土壤中蕴藏着高度的微生物多样性,在陆地生态系统中发挥着非常重要的功能,加强对土壤微生物资源的综合管理与开发应用是提升生态系统稳定性与生产力及农产品质量的重要途径。首先,土壤微生物多样性具有全球性的重大意义,有待完善对土壤微生物的检测与监测技术研究,进而实现土壤微生物多样性与土壤功能的耦合以及对土壤质量的评定;其次,土壤微生物作为一种宝贵的生产资料和可持续资源,要加强其在土壤肥力强化与保育、土壤障碍消减与调节、土壤污染控制与修复等3个领域的应用研究。最后,未来土壤微生物学发展将会形成土壤微生物系统学、土壤微生物过程学与土壤微生物功能学3个子学科,要建立土壤微生物种质资源库与遗传信息库,推进土壤微生物生理代谢过程、生物化学过程及生态行为过程的研究,联结土壤微生物与土壤功能的关系,并从土壤中的功能微生物出发对环境变化作出积极响应和主动调控。此外,原创性方法的建立与应用是限制土壤微生物学发展的技术瓶颈,联合生物地理学与生物信息学破译重要基因的特定生态功能,并将其应用到生态模型以及生态系统未知领域的研究中去,是土壤微生物学面临的挑战。  相似文献

Microbiological and biogeochemical investigations of the coastal zone and the littoral of the Kandalaksha Bay of the White Sea were carried out. The material for investigations was obtained in the series of expeditions of the Institute of Microbiology, Russian Academy of Sciences, in August 1999, 2000, 2001, and in March 2003. The studies were conducted on the littoral and in the water area of the Kandalaksha Preserve, the Moscow University Belomorsk Biological Station, and the Zoological Institute Biological Station, Russian Academy of Sciences. Sediment sampling on the littoral was carried out in the typical microlandscapes differing in the sediment properties and macrobenthos distribution. The maximal sulfate reduction rate (SRR) was shown for the shallow part of the Chernorechenskaya Bay (up to 2550 g S/(dm3 day)) and in the Bab'ye More Bay (up to 3191 g S/(dm3 day)). During the winter season, at a temperature of –0.5 × 0.5°C, the SRR in the sediments of the Kartesh Bay was 7.9 × 13 g S/(dm3 day). In the widest limits, the SRR values varied in the sediment cores sampled on the littoral. The minimal values (11 g S/(dm3 day)) were obtained in the core samples on the silt–sandy littoral. The littoral finely dispersed sediments rich in organic matter were characterized by high SRR values (524–1413 g S/(dm3 day)). The maximal SRR values were shown for the sediments present within the stretch of decomposing macrophytes, in local pits at the lower littoral waterline, and in the mouth of a freshwater stream (51–159 mg S/(dm3 day)). A sharp difference in the level of H2S production in the type microlandscapes was shown. The average hydrogen sulfide production in finely dispersed sediments constituted 125 mg S/(m2 day); in stormy discharge deposits, 1950 mg S/(m2 day); in depressions under stones and in silted pits, 4300 mg S/(m2 day). A calculation made with regard to the area of microlandscapes with increased productivity shows that the daily H2S production per 1 km2 of the littoral (August) is 60.8 to 202 kg S/(km2 day), while the organic carbon consumption for sulfate reduction per 1 km2 of the littoral is 46 to 152 kg Corg/(km2 day).  相似文献

Over the last 15 years, it has become increasingly apparent that a single class of compounds, the acylated homoserine lactones (AHLs), elicit effects on many levels of biological and ecological organization. Despite the fact that the distribution of AHL production in the prokaryotic phylogenetic tree is restricted to a small set of genera, representatives of these genera are abundant in the environment and are responsible for processes of much interest to humans. As well as driving interactions between clones, AHLs have been shown to mediate interactions between different species of bacteria and between bacteria and higher organisms, either through the phenotypes they regulate or directly through their own chemical behaviour. Understanding the biological activity of AHLs and the ecological consequences of these activities may provide us with an opportunity to manipulate the composition and function of complex biological assemblages. Ultimately, this broadens the biotechnological focus of AHL-based research beyond the attenuation of virulence in humans and plant pathogens.  相似文献

Bacteriocins are the most abundant and diverse defense systems in bacteria. As a result of the specific mechanisms of bacteriocin recognition and translocation into the target cell it is assumed that these toxins mediate intra-specific or population-level interactions. However, no published studies specifically address this question. We present here a survey of bacteriocin production in a collection of enteric bacteria isolated from wild mammals in Australia. A subset of the bacteriocin-producing strains was assayed for the ability to kill a broad range of enteric bacteria from the same bacterial collection. A novel method of estimating killing breadth was developed and used to compare the surveyed bacteriocins in terms of the phylogenetic range over which they kill. The most striking result is that although bacteriocin-producers kill members of their own species most frequently, some kill phylogenetically distant taxa more frequently than they kill closer relatives. This study calls into question the role these toxins play in natural populations. A significant number of bacteriocins are highly effective in killing inter-specific strains and thus bacteriocins may serve to mediate bacterial community interactions.  相似文献

Rhododendron aureum Georgi is a perennial evergreen dwarf shrub that grows at all elevations within the alpine tundra of northern China. Previous research has investigated the plant communities of R. aureum; however, little information is available regarding interspecific competition and underground soil microbial community composition. The objective of our study was to determine whether the presence of R. aureum creates a unique soil microbiome and to investigate the relationship between R. aureum and other plant species. Our study site ranged from 1,800 to 2,600 m above sea level on the northern slope of the Changbai Mountain. The results show that the soil from sites with an R. aureum community had a higher abundance of nitrogen‐fixing bacteria and a higher resistance to pathogens than soils from sites without R. aureum. We emphasize that R. aureum promotes a unique soil microbial community structure that is distinct from those associated with other plants. Elevation and microbial biomass were the main influencing factors for plant community structure. Analysis of interspecific relationships reveals that R. aureum is negatively associated with most other dominant shrubs and herbs, suggesting interspecific competition. It is necessary to focus on other dominant species if protection and restoration of the R. aureum competition is to occur. In the future, more is needed to prove whether R. aureum decreases species diversity in the tundra ecosystems of Changbai Mountain.  相似文献

The effect of diets varying in carbohydrate and protein content on the structure and function of the hindgut microbiota of crickets was evaluated by determining bacterial densities, fermentation activity, and guanine plus cytosine (G + C) profiles of the DNA extracted from the microbial hindgut community. DNA isolated from the gut community was fractionated and quantified according to G + C content as a comprehensive, coarse-level measure of the composition and structure of the community. The bacterial densities measured by direct counts were not significantly different among the four diets. The crickets were initially reared in the laboratory on cricket chow, which resulted in a hindgut community dominated by bacteria with a G + C content between 32% and 57%. Crickets shifted to an alfalfa diet showed a similar hindgut community G + C profile, although microbial populations with DNA between 35% and 45% G + C were more abundant in alfalfa- than chow-fed crickets. The apparent complexity of the gut community was reduced in crickets fed beet-pulp and protein-based diets compared to those fed chow and alfalfa, and was dominated by populations with a low percentage G + C content. Hindgut communities in crickets fed pulp and protein diets also showed a decrease in hydrogen and carbon dioxide production, suggesting that these diets affected the biochemical activity of the hindgut community. The protein-based diet resulted in a decrease in the rate of evolution of volatile fatty acids, while the ratio of butyrate production to acetate and propionate production was significantly higher in these crickets. Our results show the emergence of a new microbial community structure concomitant with changes in microbial biochemical activity due to shifts in the cricket's dietary regime.  相似文献

单一微生物降解水稻秸秆效果不明显,多种微生物组合在一起的微生物菌群能够有效降解水稻秸秆,是当前降解秸秆类废物的首要选择。【目的】探究微生物菌群比单一菌株转化秸秆效率提高的种间协作机制,为改善秸秆类物质的生物降解过程提供理论参考。【方法】通过菌种组合,以水稻秸秆减重率为指标人工构建出降解效果优于单菌培养的微生物群体组合,利用分子生物学方法对实验菌株进行鉴定,结合纤维素酶活性、发酵产物GC-MS分析等指标得出水稻秸秆降解时微生物群体的种间协作机制。【结果】菌株B (Bacillus cereus)在30℃培养8 d水稻秸秆降解率为50.9%,菌株B与菌株D_2(Bacillus amyloliquefaciens)、W_1(Ochrobactrum intermedium)、G_1降解(Bacillus licheniformis)组合构成4株菌BD_2W_1G_1的复合菌群,在30℃培养8 d水稻秸秆降解率为73.3%,比B单菌株分解能力提高22.4%。发酵产物分析结果表明,菌株B单独培养产生大量酸、酚等物质。B+D_2联合培养,发酵液内酸类相对减少87.4%,酚类相对减少61.9%(十五烷酸、正十六烷酸、2,4二叔丁基酚和6-叔丁基对甲酚减少明显),水稻秸秆降解率为64.6%。当B+D_2+W_1联合培养,发酵液内酚类进一步减少15.7%(6-叔丁基对甲酚减少明显),水稻秸秆降解率为71.0%。当B+D_2+W_1+G_1联合培养,酚类继续减少10.7%,水稻秸秆降解率为73.3%。【结论】十五烷酸、正十六烷酸、2,4二叔丁基酚和6-叔丁基对甲酚等酸、酚类物质会抑制菌株B分解水稻秸秆的效率,菌株D_2、W_1、G_1能够减少包括4种物质在内的酸、酚类含量(酸类共减少82.9%,酚类共减少88.2%),来提高分解效率(共提升22.4%的分解效果)。水稻秸秆成分复杂,具有不同功能的菌种组合成复合菌剂,减少反馈抑制形成代谢互补,能有效提高水稻秸秆生物降解效果。  相似文献

为探明不同施肥处理对早稻和晚稻各个生育时期稻田土壤微生物生物量碳、氮和微生物熵的影响,以湖南宁乡长期定位试验为平台,应用氯仿熏蒸-K_2SO_4提取法和化学分析法系统分析了定位长达29年5种施肥处理之间(化肥、秸秆还田+化肥、30%有机肥+70%化肥、60%有机肥+40%化肥和无肥)双季稻田土壤微生物生物量碳、氮和微生物熵的差异。结果表明,早稻和晚稻各主要生育时期,长期施肥均能提高土壤微生物生物量碳、氮含量和微生物熵,各施肥处理土壤微生物生物量碳、氮含量和微生物熵均随水稻生育期推进呈先增加后降低的变化趋势,均于齐穗期达到最大值,成熟期达到最低值;其中,以60%有机肥和30%有机肥处理双季稻田土壤微生物生物量碳、氮含量和微生物熵均为最高,均显著高于其他处理,其大小顺序表现为60%有机肥30%有机肥秸秆还田化肥无肥。长期有机无机配施可以提高土壤微生物生物量碳、氮和微生物熵,有机肥与化肥配施对提高土壤肥力效果最好。土壤微生物生物量碳、氮及微生物熵可以反映土壤质量的变化,可作为评价土壤肥力的生物学指标。  相似文献

One hundred microorganisms have been screened for their abilities to selectively modify the structure of the sesquiterpene lactone known as quadrone. The only products obtained were those formed when the 4-ketone functional group was reduced to the stereoisometric 4-quadronols. Quadrone alcohol isomers of (S) or (R) absolute configurations were identified by proton and carbon n.m.r., and high performance liquid chromatography (h.p.l.c.) was used to separate and quantitate these compounds in extracts of fermentations. Microorganisms were categorized according to their abilities to achieve Re- or Si-face carbonyl reduction to yield (S)- or (R)-alcohol isomers by h.p.l.c. Three groups of microorganisms were identified: those yielding only the (R)-alcohol isomer; those yielding only the (S)-alcohol isomer; and those providing mixtures of the two alcohol isomers. With quadrone as substrate, Mucor and Curvularia spp. may contain either Re- or Si-face reductases. The selection of microorganisms for their abilities to achieve enantiospecific reductions of ketones to alcohol products is discussed.  相似文献

微生物总活性是指在某一时段内微生物所有生命活动的总和,它直接决定着微生物行使生理、生态功能的能力,是微生物学研究的热点,也是难点。迄今为止,还没有建立直接测定微生物总活性的方法,只能用一些相关指标来间接反映它。目前常用的指标主要包括微生物的呼吸速率、生长速率以及胞内RNA含量等。与其它一些基质和环境相比,测定土壤中的微生物总活性更为困难。通过总结研究土壤微生物总活性常用的3种方法,在简略概括传统的土壤微生物呼吸测定法的基础上,详细介绍了放射性同位素标记法和RNA直接表征法的原理和操作流程,整理归纳了一些重要应用案例,比较分析了不同方法的优缺点,以期为选择研究土壤微生物总活性的适宜方法提供依据。  相似文献

植物的叶际是一个复杂的生态系统,微生物的生存环境条件严苛。其可被利用的营养成分较少,温湿度波动大。此外,较强的紫外线辐射对于叶际微生物的生存也有很大影响。但是植物叶际却有着丰富的微生物多样性,其中还有许多有益细菌和真菌。它们通过和植物寄主的互作,改善着叶际微生物的栖居环境;其对植物病原体的拮抗亦可提高植物的抗病性。植物叶际的微生物还可以产生激素以促进植物生长,还有一些微生物可以利用农药等污染有机物作为营养物质,在污染物的环境生物修复方面显示巨大的潜力。此外,叶际微生物作为一种生态学指标在生态稳定与环境安全评价中开始发挥显著的作用。  相似文献

The impact of urine on the microbial biomass, activity and community structure was compared in the soil beneath two pastures in the Scottish uplands; Fasset, a natural Agrostis capillaris–Festuca ovina–Galium saxatile grassland and Strathfinella, a semi-natural grassland, improved with fertiliser addition. Community level physiological profiles (CLPP) were used to characterise the microbial communities. The utilisation of sugars, oligosaccharides, alcohols, carboxylic acids, long chain aliphatic acids, acidic, basic and neutral amino acids, amide N, phenolic acids and long chain aliphatic acids was used to compare the soils and the impact of synthetic urine addition. In the untreated soils, the utilisation of all the substrates decreased from the first week in May through to October. Averaged over all times and urine treatment, the potential utilisation of all substrates except for phenolic acids, long chain aliphatic acids and carboxylic acids was greater in the improved and more intensively grazed Strathfinella site. When averaged over all sample times, urine increased the utilisation of sugars, oligosaccharides, basic amino acids and amide N and the increases were greater in the unimproved, less intensively grazed, Fasset soil than that at Strathfinella. The effect of urine tended to be greatest during the period between 2 and 5 weeks after urine addition when utilisation of alcohols, acidic and neutral amino acids was also increased. Microbial biomass C in the control soils was 155.9 and 112.7 g C m−2 at Fasset and Strathfinella, respectively. Values did not change significantly with time and were unchanged by the addition of urine. However, urine addition significantly increased basal respiration rates at Fasset and decreased them at Strathfinella. Urine also increased bacterial numbers in both soils, but had no consistent effect on fungi or yeasts. The significance of these findings for studies of soil microbial community structure and activity in grazed upland grasslands is discussed. Anaerobacter polyendosporuscells do not have typical mesosomes. However, the analysis of this anaerobic multispore bacterium by electron microscopic cryofractography showed that its cytoplasmic membrane contains specific intramembrane structures in the form of flat lamellar inverted lipid membranes tenths of nanometers to several microns in size. It was found that these structures are located in the hydrophobic interior between the outer and inner leaflets of the cytoplasmic membrane and do not contain intramembrane particles that are commonly present on freeze-fracture replicas. The flat inverted lipid membranes were revealed in bacterial cells cultivated under normal growth conditions, indicating the existence of a complex-type compartmentalization in biological membranes, which manifests itself in the formation of intramembrane compartments having the appearance of vesicles and inverted lipid membranes.  相似文献

土壤微生物作为陆地生态系统的重要组成部分,直接或间接地参与几乎所有的土壤生态过程,在物质循环、能量转换以及污染物降解等过程中都发挥着重要作用。对土壤微生物时空演变规律及其形成机制的研究,不仅是微生物演变和进化的基础科学问题,也是预测微生物及其所介导的生态功能对环境条件变化响应、适应和反馈的理论依据。讨论了土壤微生物群落的定义、测度方法和指标,认为群落是联系动植物宏观生态学与微生物生态学的基础,群落构建机制是宏观和微观生态学都需要研究的核心科学问题;从生态学的群落构建理论出发,阐述了包括生态位理论/中性理论、过程理论和多样性-稳定性理论在土壤微生物时空演变研究中的应用,以及微生物群落在时间和空间上的分布特征及其尺度效应;确立了以微生物群落构建理论为基础、不同时空尺度下土壤微生物群落演变特征为主要内容的微生物演变研究的基本框架。  相似文献
