 isolates from Desmarestiaceae, representing 17 taxa of Desmarestia and the monotypic Antarctic genera Himantothallus and Phaeurus. Phaeurus and Arthrocladia were used as outgroups. Parsimony analyses of nuclear ribosomal DNA internal transcribed spacer (ITS1 and ITS2) sequences, in which gaps were both included and excluded, yielded well-resolved trees with a consistent general branching pattern. A parallel analysis of nine morphological and life-history characters and three ecological characters yielded a similar tree but provided little resolution in the terminal clades. The position of the monotypic Arthrocladia villosa within the Desmarestiales is consistent with monophyly for the order, but its position as the most primitive desmarestialean is not resolvable from the molecular data set. The basal position of Phaeurus, the Antarctic Desmarestia species, and Himantothallus is consistent with the hypothesis of a Southern Hemisphere origin for the family Desmarestiaceae. The more recent Northern Hemisphere “aculeata” clade evolved from an Antarctic ancestor. A “D. aculeata-like” species was ancestral to a lineage characterized by annual sporophytes with high sulfuric acid content, which radiated into many species, widely distributed in both hemispheres. Mapping of morphological and ecological characters onto the molecular tree confirm the informativeness of sulfuric acid-containing vacuoles and unilocular sporangial types. There is good congruence between phylogenetic tree topology and temperature impints in relation to biogeographic distribution, supporting the theory that temperature tolerance is a conservative trait.  相似文献

The mature male gametophyte of Ginkgo biloba can be divided into two regions: a large saccate structure that is suspended within the fertilization chamber above the archegonia, and a pervasive, highly branched haustorial system that ramifies through the intercellular air spaces of the apex of the nucellus. This morphology appears to differ in many ways from the simpler more typical male gametophytes of most other groups of seed plants. Growth and development of the male gametophyte of Ginkgo biloba were studied using computer reconstruction techniques to generate images of the gametophyte from data derived from serial sections through the ovule. These investigations reveal that morphological development of the male gametophyte of Ginkgo biloba is divided into three distinct phases: 1) Germination, characterized by an initial brief period of diffuse growth. This phenomenon has not been described for any other seed plant male gametophyte; 2) Initiation of tip growth and the formation of a tubular body, as typifies all seed plant male gametophytes. In Ginkgo, this is accompanied by a high degree of branching, giving rise to an extensively branched haustorial system; 3) Late swelling of the proximal unbranched portion of the gametophyte resulting in formation of the saccate structure that is characteristic of the mature gametophyte. This process appears to be very similar to late development in cycad male gametophytes. Thus, despite the seemingly anomalous morphology of the mature male gametophyte of Ginkgo biloba, specific patterns of growth and development are in many ways similar to growth processes expressed by the male gametophytes of some or all major groups of seed plants.  相似文献

Exudates from the brown algaeCaepidium antarcticum andDesmarestia sp. were investigated for their ability to associate with hydrophobic pollutants such as polychlorinated biphenyls (PCB s). The percentage of PCB associated with algal exudates ranged from 79% for decachlorobiphenyl to 23% for the pentachlorobiphenyl congener No. 95. An improved method for the preparation of chromosomes from the male gametophyte of the alga Laminaria japonica Aresch. was described. The male gametophyte was pretreated with pDB (p-dichlorobenzene) and 8-hydroxyquinoline in order to clear cell wall and soften cytoplasm. The samples were treated by mordant iron alum [FeNH4 (SO4)2⋅6H2O] followed by staining with haemotoxylin. Well-spread and highly stained chromosomes were observed without precipitation. The chromosome number of male gametophyte of L. japonica was estimated to be 31. %

The female gametophyte of Ginkgo biloba is the only seed plant gametophyte known to contain chlorophyll. Measurements of photosynthetically active radiation (PAR) indicate that a gametophyte growing within an ovule can receive significant quantities of light (70 μmol photons m–2 s–1). Under the conditions of our experiments, whole gametophytes dissected free from ovules were capable of gross photosynthesis, but not net photosynthesis. On a dry wt basis, the maximum rate of carbon fixation under near saturating light intensities was 3.64 × 10–3 μmol CO2g–1 s–1. The unique ability of the female gametophyte of Ginkgo biloba to produce chlorophyll and engage in photosynthesis appears to result from its exposure to sufficient levels of light and a predisposition to react to this stimulus by the development of a functional photosynthetic apparatus.  相似文献

The development of male gametophyte and female gametophyte within a floret of rye (Secale cereale L.) was examined. Generally, meiosis in microsporocytes and in megasporocytes occurs simultaneously in most florets, but the period from zygotene to tetrad meiosis in the megasporocyte progresses more slowly than that in the microsporocyte. When the female gametophyte has one nucleus and no vacuoles, the male gametophyte has a single, eccentric nucleus. By the time the female gametophyte develops to the vacuolated one-, two-, four-, and eight-nucleate stages and to the growth and differentiation of the egg apparatus stage, the male gametophyte reaches the two-celled pollen stage. As the female gametophyte matures, the male gametophyte also reaches maturity. The duration of male gametophyte development from microspore mother cell and the duration of female gametophyte development from megaspore mother cell are the same in most florets. The relationship between sexual development of cross-pollinated rye is similar to that of self-pollinated wheat (Triticum aestivum L.). The spore germination, protonemal development, and gametophyte differentiation of Hypnum pacleseens were observed in cultivation. Photomicrographs showed that spore germination of Hypnum pacleseens occured within the exospore. Its protonema is massive with filamentous chloronema formed inside. The terminal part of the chloronema differentiated into filamentous caulonema and its rhizoid was derived from the apical cell of the filamentous chloronema. The initial cell of gametophyte differentiated from chloronema and caulonema. Sporeling-type of Hypnum pacleseens is developmentally similar to Glyphmitrium-type.  相似文献

为弄清罗汉果(Siraitia grosvenorii)大孢子发生、雌配子体发育过程与花部形态特征、胚珠的关系,运用石蜡切片法对罗汉果子房进行了显微观察。结果表明,罗汉果的胚珠倒生,双珠被,厚珠心,大孢子四分体呈线型排列,合点端一个大孢子分化为功能大孢子,成熟胚囊为蓼型。花蕾形态、胚珠变化与大孢子发生、雌配子体的发育时期具有一定相关性,当子房长度为7.0 mm≤L<9.0 mm,珠心呈椭圆形时,约有45.83%的大孢子母细胞处于减数分裂时期。因此,依据罗汉果花部形态可有效确定大孢子发生与雌配子体发育的时期。  相似文献

All combinations of individuals of Macrocystis pyrifera (L.) C. Agardh, M. integrifolia Bory, and M. angustifolia Bory hybridized. Gametophyte isolates obtained from 18 individuals were used, including M. pyrifera and M. integrifolia from the extremes of their Northern Hemisphere ranges along the Pacific Coast of North America and M. pyrifera and M. angustifolia from Tasmania, Australia. All combinations of gametophytes produced sporophytes of normal morphology, with the exception of crosses involving three gametophyte isolates. One female (M. integrifolia) and two male (M. pyrifera and M. angustifolia)gametophyte isolates were unable to produce normal sporophytes in combination with gametophytes of the opposite sex. Some cultures of female gametophytes produced abnormally shaped parthenogenetic sporophytes. Gametophytes and sporangia of M. pyrifera had n= 16 chromosomes. The M. integrifolia female gametophyte that was unable to produce normal sporophytes had n = ca. 32 chromosomes. These results show that these species of Macrocystis have not become reproductively isolated. Although these species may be considered conspecific according to the biological species concept, we recommend that they continue to be recognized as separate species based on morphological differences.  相似文献

Propagation of gametophytes and sporophytes using mechanical fragmentation has been considered a suitable method for mass production of ferns. This study aimed to develop a practical propagation method for Lemmaphyllum microphyllum C. Presl, which is a fern of significant ornamental and medicinal value. Gametophytes were obtained through in vitro spore germination and used for propagation experiments. The gametophyte was mechanically fragmented using a scalpel into small fragments, which were then used to investigate gametophyte proliferation. In addition, the gametophyte was fragmented using a blender and then used to study sporophyte formation. Optimal proliferation conditions of the gametophyte were determined using Murashige and Skoog (MS) basal medium (double-, full-, half-, quarter-strength), Knop medium, and medium components (sucrose, nitrogen sources, activated charcoal), at various concentrations. The fresh weight of the gametophyte was 14-fold higher than that of gametophytes (300 mg) used as culture material, when cultured on double-strength MS. Moreover, 1 g of the gametophyte fragmented in 25 mL of distilled water formed more than 430 sporophytes in a soil mixture in an area of 7.5 cm2. The sporophytes were successfully cultivated in the greenhouse after acclimation. A large-scale production method for L. microphyllum that can be easily implemented in a fern production farm is outlined.

Eighteen polymorphic microsatellite DNA markers were developed for Laminaria japonica, a brown alga cultured intensively in China. These markers are independent from each other and are moderately variable in L. japonica. The number of alleles and gene diversity detected in 53 gametophyte clones representing the varieties of L. japonica cultured once or currently in China range from two to nine and from 0.355 to 0.768, respectively. These markers will certainly facilitate the management and exploitation of the germplasm resource of L. japonica conserved indoor as gametophyte clones and the determination of the genetic diversity of L. japonica naturally distributed.  相似文献

The sublittoral zonation of macroalgae and abundant animals in Potter Cove, King George Island, is described in relation to substrate, exposure to turbulence and impact of grounding icebergs. Implications on the ecological niches of the most prominent phaeophytes are discussed. It is concluded that Desmarestia anceps and D. menziesii exclude Himantothallus grandifolius under favourable conditions. However, Himantothallus, by its potential to inhabit unstable substrates, may be better adapted to withstand the ice impact. The replacement of Desmarestia by Himantothallus at greater depth can be explained only partially at present.  相似文献

The development of the female gametophytes of Epipactis atrorubens, E. helleborine, and E. palustris have been investigated using confocal scanning laser microscopy. These species have T-tetrads and eight-nucleate female gametophytes in common. The development of the female gametophyte is monosporic in accordance with the Polygonum type. Furthermore, the outer integument grows slowly and reaches to the middle of the inner integument, when the female gametophyte is mature. In the first two species, T-tetrads develop prior to anthesis, which may be correlated with autogamy. The development of the female gametophyte of E. palustris appears to be similar in plants from different localities.  相似文献

During anther development the male gametophyte develops inside the locule and the tapetal cells provide all nutrients for its development. Magnesium Transporter 5 (MGT5) is a member of the MGT family and has dual functions of Mg export and import. Here, we show that male gametophyte mitosis and intine formation are defective in a mgt5 mutant. The transient expression of GFP‐MGT5 revealed that MGT5 is localized in the plasma membrane. These findings suggest that in the male gametophyte MGT5 plays a role in importing Mg from the locule and that Mg is essential for male gametophyte development. The expression of MGT5 in the knockout ABORTED MICROSPORES (AMS) mutant (AMS being an essential regulator of tapetum) is tremendously reduced. Chromatin immunoprecipitation and mobility shift assay experiments demonstrated that AMS can directly bind the promoter of MGT5. An immunoelectron microscopy assay revealed that MGT5‐His is localized to the plasma membrane of the tapetum. These findings suggest that AMS directly regulates MGT5 in the tapetum and thus induces export of Mg into the locule. The mgt5 plant exhibits severe male sterility while the expression of MGT5 under the tapetum‐specific promoter A9 partly rescued mgt5 fertility. mgt5 fertility was restored under high‐Mg conditions. These findings suggest that the mgt5 tapetum still has the ability to export Mg and that a sufficient supply of Mg from the tapetum can improve the importation of Mg in the mgt5 male gametophyte. Therefore, MGT5 plays an important role in Mg transport from the tapetum to the microspore.  相似文献

Fern gametophytes have often been neglected in research; however, studies on gametophytes are crucial for a better understanding of the evolution of ferns. During their life cycle, some gametophytes produce large and long‐lived populations without producing sporophytes and reproduce independently through asexual means, such as through the formation of gemmae. In this study, we investigated independent gametophytes on the Jeju Island of Korea, which was located on the land bridge between East China and Japan during the glacial periods. Fourteen gametophyte populations were collected from seven sites, of which 13 populations were clearly identified as belonging to four fern species known to occur in Jeju Island with BLAST searches using rbcL and trnL‐F sequences. Surprisingly, the last remaining population constituted undescribed taxa in Korea. We presented the first report of the independent gametophytes of Antrophyum obovatum Baker which has not been previously recorded in Korea. It has been supposed that many ferns sought suitable habitat throughout the land bridge between China and Japan. However, Jeju Island might be unsuitable for vittarioid ferns that prefer a tropical or subtropical environment. Consequently, only two species of vittariod ferns (A. obovatum and Haplopteris flexuosa (Fée) E.H. Crane) in the form of a gametophyte and sporophyte, respectively, exist on Jeju Island. Therefore, this gametophyte population must be protected and managed from a conservation perspective. In the case of the independent gametophyte of Hymenophyllum wrightii Bosch, haplotype analysis was conducted based on the rbcL sequences and the result supported that the North American populations were migrated from Japan through land bridge during the glacial periods and Jeju populations were recently established by long‐distance dispersal of the Japanese populations.  相似文献

A tentative amplified fragment length polymorphism–simple sequence repeat (AFLP–SSR) linkage map of Laminaria was constructed using a haploid population of 40 gametophyte clones isolated from an individual of Dongfang No. 2, the first commercially cultured hybrid of a female gametophyte clone of L. japonica Aresch. [=Saccharina japonica (Aresch.) C. E. Lane, C. Mayes et G. W. Saunders] and a male one of L. longissima Miyabe [=Saccharina longissima (Miyabe) C. E. Lane, C. Mayes et G. W. Saunders]. To the map, 263 markers (255 AFLP, seven SSR, and the gametophyte sex) were assigned. The map consisted of 25 linkage groups (LGs) with ≥ four markers, five triplets, and 15 doublets, which is 1,629.0 centiMorgans (cM) in length, covering 66% of Laminaria genome. The maximum space between loci is 24.63 cM. A putative sex‐determining region was identified in LG2, which was characterized by a dense marker distribution around the gametophyte sex locus. The linkage map itself and the methodology associated with its construction will facilitate the genetic study and further trials of the linkage map construction of Laminaria.  相似文献

金毛狗[Cibotium barometz(L.)J.Sm.]是珍稀观赏蕨类的重要类群,为国家二级重点保护野生植物。该研究以金毛狗孢子为试验材料,探究培养密度、外源赤霉素以及光质等不同环境因子对金毛狗配子体性别分化的影响,为金毛狗人工繁育和蕨类植物配子体性别决定机制研究提供技术支持。结果表明:(1)低配子体培养密度(1个/cm2和5个/cm2)有利于颈卵器和雌配子体形成,随着配子体培养密度增加,颈卵器平均数量及雌配子体比率下降,精子器平均数量以及雄配子体和两性配子体比例增加,但配子体培养密度过高(80个/cm2)会导致大量无性配子体产生。(2)不同配子体培养密度下,随着培养时间延长,两性配子体比率均有增加,且增加幅度基本一致。(3)外源GA4显著抑制颈卵器和雌配子体形成,并显著促进精子器和雄配子体形成;外源GA3对金毛狗配子体性别分化没有显著影响。(4)白光、红光、蓝光等不同光质对金毛狗配子体性别分化未产生显著影响,但会影响配子体的发育和形态建成。  相似文献

Abstract

(1) The ratio of 14CO2 uptake of fully expanded capsules to that of corresponding gametophytes in the species studied was about 1 : 15 in Mnium hornum, 1 : 35 in Pleurldium acuminatum and around 1 : 1 in Funaria hygrometriea.

(2) Translocation of photosynthate from gametophyte to sporophyte took place within 1 to 4 days of exposure to 14CO2; in expanded capsules the final activity was at least equal to that remaining in the gametophyte.

(3) In M hornum there was no evidence of reverse translocation from sporophyte to gametophyte.

(4) Translocation from gametophyte continued at a high level throughout the expansion and differentiation of the capsule in M. hornum. The activity reaching the sporophyte was approximately proportional to its fresh weight.

(5) Both in situ photosynthesis and translocation to the capsule of F. hygrometrica rose to a maximum during the latter part of capsule expansion and ensuing differentiation, subsequently declining as the green tissues of the capsule senesced.

(6) Fully expanded (but premeiotic) capsules of M. hornum contributed approximately 20% of the assiinilate necessary for their growth. The corresponding figure for P. acuminatum was about 10%, and for F. hygrometrica about 50%.  相似文献
