Saprolegnia diclina andS. parasitica isolated from three sources could germinate in strong acidic conditions. Growth ability correlated with the species of fungi rather than with the sources from which they were isolated.S.diclina isolates appeared to germinate at a pH condition as low as 3.5, whereasS. parasitica isolates could not germinate at below pH 3.8.S. parasitica isolates from visceral mycoses still showed good growth at 30°C, whereas other isolates did not. Also,S. parasitica isolates from visceral mycoses produced more abundant motile zoospores, and continued to do so for a longer period of time (28 d), thanS. parasitica isolates from external saprolegniasis andS. diclina isolates.  相似文献

Parasitological monitoring was carried out from 1997 to 1999 in a highly saline (41–45 g/l) lake in southeastern California, Salton Sea. A total of 1473 fishes were examined. Young tilapia, Oreochromis mossambicus, croaker, Bairdiella icistia, and mudsucker, Gillichthys mirabilis, were found infected by ectoparasites. Some persistent foci of fish infestations were found around the perimeter of the lake. The diversity of parasites was limited to three protozoan species, Amyloodinium ocellatum (Dinoflagellida), Ambiphrya ameiuri (Peritricha), Cryptobia branchialis (Bodonida: Kinetoplastida), and two metazoans, the monogeneans Gyrodactylus olsoni and G. imperialis. Both A. ocellatum and A. ameiuri infested fish from spring through fall. The greatest infestations occurred in summer (29–40°C) in the case of A. ocellatum and in spring and autumn (22–27°C) in the case of A. ameiuri. High parasite loads caused severe damage to such respiratory organs as gills and skin. They may depress respiration and osmoregulation and, in combination with other environmental factors, cause fish suffocation and death. These parasites may play a major role in juvenile fish mortality.  相似文献

Three psychrotrophic Arthrobacter strains, isolated from Antarctic lake sediment (An24, An25T) and deep-sea sediment (ZX6) were studied. Their 16S rRNA gene sequences showed highest similarities (97.0–97.9%) with those of A. nicotianae and A. protophormiae. All three strains underwent rod–coccus morphological change, had high mol% G+C content, were aerobic to slightly anaerobic, and grew between 0°C and 30°C, with optimal growth temperature around 25°C. The cell wall peptidoglycan was A4 variant. DNA–DNA hybridization, physiological and chemotaxonomic studies indicated that these three strains constituted a new homogeneous genomic species within the genus Arthrobacter, for which the name Arthrobacter ardleyensis, with the type strain An25T (CGMCC 1.3685, JCM 12921) was proposed.M. Three species of Lagenidiales were isolated from the eggs and zoeae of the marine crabPortunus pelagicus. One of them,Lagenidium callinectes, is reported for the first time in Japan, with fungal infection in the eggs.Haliphthoros milfordensis was discovered from a zoea of the crab.Atkinsiella okinawaensis was also isolated from a zoea of the crab and described as a new species. Growth temperature range and optimum temperature of the fungi were examined. All of the isolates grew at various concentrations of NaCl or KCl, and optimum growth was observed on PYGS agar containing seawater. Pathogenicity to zoeae of the swimming crabPortunus trituberculatus was demonstrated by using a zoospore suspension of each fungus at 25°C.  相似文献

Carboxypeptidase produced by Monascus purpureus IFO 4478 was purified to homogeneity. The purified enzyme is a heterodimer with a molecular mass of 132 kDa and consists of two subunits of 64 and 67 kDa. It is an acidic glycoprotein with an isoelectric point of 3.67 and 17.0% carbohydrate content. The optimum pH and temperature were 4.0 and 40 °C, respectively. The enzyme was stable between pH 2.0 and 8.0 at 37 °C for 1 h, and up to 50 °C at pH 5.0 for 15 min. The enzyme was strongly inhibited by piperastatin A, diisopropylfluoride phosphate (DFP), phenylmethylsulfonylfluoride (PMSF), and chymostatin, suggesting that it is a chymotrypsin-like serine carboxypeptidase. Monascus purpureus carboxypeptidase was also strongly inhibited by p-chloromercuribenzoic acid (PCMB) but not by ethylenediaminetetraacetic acid (EDTA) and 1,10-phenanthroline, indicating that it requires cysteine residue but not metal ions for activity. Benzyloxycarbonyl-l-tyrosyl-l-glutamic acid (Z-Tyr-Glu), among the substrates tested, was the best substrate of the enzyme. The Km, Vmax, Kcat, and Kcat/Km values of the enzyme for Z-Tyr-Glu at pH 4.0 and 37 °C were 0.86 mM, 0.917 mM min–1, 291 s–1, and 339 mM–1 s–1, respectively.  相似文献

Moina mongolica, 1.0-1.4 mm long and 0.8 mm wide, is an Old World euryhaline species. This paper reviewed the recent advances on its autecology, reproductive biology, feeding ecology and perspective as live food for marine fish larviculture. Salinity tolerance of this species ranges from 0.4–1.4 to 65.2–75.4. Within 2–50 salinity, Moina mongolica can complete its life cycle through parthenogenesis. The optimum temperature is between 25 °C and 28 °C, while it tolerates high temperature between 34.4 °C and 36.0 °C and lower temperature between 3.2 °C and 5.4 °C. The non-toxic level of unionised ammonia (24 h LC50) for M. mongolica is <2.6 mg NH3–N l–1. Juvenile individuals filter 2.37 ml d–1 and feed 9.45×106 algal cells d–1, while mature individuals filter 9.45 ml d–1 and consume 4.94×106 algal cells d–1. At 28 °C, M. mongolica reaches sex maturity in 4 d and gives birth once a day afterward; females carry 7.3 eggs brood–1 and spawn 2.8 times during their lifetime. A variety of food can be used for M. mongolica culture including unicellular algae, yeast and manure, but the best feeding regime is the combination of Nannochloropsis oculata and horse manure. Moina mongolica reproduces parthenogenetically during most lifetime, but resting eggs can be induced at temperature (16 °C) combined with food density at 2000–5000 N. oculata ml–1. The tolerance to low dissolved oxygen (0.14–0.93 mg l–1) and high ammonia makes it suitable for mass production. Biochemical analyses showed that the content of eicospantanoic acid (20:53) in M. mongolica accounts for 12.7% of total fatty acids, which is higher than other live food such as Artemia nauplii and rotifers. This cladoceran has the characteristics of wide salinity adaptation, rapid reproduction and ease of mass culture. The review highlights its potential as live food for marine fish larvae.  相似文献

Synopsis Contraction time of an isolated white muscle from the temperate water Girella tricuspidata is proportional to temperature and inversely proportional to fish size. Between ambient (14°C) and 8° C muscle from all sizes of fish is similary affected by temperature; the lower the temperature the more the contraction time is slowed. Below 8° C muscle from large fish is affected more than is muscle from small fish. Contraction time of white muscle in the antarctic notothenioid Pagothenia borchgrevinki is about twice as fast as that of Girella tricuspidata at temperatures between 2–12°C, but at normal body temperature, contraction time of muscle from Girella tricuspidata (14°C) is about twice as fast as that of Pagothenia borchgrevinki (–1.9°C).  相似文献

Aprostocetus vaquitarum (Wolcott) causes 78–91 percent mortality to eggs of Diaprepes abbreviatus (L.), under field conditions in southern Florida. In the laboratory, A. vaquitarum was reared on D. abbreviatus eggs at 25 °C, a photoperiod of 12:12 (L:D) and with abundant hosts, A. vaquitarum adult females lived around 15 days. Oviposition was significantly affected by the age of the host egg mass. Egg masses aged 0- to 3-day-old were accepted significantly better than those aged 4–6 days. The mean number of eggs deposited per female was around 53, with extreme values of 124 and 19 eggs per female. Using these data in combination with the sex ratio observed in the field (0.16) and the duration of the preimaginal stages, rm (0.168–0142 day−1), T (22.39–22.89 days), and R0 (43.03–25.81 females per female) were calculated.  相似文献

Saprolegnia species are destructive pathogens to many aquatic organisms and are found in most parts of the world. Reports based on phylogenetic analysis suggest that Saprolegnia strains isolated from aquatic animals such as crustaceans and frogs are close to Saprolegnia strains isolated from infected fish or fish eggs and vice versa. However, it has often been assumed that host specificity occurs for each individual isolate or strain. Here we demonstrate that Saprolegnia spp. can have multiple hosts and are thus capable of infecting different aquatic organisms. Saprolegnia delica, Saprolegnia hypogyna, and 2 strains of Saprolegnia diclina were isolated from aquatic insects and amphipods while S. delica, Saprolegnia ferax, Pythium pachycaule, and a Pythium sp. were isolated from the water of a medium to fast flowing river. The ITS region of the rRNA gene was sequenced for all isolates. In challenge experiments, all four isolates from insects were found to be highly pathogenic to eggs of Atlantic salmon (Salmo salar) and embryos of the African clawed frog (Xenopus laevis). We found that Saprolegnia spp. isolated from salmon eggs were also able to successfully establish infection in nymphs of stonefly (Perla bipunctata) and embryos of X. laevis). These results suggest that Saprolegnia spp. are capable of infecting multiple hosts, which may give them an advantage during seasonal variation in their natural environments.  相似文献

The influence of relative humidity (RH) and temperature on growth and metabolism of eight microfungi on 21 different types of building material was investigated. The fungi were applied as a dry mixture to the materials, which were incubated at 5°C, 10°C, 20°C and 25°C at three humidity levels in the range 69–95% RH over 4–7 months. The lower limit for fungal growth on wood, wood composites and starch-containing materials was 78% RH at 20–25°C and increased to 90% RH at 5°C. An RH of 86% was necessary for growth on gypsum board. Ceramic materials supported growth at RH >90%, although 95% RH was needed to yield chemically detectable quantities of biomass. Almost exclusively only Penicillium, Aspergillus and Eurotium (contaminant) species grew on the materials. Production of secondary metabolites and mycotoxins decreased with humidity and the quantities of metabolites were insignificant compared with those produced at high RH (RH >95%), except in the case of Eurotium.  相似文献

The reproduction of Nassarius festivus (Gastropoda: Nassariidae) has been studied on three contrasting beaches in Hong Kong. There was no significant difference in shell heights between females and males of N. festivus at three study sites and the sex ratio was always female biased. The highest female sex ratio was recorded from Starfish Bay, followed by Tai Tam Bay and then Tai Mong Tsai. The sexual cycle of N. festivus is completed in 1 year in Hong Kong. It is a winter spawner with a reproductive period ranging from September to May. The gonads progressively mature in autumn with maturity being achieved in winter. Spring and summer are times of gonadal decline and gametogenesis initiation, respectively. The ranges in water temperature and salinity at which mature individuals occur were 13.7–28.0°C and 19.3–29.0‰, respectively. N. festivus produces between 16 and 46 eggs·capsule−1. Over its short life span of 1.21 years, therefore, N. festivus attempts to maximize reproductive output. The eggs hatch as veligers which remain in the plankton for between 33 and 41 days. In terms of its reproductive biology, it is concluded that N. festivus behaves as a typical sub-tropical species with a mix of tropical and temperate characteristics seen in other nassariids from these latitudes.  相似文献

Induction of diapausing amictic eggs in Synchaeta pectinata   总被引:2,自引:0,他引:2  
Amictic females of a clone of S. pectinata from Star Lake (Norwich, Vermont) may produce diapausing as well as non-diapausing (subitaneous) eggs. The proportion of diapausing eggs produced in cultures was unaffected by temperature (12 vs 19 °C) or rotifer population density (minima of 0.33 vs 3 ind. ml–1) at 19 °C. However, at 19 °C this proportion was higher in cultures maintained at a low food level suppressing reproduction (5 × 103 cells ml–1 Cryptomonas erosa) than in those maintained at a high food level (2 × 104 cells ml–1); the treatment effect was marginally significant (p=0.067). Consistent with the effect of low food availability, a period of starvation was very effective in inducing the development of diapausing eggs. None of 19 females cultured individually from hatching at 19 °C on C. erosa (2 × 104 cells ml–1) in 1-ml volumes produced any diapausing eggs in 4 days (0 out of 349 eggs), while 13 out of 16 females subjected to a 15-hour starvation period 6 hours after birth produced one or more diapausing eggs during that time (34% of the 158 eggs produced by the 16 females were diapausing). Diapausing eggs produced and left at 19 °C hatched after 4 to 13 days. Those produced in cultures with a low food level took significantly longer to hatch (9.7 days) than those produced in cultures with a high food level (8.1 days) (p=0.022). In natural communities, S. pectinata should be able to respond directly and rapidly to poor food conditions by producing eggs that undergo an obligatory dormant period before resuming development.  相似文献

To develop an effective fermentation system for producing Escherichia coliphytase AppA2, we expressed the enzyme in three inducible yeast systems: Saccharomyces cerevisiae (pYES2), Schizosaccharomyces pombe (pDS472a), and Pichia pastoris (pPICZ A), and one constitutive system: P. pastoris (pGAPZA). All four systems produced an extracellular functional AppA2 phytase with apparent molecular masses ranging from 51.5 to 56 kDa. During 8-day batch fermentation in shaking flasks, the inducible Pichia system produced the highest activity (272 units ml–1 medium), whereas the Schizo. pombesystem produced the lowest activity (2.8 units ml–1). The AppA2 phytase expressed in Schizo. pombehad 60–75% lower Kmfor sodium phytate and 28% higher heat-stability at 65 °C than that expressed in other three systems. Two serine carboxypeptidases, MpiCP-1 and MpiCP-2, were purified to homogeneity from Monascus pilosus IFO 4480. MpiCP-1 is a homodimer with a native molecular mass of 125 kDa composed of two identical subunits of 61 kDa, while MpiCP-2 is a high mass homooligomer with a native molecular mass of 2,263 kDa composed of about 38 identical subunits of 59 kDa. This is unique among carboxypeptidases and distinguishes MpiCP-2 as the largest known carboxypeptidase. The two purified enzymes were both acidic glycoproteins. MpiCP-1 has an isoelectric point of 3.7 and a carbohydrate content of 11%, while for MpiCP-2 these values were 4.0 and 33%, respectively. The optimum pH and temperature were around 4.0 and 50°C for MpiCP-1, and 3.5 and 50°C for MpiCP-2. MpiCP-1 was stable over a broad range of pH between 2.0 and 8.0 at 37°C for 1 h, and up to 55°C for 15 min at pH 6.0, but MpiCP-2 was stable in a narrow range of pH between 5.5 and 6.5, and up to 50°C for 15 min at pH 6.0. Phenylmethylsulfonylfluoride strongly inhibited MpiCP-1 and completely inhibited MpiCP-2, suggesting that they are both serine carboxypeptidases. Of the substrates tested, benzyloxycarbonyl-l-tyrosyl-l-glutamic acid (Z-Tyr-Glu) was the best for both enzymes. The Km, Vmax, Kcat and Kcat/Km values of MpiCP-1 for Z-Tyr-Glu at pH 4.0 and 37°C were 1.33 mM, 1.49 mM min–1, 723 s–1 and 545 mM–1 s–1, and those of MpiCP-2 at pH 3.5 and 37°C were 1.55 mM, 1.54 mM min–1, 2,039 s–1 and 1,318 mM–1 s–1, respectively.  相似文献

A proteolytic thermophilic bacterial strain, designated as strain SF03, was isolated from sewage sludge in Singapore. Strain SF03 is a strictly aerobic, Gram stain-positive, catalase-positive, oxidase-positive, and endospore-forming rod. It grows at temperatures ranging from 35 to 65°C, pH ranging from 6.0 to 9.0, and salinities ranging from 0 to 2.5%. Phylogenetic analyses revealed that strain SF03 was most similar to Saccharococcus thermophilus, Geobacillus caldoxylosilyticus, and G. thermoglucosidasius, with 16S rRNA gene sequence identities of 97.6, 97.5 and 97.2%, respectively. Based on taxonomic and 16S rRNA analyses, strain SF03 was named G. caldoproteolyticus sp. nov. Production of extracellular protease from strain SF03 was observed on a basal peptone medium supplemented with different carbon and nitrogen sources. Protease production was repressed by glucose, lactose, and casamino acids but was enhanced by sucrose and NH4Cl. The cell growth and protease production were significantly improved when strain SF03 was cultivated on a 10% skim-milk culture medium, suggesting that the presence of protein induced the synthesis of protease. The protease produced by strain SF03 remained active over a pH range of 6.0–11.0 and a temperature range of 40–90°C, with an optimal pH of 8.0–9.0 and an optimal temperature of 70–80°C, respectively. The protease was stable over the temperature range of 40–70°C and retained 57 and 38% of its activity at 80 and 90°C, respectively, after 1 h.  相似文献

Quantitative population dynamical information derived from laboratory- and field-based experiments is provided for the fish-parasitic copepod, Lernaeocera branchialis, infecting flounder (Platichthys flesus) and whiting (Merlangius merlangus). Adult, post-metamorphosis females from whiting can produce more than one set of egg-strings. The mean number of eggs in each egg-string pair was 1445. At 10 °C these eggs took about 12.7 d after extrusion before hatching of NI nauplii began. Hatching took up to 12 days to be completed with an exponentially declining pattern of output over this period. In the laboratory about 44% of the egg-string egg population successfully passed through the NI to NII nauplius moult and the NII to copepodid moult to produce infective copepodids, a process lasting about 2 d. The non-feeding copepodids had a maximum survival time at 10 °C of 18 d, with a time to 50% survival of 7.5 d. In laboratory infection experiments at 10 °C, copepodids infected flounder and passed through all their developmental stages to adulthood and copulation in a minimum of 25 d. Field experiments on the seabed off Lowestoft in June 1987 with a sea temperature of about 16 °C suggested that the developmental period in those conditions could be as short as 11 d. Previously uninfected flounder in the field experiments became naturally infected with copepodids at a mean rate of not less than 30 parasites per fish d–1.  相似文献

The gene, encoding malate synthase (MS), aceB, was cloned from the thermophilic bacterium Streptomyces thermovulgaris by homology-based PCR. The 1,626-bp cloned fragment encodes a protein consisting of 541 amino acids. S. thermovulgaris malate synthase (stMS) gene was over-expressed in Escherichia coli using a glutathione-S transferase (GST) fusion vector (pGEX-6P-1), purified by affinity chromatography, and subsequently cleaved from its GST fusion partner. The purified stMS was characterized and compared to a mesophilic malate synthase (scMS) from Streptomyces coelicolor. stMS exhibited higher temperature optima (40–60 °C) than those of scMS (28–37 °C). It was more thermostable and very resistant to the chemical denaturant urea. Amino acid sequence comparison of stMS with four mesophilic streptomycete MSs indicated that they share 70.9–91.4% amino acid identities, with stMS possessing slightly more charged residues (~31%) than its mesophilic counterparts (~28–29%). Seven charged residues (E85, R187, R209, H239, H364, R382 and K520) that were unique to stMS may be selectively and strategically placed to support its peculiar characteristics.  相似文献

A thermotolerant methylotrophicBacillus sp. (KISRI TM1A, NCIMB 40040), isolated from the Kuwaiti environment and belonging to the group II spore-forming, bacilli, could not be correlated with any knownBacillus sp. It may, therefore, be a new species. It grew at temperatures from 37° to 58°C from pH 6.5 to 9.0 and on methanol up to 40 g l–1. It grew well in a chemostat. Its biomass yield coefficient was improved by about 30% by optimization of medium and growth conditions, reaching a maximum of 0.44g g–1 at 45°C pH 6.8 to 7.0, dilution rate 0.25 h–1 with methanol at 10 g l–1. Average crude protein and amino acid content were 84% and 60%, respectively, and maximum productivity attained under laboratory conditions was 5.06 g l–1h–1. It was concluded that this strain has good potential for use in single-cell protein production.  相似文献
