实验针对三株海洋富油微藻:球等鞭金藻(Isochrysis galbana CCMM5001)、一种等鞭金藻(Isochrysis sp. CCMM5002)和一种微拟球藻(Nannochloropsis sp. CCMM7001),研究了它们在通入0.03%(空气)、5%、10%三个浓度CO2培养条件下的生长特性,同时考察了其总油脂及中性脂的累积情况。结果显示,富碳培养有利于这三株海洋微藻的生长,但最适生长的CO2浓度不同。球等鞭金藻(Isochrysis galbana CCMM5001)和等鞭金藻(Isochrysis sp. CCMM5002)在通入10% CO2时具有最大产率,分别达到(182.287.07) mg/(Ld)和(164.227.10) mg/(Ld),而微拟球藻在通入5%时具有最大产率,达到(122.251.17) mg/(Ld),随着CO2浓度的增加,三株海洋微藻的总脂含量和中性脂含量有明显提高。在通入10% CO2条件下,球等鞭金藻(Isochrysis galbana CCMM5001)、等鞭金藻(Isochrysis sp. CCMM5002)和微拟球藻(Nannochloropsis sp. CCMM7001)的总脂含量分别达到(45.154.03)%、(47.151.20)%和(41.201.69)%;从中性脂的累积规律来看,三株藻均在平台期的累积达到最大,脂肪酸分析结果表明三株藻种适合制备生物柴油的C14-C18系脂肪酸相对含量在不同CO2条件下基本保持不变,维持在90%左右。实验结果显示,研究的藻株作为富油高固碳优良藻株,具备用于海洋生物质能耦合CO2减排开发的潜力。    相似文献

Total biomass dry weight (DW) and ash free dry weight (AFDW) of five species of marine microalgae, Dunaliella sp., Isochrysis galbana, Nannochloropsis sp., Nitzschiaclosterium and Porphyridium cruentum, retained on filter paper, were determined. Dunaliella and Isochrysis cells have no cell wall; Nannochloropsis, Nitzschia and Porphyridium possess a cell wall and Nitzschia and Porphyridium cells are covered by silica and mucilage coating, respectively. In all these algae, DW of non-washed samples was at least 1.2 times higher than those washed by distilled water,0.9% sodium chloride, 0.5 M ammonium formate or 0.5 M ammonium bicarbonate. DW of 0.9% sodium chloride washed samples was more than 0.8 times higher than the other three washed samples. In most of the cases, there was no significant difference between DW of samples washed by ammonium formate and ammonium bicarbonate solutions (p>0.05). The AFDW of the non-washed algal samples was about twice that washed samples, and could be accounted for by volatile component in the sea water medium. Isotonic solution of ammonium bicarbonate is a satisfactory washing agent for algal cells for dry weight determination. The aim of this research was to study the impact of nitrogen starvation on the production of two major secondary metabolites, fatty acids and carbohydrates, in two microalgae: Nannochloropsis sp. and Haematococcus pluvialis. The major response to nitrogen starvation in both algae occurred within the first 2 days, accompanied by a sharp reduction in chlorophyll content. However, the pattern of the response differed between the two microalgae. In H. pluvialis, the first response to nitrogen starvation was intensive production of carbohydrates, accumulating to up to 63% of dry weight by day 1; on day 2, the total carbohydrate content decreased and was partially degraded, possibly to support fatty acid synthesis. Under these conditions, H. pluvialis accumulated up to 35% total fatty acids in the biomass. In Nannochloropsis sp., the immediate and major response, which was maintained throughout the entire period of exposure to stress, was production of fatty acids, accumulating up to 50% of dry weight, while carbohydrate content in the biomass remained stable at 18%. In addition, we tested the effect of the lipid-synthesis inhibitor sesamol, known to inhibit malic enzyme, on the balance between total fatty acid and carbohydrate contents in H. pluvialis and Nannochloropsis sp. In both cultures, sesamol inhibited fatty acid accumulation, but the carbohydrate content was reduced as well, albeit to a lesser extent. These findings demonstrate the complexity of the stress-response and the potential link between fatty acid and carbohydrate synthesis.  相似文献

Three morphologically different anaerobic fungi, a Neocallimastix sp. strain (LM-1), a Piromonas sp. strain (SM-1), and a Sphaeromonas sp. strain (NM-1), were isolated from the rumens of sheep. Growth studies were conducted with each isolate in batch cultures by using an anaerobic semidefined medium that lacked ruminal fluid and contained 0.5% cellobiose. Cultures were incubated for periods of up to 10 days, and fungal growth was assessed at regular intervals by dry weight measurements. Samples of fungal biomass were also analyzed for cell-associated protein and, after acid hydrolysis, for chitin as hexosamine. The isolates produced similar yields of dry weight and contained similar amounts of protein. However, strain LM-1 grew at a higher rate and contained less than half the level of chitin compared with the other two isolates. There were high positive correlations between chitin and protein for all three fungi, but comparisons of these parameters with dry weights were affected by the presence of variable amounts of storage carbohydrate. The amount of storage carbohydrate reached maximum levels in strain LM-1 during mid-growth phase and then quickly declined thereafter. When dry weight yields for strain LM-1 were adjusted for changes in storage carbohydrate, high positive correlations were obtained between dry weight and protein or chitin. The storage carbohydrate was probably an alpha-1,4-glucan with alpha-1,6 branches.  相似文献

Protein, lipid, water and caloric contents of immature rainbow trout, relative to size of whole fish, growing at different rates were examined by use of allometric analysis (y = axb, Huxley, 1932). Fish grew at different rates as a result of differences in ration size (satiation, or 4–5% of dry body weight), temperature (7 and 12°C) and bGH (bovine growth hormone) administration. In fingerlings, protein, lipid and caloric contents tended to increase (v. body weight) as a percentage of body composition, whereas above fingerling size, protein decreased while lipid and caloric contents still increased. These trends occurred regardless of growth rate differences. The correlations between protein, lipid, caloric contents and body weight were high so reliable estimates of body components can be made from body weight for all experimental treatments. At satiation rations (7 and 12°C), there were no significant differences in protein content, but lipid and caloric contents were significantly higher in control fish. At low rations, protein and caloric contents were lower than those at satiation rations and lipid was lower than in the control group. On a dry weight basis, in uninjected fish at 12°C, ration size did not influence the percentage composition (protein and lipid) but the low ration group had lower energy values per unit of body dry weight. At low temperature (7°C satiation), fish had lower lipid and higher protein content (dry weight basis) than their controls (12°C satiation), thereby resembling bGH injected fish at satiation rations. Values of caloric content estimated from protein and lipid values by use of standard conversion factors differed sufficiently from caloric values directly determined by bomb calorimetry to suggest that caloric values of lipids may change during growth. The increase in body caloric content during growth apparently results mainly from an increase in the percentage of lipid. The similarity of body composition between different growth rate groups implied a tendency for conservation of relative proportions of components such that body composition can be approximated from body weight. Equations were also given to describe the relationship between water content and body constituents, and the relationship between condition factor (K) and both the body dry weight and lipid content.  相似文献

在温度23.5~24.0 ℃和盐度29.5~30.0条件下,研究了球等鞭金藻、小球藻、牟氏角毛藻和新月菱形藻4种单细胞藻类单独及组合投喂24 d后对魁蚶稚贝生长与存活的影响.结果表明: 不同饵料投喂条件下,稚贝存活率均在95%以上,不同处理组间差异不显著.单一饵料投喂组中,球等鞭金藻组的稚贝生长最快,小球藻组的稚贝生长最慢.组合投喂组中,球等鞭金藻与其他3种单胞藻混合(1∶1)投喂组稚贝壳长及特定生长率均优于其他组,其中与小球藻混合投喂的效果最好,稚贝壳长和壳高的特定生长率分别为5.6%·d-1和6.4 %·d-1.研究结果可为魁蚶人工苗种培育技术的优化提供参考依据.

Forty psychrophile or psychrotrophic crude-oil-utilizing marine bacteria were investigated for their ability to accumulate lipid storage compounds in the cytoplasm during cultivation under nitrogen-limiting conditions. Most of them (73%) were able to accumulate specialized lipids like polyhydroxyalkanoic acids (PHA) while other lipids such as wax esters occurred in two isolates. Accumulation of PHA occurred predominantly at low temperatures (4–20 °C) as demonstrated for three isolates. Electron microscopy revealed polyphosphate inclusions occurring in two isolates in addition to PHA. Cells of the isolate Acinetobacter sp. 211 were able to synthesize and accumulate lipid inclusions during growth on acetate, ethanol, olive oil, hexadecanol and heptadecane. The composition of the lipid inclusions depended on the compounds provided as carbon source. Wax esters and acylglycerols occurred mainly during the cultivation on olive oil; in contrast, wax esters and free alcohols occurred during cultivation on hexadecanol. Total fatty acids in cells of the Acinetobacter sp. 211 amounted to 25% of the cellular dry weight in olive-oil-grown cells. Palmitic acid was the main fatty acid in the lipids when the cells were cultivated on acetate or ethanol (44% and 32% of total fatty acids respectively). In contrast, fatty acids occurring in the lipids during cultivation on hexadecanol, heptadecane or olive oil were related to the carbon source. The fatty acids present in the accumulated lipids consisted predominantly of saturated and unsaturated straight-chain fatty acids with a chain length ranging from 12 to 18 carbon atoms. Analysis of the lipid-granule-associated proteins in cells of Acinetobacter sp. 211 revealed a protein of 39 kDa as the predominant protein species. Three marine phytoplankters (Isochrysis galbana, Chaetoceros calcitrans andThalassiosira pseudonana), commonly used in the culture of bivalve larvae, were grown in batch or semi-continuous cultures. Changes in protein, carbohydrate, lipid and some fatty acids were measured as growth became limited by nitrogen, silicon, phosphorus or light. Under N starvation (2 d) the % lipid remained relatively constant, while% carbohydrate increased and% protein decreased in all 3 species compared to cells growing under no nutrient limitation. Under Si starvation (6 h) there was no change in lipid, protein or carbohydrates. The amount of two fatty acids, 20 : 53 and 22 : 63 remained relatively constant under N, P and Si starvation, exept for a sharp drop in the cells of P-starvedT. pseudonana. However, there were pronounced species differences withI. galbana containing significantly less 20 : 5 3 thanC. calcitrans orT. pseudonana. Under light limitation the amount of lipid per cell showed no consistent trend over a range of irradiances for all 3 species. The amount of N per cell (an index of protein content) as a function of irradiance, was relatively constant forI. galbana andT. pseudonana, while the amount of N per cell was lower under low irradiances forC. calcitrans. These examples of changes in protein, carbohydrate, lipid and certain fatty acids under nutrient (N, Si or P) or light limitation, emphasize the importance of knowing the phase (e.g. logarithmic vs stationary) of the growth curve in batch cultures, since the nutritional value of the phytoplankters could change as cultures become dense and growth is terminated due to nutrient or light limitation.Presented at the XIIIth International Seaweed Symposium, University of British Columbia, Vancouver, Canada, August 1989.  相似文献

Aloe greatheadii var. davyana is the most important indigenous South African bee plant. Fresh, bee-collected and stored pollen of this aloe was collected and analysed for its nutritional content, including amino acid and fatty acid composition. Highly significant differences were found between the three types of pollen. Collection and storage by the bees resulted in increased water (13-21% wet weight) and carbohydrate content (35-61% dry weight), with a resultant decrease in crude protein (51-28% dry weight) and lipid content (10-8% dry weight). Essential amino acids were present in equal or higher amounts than the required minimum levels for honeybee development, with the exception of tryptophan. Fatty acids comprised a higher proportion of total lipid in fresh pollen than in bee-collected and stored pollen. This study is the first to compare the changes that occur in pollen of a single species after collection by honeybees.  相似文献

Three species of microalgae commonly used in mariculture —Isochrysis sp. (clone T.ISO) Parke,Pavlova lutheri (Droop) Green andNannochloropsis oculata (Droop) Green — were grown in batch and semicontinuous modes to compare their biochemical composition and production rates.In batch mode, logarithmic-phase cultures of all species had high levels of protein (25.2 to 41.1) and low levels of carbohydrate (7.1 to 10.3) and lipid (8.8 to 14.9). At stationary phase, cultures ofIsochrysis sp. (clone T.ISO) andN. oculata contained significantly less protein (21.8 and 20.3, respectively), all species contained more carbohydrate (14.8 to 30.6), andP. lutheri contained more lipid (16.6). In semi-continuous mode, cultures maintained at late logarithmic-phase contained more carbohydrate,Isochrysis sp. (clone T.ISO) contained less protein, andP. lutheri more lipid than logarithmic-phase batch cultures of the same species. Neither growth phase nor harvest regime affected the amino acid composition of the microalgae significantly. However, the concentration of proline inN. oculata was higher in batch cultures in logarithmic phase (9.4), than in either semi-continuous cultures in logarithmic phase (5.8 to 7.9) or batch cultures in stationary phase (5.6 to 5.9).The production rates from batch and semi-continuous logarithmic-phase cultures were not significantly different for any of the species, and there were only minor differences in the production rates of the species (range 12.4 to 17.1 mg algae dry weight 1–1 d–1). The different culture and harvest regimes produced significant differences in the proportions of protein and carbohydrate in the microalgae. In the present study, different water samples from Red Sea coastal area at Rabigh city, Saudi Arabia were studied for their dominant algal species. Microalgal isolation was carried out based on dilution method and morphologically examined using F/2 as a growth medium. Dry weight and main biochemical composition (protein, carbohydrates, lipids) of all species were performed at the end of the growth, and biodiesel characteristics were estimated. Nannochloropsis sp., Dunaliella sp., Tetraselmis sp., Prorocentrum sp., Chlorella sp., Nitzschia sp., Coscinodiscus sp., and Navicula sp. were the most dominant species in the collected water samples and were used for further evaluation. Nannochloropsis sp. surpassed all other isolates in concern of biomass production with the maximum recorded dry weight of 0.89 g L?1, followed by Dunaliella sp. (0.69 g L?1). The highest crude protein content was observed in Nitzschia sp. (38.21%) and Dunaliella sp. (18.01%), while Nannochloropsis sp. showed 13.38%, with the lowest recorded lipid content in Coscinodiscus sp. (10.09%). Based on the growth, lipid content, and biodiesel characteristics, the present study suggested Dunaliella sp. and Nitzschia sp. as promising candidates for further large-scale biodiesel production.  相似文献

Protein, lipid, carbohydrate, chitin, ash, and caloric energy contents of the Arctic marine benthic amphipod Onisimus (= Boeckosimus) affinis H.J. Hansen were determined at intervals over two and one half years. The lipid content exhibited the most pronounced seasonal change, from a minimum of about 14–17% of dry wt to a maximum of 25–27% of dry wt. An increase in lipid content in the spring coincided with the reproductive period of the species. Lipid and caloric content decreased during the summer to levels that were maintained through the winter, indicating that the animals do not store significant nutritional reserves for use during the winter. The dry weight proportions of other components were: protein 27.7–38.3%; carbohydrate 1.8–4.0%; chitin 7.8–8.2%; ash 21.3–27.2%. Caloric content varied between 3.29 and 4.18 cal/mg dry wt.  相似文献

Diatom cell quantity and their biochemical composition vary among species and are greatly affected by harvest stage or culture conditions. This study compares growth pattern, cell attachment, and biochemical composition of four diatoms suitable for abalone post-larvae: Navicula incerta, Proschkinia sp., Nitzschia sp., and Amphora sp. The four diatoms were grown in F/2 medium at 28.5?±?1.4°C, under 62?±?8?μmol?photons?m?2?s?1, at different original inoculating densities (0.05?×?106, 0.10?×?106, and 0.25?×?106?cells?mL?1) and were harvested in log and stationary phase of growth for biochemical analysis. Total protein, carbohydrate, lipid, and ash composition, as well as fatty acid composition, were determined. All diatoms grew better when inoculated at 0.10?×?106?cell?mL?1 with Proschkinia sp. reaching the highest cell density of 6.56?×?106?cells?mL?1 in log phase. Amphora sp. had the highest cell attachment capacity when inoculated at 0.10?×?106?cell?mL?1 (11,580?cells?mm?2), whereas N. incerta had the lowest (7,750?cells?mm?2). Protein and lipid (percent dry weight) contents were generally highest in cells during log phase of growth; Amphora sp. in log phase of growth had the highest lipid content of 9.74% DW, whereas significant differences in carbohydrate between the two growth phases were only observed for Proschkinia sp. Besides, all diatoms had higher energy contents in log phase of growth. There were no significant differences in ash content among the four diatoms. Polyunsaturated fatty acid (PUFA) content ranged from 23.25% to 38.62% of the total fatty acids, and the four diatoms tested were richer in n-3 PUFA than in n-6 PUFA. All the diatoms had significant quantities of 20:5n-3 (EPA) (between 12.69% and 17.68% of TFA), and Proschkinia sp., in log phase of growth, had the highest quantity of arachidonic acid (20:4n-6; ARA). The results highlight the influence of culture conditions and harvest protocols on diatom nutritive value and enabled a preliminary approach towards the selection of novel diatom species.  相似文献

The macromolecular composition of macroalgae influences nutrient flow and food quality in aquatic ecosystems and the value of macroalgae species for human consumption, aquaculture, biofuels, and other applications. We used literature data (125 publications, 1,117 observations) and a hierarchal Bayesian statistical model to estimate the average macromolecular composition, protein, lipid, and carbohydrate of macroalgae as a whole and at the phylum level. Our focus was on marine, noncalcified macroalgae sampled from wild‐grown populations in the field. We found that the median macromolecular composition is 9.98% protein, 2.7% lipid, 48.5% carbohydrate, and 31.8% ash as percent dry weight. We compared the median macromolecular content of macroalgae to microalgae and herbaceous plants and test for differences in macromolecular content across macroalgal phyla. Macroalgae were much more enriched in carbohydrate and minerals than the microalgae and lower in protein and lipid than many herbaceous plants. Rhodophyte macroalgae have significantly less lipid and more protein and the Ochrophyte macroalgae have significantly less protein than the average.  相似文献

Interaction between arbuscular mycorrhizal fungus Glomus mosseae and plant growth promoting fungus Phoma sp. was studied for its effect on their root colonization and plant growth of cucumber. Two isolates of Phoma sp. (GS8-2 and GS8-3) were tested with G. mosseae. The percent root length colonized by G. mosseae was not adversely affected by the presence of Phoma isolates. In contrast, the root colonization of both isolates GS8-2 and GS8-3 in 4-week-old plants was significantly reduced (80.7% and 84.3%, respectively) by added G. mosseae. Inoculating plants with each Phoma isolate significantly increased the shoot dry weight. However, dual inoculation of each Phoma isolate with G. mosseae had no significant effect on growth enhancement.  相似文献

The variability in cultural characteristics and the virulence among three isolates of Fusarium oxysporum f. sp. ciceri causing vascular wilt in chickpea was studied under laboratory conditions. The three isolates (Foc-1, Foc-2 and Foc-3) did not show any significant difference in their mycelial dry weight production at any temperature regimes, pH level or the growth media tested. The radial growth on PDA also did not differ significantly in the three isolates. However, some quantitative differences were noted in colony characters and septations in macroconidia of these isolates. The isolate Foc-1 exhibited dull white, thin and flat hairy growth, spreading out like thread, Foc-2 showed a white fluffy colony with irregular aerial margin, while Foc-3 exhibited a pinkish white, slightly fluffy colony with regular margin. Conidia also differed with regard to septation. Three to six septa were present in Foc-2, while there were 2–3 in isolates Foc-1 and Foc-2. These isolates differed significantly with regard to their virulence on test varieties. Isolate Foc-1 was more virulent that Foc-2 or Foc-3 and produced abundant spores.  相似文献

Apiotrichum curvatum ATCC 20509, an oleaginous yeast that can accumulate up to 60% of its cellular dry weight as intracellular lipid when grown with excess carbon, was grown in nitrogen-limited, balanced, and lactose-free medium with asparagine as nitrogen source and lactose as carbon source. Biomass and lipid accumulation were measured, cell composition was analyzed, and catalase activity was followed as marker enzyme for peroxisomes. The organism accumulated 54% of its dry weight as total cellular lipid when grown under nitrogen limitation and accumulated only 20-25% of its dry weight as lipid when grown in balanced medium. When starved for carbon, cells utilized endogenous lipid and carbohydrate as carbon and energy sources; the intracellular contents of lipid and carbohydrate decreased by 31 and 26%, respectively. Intracellular carbohydrates also seemed to be used as intermediates for lipid accumulation and lipid turnover. Catalase activity was strongly induced (over 10-fold increase in specific activity) when cells metabolized endogenous lipid. The lipid content of cells was inversely related to catalase activity and to intracellular protein or total nitrogen content. Lipid content showed no correlation with intracellular carbohydrate content.  相似文献
