The aim of the present study was to estimate and compare the growth and survival traits of the hybrids and purebreds produced by crossing the Siberian sturgeon (Acipenser baerii) (S) and sterlet (Acipenser ruthenus) (St) in order to determine the heterosis effect in the F1 generation. We compared the breeding conditions, mean body weight (BW) and cumulative survival in the artificially produced hybrid crosses of sterlet and Siberian sturgeon with respect to their pure parental species in indoor and outdoor aquaculture systems at different developmental stages. Fertilization and hatching rates were found to be significantly higher in S × S purebred compared to St × S hybrid. The highest values of BW were recorded in St × S hybrid (557.54 ± 179.7 g) on 862 days post-hatch (dph) while the highest cumulative survival was recorded in S × S purebred (14.3%). The recorded cumulative survival and mean BW was significantly lower in St × St purebred. The highest positive heterosis was recorded for mean BW of St × S hybrid (51.3% on 862 dph) throughout the sampling points. The studied sturgeon hybrids had higher mean BW compared to St × St purebred under suboptimal rearing conditions. Although there was no clear demonstration of the superiority in performance of reciprocal hybrids over purebreds, the St × S hybrid can be used for achieving better productivity in aquaculture systems.  相似文献

In 2009 through 2011, among 730 individuals of kaluga and Amur sturgeon collected in the lower reaches of the Amur River and the Amursky Liman, 17 morphologically intermediate individuals (hybrids) with the body length of 56 to 202 cm (median, 81 cm) were identified, including 11 individuals (4.6%) found in 2009, three individuals (1.6%) found in 2010, and three individuals (1.1%), in 2011. In 16 hybrids, 819 bp of the mtDNA control regions were sequences and 11 haplotypes were identified. Since all these haplotypes were from the mtDNA lineages of kaluga, it was concluded that hybridization occurred in one direction, kaluga (♀) × Amur sturgeon (♂). This asymmetry could be caused by the large size differences between these species. Since the earlier examined morphologically typical Amur sturgeons showed the absence of alien haplotypes (Shedko et al., 2015), the absence of the mtDNA introgression is claimed. This can be caused by low viability or sterility of the backcross females (kaluga (♀) × Amur sturgeon (♂)) × Amur sturgeon (♂). The samples of hybrids and typical kaluga individuals demonstrated no differences in the frequency spectra of the mtDNA haplotypes. However, haplotype and nucleotide diversity in the first sample was somewhat higher than in the second one (0.950 versus 0.927 and 0.0054 versus 0.0044, respectively). The data obtained will be useful for population monitoring of kaluga and Amur sturgeon, Amur River endemics, which are classified as critically endangered by the IUCN Red List of Threatened Species.  相似文献

Dispermic androgenesis was used to produce, for the first time, an androgenetic progeny of the Siberian sturgeon (Acipenser baerii) and the androgenetic nucleo-cytoplasmic hybrids (Siberian sturgeon, A. baerii × Russian sturgeon, A. gueldenstaedtii) using cryopreserved sperm. Microsatellite DNA analysis confirmed exclusively paternal inheritance in the androgenetic progeny of Siberian sturgeon. Heterozygotes for certain microsatellite loci were detected among the androgenetic hybrids, thereby confirming a dispermic nature of androgenesis. According to the data of comparative morphological analysis, the obtained androgenetic hybrid, by the age of 15 months old, was completely identical to the paternal species. Both a female and a male were detected in the androgenetic sturgeon progenies, which is of interest for producing bisexual progenies via androgenesis. The data of this study confirm the feasibility of dispermic androgenesis using cryopreserved sperm to preserve and recover the gene pools of endangered sturgeon species.  相似文献

The method of polymerase chain reaction with random primers (RAPD PCR) was used to identify the progeny of the crosses between three sturgeon species, Amur sturgeon (Acipenser schrenckii Brandt, 1869), Siberian sturgeon (A. baerii Brandt, 1869), and sterlet (A. ruthenus Linnaeus, 1758). Using ten primers, genetic variation in 70 yearlings, produced in seven individual crosses: Acipenser schrenckii × A. schrenckii, A. baerii × A. baerii, A. ruthenus × A. ruthenus, A. schrenckii × A. baerii, A. baerii × A. schrenckii, A. schrenckii × A. ruthenus, and A. ruthenus × A. schrenckii was described and evaluated. It was demonstrated that the samples composed of hybrids from individual crosses were more variable than the samples of parental species. On the other hand, pooled samples of hybrids from two cross directions were genetically less variable than the pooled samples of their parents. The three main features of the hybrid RAPD profiles identified included: (1) preservation of marker DNA fragments of both parents in one genome; (2) presence of specific DNA fragments, absent from both parents; and (3) dependence of the frequency of some DNA fragments from the cross direction. Multidimensional scaling clearly distinguishes in the space of three coordinates the individuals of original species and the hybrid progeny with differentiation in the groups of direct and backcross hybrids. Analysis of relationships (UPGMA and NJ) pointed to substantial differentiation between the species, as well as between the species and hybrid progeny. Close genetic relationships between direct and backcross hybrids were demonstrated. Multilocus RAPD markers in association with statistical methods are considered to be the useful tool for discrimination of interspecific hybrids of sturgeon. The diet of farm‐escaped sturgeon accidentally introduced into the Yangtze River ecosystem was investigated from samples collected over 10 days. The fish had consumed adequate quantities of natural food, with prey type related to fish body size. The fish were categorized according to their primary dietary component: demersal fish were most common prey of kaluga Huso dauricus and H. dauricus × Acipenser schrenckii hybrids of total length 120.3–188.7 cm. Shrimp Macrobrachium nipponensis made up the major portion of the diet of H. dauricus × A. schrenckii and Acipenser baerii × A. schrenckii of total length 91.1–106.8 cm. Gammarid amphipods were consumed by A. baerii × A. schrenckii of total length 35.0–81.2 cm. The body length of prey was proportional to the fork length of the sturgeon, Y = 0.19X‐9.46, R2 = 0.997. Escaped sturgeon had travelled at least 898 km downstream from the point of escape in the Yangtze River and were feeding on native macrobenthos and demersal fish.  相似文献

Diploid gynogenetic progenies were obtained in the stellate sturgeon Acipenser stellatus, Russian sturgeon, A. gueldenstaedtii, and sterlet A. ruthenus by means of insemination of the eggs with UV-irradiated spermatozoa and suppression of the second meiotic division by heat shock. The gynogenetic nature of experimental fish was confirmed by RAPD-PCR analysis of DNA. Effective photoreactivation of UV-induced lesions of spermatozoa was shown in the case of illumination of the fertilized eggs with visible light. This phenomenon should be taken into account when determining the doses of irradiation that allow inactivation of the male chromosomes and incubating gynogenetic embryos. Gynogenetic stellate and Russian sturgeons are viable and can be reared in order to study the mechanism of sex determination in sturgeons.  相似文献

Amur sturgeon Acipenser schrenckiii Brandt 1869 and kaluga Huso dauricus (Georgi 1775) are critically endangered species with populations showing significant decline from historical levels due to overexploitation, yet little is known about their population structure. Adults are not often captured in the Fuyuan reach of the Amur River, Heilongjiang Province, Northeast China, and the government prioritizes juvenile sturgeon management. This study was conducted to determine the age and length/weight relationships of juvenile Amur sturgeon and kaluga in the Fuyuan reach of the Amur River. We estimated age using pectoral fin spine sections obtained from 65 juvenile Amur sturgeon and 50 juvenile kaluga. We compared the age estimates from two readers, and found 100% between‐reader agreement in 67.7% of the Amur sturgeon and 64.0% of the kaluga. The majority of differences in estimated age were within 1 year. Length/weight relationships (LWR) were calculated, and the LWR of the Amur sturgeon and kaluga were W = 0.0025L3.106 (r2 = 0.966)and W = 0.0022L3.175 (r2 = 0.989), respectively. Our study provides the age structure and LWR in juveniles of two sturgeon species.  相似文献

An unusually high frequency of spontaneous diploidization in a maternal chromosome set (SDM) was discovered in one ornamental carp (koi) female in an experiment on these fish for induced gynogenesis. Spontaneous appearance of diploid embryos in the gynogenetic offspring (intact eggs × irradiated sperm) and appearance of triploids among control fish (intact eggs × intact sperm) obtained from koi female and males of edible carp indicated spontaneous diploidization of maternal chromosomes (SDM). Possible cytological processes causing this phenomenon are discussed.  相似文献

The results are presented on haploid androgenesis in Siberian sturgeon and sterlet induced by UV irradiation of ovicells. During irradiation, the cells in Ringer solution were rotated around a UV lamp. The efficiency of genetic inactivation of ovicells was estimated by the following parameters: manifestation of Hertwig effect, the fraction of embryos demonstrating haploid syndrome at final developmental stages, by arrest of embryonic development in hybrids Siberian sturgeon × great sturgeon, and by absence of maternal alleles of microsatellite loci in embryos. The dose-effect curve suggests that, during UV irradiation of ovicells of Siberian sturgeon, the complete genetic inactivation is attained at exposition of 120 s, while that in sterlet is 90 or 105 s. The results show a principal possibility of inactivation of ovicells by UV irradiation and use of such cells for producing androgenetic progeny of acipenserids.  相似文献

This study investigated the gynogenetic cytobiological behavior of the third gynogenetic generation (G3), which was generated from the diploid eggs produced by the second gynogenetic generation (G2)of red crucian carp × common carp, and determined the chromosomal numbers of G3, G2×scatter scale carp and G2×allotetraploid hybrids of red crucian carp × common carp. The results showed that the diploid eggs of G2 with 100 chromosomes, activated by UV-irradiated sperm from scatter scale carp and without the treatment for doubling the chromosomes, could develop into G3 with 100 chromosomes.Similar to the first and second gynogenetic generations (G1 and G2), G3 was also diploid (2n=100) and presented the hybrid traits. The triploids (3n=150) and tetraploids (4n=200) were produced by crossing G2 with scatter scale carp, and crossing G2 with allotetraploids, respectively. The extrusion of the second polar body in the eggs of G2 ruled out the possibility that the retention of the second polar body led to the formation of the diploid eggs. In addition, we discussed the mechanism of the formation of the diploid eggs generated by G2. The establishment of the diploid gynogenesis clonal line (G1, G2 and G3) provided the evidence that the diploid eggs were able to develop into a new diploid hybrid clonal line by gynogenesis. By producing the diploid eggs as a unique reproductive way, the diploid gynogenetic progeny of allotetrapioid hybrids of red crucian carp × common carp had important significances in both biological evolution and production application.  相似文献

The karyology of species of sturgeon from the Russian Far East demonstrates that the karyotype of the Sakhalin sturgeon (Acipenser mikadoi) includes 262 ± 4 chromosomes with 80 biarmed chromosomes and the number of chromosome arms (NF) 342 ± 4, the karyotype of the Amur sturgeon (A. schrenckii) includes 266 ± 4 chromosomes with 92 biarmed chromosomes and NF 358 ± 4, and the karyotype of the kaluga (A. dauricus) consists of 268 ± 4 chromosomes with 100 biarmed chromosomes and NF 368 ± 4. These results prove that all western Pacific sturgeon species are from a tetraploid origin, based on a recent ploidy scale. This suggests that at least three polyploidization events have occurred during the evolution of Acipenseridae. However, if polyploid species originated by hybridization between diploid species, there may have been more polyploidization events in this group of fishes.  相似文献

Sturgeons were cultured at the Solombalsky cellulose and paper plant (CPP) in Arkhangelsk over a 3 year period. The warm-water effluents of the biological treatment unit were utilized to rear the Siberian sturgeon (Acipenser baerii) and its hybrids with the sterlet (Acipenser ruthenus). Another culture trial included the Russian sturgeon (Acipenser gueldenstädtii) and its hybrids with the beluga (Huso huso). The most profitable culture (in terms of growth rates) was with the Siberian sturgeon and its hybrids with the sterlet. After the second year of culture, Siberian sturgeon and its hybrid averaged about 2.3 kg in wet weight while Russian sturgeons gained less (1.5 kg). One- and two-year old sturgeons grown at the Solombalsky CPP met practically all requirements needed for a successful culture.  相似文献

Hybridization experiments between Haemonchus contortus from sheep and H. placei from cattle indicate that the H. placei vulvar morph type and the inability of the eggs to hatch and develop at 11°C were inherited as dominant traits in the hybrids. The size of the third stage larvae was similar to H. placei in hybrids from the mating of male H. placei × female H. contortus while larvae from the reciprocal mating were intermediate in size. Hybrids produced by the mating of thiabendazole resistant female H. contortus × non-resistant male H. placei were also resistant. The F1 males of the mating between male H. contortus and female H. placei were sterile. Male sterility did not occur in the reciprocal cross until the F2 generation. Female hybrids from these generations had a low level of fertility when backcrossed to males of either parent species. Cytological studies of the hybrid males indicated that sterility was due to several kinds of meiotic disturbance and that spermatogenesis stopped during metaphase I. The chromosomes in eggs of unfertilized females did not undergo meiosis and polar body formation; instead they increased in number by a process of endomitosis. Unfertilized eggs in the faeces were characterized by uneven cytoplasmic division and abnormal shape. It is proposed that hybrid sterility can lead to the local eradication of one species of Haemonchus by the other. Furthermore, the outcome of which species is eradicated can be influenced by such variables as initial population sizes, climate, and the local ratio of sheep to cattle.  相似文献

Blue mussels of the genus Mytilus form extensive hybrid zones in the North Atlantic and elsewhere where the distributions of different species overlap. Mytilus species transmit both maternal and paternal mtDNA through egg and sperm, respectively, a process known as doubly uniparental inheritance (DUI), and some females produce offspring with extremely biased sex ratios. These two traits have been shown to be linked and maternally controlled, with sex determination involving nuclear–cytoplasmic interactions. Hybridization has been shown to disrupt DUI mitochondrial inheritance and sex ratio bias; however, the effect of hybridization on reproductive fitness has not previously been examined. We investigated this effect in M. edulis × M. trossulus crosses through histological examination of mature F1 progeny, and spawning of F1 hybrids to monitor survival of their progeny through to the D stage of larval development. For progeny produced from mothers with a strong bias toward female offspring (often 100%) in pure-bred crosses, there was a clear breakdown in female dominance of progeny and significantly more hermaphrodites in the hybrid crosses produced from sperm with the M-tr1 mitotype. We also found significant sex-specific differences among hybrid progeny, with females producing normal eggs while males and hermaphrodites evidenced impaired gonadal development with significantly greater numbers of Sertoli cells, phagocytic hemocytes, and degenerating germ cells, all associated with gonad resorption. Males from crosses where DUI was disrupted and where male progeny were homoplasmic for the female mtDNA were the most severely compromised. Allelic incongruity between maternal and paternal mitotypes in hybrid crosses was associated with significant disruption of male gonadal development.  相似文献

The efficacy of ploidy analysis for separating progeny of Siberian sturgeon Acipenser baerii after induced gynogenesis was demonstrated using sperm of a paternal species differing in ploidy level from the maternal species. Gynogenesis was induced in tetraploid A. baerii with UV‐C irradiated sperm from the diploid sterlet Acipenser ruthenus and vice‐versa. The success of sperm UV irradiation and diploidy restoration by heat‐shock was estimated based on the ploidy level of progeny, confirmed by microsatellite parentage assignment. Hatching rates of interspecific gynogenotes were comparable with rates reported for gynogenesis induction using sperm and eggs of the same species. Juvenile mortality was similar to that observed in the control hybrids. The efficiency and reliability of this method may foster its use for production of gynogenotes in aquaculture, potentially allowing interspecific gynogenesis to replace intraspecific.  相似文献

The aim of this study was to further test the viability of the roach Rutilus rutilus × common bream Abramis brama hybrid in terms of reproductive behaviour and sexual production. Egg release, mating and aggressive acts in reproductive behaviour, as well as absolute fecundity and sperm density in milt for sexual production were examined in the first generation of these hybrids at their first sexual maturity. The F2 and backcrosses of hybrids were also studied. The results revealed that these hybrids expressed a normal and typical mating behaviour, producing viable gametes. Under experimental reproduction between hybrids (hybrid reproduction), the number of egg‐release acts (range, 21–66) was nearer (χ2 test, P > 0.05) the number of mating acts (11–65). Moreover, hybrid males exhibited territorial and aggressive behaviours. However, in experimental reproduction of female and male hybrids mixed with parental males (mixed reproduction), the egg‐release act and the mating act were inhibited by the intense territorial and aggressive activities of the common bream male. Absolute fecundity values (median, <2.2 × 103 eggs) and sperm density (<7 × 109 spermatozoa ml?1) of hybrids showed a greater decrease (U test, P < 0.05) than in parental species (median, >6.0 × 103 eggs and >14 × 109 spermatozoa ml?1, respectively). Nevertheless, these hybrids were fertile. F2 and backcross generations were produced, although with a significantly lower viable hatching rate (FEP test, P < 0.05) in F2 individuals from the female and its corresponding hybrid male (<6%), indicating a very low chance of survival in rivers.  相似文献

The genetic basis of male and female sterility in hybrids of Drosophila pseudoobscura-Drosophila persimilis was studied using backcross analysis. Previous studies indirectly assessed male fertility by measuring testis size; these studies concluded that male sterility results from an X chromosome-autosome imbalance. By directly scoring for the production of motile sperm, male sterility is shown to be largely due to an incompatibility between genes on the X and Y chromosomes of these two species. These species have diverged at a minimum of nine loci affecting hybrid male fertility. Semisterility of hybrid females appears to result from an X chromosome-cytoplasm interaction; the X chromosome thus has the largest effect on sterility in both male and female hybrids. This is apparently the first analysis of the genetic basis of female sterility, or of sterility/inviability affecting both sexes, in an animal hybridization.  相似文献

Clonal full-sib progeny groups of Atlantic salmon Salmo salar × brown trout Salmo trutta hybrids were produced by gynogenesis. Eggs obtained from two 3-year-old Atlantic salmon (female) × brown trout (male) F1 hybrids were activated with UV-irradiated rainbow trout Oncorhynchus mykiss sperm. Fecundity, percentage egg activation and percentage survival to completion of yolk-sac absorption were similar for the two females, and averaged 800 eggs kg−1, 90 and 65%, respectively. Flow cytometric and protein electrophoretic analyses confirmed the progeny to be diploid hybrids. Isogenicity within progeny groups and to the maternal parent was indicated by identical DNA fingerprint patterns detected with multilocus oligonucleotide probes–GATA(5) and ACTG(n). Isogenicity was also observed in the gynogenetic progeny of a third female spawned the following year. It appeared that a large portion of the oocytes in females of this hybrid underwent a premeiotic chromosome doubling, or possibly a complete suppression of meiosis. The result was ovulation of diploid eggs, each possessing a full set of both Atlantic salmon and brown trout chromosomes identical to those in the maternal somatic cells. Lines of clonal hybrids could therefore be perpetuated by gynogenesis and would have potential both as experimental animals and in commercial aquaculture.  相似文献
