Unlike ZrCl4, ZrBr4 is not ammonolysed in liquid ammonia at temperatures up to −33 °C. The existence of ammoniates ZrBr4nH3 (n = 17, 12 and 9) at −36 °C has been established; at room temperature, the hexammine ZrBr4 · 6NH3 is the stable species which becomes ZrBr4 · 2NH3 at 200 °C. When treated with an excess of NH4CN in liquid ammonia, complete replacement of bromide ions by cyanide occurs to give an inseparable mixture of Zr(CN)4 · 2NH3 and NH4Br. The chloride and bromide of zirconium(III) also undergo no ammonolysis in liquid ammonia; the ammoniates stable at room temperature are ZrCl3 · 2.5NH3 and ZrBr3 · 6NH3.  相似文献

Barley hull, a lignocellulosic biomass, was pretreated using aqueous ammonia, to be converted into ethanol. Barley hull was soaked in 15 and 30 wt.% aqueous ammonia at 30, 60, and 75 °C for between 12 h and 11 weeks. This pretreatment method has been known as “soaking in aqueous ammonia” (SAA). Among the tested conditions, the best pretreatment conditions observed were 75 °C, 48 h, 15 wt.% aqueous ammonia and 1:12 of solid:liquid ratio resulting in saccharification yields of 83% for glucan and 63% for xylan with 15 FPU/g-glucan enzyme loading. Pretreatment using 15 wt.% ammonia for 24–72 h at 75 °C removed 50–66% of the original lignin from the solids while it retained 65–76% of the xylan without any glucan loss.

Addition of xylanase along with cellulase resulted in synergetic effect on ethanol production in SSCF (simultaneous saccharification and co-fermentation) using SAA-treated barley hull and recombinant E. coli (KO11). With 3% w/v glucan loading and 4 mL of xylanase enzyme loadings, the SSCF of the SAA treated barley hull resulted 24.1 g/L ethanol concentration at 15 FPU cellulase/g-glucan loading, which corresponds to 89.4% of the maximum theoretical yield based on glucan and xylan.

SEM results indicated that SAA treatment increased surface area and the pore size. It is postulated that these physical changes enhance the enzymatic digestibility in the SAA treated barley hull.  相似文献

Potential environmental hazards from the excess accumulation of swine (Sus scrofa domesticus) manure in eastern North Carolina and new state guidelines on treatment alternatives have necessitated the reevaluation of best management practices for disposal of swine waste (manure and effluent) as a fertilizer source on local crop land. Creation of a value-added product is one viable means of utilizing and economically redistributing the nutrients in swine manure. Incubation studies using four agricultural soils from eastern North Carolina were conducted with pelletized processed swine lagoon solids (PSLS) (1.7% N, 2.5% P, 0.12% Cu and 0.18% Zn) composed of dewatered swine lagoon sludge plus rock flour. The PSLS was added at three application rates (0, 200, 400 mg N kg−1 soil) and incubated for 16 weeks at 25°C. The soil was sampled at 0, 1, 2, 4, 8, 12, and 16 weeks and analyzed for NO3–N, NH4–N and Mehlich III-extractable P, Zn, and Cu. High concentrations of NH4–N and low concentrations of NO3–N were present in the soils shortly after addition of PSLS. After the second week, extractable NH4–N dropped to <4 mg kg−1, while the soil concentration of NO3–N increased rapidly. The amount of NO3–N generated reached 90% of its final value after the fourth week. Across the four soils, 24–35% of the added N, 15–50% of the added P, 20–50% of the added Zn and 15–20% of the added Cu was extractable after 8 weeks. The PSLS is an excellent source of P, but may require additional N if used as a fertilizer source for most row crops.  相似文献

A multi-segmental mathematical model of human thermoregulation was tested for its capability to predict individualized physiological responses. We compared the model predictions obtained for an average person with measured individual responses of subjects exposed to mild cold. Secondly, body composition (BC) data, the resting metabolic rate (MR), and the actual measured MR during the test were used as input into the model.

The data was obtained from 20 subjects (age: 19–36 years; BMI: 17–32 kg/m2). BC, MR, rectal and skin temperatures were measured for 1 h at 22 °C, followed by 3 h at 15 °C.

A mean bias of 1.8 °C, with a standard error of 0.7 °C, resulted for the mean skin temperature of an average person at 15 °C. When subjective BC and measured MR were incorporated the bias was −0.2±0.9 °C. For the hand-back skin temperature the bias ± standard error fell from 5.3±2.8 °C for an average person to 2.0±2.5 °C, when using individualized characteristics. Trunk skin temperatures were not significantly affected by the adjustments.

In conclusion, this study shows that on a group level predictions of skin temperatures can be improved when adopting individualized body characteristics and measured MR, but the predictions on an individual level were not improved.  相似文献

Wheat straw consists of 48.57 ± 0.30% cellulose and 27.70 ± 0.12% hemicellulose on dry solid (DS) basis and has the potential to serve as a low cost feedstock for production of ethanol. Dilute acid pretreatment at varied temperature and enzymatic saccharification were evaluated for conversion of wheat straw cellulose and hemicellulose to monomeric sugars. The maximum yield of monomeric sugars from wheat straw (7.83%, w/v, DS) by dilute H2SO4 (0.75%, v/v) pretreatment and enzymatic saccharification (45 °C, pH 5.0, 72 h) using cellulase, β-glucosidase, xylanase and esterase was 565 ± 10 mg/g. Under this condition, no measurable quantities of furfural and hydroxymethyl furfural were produced. The yield of ethanol (per litre) from acid pretreated enzyme saccharified wheat straw (78.3 g) hydrolyzate by recombinant Escherichia coli strain FBR5 was 19 ± 1 g with a yield of 0.24 g/g DS. Detoxification of the acid and enzyme treated wheat straw hydrolyzate by overliming reduced the fermentation time from 118 to 39 h in the case of separate hydrolysis and fermentation (35 °C, pH 6.5), and increased the ethanol yield from 13 ± 2 to 17 ± 0 g/l and decreased the fermentation time from 136 to 112 h in the case of simultaneous saccharification and fermentation (35 °C, pH 6.0).  相似文献

An experiment examined the intake, growth responses and rumen digestion of young sheep fed ad libitum oat or barley straws alone or supplemented with approximately isonitrogenous amounts of barley grain and urea (Bar/N), safflower seed meal (SAF) or linseed meal (LIN) supplements provided at 3 day intervals. The supplements comprised 15–22% of total dry matter (DM) intake. Sheep offered either of the straws alone consumed 35.0–37.2 g DM/kg liveweight (LW0.75) per day of straw and an estimated 2.03–2.07 MJ metabolizable energy (ME) per day, and lost 85–97 g LW per day. Supplements increased (P<0.05 or <0.001) voluntary intake of straw and of total DM, and the organic matter (OM) digestibility of the entire diet. Each of the supplements increased (P<0.001) the estimated ME intake to a similar extent and changed the rapid LW loss of sheep fed straw alone to approximate LW maintenance. Rumen ammonia concentrations in sheep fed barley and oat straws alone (12 and 24 mg NH3/l, respectively) were expected to be deficient for microbial activity, but were increased (P<0.001) by provision of the supplements. Digestion of straw in synthetic fibre bags incubated in the rumen was markedly increased (P<0.01 or <0.001) when supplements were provided. Rumen pH was depressed briefly to pH <6.0 by the Bar/N, but not by the LIN or SAF, supplements. In young sheep fed cereal straws and losing LW rapidly the oilseed meal supplements increased wool growth more than the barley grain–urea supplements, but both types of supplement increased ME intake similarly and were equally effective to reduce the extent of LW loss.  相似文献

Investment by bladderwort (Utricularia foliosa) in carnivory, in terms of total C and N of bladders per leaf, was estimated in places with different nutrient concentrations from the Yahuarcaca Creek in the Colombian Amazon. The aims were to determine whether nutrient limiting conditions stimulate the investment in carnivory, and the relative balance between C and N invested in carnivory versus C and N obtained from prey. There were no significant differences either for phosphate (PO43−) concentration or for ammonia (NH4+) concentration among five sampling areas, along approximately 5 km long stretch of the creek, with a pooled mean ± S.D. of 0.19 ± 0.06 and 8.6 ± 3.0 μM, respectively. However, there were significant differences in the nitrate (NO3) concentration ranging from 0.6 to 2.5 μM. Total C and N of bladders per leaf increased with decreasing NO3. This corroborates the hypotheses that the carnivorous plant U. foliosa optimises its investment in carnivory according to nutrient availability in the water, and that N is a limiting factor that stimulates the investment in carnivory. The numbers of prey per bladder were also higher under NO3 limitation, thus enhancing the input of nutrients toward the plant through the bladders. The ratio of total C of prey captured/total C invested in bladders was always lower than 1. However, the efficiency of N was higher since when NO3 concentration was lower than 1 μM, the ratio of total N of prey captured/total N invested in bladders ranged between 0.97 and 1.67.  相似文献

Water-soluble non-starch polysaccharides were extracted from a Canadian malting barley (cv. Harrington) by sequential treatment with water at 40 °C (WE40) and 65 °C (WE65). The yields were 1.4 and 1.3% (w/w), respectively, of the dry barley grist. The WE40 extract was composed of 82.5% glucose, 8.9% xylose, and 7.0% arabiose residues, whereas WE65 contained 93.3% Glc, 3.3% Xyl, and 2.5% Ara. Only minute amounts of mannose and galactose residues were found in either fraction. Both extracts were further fractionated by stepwise (NH4)2SO4 precipitation into several polysaccharide populations. Subfractions from both extracts, obtained up to 45% saturation with (NH4)2SO4, contained mostly β-glucans, whereas subfractions precipitated at increasing saturation levels of (NH4)2SO4 (45–100%) contained progressively more arabinoxylans and less β-glucans. Compared to WE40, the WE65 extract was enriched in β-glucan populations with higher molecular size, higher limiting viscosity values, and higher content of β-(1 → 4) linkages. The ratio of tri-/ tetrasaccharide oligomers was also higher in β-glucans extracted at 65 °C than those extracted at 40 °C. Arabinoxylans in both extracts, WE40 and WE65, were highly substituted and contained large proportions of doubly substituted xylose residues.  相似文献

Cross-linked waxy maize (CWM) starch dispersions (STDs) of concentration 50 g kg−1 were heated in sucrose solutions containing 0–600 g kg−1 (g sucrose/kg dispersion) at 85 °C at low shear and in intermittently agitated cans at 110 °C. The STDs heated in 0–300 g kg−1 sucrose exhibited antithixotropic behavior, while those heated in 400–600 g kg−1 sucrose exhibited thixotropic behavior. The mean starch granule diameter of the starch dispersions did not show strong dependence on sucrose concentration. The dispersions, especially those with high sucrose concentrations and heated at 110 °C, exhibited G′ versus frequency (ω) profiles of gels. The STDs exhibited first normal stress differences that increased in magnitude with the concentration of sucrose. Values of the first normal stress coefficient of canned dispersions calculated from dynamic rheological data plotted against ω and experimental values plotted against shear rate of some of the STDs overlapped.  相似文献

Pre-cooling improves heat tolerance and time to exhaustion in the heat. We tested the possibility that reduced tissue temperatures may explain this phenomenon, using three whole-body treatments: pre-cooling, thermoneutral (control) and pre-heating. Pre-cooling reduced muscle temperature (Tm) by 6.3 °C while pre-heating increased Tm 3.4 °C, relative to control. Despite this offset, Tm climbed towards a common asymptote, with pre-cooling offering no thermal protection beyond 40 min. Following pre-cooling, exercising oesophageal temperature (Tes) initially increased at 0.09 °C min−1, being significantly faster than control (0.05 °C min−1) and pre-heated conditions (0.03 °C min−1). Pre-cooling lowered the sweat threshold and also resulted in a reduced cardiac frequency across the exercise-heat exposure. Our observations do not support the hypothesis that pre-cooling reduces Tm at the end of an exercise-heat exposure, thereby delaying the development of fatigue.  相似文献

Safflower (Charthamus tinctorius L.) seed press cake was pyrolysed in a fixed-bed reactor. The effects of pyrolysis temperature, heating rate and sweep gas flow rates on the yields of the products were investigated. Pyrolysis runs were performed using pyrolysis temperatures between 400 and 600 °C with heating rates of 10, 30 and 50 °C min−1. The obtained bio-char, gas and bio-oil yields ranged between 25 and 34 wt%, 19 and 25 wt%, and 28 and 36 wt%, respectively, at different pyrolysis conditions. The highest liquid yield was obtained at 500 °C pyrolysis temperature with a heating rate of 50 °C min−1 under the sweep gas of N2 with a flow rate of 100 cm3 min−1. Employing the higher heating rate of 50 °C min−1 results in maximum bio-oil yield, probably due to the decrease in mass transfer limitations. According to the results obtained under the conditions of this study, the effects of pyrolysis temperature and sweep gas flow rate are more significant than the effect of heating rate on the yields.  相似文献

In this communication, pretreatment of the anaerobically digested (AD) manure and the application of the pretreated AD manure as liquid medium for the simultaneous saccharification and fermentation (SSF) were described. Furthermore, fermentation of pretreated maize silage and wheat straw was investigated using 2 l bioreactors. Wet oxidation performed for 20 min at 121 °C was found as the most suitable pretreatment conditions for AD manure. High ammonia concentration and significant amount of macro- and micro-nutrients in the AD manure had a positive influence on the ethanol fermentation. No extra nitrogen source was needed in the fermentation broth. It was shown that the AD manure could successfully substitute process water in SSF of pretreated lignocellulosic fibres. Theoretical ethanol yields of 82% were achieved, giving 30.8 kg ethanol per 100 kg dry mass of maize silage.  相似文献

Cordyceps militaris mycelium produced mainly Cu, Zn containing superoxide dismutase (Cu, Zn-SOD). Cu, Zn-SOD activity was detectable in the culture filtrates, and intracellular Cu, Zn-SOD activity as a proportion protein was highest in early log phase culture. The effects of Cu2+, Zn2+, Mn2+ and Fe2+ on enzyme biosynthesis were studied. The Cu, Zn-SOD was isolated and purified to homogeneity from C. militaris mycelium and partially characterized. The purification was performed through four steps: (NH4)2SO4 precipitation, DEAE-sepharose™ fast flow anion-exchange chromatography, CM-650 cation-exchange chromatography, and Sephadex G-100 gel filtration chromatography. The purified enzyme had a molecular weight of 35070 ± 400 Da and consisted of two equal-sized subunits each having a Cu and Zn element. Isoelectric point value of 7.0 was obtained for the purified enzyme. The N-terminal amino acid sequence of the purified enzyme was determined for 12 amino acid residues and the sequences was compared with other Cu, Zn-SODs. The optimum pH of the purified enzyme was obtained to be 8.2–8.8. The purified enzyme remained stable at pH 5.8–9.8, 25 °C and up to 50 °C at pH 7.8 for 1.5 h incubation. The purified enzyme was sensitive to H2O2, KCN. 2.5 mM NaN3, PMSF, Triton X-100, β-mercaptoethanol and DTT showed no significant inhibition effect on the purified enzyme within 5 h incubation period.  相似文献

(1)Final temperature preferendum of juvenile (0.9–1.9 g) and adult (5.2–12.5 g) angelfish Pterophyllum scalare were determined with acute and gravitation methods. The final preferenda were similar, independent of the method and development stage (29.0–31.1°C).
(2)The critical thermal maxima (CTMax) for juveniles were 36.9°C, 37.6°C, 40.6°C, 40.8°C and for adults 38.4°C, 38.6°C, 41.0°C, 42.1°C. Adult angelfish CTMax was slightly higher than in juveniles (1°C; P<0.05); the endpoint of CTMax was the onset of spasms.
(3)The acclimation response ratio for both stages had an interval of 0.33–0.44; these values are in agreement with results for subtropical and tropical fishes.
(4)Therefore it is recommended that angelfish cultivation should be consistent with temperatures that do not change abruptly throughout the year and temperature maximum does not exceed 30°C.

Estimation of the ammonia production of the shrimp C. crangon in two littoral ecosystems (oligotrophic sand and eutrophic mud) was determined in winter and summer conditions from laboratory observations in experimental microcosms. The ammonia excretion rate of C. crangon was not influenced by either the sediment type or the ammonia concentration of the overlying water; on the other hand, the mean excretion rate and the response to initial handling stress increased markedly as shrimp were deprived of soft substratum.

The daily ammonia production of C. crangon was 16 μmol NH3 · g −1 wet wt · day −1 in winter and 40 μmol in summer. A gross production of 12 μmol NH3 · m−2 · day −1 and 300–700 μmol μ m−2 · day−1, respectively, could be expected in the two ecosystems studied. This would account for 5% (winter) and 2–4% (summer) of the total NH+4 flux at the sediment-water interface. The contribution of the excretion of all macrofauna to the NH+4 flux from the sediment is discussed.  相似文献

The effects of temperature and concentration on the viscosity of orange peel pectin solutions were examined at five different temperatures between 20 and 60°C and five concentration levels between 2.5–20 kg/m3. The effects of temperature was described by an Arrhenius-type equation. The activation energy for viscous flow was in the range 19.53–27.16 kJ/mol, depending on the concentration. The effect of concentration was described by two types of equation, power-law and exponential. Equations were derived which describes the combined effects of temperature and concentration on the viscosity for two different models in the range of temperatures and concentrations studied. Orange peel pectin was extracted by using HCl (pH 2.5, 90°C, 90 min) ammonium oxalate (0.25%, pH 3.5, 75°C, 90 min) and EDTA (0.5%, 90°C, 90 min) extraction procedures. The best result was obtained with ammonium oxalate extraction in which the pectin content of the final product was 30.12%, although the efficiency among the procedures varied.The average molecular weight was measured by light scattering technique. Magnitudes of intrinsic viscosity and molecular weight of pectins obtained by extraction with HCl, ammonium oxalate and EDTA were 0.262, 0.281, 0.309 m3/kg and 84 500, 91 400, 102 800 kg/kgmol, respectively. The molecular weight dependence of the intrinsic viscosity of the orange peel pectin solutions was expressed by Mark–Houwink–Sakurada equation. The data were fitted to equation as ηi=2.34×10−5(Mw,ave)0.8224 which helps to evaluate the average molecular weight of pectin solutions from orange peel with a knowledge of their intrinsic viscosity.  相似文献

Eight hemicellulosic fractions were obtained by sequential treatment of dewaxed barley straw with 0.1 M NaOH at 45 °C for 3 h, 0.25, 0.5, 1.0, 1.5, 2.0, and 3.0% H2O2 at 45 °C for 3 h at pH 11.5, and 10% KOH–1% Na2B4O7·10H2O at 28 °C for 15 h under continuous agitation. The yields of the fractions were 8.0, 3.1, 3.3, 3.3, 2.2, 2.0, 2.0, and 9.9%, respectively, of the initial amount of barley straw, corresponding to the dissolution of 21.6, 8.4, 8.9, 8.9, 5.9, 5.4, 5.4, and 26.7% of the original hemicelluloses. Meanwhile, the successive treatment also solubilized 29.1, 15.8, 14.6, 10.8, 4.5, 3.2, 2.7, and 3.7% of the original lignin, respectively. This sequential extraction together resulted in dissolution of 91.1% of the original hemicelluloses and 84.8% of the original lignin. The 0.1 M NaOH-soluble hemicellulosic fraction contained mainly xylose, glucose, and arabinose, 44.2, 15.7, and 15.2%, respectively, while the 10% KOH–1% Na2B4O7·10H2O-soluble fraction predominated in xylose, 75.0%. The six alkaline peroxide-soluble fractions were composed of 50.3–54.4% xylose, 14.7–16.9% arabinose, 6.8–10.7% glucose, 6.8–8.5% glucuronic acid or 4-O-methyl- -glucuronic acid, 0.4–1.5% mannose, and 0.3–1.2% rhamnose. All the hemicellulosic fractions contained substantial amounts of glucuronoarabinoxylans and noticeable quantities of β-glucans. In comparison, the six hemicellulosic fractions, isolated with alkaline peroxide, had much higher molecular weights (56,890–63,810 g mol−1) than those of the two hemicellulosic preparations (28,000–29,080 g mol−1), isolated with alkali in the absence of hydrogen peroxide. The thermal stability of the hemicelluloses increased with an increment of their molar mass.  相似文献

In order to study the effects of storage media and time of storage on the viability of unfertilized eggs of endangered Caspian brown trout (Salmo trutta caspius), the ova of this fish was stored in coelomic fluid and Cortland artificial media at 2–3 °C for 120 h. In this research, Cortland artificial medium was buffered with 20 mM of three different buffers: Hepes (C8H18N2O4S), Tris–HCl (C4H11NO3–HCl) and sodium salt Hepes (C8H17N2O4SNa). The pH of these media were adjusted according to natural pH of coelomic fluid. The eggs that stored in these media fertilized at times 0 h (eggs fertilized prior to storage), 48, 72 and 120 h of post-stripping, using fresh and pooled sperm obtained from four to six males. According to the results of present study, time of storage showed a significant (p < 0.05) main effect on eyeing, hatching and eyed eggs mortality rates. Eyeing and hatching rates significantly (p < 0.05) decreased from 97.4 ± 2.1% and 95.1 ± 4.4% at time 0 (eggs fertilized prior to storage) to 77.9 ± 3% and 65.5 ± 5% after 120 h of storage. Within a similar period of time, eyed eggs mortality significantly (p < 0.05) increased from 2.4 ± 2.4% to 17.2 ± 3.9%. No significant (p > 0.05) main effect was found among media buffered with Tris–HCl (82.8 ± 3.2%, 73.4 ± 5.4%, 12.1 ± 4.5%), Hepes (88.2 ± 3.4%, 80.7 ± 5.5%, 9.3 ± 3.4%), sodium salt Hepes (77.8 ± 3.8%, 69.3 ± 5.7%, 12.2 ± 3.9%) and coelomic fluid (84.8 ± 3.8%, 77.7 ± 5.1%, 8.9 ± 2.7%) for eyeing, hatching and eyed eggs mortality rates. There was a negative correlation (r = −0.895, p < 0.001) between eyed eggs mortality and hatching rates. In conclusion, unfertilized eggs of endangered Caspian brown trout can be successfully stored for 48 h without significant loss of fertility. But, storage for 120 h results in the falling of hatching rate. In addition, no significant difference was found between viability rates of ova stored in coelomic fluid and artificial media, 120 h post-storage. It reveals that artificial media could be substituted for coelomic fluid as storage medium at least for 120 h in Caspian brown trout.  相似文献

Nitrogen excreted as ammonium, urea, and dissolved primary amines (DPA), and nitrogen ingested by the planktonic calanoid copepod, Acartia tonsa, were measured while fed 4 foods with different N/C ratios in high (500 μg C l− 1) and low (50 μg C l− 1) concentrations. Adult copepods were fed the ciliate, Uronema marinum (N/C = 0.26), the diatom, Thalassiosira weissflogii, in log-phase growth (N/C = 0.20), and in senescent-phase growth (N/C = 0.12), and detritus derived from the saltmarsh grass, Spartina alterniflora, (N/C = 0.04). Total nitrogen excreted ranged from 0.06 to 0.18 μg N copepod− 1 d− 1 whereas nitrogen ingested exhibited considerably more variation (0.01 to 0.39 μg N copepod − 1d − 1). Ammonium was the dominant form of nitrogen excreted and was influenced by both food concentration and N/C ratio. Copepods fed foods with N/C ratios resembling their own body composition (log-phase diatoms and ciliates) excreted more ammonium when fed higher concentrations of food. In contrast, copepods fed foods with lower N/C ratios than their own body composition excreted more ammonium when fed lower concentrations of food, suggesting that they were catabolizing body protein for survival. Excretion of urea varied with food N/C ratio, with more urea excreted when the copepods were fed higher N/C foods. The excretion of DPA did not vary with either food concentration or food N/C ratio. Homeostasis serves to conserve the N/C ratio of copepods. Thus nitrogen excretion by healthy copepods should be expected to increase with ingestion only when copepods have high quantities of nitrogen-rich foods relative to the body composition of the copepods.  相似文献

Protonation of Na3[Ta(CO)5] in liquid ammonia provides the thermally unstable Na[Ta(CO)5NH3], which may be isolated as the crystalline and deep violet salt [Ph4As][Ta(CO)5NH3]. Sodium amminepentacarbonyltantalate(1−) reacts with PMe3, PPh3, P(OMe)3, AsPh3, SbPh3, CNtBu and CN at about 0°C in NH3/THF to give exclusively the corresponding [Ta(CO)5L]z.
