A data set concerning 1,816 subjects entered in the Italian Horse Registry from 1925 to 2002 was analyzed to investigate the morphological evolution of the Murgese horse and to obtain useful elements to enhance breeding practices. Three basic body measurements (height at withers, chest girth, and cannon bone circumference) were considered for each subject. Heritabilities were calculated for each parameter to infer the growth and development traits of this breed. Over the past 20 years the Murgese horse has undergone considerable changes, passing from a typical mesomorphic structure (height at withers: 156.30 and 151.04 cm; chest girth: 185.80 and 176.11 cm; cannon bone: 21.10 and 19.82 cm for males and females, respectively) to a mesodolichomorphic structure (height at withers: 160.31 and 156.44 cm; chest girth: 187.89 and 182.48 cm; cannon bone: 21.07 and 20.37 cm, for males and females, respectively). Due to these changes and to its characteristic strength and power, the Murgese, which was once used in agriculture and for meat production (at the end of its life), is now involved in sports, mainly in trekking and equestrian tourism. PCR–RFLP was applied to analyse polymorphisms within the MRF4 and heart fatty acid-binding protein (H-FABP) gene for correlation studies with growth traits in three-month-old Qinchuan (QQ), Qinchuan × Limousin (LQ) and Qinchuan × Red Angus (AQ) cattle. The results showed that 874 bp PCR products of MRF4 digested with XbaI and 2,075 bp PCR products of H-FABP digested with HaeIII were polymorphic in the three populations. Moreover, the frequencies of allele A at MRF4 locus and allele B at H-FABP locus in the QQ, AQ, and LQ populations were 0.8358/0.8888/0.8273 and 0.8358/0.7500/0.8195 respectively. Allele A at MRF4 locus and allele B at H-FABP locus were dominant in the three populations. No statistically significant differences in growth traits were observed among the genotypes of the all three populations at H-FABP locus. However, the association of MRF4 polymorphism with growth traits was then determined in all three populations. The body weight, withers height, heart girth and height at hip cross of individuals with genotype AA were higher than those with genotype AB or BB (P < 0.05). Therefore, we suggest that the MRF4 gene may function in the control or expression of growth traits, particularly body weight, withers height, heart girth and height at hip cross.  相似文献

Insulin-like growth factor 2 (IGF2) is a potent cell growth and differentiation factor and is implicated in mammals’ growth and development. The objective of this study was to evaluate the effects of the mutations in the bovine IGF2 with growth traits in Chinese Qinchuan cattle. Four single nucleotide polymorphisms (SNPs) were detected of the bovine IGF2 by DNA pool sequencing and forced polymerase chain reaction–restriction fragment length polymorphism (forced PCR–RFLP) methods. We also investigated haplotype structure and linkage disequilibrium (LD) coefficients for four SNPs in 817 individuals representing two main cattle breeds from China. The result of haplotype analysis showed eight different haplotypes and 27 combined genotypes within the study population. The statistical analyses indicated that the four SNPs, combined genotypes and haplotypes are associated with the withers height, body length, chest breadth, chest depth and body weight in Qinchuan cattle population (P < 0.05 or <0.01). The mutant-type variants and mutant haplotype (Hap 8: ATGG; likely to be the beneficial QTN allele) was superior for growth traits; the heterozygote diplotype was associated with higher growth traits compared to wild-type homozygote. Our results provide evidence that polymorphisms in the IGF2 gene are associated with growth traits, and may be used for marker-assisted selection in beef cattle breeding program.  相似文献

In the present study the relationships between five body structural and nine udder morphological traits with milk yield and non-yield traits, i.e. milk contents, somatic cell count and body weight in n = 171 Frizarta ewes were investigated. Positive correlations of moderate to high magnitude were observed among body measurements ranging from 0.25 (body length-chest width) to 0.61 (chest width-chest girth). High correlations were observed between udder width and udder circumference (r = 0.69) as well as between udder circumference at base and middle (r = 0.62). Udder height was found to be positively correlated with cistern depth (r = 0.32), udder width (r = 0.40) and udder circumference (r = 0.42) as well as with teat measurements (0.19 and 0.25 with teat length and teat circumference, respectively). Cistern depth was positively and highly correlated with teat angle (r = 0.64). The correlation between teat length and circumference was also high (r = 0.57). Teat angle was found to be negatively correlated with teat measurements (r = −0.39 and −0.23 for teat length and teat circumference, respectively). Principal components analysis revealed four components that jointly explained 67% of the variance of all the body and the udder morphological traits. The first principal component interpreted as the body dimensions was found to be associated with body weight of ewes. The second component interpreted as global and rear measurements of the udder was significant for milk yield traits. The third and fourth principal components were interpreted as the udder type and teat dimensions, respectively. Ewes’ body weight could be adequately predicted by chest girth, body length, chest width and withers height. Test day milk yields were found to be associated with udder circumference and udder width as well as with udder height and teat length. None of the udder measurements and/or teat characteristics was found to be correlated with milk content(s) and/or somatic cell count. New statistical techniques can be successfully implemented in investigation of relationships between various traits in dairy sheep.  相似文献

A multilocus GWAS was performed to explore the genetic architecture of four growth traits in yak. In total, 354 female yaks for which measurements of body weight (BW), withers height (WH), body length (BL) and chest girth (CG) at weaning were available underwent genotyping with the Illumina BovineHD BeadChip (770K). After quality control, we retained 98 688 SNPs and 354 animals for GWAS analysis. We identified seven, 18, seven and nine SNPs (corresponding to seven, 17, seven and eight candidate genes) associated with BW, WH, BL and CG at weaning respectively. Interestingly, most of these candidate genes were reported to be involved in growth-related processes such as muscle formation, lipid deposition, feed efficiency, carcass composition and development of the central and peripheral nervous system. Our results offer novel insight into the molecular architecture underpinning yak growth traits. Further functional analyses are thus warranted to explore the molecular mechanisms whereby these genes affect these traits of interest.  相似文献

Liver X receptor α (LXRα) has emerged as an important regulator of lipid and energy metabolism. In this study, to better understand the effects of LXRα gene on growth traits in cattle, the mRNA tissue expression patterns and the polymorphisms of some exons of LXRα were revealed. The expression profile of the bovine LXRα gene was detected by quantitative real-time polymerase chain reaction (qRT-PCR) in 11 different Jiaxian cattle tissues and was found mainly expressed in spleen, liver, fat tissue, kidney, muscle, and lung. Meanwhile, it showed that four single nucleotide polymorphisms (SNPs), named g.1028 T>C, g.1514 T>C, g.2929G>A, and g.3493 T>C, were detected and 12 different haplotypes were constructed. Haplotype with CCGT was dominant with frequency of 40.8 %. There was a strong link between g.1028 T>C and g.1514 T>C (r 2?=?0.374). Association analysis of SNPs with growth- and body-related traits was carried out in 445 Chinese native cattle. The results displayed that the heterozygous genotypes of g.1028 T>C and g.1514 T>C showed a molecular heterosis on four performance traits related to body size: height at withers, body length, hipbone width, and hip width (P?<?0.05). The multiple effects of four sites showed that the height at withers, body length, hipbone width, and hip width of individuals of TC-TC-GG-TT combined genotypes were significantly higher than other genotypes (P?<?0.05). The effects of the four loci genotype combination on conformation traits were consistent with the effects of g.1028 T>C and g.1514 T>C loci. The SNPs of g.1028 T>C and g.1514 T>C of the bovine LXRα gene could be potential genetic markers for growth traits in cattle. These results suggest that LXRα gene is expressed in many tissues and may provide primary molecular information for further studies on body size traits in Chinese indigenous cattle.  相似文献

Several studies have reported positive correlations between the degree of enzyme heterozygosity and fitness-related traits. Notable among these are the correlations between heterozygosity and growth rate in marine bivalves. Whether the correlation is the result of intrinsic functional differences between enzyme variants at the electrophoretic loci scored or arises from non-random genotypic associations between these loci and others segregating for deleterious recessive genes (the associative overdominance hypothesis) is a matter of continuing debate. A prediction of the associative overdominance hypothesis, not shared by explanations that treat the enzyme loci as causative agents of the correlation, is that the correlation is not specific to the type of genetic marker used. We have tested this prediction by scoring heterozygosity at single locus nuclear restriction fragment length polymorphisms (RFLPs) in a cohort of juvenile scallops (Placopecten magellanicus) in which growth rate was known to be positively correlated with an individual's degree of allozyme heterozygosity. A total of 222 individuals were scored for their genotypes at seven allozyme loci, two nonspecific protein loci of unknown function and eight nuclear RFLPs detected by anonymous cDNA probes. In contrast to the enzyme loci, no correlation was observed between growth rate and the degree of heterozygosity at the DNA markers. Furthermore, there was no relationship between the magnitude of heterozygote deficiency at a locus and its effect on the correlation. The differences observed between the effects of allozyme and RFLP heterozygosity on growth rate provide evidence against the associative overdominance hypothesis, but a strong case against this explanation must await corroboration from similar studies in different species.  相似文献

阿黑皮素原(Pro-opiomelanocortin, POMC)在动物采食和能量平衡调控中发挥重要作用, 文章对绵羊POMC基因外显子3进行扩增和测序, 筛选多态性位点, 并分析多态位点与湖羊和东弗里生×湖羊杂种羊生长性状的相关性。测序后发现湖羊POMC基因外显子3有2个单碱基突变(g.273 T/C和g.456 G/A), 根据273位点处发生的T/C突变, 建立PCR-RFLP分析方法, 并对162只湖羊和130只东湖杂种羊进行检测分析。结果发现, 在湖羊群体中检测到TT(0.469)、TC(0.438)和CC(0.093)3种基因型, 而在东湖杂种羊群体中仅检测到TT(0.754)和TC(0.246)两种基因型。POMC基因外显子3的273位点多态性与生长性状的相关性研究结果显示:湖羊群体中CC基因型个体的2月龄断奶重、4月龄尻高及TC基因型个体4月龄体长和管围均显著高于TT型个体(P<0.05); CC基因型个体的4月龄重、6月龄重极显著高于TT和TC基因型个体(P<0.01); CC基因型个体的4月龄体高和体长极显著高于TT型个体(P<0.01), 且显著高于TC基因型个体(P<0.05)。此外, CC型个体的管围极显著高于TT基因型个体(P<0.01)。东湖杂种羊群体中TC基因型个体的2月龄断奶重、4月龄重及4月龄体高、体长、胸深和管围都显著高于TT型个体(P<0.05), TC型个体的6月龄重极显著高于TT型个体(P<0.01)。研究结果表明, POMC基因外显子3与绵羊生长性状相关, C等位基因对体重及体尺性状的增加更有利。该结果为进一步探讨POMC基因作为绵羊生长性状的辅助选育标记奠定了基础。  相似文献

Body measurement traits, influenced by genes and environmental factors, play numerous important roles in the value assessment of productivity and economy. There has been some indication that ANAPC13 influences adult height. We used PCR-SSCP and DNA sequencing technology to identify polymorphisms in the ANAPC13 gene. A polymorphism in intron 1 (A > G at base 17) was identified and an additional polymorphic site (C > T at base 42) was also uncovered, which accompanied the previous polymorphism in more than 98% of the subjects. The two novel polymorphisms in exon 1 were assayed and potential associations with body measurement traits were evaluated in 404 individuals. Three genotypes were detected in the study group, named AACC, AGCT and GGTT. Significant differences were observed between genotypes AACC and AGCT for body length, withers height, hip height, hip width, heart girth, pin bone width. However, no associations were found among any genotypes and chest depth. We conclude that polymorphisms and mutations in non-coding regions of the ANAPC13 gene significantly affect body measurement traits.  相似文献

PCR-RFLP was applied to analyze the effect of the genetic variations of the POU1F1 gene on growth traits of 100 Nanyang cattle. The results showed that the 451 bp PCR product digested with Hinf I demonstrated polymorphism in the population, which was at Hardy-Weinberg equilibrium. Moreover, the frequencies of alleles A/B in the Nanyang population were 0.465/0.535. The association of the variations of the POU1F1 gene with the growth traits in the population was analyzed. The following parameters were greater in individuals with a genotype BB than in those with an genotype AB: birth weight, average weight increase before ablactation, body height at 12 months, body weight, body length, and chest girth at 6 months and 12 months (P<0.05). The body weight at 12 months was higher in the BB individuals than in the AA individuals (P <0.05). The body weight and body sizes also showed a trend of allele B> allele A in the other age groups. Therefore, the genotype BB maybe a dominant genotype and the allele B may be a dominant allele. These results imply that the allele B of the POU1F1 gene is likely to positively affect the growth traits.  相似文献

Stearoyl-CoA desaturase (SCD) is an iron-containing enzyme involving in the biosynthesis of monounsaturated fatty acids (MUFA) in mammary gland and adipose tissue, while decorin (DCN) consists of a protein core and a single dermatan or chondroitin sulfate glycosaminoglycan chain, contributing multifunctionally to matrix assembly, modulation of the activity of growth factors and cell migration and proliferation. However, few studies have focused on the genetic variability of them in goat. Herein, five Chinese goat breeds (1229 animals) were analyzed. Based on DNA pooling and PCR-RFLP, three nucleotide substitutions, one of which caused a amino acid substitution, were detected in SCD gene and three haploids (A, B, C) were constructed. According to SSCP analysis and DNA sequencing methods, a 2-bp deletion and two other SNPs were found existing in another analyzed gene DCN, and three haploids (X, Y, Z) were built. Associations between the genotypes and the growth traits (body length, body height, chest circumference, cannon circumference) were also analyzed. For SCD gene, genotype CC individuals had significant greater body height in Guanzhong and body length in both Guanzhong and Xinong saanen than genotype BC individuals (P < 0.05). For DCN gene, individuals with genotype XX was obviously higher than that with genotype XY (P < 0.05). These results indicated that genotype CC of SCD gene and genotype XX of DCN gene could be used for the breeding of new breeds of goat in China.  相似文献

The objective of the study was to assess the dimensionality of seven linear traits (body condition score, body stature, body length, heart girth, navel height, body depth and flank circumference) in Nguni cows using factor analysis and indicate the relationship between the extracted latent variables and calving interval (CI) and age at first calving (AFC). The traits were measured between December 2012 and November 2013 on 1559 Nguni cows kept under thornveld, succulent karoo, grassland and bushveld vegetation types. Low partial correlations (−0.04 to 0.51), high Kaiser statistic for measure of sampling adequacy scores and significance of the Bartlett sphericity test (P<0.01) showed that there were significant phenotypic correlations between the linear traits and the data were suitable for factor analysis. Two factors had eigenvalues >1. Factor 1 included body condition score, body depth, flank circumference and heart girth and represented body capacity of cows. Factor 2 included body length, body stature and navel height and represented frame size of cows. CI and AFC decreased linearly with increase of factor 1. There was a quadratic increase in AFC as factor 2 increased (P<0.05). It was concluded that the linear type traits under study can be grouped into two distinct factors, one linked to body capacity and the other to the frame size of the cows. Small-framed cows with large body capacities have shorter CI and AFC.  相似文献

Long‐term selection of goats for a certain production system and/or different environmental conditions will be reflected in the body morphology of the animals under selection. To investigate the variation contributing to different morphological traits and to identify genomic regions that are associated with body morphological traits in Sudanese goats, we genotyped 96 females belonging to four Sudanese goat breeds with the SNP52 BeadChip. After quality control of the data, the genome‐wide association study was performed using 95 goats and 24 027 informative single nucleotide polymorphisms (SNPs). Bicoastal diameter was significantly associated (LOD = 6.32) with snp10185‐scaffold1365‐620922 on chromosome 2. The minor allele has an additive effect, increasing the bicoastal diameter by 2.6 cm. A second significant association was found between body length and snp56482‐scaffold89‐467312 on chromosome 3 (LOD = 5.65). The minor allele is associated with increased body length. Additionally, five regions were suggestive for cannon bone, head width, rump length and withers height (LOD > 5). Only one gene (CNTNAP5) is located within the 1‐Mb region surrounding the significant SNP for bicoastal diameter on chromosome 2. The body length QTL on chromosome 3 harbors 49 genes. Further research is required to validate the observed associations and to prioritize candidate genes.  相似文献

Recently, a SNP (BIEC2-808543) was demonstrated to be associated with equine body size in horses. In this study, we genotyped BIEC2-808543 SNPs in 314 Yili horses in order to evaluate the association between genotype and body composition traits, such as body weight, withers height, chest circumference, and cannon circumference. Results indicate significant associations between polymorphisms of this SNP and body conformation in Yili horse populations. Based on these results, we hypothesize that BIEC2-808543 is strongly related to body conformation of Yili horses and has the potential to be used for marker-assisted selection.  相似文献

Secondary sexual traits, such as horns in ungulates, may be good indicators of genetic quality because they are costly to develop. Genetic effects on such traits may be revealed by examining correlations between multilocus heterozygosity (MLH) and trait value. Correlations between MLH and fitness traits, termed heterozygosity-fitness correlations (HFC), may reflect inbreeding depression or associative overdominance of neutral microsatellite loci with loci directly affecting fitness traits. We investigated HFCs for horn growth, body mass and faecal counts of nematode eggs in wild Alpine ibex (Capra ibex). We also tested if individual inbreeding coefficients (f') estimated from microsatellite data were more strongly correlated with fitness traits than MLH. MLH was more strongly associated with trait variation than f'. We found HFC for horn growth but not for body mass or faecal counts of nematode eggs. The effect of MLH on horn growth was age-specific. The slope of the correlation between MLH and yearly horn growth changed from negative to positive as males aged, in accordance with the mutation accumulation theory of the evolution of senescence. Our results suggest that the horns of ibex males are an honest signal of genetic quality.  相似文献

We looked for novel genetic variations within the Six6 gene by PCR-SSCP, DNA sequencing and forced RFLP-PCR and estimated their associations with production traits in 2132 goats of eight indigenous Chinese breeds. A novel single nucleotide polymorphism (NM_001104993.1: g.232T>C) within the goat Six6 gene was identified. The frequencies of allele "C" varied from 0.8621 to 1.000, which were in Hardy-Weinberg equilibrium. Genotype and allele frequencies were found to be significantly different in the breeds. Association analysis revealed a significant relationship between genotypes and cannon circumference in Hainan black goats (P = 0.047); adult individuals with genotype CT showed higher cannon circumference than those with genotype CC. Another significant association of genotypes with five-year-old fiber length was found in Inner Mongolia white cashmere goats (P = 0.002). In addition, individuals with genotype CT had longer fiber length than those with genotype CC. The data revealed that the Six6 gene positively affects growth traits and cashmere traits. Polymorphism of Six6-PstI could be useful as a DNA marker for goat breeding and genetics via marker-assisted selection.  相似文献

摘要: ZAG基因的功能主要是促进脂肪分解, 减少脂肪含量。文章利用PCR-SSCP和DNA测序技术研究了145头郏县红牛ZAG基因编码区4个位点(Z1、Z2、Z3、Z4)的多态性, 发现Z1、Z3、Z4 位点存在SSCP多态。对不同SSCP带型的对应片段进行了测序分析, 共发现6个新的SNP多态位点(C115T、A3257G、A4013G、T4027C、C4032T、T4120C)。Z3位点处于Hardy-Weinberg平衡状态, Z1、Z4 位点处于非平衡状态。不同基因型与生长发育性状的相关性分析显示, Z4位点上, AC基因型个体的体斜长、胸围、管围、体重指标显著(P<0.05)或极显著(P<0.01), 大于AA、AB基因型个体, 暗示该位点有可能作为郏县红牛生长性状标记辅助选择的标记之一。  相似文献

The involvement of melanocortin-3 receptor (MC3R) is well recognized in the regulation of feeding efficiency, body weight, and energy homeostasis. The objective of this study was to investigate the associations between MC3R gene polymorphisms and growth traits. Three novel SNPs (c.24C→T, c.220T→A, c.734G→C) and five haplotypes were identified in 234 Xiangxi cattle. The associations between MC3R gene polymorphisms and growth traits indicated that the individuals with TT and AT genotypes maintained higher body weight than those with the AA genotype at the c.220T→A locus (P < 0.05). The animals with GG and CG genotypes had higher heart girth and body weight than those with the CC genotype at c.734G→C (P < 0.05). The animals with H3H3 and H2H3 haplotype combinations had higher body weight than those with other haplotype combinations (P < 0.05). The results suggest that these SNPs in the MC3R gene might be useful genetic markers for marker-assisted selection and cattle breeding.  相似文献

Heterozygosity-fitness correlations (HFCs) have been reported in populations of many species. We provide evidence for a positive correlation between genetic variability and growth rate at 12 allozyme loci in a catadromous marine fish species, the European eel (Anguilla anguilla L.). More heterozygous individuals show a significantly higher length and weight increase and an above average condition index in comparison with more homozygous individuals. To a lesser extent, six microsatellite loci show a similar pattern, with positive but not significant correlations between heterozygosity and growth rate. The HFCs observed could be explained by an effect of either direct allozyme over-dominance or associative overdominance. Selection affecting some of the allozyme loci would explain the greater strength of the HFCs found at allozymes in comparison with microsatellites and the lack of correlation between MLH at allozymes and MLH at microsatellites. Associative overdominance (where allozyme loci are merely acting as neutral markers of closely linked fitness loci) might provide an explanation for the HFCs if we consider that allozyme loci have a higher chance than microsatellites to be in linkage disequilibrium with fitness loci.  相似文献
