Eu3+-tetracycline complex (EuT) increased the chemiluminescence (CL) intensity of linolenic acid micells (UFA-somes) oxidized with lipoxygenase and CL of the lecithin liposomes peroxidized with Fe2+ ions by 3 orders of magnitude. In the systems producing oxygen radicals (xanthine + xanthine oxidase and Fenton's reagent) EuT was ineffective. Luminol increased CL intensity up to 4 orders of magnitude in Fenton's reagent and by 2 orders of magnitude in xanthine oxidase reaction. The sensitization of CL in Fe2+-induced lipid peroxidation (LPO) of liposomes was by a factor 40, while in lipoxygenase reaction very low sensitization was observed. By means of cut-off light filter OS-12 (Soviet) having short wave-length transmittance limit at 560 nm it was possible to measure separately in the same sample the luminol-sensitized CL (maximal emission near 480 nm) and EuT-sensitized CL (maximum at 620 nm); these two CL components reflect, correspondingly, the production rate of oxygen- and lipid-free radicals. Mannitol, the OH radical scavenger, inhibited luminol-dependent component of CL in peroxidized liposomes and did not inhibited EuT sensitized CL in the same system. Apparently, hydroxyl radicals are produced in LPO reactions and responsible for the effect of CL sensitization by luminol, but are not involved in the chain LPO process.  相似文献

Two chemiluminescence approaches have been used for study of active oxygen species produced by the TiO2 photocatalytic reaction. One is based on flow injection analysis (FIA)-luminol chemiluminescence (CL); another is a time-resolved CL method. In the FIA-CL experiment, an UV-illuminated TiO2 suspension and water were passed into a mixing cell by two separate flow lines. Luminol solution was injected into the water flow line at different times. The injected luminol reacted with active oxygen species generated by the TiO2 photocatalytic reaction in a mixing coil and produced CL. It was found that the maximum CL was detected at the first injection of luminol. CL intensity decreased with time of injection. When the luminol was injected after 5 min, the CL intensity was almost unchanged. Addition of scavengers of active oxygen species indicated that the CL produced early in the 5 min was caused by O2- and H2O2, while CL after 5 min was only from H2O2. In the time-resolved CL, the third harmonic wavelength of Nd:YAG laser (355 nm) was used as a UV light source, and CL was detected by a PMT and recorded in a millisecond time scale using a digital oscilloscope. It was found that CL induced by the photocatalytic reaction increased with concentration of the TiO2 suspension. Scavengers of active oxygen species of *OH, O2- and H2O2 were added to study the involvement of the active oxygen species.  相似文献

Cardiolipin (CL), a unique mitochondrial phospholipid synthesized by CL synthase (CLS), plays important, yet not fully understood, roles in mitochondria-dependent apoptosis. We manipulated CL levels in HeLa cells by knocking down CLS using RNA interference and selected a clone of CL-deficient cells with ~ 45% of its normal content. ESI–MS analysis showed that the CL molecular species were the same in CL-deficient and CL-sufficient cells. CL deficiency did not change mitochondrial functions (membrane potential, reactive oxygen species generation, cellular ATP levels) but conferred resistance to apoptosis induced by actinomycin D (ActD), rotenone, or γ-irradiation. During ActD-induced apoptosis, decreased CL peroxidation along with suppressed cytochrome (cyt) c release was observed in CL-deficient cells, whereas Bax translocation to mitochondria remained similar to that in CL-sufficient HeLa cells. The amounts of loosely bound cyt c (releasable under high ionic strength conditions) were the same in CL-deficient and CL-sufficient cells. Given that CL peroxidation during apoptosis is catalyzed by CL/cyt c complexes and CL oxidation products are essential for cyt c release from mitochondria, our results suggest that CL deficiency prevents adequate assembly of productive CL/cyt c complexes and CL peroxidation, resulting in increased resistance to apoptosis.  相似文献

The effect of heparin on peroxidation of cardiolipin (CL) initiated by ferrous iron was studied in vitro using detergent-solubilized CL, liposomal CL, or CL bound to isolated cytochrome c oxidase (CcO). Heparin increased both the rate and the extent of CL peroxidation for detergent-solubilized CL and for CcO-bound CL. The effect of heparin was time- and concentration-dependent as monitored by the formation of conjugated dienes or thiobarbituric acid reactive substances. The results showed great similarity between the effect of heparin and the effect of certain iron chelators, such as ADP, on phospholipid peroxidation. Heparin increased the peroxidation of CcO-bound CL only when tertiary butyl hydroperoxide was also present. The enzyme activity of the resulting CcO complex decreased 25 %, in part due to peroxidation of functionally important CL. In contrast to peroxidation of detergent-solubilized CL, peroxidation of liposomal CL was inhibited by heparin, suggesting that the effect of heparin and ferrous iron depends on their proximity to the acyl chains of CL.  相似文献

The correlation between the oxidative processes in tert-butyl hydroperoxide (tBHP)-exposed red blood cells and the reactions of oxygen consumption and release were investigated. Red blood cell exposure to tBHP resulted in transient oxygen release followed by oxygen consumption. The oxygen release in red blood cells was associated with intracellular oxyhaemoglobin oxidation. The oxygen consumption proceeded in parallel with free radical generation, as registered by chemiluminescence, but not to membrane lipid peroxidation. The oxygen consumption was also observed in membrane-free haemolyzates. The order of the organic hydroperoxide-induced reaction of oxygen release with respect to the oxidant (tBHP) was estimated to be 0.9 +/- 0.1 and that of the oxygen consumption reaction was determined to be 2.4 +/- 0.2. The apparent activation energy values of the oxygen release and oxygen consumption were found to be 107.5 +/- 18.5 kJ/mol and 71.0 +/- 12.5 kJ/mol, respectively. The apparent pKa value for the functional group(s) regulating the cellular oxyHb interaction with the oxidant in tBHP-treated red blood cells was estimated to be 6.7 +/- 0.2 and corresponded to that of distal histidine protonation in haemoprotein. A strong dependence of tBHP-induced lipid peroxidation on the oxygen concentration in a red blood cell suspension was observed (P50 = 32 +/- 3 mmHg). This dependence correlated with the oxygen dissociation curve of cellular haemoglobin. The order of the membrane lipid peroxidation reaction with respect to oxygen was found to be 0.5 +/- 0.1. We can conclude that the intensity of the biochemical process of membrane lipid peroxidation in tBHP-exposed erythrocytes is controlled by small changes in such physiological parameters as the oxygen pressure and oxygen affinity of cellular haemoglobin. NADPH-menadione reductase activity by rat brain microsomes (Ms) was decreased 40-50% by 10 microM dicumarol, a potent inhibitor of DT-diaphorase, whereas no change in NADPH-paraquat (PQ) and -diquat (DQ) reductase activity was observed. NADPH-DQ reductase activity in brain Ms was 2.5-fold higher than NADPH-PQ reductase activity. The formation of PQ and DQ radicals was verified optically and observed directly by ESR spectroscopy in the NADPH-PQ and -DQ reductase reactions by brain Ms under anaerobic conditions. PQ- and DQ-induced superoxide formation was confirmed by the detection of DMPO-OOH ESR signals and followed by chemiluminescence (CL) of a Cypridina luciferin analogue (CLA). The kinetics and intensity of the CL were consistent with the observations that the reduction in DQ is faster than that in PQ. Thiobarbituric acid reactive substances (TBARS) and phospholipid hydroperoxides in brain Ms increased in the presence of NADPH and Fe3+. The generation of both lipid peroxidation products derived from brain Ms decreased with increasing concentrations of PQ and DQ. The inhibitory effect of DQ is more pronounced than that of PQ. The formation of PQ- and DQ-induced reactive oxygen species was not associated with lipid peroxidation in rat brain Ms.  相似文献

Light emission (chemiluminescence; CL) was observed in the reaction of anthocyanins with tert-butyl hydroperoxide (t-BuOOH) in the presence of acetaldehyde. The intensity of the CL of the anthocyanins was in the order of nasunin > rubrobrassicin > delphinidin > malvin = cyanidin > malvidin, indicating that glucosylation at C-3 and C-5 of the anthocyanin skeleton enhances the CL of the parent compound. CL intensity was enhanced at alkaline pH. The results suggest that the antioxidant effect of anthocyanins on lipid peroxidation, which is observed in the linoleic acid-β-carotene-lipoxcygenase system, is at least partly due to their strong reactivity with hydroperoxides.  相似文献

Rat liver microsomal suspension (1 mg protein per ml) was incubated at 37 degrees C with 5 mM salicylic acid and 0.2 mM NADPH. The amounts of thiobarbituric acid reactive substances (TBARS) and 2,5-dihydroxybenzoic acid (2,5-DHB), an oxidative metabolite of salicylic acid increased with the incubation time. Simultaneously spontaneous chemiluminescence (CL) was found to be generated there. The addition of SKF-525A, an inhibitor of cytochrome P450 (P450), to the reaction mixture inhibited the CL generation together with the inhibition of the oxidative metabolism. The anti-oxidants and singlet oxygen scavengers like N,N-diphenylphenylenediamine (DPPD) and histidine suppressed the CL generation. The addition of 1,4-diazabicyclo [2.2.2] octane (DABCO), a singlet oxygen quencher, to the reaction mixture generating CL enhanced CL transiently and then CL decreased markedly. Thus CL observed here may possibly originate from the singlet oxygen. The CL generation was suggested to be closely related with salicylic acid-induced lipid peroxidation, and to be coupled with the oxidative metabolism mediated by P450 in rat liver microsomes.  相似文献

Bilirubin in an alkaline solution exhibits a weak chemiluminescence (CL) under aerobic conditions. This spontaneous CL was markedly enhanced by the addition of various aldehydes. The fluorescent emission spectrum of bilirubin, excited by weak intensity light at 350 nm, coincided with its CL emission spectrum (peak at 670 nm). CL emission from bilirubin was not quenched by active oxygen scavengers. This suggests that triplet oxygen reacts with bilirubin, and forms an oxygenated intermediate (hydroperoxide) as a primary emitter (oxidative scission of tetrapyrrole bonds in bilirubin is not involved in this CL). The Ehrlich reaction (test for monopyrroles) and hydrolsulphite reaction (test for dipyroles) on the CL reaction mixture and unreacted bilirubin showed no differences. When the CL was initiated by singlet oxygen, rather than superoxide anion, monopyrrole, was detected in the reaction products by gel chromatography. The inhibitory effect of a scavenger of singlet oxygen on CL was eliminated in the presence of formaldehyde. Therefore, triplet carbonyl, formed by singlet oxygen through the dioxetane structure in bilirubin, is not an emitter. The reaction mechanism of bilirubin CL and the formation of a hydroperoxide intermediate is discussed in relation to the chemical structure of luciferin molecules from bioluminescent organisms.  相似文献

紫外A(UVA,320 nm-400 nm)诱发的脂质过氧化反应是通过活性氧(ROS)介导的。在UVA照射之后,单线态氧(1O2)和超氧阴离子(O2-.)是细胞内最初产生的ROS,它们进一步生成过氧化氢(H2O2),羟自由基(.OH)等其它自由基。为了探讨UVA照射后最早生成的1O2和O2-.与细胞氧化损伤后果的关系,我们采用一种特异性检测1O2和O2-.的高灵敏度化学发光探针MCLA(2-methyl-6-(4-methoxyphenyl)-3,7-dihydroimid-azo[1,2-α]pyrazin-3-one hydrochloride)检测人外周血淋巴细胞经UVA照射后的化学发光变化。发现不同剂量UVA照射后,细胞MCLA化学发光变化和MDA浓度变化一致。结果表明UVA照射后1O2和O2-.的水平与由此引发的脂质过氧化损伤存在正相关关系。因此,MCLA化学发光方法可望作为一种检测UVA诱发脂质过氧化水平的简单快速方法。  相似文献

Using a highly sensitive single photon counter, a spontaneous chemiluminescence (CL) study on rice (Oryza sativa L.) seeds stored in different years was carried out. We first observed that the degree of ageing in rice seeds was related to the intensity of spontaneous CL during early imbibition (0–30 min). Rice seeds stored for a shorter time had a stronger intensity of CL in early imbibition. The germination rate of rice seeds showed an obvious positive correlation with the intensity of spontaneous CL. Singlet oxygen (1O2) in rice seeds during early imbibition was investigated by a CL method using a cypridina luciferin analogue, 2‐methyl‐6‐(p‐methoxyphenyl)‐3,7‐dihydroimidazo [1,2α] pyrazin‐3‐one (MCLA), as a selective CL probe. Additional experimental evidence for the formation of 1O2 came from the quenching effect of sodium azide (NaN3) on MCLA‐mediated rice seeds' CL. Analysis based on the experimental results demonstrated that spontaneous CL in rice seeds during early imbibition was mainly contributed by singlet oxygen (1O2). The healthy intact polymorphonuclear leukocytes (PMNs) were labeled with 4-maleimide-TEMPO spin labeling compound (MAL) to study the effects of oxygen radicals produced by phorbol myristate acetate (PMA)-stimulated PMNs on the conformation of sulfhydryl (SH) groups of PMN membrane proteins. The lipid peroxidation induced by PMA-stimulated PMNs was detected by evaluating the formation of malonaldehyde (MDA) with the thiobarbituric acid (TBA) test. From the experiments of luminol-dependent chemiluminescence (CL) and fluorometry, it was found that Chinese herbs schizandrin B (Sin B) and quercetin (Q) possessed scavenging properties for oxygen radicals produced during the PMN respiratory burst. These two herbs can also inhibit the conformation changes in SH binding sites on the PMN membrane proteins caused by oxygen radicals produced by the PMNs themselves. They also decreased the amount of MDA, which was a final product formed during lipid peroxidation.  相似文献

Extra-weak chemiluminescence (CL) from amino-carbonyl reactions of L-lysine with various sugars and aldehydes in aqueous solution was examined. Amongst the aldehydes and sugars tested, glycolaldehyde and D -arabinose produced the highest CL intensity. The CL of the amino-carbonyl reaction reached a maximum after about 60 minutes. The CL was pH dependent, and a linear relation between CL intensity and hydroge-ion concentration was demonstrated. Low oxygen levels inhibited CL and no CL was produced in nitrogen purged solutions. Addition of cupric or ferrous ion, decreased the CL. The involvement of free radical intermediates was demonstrated by ESR. Our findings suggest that the CL of the amino-carbonyl reaction arises from free radicals derived from melanoidines or their intermediates. CL should prove useful for evaluating the stability of crude drugs extracted from natural resources that contain various amino acid derivatives protein and sugar components.  相似文献

An acridinium ester (AE) alkaline solution can react with Mn(II) to generate a strong chemiluminescence (CL) centered at 435 nm. The effects of reaction conditions such as pH and Mn(II) concentration on CL intensity were examined. In order to explore the CL mechanism, the effect of oxygen on the CL reaction was examined and an X‐ray photoelectron spectroscopy study of the reaction precipitate was carried out. The results indicated that oxygen participated in the CL reaction and Mn(IV) was the primary product in the system. A possible mechanism was proposed that involved two pathways: (1) dissolved oxygen was reduced to reactive oxygen radicals by Mn(II), these reactive intermediates then reacted with AE to produce excited state acridone; (2) Mn(II) could reduce AE to partly reduced AE, which then reacted with oxygen to form excited state acridone. The reactions of other metal ions with AE were also tested, and only Mn(II) was shown to trigger strong CL emission of AE, which indicated that the system had good selectivity for Mn(II). It has been reported that weak chemiluminescence (CL) from crude extracts of soybean seedlings is remarkably enhanced with the addition of various aldehydes (Biochim. Biophys. Acta 1058, 209-216). The reactivity of certain emitter(s) with oxygen species was examined in the autoclaved extracts of seedlings. When samples were reduced by the addition of hydrosulfite, two different types of reactivities in CL were defined. One type showed an initial rapid increase and a subsequent fast decay in CL upon mixing with oxygen. This rapid increase in CL intensity was independent of the presence of aldehydes, and was significantly suppressed by SOD. However, the subsequent slow decay phase in CL was dependent on the presence of aldehydes. In the sample reduced more moderately by borohydride, the same slow decay of CL appeared upon mixing with acetaldehyde and oxygen. This second type of CL was not inhibited by active oxygen scavengers. Hydrogen peroxide added to unreduced (oxidized) samples also elicited CL. Three types of primary emitters may be oxidized to form transient hydroperoxide, and excited for light emission by slightly different ways: two of them are excited by abstraction of one atomic oxygen from the hydroperoxy intermediate with aldehyde or hydrogen peroxide, leading to formation of an excited hydroxide intermediate. The third is excited directly on the binding of superoxide anion to the reduced primary emitter.  相似文献

Previously it was shown that lipophilic analogs of a free-radical scavenger, 2(3)-tert-butyl-4-hydroxyanisole (BHA), inhibit ornithine decarboxylase (ODC) activity which is induced by 12-O-tetradecanoylphorbol-13-acetate (TPA) in mouse epidermis. With regard to this antitumor-promoting effect, eight analogs of BHA (2- and 3-BHA, 2-t-butyl-1, 4-dimethoxybenzene methyl-BHA), t-butylhydroquinone (t-BHQ), p-hydroquinone (HQ), 4-hydroxyanisole, phenol and 2-t-butylphenol) are evaluated herein for their antioxidant capacities for scavenging superoxide anions (O-2), of inhibiting lipid peroxidation and of inhibiting chemiluminescence (CL) in TPA-activated polymorphonuclear leukocytes (PMNs), an event associated with oxy-radical production. None of the analogs reacted with O-2, while 2- and 3-BHA suppressed the formation of O-2 by TPA-activated PMNs. T-BHQ underwent autoxidation in aqueous solution, reducing molecular oxygen and increasing the levels of O-2 that were formed chemically, enzymatically and cellularly. However, all of the phenolic antioxidant analogs of BHA inhibited TPA-stimulated CL in PMNs and ascorbate-initiated lipid peroxidation, while methyl-BHA (a non-antioxidant analog) was inactive. The inhibitory activities of these analogs for lipid peroxidation were related to both their lipophilic and antioxidant properties and corresponded favorably with their inhibitory activities for TPA-induced ODC activities in mouse epidermis. On the other hand, inhibition of the CL response by these antioxidants was independent of their lipophilicity and compared less favorably with their capacities to antagonize phorbol ester-induced ODC activity. These results imply that lipophilic BHA analogs inhibit TPA-induced ODC activity by scavenging free radicals other than O-2. Furthermore, the fact that t-BHQ was the most potent inhibitor of CL, lipid peroxidation and ODC activity and simultaneously reduced molecular oxygen, suggests the possibility that O-2 may act as a precursor to the formation of free radicals which are reactive with t-BHQ and more directly involved in the process of tumor promotion.  相似文献

Hydrogen peroxide formed during the course of the copper(II)-catalysed oxidation of cysteamine with oxygen was continuously determined by a peroxidase (POD)-catalysed luminol chemiluminescence (CL) method. Horseradish peroxidase (HRP), lactoperoxidase (LPO) and Arthromyces ramosus peroxidase (ARP) were used as a CL catalyst. The respective PODs gave specific CL intensity-time profiles. HRP caused a CL delay, and ARP gave a time-response curve which followed the production rate of H2O2. LPO gave only a weak CL flash which decayed promptly. These differences of CL response curves could be explained in terms of the different reactivities of PODs for superoxide anion and the different formation rate of luminol radicals in the peroxidation of luminol catalysed by POD.  相似文献

Lucigenin-enhanced chemiluminescence (CL) makes it possible to study directly the reactions of superoxide anion radical (O 2 ? ) produced by mitochondria and is used for detection of the superoxide generated in enzyme and membrane systems, isolated mitochondria and cells, and whole animal organs. Lucigenin-enhanced CL is applicable to examination of human tissues; this assay requires small fragments as, for example, biopsies; however, no systematic studies with these objects have been conducted. In this work, we studied the lucigenin-enhanced CL in tissue specimens of various rat organs to determine its dependence on the degree of tissue fragmentation, storage time, and oxygen access. Addition of lucigenin to tissue fragment, slurry, or homogenate drastically increased CL; note that the whole tissue fragment displayed considerably lower (by 1–1.5 order of magnitude) CL intensity than the homogenate or slurry. The lucigenin-enhanced CL in tissue in the absence of stirring of the surrounding solution rapidly decreased, presumably because of a drop in the tissue oxygen level as a result of its consumption. The CL had two components, namely, lucigenin-enhanced CL and lucigenin-independent (intrinsic) CL. The tissue itself was the source of lucigenin-enhanced CL, and this CL was observed only in the case of sufficient oxygen access. The lucigenin-independent CL component was virtually independent of oxygen and was determined by the components released from the tissue specimen into the surrounding solution. Nitric oxide inhibited CL with increase in its concentration but had no essential effect on the rate of tissue oxygen consumption. These results suggest that lucigenin can be used as a very efficient chemiluminescent probe for superoxide radicals generated by tissue fragments.  相似文献

The chemiluminescence (CL) phenomena of lanthanide (Ln) ions and their coordinate complexes in peroxomonosulfate system and the energy transfer mechanism during the process were investigated in this work. A strong and sharp CL signal was yielded when the Eu(III) or Tb(III) solution was added to the peroxymonosulfate solution. The CL intensity was greatly enhanced by 2,6‐pyridinedicarboxylic acid (DPA) ligand [maximum enhancement reached when Ln(III):DPA was 1:1] and hexadecyltrimethylammonium chloride micelles. The degree of enhancement of DPA and micelles on Ln(III) CL was related to the fluorescence lifetimes of Ln(III) in different media. According to the ESR spin‐trapping experiments of 2,2,6,6‐tetramethyl‐4‐piperidone and the specific quenching experiments of 1,4‐diazabicyclo[2.2.2]octane and sodium azide, singlet oxygen was generated though the Ln(III) ion‐catalyzed decomposition of peroxymonosulfate. From the comparisons of the fluorescence and CL spectra, lanthanide ions were the luminescence emitter and the ligand DPA absorbed the energy from singlet oxygen and transferred it to Ln(III) ions in the coordinate complexes. Micelles can enhance the CL intensity by improving intermolecular energy transfer efficiencies, removing the quenching effect of water and prolonging the lifetime of singlet oxygen. The effect of alpha-tocopherol, ascorbate, rutin and dihydroquercetin on chemiluminescence (CL) accompanying the Fe2+-induced peroxidation of unsaturated fatty acids in phospholipid liposomes has been investigated. The amplitude of CL decreased and the latent period increased in the presence of alpha-tocopherol, rutin and dihydroquercetin which is typical of peroxide radical traps. Ascorbate also reduced the CL amplitude but only at small concentrations up to about 4 microM. A further increase of ascorbate concentration had a negligible effect on the amplitude. At the same time, the latent period in CL development increased with the growth of ascorbate concentration, apparently, as a result of recycling of divalent iron oxidized in the course of lipid peroxidation. The effects of rutin and dihydroquercetin on the liposomal CL in the presence of alpha-tocopherol and ascorbate in all experiments were almost the same as when these compounds were added individually. The antioxidant effects were merely summed up without any mutual enhancement or inhibition of each other's action.  相似文献
