Two amino acid substitutions (L1014F and M918T) in the voltage-gated sodium channel confer target-site resistance to pyrethroid insecticides in the peach potato aphid, Myzus persicae. Pyrethroid-resistant and -susceptible M. persicae clones with various combinations of these mutations were crossed under laboratory conditions, and the genotypes of aphid progeny were analysed by direct DNA sequencing of the IIS4-S6 region of the sodium channel gene. Segregation patterns showed that in aphids heterozygous for both L1014F and M918T, both mutations were present in the same resistance allele. Despite these mutations appearing largely recessive in other pest species, such aphids exhibited strong resistance to pyrethroids in leaf-dip bioassays. These results have important implications for the spread and management of pyrethroid resistance in field populations.  相似文献

Target‐site mutations and detoxification gene overexpression are two major mechanisms conferring insecticide resistance. Molecular assays applied to detect these resistance genetic markers are time‐consuming and with high false‐positive rates. RNA‐Seq data contains information on the variations within expressed genomic regions and expression of detoxification genes. However, there is no corresponding method to detect resistance markers at present. Here, we collected 66 reported resistance mutations of four insecticide targets (AChE, VGSC, RyR, and nAChR) from 82 insect species. Next, we obtained 403 sequences of the four target genes and 12,665 sequences of three kinds of detoxification genes including P450s, GSTs, and CCEs. Then, we developed a Perl program, FastD, to detect target‐site mutations and overexpressed detoxification genes from RNA‐Seq data and constructed a web server for FastD (http://www.insect-genome.com/fastd). The estimation of FastD on simulated RNA‐Seq data showed high sensitivity and specificity. We applied FastD to detect resistant markers in 15 populations of six insects, Plutella xylostella, Aphis gossypii, Anopheles arabiensis, Musca domestica, Leptinotarsa decemlineata and Apis mellifera. Results showed that 11 RyR mutations in P. xylostella, one nAChR mutation in A. gossypii, one VGSC mutation in A. arabiensis and five VGSC mutations in M. domestica were found to be with frequency difference >40% between resistant and susceptible populations including previously confirmed mutations G4946E in RyR, R81T in nAChR and L1014F in VGSC. And 49 detoxification genes were found to be overexpressed in resistant populations compared with susceptible populations including previously confirmed detoxification genes CYP6BG1, CYP6CY22, CYP6CY13, CYP6P3, CYP6M2, CYP6P4 and CYP4G16. The candidate target‐site mutations and detoxification genes were worth further validation. Resistance estimates according to confirmed markers were consistent with population phenotypes, confirming the reliability of this program in predicting population resistance at omics‐level.  相似文献

We examined the effect of fertilization on population growth and within‐plant distribution of melon or cotton aphid, Aphis gossypii Glover, on potted chrysanthemum, Dendranthema grandiflora (Tzvelev). In terms of pest management implications, we also investigated the effect of fertilization on the number of insecticide applications needed to control A. gossypii on potted chrysanthemum. Population growth rate of A. gossypii increased with fertilization levels from 0 to 38 ppm N and reached a plateau from 38 to 488 ppm N. Increased fertilization beyond 38 ppm N, 10% of the commercial standard, did not result in higher aphid number. Aphids responded to nutrient availability of plants by distributing themselves in areas with higher level of nitrogen. More aphids were found in the apical and middle strata of the plants than the basal stratum, which had the lowest nitrogen content. Leaf nitrogen content increased with increased fertilization level and was consistently higher in the apical and middle strata than the basal stratum. Increased fertilization from 0 to 375 ppm N did not result in higher number of insecticide applications. All three insecticides (bifenthrin, kinoprene or pymetrozine) were effective in keeping the aphid infestation below a pre‐determined level, five aphids per plant, but pymetrozine required the least number of applications. For chrysanthemum, a fast‐growing crop and heavy utilizer of nitrogen, increased fertilization shortened the time to flowering, which would allow growers to harvest their crop sooner and reduce the time for aphid population growth. Reduction in time to harvest could result in significant reduction of insecticide usage by reducing the time for aphid population growth. As a result, high fertilization together with minimal runoff may be a useful tactic to an integrated pest management (IPM) programme for managing A. gossypii on potted chrysanthemums.  相似文献

The broad-spectrum insecticides greatly influence the control of cotton aphids; however, due to frequent chemical control, Aphis gossypii (Hemiptera: Aphididae) has developed resistance against several classes of synthetic insecticides. In this study, we explored the sub-lethal effects of imidacloprid and pirimicarb, two commonly used insecticides for aphid control, on a parasitoid wasp, Lysiphlebus fabarum (Marshall) (Braconidae: Aphidiinae), when simultaneously used to control melon aphid on cucumber plants, as part of a comprehensive study for integrated pest management. Bioassays of imidacloprid and pirimicarb were performed to calculate LC50 with third instars of A. gossypii. The LC50 of these insecticides (110.55 and 250.89 μg/lit, respectively) were used to expose the wasp larvae, pupae, and adult parasitoids on a cucumber leaf. The percent mortality, percent adult emergence, and sex ratio were calculated during each exposure test. Moreover, the body size, egg load, and mature egg size of wasps surviving the insecticide treatments, as well as the sex ratio of the second generation was evaluated. Regardless of the host aphid mortality, none of the insecticides caused mortality of larval stage of the parasitoid. The insecticide application on pupal stage revealed that the percentage of mortality, sex ratio, body size, and egg load of surviving wasps, as well as the sex ratio of their offspring was adversely affected by imidacloprid, but not by pirimicarb. The present study suggests pirimicarb as a preferred insecticide, with less harmful effects on the fitness components of L. fabarum, for integrated pest management of cotton aphids.  相似文献

1 Control failures of insecticides used against the tomato leafminer Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) in Brazil led to the investigation of the possible occurrence of resistance of this insect pest to abamectin, cartap, methamidophos and permethrin. 2 The insect populations were collected from seven sites in the states of Minas Gerais, Rio de Janeiro, and São Paulo. These populations were subjected to concentration–mortality bioassays using insecticide‐impregnated filter papers. 3 We were unable to obtain a single population which provided a susceptibility standard for all insecticides tested. Therefore, the resistance levels were estimated in relation to the most susceptible population to each insecticide. Resistance to abamectin and cartap were observed in all populations when compared with the susceptible standard population, with resistance ratios ranging from 5.2‐ to 9.4‐fold and from 2.2‐ to 21.9‐fold for abamectin and cartap, respectively. Resistance to permethrin was observed in five populations with resistance ratios ranging from 1.9‐ to 6.6‐fold, whereas resistance to methamidophos was observed in four populations with resistance ratios ranging from 2.6‐ to 4.2‐fold. 4 The long period and high frequency of use of these insecticides against this insect pest suggest that the evolution of insecticide resistance on them has been relatively slow. Alternatively, the phenomenon might be widespread among Brazilian populations of T. absoluta making the finding of suitable standard susceptible populations difficult and leading to an underestimation of the insecticide resistance levels in this pest. 5 Higher levels of resistance to abamectin, cartap and permethrin are correlated with greater use of these compounds by growers. This finding suggests that local variation in insecticide use was an important cause of variation in susceptibility.  相似文献

Insecticides are often implicated in causing outbreaks of the cotton aphid, Aphis gossypii (Glover) (Homoptera: Aphididae), through stimulation of reproduction. In this study we report the sublethal effects of dosages of bifenthrin, acephate, carbofuran or pyriproxifen on cotton aphid reproduction. We could not detect any increase or decrease in the intrinsic rate of increase of cotton aphids exposed to bifenthrin, acephate or carbofuran. However, we did detect some increases in the net reproductive rate of aphids treated with bifenthrin justifying further investigation of the effect on reproduction by this insecticide. Trends based on simple linear regression models suggest that sublethal dosages of bifenthrin or carbofuran have a negative impact on aphid population growth as dosages increase. These data suggest that stimulation of reproduction by these insecticides probably does not play a major role in cotton aphid outbreaks or resurgence. Pyriproxyfen is a juvenoid currently used for control of whiteflies in cotton. It demonstrated significant activity towards cotton aphid reared on treated cotton in our bioassays. Pyriproxyfen caused sterility in most aphids exposed to dosages exceeding 1 ppm, and reduced aphid longevity by approximately 50%. However, it did not appear to greatly influence the reproductive potential or longevity of reproductively mature aphids. A field study indicates that pyriproxyfen affects cotton aphid population structure and may have potential in managing cotton aphid outbreaks. Modifying aphid population structure and growth through the use of juvenoids such as pyriproxifen may prove to be an effective proactive approach to pest control without adversely impacting beneficial organisms or causing pest resurgence.  相似文献

The Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Liviidae), is an important insect pest of citrus. It is the vector of ‘Candidatus’ Liberibacter asiaticus, a phloem‐limited bacterium that infects citrus, resulting in the disease Huanglongbing (HLB). Disease management relies heavily on suppression of D. citri populations with insecticides, including pyrethroids. In recent annual surveys to monitor insecticide resistance, reduced susceptibility to fenpropathrin was identified in several field populations of D. citri. The primary target of pyrethroids is the voltage‐gated sodium channel (VGSC). The VGSC is prone to target‐site insensitivity because of mutations that either reduce pyrethroid binding and/or alter gating kinetics. These mutations, known as knockdown resistance or kdr, have been reported in a wide diversity of arthropod species. Alternative splicing, in combination with kdr mutations, has been also associated with reduced pyrethroid efficacy. Here we report the molecular characterization of the VGSC in D. citri along with a survey of alternative splicing across developmental stages of this species. Previous studies demonstrated that D. citri has an exquisite enzymatic arsenal to detoxify insecticides resulting in reduced efficacy. The results from the current investigation demonstrate that target‐site insensitivity is also a potential basis for insecticide resistance to pyrethroids in D. citri. The VGSC sequence and its molecular characterization should facilitate early elucidation of the underlying cause of an established case of resistance to pyrethroids. This is the first characterization of a VGSC from a hemipteran to this level of detail, with the majority of the previous studies on dipterans and lepidopterans.  相似文献

【目的】测定湖北草地贪夜蛾田间种群对4种杀虫剂的敏感性,并对杀虫剂靶标Ace-1,VGSC和RyR的基因型和突变频率进行检测,以明确湖北草地贪夜蛾田间种群药剂敏感性现状,进而指导田间科学用药。【方法】采集湖北黄冈、武穴、咸宁和荆州4个地区玉米田中的草地贪夜蛾幼虫,采用浸叶法测定其对氯虫苯甲酰胺、乙基多杀菌素、茚虫威和甲维盐4种杀虫剂的敏感性;提取4个种群80头个体的单头总RNA合成cDNA,利用特异性引物进行PCR扩增,获得目的基因片段,根据序列比对和测序峰图分析,确定Ace-1,VGSC和RyR的基因型和靶标突变位点的突变频率。【结果】生测结果表明,湖北这4个田间种群的草地贪夜蛾幼虫对氯虫苯甲酰胺、乙基多杀菌素、茚虫威和甲维盐均比较敏感,敏感性从高到低依次为:甲维盐>乙基多杀菌素>氯虫苯甲酰胺>茚虫威。分子检测结果表明,湖北草地贪夜蛾这4个田间种群Ace-1基因在A201S, G227A和F290V位点均存在抗性杂合突变,且在F290V位点检测到抗性纯合突变,而VGSC和RyR两个基因均没有检测到靶标位点突变。【结论】湖北草地贪夜蛾4个田间种群对氯虫苯甲酰胺、乙基多...  相似文献

Neonicotinoid insecticides, such as imidacloprid, are selective agonists of insect nicotinic acetylcholine receptors (nAChRs) and are used extensively to control a variety of insect pest species. Previously, we have identified a nAChR point mutation (Y151S) associated with insecticide resistance in the brown planthopper Nilaparvata lugens . Although this mutation has been identified in two different N. lugens nAChR subunits (Nlα1 and Nlα3) because of difficulties in heterologous expression of Nlα3; its influence on agonist potency has been examined only in Nlα1-containing nAChRs. Here we describe the cloning of a novel nAChR subunit from N. lugens (Nlα8), together with evidence for its co-assembly with Nlα3 in native and recombinant nAChRs. This has, for the first time, enabled the functional effects of the Nlα3Y151S mutation to be examined. The Nlα3Y151S mutation has little effect on agonist potency of acetylcholine but has a dramatic effect on neonicotinoid insecticides (reducing I max values and increasing EC50 values). The apparent affinity of neonicotinoids was higher and the effect of the Y151S mutation on neonicotinoid agonist potency was more profound in Nlα3-containing, rather than Nlα1-containing nAChR. We conclude that Nlα3- and Nlα1-containing nAChRs may be representative of two distinct insect nAChR populations.  相似文献

The fall armyworm (FAW), Spodoptera frugiperda, is a major pest native to the Americas that has recently invaded the Old World. Point mutations in the target-site proteins acetylcholinesterase-1 (ace-1), voltage-gated sodium channel (VGSC) and ryanodine receptor (RyR) have been identified in S. frugiperda as major resistance mechanisms to organophosphate, pyrethroid and diamide insecticides respectively. Mutations in the adenosine triphosphate-binding cassette transporter C2 gene (ABCC2) have also been identified to confer resistance to Cry1F protein. In this study, we applied a whole-genome sequencing (WGS) approach to identify point mutations in the target-site genes in 150 FAW individuals collected from China, Malawi, Uganda and Brazil. This approach revealed three amino acid substitutions (A201S, G227A and F290V) of S. frugiperda ace-1, which are known to be associated with organophosphate resistance. The Brazilian population had all three ace-1 point mutations and the 227A allele (mean frequency = 0.54) was the most common. Populations from China, Malawi and Uganda harbored two of the three ace-1 point mutations (A201S and F290V) with the 290V allele (0.47–0.58) as the dominant allele. Point mutations in VGSC (T929I, L932F and L1014F) and RyR (I4790M and G4946E) were not detected in any of the 150 individuals. A novel 12-bp insertion mutation in exon 15 of the ABCC2 gene was identified in some of the Brazilian individuals but absent in the invasive populations. Our results not only demonstrate robustness of the WGS-based genomic approach for detection of resistance mutations, but also provide insights for improvement of resistance management tactics in S. frugiperda.  相似文献

Aphis gossypii Glover is a cosmopolitan and polyphagous aphid species. When neonicotinoid-resistant clones of A. gossypii were recently found in Japan, it was subsequently discovered that a point mutation (R81T) in the D-loop region of the β1 subunit of the nicotinic acetylcholine receptor gene had conferred this resistance. To quickly identify this neonicotinoid-resistant allele, we developed a molecular diagnostics test using a multiplex polymerase chain reaction method that targeted the R81T mutation.  相似文献

1 Spatial and temporal habitat heterogeneity represented by annual crops is a major factor influencing population dynamics of phytophagous insect pests such as the cotton aphid Aphis gossypii Glover. We studied the effects of instability of the cotton agroecosystem resulting from the temporary availability of the plant resource and the repeated use of insecticides on the genetic variability of the cotton aphids.
2 Samples of A. gossypii were collected in cotton plots, treated or not with insecticides and from vegetable crops (Malvaceae, Cucurbitaceae and Solanaceae) within the cotton growing area of northern Cameroon. The genetic structure of the samples was assessed using eight microsatellite markers. Insecticide resistance was estimated through the detection of two mutations in the ace -1 gene that are associated with insensitivity of acetylcholinesterase to carbamate and organophosphate insecticides.
3 The results obtained show that both host plants and insecticides act in genetic structuring of A. gossypii . Ninety-three percent of aphids collected on cotton were characterized by the same microsatellite multilocus genotype, Burk1 , which also displays the insecticide resistant alleles.
4 During the dry season, the cotton crop season after, the genotype Burk1 was principally found on two other malvaceous cultivated plants, rosella and okra, acting as suitable reservoir plants. The ability of the cotton aphid to move among asynchronous suitable habitats in response to changes in resource availability enables the pest to exploit unstable cropping systems. An understanding of the cotton aphid life system may aid to improve strategies for integrated resistance management.  相似文献

Recent advances in the characterisation of insect sodium channel gene sequences have identified a small number of point mutations within the channel protein that are implicated in conferring target-site resistance to pyrethroid insecticides (so-called knockdown resistance or kdr). The L1014F (leucine-to-phenylalanine) mutation located in the centre of segment 6 of the domain II region (IIS6) of the sodium channel (the so-called kdr trait) has been detected in the peach-potato aphid, Myzus persicae (Sulzer), and is considered to be the primary cause of pyrethroid resistance in this species. Here we report on the characterisation of a second mutation, M918T (methione-to-threonine), within the nearby IIS4-S5 intracellular linker (the so-called super-kdr trait) in a field clone also possessing L1014F, with both mutations present in heterozygous form. The resistance phenotype of M. persicae clones possessing various combinations of L1014F and M918T to a wide range of pyrethroids (both Type I and II) was assessed in leaf-dip bioassays and to lambda-cyhalothrin applied at up to ten times the recommended field rate as foliar sprays to aphids feeding on whole plants. Bioassay results demonstrated that presence of both mutations was associated with extreme resistance to all the pyrethroids tested relative to aphids lacking the mutations. Furthermore, this resistance well exceeded that shown by aphids that were homozygous for L1014F but lacking M918T. However, pre-treatment with piperonyl butoxide in the leaf-dip bioassays failed to suppress pyrethroid resistance in aphids carrying one or both of the mutations. The relevance of these findings for monitoring and managing pyrethroid resistance in M. persicae populations in the field is discussed.  相似文献

The Colorado potato beetle (Leptinotarsa decemlineata) is a major agricultural pest of solanaceous crops. An effective management strategy employed by agricultural producers to control this pest species is the use of systemic insecticides. Recent emphasis has been placed on the use of neonicotinoid insecticides. Despite efforts to curb resistance development through integrated pest management approaches, resistance to neonicotinoids in L. decemlineata populations continues to increase. One contributing factor may be alterations in insect fatty acids, which have multiple metabolic functions and are associated with the synthesis of xenobiotic-metabolizing enzymes to mitigate the effects of insecticide exposure. In this study, we analyzed the fatty acid composition of L. decemlineata populations collected from an organic production field and from a commercially managed field to determine if fatty acid composition varied between the two populations. We demonstrate that a population of L. decemlineata that has a history of systemic neonicotinoid exposure (commercially managed) has a different lipid composition and differential expression of known metabolic detoxification mechanisms relative to a population that has not been exposed to neonicotinoids (organically managed). The fatty acid data indicated an upregulation of Δ6 desaturase in the commercially managed L. decemlineata population and suggest a role for eicosanoids and associated metabolic enzymes as potential modulators of insecticide resistance. We further observed a pattern of delayed emergence within the commercially managed population compared with the organically managed population. Variations in emergence timing together with specific fatty acid regulation may significantly influence the capacity of L. decemlineata to develop insecticide resistance.  相似文献

Cotton is an economically important crop and its production is challenged by the diversity of pests and related insecticide resistance. Identification of the conserved target across the cotton pest will help to design broad spectrum insecticide. In this study, we have identified conserved sequences by Expressed Sequence Tag profiling from three cotton pests namely Aphis gossypii, Helicoverpa armigera, and Spodoptera exigua. One target protein arginine kinase having a key role in insect physiology and energy metabolism was studied further using homology modeling, virtual screening, molecular docking, and molecular dynamics simulation to identify potential biopesticide compounds from the Zinc natural database. We have identified four compounds having excellent inhibitor potential against the identified broad spectrum target which are highly specific to invertebrates.  相似文献

The cotton aphid, Aphis gossypii Glover (Hem: Aphididae), is one of the most injurious pests of fruits, vegetables and ornamental plants worldwide, both outdoor and indoor. Currently, the main method of control of this pest is through application of pesticides which is mostly accompanied by the resistance of the pest against pesticide(s). The resurgence of resistant aphid populations brings about further contamination of foodstuff and environment. Essential oils obtained from the aerial parts of plants may have the potential to be an alternative to synthetic pesticides, since they have been demonstrated to possess a wide range of bioactivities against insects and mites. So, the aim of the current study was to investigate the effect of essential oils extracted from three different plants namely: Azadirachta indica Adr. Juss. (Meliaceae), Eucalyptus camaldulensis Dehn. (Myrtaceae) and Laurus nobilis L. (Lauraceae) against A. gossypii. The LC50 values of essential oils of A. indica, E. camaldulensis and L. nobilis against A. gossypii were 1.96, 2.28 and 3.16?μl L?1 air, respectively. This shows that A. indica possesses the highest lethal activity whereas L. nobilis the lowest. These data suggest that essential oils of all the three plants have the potential to be employed in the pest management programmes designed for a control of A. gossypii under greenhouse conditions.  相似文献

昆虫烟碱型乙酰胆碱受体(nicotinic acetylcholine receptor, nAChR)是杀虫剂的重要作用靶标之一。本研究利用简并引物PCR和半巢式PCR技术从麦长管蚜Sitobion avenae (Fabricius)中克隆nAChR基因,成功地获得了5个α型nAChR亚基的cDNA片段。根据5个α亚基片段设计特异引物,结合快速扩增cDNA末端(RACE)技术,成功克隆了5个α型亚基的全长,并发现α5亚基有两种存在形式,它们仅在胞外区有一段175 bp的片段有差异。序列分析发现,这些基因均具有nAChR基因家族的典型特征,并与已报道的其他昆虫的烟碱型乙酰胆碱受体的相应亚基具有很高的同源性。该研究为进一步利用基因表达技术研究昆虫nAChR的天然亚基组成,以及分析麦长管蚜对新烟碱类杀虫剂的靶标抗性,奠定了基础。  相似文献

Helicoverpa armigera has been controlled effectively with chemical insecticides in the major cotton crop production areas of northern Greece for many years. However, a resurgence of the pest was observed in 2010, which significantly affected crop production. During a 4‐year survey (2007 – 2010), we examined the insecticide resistance status of H. armigera populations from two major and representative cotton production areas in northern Greece against seven insecticides (chlorpyrifos, diazinon, methomyl, alpha‐cypermethrin, cypermethrin, gamma‐cyhalothrin and endosulfan). Full dose‐response bioassays on third instar larvae were performed by topical application. Lethal doses at 50% were estimated by probit analysis and resistance factors (RF) were calculated, compared to a susceptible laboratory reference strain. Resistance levels were relatively moderate until 2009, with resistance ratios below 10‐fold for organophosphates and carbamates and up to 16‐fold for the pyrethroid alpha‐cypermethrin. However, resistance rose to 46‐ and 81‐fold for chlorpyrifos and alpha‐cypermethrin, respectively in 2010, when the resurgence of the pest was observed. None of the known pyrethroid resistance mutations were found in the pyrethroid‐resistant insects.
