We used a combination of molecular-phylogenetic inference of 82 ITS rDNA sequences and anatomical approach to describe three new west African thelephoroid species, namely Tomentella afrostuposa, T. guineensis and T. guinkoi. Anatomically, T. afrostuposa is reminiscent of T. stuposa with globose to broadly ellipsoid large basidiospores of 8–14 μm, long aculei of up to 3 μm and prominent apiculi of 2 μm width. Molecular-phylogenetically, it falls within the T. stuposa complex. However, T. afrostuposa deviates by at least 7.80–10.74% from T. stuposa in regard with the ITS rDNA sequences. Tomentella guineensis is characterised by long (up to 85 μm) utriform basidia, the presence of reniform basidiospores in lateral view (up to 9 μm) with aculei not exceeding 1 μm and a strong cyanescent reaction of the subhymenial hyphae and basidia in 2.5% KOH. It forms a sister species of the newly described species Tomentella maroana; however, deviating from the last species by at least 9.75–10.04%. The very short, inflated (up to 14 μm) and thick-walled septate (septa up to 1.5 μm) subhymenial hyphae combined with ellipsoid basidiospores (up to 8 μm) and short aculei not exceeding 0.5 μm characterise Tomentella guinkoi. Anatomically, T. guinkoi recalls T. ellisii. Genetic distance between both species ranges from 12.67 to 13.73% according to ITS rDNA sequences analyses. Tomentella guinkoi forms a sister species of the group composed of T. ellisii, T. hjortstamiana and T. pisoniae. Detailed anatomical comparisons between the newly described species and their close relatives are given.  相似文献

In this study, a new tropical African resupinate thelephoroid species, namely Tomentella furcata, is described. Microscopically, it is characterised by mostly simple septate and thick-walled (1–2 μm) subicular and subhymenial hyphae, suburniform basidia with transverse septa and/or infrequently with an internal hypha, large sterigmata of 2–4 μm at base, and distinctly yellow, subglobose to globose basidiospores (both in frontal and lateral view) with long (1–2 μm) and dense, simple or forked spines. Phylogenetically, T. furcata falls within terminal clades of investigated Tomentella species. We discuss ornamentation types of basidiospores for discrimination of resupinate thelepohoroid genera and confirm the limitation of ornament bifurcation as a diagnostic feature of Pseudotomentella. Anatomical dissimilarities between T. furcata and close species are given. Taxonomical novelty: Tomentella furcata Yorou & Agerer.  相似文献

A common resupinate thelephoroid fungus was collected in northern Guinean seasonal forests in central and north of Benin (West Africa). The species is reminiscent of Tomentella umbrinospora with respect to the color and thickness of basidiomata and rhizomorphs, the shape of basidiospores in frontal view and the size of subicular hyphae. Both species fall phylogenetically within two clades. Based on detailed anatomical comparison (mostly of rhizomorphs and basidiospores) with the holotype of T. umbrinospora and phylogenetic analyses including ITS rDNA sequences of 40 Tomentella species, T. africana is described as a new species. Genetic distance between the newly described species and T. umbrinospora is 12.1-12.9%, based on ITS rDNA sequences. T. africana is characterized anatomically by yellow-brown thick (0.3-0.8 mm) monomitic rhizomorphs that are commonly covered by irregularly shaped thin hyphae, thin- to thick-walled subicular hyphae of two size ranges, clavate and clamped basidia of 30-60 microm and light yellow to pale brown echinulate basidiospores with irregular shape in frontal view. Detailed anatomical and molecular dissimilarities between T. africana and close species are discussed. Differences between irregularly shaped surface thin hyphae and skeletal ones are highlighted. We stress the relevance of rhizomorphal structures in the discrimination of resupinate thelephoroid fungi.  相似文献

An anatomical approach coupled with molecular phylogeny of 84 sequences of thelephoroid taxa have been used to describe two new West African resupinate Thelephorales, namely, Tomentella agereri and Tomentella maroana. T. agereri presents a maximal sequence similarity of 94% with its genetically closest species, Tomentalla pilosa, according to a Blastn search in public GenBanks. By molecular phylogenetics, it is nested within the T. pilosa complex, a well-supported (bootstrap support of 100%) monophyletic clade composed of cystidiate and differentiated rhizomorphic species, although it presents contrasting anatomical features including the lack of cystidia, the presence of undifferentiated rhizomorphs, and basidiospores with very short aculei, up to 0.5 μm. Tomentalla maroana is close, by molecular phylogenetic study, to T. ellisii, T. pisoniae, and T. hjortstamiana. The phylogenetic proximity between T. maroana and T. ellisii is supported by morphological characters between the two species, namely, a crustose adherent basidiocarp, a differentiated sterile margin, and a granular hymenium. The two species deviate from each other by 11.38–12.37% with regard to the ITS rDNA sequences, whereas the intraspecific genetic distances vary from 1.68% to 2.9% among the three specimens assigned to T. maroana. Discriminating characters as well as genetic distance between the new species and the closely related species are discussed in detail.  相似文献

This paper presents Tomentella capitata and Tomentella brunneocystidia as new species based on molecular data and anatomical features. Both T. capitata and T. brunneocystidia form sister species with Tomentella pilosa. All three taxa are well supported by bootstrap values. Anatomically, T. capitata and T. brunneocystidia are very close and are similar in shape, size, ornamentation of basidiospores, and size and colour of subicular hyphae. Monomitic rhizomorphs sometimes covered by irregularly shaped thin hyphae are present in both species. Shape and pigmentation of the cystidia are the most discriminating features between T. capitata and T. brunneocystidia. The cystidia of T. capitata are maximum 35 μm long, show a distinctive globose apex and are sometimes covered with dark brown pigmentation and/or encrustation, whereas cystidia of T. brunneocystidia are bigger, up to 55 μm long, with a sub-capitate shape and dark blue to dark green contents all over their length. The differences to species, already described as having capitate and clavate cystidia, are discussed. A key for the identification of cystidioid Tomentella species is given. A new poroid wood-inhabiting basidiomycete, Perenniporia nanlingensis, collected in Guangdong Province, southern China, is described and illustrated on the basis of three collections. Both the morphology and phylogenetic analysis of rDNA ITS sequences support this new species. Macroscopically, the new species is characterized by an annual growth habit, resupinate basidiocarps with pinkish buff to cinnamon-buff pore surface when dry. Microscopically, it has a trimitic hyphal system, slightly dextrinoid and cyanophilous skeletal and binding hyphae, and its basidiospores are ellipsoid, truncate, strongly dextrinoid and cyanophilous, 9.0–9.8 × 5.0–5.9 μm.  相似文献

Ypsilonidium bananisporum sp. nov. belonging to Ceratobasidiales is described and illustrated. This fungus has all the characteristics of the genusYpsilonidium including reticulate-hypochnoid basidiomes, broad hyphae branching at right angles, broadly clavate basidia with two sterigmata, and basidiospores germinating by repetition. It differs from all hitherto known species in the genus by producing suballantoid to banana-shaped basidiospores, measuring 19.5–22×5.5–6 μm. Lignosus hainanensis sp. nov., is described and illustrated on the basis of collections originating from tropical forest in Hainan Province, southern China. Both the morphology and phylogeny support this new species. It is characterized by its centrally stipitate basidiocarps arising from a distinct sclerotium, yellowish brown to cinnamon-brown pileal surface, cream to cream-buff pore surface, trimitic hyphal system with clamped generative hyphae, and oblong ellipsoid to cylindrical basidiospores. Lignosus hainanensis may be confused with L. rhinocerus, also occurring in southern China, but L. rhinocerus can be distinguished by its smaller pores (6–8 per mm) and smaller and distinctly broadly ellipsoid basidiospores (4–5 × 2.8–3 μm).  相似文献

A new homobasidiomycete, Asterostroma macrosporum, was found in mangrove forests of Iriomote Island, Japan. This species is morphologically characterized by having resupinate basidiomata, a monomitic (asterodimitic) hyphal system, simple septate generative hyphae, dextrinoid asterosetae, four sterigmate basidia and globose, tuberculate and amyloid basidiospores measuring 8.5–11 × 7.5–9 μm. It is similar to A. muscicola, but basidiospores in the latter are smaller (7–8 × 5.5–7 μm). Furthermore, phylogenetic analysis using internal transcribed spacer (ITS) region revealed that A. macrosporum is distinctly separated from A. muscicola. In Japan, A. muscicola is widely distributed in warm-temperate to subtropical regions, growing on a variety of broadleaved trees including mangroves, while A. macrosporum has been found only on mangroves.  相似文献

Three new species of Inonotus are described and illustrated. Inonotus acutus sp. nov. is characterized by having small, thin basidiocarps with a strongly contracted base, a sharp and undulate pileus margin, ventricose hymenial setae, and ellipsoid, yellowish to yellow-brown, slightly thick-walled basidiospores. Inonotus chrysomarginatus sp. nov. differs in having an annual to perennial growth habit, pileate basidiocarps with a distinctly yellowish buff to golden-yellow margin, distinct setal hyphae and hooked hymenial setae, and broadly ellipsoid to subglobose, pale yellowish, slightly thick-walled and weakly cyanophilous basidiospores. Inonotus rigidus sp. nov. is distinguished by its resupinate and hard rigid basidiocarps, the honey-yellow pore surface, and smaller pores; microscopically, it has ellipsoid, yellowish brown and thick-walled basidiospores, and lacks both setal hyphae and hymenial setae. An identification key to the Chinese species of Inonotus sensu stricto is provided.  相似文献

Several scientists collected extensively cyphellaceous Hymenomycetes, Basidiomycota, with cup-shaped fruitbodies for the ‘Flora Neotropica’ project. A distinctive group within these fungi comprises genera with characteristically encrusted, non-ramified hyphae on the outer side of their fruitbodies, called surface hyphae. The genus Flagelloscypha, typically provided with whiplash-like distal ends on these hyphae, is one of the better known genera of this group. Here, we propose F. crassipilata as a new species, which is characterised by (3.5)4–6.5(8) μm thick surface hyphae, by extremely big basidia measuring 36–40(44) × 8.5–10.5 μm, and asymmetrically ovoid to asymmetrically ellipsoid, 10.5–12 × (4.5)5–5.5 μm large spores. Differences to similar species are discussed and a short introduction to genera related to Flagelloscypha is given.  相似文献

Zygophiala qianensis is described as a new fungal species associated with the cuticle of apple fruit sampled from an orchard in Shaanxi Province, China. Conidiophores were separate, arising from superficial hyphae, erect, scattered, subcylindrical, irregularly flexuous, consisting of four parts: a hyaline supporting cell that gives rise to a smooth, dark brown stipe, terminating in a finely verruculose, medium brown apical cell that gives rise to (1–)2(–3) medium brown, finely verruculose, doliiform to ellipsoidal, polyblastic conidiogenous cells, with 1–2 prominent scars, apical and lateral, darkened, thickened. Conidia were solitary, fusiform to obclavate, hyaline, smooth and thick-walled, transversely (0–)1(–7)-septate, mostly 1–2-septate, prominently constricted at the septum; apex obtuse, base subtruncate, with a darkened, thickened hilum. Zygophiala qianensis is compared morphologically to other species of Zygophiala, and a phylogenetic analysis of their DNA sequence data is presented.  相似文献

The ectomycorrhizal status of Sistotrema sp. is shown by morphological–anatomical and molecular identification, confirming earlier reports about Sistotrema DNA in ectomycorrhizae (ECM). For molecular identification of the ECM nuclear rDNA ITS sequences obtained from mycorrhizal root tips and fruitbodies of Sistotrema sp. were compared. Blast searches using the Sistotrema sp. sequences as query were performed in GenBank and UNITE for comparison with previously published Sistotrema sequences. The morphological–anatomical characterization of the ECM used well-established protocols for the examination of all mantle parts and rhizomorphs in different sections and views including detailed illustrations. The ECM are irregularly monopodial–pyramidal, whitish ochre to yellow ochre, and woolly. Older ones become more greyish and silvery at some patches. Diagnostic anatomical characteristics are irregularly inflated emanating and rhizomorph hyphae, ampullately inflated clamps, and the occurrence of yellow drops within the hyphae. The plectenchymatous mantle shows ring-like arranged hyphae, and a slightly gelatinous matrix. The ECM of Sistotrema sp. are compared to those of other species that form distinctly ampullate hyphae in rhizomorphs, too. The anatomically most similar ECM to those of this Sistotrema specimen are those of Hydnum repandum.  相似文献

Amyloporia pinea sp. nov. is described and illustrated on the basis of collections from southern China. Morphology and phylogenetic analysis of rDNA ITS sequences support this new species. Morphologically, it is characterized by resupinate, annual basidiocarps, cream to yellowish buff pore surface when fresh, which becomes yellowish brown to clay-buff upon drying, a dimitic hyphal system with clamped generative hyphae and inamyloid skeletal hyphae, fusoid cystidioles, and cylindrical basidiospores; moreover, it causes a brown rot. Molecular phylogeny inferred from ITS sequence data suggested a close relationship between A. pinea and Amyloporia crassa sensu lato. Antrodia subxantha has amyloid skeletal hyphae, and grouped within the Amyloporia clade, hence, it is transferred to Amyloporia, and a new combination Amyloporia subxantha is proposed.  相似文献

A new coprophilous species, Coprinellus radicellus, is presented and described on the basis of morphological characters and a species phylogeny inferred from ITS1–5.8S–ITS2 and beta-tubulin gene sequences. The species is characterized by lageniform pileo- and caulocystidia, 8- to 11-μm long ellipsoid basidiospores with a central germ-pore, globose cheilocystidia, an often rooting stipe, the lack of pleurocystidia and velar elements. These characters distinguish C. radicellus from all other described species in subsection Setulosi. It comes closest to C. brevisetulosus (Arnolds) Redhead, Vilgalys & Moncalvo and C. pellucidus (P. Karst.) Redhead, Vilgalys & Moncalvo both morphologically and phylogenetically, but is distinct from both. Another five representatives of morphologically recognizable groups of subsection Setulosi were included in the phylogenetic analyses and found were distinct from C. radicellus, both morphologically and phylogenetically. To date, this new species is known only from Scandinavia, which is surprising in view of the uniform geographical distributions of most setulose Coprinellus species. A key to coprophilous taxa of Coprinellus with pileo- and caulocystidia is presented. Three new combinations are proposed.  相似文献

A species of Aphanomyces was isolated from juvenile soft-shelled turtles, Pelodiscus sinensis, cultured in Japan. Typically, an infected turtle showed small whitish maculae on the carapace. Many hyphae were observed in the epidermis. The hyphae were isolated using glucose–yeast (GY) agar plates. The morphological characteristics were very similar to those of Aphanomyces laevis, but a clear nuclear spot was observed in the center of the oospore in the strains isolated from the soft-shelled turtles. The optimal growth temperature for the isolates was 25–30°C and the optimum pH was 6–9. Experimental infection tests with isolates produced small whitish maculae on the carapace, and soft-shelled turtles artificially infected with the zoospores showed high mortality, especially in the high-dose group. Phylogenetic analysis based on the internal transcribed spacer (ITS) region of ribosomal DNA (rDNA) indicated that the isolates from the soft-shelled turtles were unidentified species of Aphanomyces. As a result, the strain was described as a new species, Aphanomyces sinensis.  相似文献

To determine the species of the yeast-like symbionts (YLS) in the brown planthoppers (BPH), Nilaparvata lugens, YLS were first isolated and purified by ultracentrifugation from the fat bodies of BPH, and then 18S rDNA and internal transcribed spacer (ITS)–5.8S rDNA sequences of YLS were amplified with the different general primers for fungi. The results showed that the two different 18S and ITS–5.8S rDNA sequences of YLS were obtained. One 2291-bp DNA sequence, which contained 18S and ITS–5.8S rDNA, showed the high similarity to Cryptococcus and was named Cryp-Like symbiotes. Another 1248-bp DNA sequence, which contained a part of 18S and ITS–5.8S rDNA, showed the high similarity to Pichia guilliermondii and was named Pichia-Like symbiotes. It was further proved that Cryp- and Pichia-Like symbiotes existed in BPH through nested PCR with specific primers for two symbiotes and in situ hybridization analysis using digoxigenin-labeled probes. Our results showed that BPH harbored more than one species of eukaryotic YLS, which suggested that diversity of fungal endosymbiotes may be occurred in planthoppers, just like bacterial endosymbiotes.  相似文献

A species of Aphanomyces was isolated from the ice fish Salangichithys microdon living in brackish water in Japan. White cotton-like growth was found on the heads and fins of the fish. Hyphae penetrated into the dermal layers, subcutaneous tissues, muscular layers, and cartilaginous tissue of the mandible and maxilla; these hyphae were associated with cellular debris and lesions in host tissue. White fluffy colonies from subcultures of these growths were isolated on glucose–yeast agar plates with 0.5% sodium chloride (NaCl). These isolates consisted of delicate, slightly wavy, and moderately branched hyphae. Zoosporangia were isodiametric with the vegetative hyphae. Oogonia were abundant and approximately 21–33 μm in diameter, with irregular short papillae. Generally they were spherical or subspherical and only rarely pyriform. Individual oogonia usually contained a single oospore, which was spherical and 19–27 μm in diameter, with a large shiny vesicle. Antheridial branches, when present, were usually androgynous; however, they were sometimes monoclinous or diclinous. The optimal growth temperature of the isolates was 20°C, and cultures grew well at low salinity (0–0.5% NaCl). Phylogenic analysis based on the internal transcribed space 1-5.8S-ITS 2 of the ribosomal RNA gene indicates that these isolates will be an as-yet unidentified species of Aphanomyces.  相似文献

Three new species of Agaricales are described and illustrated from eastern Honshu, Japan:Agrocybe pseudoerebia sp. nov. (sectionTriviales of subgenusRussularia), forming a pale grayish, strongly glutinous pileus and watery, latex without discoloration, was found on the ground in a lowland forest dominated byQuercus myrsinaefolia andQuercus serrata; Tricholoma foliicola sp. nov. (close to sectionAlbobrunnea), forming a reddish brown, hygrophanous, dry, glabrous pileus, almost adnate, densely crowded lamellae, small, ellipsoid basidiospores, and clampless hyphae, was found on leaf litter of a broad-leaved forest.  相似文献

On the family Brassicaceae, the causal agent responsible for downy mildew disease was originally regarded as a single species, Peronospora parasitica (now under Hyaloperonospora), but it was recently reconsidered to consist of many distinct species. In this study, 11 specimens of Peronospora drabae and P. norvegica parasitic on the genus Draba were investigated morphologically and molecularly. Pronounced differences in conidial sizes (P. drabae: 14–20 × 12.5–15.5 μm; P. norvegica: 20–29 × 15.5–22 μm) and 7.8% sequence distance between their ITS1-5.8S-ITS2 rDNA sequences confirmed their status as distinct species. Based on ITS phylogeny and morphology (monopodially branching conidiophores, flexuous to sigmoid ultimate branchlets, hyaline conidia and lobate haustoria), the two species unequivocally belong to the genus Hyaloperonospora and not to Peronospora to which they were previously assigned. Therefore, two new combinations, Hyaloperonospora drabae and H. norvegica, are proposed. The two taxa are illustrated and compared using the type specimen for H. norvegica and authentic specimens for H. drabae, which is lectotypified.  相似文献
