Enterocytozoon salmonis was transmitted to chinook salmon Oncorhynchus tshawytscha by feeding tissues infected with the parasite and by cohabitation of noninfected fish with experimentally infected fish. Affected fish (dead and survivors) in both transmission trials had gross and microscopic signs of the disease and merogonic and sporogonic stages of the parasite. There were no morbidities or mortalities, or evidence of the parasite among control fish in either study. Results suggest that the parasite may be contracted by indirect contact among healthy and infected fish held in crowded ponds or net pens or by direct ingestion of spores found in the water.  相似文献

ABSTRACT. The microsporidium, Enterocytozoon bieneusi , causes a severe, debilitating, chronic diarrhea in patients with the acquired immunodeficiency syndrome. Specific diagnosis of intestinal microsporidiosis, especially due to Enterocytozoon , is difficult and there is no known therapy that can completely eradicate this parasite. Preliminary studies indicate that a short term (about 6 months) in vitro culture of this parasite yielding low numbers of spores, may be established by inoculating human lung fibroblasts and/or monkey kidney cell cultures with duodenal aspirates and or biopsy from infected patients. The cultures may subsequently be used for the isolation and molecular analysis of parasite DNA.  相似文献

小鼠卵母细胞体外成熟、体外受精的效果观察   总被引:2,自引:0,他引:2  
目的 研究不同培养条件对小鼠卵母细胞体外成熟及体外受精率的影响。方法 小鼠卵母细胞分别在含有FSH、BSA和胰岛素的培养液中体外成熟,在Whitten 氏液中体外受精,比较体外成熟率、体外受精率。结果 1- 裸卵(DO) 的体外成熟率、体外受精率(81-4% ,31-0 % ) 均高于卵丘卵母细胞复合体(COC)(48-6 % ,27-1% ) 。2- 在培养液中添加FSH、胰岛素和BSA,卵母细胞的体外成熟率为77-9 % ,82-3% 、60-7% ;体外受精率为77-2 % 、72-6 % 、26-7% ;2 - 细胞率为49-2 % 、34-2 % 、10-0% 。胰岛素组的卵母细胞IVM 率最高,但IVF率、2 - 细胞率低于FSH 组。3- 添加BSA的两组的体外受精率只有26-7 % 、25-8 % ,显著低于其他组,其体外成熟率也较添加FSH 和胰岛素的组成。4- 排出第一极体(PbI) 的卵母细胞的体外受精率和2 - 细胞率(85-9 % ,22-4% ) 均高于GV期卵母细胞(71-1 % ,12-9 % ) 。结论 1- 卵丘卵母细胞(COC) 较裸卵(DO) 的体外成熟率、体外受精率都低,差异显著(P成熟< 0-01;P受精< 0-05) 。2-FSH 和胰岛素均能提高小鼠卵母细胞的体外成熟率、体外受精率。3-BSA可以降低小鼠卵母细胞体外受精率,差异极显著。4-GV 期卵母细胞的体外受精率显著低于体外培养的排出第一极体的卵母细胞(P2 - cell < 0-05,P受精<0-05)  相似文献

Tubulinosema ratisbonensis is a microsporidian pathogen of Drosophila melanogaster belonging to the family Tubulinosematidae. The microsporidia in this family mainly cause infections in invertebrate hosts, but two members of this family, Brachiola vesicularum and Brachiola algerae, have been found to cause infections in humans as well. Moreover, B. algerae can be transmitted to immunodeficient mice and grows in mammalian cell cultures. Thus, the examination of the opportunistic properties of other members of the family Tubulinosematidae is important. Spores of T. ratisbonensis, isolated from infected fruit flies, were used to inoculate mammalian and insect cell cultures. Parasite growth was only seen in human lung fibroblasts. No growth was seen in Vero cells or insect cell cultures. Comparison of growth kinetics at 31 degrees C and 37 degrees C showed that there were fewer and smaller parasitic foci in cultures incubated at 37 degrees C. Transmission electron microscopy revealed the typical ultrastructure of T. ratisbonensis, and scanning electron microscopy showed oval or slightly pyriform spores, with some spores having extruded their polar tubes. The PCR-amplified sequences of rDNA fragments from infected cell cultures were 100% identical to the original T. ratisbonensis rRNA sequence. As T. ratisbonensis is able to proliferate in mammalian cell cultures, it may have the opportunistic properties of other members of the family Tubulinosematidae.  相似文献

以美国引进的越橘属植物为材料,取其成龄植株的茎段或茎尖为外植体,培养在添加各种激素配比的改良WPM的培养基上,研究了影响克隆体外繁殖的因素,筛选离体最佳增殖和生根培养基及培养条件.结果表明:在改良WPM添加Zt 1.0 mg/L的增殖培养基上,增殖系数最大达50,在1/2WPM IBA 0.3 mg/L的生根培养基上生根率因外植体的基因型不同而有差异,可达到30%~70%.通过实验室的半无菌炼苗后,试管苗移栽到温室的成活率达60%.  相似文献

对用于猪体外受精(IVF)的研究方法和技术,如传统的液滴IVF、透明带下注射精子受精(SUZI)、卵母细胞质内单精注射受精(ICSI)及细管IVF等进行了简述。与其它动物相比,进行猪卵的体外受精研究,多精受精现象特别明显。大量的研究表明,猪卵的多精受精不但与其品种特性有关,而且与卵母细胞成熟的程度、透明带的异常、受精时获能精子的浓度、输卵管分泌物、受精液蛋白添加成分、NaHCO3浓度、咖啡因、pH值以及温度等因素密切相关。  相似文献

采用75分钟和150分钟两种精卵作用时间,对经6℃低温处理2小时和6小时的小鼠卵母细胞进行体外受精。精卵作用75分钟后,经6℃处理的卵子无一受精,而对照组的受精率为30.7%。作用150分钟后,低温处理2小时、6小时和对照组的受精率分别为62.0%,36.55%和76.0%。并对试验所出现的现象作了讨论。  相似文献

ABSTRACT. The effect of low incubation temperature on synchronized cultures of Plasmodium falciparum was studied. Young trophozoites that were maintained at 28°C matured slowly and invaded poorly. Growth seemed to arrest when parasites reached a maturation equivalent to 30 h, although they reestablished their growth normally when returned to 37°C. On the other hand, 36-h synchronized parasites that were transferred to 28°C completed their cell cycle with a 12-16 h delay, but without changes in the parasite as seen by light microscopy and without a diminution in the efficiency of the invasion or in the incorporation of 35S-methionine. These results might be useful for obtaining parasites at defined stages of development at the desired time.  相似文献

Abstract: The effect of l -glutamate on the adrenergic-stimulated release of melatonin in the rat pineal gland was examined using an in vitro perfusion system. l -Glutamate by itself had no effect on melatonin secretion whereas l -glutamate administered prior to (–)-isoproterenol (β-adrenergic agonist) and l -phenylephrine (α-adrenergic agonist) inhibited melatonin production by 42%. l -Glutamate did not inhibit melatonin secretion when glands were stimulated with (–)-isoproterenol alone. d -Glutamate, as well as the l -glutamate agonists kainate, N -methyl- d -aspartate, quisqualate, and trans -1-aminocyclopentane-1, 3-dicarboxylic acid, had no effect on the (–)-isoproterenol-and l -phenylephrine-stimulated secretion of melatonin, which suggests that the inhibitory effects of glutamate are not mediated via any of the known glutamate receptor subtypes. The possibility that l -glutamate may be converted to another neuroactive compound (GABA) prior to the addition of (–)-isoproterenol and l -phenylephrine is suggested by the observation that simultaneous administration of l -glutamate with (–)-isoproterenol and l -phenylephrine did not inhibit melatonin production.  相似文献

Effects of Neurotransmitters on Astrocyte Glycogen Stores In Vitro   总被引:2,自引:4,他引:2  
We have used receptor binding assays to determine the presence of three neurotransmitter receptors in a crude membrane fraction derived from neonatal rat cortical astrocyte cultures and subsequently determined the effects of transmitter receptor activation on astrocyte glycogen content in vitro. beta-Adrenergic (KD = 88 pM; Bmax = 51 fmol/mg of protein), serotonin (KD = 70 nM; Bmax = 44 pmol/mg of protein), and muscarinic cholinergic receptors (KD = 79 pM; Bmax = 44 fmol/mg of protein) were found to be present on astrocyte membranes using [3H]dihydroalprenolol, [3H]serotonin, and [3H]quinuclidinyl benzilate, respectively, as ligands. Astrocyte cultures exposed to noradrenaline but not specific alpha- and beta-receptor agonists contained 33% less glycogen than controls. Neither serotonin nor carbachol caused alterations in astrocyte glycogen content under normal conditions. Reserpine-treated cultures, however, responded to serotonin with a 28% decrease in glycogen content and contained higher levels of glycogen than non-reserpine-treated controls (a 55% increase). These results show that both noradrenaline and serotonin can evoke astrocyte glycogenolysis and that noradrenergic control of glycogen metabolism is probably exerted through both alpha- and beta-receptors. Neurotransmitter control of astrocyte glycogen turnover may represent a form of neuron-astrocyte signalling in addition to that provided by changes in external potassium concentration.  相似文献

Abstract: We have studied the posttranslational processing of actin molecules synthesized in a cell-free system. The results of these experiments indicate that during the in vivo synthesis of the actins from rat brain the primary translational products undergo two different and sequential posttranslational modifications. These modifications are accompanied by slight changes in the isoelectric points of the proteins and can be detected by isoelectric focusing analysis. The same posttranslational modifications can be detected during the in vitro synthesis of chick embryo skeletal muscle actin. The evidence presented suggest that the first posttranslational modification may correspond to the methylation of a histidine residue, and the second modification most likely corresponds to the acetylation of the NH2-terminal amino acid residues of actin molecules.  相似文献

本研究以羔羊卵巢母细胞为材料,把体外受精技术应用到绒山羊育种工作中,观察了羔羊体外受精卵的体外发育及移植产羔的效果,以便探讨通过体外受精技术缩短绒山羊育种周期的技术途径。对406 枚羔羊体外受精卵用M199 hcG、M199 LH 和M199 hcG EGF三种培养基进行体外发育培养。结果是,2~4 细胞期胚的发育率平均分别为40-0 % 、43-6 % 和49-6 % ;桑椹胚和囊胚的发育率平均分别为17-5 % 、15-8 % 和18-8 % 。将49 枚F1 代羔羊2~8 细胞期胚移植给31 只受体母羊,结果有11 只妊娠(35-5 % ) 并产羔12 只(F2 代) ;将24 枚F2 代羔羊2~4 细胞期胚移植给15 只受体母羊,结果有2 只妊娠(13-3 % ) 并产F3 代羔羊2 只。  相似文献

The use of nuclear magnetic resonance (NMR) spectroscopy for characterising microencapsulated tuna oil powders (25% and 50% w/w oil) and assessing the behaviour of the microcapsules on their exposure to water, simulated gastric fluid or to sequential exposure of simulated gastric and intestinal fluids was examined. The matrices used for encapsulation were physical mixtures of casein or whey protein in combination with carbohydrates (dextrose monohydrate with either dried glucose syrup or a physically modified resistant starch) or heated mixtures of these matrices. Solid-state 13C cross-polarised magic angle spinning NMR and dipolar de-coupled magic angle spinning NMR record the 13C NMR signals of the encapsulant material and that of the encapsulated oil, respectively. 1H and 13C solution NMR were used to investigate the relative increase in mobility of the various encapsulant matrices due to their dissolution on exposure to gastrointestinal fluids. The results suggested that the dissolution characteristics of matrices of microencapsulated oil powders were dependent on the type of milk protein and carbohydrate used and whether the protein–carbohydrate matrices were heat-treated prior to encapsulation of the oil.  相似文献

To assess the drug transport across the blood-brain barrier (BBB), we compared the maximal brain extraction values at time 0 [E(0) values] obtained using either in vitro or in vivo methods. The in vitro BBB model consisted of a coculture of brain capillary endothelial cells growing on one side of a filter and astrocytes on the other. The in vivo model used intracarotid injection in anesthetized rats. Eleven compounds were tested. They were selected because they exhibit quantitatively different brain extraction rates: very low for inulin and sucrose, low for oxicam-related nonsteroidal antiinflammatory drugs and diclofenac, and high for propranolol and diazepam. As these compounds are apparently transferred by a passive diffusion mechanism, two others, glucose and leucine, were added that cross the BBB by a known carrier-mediated process. The in vivo and in vitro E(0) values showed a strong correlation as indicated by the Spearman's correlation coefficient (r = 0.88, p less than 0.01). The relative ease with which such cocultures can be produced in large quantities could facilitate the screening of new centrally acting drugs.  相似文献
