以菲和芘为多环芳烃(PAHs)代表物,采用水培体系研究了黑麦草(Lolium multiflorum Lam)对水中PAHs的吸收作用,重点研究了植物吸收菲和芘的时间动态.水中菲和芘起始浓度分别为1.00mg/L和0.12mg/L.0~288h内,黑麦草根和茎叶中菲和芘含量均先快速增加而后降低,积累量不断增大,植物根系和茎叶富集系数则先快速升高而后趋于稳定.茎叶中菲和芘含量、茎叶对菲和芘的富集系数比根低1~3数量级,积累量也明显小于根系.黑麦草根系对水中芘有更强的富集能力,其根系富集系数比菲大85%~179%;而其茎叶对菲的富集作用则略强.菲和芘在植物体内有明显的传导作用.0~288h,传导系数(TF)先显著升高而后趋于恒定;但实验条件下,菲和芘的TF值均很小,分别不高于0.031和0.009,且芘的TF值明显小于菲,表明供试植物对芘的传导能力更弱.  相似文献

测定了草甸棕壤条件下 ,菲、芘对小麦、白菜、西红柿 3种植物根伸长抑制率 ,以及复合污染毒性效应。结果表明 ,菲、芘浓度与植物根伸长抑制率呈显著线性或对数相关 (P =0 0 5 )。 2种有机物对植物根伸长抑制强度为菲 >芘。这与菲、芘的水中溶解度明显相关。与土壤脱氢酶活性和蚕豆根尖微核实验结果相比 ,植物根伸长对菲、芘毒性更敏感。小麦为有机污染的最敏感指示植物。菲、芘复合污染产生明显协同作用。  相似文献

为了探讨菲胁迫下植物的生理反应及耐受机制,以大豆为供试材料,采用叶片喷施试验,分别从表型、生物量、氧化损伤以及抗氧化酶活性等方面研究不同浓度菲处理对大豆幼苗生长的影响。结果表明:低浓度菲(≤50 μmol/L)能够增加叶片生物量,提高叶绿素及类胡萝卜素含量。同时,幼苗叶片中的丙二醛(Malondialdehyde,MDA)含量、脯氨酸含量,超氧化物歧化酶(Superoxide dismutase,SOD)活性以及过氧化物酶(Peroxidase,POD)的活性明显高于对照组。在高浓度菲(75-100 μmol/L)处理下,MDA含量继续增大,大豆幼苗的生物量、叶绿素含量、渗透胁迫物质含量及SOD活性都受到抑制,而叶片中的类胡萝卜素含量及POD活性较高。以上结果表明,植物通过提高SOD和POD活性,以及类胡萝卜素和渗透胁迫物质含量来减轻叶片施用菲对植物生物量、叶绿素含量及氧化胁迫的影响。此外,POD和类胡萝卜素在高浓度菲胁迫下仍保持较高水平,说明它们在菲胁迫中起到较为稳定的作用。  相似文献

多年生黑麦草抗氧化酶和植物络合素对Cd2+胁迫的应答   总被引:1,自引:0,他引:1  
采用水培方法研究了5 mg· L-1 Cd2+胁迫下,Cd在多年生黑麦草中的积累和Cd2+对多年生黑麦草抗氧化酶活性和植物络合素等巯基化合物浓度的影响.将具有3片展开叶的多年生黑麦草实生苗转至1/2霍格兰营养液中培养2周后,对其进行5 mg-L-1 Cd2+处理,分别在处理后的0、0.25、1、3、6d取样测定根系和叶片的Cd浓度、抗氧化酶活性和植物络合素等巯基化合物的浓度.结果表明,Cd2+处理多年生黑麦草6d后,根系中Cd浓度达到2.59 mg·g-1,叶片中Cd浓度达到0.24 mg·g-1,根中Cd向叶片的转运系数力0.093,叶中Cd的富集系数为48,多年生黑麦草属Cd高积累植物,具备在植物修复上应用的前景.Cd2+胁迫下,多年生黑麦草根叶中丙二醛(MDA)含量无显著变化,根中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性无显著变化,抗坏血酸过氧化物酶(APX)对Cd2+敏感,处理后6d活性较0d显著下降67.19%.Cd2+处理1d内,叶中SOD、APX、CAT活性显著降低.Cd2+处理后3d,叶中的抗氧化酶系统对叶中Cd浓度的升高做出了正反馈,SOD、APX、CAT的活性分别较处理后1d显著上升了14.19%、76,82%、99.26%,Cd2+处理时间延长至6d,SOD活性较处理后3d显著下降了18.58%,APX、CAT活性无显著变化.Cd2+处理后6d,多年生黑麦草根中半胱氨酸(Cys)、谷胱甘肽(GSH)、植物络合素2(PC2)、植物络合素3(PC3)、植物络合素4(PC4)、植物络合素5(PC5)和植物络合素6(PC6)浓度分别较处理0d提高了2.19、1.57、2.06、16.08、5.73、6.03和4.31倍,叶中Cys、GSH、PC2、PC3和PC4浓度分别较处理0d提高了0.69、3.21、1.64、5.73和0.27倍.根中PC3巯基比例最大,叶中GSH的巯基比例最大,二者是根、叶中巯基存在的主要形式.随着Cd2+处理时间的延长,根系和叶片中各巯基化合物的总巯基浓度显著升高,根系和叶片中植物络合素总巯基浓度与Cd浓度显著正相关.多年生黑麦草通过植物络合素等巯基化合物的快速合成降低了根叶中自由Cd2+的比例,保护了根叶中抗氧化酶的活性,间接维持了活性氧代谢的平衡.  相似文献

用含不同浓度Pb(NO3)2(0~40 mmol·L-1)的Hoagland营养液处理沙培中的木榄(Bruguiera gymnorrhiza)幼苗,2个月后测定幼苗过氧化物酶(POD)、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性并对其同工酶进行电泳分析。结果表明,1~10 mmol·L-1 Pb促进幼苗根和叶中抗氧化酶活性,但在20~40 mmol·L-1 Pb高浓度下,酶活性下降。电泳结果显示,Pb主要影响阴离子型POD同工酶活性。20~40 mmol·L-1Pb强烈抑制根中所有CAT同工酶的活性;30~40 mmol·L-1 Pb诱导出1条新的Cu/Zn-SOD同工酶LS-3和3条新的CAT同工酶RC-3、LC-2和LC-3。  相似文献

本文通过小区实验,研究再生水灌溉对葡萄叶片抗氧化酶及土壤酶的影响。结果表明,再生水灌溉的葡萄叶片MDA含量和CAT活性均高于清水对照的,而清水对照的葡萄叶片SOD和POD活性高于再生水灌溉的,但差异均不显著。再生水灌溉的土壤CAT活性比清水对照低;2011年的脲酶活性显著高于其他两年,而同年份间不同处理的土壤脲酶活性无显著差异。2011年再生水灌溉的土壤蔗糖酶低于清水对照,后两年则逐渐高于对照。土壤磷酸酶高于清水对照。再生水灌溉的葡萄产量高于清水对照的。  相似文献

农田中不断积累的多环芳烃不仅严重影响作物生长,同时增加粮食安全风险。筛选兼具促进植物生长特性和降解污染物功能的微生物菌株是解决上述问题的一种有效手段。从油田附近生长的植物根表分离得到一株具有芘降解能力,同时还具有溶磷、产吲哚乙酸和铁载体等植物促生特性的菌株PR3,经16S rDNA序列同源性分析确定为假单胞菌(Pseudomonas sp.)。菌株PR3在无机盐培养液中生长14 d后,对芘(20 mg/L)的降解率可达94%,对萘(50 mg/L)、菲(50 mg/L)、苯并(a)芘(10 mg/L)的降解率也分别达到92%,84%和47%。同时,该菌株7 d内最大溶磷量为756.25 mg/L,2 d内IAA合成量可达14.46 mg/L,4 d内生成铁载体的活性单位可达58.53%。在不同芘污染浓度处理下的盆栽实验表明,接种PR3可有效促进水稻生长并提高根际土壤中芘的降解,去除率可达72.02%-86.22%,同时显著降低水稻根及地上部中的芘含量,分别为21.81%-53.01%和49.81%-57.17%。因此,菌株PR3有助于实现芘污染土壤的生态修复以及降低作物芘暴露的风险。  相似文献

测定了草甸棕壤条件下,菲、芘、1,2,4—三氮苯对高等植物(小麦、白菜、西红柿)根伸长抑制串以及复合污染毒性效应。结果表明,菲、芘、1,2,4—三氮苯浓度与植物根伸长抑制串呈显著线性或对数相关(p=0.05)。3种化学品对植物根伸长抑制的强弱顺序为1,2,4—三氮苯>菲>芘。这与3种化学品的水中溶解度大小显著相关。小麦是3种供试植物中对有机污染物最敏感植物。菲、芘、1,2,4—三氮苯复合污染主表现为协同作用。  相似文献

以健康性成熟黑斑蛙为供试动物,以精巢组织丙二醛(MDA)含量、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)活性为指标,进行了水体铅、镉暴露的生态毒性响应研究.结果表明:(1)精巢MDA含量随铅、镉暴露浓度的升高而明显增加,且呈明显的浓度-效应关系.说明低水平铅、镉的长期暴露对黑斑蛙精巢具有一定的损伤作用;(2)SOD活性在各处理组响应变化不明显,CAT、GSH-Px活性则被显著诱导,说明GSH-Px、CAT在铅、镉引起的精巢抗氧化损伤中起着重要作用;(3)3种抗氧化酶相比,GSH-Px活性对铅、镉暴露响应最敏感,SOD活性的响应最不明显,精巢GSH-Px活性是指示铅、镉暴露的优选生物标志物。  相似文献

采用盆栽试验法,研究了蚯蚓(Pheretima hupeiensis)在植物修复芘污染土壤中的作用。结果显示,试验浓度(20.24-321.42 mg/kg) 范围内,蚯蚓活动促进了芘污染土壤中修复植物黑麦草(Lolium multiforum)黑麦草的生长,其根冠比明显增大。添加蚯蚓72 d后,种植黑麦草的土壤中芘的去除率高达60.01%-86.26%,其平均去除率(74.66%)比无蚯蚓活动的土壤-植物系统(64.55%)提高10.11%,比无植物对照组(18.24%)提高56.42%。各种生物、非生物修复因子中,植物-微生物交互作用对芘去除的平均贡献率(51.75%)最为突出,比无蚯蚓活动时(44.94%)提高6.81%。说明蚯蚓活动可强化土壤-植物系统对土壤芘污染的修复作用。  相似文献

The influence of arsenic (As) on the growth and the antioxidant system of Lemna gibba L. exposed to five concentrations of As (0.0, 0.25, 0.5, 1.0, and 1.5 mg L?1) was studied. Although As exposure reduced relative growth rate, L. gibba continued to grow even after the high accumulation of this element after five days of exposure. The concentration of the superoxide anion was unaffected by As, whereas an increase of hydrogen peroxide concentrations was observed with the increasing of As concentration. Increasing concentrations of As also increased the enzyme activity of superoxide dismutase, peroxidase, and glutathione reductase and the total and nonprotein thiols, up to 0.5 mg L?1, whereas the anthocyanin content increased constantly with As concentration. Catalase and ascorbate peroxidase activities as well as the content of chloroplastic pigments were reduced in plants exposed to all As concentrations. These results support a major role of anthocyanins, nonprotein thiols, and antioxidant scavenging enzymes in L. gibba tolerance to toxic As concentrations.  相似文献

Sorption of phenanthrene (Phen) and benzo[α]pyrene (BaP) by humic acids (HAs) extracted from four typical soils in China, including cinnamon soil, fluvo-aquic soil, red soil, and mountain meadow soil were investigated. All sorption data were fitted well by the Freundlich model, but BaP exhibited stronger and more nonlinear sorption than Phen by a given HA. For Phen isotherms, there was a positive relation between K oc values and aliphaticity of HAs, whereas a negative correlation was observed between n values and aliphaticity. This indicated the importance of aliphatic groups in Phen sorption capacity and nonlinearity. Compared to Phen, a similar trend was obtained between n values of BaP and aliphaticity or aromatic carbon. However, no correlation existed between K oc values for BaP and aliphaticity or aromatic carbon.  相似文献

In the present study, the impact of potassium phosphite on response of cucumber plants inoculated with Pythium ultimum var. ultimum was assessed. Variations in the accumulation of both antioxidant enzymes and growth parameters were investigated. The results revealed that fresh and dry weights of shoot and root exhibited up to 2 fold increase in potassium phosphite-treated plants. The concentrations of peroxidase, superoxide dismutase and catalase were significantly higher in the plants treated with 4 g L?1 of potassium phosphite compared to other treatments, at 48 h after inoculation. It was demonstrated that both 2 and 4 g L?1 treatments could alleviate the disease damage to a high extent, while control plants were severely damaged by the pathogen. The results of this study suggest that the increased induction of antioxidant enzymes (2.2, 2.8 and 4 fold increase for superoxide dismutase, catalase and peroxidase, respectively) might have alleviated damping-off symptoms leading to increased plant growth and yield.  相似文献

Lung cancer is the leading cause of cancer related mortality worldwide. Crocetin, saffron plant derivative known to play a role in cancer chemoprevention. In the present study the effects of crocetin was tested against lung cancer-bearing mice in both pre-initiation and post-initiation periods. Healthy male Swiss albino mice (6–8 weeks old) were used throughout the study. Experiment was designed with the treatment regimen of crocetin [20 mg/kg body weight dissolved in dimethyl sulphoxide (DMSO)] for 4 weeks before (pre-initiation) and from 12th week after Benzo(a) pyrene B(a)p (50 mg/kg body weight) induced lung carcinoma(post-initation). The level of lipid peroxidation (LPO) and marker enzymes markedly increased in carcinogen administered animals, which was brought back to near normal by crocetin treatment. The activities of the enzymic antioxidants and glutathione metabolizing enzymes were decreased in B(a)p induced animals and increased upon drug treatment. Crocetin profoundly reverted back the pathological changes observed in cancerous animals. From the results crocetin proves to scavenge free radical and plays an important role in cellular function. Tumor incidence and histopathological studies proves crocetin is a potent antitumour agent.  相似文献

Lipid peroxidation induced by metals at sub-lethal levels, alter physiological and biochemical characteristics of biological systems. To counter the detrimental effects of the prooxidant activity of metals, a group of antioxidant enzyme systems function in the organisms. The present study was performed to investigate into the lipid peroxidation product formation due to the exposure to effects of the metals namely aluminium, lead and cadmium at sub-lethal concentrations and the biological response through protective antioxidant enzyme activity in the marine mussels,Perna viridis Lin. This organism is a known bioindicator and bioconcentrator of metals in the environment.The results of the present study were: (a) accumulation of lead showed a definite linear increase during the period of exposure whereas aluminium and cadmium showed fluctuations. Mantle and gill tissues showed greater accumulation of metals when compared to digestive gland; (b) lead and aluminium induced lipid peroxidation was greater in tissues than the peroxidation induced by cadmium. Cadmium induced peroxidation was observed only after the day 7 of the exposure; (c) anti-oxidant enzymes activity levels were significantly higher in digestive gland and mantle than gills; (d) mantle was observed to significantly contribute to the organismal response to lipid peroxidation as indicated by high activity levels of anti-oxidant enzymes.  相似文献

Summary The activities of three antioxidant enzymes, superoxide dismutase, catalase, and glutathione peroxidase, were monitored in isolated human renal adenocarcinoma tissues and in cultured human renal adenocarcinoma cells. The results were compared to the activities of these enzymes in the proposed cell of origin, isolated human proximal tubular tissues, and cultured proximal tubular epithelial cells. Strong modulation of these enzymes by culture conditions was observed in normal cells but not in carcinoma cells. Low levels of cellular lipid peroxidation, as assessed by levels of malondialdehyde (MDA), were observed in adenocarcinoma cells under the culture conditions tested with one exception: greatly elevated MDA was observed in renal adenocarcinoma cells growth on plastic in serum-free, chemically defined medium. This increased lipid peroxidation correlated with a loss of cell viability under these conditions. This work was supported by a grant from the Veterans Adminsitration (T. D. The research aimed to verify the important physiological effect of nitrogen (N) on plants exposed to cadmium (Cd). The poplar plants were grown in a Hoagland nutrient solution and treated with extra N, Cd, and N + Cd. After treatment, plant growth and chlorophyll content were recorded. The oxidative stress, the activity of antioxidant enzymes, and the expression of related genes were also examined. The results indicated the plants treated with sole Cd presented obvious toxicity symptoms, i.e. growth inhibition, reactive oxygen species accumulation, and chlorophyll content decrement. However, when N was added to the plants under Cd stress, plant growth was enhanced, chlorophyll synthesis was promoted, and the oxidative stress was alleviated. Further, the expression of antioxidant enzymes genes was upregulated by N. The results indicated that N partially reversed the toxic effect of Cd on poplar plants, which can provide new methodology to enhance the phytoremediation technology for heavy metal pollution soil.  相似文献

飞机草(Eupatorium odoratum)原产于中南美洲,在我国最早发现于1934年,目前已经分布到云南、海南、台湾、广东等省区,是一种典型的外来入侵种。研究其在逆境条件下的生理适应机制对于有效治理该入侵种具有十分重要的意义。研究了其在不同环境胁迫条件(高温、低温和干旱)下抗氧化酶系统的变化。结果表明:超氧化物歧化酶(SOD)、抗坏血酸过氧化物酶(APX)和脱氢抗血酸还原酶(DHAR)在3种环境胁迫下酶活性都升高;过氧化氢酶(CAT)在高温胁迫下升高,而在低温和干旱胁迫下酶活性降低;过氧化物酶(POD)和单脱氢抗坏血酸还原酶(MDAR)在高温和干旱胁迫下酶活性升高,而在低温胁迫下酶活性降低;谷胱甘肽还原酶(GR)在高温和干旱胁迫下酶活性升高,而在低温胁迫下保持不变。这些研究结果可能有助于寻找到一条控制飞机草的新途径。  相似文献

Objectives: This study was designed to compare the levels of copper/zinc superoxide dismutase (Cu/Zn SOD), peroxidase (POx) and glutathione peroxidase (GSH‐Px) in saliva of smokers and those in saliva of non‐smokers. Methods: Unstimulated saliva samples were collected from 88 elderly males (65 years old or over) who visited a private dental clinic. Forty‐four subjects were current smokers (more than 20 cigarettes daily for at least 30 years) and 44 were non‐smokers. The levels of salivary thiocyanate, Cu/Zn SOD, GSH‐Px, and POx activity were measured using standard procedures. Results: The mean levels of salivary thiocyanate (SCN?) and SOD were significantly higher (p < 0.01) in the smoking group than in the non‐smoking group, whereas the specific activity levels of POx and GSH‐Px were significantly higher (p < 0.05) in the non‐smoking group than in the smoking group. Significant correlation coefficients were found between the levels of SCN? and SOD (r = 0.37, p < 0.001). In the non‐smoking group, a significant positive association was found between specific activity of POx and age (r = 0.33, p < 0.05). Conclusion: Measurement of SCN? and Cu/Zn SOD in human saliva might be useful for estimating the level of oxidative stress caused by cigarette smoke. Despite increased H2O2 level as a defense system induced by SOD, detoxification of H2O2 might be deteriorated in the oral cavity of elderly smokers.  相似文献
