Recent advances in DNA analysis allow us to identify an unprecedented number of insect samples collected by mass sampling techniques such as insect traps. In these circumstances, a preservative that can be applied from trap to storage is necessary to prevent degradation of DNA before analysis and save on the cost of labor for collecting insects from traps. Propylene glycol has a prominent feature as a trap solution. We aimed to examine the DNA preservability of 98% propylene glycol at 2 weeks and more than 6 months after initial collection in comparison with 99.5% ethanol, which is commonly used for storage of specimens for genetic analysis. We compared amplification performance of PCR targeting a specific region of the mitochondrial cytochrome c oxidase subunit I (COI) gene in the orders Hymenoptera, Diptera, and Coleoptera using two extraction methods varying in extraction efficiency. Even after 6 months, more than 75% of samples were recognized to have succeeded in PCR amplification irrespective of preservatives by the extraction method with higher extraction efficiency. It suggested that mitochondrial DNA was preserved in both solutions. However, dim bands in the electrophoreses of PCR products increased with time in extracts by another method with lower extraction efficiency. In Diptera and Coleoptera, the rate of dim bands increased more rapidly for ethanol-preserved than for propylene glycol-preserved specimens, indicating higher DNA preservability of propylene glycol over time for these taxa. On the other hand, in Hymenoptera, the preservatives did not affect PCR amplification performance. Considering its safer characteristics and high DNA preservability in a wide range of taxa, propylene glycol can be a promising solution from trapping of insects to storage for genetic analysis.  相似文献

Globally, many insect populations are declining, prompting calls for action. Yet these findings have also prompted discussion about sampling methods and interpretation of long‐term datasets. As insect monitoring and research efforts increase, it is critical to quantify the effectiveness of sampling methods. This is especially true if sampling biases of different methods covary with climate, which is also changing over time. We assess the effectiveness of two types of flight intercept traps commonly used for beetles, a diverse insect group responsible for numerous ecosystem services, under different climatic conditions in Norwegian boreal forest. One of these trap designs includes a device to prevent rainwater from entering the collection vial, diluting preservatives and flushing out beetles. This design is compared to a standard trap. We ask how beetle capture rates vary between these traps, and how these differences vary based on precipitation levels and beetle body size, an important species trait. Bayesian mixed models reveal that the standard and modified traps differ in their beetle capture rates, but that the magnitude and direction of these differences change with precipitation levels and beetle body size. At low rainfall levels, standard traps catch more beetles, but as precipitation increases the catch rates of modified traps overtake those of standard traps. This effect is most pronounced for large‐bodied beetles. Sampling methods are known to differ in their effectiveness. Here, we present evidence for a less well‐known but likely common phenomenon—an interaction between climate and sampling, such that relative effectiveness of trap types for beetle sampling differs depending on precipitation levels and species traits. This highlights a challenge for long‐term monitoring programs, where both climate and insect populations are changing. Sampling methods should be sought that eliminate climate interactions, any biases should be quantified, and all insect datasets should include detailed methodological metadata.  相似文献

Malaise traps are typically used to sample a range of flying insect groups; however non-target taxa such as spiders may also be collected in large numbers. In this study, spiders were sampled in peatlands and wet grasslands and catches in Malaise and pitfall traps were compared in order to determine the adequacy of Malaise traps for use in spider biodiversity assessment. Overall, the number of species and individuals caught in Malaise and pitfall traps were comparable, although more species were sampled in Malaise traps in locations with a greater structural diversity of the vegetation. The spider fauna sampled by the Malaise traps differed from that of the pitfall traps, but both methods consistently separated the species assemblages by biotope. These results demonstrate that Malaise traps are effective at sampling spiders and indicate that they can be used in biodiversity assessment. In addition the complementary species sampled by each method mean that employing both techniques will be useful where a full inventory of the species is required. The authors do not suggest that Malaise traps should be used solely to sample spiders; however, if traps are set to collect insects, identification of the spiders sampled may reduce the need to employ additional sampling techniques.  相似文献

We present experimental results and modeling on the efficacy of dielectrophoresis-based single-particle traps. Dielectrophoretic forces, caused by the interaction of nonuniform electric fields with objects, have been used to make planar quadrupole traps that can trap single beads. A simple experimental protocol was then used to measure how well the traps could hold beads against destabilizing fluid flows. These were compared with predictions from modeling and found to be in close agreement, allowing the determination of sub-piconewton forces. This not only validates our ability to model dielectrophoretic forces in these traps but also gives insight into the physical behavior of particles in dielectrophoresis-based traps. Anomalous frequency effects, not explainable by dielectrophoretic forces alone, were also encountered and attributed to electrohydrodynamic flows. Such knowledge can now be used to design traps for cell-based applications.  相似文献

Abstract: Standardized, effective sampling methods are required to monitor amphibian population trends and community composition. Funnel traps have been used to ostensibly estimate species richness and relative abundance of larval amphibians. We tested whether funnel traps can be used to provide unbiased estimates of amphibian community composition in playa wetlands by comparing seining—dip netting and passive funnel-trapping results. Plains spadefoots (Spea bombifrons) were more prone to be captured in funnel traps whereas New Mexico spadefoots (S. multiplicata) were less likely captured by funnel traps than by seines and dip nets. In playas funnel traps should be used only for collecting specimens and not for estimating amphibian community composition.  相似文献

The effect of three food preservatives, sorbic acid and methyl and butyl esters of p-hydroxybenzoic acid, on the protonmotive force in Escherichia coli membrane vesicles was investigated. Radioactive chemical probes were used to determine the two components of the protonmotive force: delta pH (pH difference) and delta psi (membrane potential). Both types of compound selectively eliminated delta pH across the membrane, while leaving delta psi much less disturbed indicating that transport inhibition by neutralization of the protonmotive force cannot be the only mechanism of action for the food preservatives tested.  相似文献

We compared naturally baited trapping systems to synthetically baited funnel traps and fallen trap trees for suppressing preoutbreak spruce beetle, Dendroctonus rufipennis Kirby, populations. Lures for the traps were fresh spruce (Picea spp.) bolts or bark sections, augmented by adding female spruce beetles to create secondary attraction. In 2003, we compared a naturally baited system ("bolt trap") with fallen trap trees and with synthetically baited funnel traps. Trap performance was evaluated by comparing total beetle captures and spillover of attacks into nearby host trees. Overall, the trap systems did not significantly differ in spruce beetle captures, although bolt traps caught 6 to 7 times more beetles than funnel traps during the first 4 wk of testing. Funnel traps with synthetic lures had significantly more spillover than either trap trees or bolt traps. The study was repeated in 2004 with modifications including an enhanced blend synthetic lure. Again, trap captures were generally similar among naturally and synthetically baited traps, but naturally baited traps had significantly less spillover. Although relatively labor-intensive, the bolt trap could be used to suppress preoutbreak beetle populations, especially when spillover is undesirable. Our work provides additional avenues for management of spruce beetles and suggests that currently used synthetic lures can be improved.  相似文献

Three new types of traps designed and made by farmers were evaluated for capture of Idiarthron subquadratum Saussure & Pictet. Bag, sack, and fabric traps were compared with previously used bamboo internode traps. A participatory methodology was used involving farmer interviews and workshops to design and make the traps. Farmer participation was useful for obtaining information on perceptions, knowledge, and control activities of I. subquadratum. The bag trap captured a greater number of individuals than the bamboo and fabric traps, but its captures were similar to those of sack trap; captures were similar in bamboo, sack, and fabric traps. When captured individuals were analyzed by stage of development, no significant differences between types of traps were detected for captured adults. The number of individuals captured in the traps showed a similar trend to that counted in nocturnal sampling. A significant positive relationship was detected between numbers of adults captured and the damage of leaves and fruits. Taylor's Power Law indicates that I. subquadratum individuals were more abundant in some traps than in others. With densities greater than five individuals per trap, a smaller number of bag traps was required to estimate the same population compared with the other types of traps. Sampling procedure that used the bag traps had the lowest cost. This study shows that resource-poor coffee, Coffea arabica L., farmers had accumulated knowledge of I. subquadratum based on unpublished data and experience and that they were capable of applying this knowledge to develop an economical and more suitable technology for their conditions.  相似文献

Trap designs for banana root borer, Cosmopolites sordidus (Germar) (Coleoptera: Curculionidae), have been done essentially on the understanding that C. sordidus rely primarily on chemical cues. Our present results indicate that these borers also rely on visual cues. Previous studies have demonstrated that among the eight differently colored traps tested in the field, brown traps were the most effective compared with the performances of yellow, red, gray, blue, black, white, and green traps; mahogany-brown was more effective than other shades of brown.In the current study, efficiency of ground traps with different colors was evaluated in the laboratory for the capture of C. sordidius. Response of C. sordidus to pheromone-baited ground traps of several different colors (used either individually or as 1:1 mixtures of two different colors) were compared with the standardized mahogany-brown traps. Traps with mahogany-brown mixed with different colors had no significant effect. In contrast, a laboratory color-choice tests indicated C. sordidus preferred black traps over other color traps, with no specific preferences for different shades of black. Here again, traps with black mixed with other colors (1:1) had no influence on the catches. Therefore, any other color that mixes with mahogany-brown or black does not cause color-specific dilution of attractiveness. By exploiting these results, it may be possible to produce efficacious trapping systems that could be used in a behavioral approach to banana root borer control.  相似文献

Lethal spring traps are widely used for killing small mammals in the UK. Many require government approval, based primarily on humaneness. However, mole traps and break-back traps for rats and mice are exempt; those available vary widely in price and apparent quality. The EU is considering implementing a Trapping Directive that would alter UK legislation, and a recent report advised the EU that trapping legislation should cover all trapped species and encourage improvement of traps. Mechanical trap performance is often used as an indicator of welfare impact. We examined the mechanical evidence for scope to improve the welfare standards of rat, mouse and mole spring traps. We measured mechanical performance among a range of rat, mouse and mole traps. Impact momentum values varied 6-8 fold, and clamping force values 4-5.5 fold, among traps for killing each species. There was considerable overlap in the performance of rat and mouse traps. Trap-opening angle and spring type were related to impact momentum and clamping force in traps for both species. There was no relationship between price and mechanical performance in traps for any species, except talpa mole traps. We are unable to judge the direct welfare impact of the traps tested, but rather the potential welfare threat associated with their exemption from approval. The wide variation in mechanical performance in traps for each species, overlap in performance between rat and mouse traps and increasing availability of weaker plastic rodent traps indicate considerable scope for improving the humaneness of spring traps for rats, mice and moles. We conclude that all such traps should be subject to the UK approval process. New welfare categories might improve trap standards further. Our results could also help improve rodent trap design and assist consumers in selecting more powerful traps. Many thousands of rats, mice and moles might benefit.  相似文献

Bee species diversity and the effectiveness of four sampling methods were investigated in a west-central Illinois restored tallgrass prairie. Bees were sampled using malaise traps, ground-level pan traps, elevated pan traps, and vane traps. A total of 4,622 bees representing 31 genera and 111 species were collected. Malaise traps collected the greatest number of bees and species, and ground-level pan traps the least. Among the pan traps and vane traps, blue-colored traps collected the greatest abundance and species richness, and yellow traps the least. Chao1 estimator and rarefaction analyses showed that substantial increases in sample sizes would be necessary to achieve asymptotic species richness levels, particularly if ground-level pan traps alone were used. Elevated pan traps and vane traps collected relatively similar species composition. Different colored pan traps at the same height collected more similar species composition than did those at different heights, but species composition of blue ground-level pan traps was relatively similar to elevated pan traps, regardless of color. Indicator species analysis revealed 22 species that were significantly associated with a specific trap type, and 11 species that were associated with a particular pan trap color/elevation. Results of this study show that elevated traps can increase the effectiveness of bee surveys in tallgrass prairie, and that a combination of trap types gives a more complete picture of the bee fauna than does a single survey method. These results should be considered along with cost, ease of use, and goals when planning and designing bee inventories.  相似文献

Overwintered adult apple blossom weevils, Anthonomus pomorum (L.) (Coleoptera: Curculionidae), colonise apple trees, Malus domestica Borkh. (Rosaceae), in early spring. Information gained from a suitable monitoring technique could serve as a guide to determining the accurate timing for control measures. To assess tree colonisation by flight and crawling, Plexiglas flight traps and commercial screen traps developed for different curculionid species were used. Refuges were mimicked by a newly designed shelter trap based on transparent bubble wrap. The trap catches were contrasted to the limb jarring technique. Microclimate and weather conditions were determined over a trial period of one and a half months. The shelter traps caught a high number of weevils irrespective of weather conditions, and thus performed better than both flight and screen traps. The temperature level within the shelter traps was slightly, but significantly, higher than on the trunk, indicating that weevils exploit microclimatic differences and occupy thermally favourable refuges. The seasonal culmination of colonisation as determined by shelter traps coincided with that as determined by the two methods used to monitor weevil movement towards host trees (flight traps and screen traps). The data indicated that not only flight, but also crawling as quantified by the screen traps contributes substantially to spring colonisation. Captures by all three types of traps over the spring season were significantly positively correlated with each other, but in most respects not with the results of the limb jarring. We conclude that the newly designed shelter trap will be useful for accurately monitoring the seasonal course of colonisation and holds promise for determining the timing of interventions.  相似文献

Fusarium circinatum, the causal agent of pitch canker disease on pines, can be disseminated by wood produced in infested areas. The purpose of the study was to evaluate the effect of wood preservatives, commonly used against sapstain and wood‐decay fungi, on growth and sporulation of Fusarium circinatum. Seven active ingredients of antisapstain and anti‐wood‐decay preservatives were evaluated by their inhibition of mycelial growth. Propiconazole, tebuconazole, and 3‐iodo‐2‐propinyl butyl carbamate (IPBC) were effective against F. circinatum, whereas hydroxycarbonate of cooper was not. An assay was also conducted to evaluate the efficacy of three commercial antisapstain and two anti‐wood‐decay preservatives on Pinus radiata sapwood blocks that were previously inoculated with Fusarium circinatum. The product with the best efficacy was an antidecay preservative composed of tebuconazole, propiconazole, and dichlofluanid. None of the antisapstain preservatives tested was effective even though they contained fungicidal ingredients. Effects of dosage, product application, and formulation on the efficacy of these preservatives are discussed.  相似文献

Ecological traps, poor-quality habitat that nonetheless attract individuals, have been observed in both natural and human-altered settings. Until recently, ecological traps were considered a kind of source–sink system, but source–sink theory does not model maladaptive habitat choice, and therefore cannot accurately represent ecological traps or predict their population-level consequences. Although recent models of ecological traps addressed this problem, they used patch-based models containing only two habitats that were very different from one another, but were internally homogeneous. These sorts of patch models may not apply to many real populations, and using them for populations in landscapes with mosaic or gradient habitat structures may be misleading. I developed models that treat source–sink dynamics and ecological traps as special cases of a single process, in which the attractiveness and quality of the habitat are separate variables that can be either positively or negatively related, and in which habitat quality varies continuously throughout the landscape. As expected, sinks are less detrimental to populations than ecological traps, in which preferential use of poor habitat elevates extinction risk. Furthermore, ecological traps may be undetected, and may even appear to be sources, when population sizes are large, but may still prevent recovery in spite of the availability of high-quality habitat when populations drop below threshold levels. Conservation biologists do not routinely consider the possibility that apparent sinks are actually traps, but since traps should be associated with the rapidly changing and novel habitat characteristics primarily produced by human activities, ecological traps should be considered an important and potentially widespread conservation concern.  相似文献

Six different types of traps were compared for capturing adults of Delia radicum. Cone traps caught so few flies that it seems inadvisable for them to be used for monitoring populations of this pest. All the other traps tested were suitable for monitoring D. radicum populations. When expressed on a trap for trap basis, the large 1800 cm2 Canadian traps caught most flies. When expressed as the numbers of flies caughthnit area of trapping surface, most flies were caught in the water traps. Similar numbers of D. radicum, D. platura, syrphids, blowflies and a tachinid, Eriothrix rufomaculatus were caught per unit area on each of three different vertical sticky traps. Although water traps have the advantage that they catch about 5 times as many females D. radicum/unit area as the most effective vertical sticky traps of a comparable size, their disadvantage is that they catch about 10 times as many beneficial syrphids. The reasons why water traps are so effective against D. radicum are discussed, with details of how to convert water-trap data to sticky-trap equivalents, and vice versa.  相似文献

Weak-acid preservatives are widely used to prevent microbial spoilage of acidic foods and beverages. Characteristically, weak-acid preservatives do not kill micro-organisms but inhibit growth, causing very extended lag phases. Preservatives are more effective at low pH values where solutions contain increased concentrations of undissociated acids. Inhibition by weak-acids involves rapid diffusion of undissociated molecules through the plasma membrane; dissociation of these molecules within cells liberates protons, thus acidifying the cytoplasm and preventing growth. By modelling preservative action in yeast, using a thermodynamic and kinetic approach, it was possible to demonstrate that: (i) inhibition depends more on the degree to which individual preservatives are concentrated within cells, rather than on undissociated acid concentration per se; (ii) it is entirely feasible for microbes to pump protons out of the cell during extended lag phase and raise internal pH (pHi), despite further influx of preservatives; (iii) the duration of the lag phase can be predicted from the model, using a Gaussian fit of proton-pumping H(+)-ATPase activity against pHi; (iv) theoretical ATP consumption for proton pumping can be directly correlated with the reduction in cell yield observed in glucose-limited cultures.  相似文献
