The pyralid moth Eldana saccharina Walker is an indigenous insect widely distributed throughout sub-Saharan Africa. Studies have shown that populations from West Africa have distinct behavioural differences compared to populations from East and southern Africa. In addition, the parasitoid guilds attacking populations in these different regions are markedly different. This marked geographical variation evoked a hypothesis of genetic differentiation. To evaluate this hypothesis a molecular analysis was conducted on populations of E. saccharina from throughout much of the species’ range, using the cytochrome c oxidase subunit I (COI) region of the mitochondrial genome. A minimum spanning network and a maximum parsimony tree separated the 21 specimens into three distinct groups. Results revealed the presence of substantial genetic differentiation that is related to geographic variation.  相似文献

Abstract:  In an attempt to set up habitat management strategy against Eldana saccharina Walker, an economically important graminaceous stem and cob borer, two laboratory experiments were carried out. The first experiment was designed to assess oviposition preference of E. saccharina for growth stages, plant parts and freshness of different host plants and between host plants species. Zea mays L., Sorghum arundinaceum (Desv.) Stapf, Andropogon gayanus Kunth, Panicum maximum Jacq., Pennisetum polystachion (Linn.) and Cyperus papyrus L. were assessed. Results showed that E. saccharina preferred wild grasses to maize for oviposition. More than 94% eggs were laid on wild grasses. Of all tested grasses, P. polystachion was the most attractive with more than 30% of collected eggs. For all plant species, post-tasseling stage was more attractive than the tasseling and pre-tasseling ones. More than 90% of total eggs were laid on dry leaves and sheaths and less than 10% on fresh leaves. Different characteristics relating to population dynamics of E. saccharina were assessed in the second experiment. Larval survival rate was 10% on Z. mays , 0.50% on S. arundinaceum and around 0% on the other wild grasses. Larval development length was longer on S. arundinaceum than on Z. mays . Females reared on Z. mays were three times more fecund than those reared on S. arundinaceum . Complete life table of females from larvae reared on Z. mays was studied.  相似文献

Abstract:  Notwithstanding the introduction of several pest management tactics, the stalk borer Eldana saccharina Walker (Lep., Pyralidae) remains the most serious pest in South African sugarcane. A novel tactic for managing this pest in sugarcane would be the use of a dead-end trap crop that attracts moths for oviposition and curtails subsequent larval development, thereby reducing pest population size. Glasshouse bioassays, in which moths chose to oviposit on maize producing Bacillus thuringiensis Cry1Ab toxin ( Bt -maize), non- Bt -maize or sugarcane of two cultivars (borer-resistant and -susceptible), showed that E. saccharina laid significantly more eggs and egg batches per dry leaf and unit mass of dry leaf on maize ( Bt or non- Bt ) than on either of the cane cultivars. When moths had a choice of ovipositing on 2-, 3-, 4- or 5-month-old maize ( Bt and non- Bt ), dry leaf number and mass of dry leaf material was significantly correlated with number of eggs and egg batches, indicating that older plants, which carried larger amounts of dry leaf matter, were more attractive for oviposition. Finally, glasshouse assays in which hatching larvae fed on 2.5-, 3.5- and 4.5-month-old Bt and non- Bt -maize plants, showed that the Cry1Ab toxin was effective in killing E. saccharina larvae in all Bt -maize plant growth stages, confirming that Bt -maize fulfilled the third requirement (curtailing larval development) of a dead-end trap crop for this pest. We argue that Bt -maize warrants further testing in the field as a trap crop, both alone and as a component of a 'push–pull' or habitat management system for E. saccharina in sugarcane.  相似文献

In Petri dish and olfactometer tests Goniozus natalensis Gordh (Hymenoptera: Bethylidae) exhibited a host-searching response upon contact with, and at a short distance from, frass of its natural host, Eldana saccharina Walker (Lepidoptera: Pyralidae), whichever the substrate on which E. saccharina had been feeding. The substrates were two host plants, sugarcane or Cyperus papyrus L.; diet media based respectively on sugarcane, papyrus, or cellulose; or a synthetic diet medium, containing no plant material. Host-searching behaviour increased in intensity with age and after mating of G. natalensis females. Parasitoids responded similarly to frass produced by male and female E. saccharina larvae. The substrates themselves elicited little response in G. natalensis. Frass of another borer, Sesamia calamistis Hamps. (Lepidoptera: Noctuidae), fed on sugarcane-based medium, also elicited little response in G. natalensis. The results suggest that frass of E. saccharina larvae contains a material, evidently volatile, to which their parasitoid G. natalensis responds, irrespective of the substrate on which the E. saccharina subsist.  相似文献

Abstract:  The effects of four silicon sources – a USA calcium silicate, a local (South African) calcium silicate, Slagment® and fly ash – on the resistance of sugarcane cultivars (two resistant and two susceptible) to Eldana saccharina Walker (Lepidoptera: Pyralidae) were studied in a potted sugarcane trial. Silicon sources were applied at 5000 or 10 000 kg/ha for the calcium silicates and Slagment; fly ash was applied at 15 000 or 30 000 kg/ha. The greatest increase in plant silicon content (particularly in stalks) was recorded for plants treated with local calcium silicate. Silicon uptake did not vary significantly between the susceptible and resistant cultivars, although the resistant cultivars had inherently higher silicon content than the susceptible ones. Treatment with silicon significantly reduced borer damage and borer performance at the higher treatment level. In general, borer damage and performance decreased with increasing rates of applied silicon and both variables were inversely related with per cent stalk silicon. On average, the higher silicon rate reduced damage by 34% in the susceptible cultivars and by 26% in the resistant cultivars, supporting the argument that susceptible cultivars benefit more from silicon treatments than resistant ones. We propose that calcium silicate amendments could be employed in the integrated, area-wide management of E. saccharina and in the management of soil acidity, both of which are widespread problems in the South African sugar industry.  相似文献

Laboratory trials were conducted to determine the efficacy of four Bacillus thuringiensis (Bt) Cry toxins at five different concentrations (0.016, 0.08, 0.4, 2, 10 μg/mL) for controlling three lepidopteran stem-borer species (i.e., the pyralid Eldana saccharina and the noctuids Busseola fusca and Sesamia calamistis) as well as to evaluate their indirect effect on the braconid larval parasitoid Cotesia sesamiae. In addition, larvae from the treatments above, after having been parasitized, were either fed a contaminated (group 1) or a toxin-free (group 2) diet and compared with a control (i.e., parasitized larvae which have never fed on Bt-toxin). All Bt Cry toxins induced larval feeding inhibition. Compared with the control, significant mortality resulted at all concentrations and for all species of Lepidoptera. Cry1Ab was the most toxic with 10 days post treatment mortalities ranging from 81% in B. fusca and S. calamistis to 100% in E. saccharina. In contrast, Cry1Ac had comparatively low toxicity particularly for B. fusca and S. calamistis (e.g., respectively, 54 and 74% mortality at 10 days at the highest concentration). In the toxin-treated group 1, percentages of C. sesamiae cocoon-producing moth larvae were higher compared to group 2 and the control, whereas clutch size was higher than in group 2 but similar to the control. In both groups, 0.016, 0.08 μg/mL CrylAc yielded lower female-biased sex ratios than the control. It was suggested that paralyses of the moth larvae caused by the toxin may have facilitated parasitization leading to higher parasitism and clutch size.  相似文献

Clinus cottoides is a fish endemic to the coast of South Africa, predominantly inhabiting rock pools. All South African clinids are viviparous, but probably breed throughout the year; as such, their dispersal may be limited, unlike species with pelagic larval stages. We analysed 343 fish from 14 localities on the west, south and east coasts using two mitochondrial genes and the second intron of the S7 ribosomal gene. Mitochondrial DNA analyses recovered significant genetic differentiation between fish populations from the east coast and other sampling locations, with a second break found between Gansbaai and Cape Agulhas on the south coast. Nuclear DNA recovered shallower, but significant, levels of population structure. Coalescent analyses suggested remarkably asymmetrical gene flow between sampling locations, suggesting that the cold Atlantic Benguela Current and Indian Ocean Agulhas counter‐current play important roles in facilitating dispersal. There was no gene flow between the east coast and the other sites, suggesting that these populations are effectively isolated. Divergence times between them were estimated to at least 68 000 years. Neutrality tests and mismatch distributions suggest recent population expansions, with the exception of peripheral western and eastern populations (possibly a consequence of environmental extremes at the edge of the species distribution). Analyses of the current South African marine protected areas network show that it is not connected and that De Hoop, one of South Africa's largest marine reserves, appears to be an important source population of recruits to both the south and southwest coasts.  相似文献

We describe a scenario of plant speciation across a relict forest archipelago in South Africa involving Pleistocene habitat expansion-contraction cycles, dispersal and adaptation to lower temperatures. This is the first population level study using molecular data in South African forests and has significant implications for conservation efforts in this area. Populations of the mesophytic forest floor herbs Streptocarpus primulifolius sensu lato and Streptocarpus rexii were sampled throughout their range in the naturally fragmented forests of eastern South Africa in order to investigate population genetic and phylogenetic patterns within the species complex, using nuclear microsatellites, nuclear ribosomal ITS (internal transcribed spacer) sequences and chloroplast genome sequences. S. primulifolius harbours high levels of genetic diversity at both the nuclear (mean HE = 0.50) and the chloroplast level (each population fixed for a unique haplotype). This is consistent with populations of these coastal species being Pleistocene relicts. In contrast, populations of S. rexii in cooler habitats at higher altitudes and lower latitudes harbour little or no nuclear genetic diversity (mean HE = 0.09) and most share a common chloroplast haplotype. The split of S. rexii from populations intermediate between the two species (S. cf. primulifolius) occurred between 0 and 0.44 million years ago according to the calibrated ITS phylogeny of the taxa. The low genetic diversity and homogeneity of S. rexii is congruent with this species having reached its current range during the Holocene. We found no evidence of monophyly of any of the taxa in this study, which we consider a consequence of recent evolution in a fragmented habitat.  相似文献

Although Porphyra is commercially farmed in many countries, in South Africa only small harvests of wild populations for sale as nori have been carried out. The discovery that Porphyra improves growth of South African abalone (Haliotis midae) farmed inland-based tanks has led to increased pressure to harvest wild populations. This paper reports on a survey of the distribution and seasonality of Porphyra in the southern Western Cape. Porphyrawas present at all sites surveyed, and showed considerable temporal variation. A significant amount of the Porphyra present is in reserves and therefore protected from harvesting. Close rexamination of one site revealed seasonal populations of Porphyra that occupied different niches dependent on season. Recruitment peaked in spring and autumn, leading to dense summer and winter populations. Summer populations generally grew lower in the eulittoral than winter populations. No pattern in the mortality of larger thalli wasde tected, though sporeling mortality was high following recruitment peaks. Although it seems that most sites in the southern Western Cape are suitable for harvesting, the taxonomy of the genus in the region urgently needs revision if populations are to be appropriately managed. SUMMARY 1. The net‐winged midges (Diptera: Blephariceridae), with highly specific habitat requirements and specialised morphological adaptations, exhibit high habitat fidelity and a limited potential for dispersal. Given the longitudinal and hierarchical nature of lotic systems, along with the geological structure of catchment units, we hypothesise that populations of net‐winged midge should exhibit a high degree of population sub‐structuring. 2. Sequence variation in the cytochrome c oxidase subunit I (COI) region of the mitochondrial DNA (mtDNA) was examined to determine patterns of genetic variation and infer historical and contemporary processes important in the genetic structuring of populations of Elporia barnardi. The DNA variation was examined at sites within streams, between streams in the same range, and between mountain ranges in the south‐western Cape of South Africa. 3. Twenty‐five haplotypes, 641 bp in length, were identified from the 93 individuals sampled. A neighbour‐joining tree revealed two highly divergent clades (~5%) corresponding to populations from the two mountain ranges. A number of monophyletic groups were identified within each clade, associated with individual catchment units. 4. The distribution of genetic variation was examined using analysis of molecular variance (amova ). This showed most of the variation to be distributed among the two ranges (~80%), with a small percentage (~15%) distributed among streams within each range. Similarly, variation among streams on Table Mountain was primarily distributed among catchment units (86%). A Mantel's test revealed a significant relationship between genetic differentiation and geographical distance, suggesting isolation by distance (P < 0.001). 5. Levels of sequence divergence between the two major clades, representing the two mountain ranges, are comparable with those of some intra‐generic species comparisons. Vicariant events, such as the isolation of the Peninsula mountain chain and Table Mountain, may have been important in the evolution of what is now a highly endemic fauna. 6. The monophyletic nature of the catchment units suggests that dispersal is confined to the stream environment and that mountain ridges provide effective physical barriers to dispersal of E. barnardi.  相似文献

A total of 1506 soil samples from different habitats in seven geographic regions of South Africa were evaluated for the presence of entomopathogenic nematodes (EPN). Nematodes were isolated from 5% of the samples. Among the steinernematids, four Steinernema sp. were recovered including Steinernema khoisanae and three new undescribed species. Although steinernematids were recovered from both humid subtropical and semiarid regions, this family accounted for 80% of EPN recovered from the semiarid climate zones characterised by sandy, acidic soils. Eight isolates of S. khoisanae were recovered from the Western Cape province. One of the new undescribed steinernematids (Steinernema sp. 1) was recovered only from the Free State and KwaZulu-Natal provinces where humid subtropical conditions prevail and soils are generally less acidic with higher clay content. A high level of adaptation, however, was noted with Steinernema sp. 2, which was recovered from a wide range of soil conditions and habitats ranging from semiarid (Western Cape province) to humid subtropical (KwaZulu-Natal province). A third undescribed steinernematid, Steinernema sp. 3, seemed better adapted to heavier soils with more than 80% of isolates recovered from fruit orchards in the Free State province. Heterorhabditis bacteriophora was the only heterorhabditid recovered during this survey. This species was particularly prevalent in four provinces ranging from humid subtropical to semiarid regions. Isolation of EPN directly from insect cadavers included Steinernema sp. 2 and one H. bacteriophora from an unidentified white grub (Scarabaeidae) cadaver (i.e., dual infection) and H. bacteriophora from the black vine weevil, Otiorhynchus sulcatus.  相似文献

The most severe mass extinction of marine species and terrestrial vertebrates and plants is associated with the Permian-Triassic boundary (∼251 Ma). The extinction interval is also marked by the disappearance of most Late Permian gymnosperm palynomorphs at a layer containing solely the abundant remains of fungi. This ‘fungal spike’ apparently represents widespread devastation of arboreous vegetation. Stratigraphic and palynological study of the Carlton Heights section in the southern Karoo Basin of South Africa revealed a 1-m-thick fungal spike zone that occurs simultaneously with the last appearance of typically Late Permian gymnosperm pollen. The plant extinction and fungal spike zone are found above the last occurrence of Late Permian mammal-like reptiles of the Dicynodont Zone at other Karoo sections. Using the fungal event as a time line in marine and non-marine sections allows placement of the marine extinctions and the extinction of terrestrial plants and reptiles within a brief crisis interval of less than about 40?000 years at the end of the Permian.  相似文献

The findings in this paper show that Arabian Gulf (Abu Dhabi, Dubai, Sharjah) corals have already been measurably affected by climate change and further negative impacts are expected. Corals in South Africa have been only weakly impacted and are expected to persist in this likely refuge. The Arabian Gulf has recently experienced high-frequency recurrences of temperature-related bleaching (1996, 1998, 2002). First evidence may suggest that bleaching patterns in corals changed due to phenotypic adaptation after two strong bleaching events in rapid succession, because Acropora, which during the 1996 and 1998 events always bleached first and suffered heaviest mortality, bleached less than all other corals in 2002 at Sir Abu Nuair and recovered at Jebel Ali and Ras Hasyan. In South Africa, reef corals largely escaped the mass mortalities observed across the tropics in the late 1990s, although bleaching has also increased since 1999. These reefs are protected by local small-scale upwelling events in summer that, if they occur at the right time, keep temperatures below bleaching levels. Both areas, the Arabian Gulf and South Africa, have rich coral faunas but little to no recent reef-framework production. It is possible that many reefs worldwide may have similar dynamics in the future, if the changed climate (recurrence of temperature anomalies, changes in aragonite saturation state, etc.) suppresses sustained reef building at least temporarily. Global climate models predict the possibility of significant environmental changes, including increases in atmospheric temperature, sea-surface temperature (SST), and sea level. Monsoon and El Niño Southeastern Oscillation (ENSO) patterns might change, but climate models are not conclusive. Sea-level rise by up to 0.88 m is expected to be a problem in some low-lying areas, like the southern Arabian Gulf. Ocean aragonite saturation state is predicted to fall throughout the ocean but may not change reef dynamics in the two study areas.  相似文献

Medicinal plants cover a broad range of taxa, which may be phylogenetically less related but morphologically very similar. Such morphological similarity between species may lead to misidentification and inappropriate use. Also the substitution of a medicinal plant by a cheaper alternative (e.g. other non-medicinal plant species), either due to misidentification, or deliberately to cheat consumers, is an issue of growing concern. In this study, we used DNA barcoding to identify commonly used medicinal plants in South Africa. Using the core plant barcodes, matK and rbcLa, obtained from processed and poorly conserved materials sold at the muthi traditional medicine market, we tested efficacy of the barcodes in species discrimination. Based on genetic divergence, PCR amplification efficiency and BLAST algorithm, we revealed varied discriminatory potentials for the DNA barcodes. In general, the barcodes exhibited high discriminatory power, indicating their effectiveness in verifying the identity of the most common plant species traded in South African medicinal markets. BLAST algorithm successfully matched 61% of the queries against a reference database, suggesting that most of the information supplied by sellers at traditional medicinal markets in South Africa is correct. Our findings reinforce the utility of DNA barcoding technique in limiting false identification that can harm public health.  相似文献

We aimed to (i) assess the extant genetic diversity of the riparian relict tree Pterocarya fraxinifolia across its current distribution range in the South Caucasus, including the past refugial areas Colchis and Hyrcan, and (ii) test if a separation of these areas is reflected in its phylogeographic history. Genetic diversity of natural populations was examined using nuclear microsatellite and plastid DNA markers. Spatial genetic structure was evaluated using Bayesian clustering methods and the reconstruction of plastid DNA networks. Divergence times of Colchic and Hyrcanian populations were estimated via divergence dating using a relaxed molecular clock. Allelic richness, private allelic richness, and expected heterozygosity were significantly higher in Hyrcan than in Colchis and the Greater Caucasus, and significant genetic differentiation was revealed between the two groups. Whereas only two plastid haplotypes were detected for the Colchic and Caucasian populations, the Hyrcanian populations displayed 11 different haplotypes. Significant isolation by distance was detected in Hyrcan. The most recent common ancestor of all P. fraxinifolia haplotypes was dated to a time well before a suggested glaciation period in the Caucasus during the late Pliocene (5.98 Ma [11.3–2.48 Ma HPD]). The widespread Colchic haplotype that also occurs along the southern slope of the Greater Caucasus and reaches south-eastern Azerbaijan has appeared more recently (0.24 Ma [1.41–0 Ma HPD]). This diversification pattern of Colchic haplotypes from ancient Hyrcanian haplotypes suggests a colonization of the region from south-east to north-west that predates the last glacial maximum (LGM). Natural populations of P. fraxinifolia show low-to-intermediate levels of genetic diversity and a significant decrease of diversity from Hyrcan to Colchis. However, the genetic differentiation between Colchic-Caucasian and Hyrcanian populations for nuclear markers suggests that independent gene pools existed in both areas at least since the LGM. Particular attention to conservation seems justified for the more diverse Hyrcanian populations.  相似文献

Largemouth bass Micropterus salmoides are a popular North American angling species that was introduced into South Africa in 1928. To enhance the largemouth bass fisheries, Florida bass Micropterus floridanus were introduced into KwaZulu Natal, South Africa, in 1980. Knowledge on the status of M. floridanus in South Africa is required, because it lives longer and reaches larger sizes than M. salmoides, which may result in heightened impacts on native biota. Because M. floridanus are morphologically similar, but genetically distinct from M. salmoides, the distribution of this species was assessed by genetically screening 185 Micropterus sp. individuals sampled from 20 localities across South Africa using the mitochondrial ND2 gene. Individuals with mitochondrial DNA matching M. salmoides were recovered from 16 localities, whereas M. floridanus mitochondrial DNA was recovered from 13 localities. At nine localities (45%), the mitochondrial DNA of both species was detected. These results demonstrate M. floridanus dispersal to multiple sites across South Africa.  相似文献

Patterns of gene flow and genetic structuring were examined in the canyon treefrog, Hyla arenicolor (Cope). Hierarchical analysis of genetic variation was performed on mitochondrial cytochrome b haplotypes from 323 individuals, representing 32 populations from previously described phylogeographic regions. Results from AMOVA revealed that 60.4-78.9% of the recovered genetic variation was the result of differences in the appointment of genetic variation between subdivisions of the primary phylogeographic regions. In contrast, populations only contained between 13.9 and 30.1% of the observed haplotypic variation. Gene flow estimates based on calculations of phi ST revealed moderate levels of gene flow within phylogeographic regions, but there was no evidence of gene flow between these regions, suggesting that geographical boundaries were probably important in the formation of phylogeographic structure in H. arenicolor. Phylogeographic regions exhibited very different patterns of gene flow. One region showed evidence of recent colonization. Another region exhibited very limited gene flow. Moderate to high estimates of gene flow were obtained for populations from two distinct phylogeographic regions characterized by mesic and xeric environments. Isolation by distance was observed in both regions suggesting that these regions are in genetic equilibrium. Because gene flow is extremely unlikely between the populations in the xeric region, this result is interpreted as historical gene flow. These results indicate that isolation-by-distance effects may still be observed even when population genetic structure and gene flow are the result of historical association.  相似文献

The monotypic South African alderfly genus Leptosialis Esben-Petersen, 1920 is reviewed and Leptosialis africana Esben-Petersen, 1920 is redescribed. In the process a new species of alderfly Leptosialis necopinatasp. n. from the Eastern Cape and KwaZulu-Natal provinces of South Africa is recognised and described. Within Sialidae the new species most closely resembles Leptosialis africana. A key to the two species of Leptosialis using both adult and larval characters is provided.  相似文献
