Summary It is well known that truncation selection is the most efficient form of directional selection in terms of changing gene frequency. In this paper we show circumstances where truncation selection followed by a balanced mating generates inbreeding effective population size smaller than that generated by a selection that assigns mating frequencies to individuals according to their breeding values, where both selection schemes give the same expected performance of selected individuals (selection differential). Breeding values of selected individuals and the weight used to determine mating frequencies are assumed to be linearly distributed on a performance scales, x. To assign mating frequencies to the individuals in the weighting system, the selected individuals are grouped using a constant , and ith group in the interval xi, xi + . With small number of groups, say 2 or 3, the weighting system in general generates inbreeding effective population size that is larger than that generated by a truncation selection. As the number of the groups increases, truncation selection generates larger effective numbers.  相似文献

A combination of stable isotope studies and 14Cdating were used to identify the main sources andprocesses controlling streamwater DOC and TIC in atemperate non-forested watershed. 13Cvalues for terrestrial (–24.9 to –29.1) and aquatic(–30.5 to –33.5) plants were similar to valuesreported in the literature for similar ecosystems.13C values for DOC in soil solution andstreamwater were consistent with soil and terrestrialvegetation, indicating that the terrestrial ecosystemis the dominant source of aquatic DOC in thiswatershed. 13C values of soil atmosphereCO2 (–17.2 to –25.2) were slightly lessnegative than would be expected for production viaaerobic soil microbial decomposition and rootrespiration. There was a close correspondence between13C values (–15.5 to –21.5) forstreamwater TIC and soil atmospheric CO2 in thecentral part of the catchment where the stream drainsCO2-rich peats. 14C dating showed thatalthough peat has been accumulating in the watershedfor at least 2700 years, DOC in soil pore water andstreamwater contains carbon of predominantly recentorigin (post-AD 1955).  相似文献

Summary The (14C)2DG autoradiographic technique has been employed to quantitatively map glucose utilization in the mesencephalon, the diencephalon and the cerebellum, of toads in response to configurational moving visual stimuli: (i) a 0.4 cm × 2.8 cm worm-like stripe (W) which elicited prey catching responses, (ii) a 8.4 cm × 8.4 cm square (S) that released predator avoidance responses, and (iii) a 2.8 cm × 0.4 cm antiworm-like stripe (A) which elicited no motor activity.For various brain nuclei different relationships were obtained: The optic tectum showed statistical significant higher 2DG uptake during worm-stimulation (¯X W) than during antiworm stimulation (¯X A), i.e.¯X W>¯X A. The latter visual pattern led to a 2DG utilization that was statistically significant stronger than during stimulation with a square (¯X S), i.e.¯X A>¯X S. There have been claims that the minor hemoglobin A2 in some primates is functionally and adaptively less vital than hemoglobin A (Boyer et al., 1971). These claims are criticized on several grounds, and evidence suggesting that selection acts on hemoglobin A2 is reviewed. An estimate is made of the proportion of mutations in the , , , and genes that could be expected to be selectively neutral. Evidence is presented that the gene is better buffered from the effects of mutation than is the gene and that is better buffered than , although quantitatively the latter difference is very small.  相似文献

Liver homogenate-supernatants from most Japanese exhibit an atypical pH optimum for ethanol oxidation at pH 8.8 instead of 10.5, the typical pH-activity optimum. It has been proposed that atypical livers contain alcohol dehydrogenase isozymes with 2 subunits while typical livers contain isozymes with 1 subunits, both produced by the ADH 2 gene. Because it is difficult to differentiate the atypical ADH2 2-2 phenotype from the ADH2 2-1 phenotype by starch gel electrophoresis, an agarose isoelectric focusing procedure was developed that clearly separated the atypical Japanese livers into two groups, A1 and A2. The isozymes in A1 and A2 livers were purified. Type A1 livers contained a single isozyme with an atypical pH-rate profile; it was designated 22. Three isozymes were isolated from A2 livers, two of which corresponded to 11 and 22. A third, absent from the typical and the atypical A1 livers, had an intermediate mobility; it was designated 21. Type A1 livers are, therefore, the homozygous ADH2 2-2 phenotype, and type A2 livers, the heterozygous ADH2 2-1 phenotype. The ADH2 2-2 phenotype was found in 53% of 194 Japanese livers, and the ADH2 2-1 phenotype, in 31%. Summary The coefficient of coancestry (fAB) between individuals A and B is the classical measure of genetic relationship. fAB is determined from pedigree records and is the probability that random alleles at the same locus in A and B are copies of the same ancestral allele or identical by descent (ibd). Recently, the proportion of molecular marker variants shared between A and B (SAB) has been used to measure genetic relationship. But SAB is an upwardly-biased estimator of fAB, especially between distantly-related lines. fAB, SAB, and adjusted (to remove bias) estimates of molecular marker similarity (f AB M ) were compared. RFLP banding patterns at 46 probe-restriction enzyme combinations were obtained for 23 maize inbred lines derived from the Iowa Stiff Stalk Synthetic (BSSS) maize (Zea mays L.) population, and for 4 non-BSSS lines. f AB M was estimated as , where A (or B) was the average proportion of RFLP variants shared between inbred A (or inbred B) and the non-BSSS lines. The average fAB among 253 pairwise combinations of BSSS lines was 0.212, whereas the average SAB was 0.397. The average f AB M was 0.162, indicating that the upward bias in SAB was effectively removed. SAB and fAB were significantly different ( = 0.05) in 76.3% of the comparisons, whereas 24.9% of the f AB M values differed significantly from fAB. The latter result suggests that selection and/or drift were present during inbred line development and that fAB may not be an accurate measure of the true proportion of ibd alleles between two lines. Cluster analyses based on S AB M and f AB M grouped lines according to pedigree, although several exceptions were noted. The presence of shared molecular marker variants between unrelated lines must be considered when setting SAB-based minimum distances for varietal protection. Summary Examination of glucagon structure-activity relationships and their use for the development of glucagon antagonists (inhibitors) have been hampered until recently by the lack of high purity of semisynthetic glucagon analogs and inadequate study of full dose-response curves for these analogs in sensitive bioassay systems. Recently a number of highly purified glucagon fragments and semi-synthetic analogs have been prepared and their full dose-response activities examined over a wide concentration range using the hepatic membrane adenylate cyclase assay, the hepatic membrane receptor binding assay, and glycogenolytic activity in isolated rat hepatocytes. The results of these studies have enabled us to identify and dissociate the structural (and in some cases conformational) features of glucagon important for binding from those most responsible for biological activity (transduction). Key findings in these studies were the observation that: (1) the C-terminal region of glucagon is primarily of importance for hormone binding to receptors; (2) glucagon1–21 and glucagon1–6 have low potency, but are essentially fully active glucagon derivatives; and (3) highly purified glucagon2–29 ([1-des-histidine]-glucagon), [1-N-carbamoylhistidine]-glucagon and [1-N-carbamoylhistidine, 12-N-carbamoyllysine]-glucagon are all partial agonists.These and other findings led us to synthesize several semisynthetic analogs of glucagon which were found to possess no intrinsic biological activity in the hepatic adenylate cyclase assay system, but which could block the effect of glucagon (competitive inhibitors) in activating adenylate cyclase in this system. Two of these highly purified analogs [1-des-histidine] [2-N-trinitrophenylserine, 12-homoarginine]-glucagon and [1-N-trinitrophenylhistidine, 12-homoarginine]-glucagon were quite potent glucagon antagonists (inhibitors) with pA2 values of 7.41 and 8.16 respectively. The latter compound has also been demonstrated to decrease dramatically blood glucose levels of diabetic animals in vivo. These results demonstrate that glucagon is a major contributor to the hyperglycemia of diabetic animals.Examination of the known and calculated conformational properties of glucagon provide insight into the structural and conformational properties of glucagon and its analogs most responsible for its biological activity. Consideration of these features and the mechanism of glucagon action at the membrane receptor level provide a framework for further developing glucagon analogs for theoretical and therapeutic applications.  相似文献

The pattern of synonymous and nonsynonymous substitutions at the reactive center of proteases (kallikrein) and their inhibitors (1-antitrypsin and serpin) was examined. In the case of 1-antitrypsin, the proportion of different nonsynonymous sites exceeds that of different synonymous sites at the reactive center for sequence pairs of recent duplication. The result indicates that the positive selection has operated after duplication to increase functional diversity. In the cases of kallikrein, serpin, and remote sequence pairs of 1-antitrypsin, the proportion of different synonymous sites at the reactive center exceeds that of different synonymous sites at the remaining region. The result indicates that gene conversion followed by natural selection is working. On the whole, it is concluded that hypervariability of amino acids at the reactive center is generated by an interaction among natural selection, random genetic drift, point mutation, and gene conversion. Gene duplication may provide potential for them to interact.  相似文献

Three layer control structure is proposed for optimal control of continuous fermentation processes. The start-up optimization problems are solved as a first step for optimization layer building. A steady state optimization problem is solved by a decomposition method using prediction principle. A discrete minimum time optimal control problem with state delay is formulated and a decomposition method, based on an augmented Lagrange's function is proposed to solve it. The problem is decomposed in time domain by a new coordinating vector. Summary The maize (Zea mays L.) improvement program of the International Maize and Wheat Improvement Center (CIMMYT) develops broad-based maize populations and, until recently, improved all of them through full-sib family selection with international testing. The purpose of this study was to estimate the genetic and genetic × environment variance components for ten of those populations and to measure expected yield improvement from full-sib selection. Mean yield ranged from 3.35–6.81 t ha–1. For five populations the average yield in the last cycle was higher than in the initial cycles. Several populations showed no improvement or yielded less in the final cycle of selection, either because selection intensity was low or because strong selection pressure was applied simultaneously for several traits. Variation resulting from differences among family means within cycles and from interaction between families and locations within cycles were significant in all populations and cycles. Results indicate that variability among full-sib families was maintained throughout the cycles for all populations. The large ge 2 / g 2 ratio shown by most populations suggests that yield response per cycle could be maximized if the environments in which progenies are tested were subdivided and classified into similar subsets. Diurnal cycle of carbon isotope ratio in soil CO2 in various ecosystems   总被引:1,自引:1,他引:0  
Our investigations of diurnal variations of the 13C/12C ratio and CO2 content in soil air were carried out in three environments during periods of high biosphere activity. It has been observed that diurnal variation of CO2 concentration is negatively correlated 13. Particularly great variations occurred at shallow soil depths (10–30 cm) when the plant cover activity was high while the soil temperature was rather low. Under such conditions the 13 variations had the magnitude of 4, while the CO2 concentration varied more than doubly. The maximum of the 13C/12C ratlo and the minimum of the CO2 concentration in a cultivated field with winter wheat took place in the afternoon, whereas in deciduous forest similar patterns were observed at dawn. In these cases soil temperatures at 10 cm depths varied less than 2°C. Hence, under wheat the variation in root respiration rate seem to be the main reason of the recorded varations. In an uncultivated grass-field during the hottest period in summer we did not measure any distinct variations of CO2 properties in spite of the fact that soil temperature varied up to 5°C. This might be due to dominant microbial respiration at the high soil temperature, which exceeded 20°C.  相似文献

The effects of CO2 concentration and the effects of growth-light conditions on Ribulose 1,5-bisphosphate carboxylase/oxygenase (Rubisco) deactivation were examined for Spinacea oleracea (spinach). Rubisco deactivation kinetics and the degree that Rubisco activation limited the rise in photosynthesis following an increase in photon flux density (PFD) were determined from gas-exchange time courses. There were no significant differences in the apparent relaxation time for Rubisco deactivation among leaves exposed to high or low CO2 (50 or 1000 mol mol-1) and low PFD (170 mol m-2 s-1) or darkness. However, when PFD was increased to 1700 mol m-2 s-1 following a period of low PFD or darkness, leaves exposed to low CO2 × low PFD showed a lower contribution to the photosynthetic induction process by the activation of Rubisco than leaves exposed to the other treatments. For the growth-light experiments, spinach was grown under high PFD × high red:far-red ratio (R:FR), low PFD × high R:FR, or low PFD × low R:FR light environments. Leaves that matured under the low PFD × low R:FR treatment showed a lower percent change in photosynthesis due to Rubisco activation than leaves exposed to the other growth-light treatments. However, there were no significant differences among the growth-light treatments in the maximum contribution of Rubisco activation to the induction response or in the apparent relaxation time for Rubisco deactivation during shade events.  相似文献

The phenotype variability and inheritance of reproductive traits were investigated in the medicinal leech. Distribution parameters were determined for the following traits: batch size (X¯ = 4.3 ± 0.2, = 1.7, CV = 40%, As = 0.23 ± 0.25, Ex = 0.19 ± 0.51), number of juveniles in a cocoon ( X¯= 10.9 ± 0.3, = 4.6, CV = 42%, As = 0.31 ± 0.15, Ex = 0.23 ± 0.30), and juvenile weight ( X¯= 32.0 ± 0.3, = 14.9, CV = 47%, As = 1.38 ± 0.05, Ex = 3.32 ± 0.11). A nonlinear negative correlation between the number of juveniles in a cocoon and their weight was found (correlation ratio R= 0.86). It was shown that the environmental variance dominated over the genotypic one in the structure of phenotypic variance of the traits studied. The genetic variability is determined mainly by additive gene interactions and, to a small extent, intralocus dominance. The narrow-sense heritability, h 2, for batch size was 0.35–0.40; for the number of juveniles in a cocoon, 0.35; for juvenile weight, 0.42.  相似文献

Summary The angular dependence of1JC,H in model compounds related to -linked oligosaccharides has been established by FPT INDO quantum chemical calculations. Values calculated for models of (1 1)-, (1 2)-, (1 3)- and (1 4)-linked disaccharides were compared, and the effect of the orientation of HO-2 elucidated. The angular dependence of1JC,H on the torsional angles H and H and the solvent dielectric constant (s) was characterized in the form:1JC,H = A cos2+B cos + C sin2 + D since + E + Fe. The1JC,H values, measured by DEPT methods for C-1-H-1 and C-X-H-X in cellobiose, cyclic trisaccharide and hexopyranoses were used to adjust the calculated angular dependences. Based on the occurrence of the conformers for agarobiose, neoagarobiose, mannobiose and methyl -xylobioside, the thermodynamically averaged <1JC,H > values were calculated. The results obtained (<1JC-1,H-1 > 162.4, <1JC-4, H-4 > 147.6 Hz for methyl -xylobioside; <1JC-1,H-1 > 162.4 and <1JC-4,H-4] > 147.6 Hz for mannobiose; <1JC-1,H-1 > 162.8 Hz for neo agarobiose and <1JC-1,H-1 > 163.2 Hz for agarobiose) agree well with the experimental values of 162.7, 147.5, 160.4, 147.2, 160.9 and 165.7 Hz, respectively.  相似文献

Tobacco (Nicotiana tabacum L.) plants transformed with antisense rbcS to decrease the expression of ribulose-1,5-bisphosphate carboxylase-oxygenase (Rubisco) have been used to investigate the contribution of Rubisco to the control of photosynthesis in plants growing at different irradiances. Tobacco plants were grown in controlled-climate chambers under ambient CO2 at 20°C at 100, 300 and 750 mol·m–2·s–1 irradiance, and at 28°C at 100, 300 and 1000 mol·m–2·s–1 irradiance. (i) Measurement of photosynthesis under ambient conditions showed that the flux control coefficient of Rubisco (C infRubisco supA ) was very low (0.01–0.03) at low growth irradiance, and still fairly low (0.24–0.27) at higher irradiance. (ii) Short-term changes in the irradiance used to measure photosynthesis showed that C infRubisco supA increases as incident irradiance rises, (iii) When low-light (100 mol·m–2·s–1)-grown plants are exposed to high (750–1000 mol·m–2·s–1) irradiance, Rubisco is almost totally limiting for photosynthesis in wild types. However, when high-light-grown leaves (750–1000 mol·m–2·s–1) are suddenly exposed to high and saturating irradiance (1500–2000 mol·m–2·s–1), C infRubisco supA remained relatively low (0.23–0.33), showing that in saturating light Rubisco only exerts partial control over the light-saturated rate of photosynthesis in sun leaves; apparently additional factors are co-limiting photosynthetic performance, (iv) Growth of plants at high irradiance led to a small decrease in the percentage of total protein found in the insoluble (thylakoid fraction), and a decrease of chlorophyll, relative to protein or structural leaf dry weight. As a consequence of this change, high-irradiance-grown leaves illuminated at growth irradiance avoided an inbalance between the light reactions and Rubisco; this was shown by the low value of C infRubisco supA (see above) and by measurements showing that non-photochemical quenching was low, photochemical quenching high, and NADP-malate dehydrogenase activation was low at the growth irradiance. In contrast, when a leaf adapted to low irradiance was illuminated at a higher irradiance, Rubisco exerted more control, non-photochemical quenching was higher, photochemical quenching was lower, and NADP-malate dehydrogenase activation was higher than in a leaf which had grown at that irradiance. We conclude that changes in leaf composition allow the leaf to avoid a one-sided limitation by Rubisco and, hence, overexcitation and overreduction of the thylakoids in high-irradiance growth conditions, (v) Antisense plants with less Rubisco contained a higher content of insoluble (thylakoid) protein and chlorophyll, compared to total protein or structural leaf dry weight. They also showed a higher rate of photosynthesis than the wild type, when measured at an irradiance below that at which the plant had grown. Summary The fluorescent intercalation complex of ethidium bromide (ETB) with DNA was used as a probe to compare the effects of various radicals with respect to impairment of the DNA base-pair region.OH radicals inhibit up to 0.7 dye intercalations perOH at low salt concentration, and for various oxidizing species the effect decreases in the orderOH > Br 2 > N 3 > $$ " align="middle" border="0"> (SCN) 2 . DNA impairment by theOH product of Met-Gly is comparable to that of N 3 , but no effect was found due to the interaction between DNA and Lys-Tyr-Lys phenoxyl radicals. Kluyveromyces lactis -galactosidase gene, LAC4, was expressed in Escherichia coli as a soluble His-tagged recombinant enzyme under the optimized culture conditions. The expressed protein was multimeric with a subunit molecular mass of 118 kDa. The dimeric form of the -galactosidase was the major fraction but had a lower activity than those of the multimeric forms. The purified enzyme required Mn2+ for activity and was inactivated irreversibly by imidazole above 50 mM. The activity was optimal at 37 and 40 °C for o-nitrophenyl--d-galactopyranoside (oNPG) and lactose, respectively. The optimum pH value is 7. The K m and V max values of the purified enzyme for oNPG were 1.5 mM and 560 mol min–1 mg–1, and for lactose 20 mM and 570 mol min–1 mg–1, respectively.  相似文献

The properties of three amidohydrolases, i.e., urease (I) EC 3.5.1.5, L-asparaginase (II) EC 3.5.1.1, and L-glutaminase (III) EC 3.5.1.2, were studied in sediment samples taken from a shallow eutrophic freshwater lake.Sediment samples were air dried (ADS) and stored for at least 3 months before being enzymically characterized. The pH optimum of I, II, and III were pH 7.0, 8.4, and 6.5–7.0, respectively, while III in soluble extracts from ADS was most active between pH 8.0 and 9.0. The temperature response of the three enzymes in ADS gave Ea values of 38.9, 41.6, and 35.9 kJmol–1 for I, II, and III, respectively. Km and Vmax values for ADS I, II, and III were 1.2 mM and 1.9mol NH3 g–1h–1; 0.8 mM and 4.1mol NH3 g–1h–1; and 1.25 mM and 17.4mol NH3 g–1h–1. Km values for all three enzymes in ADS extracts were at least an order of magnitude greater than those of the ADS. The susceptability of each enzyme to proteolysis was followed in ADS and fresh wet sediment and compared with that of III in an ADS extract. All sediment enzymes were found to be more resistant than the commercial preparation of bacterial L-glutaminase subjected to the same treatment. These results suggested that I, II, and III all exist to some extent as colloid-immobilized enzyme fractions in freshwater sediments and are analogous to the stable enzyme fractions in soils.  相似文献

The objective of this study was to discern the source of higher than usual concentrations of chloride in drainage water collected from experimental forest plots after clear-cutting. When the sandbox experiments were initiated at the Hubbard Brook Experimental Forest Station three vegetation types were established: red pine, grass, and minimally vegetated (scattered lichens and bryophytes) as the bare control plot. After 15 years of growth the trees were cut down and above-ground biomass removed from the red pine sandbox. For several years prior to the cut, high concentrations ( 75 M) of dissolved Cl in drainage waters occurred in November/December. This is attributed to the buildup of rainfall-derived Cl due to evapotranspiration that depletes soil moisture to low levels resulting in a lack of drainage during this period. The excess Cl is quickly flushed out by subsequent drainage over a few weeks and Cl concentrations return to values characteristic of rainfall and throughfall. After the trees were removed in May, 1998, Cl continued to be leached from the system. The concentration of Cl peaked (175 M) in Sept. 1998 and did not return back to base level concentration until Dec. 1999. The Cl release pattern is distinctly different from that of dissolved NO3 , which peaked about one year later than Cl. An excess (over that of the control sandbox) of 78 g Cl was released in the 1.5 year period after clear-cut, showing that a large amount of leachable chloride is stored in the bulk soil/root/organic matter fraction. Lack of uptake by trees may be part of the reason for this chloride pulse. But an analysis of chloride content in roots and litter indicates that as much as 50% of the chloride leached from the sandbox may have come from the decaying roots and litter. Additional chloride may have been released from the soil organic matter by decomposition. The biochemical behavior of Cl in systems such as this should be evaluated before assuming Cl to be conservative for purposes of hydrological transport or soil weathering studies.  相似文献

Direct evidence for the excitotoxicity of -N-oxalyl-L-,-diaminopropionic acid (ODAP), the Lathyrus sativus neurotoxin has been studied by examining the binding of chemically synthesized [2,3 3H]ODAP ([3H]ODAP) to synaptic membranes. [3H]ODAP binding to membranes was mostly nonspecific, with only a very low specific binding (15–20% of the total binding) and was also not saturable. The low specific binding of [3H]ODAP remained unaltered under a variety of assay conditions. A low Bmax of 3.2 ± 0.4 pmol/mg and Kd 0.2 ± 0.08 M could be discerned for the high affinity interactions under conditions wherein more than 80–90% of the binding was nonspecific. While ODAP could inhibit the binding of [3H]glutamate to chick synaptic membranes with a Ki of 10 ± 0.9 M, even L-DAP, a non neurotoxic amino acid was also equally effective in inhibiting the binding of [3H]glutamate. The very low specific binding of [3H]ODAP to synaptic membranes thus does not warrant considering its interactions at glutamate receptors as a significant event. The results thus suggest that the reported in vitro excitotoxic potential of ODAP may not reflect its true mechanism of neurotoxicity.  相似文献
