The repair-deficient mutant, mei-9a in Drosophila melanogaster was investigated regarding its effect on spontaneous and X-ray-induced chromosome loss in male postmeiotic cells. From matings of males carrying a mei-9a or an ordinary ring-X and a doubly marked Y chromosome (BSYy+) with mei-9a or ordinary females, the spontaneous frequencies of complete loss, partial loss, and inferred ring-X loss (based on shifts in sex ratio female:male) were significantly higher with mei-9a than with non-mei-9a. When males were given 3000 rad X-irradiation, frequencies of induced partial loss, inferred ring-X loss and the reduction in the number of progeny per female were significantly greater with mei-9a than with non-mei-9a. The results provide evidence that the mei-9a is a potentiator of both spontaneous and X-ray-induced chromosome lesions in sperm of the Drosophila male. Evidence is presented which implicates the presence of mei-9a in the P1 female and not the male as (at least) largely responsible for the characteristic mei-9a effects.  相似文献

The influence of the maternal genotype (Canton-S, proficient in the repair of X-ray-induced chromosome breaks and ebony, less proficient in this regard) on the recovery of X-ray-induced autosomal (II–III) translocations and ring-X chromosome losses in mature spermatozoa was studied. In the first series of experiments, males carrying appropriate markers on their second and third chromosomes were irradiated and mated to Canton-S or ebony females and the frequencies of II–III translocations were determined. In the second series of experiments, males carrying ring-X chromosomes were irradiated in N2 or in O2, mated to Canton-S or ebony females and the frequencies of XO males were determined; additionally, under similar gas-treatment and radiation conditions, the pattern of egg-mortality was also assessed.

The data on translocations show that the yields are higher with ebony than with Canton-S females; these and earlier results on dominant lethals and sex-linked recessive lethals support the interpretation that the maternal repair system in the ebony strain is less proficient and more error-prone than that of the Canton-S strain.

Those on the losses of ring-X chromosomes demonstrate that (i) the absolute yields of XO males are lower with ebony than with Canton-S females irrespective of whether the parental males are irradiated in N2 or in O2; (ii) the exposure-frequency relationships are all linear, but the slopes are higher when the males are irradiated in O2 and are consistent with an oxygen-enhancement-ratio of about 1.5 and (iii) the relationships between the logarithm of egg-survival and XO male frequency are also linear, but the slopes for the O2 groups are lower than those for the N2 groups (slope ratios of 0.86–0.87).

The finding that at given survival levels, the XO frequencies are lower in the O2 than in the N2 groups of both the Canton-S and ebony series viewed in the context of the mechanisms that have been postulated to explain the loss of ring-X chromosomes in irradiated mature spermatozoa permits the following interpretation for the observed results: (i) a higher proportion of potential XO zygotes is lost through dominant lethality in the O2 groups than in the N2 ones presumably because the chromosome breaks induced in O2 are qualitatively different in the sense that they have higher probability to undergo reunions relative to restitution, compared with breaks induced under anoxia and (ii) this leads to lower than expected oxygen-enhancement ratios (i.e., expected on the basis of published data on sex-linked recessive lethals, another kind of genetic damage which shows a linear exposure-frequency relationship).  相似文献

1. 1. The purpose of the study was to investigate the preferred temperature of the elderly after cold and heat exposures.

2. 2. Eight elderly and 9 young females wearing the same type of clothing were exposed to cold (10°C), moderate (25°C) or hot (35°C) environments for 30 min in the exposure room.

3. 3. Then they moved to the self-control room in which the temperature was set at 25°C, and the room temperature increased or decreased continuously by 0.4°C every minute.

4. 4. The subjects were instructed to operate the switch when they felt uncomfortably warm or cool during a 90-min period.

5. 5. In operating the switch, the changing in room temperature shifted to the opposite direction.

6. 6. The ambient temperature was recorded continuously and analyzed as the preferred temperature, which was defined as the midpoint temperature of the crest and trough of temperature records.

7. 7. The preferred temperatures after the cold exposure were significantly higher than those of other exposure conditions in the elderly.

8. 8. On the other hand, in the young, there was no significant difference in the preferred temperature among the exposure conditions.

9. 9. Although the effect of exposure to cold or hot environments decreased in the latter parts of self-control, the elderly still preferred the higher temperature after cold exposure.

Postmeiotic cell stages of repair-proficient ring-X (RX) males were treated with methyl methanesulfonate (MMS), ethyl methanesulfonate (EMS), diethylnitrosamine (DEN) or ethylnitrosourea (ENU) and then mated to either repair-defective (mei-9L1) or to repair-competent females (mei-9+). Absence of the mei-9+ function resulted in a hypermutability effect to all alkylating agents (AAs) when they were assayed for their ability to induce chromosomal aberrations (chromosome loss; CL), irrespective of marked differences in distribution of DNA adducts brought about by these AAs. This picture is different from that described previously for the induction of point mutations (Vogel et al., 1985a). There, evidence was presented indicating that reduction in DNA excision repair does not affect point mutation induction (recessive lethals) by those AAs most efficient in ring-oxygen alkylation such as ENU, DEN, N-ethyl-N'-nitro-N-nitrosoguanidine (ENNG), and isopropyl methanesulfonate (iPMS): the order of hypermutability of AAs with mei-9L relative to mei-9+ was MMS greater than MNU greater than DMN = EMS greater than iPMS = ENU = DEN = ENNG. When the percentage of lethal mutations induced in mei-9L1 females were plotted against those determined for mei-9+ females, straight lines of following slopes were obtained: MMS = 7.6, MNU = 5.4, DMN = 2.4, EMS = 2.4, and iPMS = ENU = DEN = ENNG = 1. Those findings, together with the recent observation that AAs do not split into two groups when assayed for their ability to cause CL, point to the involvement of different DNA alkylation products in ENU- and DEN-induced chromosome loss vs. that of point mutations. It is concluded that with ENU and DEN chromosomal loss results from N-alkylation products whereas point mutations (SLRL) are the consequence of interactions with oxygen-sites in DNA. Thus, as a consequence of a very dominating role of O-ethylguanine (and possibly O4-alkylation of thymine), N-alkylation in DNA does not contribute measurably to mutation induction in the case of ENU-type mutagens while O-alkylation, very clearly, does not show a positive correlation with the formation of chromosome breakage events in Drosophila. Conversely, it appeared that with MMS-type mutagens (MMS; dimethyl sulfate, DMS; trimethyl phosphate, TMP), alkylation products such as 7-methylguanine and 3-methyladenine, if unrepaired or misrepaired, are potentially mutagenic lesions causing both mutations and chromosomal aberrations.  相似文献

1. 1.|Body temperatures (Tb) and contaneous evaporative water loss rates (CWL) were measured in tree frogs (Hyla cinerea) and toads (Bufo valliceps) exposed to cyclical ramp changes in water vapor density (WVD) between 7.5 and 9.8 gm−3 (1 cycle h−1 at an air temperature of 27.0°C.

3. 3.|Tb in toads cycled directly with WVd; WVD accounted for 98% of the variation in toad Tb.

2. 2. There are nearly 4000 cold storage facilities and about 80% are being kept at temperatures below −20°C.

1. 1.|The instantaneous addition of phospholipase C to goldfish brain inner mitochondrial vesicles resulted in a partial loss of ferricyanide reduction. The retained activity rose when the phospholipase concentration and incubation period were increased.

2. 2.|No impairment of ferricyanide reduction was observed when Antimycin A-treated inner membrane vesicles were incubated with the phospholipase at 0.05 and 0.2 U mg−1 to 20 min. However, higher concentrations of the hydrolytic enzyme caused a considerable elevation in reduction and is attributed to a deterioration in vesicular integrity.

3. 3.|The hydrolysis of phospholipids from the inner membrane revealed that the phospholipase, at concentrations above 0.2 U mg−1 (minimum of 10 min incubation), resulted in removal of over 50% lipid phosphate. It was suggested that the phospholipase, at such levels, is accessible to the interior of the vesicles.

4. 4.|With the exception of cardiolipin and phosphatidyl inositol, the phospholipase was observed to be non-specific to the phospholipids from mitochondrial inner membrane treated with Triton X-100.

5. 5.|The phospholipids are asymmetrically arranged across the inner membrane and their distribution is dependent on acclimation temperature.

6. 6.|It is suggested that the compositional redistribution of the phospholipids, when acclimation temperature was changed, is necessary to stabilize the bilayer conformation of the membrane in concurrence with maintaining fluidity and functional capacity.

1. 1.|Atropine administration resulted in higher skin temperatures in both sensible and insensible environments and a higher core temperature in the hot environment, due to the reduction in whole body sweating. Exercise time was reduced 28 min following atropine in the hot environment, but was not affected in the humid environment.

2. 2.|The effect of heat storage (significantly higher after atropine) was shown to be greater in the hot environment due to inadequate sweat secretion for subsequent evaporative cooling. In the warm environment, enhanced sensible heat loss resulted in more effective thermoregulation.

3. 3.|Based on the effective temperature (ET*) it is suggested that exercise in the heat can be accomplished during environmental stress at warm temperatures after atropine treatment.

To examine the influence of different bed conditions (ondol sleep, bed sleep on ondol with same bedding) of the Korean ondol traditional heating system on human response during sleep, bed climates and physiological responses such as skin and rectal temperatures, weight loss, body movement and subjective sensation were measured with 4 grown-up females as subjects while they were sleeping for 7 h.

2. 2. Bed climate: Temperatures under the mattress and inside the quilt were higher on ondol while temperatures on the mattress and humidity inside the quilt were higher on the bed.

3. 3. Rectal temperature was significantly higher on ondol; skin temperature showed no major differences in relation to bed conditions. The frequency of body movements had the highest correlation with bed climate of the parameters measured.

4. 4. Mattress weight decreased on ondol and increased on the bed.

5. 5. The frequency of body movements was significantly higher in ondol sleep.

6. 6. The subjects sensation showed difference on cushion sensation between the two types of bed condition.

7. 7. To obtain the same level of comfort on both ondol and bed sleeping conditions less thermal insulating value is needed for ondol sleep.

1. 1. Our previous experimental results showed the thermal sensation vote was much less than the values of PMV and SET* at air velocities above 0.5 m/s.

2. 2. The method to modify SET* is presented from the results of subjective experiments taking account of decrease in clo value of summer clothing and decrease in skin wettedness due to increased air velocity.

3. 3. Thermal resistance under increased air movement on a standard summer clothing ensemble was measured. Basic thermal insulation of the summer ensemble was reduced by 25% at air velocity of 1.0 m/s.

4. 4. Thirty-two subjects were exposed at operative temperatures of 27 and 30°C under 1 m/s air movement in order to determine the amount of skin diffusion. Measured evaporation heat loss from skin surface was much smaller at air velocity of 1 m/s than that predicted by SET*.

5. 5. Estimated thermal sensation vote using modified SET* agreed well with our previous experimental results under different air velocities for the same clothing.

1. 1.Hypothalamic temperature (Thy), nonevaporative heat loss (R + C + K), evaporative heat loss (E), thermal conductance (k), metabolic heat production (M) and heat storage (S) of rats were simultaneously measured by direct and indirect calorimetry during internal heat loading (2 W per rat) with an intraperitoneal electric heater.

2. 2.The tests were made twice a day; once during the day (1000–1200 h) and once at night (2200–2400 h) at an ambient temperature of 24°C.

3. 3.The resting values of Thy, colonic temperature, (R + C + K), E, M and heart rate, and the Thy threshold for tail skin vasodilation (Tth) during internal heat load were significantly higher at night than during the day.

4. 4.The slopes showing the relationshiop between (R + C + K), k or M and Thy were significantly steeper during the day than at night after Thy exceeded Tth.

5. 5.The slopes showing the relationship between E or S and Thy were not different during the day and at night.

6. 6.These results indicate that the responses of nonevaporative heat loss and heat production to internal heat load vary with the time of day in rats.

The specificity and nature of the reaction between salicylaldehyde and myosin and the effect of salicylalation on the molecular parameters of myosin were studied. The following observations were made.

1. 1. The reaction of salicylaldehyde with the lysyl residues of myosin is specific, since no salicylaldehyde is bound if the lysyl residues of myosin are trinitrophenylated.

2. 2. Salicylaldehyde is bound by myosin through the formation of an azomethine linkage (Schiff's base). This was established from the measured difference absorption spectrum of the myosin-salicylaldehyde complex.

3. 3. Three groups of lysyl residues can be distinguished with respect to the reaction with salicylaldehyde, namely, (a) residues with high association constant (Kass = 1.8 ± 0.9·105 M-1), (b) residues with moderate association constant (Kass = 2.2·103 M-1) and (c) residues that react with salicylaldehyde only after the denaturation of the protein. Their numbers could be estimated as 10 ± 5, 130 ± 5 and 260 ± 5 per mole myosin, respectively. The first group of residues was found to be absent from heavy and light meromyosin, the proteolytic fragments of myosin.

4. 4. The reaction is reversible. The complex formation rate constant, evaluated from the formula for second order reaction, is 2.2 sec-1·M-1, and the decomposition rate constant for first order reaction is 1.1·10-3 sec-1 at 22°.

5. 5. The reaction is pH dependent, the reaction yield increasing at higher pH.

6. 6. The solubility of myosin at low ionic strength decreases with increasing degree of salicylalation at slightly alkaline pH.

7. 7. The intrinsic viscosity of myosin does not change on salicylalation.

8. 8. A second peak due to polymerization appears on the sedimentation profile of the protein if more than 70 lysyl residues are salicylalated per mole of myosin.

1. The heat flow of paraplegic (PA) and able-bodied (AB) subjects were determined at rest in cool and warm conditions.

2. During heat exposure upper body sites for both groups showed heat loss, whereas the lower body sites of the PA groups showed heat gain.

3. During heat exposure, a systematic difference between groups in the relationship between heat flow and calf-skin temperature existed.

4. In conclusion, heat storage appears to be localised in PA subjects at rest and centralised for AB subjects.

1. 1. Experiments were carried out concerning the characteristics of wettedness revealed under constant average skin temperature using sitting-resting nude subjects. From the basic measurements of both environmental parameters and human physiological responses, the conclusions detailed below were proposed regarding the changes of wettedness under constant average skin temperature.

2. 2. There is positive correlation between the wettedness and environmental humidity, and negative correlation between the wettedness and air temperature.

3. 3. There is positive correlation between the evaporative heat loss from the skin surface and air temperature, and negative correlation between the evaporative heat loss and environmental humidity.

4. 4. There is negative correlation between the wettedness and evaporative heat loss.

5. 5. Wettedness is not constant but takes varying values, that is, corresponding to each average skin temperature both the maximum and the minimum wettedness values occur.

6. 6. Deriving from the items mentioned above, the theoretical locus of equal average skin temperature is not a straight line, but is a curved line plotted on the psychrometric chart.

1. 1. Seven thermal conditions were imposed on male sitting subjects (slightly clothed: 0.6 clo).

2. 2. A thermal mannikin was also used to determine the exact operative temperature, T0.

3. 3. Conditions were: uniform (UN: all parameters at 24.5°C, air velocity at 0.15 ms−1), heated ceiling (HC at 45°C), heated floor (HF at 34°C), cold floor (CF at 14°C), two conditions of one cold wall at 6°C (CW1 and CW2 respectively with and without air temperature compensation) and increased air velocity (AV at 0.4 ms−1).

4. 4. Local skin temperatures and answers to questionnaires were obtained.

5. 5. Skin temperature variations were affected by conditions and slight T0 changes.

6. 6. Comfort judgments were fairly well related to T0, especially when expressed as differences between actual non-uniform environment and the uniform one.

7. 7. It is concluded that, in case of non-uniform environments close to thermoneutral zone, thermal comfort or discomfort reflects the climate alterations better than the thermal sensation does.

1. 1.|The hyperhermia induced haemolysis of cells and resealed ghosts suspended in isotonic NaCl/sucrose media was studied upon transient heating.

2. 2.|At 61.5°C a process of temperature accelerated disturbance of membrane permeability barrier was initiated, wich was sensed by the consequent volume changes. Concomitantly with this process the thermohaemolysis appeared as a threshold colloid-osmotic lysis.

3. 3.|The initial temperature of this successive barrier disturbance was decreased linearly by ethanol. At 18% ethanol this barrier disturbance took place at 39°C while spectrin was denaturated at about 45°C. Apparently, the spectrin denaturation was not sufficient, nor was involved in, the initiation of this membrane disturbance.

4. 4.|The membrane of cells made ion permeable in the presence of 18% ethanol by heating to 39°C contained irreversible pores with a radius of about 0.45 nm.

5. 5.|This suggests a conformational change of a protein(s) in their formation, but not spectrin nor the anion channel.

6. 6.|Using specific amino reagents it was ascertained, that a superficial NH3+ group dissociable at neutral pH impeded this thermo-induced pore formation.

7. 7.|Consistent results show that this formation of membrane pores initiated at 61.5°C may be included in the still unknown mechanism of thermohaemolysis.

1. 1. This study was conducted to investigate the effects of forced vital capacity on breathing pattern and subjective responses to inspiratory resistance.

2. 2. The subjects were divided into two groups according to their %FVC [large (L) and small (S) group; five subjects in each].

3. 3. Added inspiratory resistances were 0.6 (control), 1.5 (R1), 2.5 (R2), 3.1 (R3) cmH2O · 1−1 · s.

4. 4. Breathing pattern was analyzed by personal computer during rest and exercise with bicycle ergometer.

5. 5. The degree of sensation of breathing difficulty was expressed in SNS reported in our previous study.

6. 6. SNS in S group increased with resistance while no tendency was observed in L group. SNS in S group was significantly greater than that in L group at R3 condition.

7. 7. The breathing pattern of S group was characterized in smaller tidal volume and faster respiratory frequency compared to those of L group with no resistive load.

8. 8. However, outstanding changes in breathing pattern were observed in S group with longer inspiratory time and lower mean inspiratory flow rate when resistive loads were added, which led to increased tidal volume and decreased respiratory frequency.

1. 1.|H2O content and local-tissue thermal conductivity were measured in cortex and medulla of 7 freshly-excised rabbit kidneys.

2. 2.|Tissue H2O content and thermal conductivity k (83.4% and 0.516 W m−t K−1, respectively) in the medulla were significantly higher than those (77.7% and 0.475 W m−1 K−1, respectively) measured in the cortex.

3. 3.|Correlations between the measured parameters are made, and the variability of previously-reported measurements of kidney-tissue thermal conductivity is discussed.

1. 1.|When Chinese hamster ovary cells are treated with cycloheximide (10 μg/ml) or puromycin (100 μg/ml) for 2 h before and during heating at 43°C for 3 h, there is protection from hyperthermic killing; i.e. the plating efficiency increases 2000-fold from 3.7 × 10−5 to (6–9) × 10−2.

2. 2.|The total intracellular levels of spermidine and spermine are not altered by the hyperthermic or drug treatments.

3. 3.|The small 30% decrease in intracellular putrescine observed after heating is not altered by drug treatment.

1. 1. Ten male students remained in a severely cold room (-25°C) for 20 min. thereafter, they transferred in a warm room (25°C) for 20 min.

2. 2. This pattern was repeated three times, total cold exposure time amounting to 60 min.

3. 3. In the warm room, the subjects removed their cold-protective jackets, or wore them continously.

4. 4. Rectal temperature, skin temperatures, manual performance and thermal comfort were measured during the experiment.

5. 5. Removing cold-protective jackets after severe cold exposure increased peripheral skin temperatures and reduced the discomfort in the warm room.

6. 6. However, these results were accompanied by a greater decrease in rectal temperature and manual performance.

7. 7. It is recommended that workers continue to wear cold-protective clothing in the warm areas outside of the cold storage to prevent decreases in deep body temperature and work efficiency caused by repated cold exposures.
