Speckled snow mold, caused by the basidiomycete Typhula ishikariensis Imai, is one of the most prominent winter diseases on perennial grasses and cereal crops in the northern hemisphere. The first linkage map of T. ishikariensis was constructed using a population of 93 sibling monokaryons derived from a single dikaryotic hybrid isolate that was created by a hyphal fusion of two monokaryotic parental isolates. The parental isolates were produced from a pathogenic dikaryotic isolate collected from a golf course in Wisconsin. The two parents exhibit significant differences in the production of aerial mycelium and sclerotia, and in their aggressiveness on creeping bentgrass (Agrostis stolonifera L.). A total of 251 loci were mapped, comprising 89 inter-simple sequence repeat (ISSR) and 160 random amplified polymorphic DNA (RAPD) markers along with 2 phenotype-based mating-type (MAT) loci. The MAT loci were mapped on linkage groups (LGs) 1 and 7. The markers were evenly distributed over 7 LGs, covering 436 cM with an average marker interval of 2.2 cM. Seven chromosomes were cytologically observed using germ tube bursting methods with acetocarmine staining. This reference linkage map of T. ishikariensis should provide a framework for the mapping of quantitatively controlled traits such as fungal growth, survival, and virulence/avirulence under low temperatures. The map should also be utilized for studying the genome organization of the cold-loving plant-pathogenic Typhula spp. and for comparative genome analysis among fungal taxa.  相似文献

The exotic pine shoot beetle, Tomicus piniperda (L.) (Coleoptera: Scolytidae), established in the north central and northeastern United States (U.S.) and adjacent regions in Canada, is regulated by a federal quarantine that restricts movement of pine material during specific times of the year based on the beetle's life history. Although climatic variation occurs across T. piniperda's range, a single set of dates is used for timing the movement of pine logs. We monitored T. piniperda spring flight, fall shoot departure, and air and internal tree temperatures at three sites along a 300-km north-south gradient in Michigan and Indiana. We also estimated dates for initial spring flight (12 degrees C threshold) and fall shoot departure (0 degrees C threshold) across an 850-km gradient using historical temperature records (1901 to 1999). Average daily temperatures in fall 1997 (8 October to 12 December) and spring 1998 (20 February to 21 April) were 1.8 to 2.4 degrees C colder, respectively, at the northern field site than at the southern field site. Fall shoot departure began at approximately the same time (day 289 to 290) at all three field sites, but complete shoot departure was extended by 3 wk at the southern site (day 336) compared with the northern site (day 317). T. piniperda adults were first captured in funnel traps on calendar day 86 at the northern site and on day 59 at the central and southern field sites. Peak flight occurred at approximately the same time (day 86) at all three sites. Within-shoot temperatures were very similar to air temperatures in the fall and aboveground inside-bark temperatures were similar to air temperatures in the spring. Average predicted dates based on historical temperature records varied by 31 d for initial shoot departure and 84 d for initial spring flight between northern Michigan and southern Indiana. Because considerable variation can occur in T. piniperda behavior across a broad geographic range, dates specified in the U.S. Federal quarantine should be adjusted according to local temperatures.  相似文献

Summary Two isolates of Pleurotus ostreatus from North America and one from Germany are interbreedable. Under identical conditions at low temperatures, their fruiting bodies are hard to distinguish. However, shape and colour and several other characters vary with culture conditions. The American stocks fruit well at temperatures from 4 to 24 °C, the German ones only below 15 °C. Four types of hybrids between German and American Pleupotus were obtained: i) The whole fruiting process is temperature sensitive as in German Pleurotus. ii) It proceeds at 4–24 °C as in American stocks, iii) Fruiting initiation is insensitive but sporophore development is sensitive to elevated temperatures, iv) Primordia formation and initial sporophore development depend on temperatures below 15 °C, but pileus expansion and spore discharge continue above 20 °. The involvement of separate genes for the single developmental steps and the use of temperature sensitivity for commercial varieties are discussed. One sporeless strain, F42x11, with considerable fruiting bodies has been obtained. In intrastock di-mon-matings this character was dominant.  相似文献

Activity patterns of Phyllophaga crinita (Burmeister), Phyllophaga congrua (LeConte), Phyllophaga crassissima (Blanchard), and Cyclocephala lurida (Bland) grubs were monitored with acoustic sensors in small pots of bluegrass, Poa arachnifera Torr, at varying and constant temperatures over multiple-day periods. Experienced listeners readily distinguished three types of sound with distinct differences in frequency and temporal patterns, intensities, and durations. Of approximately 3,000 sounds detected from P. crinita larvae, 7% were identifiable as snaps, with large amplitudes and short durations typically associated with root breakage or clipping activity. Approximately 60% were identifiable as rustles, suggestive of surfaces sliding or rubbing past each other during general movement activity. Another 2% of sounds contained patterns of repeated pulses suggestive of surfaces scraping across a pointed ridge. The remaining 31% had spectral or temporal patterns that fell outside the ranges of easily recognizable sound types. Because the behavioral significance of the different sound types has not yet been fully established, the classified and unclassified sounds were pooled together in analyses of the effects of species, temperature, weight, and time of day. Grubs of all four species produced detectable sounds at rates that increased with temperature [0.45 sounds/((min)(degrees C))] and larval weight [6.3 sounds/((min)(g))]. Mean sound rates were independent of species and time of day. At temperatures <9 degrees C, mean sound rates fell below the typical levels of background noise observed under field conditions. This reduced activity at low temperatures is likely to reduce the effectiveness of acoustic monitoring in the field in cold weather. The consistency of results obtained in these tests over multiple-day periods suggests that acoustic systems have potential as tools for nondestructive monitoring of the efficacy of insect management treatments as well as for biological and ecological studies.  相似文献

To investigate the effect of culture temperature on erythropoietin (EPO) production and glycosylation in recombinant Chinese hamster ovary (CHO) cells, we cultivated CHO cells using a perfusion bioreactor. Cells were cultivated at 37 degrees C until viable cell concentration reached 1 x 10(7) cells/mL, and then culture temperature was shifted to 25 degrees C, 28 degrees C, 30 degrees C, 32 degrees C, 37 degrees C (control), respectively. Lowering culture temperature suppressed cell growth but was beneficial to maintain high cell viability for a longer period. In a control culture at 37 degrees C, cell viability gradually decreased and fell below 80% on day 18 while it remained over 90% throughout the culture at low culture temperature. The cumulative EPO production and specific EPO productivity, q(EPO), increased at low culture temperature and were the highest at 32 degrees C and 30 degrees C, respectively. Interestingly, the cumulative EPO production at culture temperature below 32 degrees C was not as high as the cumulative EPO production at 32 degrees C although the q(EPO) at culture temperature below 32 degrees C was comparable or even higher than the q(EPO) at 32 degrees C. This implies that the beneficial effect of lowering culture temperature below 32 degrees C on q(EPO) is outweighed by its detrimental effect on the integral of viable cells. The glycosylation of EPO was evaluated by isoelectric focusing, normal phase HPLC and anion exchange chromatography analyses. The quality of EPO at 32 degrees C in regard to acidic isoforms, antennary structures and sialylated N-linked glycans was comparable to that at 37 degrees C. However, at culture temperatures below 32 degrees C, the proportions of acidic isoforms, tetra-antennary structures and tetra-sialylated N-linked glycans were further reduced, suggesting that lowering culture temperature below 32 degrees C negatively affect the quality of EPO. Thus, taken together, cell culture at 32 degrees C turned out to be the most satisfactory since it showed the highest cumulative EPO production, and moreover, EPO quality at 32 degrees C was not deteriorated as obtained at 37 degrees C.  相似文献

Despite their close association with human activities, plant pathogenic fungi have rarely been found in archaeological excavations. We report here that a fungus was closely associated with human activities even in prehistoric times. Sclerotium-like objects were found at historical sites (4000 to 400 BP) on the island of Hokkaido, northern Japan. They were spherical, 0.3-1.0 mm in diameter, and had a medulla and rind. Some had leaf fragments on the surface or a protuberance that resembled emerging sporocarp primordia. These traits indicated that they were sclerotia of the snow mold fungus, Typhula ishikariensis biotype B.  相似文献

Stop-flow techniques were used to determine how temperature affected the axonal transport of dopamine-beta-hydroxylase (DBH) activity in rabbit sciatic nerves in vitro. These nerves were cooled locally to 2 degrees C for 1.5 hr, which caused a sharp peak of DBH activity to accumulate above the cooled region. Accumulated DBH was then allowed to resume migration at various temperatures. From direct measurements of the rate of migration, we found that the axonal transport velocity of DBH was a simple exponential function of temperature between 13 degrees C and 42 degrees C. Over this range of temperatures, the results were well described by the equation: V=0.546(1.09)T, where V is velocity in mm/hr, and T is temperature in degrees centigrade. The Q10 between 13 degrees and 42 degrees C was 2.33, and an Arrhenius plot of the natural logarithm of velocity versus the reciprocal of absolute temperature yielded an apparent activation energy of 14.8 kcal. Transport virtually halted when temperature was raised to 47 degrees C, although only about half of the DBH activity disappeared during incubation at this temperature. Another transition occurred at 13 degrees C; below this temperature, velocity fell precipitously. This was not an artifact peculiar to the stop-flow system since the rate of accumulation of DBH activity proximal to a cold-block also decreased abruptly when the temperature above the block was reduced below 13 degrees C.  相似文献

Five complimentary studies were undertaken with the overall aim to examine the ability of free-living stages of Haemonchus contortus to over-winter and tolerate cold stress. Two studies deal with the development and long-term survival of eggs and infective larvae of two geographically different isolates (Kenya and Sweden). Eggs and larvae were monitored in climatic chambers at temperatures that fluctuated daily between -1 degrees C and 15 degrees C, or at constant temperatures of 5 degrees C and 15 degrees C. The development from egg to larvae was dependent on temperatures over 5 degrees C. The long time survival was favoured at lower temperatures. Furthermore, the overwintering capacity of the free-living stages of these isolates was estimated under Swedish field conditions. Two groups of lambs were experimentally infected with different isolates, and kept separated on previously ungrazed plots. In early May the following year, two parasite-naive tracer lambs were turned out on each of the plots to estimate the pick up of overwintered larvae. This experiment was replicated in central and southern Sweden. In addition, two experiments were performed in 2003 on pasture previously grazed by naturally infected sheep. One trial was on a pasture in southern Sweden grazed by a commercial flock, where extreme numbers of H. contortus were found towards the end of the grazing season 2002. The other study was on a pasture plot in central Sweden grazed by a hobby flock in 2002, where three of six lambs died due to haemonchiasis. Overwintered H. contortus was recorded on three of four experimental sites. Worm burdens were in all instances extremely low. No differences in development and survival were found between the isolates. Consequently, overwintering on pasture is of no practical significance in the transmission of H. contortus between grazing-seasons in Sweden.  相似文献

Campylobacter jejuni is a leading cause of bacterial gastroenteritis in humans, and contamination of poultry has been implicated in illness. The bacteria are fastidious in terms of their temperature requirements, being unable to grow below ca. 31 degrees C, but have been found to be physiologically active at lower temperatures and to tolerate exposure to low temperatures in a strain-dependent manner. In this study, 19 field isolates of C. jejuni (10 of clinical and 9 of poultry origin) were studied for their ability to tolerate prolonged exposure to low temperature (4 degrees C). Although substantial variability was found among different strains, clinical isolates tended to be significantly more likely to remain viable following cold exposure than poultry-derived strains. In contrast, the relative degree of tolerance of the bacteria to freezing at -20 degrees C and freeze-thawing was strain specific but independent of strain source (poultry versus clinical) and degree of cold (4 degrees C) tolerance.  相似文献

Responses of broiler chickens to a high ambient temperature (35 degrees C) were measured in two experiments. In one experiment temperatures were increased abruptly from 21 degrees C to a daily range of 21-35 degrees C whereas, in the other, temperatures were increased more gradually over 6 days. The high temperatures were maintained for 5 h/day. In both experiments, birds exposed to the high temperatures ate less food and gained less liveweight than birds maintained at 21 degrees C. Efficiency of food conversion to liveweight gain and body composition were not affected by high temperature but there was a tendency for thyroid weight to decrease. Overall, the plasma concentration of triiodothyronine (T3) decreased and the plasma concentration of thyroxine (T4) increased, resulting in a decreased T3/T4 molar ratio, during exposure to high temperature. The concentration of plasma growth hormone, but not plasma reverse T3, was increased by high temperature. The initial responses to increased temperature were variable, with birds exposed more gradually adjusting relatively well until the maximum temperature was increased to 35 degrees C. All heated birds readjusted quickly to the daily reduction in temperature to 21 degrees C.  相似文献

Formation of reactive oxygen species (ROS) in mitochondrial isolates from gill tissues of the Antarctic polar bivalve Laternula elliptica was measured fluorimetrically under in vitro conditions. When compared to the rates measured at habitat temperature (1 degrees C), significantly elevated ROS formation was found under temperature stress of 7 degrees C and higher. ROS formation correlated significantly with oxygen consumption in individual mitochondrial preparations over the entire range of experimental temperatures (1-12 degrees C). ROS generation per mg of mitochondrial protein was significantly higher in state 3 at maximal respiration and coupling to energy conservation, than in state 4+, where ATPase-activity is inhibited by oligomycin and only proton leakage is driving the residual oxygen consumption. The percent conversion of oxygen to the membrane permeant hydrogen peroxide amounted to 3.7% (state 3) and 6.5% (state 4+) at habitat temperature (1 degrees C), and to 7% (state 3) and 7.6% (state 4+) under experimental warming to 7 degrees C. This is high compared to 1-3% oxygen to ROS conversion in mammalian mitochondrial isolates and speaks for a comparatively low control of toxic oxygen formation in mitochondria of the polar bivalve. However, low metabolic rates at cold Antarctic temperatures keep absolute rates of mitochondrial ROS production low and control oxidative stress at habitat temperatures. Mitochondrial coupling started to fall beyond 3 degrees C, closely to pejus temperature (4 degrees C) of the bivalve. Accordingly, the proportion of state 4 respiration increased from below 30% at 1 degrees C to over 50% of total oxygen consumption at 7 degrees C, entailing reduced ADP/O ratios under experimental warming. Progressive mitochondrial uncoupling and formation of hazardous ROS contribute to bias mitochondrial functioning under temperature stress in vitro. Deduced from a pejus temperature, heat stress commences already at 5 degrees C, and is linked to progressive loss of phosphorylation efficiency, increased mitochondrial oxygen demand and elevated oxidative stress above pejus temperatures.  相似文献

We report the isolation and properties of several species of bacteria from Siberian permafrost. Half of the isolates were spore-forming bacteria unable to grow or metabolize at subzero temperatures. Other Gram-positive isolates metabolized, but never exhibited any growth at - 10 degrees C. One Gram-negative isolate metabolized and grew at - 10 degrees C, with a measured doubling time of 39 days. Metabolic studies of several isolates suggested that as temperature decreased below + 4 degrees C, the partitioning of energy changes with much more energy being used for cell maintenance as the temperature decreases. In addition, cells grown at - 10 degrees C exhibited major morphological changes at the ultrastructural level.  相似文献

AIMS: The effects of water activity (0.90-0.99 a(w)), temperature (15-37 degrees C), and their interaction on growth and ochratoxin A (OTA) production by eight isolates of Aspergillus carbonarius were investigated on synthetic nutrient medium (SNM) with composition similar to grapes. METHODS AND RESULTS: Growth data were modelled by an multiple linear regression and response surface models were obtained. Aspergillus carbonarius grew much faster at 30 degrees C than at the other temperature levels tested and its growth rate increased with increasing a(w), maximum growth rate being between 0.95 and 0.99 a(w). In general, isolates grew faster at 35-37 degrees C than at 20 degrees C, although no significant differences were found between these temperatures. OTA accumulation was also favoured by high a(w) levels, and although it was observed in the whole range of temperatures, maximum amounts were detected at 20 degrees C. No OTA was found at the most unfavourable growth conditions. CONCLUSIONS: Optimum a(w) level for growth seems to correspond with optimum for OTA production, meanwhile the most propitious temperature for the toxin production was below the best one for growth. SIGNIFICANCE AND IMPACT OF THE STUDY: Prediction of A. carbonarius growth would allow estimating their presence and therefore, the OTA production, as it was found that conditions for the toxin production were more limited than those permitting growth.  相似文献

Body temperatures during hot flashes were measured in a menopausal woman. Internal temperatures fell after each flash; lowest: rectal, 35.6 degrees C; vaginal, 35.6 degrees C; tympanic, 35.2 degrees C. Where sweating occurred, the skin temperature fell during the flash and rose after it. Finger and toe temperatures always showed a sharp rise at the onset of a flash with a slower fall after the flash. Only the cheeks showed additional temperature rises; maximum, 0.7 degrees C. The heart accelerated 13% at the onset of the flash but slowed immediately thereafter. The flash interval was sharply demarcated by undulations in the ECG baseline. There was never any premonitory sign of the imminence of a flash. A central excitatory state seemed to build up, perhaps by the accumulation of a chemical compound, but not of heat, which was explosively dischargedmthe thermal distress was probably evoked by vascular warming in the cheeks. Dabbing the malar prominences with cold water brought prompt relief.  相似文献

The effects of sodium bicarbonate concentration, population density, and temperature on the maintenance of an outdoor monoculture of the cyanobacterium Spirulina platensis were studied. A clear response by Spirulina to the concentration of bicarbonate was evident, with 0.2M bicarbonate representing the lowest concentration in which a monoculture could be maintained. When the temperatures fell during the winter period to some 20-25 degrees C below the optimum for Spirulina, Chlorella sp. gradually increased and became the dominant species in the culture. Raising the temperature by covering the pond with transparent polyethylene resulted in a sharp decline in the population of Chlorella, and a gradual resumption of species dominance by Spirulina. In winter, there was an inverse relationship in the pond between the population density of Spirulina and the extent of contamination by Chlorella sp.; but no such effect was observed under field conditions at temperatures higher than 25 degrees C.  相似文献

The growth of the snow mould fungi Fusarium nivale (Fr.) Ces. and Typhula ishikariensis Imai at different water potentials was measured on potato dextrose broth supplemented with KCl or polyethylene glycol 6000. The dew point hygrometer method was used for determination of water potentials in both artificial growth media and plant material. Both fungi had a considerable decrease in dry weight production when water potentials of growth media were decreased from −0.7 to −3 MPa. Crown tissue from hardened plants of Phleum pratense L. (timothy, cvs Engmo, Grindstad) and Phalaris arundinacea L. (reed canary grass, cvs Hansvoll, American common, Vantage) showed a significant decrease in water potentials compared with unhardened plants. Thus, reduced availability of water may partly explain increased resistance to fungal pathogens in grasses after hardening.  相似文献

The infectivity of 4 isolates of Metarhizium anisopliae to puparia of Ceratitis capitata treated as late third-instar larvae in unsterilized soil was investigated in the laboratory under controlled temperature and moisture. At 20-30 degrees C, mortality in puparia was highest at water potential of -0.1 and -0.01 mega Pascal (MPa) and lowest at water potential of -0.0055 and -0.0035 MPa in all the isolates. In wetter soil however, isolates ICIPE 20 and 60 caused significantly higher mortality than ICIPE 18 and 69. The survival of conidia in drier soil (-0.1 MPa) was not adversely affected at all temperatures. However, in wet soil (-0.0035 MPa) there was drastic reduction in colony counts in ICIPE 18 and 69 at 25 and 30 degrees C but conidial density in ICIPE 20 and 60 remained at the initial level at 14 days after inoculation at all temperatures. When ICIPE 20 was evaluated against three other fruit fly species (Ceratitis cosyra, Ceratitis rosa, and Ceratitis fasciventris), significant reduction in adult emergence and higher pupal mortality occurred in C. cosyra and C. fasciventris than in C. rosa at a combination of 15 and 20 degrees C and -0.1 and -0.0035 MPa. However, at higher temperature and the same moisture level, the isolates were equally pathogenic across the 3 species. It is probable that in addition to pathogen cycling and multiplication from dead infected insects in the soil, a balance between microbial degradation and replenishment of inoculum of virulent isolates occur through fluctuations in, and intricate interactions between temperature and moisture levels. This study is indicative of the potential of using isolate ICIPE 20 for soil inoculation against pupariating third-instar larva of fruit flies, thus providing a novel alternative to chemical soil application.  相似文献

Three isolates of Fusarium graminearum (DAOM 180377, 180378, and 180379) were screened for their ability to produce mycotoxins on the solid substrates corn and rice. They all produced deoxynivalenol and zearalenone on corn. On rice, only DAOM 180378 and 180379 produced significant amounts of these mycotoxins, with levels of deoxynivalenol being much higher than those of zearalenone. The effects of the initial moisture content before autoclaving, incubation temperature, and time were studied with isolate DAOM 180378. At 19.5 degrees C the main product was zearalenone, whereas at 25 degrees C both deoxynivalenol and zearalenone were formed. Higher incubation temperatures (28 degrees C) favored deoxynivalenol formation, the maximum amount being 515 ppm (515 micrograms/g) formed after 24 days at an initial moisture content of 40%. The maximum level of zearalenone produced at the same temperature was 399 ppm, but at an initial moisture content of 35%. Other factors, such as pH, oxygen and carbon dioxide concentrations, and size of the culture flask also appeared to affect the production of mycotoxins.  相似文献

以华北平原冬小麦农田为研究对象,采用涡度相关技术和热红外遥感技术,研究了不同环境条件下土壤含水量与农田蒸散量及作物冠层温度的关系.结果表明,冬小麦在农田郁闭(LAI≥3)、晴天和土壤相对含水量低于田间持水量65%的情况下,蒸发比值日变化正午前后出现相对较低且平稳的变化趋势.在晴天情况下,农田潜热通量与作物冠层温度日变化和季节变化均呈极显著的非线性相关关系,而冠气温差、农田相对蒸散量则与0~100 cm土层的土壤相对含水量密切相关.以13:30~14:00的平均冠层温度值Tc、日最高气温Ta max和日净辐射总量Rnd为统计数据,确立了冬小麦农田日蒸散量ETd (mm)估算简化模式参数.  相似文献

Allocation of organic carbon (OC) to primary energetic pathways was estimated under seasonal and artificially elevated ambient temperatures for a field population of a freshwater pulmonate snail, Physella virgata. Allocation to respiration increased with temperature. Snails allocated most assimilated OC to reproduction within their natural temperature range (15 degrees -35 degrees C), where assimilation efficiencies remained relatively stable at 25%-35%. However, in artificially heated waters exceeding 35 degrees C, declining assimilation rates and increasing respiratory demands inhibited allocation to reproduction and growth. At the species' 40 degrees C upper thermal limit, assimilation efficiencies fell below 10%, while average consumption levels more than doubled relative to snails unaffected by the thermal effluent. Ambient temperature substantially influenced OC allocation over P. virgata's natural temperature range and negatively affected growth and reproduction at temperatures approaching or exceeding maximum natural levels.  相似文献
