Physiological and pathological functions of mitochondria are highly dependent on the properties and regulation of mitochondrial ion channels. There is still no clear understanding of the molecular identity, regulation, and properties of anion mitochondrial channels. The inner membrane anion channel (IMAC) was assumed to be equivalent to mitochondrial centum picosiemens (mCS). However, the different properties of IMAC and mCS channels challenges this opinion. In our study, we characterized the single-channel anion selectivity and pH regulation of chloride channels from purified cardiac mitochondria. We observed that channel conductance decreased in the order: Cl? > Br? > I? > chlorate ≈ formate > acetate, and that gluconate did not permeate under control conditions. The selectivity sequence was Br? ≥ chlorate ≥ I? ≥ Cl? ≥ formate ≈ acetate. Measurement of the concentration dependence of chloride conductance revealed altered channel gating kinetics, which was demonstrated by prolonged mean open time value with increasing chloride concentration. The observed mitochondrial chloride channels were in many respects similar to those of mCS, but not those of IMAC. Surprisingly, we observed that acidic pH increased channel conductance and that an increase of pH from 7.4 to 8.5 reduced it. The gluconate current appeared and gradually increased when pH decreased from pH 7.0 to 5.6. Our results indicate that pH regulates the channel pore diameter in such a way that dilation increases with more acidic pH. We assume this newly observed pH-dependent anion channel property may be involved in pH regulation of anion distribution in different mitochondrial compartments.  相似文献

The objectives of this study were to determine (1) the individual ad libitum intake of mineral mix by beef cows managed under a year-long, fall-calving, forage-based production regimen and (2) if Se form in the mineral mix affected the blood Se concentrations of cows and suckling calves. Twenty-four late-gestation (6 to 8 months) Angus-cross cows (2.7?±?0.8 years; body weight [BW]?=?585?±?58 kg) were blocked by BW and randomly assigned (n?=?8) to a mineral supplement treatment (TRT) containing 35 ppm Se as either inorganic (ISe; sodium selenite), organic (OSe; Sel-Plex®), or a 1:1 combination of ISe/OSe (MIX). Cows commonly grazed a 10.1-ha predominately tall fescue pasture and had individual ad libitum access to TRT using in-pasture Calan gates. Cows calved from August to November and calves had common ad libitum access to creep feed and a mineral supplement that lacked Se. Cow jugular blood was taken at 28-day intervals (13 periods) and calf blood was taken with cows from birth through weaning. Individual cow mineral mix (mean?=?54.0?±?7.0 g/day, range?=?97.3 to 27.9?±?7.4 g/day) and Se (mean?=?1.82?±?0.25 mg/day, range?=?3.31 to 0.95?±?0.25 mg/day) intakes were affected by period (P?<?0.0001), but not by cow Se TRT (P?>?0.30). Cow blood Se (0.109 to 0.229?±?0.01 μg/mL) was affected (P?<?0.002) by period, Se form, and their interaction, with ISe?< MIX for periods 8 and 11, ISe?<?OSe for all periods except period 1, and MIX?<?OSe for periods 2 to 4, 7, 8, 10, and 12. Calf blood Se (in micrograms Se per milliliter) was correlated with cow blood Se and affected (P?<?0.0001) by cow Se TRT, with ISe (0.07 to 0.11)?<?MIX (0.10 to 0.15)?=?OSe (0.16 to 0.19). These data reveal that (1) mean supplemental ad libitum cow mineral intake was 36 % less than the typical formulation intake expectations (85 g/day) and, correspondingly, mean supplemental Se intake was 33 % less than that allowed by the FDA and (2) cow Se TRT differentially affected both cow and calf blood Se concentrations, resulting in adequate concentrations for all cows but inadequate concentrations for ISe calves.  相似文献

Background

Non-invasive imaging tests are widely used in the evaluation of stable angina pectoris (SAP). Despite these tests, non-significant coronary lesions are not a rare finding in patients undergoing elective coronary angiography (CAG). Two-dimensional (2D) speckle tracking global longitudinal strain (GLS) imaging is a more sensitive and accurate technique for measuring LV function than conventional 2D methods. Layer-specific strain analysis is a relatively new method that provides endocardial and epicardial myocardial layer assessment. The aim of the present study was to evaluate longitudinal layer-specific strain (LSS) imaging in patients with suspected SAP.

Methods

Patients who underwent CAG for SAP were retrospectively screened. A total of 79 patients with no history of heart disease and wall motion abnormalities were included in the study. Forty-three patients with coronary lesions >?70% constituted the coronary artery disease (CAD) group and 36 patients without significant CAD constituted the control group. Layer-specific GLS transmural, endocardium, and epicardium values (GLS-trans, GLS-endo, and GLS-epi, respectively) were compared between the groups.

Results

Patients in the CAD group had significantly lower GLS values in all layers (GLS-trans: -18.2 + 2.4% vs -22.2 + 2.2% p?<?.001; GLS-endo: -20.8 + 2.8% vs -25.3 + 2.6%, p?<?.001; GLS-epi: 15.9 + 2.4% vs -19.5 + 1.9%, p?<?.001). Multivariate adjustment demonstrated GLS-trans as the only independent predictor of CAD [OR:0.472, CI (0.326–0.684), p?<?.001]. Additionally, the GLS values were all lower in myocardial perfusion scintigraphy (MPS) true-positive patients compared with MPS false-positive patients (GLS-trans: -17.7?±?2.4 vs. -21.9?±?2.4%, p?<?.001; GLS-endo: -20.2?±?2.9% vs -24.9?±?2.9%, P?<?.001; GLS-epi: 15.4?±?2.6% vs. -19.2?±?1.8%, P?<?.001).

Conclusion

Resting layer-specific strain as assessed by 2D speckle tracking analysis demonstrated that GLS values were reduced in all layers of myocardium with SAP and with no wall motion abnormalities. LSS analysis can improve the identification of patients with significant CAD but further prospective larger scale studies are needed to put forth the incremental value of LSS analysis over transmural GLS.

In the present study, the influence of humic acid (HA) molecular fractions (HA?<?30 kDa and HA?>?30 kDa) on enhancing the tolerance of seedlings of soybean [Glycine max (L.) Merr.] Progres and Nawiko cultivars to salt stress (50 mM NaCl) was investigated. HA were extracted from mountain fen soil and then were separated into two molecular fractions by membrane filtration and characterized by diffusion coefficient (Dapp), electrolytic conductivity (κ) and electrophoretic mobility (Ue). The following biometric parameters of tested plants were determined: total leaf area, height of plants, fresh and dry mass of the over ground part and roots as well as length of shoots cells and length of stomas. The chlorophyll content in ground tissue as well as the macro and microelements content in tested plants also were determined. The results showed that the κ, Ue and Dapp for HA?>?30 kDa were lower than these for HA?<?30 kDa. Adding NaCl caused increase κ and decrease Dapp and Ue. The salt stress caused a major decrease in biometric parameters in tested plants. HA?>?30 kDa reduced the uptake of macro and microelements in the soybean Progres cultivar. In soybean Nawiko cultivar, it caused significant uptake of Fe and Zn. Soybean cultivars showed strong reaction to salt stress. HA molecular fractions reduced or eliminated the influence of the salt stress. However, HA?>?30 kDa was more effective than HA?<?30 kDa, due to its properties.  相似文献

Metal removal potential of indigenous mining microorganisms from acid mine drainage (AMD) has been well recognised in situ at mine sites. However, their removal capacity requires to be investigated for AMD treatment. In the reported study, the capacity of an indigenous AMD microbial consortium dominated with Klebsormidium sp., immobilised in a photo-rotating biological contactor (PRBC), was investigated for removing various elements from a multi-ion synthetic AMD. The synthetic AMD was composed of major (Cu, Mn, Mg, Zn, Ca, Na, Ni) and trace elements (Fe, Al, Cr, Co, Se, Ag, Mo) at initial concentrations of 2 to 100 mg/L and 0.005 to 1 mg/L, respectively. The PRBC was operated for two 7-day batch periods under pH conditions of 3 and 5. The maximum removal was observed after 3 and 6 days at pH 3 and 5, respectively. Daily water analysis data demonstrated the ability of the algal–microbial biofilm to remove an overall average of 25–40 % of the major elements at pH 3 in the order of Na?>?Cu?>?Ca?>?Mg?>?Mn?>?Ni?>?Zn, whereas a higher removal (35–50 %) was observed at pH 5 in the order of Cu?>?Mn?>?Mg?>?Ca?>?Ni?>?Zn?>?Na. The removal efficiency of the system for trace elements varied extensively between 3 and 80 % at the both pH conditions. The batch data results demonstrated the ability for indigenous AMD algal–microbial biofilm for removing a variety of elements from AMD in a PRBC. The work presents the potential for further development and scale-up to use PBRC inoculated with AMD microorganisms at mine sites for first or secondary AMD treatment.  相似文献

Litterfall production, decomposition and nutrient use efficiency in three different tropical forest ecosystems in SW China were studied for 10 years. Annual mean litterfall production in tropical seasonal forest (TSF) (9.47?±?1.65 Mg ha?1) was similar to that in man-made tropical forest (MTF) (9.23?±?1.29 Mg ha?1) (P?>?0.05) but both were significantly lower than that in secondary tropical forest (STF) (12.96?±?1.71 Mg ha?1) (P?<?0.05). The annual variation of litterfall was greater in TSF (17.4%, P?<?0.05) than in MTF (14.0%) or STF (13.2%). The annual mean decomposition rate of litterfall increased followed the order of MTF (2.72)?<?TSF (3.15)?<?STF (3.50) (P?<?0.05), which was not correlated with annual precipitation or annual mean temperature, but was rather related to litter quality. The nutrient use efficiency was found to be element-dependent and to vary significantly among the three forest types (P?<?0.05). These results indicate that litterfall production and decomposition rates in different tropical forest systems are related to plant species composition and are influenced strongly by coexisting species and their life stage (age) but less so by the species richness. Constructing multi-species and multistory man-made tropical forest is an effective way to enhance biological productivity and maintain soil nutrients on degraded tropical land.  相似文献

Since the 1990s, blood donors have been scanned for anti-hepatitis C virus (anti-HCV) antibodies, which can be defined by enzyme immunoassay as a screening test. In this population, false-reactive ratios have been high. Recently, some authors have aimed to find a cutoff value for anti-HCV different from those established by test manufacturers to predict HCV infection. In this study, 321 patients, after two repeating tests, had reactive results in s/co <10 titers on anti-HCV test. The patients were 29.6 % (n?=?95) in women and 70.4 % (n?=?226) in men. The patients were classified into three groups by Western blot (WB) results (PS, positive; NG, negative; and ID, indeterminate). The average anti-HCV titer of the whole group was 2.61?±?1.96. Anti-HCV titers of subgroups were 2.43?±?1.95 in NG, 4.93?±?2.53 in PS, and 2.50?±?1.65 in ID (p?<?0.001). There was a significant difference between NG and PS and between PS and ID subgroups (p?<?0.001). There was a positive correlation between WB and anti-HCV titers in all patients (r?=?0.298, p?<?0.001), in women (r?=?0.282, p?<?0.001), and in men (r?=?0.337, p?=?0.002). According to receiver operator characteristic curve analysis, the cutoff value of anti-HCV titer to predict hepatitis C infection was >2.61 s/co, with 74.1 % sensitivity and 71.6 % specificity (area under the curve, 0.820; 95 % confidence interval, 0.753 to 0.887). We suggest that an effective cutoff value for anti-HCV other than that established by the manufacturer cannot be assigned to predict hepatitis C infection for blood donors in low-prevalence areas.  相似文献

The effects of trace elements of gallium (Ga), germanium (Ge), and strontium (Sr) on ovariectomized (OVX) osteopenic rats were studied in this paper. The urine calcium content, serum calcium, and phosphorus contents, bone mineral content, mineral dissolution, and mechanical strength of the osteopenic rats were analyzed respectively. After the rats were fed with Ga, Ge, and Sr diet for 8 weeks, respectively, the urine calcium content decreased (P?<?0.01). Plasma calcium and phosphate concentrations decreased in the order of OVX group?>?Ge group?>?Sr group?>?Ga group?>?Sham group. Mineral content increased in the order of OVX group?<?Ge group?<?Sr group?<?Ga group?<?Sham group. A dramatic decrease in calcium solubility was found both in the gallium and strontium treated animals (P?<?0.05). However, the same result did not occur in germanium treated groups. The data provide an important proof of concept that gallium and strontium might be a new potential therapy for the management of postmenopausal osteoporosis in humans.  相似文献

This study evaluated the effects of supplemental dietary chromium (Cr) on the performance, carcass traits, and some serum parameters of broilers under a heat stress (23.9 to 37 °C cycling) condition. A total of 150 1-day-old broiler chicks (Cobb 500) according to a completely randomized design were assigned into five treatment groups. Each treatment consisted of three replicates and each replicate contained ten chicks. Treatments were supplemented with 0 (control), 600, and 1,200 μg kg?1 Cr in the form of Cr chloride (CrCl3) and Cr L-methionine from 1 to 49 days of age. Blood samples were collected from two birds in each replicate to determine serum parameters at 35 and 49 days of age. The body mass, feed intake, and conversion ratio were not influenced by dietary Cr (P?>?0.05). Dietary supplementation of Cr from either CrCl3 or Cr L-methionine caused increased serum concentrations of Cr (P?<?0.05), but had no effect on serum insulin and glucose concentrations at both sampling times (P?>?0.05). Serum triglycerides, very low-density lipoprotein cholesterol, low-density lipoprotein cholesterol, and high-density lipoprotein cholesterol were also not significantly affected (P?>?0.05) by dietary treatments, whereas total cholesterol concentration decreased in chicks fed Cr L-methionine compared to the control (P?<?0.05).  相似文献

A study was conducted using 360 broiler chickens to evaluate the effects of dietary vitamin E (0, 125 and 250 mg/kg), selenium (Se, 0, 0.5 and 1 mg/kg), or their different combinations on immune response and blood biological parameters of broilers raised under either thermoneutral (TN, 23.9 °C constant) or heat stress (HS, 23.9 to 37 °C cycling) conditions. Humoral immunity was assessed by intravenous injection of 7 % sheep red blood cell (SRBC) followed by evaluation of serum for antibody titers in primary and secondary responses. Heterophil to lymphocyte (H/L) ratio also determined as an indicator of stress. Furthermore, at the end of the experiment, birds were bled for determination of some biological parameters. There was a significant reduction in body weight and feed intake, but the feed conversion ratio increased when the birds were exposed to HS (P?<?0.05). Body weight and feed intake were not influenced significantly by dietary vitamin E and Se (P?>?0.05), whereas feed conversion was improved significantly by 125 mg/kg vitamin E (P?<?0.05). The liver and lymphoid organ weights as well as IgM and IgG, antibody titers for primary and secondary antibody responses to SRBC were reduced significantly under HS (P?<?0.05). Heat stress also resulted in a significant increase in H/L ratio (P?<?0.05). Dietary vitamin E resulted in improvement of primary and secondary antibody responses both in TN and HS broilers (P?<?0.05). The HS birds also showed an improved antibody titer in secondary response with high concentration of Se (P?<?0.05). Vitamin E and Se had interactive effects on anti-SRBC titers; however, no consistent differences were found between dietary levels during the study. The H/L ratio decreased by feeding vitamin E at both levels either under HS or TN conditions (P?<?0.05). A great concern exists about the lifetime, cost, low-temperature performance, and safety of Li-ion batteries. Na-ion batteries (NIB) are an alternative to the Li-ion batteries due to the wide availability of sodium, its low cost, and nontoxicity. Here, we examined the Na and Na+ adsorption on nanosheets of carbon (graphene), AlN, BN, and SiC to explore their potential use as an anode in NIBs. The interaction of atomic Na was found to play the main role in producing different nanosheet cell voltages. Unlike the graphene and SiC nanosheets, the lone pairs on the surface of the AlN and BN nanosheets hinder the Na adsorption and significantly increase the cell voltage. The order of magnitude of the nanosheet cell voltage as an anode in NIBs is as follows: AlN (1.49 V)?>?BN (1.46 V) >?>?C (0.69 V)?>?SiC (0.61 V). The AlN and BN nanosheets may be appropriate compounds for NIBs and their cell voltages are comparable with carbon nanotubes.  相似文献

A 3?×?2 factorial experiment consisting three levels (0, 200, and 400 μg/kg) of chromium (chromium picolinate) and two levels (0 and 250 mg/kg) of vitamin C was employed to evaluate the effects of these dietary supplements on performance, egg quality traits, and serum biochemical parameters of heat-stressed laying hens (Lohmann LSL-Lite) from 66 to 74 weeks of age. Feed intake increased when birds were given either 400 μg/kg chromium or 250 mg/kg vitamin C (P?<?0.05), but the birds that received both chromium and vitamin C consumed feed similar to those that received only chromium. Dietary treatments had no effect on egg production, egg mass, egg volume, feed conversion ratio, and body mass (P?>?0.05). The birds that fed on diet with chromium or vitamin C produced eggs with higher shell mass and thickness compared to the control. Both eggshell mass and thickness decreased when vitamin C and chromium were supplemented simultaneously, and birds given the diet supplemented with 400 μg/kg chromium and 250 mg/kg vitamin C had eggshell mass and thickness similar to those of the control group. The serum concentration of chromium increased due to increasing level of dietary chromium (P?<?0.05). The birds that received diet with chromium and vitamin C had higher serum concentrations of chromium compared to those that received only chromium (P?<?0.05). Similarly, the hens that received chromium and vitamin C had higher serum concentrations of calcium and phosphorus compared to the hens fed with other treatments (P?<?0.05). Curcumin, a polyphenol, has pharmacological effects including antioxidant, anti-inflammatory and anti-cancer features. In this study, we have performed comparative in vivo evaluations of CDF (curcumin difluorinated) and curcumin in cisplatin-induced nephrotoxicity in rats. Male Wistar rats were divided into four groups: (1) Control; (2) Cisplatin (7 mg/kg body wt, intraperitoneal as a single dose); (3) Cisplatin and CDF (50 mg/rat/day; for 12 days); (4) Cisplatin and curcumin (50 mg/rat/day), for 12 days). Cisplatin treated rats exhibited kidney injury manifested by increased serum N-urea and creatinine (P?<?0.001). Kidney from cisplatin treated rats also exhibited significant increase in malondialdehyde (MDA) and 8-isoprostane levels (P?<?0.001). Treatment with CDF and curcumin prevented the rise in serum N-urea, creatinine, MDA and 8-isoprostane as compared to experimental control group in kidney (P?<?0.05). Compared to curcumin, CDF had greater potential in suppressing cisplatin-induced pro-inflammatory factors NF-κB and COX-2 as well as downstream markers Nrf2 and HO-1 (P?<?0.05) in kidney. The analysis on anion transport markers (OAT1 and OAT3) showed a similar trend (CDF?>?curcumin). CDF could reduce the expression of multi-drug resistance markers OCT1, OCT2, MRP2 and MRP4 to a much greater extent than curcumin (P?<?0.05). We also demonstrate that CDF influenced the expression of p-mTOR, p-p70S6K1, p-4E-BP1 and p-Akt. These data suggest that CDF can potentially be used to reduce the chemotherapy induced nephrotoxicity thereby enhancing the therapeutic window of cisplatin. The results also proved that compared to curcumin, CDF has superior protective effect in nephrotoxicity.  相似文献

Forty weaned male guinea pigs (Cavia porcellus) of 152.6?±?7.96 g mean body weight were divided into four equal groups and fed a common basal diet comprised of 25% ground cowpea (Vigna unguiculata) hay, 30% ground maize (Zea mays) grain, 22% ground gram (Cicer arietinum) grain, 9.5% deoiled rice (Oryza sativa) bran, 6% soybean (Glycine max) meal, 6% fish meal, 1.5% mineral mixture (without Se), and ascorbic acid at 200 mg/kg to meet their nutrient requirements along with 0, 0.1, 0.2, and 0.3 ppm of organic selenium (Se) in groups I, II, III, and IV, respectively. Experimental feeding lasted for a period of 10 weeks, during which, daily feed intake and weekly body weights were recorded. Intake and digestibility of dry matter, organic matter, ether extract, crude fiber, and nitrogen-free extract as well as uptake of calcium and phosphorus were similar (P?>?0.05) among the four groups. Feed:gain ratio was also similar (P?>?0.05) in the four groups. However, digestibility of crude protein was significantly (P?<?0.001) higher in group II supplemented with 0.1 ppm organic Se as compared to other three group. Intake and absorption of Se was significantly (P?<?0.001) higher in all the Se supplemented groups as compared to control group. Average daily gain (ADG) was significantly (P?<?0.05) higher in group II (3.16 g/day) and III (3.38 g/day) as compared to group I (2.88 g/day). However, ADG in group IV (supplemented 0.3 ppm organic Se) was significantly (P?<?0.05) lower (2.83 g/day) than group II and III, but comparable (P?>?0.05) to group I. Findings of the present experiment suggests that Se requirements of guinea pigs are ≥0.2 ppm, as supplementation of 0.1 ppm organic Se in the diet (having 0.1 ppm Se) not only enhanced their growth rate but also improved the protein utilization.  相似文献

There has been considerable interest in cultivation of green microalgae (Chlorophyta) as a source of lipid that can alternatively be converted to biodiesel. However, almost all mass cultures of algae are carbon-limited. Therefore, to reach a high biomass and oil productivities, the ideal selected microalgae will most likely need a source of inorganic carbon. Here, growth and lipid productivities of Tetraselmis suecica CS-187 and Chlorella sp were tested under various ranges of pH and different sources of inorganic carbon (untreated flue gas from coal-fired power plant, pure industrial CO2, pH-adjusted using HCl and sodium bicarbonate). Biomass and lipid productivities were highest at pH 7.5 (320?±?29.9 mg biomass L?1 day?1and 92?±?13.1 mg lipid L?1 day?1) and pH 7 (407?±?5.5 mg biomass L?1 day?1 and 99?±?17.2 mg lipid L?1 day?1) for T. suecica CS-187 and Chlorella sp, respectively. In general, biomass and lipid productivities were pH 7.5?>?pH 7?>?pH 8?>?pH 6.5 and pH 7?>?pH 7.5?=?pH 8?>?pH 6.5?>?pH 6?>?pH 5.5 for T. suecica CS-187 and Chlorella sp, respectively. The effect of various inorganic carbon on growth and productivities of T. suecica (regulated at pH?=?7.5) and Chlorella sp (regulated at pH?=?7) grown in bag photobioreactors was also examined outdoor at the International Power Hazelwood, Gippsland, Victoria, Australia. The highest biomass and lipid productivities of T. suecica (51.45?±?2.67 mg biomass L?1 day?1 and 14.8?±?2.46 mg lipid L?1 day?1) and Chlorella sp (60.00?±?2.4 mg biomass L?1 day?1 and 13.70?±?1.35 mg lipid L?1 day?1) were achieved when grown using CO2 as inorganic carbon source. No significant differences were found between CO2 and flue gas biomass and lipid productivities. While grown using CO2 and flue gas, biomass productivities were 10, 13 and 18 %, and 7, 14 and 19 % higher than NaHCO3, HCl and unregulated pH for T. suecica and Chlorella sp, respectively. Addition of inorganic carbon increased specific growth rate and lipid content but reduced biomass yield and cell weight of T. suecica. Addition of inorganic carbon increased yield but did not change specific growth rate, cell weight or content of the cell weight of Chlorella sp. Both strains showed significantly higher maximum quantum yield (Fv/Fm) when grown under optimum pH.  相似文献

A study was conducted using 240 female day-old broiler chicks to evaluate the effects of dietary chromium picolinate (CrPic), peppermint essential oil (P.mint), or their combination on growth performance and blood biochemical parameters of female broiler chicks raised under heat stress conditions (HS, 23.9 to 38 °C cycling). Average daily gain (ADG), average daily feed intake (ADFI), and feed conversion ratio (FCR) were obtained from 1 to 42 days of age. Furthermore, at the end of the experiment (day 42), birds were bled to determine some blood biochemical parameters and weighed for final body weight (BW). ADFI, ADG, and BW were not influenced significantly by dietary CrPic and P.mint (P?>?0.05). A significant interaction between dietary CrPic and P.mint on FCR (P?=?0.012) was detected. FCR significantly decreased in chicks fed the diet including both CrPic and P.mint compared with the CrPic group. Significant interaction between dietary P.mint and CrPic on serum concentrations of triglycerides, glucose, and albumin were observed (P?<?0.05), but the other measured blood biochemical parameters were not statistically affected by dietary treatments (P?>?0.05). The serum concentrations of glucose, triglycerides were decreased (P?<?0.05) in broilers fed the diet including both CrPic and P.mint. Thirty Chlorella and 30 Scenedesmus strains grown in nitrogen-stressed conditions (70 mg L?1 N) were analyzed for biomass accumulation, lipid productivity, protein, and fatty acid (FA) composition. Scenedesmus strains produced more biomass (4.02?±?0.73 g L?1) after 14 days in culture compared to Chlorella strains (2.57?±?0.12 g L?1). Protein content decreased and lipid content increased from days 8 to 14 with an increase in triacylglycerol (TAG) accumulation in most strains. By day 14, Scenedesmus strains generally had higher lipid productivity (53.5?±?3.7 mg lipid L?1 day?1) than Chlorella strains (35.1?±?2.8 mg lipid L?1 day?1) with the lipids consisting mainly of C16–18 TAGs. Scenedesmus strains generally had a more suitable FA profile with higher amounts of saturated fatty acids and monounsaturated fatty acids (MUFAs) and a smaller polyunsaturated fatty acid (PUFA) component. Chlorella strains had a larger PUFA component and smaller MUFA component. The general trend in the FA composition of Chlorella strains was oleic > palmitic > α-linolenic = linoleic > eicosenoic > heptadecenoic > stearic acid. For Scenedesmus strains, the general trend was oleic > palmitic > linoleic > α-linolenic > stearic > eicosenoic > palmitoleic > heptadecenoic acid. The most promising strains with the highest lipid productivity and most suitable FA profiles were Scenedesmus sp. MACC 401, Scenedesmus soli MACC 721, and Scenedesmus ecornis MACC 714. Although Chlorella sp. MACC 519 had lower lipid productivity, the FA profile was good with a lower PUFA component compared to the other Chlorella strains analyzed and a low linolenic acid concentration.  相似文献

Urbanized watersheds in colder climates experience episodic salinization due to anthropogenic salt inputs and runoff from impervious surfaces. Episodic salinization can be manifested as a ‘pulse’ in concentrations and fluxes of salt ions lasting from hours to days after snowstorms in response to road salting. Episodic salinization contributes to freshwater salinization syndrome, characterized by cascading mobilization of chemicals and shifting acid–base status. We conducted laboratory experiments and analyzed high-frequency sensor data to investigate the water quality impacts of freshwater salinization syndrome and episodic salinization across 12 watersheds draining two major metropolitan regions along the U.S. East Coast. Sediments from 12 watersheds spanning land use gradients across two metropolitan regions, Baltimore, Maryland and Washington DC, were incubated across a range of replicated salinity treatments (0–10 g/L sodium chloride). There were statistically significant linear increasing trends in calcium and potassium concentrations with experimental salinization across all 12 sites and in magnesium concentrations at 11 of 12 sites (p?<?0.05), with mean rates of increase of 1.92?±?0.31 mg-Ca per g-NaCl, 2.80?±?0.67 mg–K per g-NaCl, and 1.11?±?0.19 mg-Mg per g-NaCl, respectively. Similarly, there were statistically significant increasing linear trends in total dissolved nitrogen (TDN) concentrations with experimental salinization at 9 of the 12 sites, with a mean rate of increase of 0.07?±?0.01 mg-N per g-NaCl. There were statistically significant increasing linear trends in soluble reactive phosphorus (SRP) concentrations with experimental salinization at 7 of the 12 sites (p?<?0.05), with a mean rate of increase of 2.34?±?0.66 µg-P per g-NaCl. The response of dissolved inorganic carbon (DIC) and organic carbon (DOC) concentrations to experimental salinization varied between sites, and dissolved silica did not show any significant response. High-frequency sensors near the experimental sites showed statistically significant positive linear relationships between nitrate concentrations, specific conductance, and chloride concentrations similar to relationships observed in laboratory incubations. Our results suggested that episodic salinization and freshwater salinization syndrome can mobilize base cations and nutrients to streams through accelerated ion exchange and stimulate different biogeochemical processes by shifting pH ranges and ionic strength. The growing impacts of freshwater salinization syndrome and episodic salinization on nutrient mobilization, shifting acid–base status, and augmenting eutrophication warrant serious consideration in water quality management.  相似文献

The primary production in the Greenland Sea, Fram Strait, Barents Sea, Kara Sea and adjacent Polar Ocean was investigated through the physically–biologically coupled, nested 3D SINMOD model with 4 km grid size for the years 1995–2007. The model had atmospheric forcing from the European Centre for Medium-Range Weather Forecasts (ECMWF) reanalysis data. Three basic gross primary production (GPP) domains were distinguished: (i) an extensive domain dominated by Atlantic Water, (ii) an elongated domain roughly corresponding to the seasonal ice zone (SIZ) and (iii) a compact perennial ice zone (>100, between 100 and 30 and <30 g C m?2 year?1, respectively). The interannual coefficient of variation for GPP in domain (i) was <0.1, and increased northwards towards >0.6 in the northwesternmost and northeasternmost fringe of the SIZ. The primary production in the northern sector of the European Arctic Corridor (EAC) region prior to 2007 was characterised by limited interannual variability, on average 75.2 ± 10% and 24.0 ± 16% g C m?2 year?1 for the EAC region at 74–80 and >80°N, respectively. The main primary production anomalies were found early in the productive season and in sections of the SIZ, generally in regions with low GPP. There was no significant trend of increasing GPP in the 1995–2007 time interval.  相似文献

Ferrous glycinate (Fe-Gly) maintains high bioavailability in animals, but its exact absorption mechanism is still unknown. Here, we studied on the absorption kinetics of ferrous glycinate and its impact on the relevant transport protein in Sprague-Dawley (SD) rats. A total of 72 SD rats (male, BW 100?±?6.25 g) were randomly allotted to three treatments. These treatments were perfused with 1 mL of normal saline, ferrous sulfate (FeSO4), and ferrous glycinate (71.35 mg/L as iron) separately. Four rats were selected from each treatment for collection of blood from the tails at certain times (15, 30, 45, 60, 75, 90, 120, 240, and 360 min) after gavage. Moreover, other six rats selected from each treatment were slaughtered for sampling after gavage at 2, 4, and 6 h to evaluate the expression of intestinal transport protein. Pharmacokinetic parameters of iron were determined by one-compartmental analysis. Compared with FeSO4, the peak plasma concentration of iron (C max) is higher in the rats given gavage with Fe-Gly (P?<?0.05). Four hours after gavage with Fe-Gly, the expression of divalent metal transporter 1 (DMT1) in the duodenum is significantly decreased (P?<?0.05), but the expression of ferroportin 1 (Fpn1) is significantly increased (P?<?0.05). This study indicates that Fe-Gly as iron sources can be absorbed more and utilized faster than FeSO4, and they had different effects on the expression of intestinal transport protein.  相似文献
