The primary sequences of the myoglobins of two rodents (the South American viscacha and the Mediterranean mole rat) have been determined. Both myoglobins exhibit one polymorphism. The two rodent sequences have been compared with each other and with other known myoglobins. The myoglobin of the viscacha is similar to those of the diving mammals and penguin in having a high arginine content. Among mammalian sequences, the arginines at positions 77 (in one of the viscacha myoglobins) and 79 have been found only in the myoglobin from viscacha. Mole rat myoglobin has a lysine at position 31, where arginine or serine is found in all other known vertebrate myoglobins. 相似文献

Summary ECG of mole rats (Spalax ehrenbergi) was recorded by chronically implanted electrodes. The average heart rate of unrestrained, resting animals (mean body mass 191 g±35 S.D.) in normoxia and at room temperature is 152 beats/min±42 S.D. It is nonrhythmic and about one third of the rate expected for an animal of this mass. ECG revealed that each heart beat is normal. From atropine and propranolol administration, it was evident that the low heart rate results from : (a) low intrinsic heart rate (285 b/min±30 S.D.), (b) high parasympathetic tone (51%±12 S.D.) and (c) low sympathetic tone (3.6%±1.6 S.D.). Unilateral vagotomy showed that the degree of left or right vagus dominancy in the mole rat differs in each individual: it may even reach a complete left vagus control, in contrast to other mammals where right vagus dominancy is apparent. 相似文献

Chromatographic separation of the non-heme proteins from the erythrocytes of the subterranean mole rat belonging to the superspecies Spalax ehrenbergi from Israel revealed two major peaks. On sequence analyses, the larger peak corresponded to a 56 kDa selenium-binding protein (SeBP) previously characterized from mouse and human liver, and the second peak to the low-activity carbonic anhydrase (CA) isozyme, CA I. There was no evidence of the high-activity CA II isozyme normally found in the red cells of all amniotes tested to date. Thus, the mole rat appears to be the first mammalian species to express both a SeBP and the low-activity CA I isozyme, as the major non-heme proteins in its red blood cells. It is possible that the absence of the high-activity CA II isozyme may be advantageous to the mole rat in adapting to the low O2 and high CO2 environment of its underground burrows. It is also likely that the 56 kDa SeBP may play an important adaptive role in the physiology of the red cell. 相似文献

The complete amino acid sequence of myoglobin from the triturative stomach of gastropodic molluscBursatella leachii has been determined. It is composed of 146 amino acid residues, is acetylated at the N-terminus, and contains a single histidine residue at position 95 which corresponds to the heme-binding proximal histidine. The E7 distal histidine, which is conserved widely in myoglobins and hemoglobins, is replaced by valine inBursatella myoglobin. The amino acid sequence ofBursatella myoglobin shows strong homology (73–84%) with those ofAplysia andDolabella myoglobins. 相似文献

Factors influencing the distribution of the Ghana mole rat Cryptomys zechi were investigated in a small part of the middle Volta basin of Ghana where it is endemic. Soil type appeared to be the most important factor controlling mole rat distribution in the area; the mole rats preferred areas where the soil was silt loam. In areas where mole rats were found, local distribution was influenced by food availability and land preparation methods for farming. The highest concentrations of mole rat colonies were found in farmlands where traditional hoe ploughing is used for land preparation and where plants with underground storage organs were more diverse; the lowest concentration was in farms where mechanized ploughing is used for land preparation. 相似文献

The amino acid sequences of the myoglobins of two rodents, the casiragua and the house mouse, have been determined. The myoglobin of casiragua differs from that of viscacha (another hystricomorph) at 6 positions. Mouse myoglobin differs from that of mole-rat (another myomorph) at 17 positions, whereas casiragua and mouse differ at 22 positions. Mouse myoglobin possesses several features unique among all known myoglobins (Gly 31, Cys 66, Thr 74 and Glu 113) and one substitution unique among known mammalian myoglobins (Glu 53).

Optimal functioning of the olfactory system is critical for survival of fossorial rodents in their subterranean lifestyle. This study examines the structure of the olfactory mucosa and olfactory bulb of two fossorial rodents exhibiting distinct social behaviors, the East African root rat and the naked mole rat. The social naked mole rat displayed simpler ethmoturbinates consisting of dorsomedial and broad discoid/flaplike parts that projected rostrally from the ethmoid bone. In the solitary root rat however, the ethmoturbinates were highly complex and exhibited elaborate branching which greatly increased the olfactory surface area. In addition, when correlated with the whole brain, the volume of the olfactory bulbs was greater in the root rat (4.24 × 10?2) than in the naked mole rat (3.92 × 10?2). Results of this study suggest that the olfactory system of the root rat is better specialized than that of the naked mole rat indicating a higher level of dependence on this system since it leads a solitary life. The naked mole rat to the contrary may have compensated for its relatively inferior olfactory system by living in groups in a social system. These findings demonstrate that structure of the olfactory system of fossorial mammals is dictated by both behavior and habitat. 相似文献

The Cape mole rat Georychus capensis is a solitary mole rat that inhabits the winter rainfall region of the Western Cape Province of South Africa. Circulating basal concentrations of luteinizing hormone (LH) were found to be significantly higher in the breeding season in both sexes. During both the breeding and non-breeding season, administration of exogenous gonadotropin-releasing hormone (GnRH) increased circulatory LH levels. The magnitude of the LH response to an overdose of exogenous GnRH both in and out of the breeding season in males and females was not significantly different. Typically, seasonally breeding species exhibit a down-regulation of the pituitary and reproductive functions out of the breeding season. It appears that there is no down-regulation of GnRH receptors at the level of the pituitary out of the breeding season, because the pituitary responds to an exogenous GnRH challenge equally both in and out of the breeding period. The Cape mole rat exhibits the potential for opportunistic breeding out of the breeding period, provided that environmental factors are favourable. This finding questions whether this mole rat is actually a seasonal breeder or whether reproduction is hindered by the ecological constraint of the lack of opportunities to burrow and find mates at certain times of the year. 相似文献

The cDNA for the unusual 41 kD myoglobin of the abaloneNordotis madaka was amplified by polymerase chain reaction (PCR), and the cDNA-derived amino acid sequence of 378 residues was determined. As with the myoglobin of the related abaloneSulculus diversicolor (Suzuki and Takagi,J. Mol. Biol.
228, 698–700, 1992), the sequence ofNordotis myoglobin showed no significant homology with any other globins, but showed high homology (35% identity) with vertebrate indoleamine 2,3-dioxygenase, a tryptophan degrading enzyme containing heme. The amino acid sequence homology betweenNordotis andSulculus myoglobins was 87%. These results support our previous idea that the abalone myoglobins evolved from a gene for indoleamine dioxygenase, but not from a globin gene, and therefore all of the hemoglobins and myoglobins are not homologous. The hemoglobin of the sea snakeMicrocephalophis gracilis was purified and the primary structure of the and chains determined. This is the first sea snake hemoglobin structure characterized, and apparently also the first complete structure of any snake hemoglobin (an chain of a viper was known), allowing judgments of reptilian variants. Variations between the sea snake form and other reptilian forms are large (52–65 differences for the chains), of similar order as those between the sea snake and avian (56–65 differences) or human (58 differences) forms. Functionally, 19 residues at / contact areas and 7 at heme contacts are exchanged in relation to the human and chains. Four positions of the sea snake hemoglobin contain residues thus far unique to this form. However, all replacements appear compatible with conserved overall functional properties. 相似文献

In a previous study the alteration in the amino acid sequence of Neurospora crassa NADP-specific glutamate dehydrogenase (GDH) resulting from two mutually compensating frameshift mutations was used to deduce the first 17 nucleotides of the coding sequence of the am gene. In the work reported here, a synthetic 17-mer corresponding to the deduced sequence was shown to hybridize strongly to a 9-kb HindIII fragment from N. crassa wild-type DNA but not to any corresponding fragment from the DNA of a mutant strain known to be deleted for most or all of the gene. Wild-type HindIII fragments were fractionated for size and a fraction centering around 9 kb was cloned in vector λL47. Two clones carrying the strongly hybridizing fragment were identified. The hybridization to the 17-mer was localized within a 2.7-kb BamHI fragment and, within this, to a 700-bp BamHI-BglII subfragment. 5' end-labelled polyadenylated RNA isolated from wild-type mycelium hybridized to the 2.7-kb BamHI fragment and not appreciably to flanking fragments. The partial sequence analysis of the BamHI-BglII fragment has confirmed that the 17-mer probe matches the coding sequence at the 5' end of the gene and has also revealed an intervening sequence 67 bp in length, interrupting codon 15. Both the 9-kb HindIII fragment and the 2.7-kb BamHI fragment have been shown to be capable of transforming the deletion mutant to prototrophy and ability to produce GDH. Analysis of one transformant showed that the am gene was integrated, together with a part of the long arm of the lambda vector, at an unusual locus. This transformant, in which the am gene does not show its normal linkage to the linkage group 5 marker inl, was found to produce GDH to about 20% of the normal level. 相似文献

Myoglobin was isolated from the radular muscle of the chitonLiolophura japonica, a primitive archigastropodic mollusc.Liolophura contains three monomeric myoglobins (I, II, and III), and the complete amino acid sequence of myoglobin I has been determined. It is composed of 145 amino acid residues, and the molecular mass was calculated to be 16,070 D. The E7 distal histidine, which is replaced by valine or glutamine in several molluscan globins, is conserved inLiolophura myoglobin. The autoxidation rate at physiological conditions indicated thatLiolophura oxymyoglobin is fairly stable when compared with other molluscan myoglobins. The amino acid sequence ofLiolophura myoglobin shows low homology (11–21%) with molluscan dimeric myoglobins and hemoglobins, but shows higher homology (26–29%) with monomeric myoglobins from the gastropodic molluscsAplysia, Dolabella, andBursatella. A phylogenetic tree was constructed from 19 molluscan globin sequences. The tree separated them into two distinct clusters, a cluster for muscle myoglobins and a cluster for erythrocyte or gill hemoglobins. The myoglobin cluster is divided further into two subclusters, corresponding to monomeric and dimeric myoglobins, respectively.Liolophura myoglobin was placed on the branch of monomeric myoglobin lineage, showing that it diverged earlier from other monomeric myoglobins. The hemoglobin cluster is also divided into two subclusters. One cluster contains homodimeric, heterodimeric, tetrameric, and didomain chains of erythrocyte hemoglobins of the blood clamsAnadara, Scapharca, andBarbatia. Of special interest is the other subcluster. It consists of three hemoglobin chains derived from the bacterial symbiont-harboring clamsCalyptogena andLucina, in which hemoglobins are supposed to play an important role in maintaining the symbiosis with sulfide bacteria. 相似文献

The amino acid sequence of plastocyanin from lettuce (Lactuca sativa L.) was determined by using a Beckman 890C automatic sequencer and by dansyl-phenylisothiocyanate analysis of peptides obtained by enzymic digestion of purified CNBr fragments. The protein consists of a single polypeptide chain of 99 residues, and shows close homology with other higher plant plastocyanins. Two trypsin inhibitors, LA-1 and LA-2, have been isolated from ridged gourd (Luffa acutangula Linn.) seeds and purified to homogeneity by gel filtration followed by ion-exchange chromatography. The isoelectric point is atpH 4.55 for LA-1 and atpH 5.85 for LA-2. The Stokes radius of each inhibitor is 11.4 å. The fluorescence emission spectrum of each inhibitor is similar to that of the free tyrosine. The biomolecular rate constant of acrylamide quenching is 1.0×109 M–1 sec–1 for LA-1 and 0.8 × 109 M–1 sec–1 for LA-2 and that of K2HPO4 quenching is 1.6×1011 M–1 sec–1 for LA-1 and 1.2×1011M–1 sec–1 for LA-2. Analysis of the circular dichroic spectra yields 40%-helix and 60%-turn for La-1 and 45%-helix and 55%-turn for LA-2. Inhibitors LA-1 and LA-2 consist of 28 and 29 amino acid residues, respectively. They lack threonine, alanine, valine, and tryptophan. Both inhibitors strongly inhibit trypsin by forming enzymeinhibitor complexes at a molar ratio of unity. A chemical modification study suggests the involvement of arginine of LA-1 and lysine of LA-2 in their reactive sites. The inhibitors are very similar in their amino acid sequences, and show sequence homology with other squash family inhibitors. 相似文献

Summary The complete amino acid sequence of the major component myoglobin from killer whale,Orcinus orca, was determined by automated Edman degradation. In this study residue 122 was found to be glutamic acid instead of glutamine as was originally reported (Castillo et al. 1977). This reassignment affects the phylogenetic relationship of killer whale myoglobin with the myoglobins from other closely related cetacean species and also affects studies concerned with the physical parameters of the protein.This is the 114th paper in a series dealing with coordination complexes and catalytic properties of proteins and related substances. The penile morphology and bacular structure of five species of African mole rat are described in relation to the method of ovulation, degree of sociality and polygynandry. We predicted that, with an increase in sociality, and a concomitant decrease in polygynandry and sperm competition, there would be a decrease in penis ornamentation (spinosity) and baculum size. In solitary species of African mole rat with marked seasonal reproduction and induced ovulation ( Bathyergus suillus and Georychus capensis ), males have numerous epidermal spines on the penis. Social, seasonally breeding, induced ovulating mole rats Cryptomys hottentotus natalensis possess less elaborate ornamentation in the form of small protrusions that are rounded at the apex. Two aseasonally breeding eusocial species with spontaneous ovulation Cryptomys damarensis and Heterocephalus glaber have ridges on the penis but lack any elaborate ornamentation. Baculae, however, showed a trend to become proportionally smaller in the solitary species. Our prediction that, with an increase in sociality and a move from induced to spontaneous ovulation, the amount and degree of penile ornamentation declines was, therefore, generally supported. 相似文献

Studies on patterns of habitat use by mammals are necessary for understanding the mechanisms involved in their distribution and abundance. In this study, we used the spool-and-line method to investigate habitat utilization by two sigmodontine rodents from Brazilian Cerrado, Necromys lasiurus and Oryzomys scotti. We conducted the study in a Cerrado area in central Brazil (15°56′S and e 47°56′W) where the animals were caught in an area of 7.68 ha of Cerrado sensu stricto. Captured individuals were marked, equipped with a spool-and-line device, and released at the same capture point. The next day we followed the thread to record their daily movements and find their nests. To investigate microhabitat selection we compared habitat characteristics along trails of each studied species with general habitat characteristics of the study area. Although the mean 24-h distance was greater for N. lasiurus (mean ± SE: 41.9 ± 42.2 m, N=3) than for O. scotti (28.7 ± 14.2 m, N=6) this difference was not significant (Mann–Whitney test, U=26, P>0.6). We detected significant differences among observed microhabitats variables of both species and available microhabitat characteristics as determined by discriminant analysis (Wilks′s lambda F=3.001; df=14, 116; P<0.001). Both species were associated to microhabitat characteristics whose values differed markedly from the overall available habitat. Along the first canonical discriminant function of the DFA both them were associated with greater grass height than the mean height available and along the second axis N. lasiurus selected areas with higher fruit availability and more shelters than those selected by O. scotti. For stronger inferences regarding differential patterns of habitat utilization by Cerrado rodents we suggest the simultaneous use of both spool-and-line and standard trapping methods. 相似文献

The ecological constraints prevalent in arid environments havepromoted the evolution of social groups with a high reproductiveskew in mole rat species occurring there. Outbred social bathyergidsface conflict between maintaining colony integrity to enhancepersonal foraging success and hence survival, and dispersalto maximum individual lifetime reproductive success (LRS). Thisconflict will be a crucial determinant of the response of colonymembers to the presence of foreign conspecifics. We investigatedhow ecological constraints, sex, and reproductive status influencethe outcome of meetings between foreign common mole rats (Cryptomyshottentotus hottentotus) in a series of dyadic encounters. Individualsfrom two localities, at the extremes of an aridity gradient,were used to assess the effects of aridity. The effects of sexand reproductive status were investigated by trials betweenindividuals of differing sex and status. The arid populationrevealed substantially higher levels of rejection than the mesicpopulation. Sex and status played a significant role in moderatingindividual response. For both populations, encounters betweendifferent-sexed individuals produced lower levels of rejectionthan those between same-sexed individuals. For the mesic site,rejection was greatest for encounters between reproductive animals.Conversely, for the arid site, the levels of rejection werecomparatively high and comparable for all combinations of reproductivestatus. Ecological constraints, sex, and reproductive statusare significant factors in interactions between foreign commonmole rats, ultimately influencing individual survival, colonyintegrity and the differential LRS of all colony members. The morphology of the spermatozoon of representative species of the subfamily Nesomyinae (Muroidea: Nesomyidae), a monophyletic group of rodents endemic to Madagascar, was examined by light and electron microscopy to determine the sperm head shape and tail length across the species. Marked interspecific differences were found to occur in both the form of the sperm head and length of the tail. The species that possess a sperm head with an apical hook, which largely contains acrosomal material, generally displayed longer sperm tails, and a species with a spatulate sperm head had the shortest tail. The association between sperm head shape and tail length mirrors that previously found in Eurasian and Australasian murine rodents. Thus, the repeated association between sperm head shape and tail length across these groups of muroid rodents clearly indicates a functional relationship between these two features. A comparison of sperm morphology of the nesomyines to that of related muroid rodents on the mainland of Africa suggests that the possession of an apical hook is the ancestral condition. J.
