For the first time, spruce shoots (Picea abies [L.] Karst.)were fumigated in vivo with 13N-labelled NO2 with the aim ofelucidating the mechanism of NO2 trapping in the apoplastof the substomatal cavity. Uptake by the needles could be monitoredon-line, and a quantitative analysis of the activity recordsdelivered a deposition velocity in agreement with the commondry deposition estimates and ruled out rapid export processes.A fast extraction procedure was applied which revealed thatNO2 did not produce any detectable traces of nitrite. In needlesin which the enzymes of nitrate reduction were not induced byprior fumigation with NO2, incorporation of NO2 was partiallyinhibited as compared to the fully induced shoots which tookup and assimilated NO2 as expected from a constant influx. Theonly labelled inorganic species found in the extracts was nitrate(60%), whereas the rest of the label (40%) was assimilated organicnitrogen.A quantitative analysis of the data shows that thereaction of NO2 in the apoplast yields at least three timesmore nitrate than nitrite, so that the existing models aboutthe apoplastic trapping reaction, disproportionation or antioxidantscavenging, which both postulate substantial production of nitrite,have to be reconsidered. Cuticles act as solution-diffusion membranes for water transport.Diffusion in pores does not contribute to cuticular transpiration.An extensive literature survey of cuticular permeances (P) andminimum leaf conductances (gmin) to water is presented. Thetwo variables cannot be distinguished with most experimentaltechniques. Results from different experiments are in good agreementwith each other for some species, for example, Fagus sytvaticaL., but not for others, such as Picea abies (L.) Karst. In adata set of 313 values of P or gmin from 200 species, distributionsof results obtained with different techniques were found todiffer significantly. Likely reasons include water loss fromincompletely closed or incompletely sealed stomata, and thedependence of P on moisture content of the cuticle and on storagetime of isolated cuticles. Contrasting evidence for an interactionbetween cuticular transpiration and stomatal sensitivity toair humidity is presented. The occurrence of unusually highgmin in trees growing at the alpine treeline and its physiologicalsignificance are discussed. It is shown that gmin is of littlevalue as a predictor for drought resistance of crops, with thepossible exception of Sorghum bicolor L. Moench. Adventitious buds were initiated on embryos of Picea abies (L.)Karst. after a pulse treatment with cytokinin. The initial stagesof bud formation could take place on culture medium lackingsucrose, but sucrose was required for further development ofmeristematic centres into bud primordia and buds. Sucrose atone per cent was optimal for adventitious bud formation. Embryoscultured on media containing sucrose started to accumulate starchduring the first day. Starch accumulation occurred especiallyin the cortex cells where starch grains were frequently presentin the chloroplasts. The starch accumulation increased withhigher sucrose concentrations in the culture medium. Embryoscultured on medium lacking sucrose did not accumulate starchbefore the formation of meristematic centres. Starch accumulationwas never observed in meristematic cells from which adventitiousbud primordia developed. Picea abies (L.) RAPD analysis was used (1) to assess the diversity of indigenousPicea abies within the French massifs (Alps, Jura and Vosges)in comparison with the Hercyno-Carpathian and Alpine domains(central Europe); and (2) to examine the molecular relationshipsamong provenances of these massifs and domains. One hundredand thirty-seven polymorphic RAPD fragments were screened. Resultsshowed that the phenotypic diversity estimates within the differentmassifs and domains analysed were similarly high. Factorialcorrespondence and cluster analyses revealed geographical structuringalong a latitudinal gradient among the French massifs and alongitudinal gradient from eastern Europe to France. Provenancesfrom the southern French Alps appeared to be genetically distinctfrom the others. The distribution of exogenously-supplied radioactive labelledindol-3-yl-acetic acid (IAA) and gibberellin A1 (GA1) in geotropicallystimulated roots of Norway spruce (Picea abies (L.) Karst.)has been demonstrated. Seedlings were positioned with theirroot tips in 2.1 x 10–6 M [14C]IAA or 1.3 x 10–8m 3H-GA1 for 4 and 20 h, respectively. After geotropic stimulationfor 90 min in the horizontal position the root tips were cutlongitudinally in 50 µm thick sections, using a freeze-microtome.The radioactivity in the 14C-IAA treated roots occurred in higherconcentration in the lower than in the upper halves (ratio 1.25:1). A similar trend was observed in the [3H]GA1-treated rootswhere the ratio lower: upper halves was 2.04: 1. The ratio ofradioactivity in right and left halves of vertical roots wasapproximately the same in roots supplied with [14C]IAA and [3H]GA1(1.09: 1). The supplied radioactive compounds were analysed chromatographicallyafter extraction in methanol of 6 mm apical root segments. Onlya small fraction (7–8 per cent) of the supplied [14C]IAAwas revealed unchanged in the segments. The major part of thechromatographed, labelled compound has not been identified,but on basis of its RF value it is suggested that it may beindol-3-acetyl-aspartic acid (IAAasp). The chromatographic analysis of the [3H]GA,-treated segmentsshowed that only small fractions of this gibberellin has beenconverted to other compounds. These results have been discussed and correlated with knowledgeof plant growth regulators and their participation in root geotropism. Leaves of Sorghum bicolor were examined at 5 d and 14 d afterinoculation with the N2-fixing endophytic bacteria Herbaspirillumseropedicae and Herbaspirillum rubrisubalbicans. Plants inoculatedwith H. rubrisubalbicans expressed symptoms of ‘red stripedisease’ i.e. red stripes along the secondary veins ofthe leaf blade close to the inoculation point and spreadingup the leaves. Infected leaves showed dense colonization byH. rubrlsubalbicans in regions showing red stripe symptoms at5 d after inoculation. The infection was confined within thevascular system, in particular, the protoxylem and associatedlacunae, which were often completely filled with bacteria, withsome of the latter expressing nitrogenase. The bacteria wererecognized using H. rubrisubalbicans-speciflc antibodies andimmunogold labelling, which also showed that the antibody reactedwith material on the surface of the bacteria, and that thismucus was released into the lumen of the xylem. At 14 d afterinoculation, disease symptoms were slightly more severe, withboth meta- and protoxylem being even more heavily colonizedin parts of the leaf showing red stripes. However, a stronghost defence response was also apparent at this stage, withgums lining the walls of the vessels and enclosing the bacteria,although the latter were still actively dividing. At the edgesof visible disease symptoms, plant gums filled the xylem; bacteriahad formed distinct colonies within these gums, with some ofthe colonies associated with the xylem walls. Plants inoculatedwith H. seropedicae either did not express the disease or showedvery mild symptoms close to the inoculation point. In the lattercase, H. seropedicae were localized within protoxylem vesselsand the metaxylem was partly occluded with plant-derived gums.By contrast with H. rubrisubalbicans, H. seropedicae was alsolocalized in leaves at 14 d without disease symptoms and didnot always appear to elicit a host response, i.e. they colonizedthe walls of metaxylem, with the xylem vessels themselves remainingunoccluded and largely free of gums. Hydraulic properties of entire root systems and isolated rootsof three contrasting sugarcane clones were evaluated using transpiration-induceddifferences in hydrostatic pressure across intact root systems,root pressure-generated xylem sap exudation, and pressure-fluxrelationships. Regardless of the measurement technique employed,the clones were ranked in the same order on the basis of theirleaf area–specific total root system hydraulic conductance(Croot). All methods employed detected large developmental changesin Grootroot with maximum values occurring in plants with approximately02 m2 total leaf area. Genotypic ranking according to Groot,was reflected as a similar ranking according to root length-specifichydraulic conductance (L) of individual excised roots. Genotypicdifferences in Groot and L were consistent with anatomical characteristicsobserved in individual roots. Patterns of Groot, during soildrying and following re-irrigation suggested that the declinein Groot, observed during soil drying occurred within the rootsrather than at the soil–root interface and may have beencaused in part by xylem cavitation in the roots. Spruce (Picea abies (L.) Karst.) seedlings were asepticallycultivated and the effects of different N-nutrition on net uptakeand reduction of nitrate were investigated. The characteristicsof nitrate uptake were calculated, Ks as 0?2 mol m–3 andVmax as 18 µmol g–1 d–1. Low pH, and Al3+ in the medium caused adecrease in nitrate uptake rate. An in vivo assay was set upwhich allowed the measurement of NRA in both roots and needlesof spruce seedlings. The in vivo nitrate reductase activitywas repressed by ammonium and stimulated by nitrate. Nitratereduction was similar to nitrate uptake, negatively affectedby low pH and ammonium. Bicarbonate-dependent O2-evolving activity in dark-grown cotyledonsof Picea abies was measured with an oxygen electrode with differentpreillumination times. The activity showed a slight linear increasewith increasing preillumination time. On the other hand, O2-evolvingactivity (Hill activity) of chloroplasts prepared from preilluminateddark-grown cotyledons exhibited a characteristic change of asteep rise followed by a gradual increase with increasing preilluminationtime. Five experiments to investigate the water relationships of Scotspine and Norway spruce sapwood are described. Field exposureof stakes, and their moisture characteristic relating wood moisturecontent to the water potential of the surroundings showed thatspruce had a lower moisture content than pine under the sameconditions. A pore size distribution derived from the characteristicsshowed more large pores in spruce than in pine, but in sprucethere were also more small pores. The inference was that thelarge pores of spruce were readily accessible to water, allowinga fast rate of water uptake, as found in the wick action andswelling investigations, while the small pores were not accessible,resulting in a low rate of water uptake and a low rate of swelling.The possibility is briefly discussed that it is the pore sizedistribution of the two woods which determines their water relationships,and the difference in pore size which determines their treatability. We have previously shown that the nitrogen-fixing endophyteof sugarcane,Acetobacter diazotrophicuslives in the sugar solutionin the intercellular-space apoplast of the stem cortex. Variousauthors have claimed that it inhabits the xylem apoplast. Thispossibility has been investigated in the clone Ja 60-5 and shownto be most unlikely for the following reasons: (1) an adequatecarbon source is lacking in the xylem sap, and cannot be suppliedfrom the intercellular-space apoplast of the cortex. Diffusionof solutes into and out of the vascular bundles is preventedby complete lignification and suberization of the bundle sheathcell walls except at the nodes. (2) Longitudinal movement ofparticles as large as bacteria is severely limited at the nodes.Vessel end walls were found in 90% of vessels at each node,and only 1% of open vessels extended through two nodes. Noneextended as far as three nodes. In addition to vessel end walls,vessel continuity at nodes was interrupted by living cells.Dye solution in the transpiration stream in metaxylem vesselsdid not pass through these living cells, but accumulated incrystals (sump formation) in the vessels below the node. Onlyin some protoxylem vessels and cavities did dye solution movethrough many nodes. It is likely that selection of sugarcaneclones such as Ja 60-5 for resistance to bacterial wilt diseaseshave selected for clones that have limited vessel continuity.(3) When culturedA. diazotrophicuswas introduced into the transpirationstream, the xylem parenchyma reacted by secreting a bright redpolymer which killed the bacteria and blocked the movement ofwater. We conclude that the xylem flow-apoplast of this cloneof sugarcane is an unsuitable habitat forA. diazotrophicusandthat additional habitats to those of the intercellular-spaceapoplast should be sought elsewhere. Summary Uptake of water and magnesium chloride solution was investigated through the outer surface of twigs of Picea abies (L.) Karst. Water uptake was determined by using pressure/volume (P/V) curves of the twigs as a basis for calculation to avoid problems of superficial extraneous water. When water was sprayed on bark and needles of 3- to 7-year-old twigs at a xylem water potential of -1.00 MPa, they absorbed as much as 80 mm3 water in 200 min/g twig dry weight as the twig water potential recovered to -0.15 MPa. With fluorescent dyes, pathways for absorption of water and solutes through the twig bark were found, particularly through the radially orientated ray tissue. In addition to uptake by mass flow, magnesium could also diffuse along a concentration gradient from the twig surface into the xylem. In the field, the magnitude of these uptake processes would depend on the concentration of elements deposited by atmospheric precipitation, the concentration gradient between the plant surface and the xylem sap, the xylem water potential and the intensity and duration of each precipitation event.  相似文献

Kouchi, H. and Higuchi, T. 1988. Carbon flow from nodulatedroots to the shoots of soybean {Glycine max L. Merr.) plants:An estimation of the contribution of current photosynthate toureides in the xylem stream.–J. exp. Bot. 39: 1015–1023. Well-nodulated, water-cultured soybean plants were allowed toassimilate 13CO2 at a constant specific activity for 10 h andthe 13C-labelling of total carbon and ureides in xylem sap wasinvestigated. Labelled carbon appeared very rapidly in the xylem stream. Percentageof labelled carbon (relative specific activity, RSA) in xylemsap was 18% at 2 h after the start of 13CO2 assimilation andreached 53% at the end of the 10 h assimilation. The amountof labelled carbon exported from nodulated roots to the shootsvia the xylem during the 10 h labelling period accounted for33% of total labelled carbon imported into the nodulated roots.Ureides (allantoin and allantoic acid) in xylem sap were stronglydependent on currently assimilated carbon. The RSA of ureidesin xylem sap had reached 83% at the end of the assimilationperiod. Labelled carbon in ureides accounted for 51% of totallabelled carbon returned from nodulated roots to the shootsvia the xylem during the 10 h assimilation period. A treatmentwith 20 mol m–3 nitrate in the culture medium for 2 ddecreased the ureide concentration in the xylem sap slightly,but greatly decreased the RSA of ureides. By comparing the data with the results of analysis of the xylemsap of nodule-detached plants, it was concluded that the majorityof labelled carbon exported to the xylem stream from noduleswas in ureide form. A considerable amount of carbon was alsoreturned from roots to shoots via the xylem stream but it wasmore dependent on (non-labelled) carbon reserved in the roottissues. The long-term effects of elevated CO2 and CO2+O3 concentrations on the growth allocation in northern provenances of Norway spruce [Picea abies (L.) Karst.], Scots pine [Pinus sylvestris (L.)] and pubescent birch clones (Betula pubescens Ehrh.) were examined in open-top chambers after a 4-year-long experiment. The total biomass responses of the tree seedlings to increased CO2 and CO2+O3 concentrations were not statistically significant and varied between the provenances and species. The seedlings of northern origin were the least sensitive in their response to treatments. The total biomass of the Norway spruce seedlings slightly decreased in response to CO2 in three provenances. Scots pine from the local provenance had a slight biomass increase after elevated CO2+O3 treatment. The slower-growing birch clone seemed to benefit from elevated CO2, whereas in the faster-growing clone, reductions in biomass accumulation were seen. The combined CO2+O3 treatment reduced the positive effects of elevated CO2, especially in the slower-growing birches. Observations of significant effects were limited to a few parameters. Carbon dioxide treatment decreased needle dry weight of Norway spruce in one northern provenance. The needle and wood dry weight increased (CO2 + O3) in local Scots pine. Significant birch response was limited to increased fine root density (O3 + CO2) in the inland clone. The diverse effects of elevated CO2 and CO2 +O3 on seedling growth and biomass provide evidence that exposure of northern trees to the enhanced variable CO2 and O3 concentrations of the future will have varied effects on the growth of these species. The direction and magnitude of those effects will differ depending on species and origins.  相似文献

Arabidopsis halleri is a Cd hyperaccumulator; however, the mechanismsinvolved in the root to shoot translocation of Cd are not wellunderstood. In this study, we characterized Cd transfer fromthe root medium to xylem in this species. Arabidopsis halleriaccumulated 1,500 mg kg–1 Cd in the shoot without growthinhibition. A time-course experiment showed that the releaseof Cd into the xylem was very rapid; by 2 h exposure to Cd,Cd concentration in the xylem sap was 5-fold higher than thatin the external solution. The concentration of Cd in the xylemsap increased linearly with increasing Cd concentration in theexternal solution. Cd transfer to the xylem was completely inhibitedby the metabolic inhibitor carbonyl cyanide 3-chlorophenylhydrazone(CCCP). Cd concentration in the xylem sap was decreased by increasingthe concentration of external Zn, but enhanced by Fe deficiencytreatment. Analysis with 113Cd-nuclear magnetic resonance (NMR)showed that the chemical shift of 113Cd in the xylem sap wasthe same as that of Cd(NO3)2. Metal speciation with Geochem-PCalso showed that Cd occurred mainly in the free ionic form inthe xylem sap. These results suggest that Cd transfer from theroot medium to the xylem in A. halleri is an energy-dependentprocess that is partly shared with Zn and/or Fe transport. The effects of CO2 on dormancy and germination were examinedusing seeds of cocklebur (Xanthium pennsylvanicum Wallr.) andgiant foxtail (Setaria faberi Herrm.). The rate of germinationof the giant foxtail seeds as well as cocklebur was promotedby exogenously applied CO2 at a concentration of 30 mmol mol-1regardless of the sowing conditions. However, seeds which failedto germinate in the presence of CO2, entered a secondary phaseof dormancy under unfavourable germination conditions. If CO2was applied to seeds under conditions such as water stress imposedwith a 200 mol m-3 mannitol solution, a hypoxic atmosphere of100 mmol mol-1 O2 or a treatment of 0·1 mol m-3 ABA,development of secondary dormancy was accelerated. These contrastedeffects of CO2 were observed in ecological studies. Under naturalfield conditions germination of buried giant foxtail seeds respondedpositively to CO2 during a period of release from primary dormancyfrom Feb. to May, but CO2 accelerated secondary dormancy commencingin early Jun. In other words, in the presence of CO2, both theenvironmental conditions and the germination states of the seedsclearly showed secondary dormancy-inducing effects. Acidic and basic peroxidases, termed as POD-A and POD-B, wereisolated from the apoplastic space of spruce (Picea abies, L.)needles and purified by acetone precipitation and anion exchangechromatography to apparent homogeneity. The molecular massesof POD-A and POD-B were 39.6 and 29.0 kDa, respectively. ThepH optimum of both isozymes ranged from 4.5 to 6. The apparentKm values of POD-A and POD-B were 460 and 210 µM for coniferylalcohol. Both isozymes acted also as NADH oxidases with apparentKm-values of 103 µM (POD-A) and 70 µM (POD-B). NAD+but not NADH was found in the apoplastic space of lignifyingneedles. Based on the lignification rate, the contents and kineticproperties of PODs, NADH oxidation by POD is not the major sourceof H2O2 required for lignin polymerisation. The effects of temperature, photoperiod and chilling on the leafing-out of Norway spruce, Picea abies (L.) Karst. were studied. High temperature promotes breakage of post-dormancy, long photoperiods having no such effect. Photoperiod and chilling cause the breakage of true dormancy. However, under field conditions, photoperiod will have no effect on leafing-out date in the spring. By use of clonal material it was possible to show substantial genetic differences between individuals in response to temperature and photoperiod. When the effect of clones was accounted for, treatments could be compared more precisely. Differences between clones were apparent in heat-sum required for leafing-out, in rapidity of response to favorable post chilling conditions, and in chilling requirement This latter quantity was given a new definition, applicable when both chilling and post chilling temperatures are controlled and specified. This is that period beyond which a further 10 days of chilling accelerates leafing-out by less than one day, i.e. the point at which the slope of the line relating days till leafing-out to chilling period, is equal to minus 0.10. Differences in leafing-out date were shown between provenances taken from throughout the range of Picea abies. These differences were related to latitude with provenances of high latitude leafing-out first.  相似文献
