Edaphic diatoms were collected from 5 representative habitats of Canary Creek salt marsh, Lewes, Delaware, from 24 July 1969 to 21 July 1970. Of the 104 taxa encountered, 32 had a general distribution on the marsh and 41 were endemic to one of the 5 habitats sampled. Three of the habitats supported stands of grasses: tall Spartina alterniflora Loisel., dwarf S. alterniflora, and Distichlis spicata (L.) Greene; and these habitats possessed the highest species diversity (H') and the greatest number of diatom species. The remaining 2 habitats, a bare bank and a panne, were devoid of macroscopic vegetation. The diatoms of these last 2 habitats were exposed to hypersaline conditions during warmer periods of the year and this was considered a contributing factor to the lower values observed for the aforementioned parameters of community structure. A comprehensive examination of the community structure characterizing the 5 habitats, employing statistical analyses and the distribution of species, showed each habitat to support its own unique and easily recognizable edaphic diatom community. A multiple regression analysis indicated that the differences between the 5 communities were closely related to differences in temperature and elevation between the habitats, and also a result of significant interactions between edaphic diatoms and filamentous algae.  相似文献

Most biominerals appear to be composites of organic material and mineral. Whether biosilica is such a composite is unresolved because of a lack of evidence for such organic components. We present evidence that organic material exists within diatom biosilica and can be extracted using HF/NH4F solutions from frustules isolated from Cyclotella meneghiniana Kütz and diatomaceous earth. To eliminate organic casing on the silicified frustules as a source of organic materials, the casing was removed by oxidation of frustules with NaOCl before extraction. The removal of the casing was confirmed in that oxidized frustules no longer displayed the ability to be stained with ruthenium red and fluorescamine. Frustules examined with EDXA showed an emission peak from sulfur before treatment but no peak following treatment, indicating that oxidation removed organic sulfur. The organic material obtained from extracts of fresh frustules contained both soluble and insoluble components. Only soluble material was evident in extracts from diatomaceous earth. The soluble material appears to contain glycoproteins with relatively high levels of serine and glycine. The soluble proteins from fresh frustules also appear to be phosphorylated. Indirect evidence is presented that suggests the soluble proteins may contain regions of primary structure enriched in anionic amino acids. The soluble extracts differ from general cell contents when the two fractions are compared, suggesting that frustules contain specialized organic material. The identification of silica-specific organic material suggests that mineralization in diatoms may be in part matrix-mediated.  相似文献

Several populations of Cymbella cistula (Ehr.) Kirchn. were studied. An epiphytic population contained smaller specimens than any reported by recent authors. The size range of members of the epiphytic population included specimens as small as 24 μ long by 11 μ wide. These small specimens could be mistaken for C. cistula var. maculata (Kütz.) V. H., but differ in having isolated puncta, which, may be indistinct, and somewhat coarser striae.  相似文献

An obligate requirement for selenium is demonstrated in axenic culture of the coastal marine diatom Thalassiosira pseudonana (clone 3H) (Hust.) Hasle and Heimdal grown in artificial seawater medium. Selenium deficiency was characterized by a reduction in growth rate and eventually by a cessation of cell division. The addition of 10−10 M Na2SeO3 to nutrient enriched artificial seawater resulted in excellent growth of T. pseudonana and only a slight inhibition of growth occurred at Na2SeO3 concentrations of 10−3 and 10−2 M. By contrast, Na2SeO4 failed to support growth of T. pseudonana when supplied at concentrations less than 10−7 M and the growth rate at this concentration was only one quarter of the maximum growth rate. The addition of 10−3 and 10−2 M Na2SeO4 to the culture medium was toxic and cell growth was completely inhibited. Eleven trace elements were tested for their ability to replace the selenium requirement by this alga find all were without effect. In selenium-deficient and selenium-starved cultures of T. pseudonana cell volume increased as much as 10-fold as a result of an increase in cell length (along the pervalvar axis) but cell width was constant. It is concluded that selenium is an indispensable element for the growth of T. pseudonana and it should be included as a nutrient enrichment to artificial seawater medium when culturing this alga.  相似文献

The morphology, ecology, and taxonomy of a pennate diatom, Cymbella cursiformis nom, nov. (family Cymbellaceae), from Cedar Bog, Champaign County, Ohio, originally described from Tasmania by Hustedt as Cymbella cistula var. gracilis, are reported. Photographs of specimens on Hustedt's type, slide, are indistinguishable from Cedar Bog specimens. Scanning electron microscopy and light microscopy reveal that, this taxon cannot be considered a variety of Cymbella cistula (Ehr.) Kirchn.; therefore it has been raised to specific rank.  相似文献

The ultrastructure of the oval, fusiform and triradiate morphotypes of Phaeodactylum tricornutum Bohlin is described. The organization and structure of the cytoplasmic organelles is similar in all three morphotypes, except that the vacuoles occupy the extra volume created by the arms of the fusiform and triradiate cells. The frustule in fusiform and triradiate cells is organic; in the oval type it may be organic or one of the valves may have a silica frustule surrounded by an organic wall. In all cells, the organic cell wall has up to 10 silica bands (13 nm wide) embedded in its surface in the girdle region, lacks girdle bands, and has an outer corrugated cell wall layer, except in the girdle region. Cell division, organic wall formation and silica deposition are described in detail. Four types of oval cells are also described. The relation to other diatoms is discussed.  相似文献

Growth rates of two clones of the freshwater planktonic diatom Asterionella formosa Hass. were measured under conditions in which external silicon concentrations controlled growth. Clone AfOH2 from Lake Ohrid, Yugoslavia, had a higher maximum growth rate (μmax= 1.11 doublings/day) and apparent half-saturation constant (Ksi] + Sio= 1.93 μM Si) than clone L262 from Lake Windermere, England. (μmax= 0.61 doublings/day; Ksi+ Sio= 1.09 μM Si). Klim, the silicon concentration at μ= 0.9 μmax, is 13.8 μM Si for clone AfOH2 and 6.5 μM Si for clone L262. These values agree well with published field observations showing A. formosa populations decreasing below 0.5 mg/l SiO2 (= 8.4 μM Si). Calculations of yield gave a range of 0.5–1.5 μM Si/106 cells for clone AfOH2 and 0.6–1.9 μM Si/106 cells for clone L262.  相似文献

The unicellular red alga Porphyridium aerugineum was shown to be encapsulated by an amorphous, water-soluble, polyanionic polysaccharide of high molecular weight. The encapsulating polysaccharide is qualitatively identical with polysaccharide found dissolved in large quantity in the culture medium. The kinetics of extracellular polysaccharide production as a function of cell age was studied. Rates of production (on a per cell basis) of both encapsulating and dissolved polysaccharides are greatest in stationary phase light-grown cultures. Dissolved polysaccharide was quantitatively isolated by precipitation with cetyl pyridinium chloride, conversion to the calcium salt, and reprecipitation with ethanol. The procedure yields a spectrally pure product, which is composed of glucose, galactose, xylose, and 2 undetermined, sugar components, and has a sulfate content of 7.6% by weight. Electron microscopy of Porphyridium revealed that Golgi vesicles transport, polymerized polysaccharides to and through the cell membrane. Similar vesicles were observed in the multicellular Pseudogloiophloea, indicating that the Golgi complex plays a crucial role in the production of extracellular polysaccharides by the red algae. H14CO3- pulse-label experiments resulted in labeled extracellular polysaccharide in which all the constituent components contained 14C. Rates of excretion of polysaccharide were found, to follow a cyclic pattern, correlated generally with the division cycle, of the cell.  相似文献

Bacteriol. 85:273-278. 1963.-The exocellular nuclease of Serratia marcescens, isolated by anion-exchange chromatography on diethylaminoethyl-Sephadex, depolymerized deoxyribonucleic acid, ribonucleic acid, and the polynucleotide which is refractory to pancreatic ribonuclease activity. The enzyme was tentatively classified as a nonspecific phosphodiesterase. Magnesium was essential for activity, which was optimal at pH 8.8. Small single‐celled Chaetoceros sp. are often widely distributed, but frequently overlooked. An estuarine diatom with an extremely high growth potential under optimal conditions was isolated from the Shinkawa‐Kasugagawa estuary in the eastern part of the Seto Inland Sea, western Japan. It was identified as Chaetoceros salsugineum based on morphological observations. This strain had a specific growth rate of 0.54 h?1 at 30°C under 700 μmol · m?2 · s?1 (about 30% of natural maximal summer light) with a 14:10 L:D cycle; there was little growth in the dark. However, under continuous light it grew at only 0.35 h?1 or a daily specific growth rate of 8.4 d?1. In addition, cell density, chlorophyll a, and particulate organic carbon concentrations increased by about 1000 times in 24 h at 30°C under 700 μmol · m?2 · s?1 with a 14:10 L:D cycle, showing a growth rate of close to 7 d?1. This very rapid growth rate may be the result of adaptation to this estuarine environment with high light and temperature. Thus, C. salsugineum can be an important primary producer in this estuary in summer and also an important organism for further physiological and genetic research.  相似文献

对我国首次在西太平洋挑战者深渊采集的硅藻软泥沉积物开展硅藻分析,结果表明硅藻软泥由数量巨大的大筛盘藻[Ethmodiscus rex(Rattray)Hendey]碎片组成,并伴生有少量热带远洋浮游种类硅藻。硅藻软泥属于大筛盘藻软泥类型,推测可能形成于更新世中后期,代表着大规模E.rex勃发事件发生。其形成机制与冰期时段南极深层水入侵西北太平洋增加水体硅含量、减弱硅藻溶蚀作用,并补充次表层水体硅含量而激发E.rex竞争优势密切相关。本文为理解低纬度海沟区硅藻微体生物随全球气候和区域海洋环境变迁而发生显著变化提供了基础材料。  相似文献

Two clones of Pinnularia brebissonii (Kütz.) Rabh. var. brebissonii were established and maintained in logarithmic phase of growth. Initial length of the cells was 37 μm. As cell division occurred, the mean length of cells in each population decreased as predicted by the MacDonald-Pfitzer hypothesis; however, the decrease in mean length was not uniform throughout the growth period. This nonuniformity is probably caused by nonrandom division of cells in the population or by a changing increment of size reduction due to division. The initial increment of size reduction was calculated as 0.7 μm/division. The smallest, cells observed were 8 μm long. As cells decrease in length, cell volume decreases and the proportion of cells with aberrant valve structure increases. More than 90% of the valves were abnormal in a population with mean length of 14 μm. The abnormalities of structure involved the raphe, the central area and the striae.  相似文献

由固体培养法得到的中国福建产银耳孢子(Tremella fuciformis Berk.),经沸水提取、三氯醋酸-正丁醇除蛋白、乙醇沉淀分离制备的多糖具有明显提高机体免疫功能的作用。应用CTAB(溴化十六烷基三甲胺)络合法进一步精制后的银耳孢子多糖干品,含糖量达80%左右。经纸层析,醋酸纤维素薄膜电泳及琼脂糖电泳分析表明,其主成分为单一斑点。组成分析发现分子中富含己糖醛酸,主要组成单糖有木糖、甘露糖、岩藻糖、葡萄糖、阿拉伯糖及葡萄糖醛酸等。组成中不含蛋白和核酸类物质。红外光谱分析证明,分子中具有典型酰基结构。  相似文献

In this study, we describe different combinations of physical separation and antibiotic treatment to remove associated bacteria from freshwater diatoms. Diatoms were purified either from natural epilithic biofilms or from unialgal cultures. We determined that for most strains, different purification procedures have to be combined individually. In a new approach, we show that for some diatom strains, the substitution of associated aquatic bacteria by an antibiotic‐sensitive Escherichia coli strain and subsequent treatment with antibiotics may be a successful strategy to obtain axenic diatom cultures. Axenic diatom cultures are essential to study the physiology and biochemistry of individual strains as well as their responses to environmental changes without interference of accompanying bacteria.  相似文献

Molecular phylogenetic analysis of the conjugating green algae (Class Zygnemophyceae) using nuclear (SSU rDNA) and chloroplast ( rbcL ) gene sequences has resolved hypotheses of relationship at the class, order, and family levels, but several key questions will require data from additional genes. Based on SSU and rbc L sequences, the Zygnemophyceae and Desmidiales are monophyletic, and families of placoderm desmids are distinct clades (Desmidiaceae, Peniaceae, Closteriaceae, and Gonatozygaceae). In contrast, the Zygnemataceae and Mesotaeniaceae are paraphyletic, although whether these two traditional families constitute a clade is uncertain. In addition, relationships of genera within families have proven resistant to resolution with these two oft-used genes. We have sequenced the cox III gene from the mitochondrial genome to address some of these ambiguous portions of the phylogeny of conjugating green algae. The cox III gene is more variable than rbc L or SSU rDNA and offers greater resolving power for relationships of genera. We present preliminary analyses of coxIII sequences from each of the traditional families of Zygnemophyceae and contrast the resulting topologies with those derived from nuclear and chloroplast genes.  相似文献
