蚯蚓中抗肿瘤蛋白组分的提取分离及其抗肿瘤活性   总被引:13,自引:0,他引:13  
以赤子爱胜蚓为原料 ,通过蛋白质的丙酮沉淀和凝胶过滤 ,分离得到一组抗肿瘤活性蛋白成分 (简称蚯蚓抽提物 ) ;这组成分富含Fe、Zn、Cu以及Se等微量元素 ,蛋白含量为 6 0 4 3± 2 36 %(n =5 ) .体外实验中 ,通过MTT法和SRB法测定了蚯蚓抽提物对多种人癌细胞株 (HCT 116、SY5Y、K5 6 2、MGc80 3和HeLa)以及正常细胞株 (HEK2 93和COS 7)的抑制杀伤率 .结果表明 ,蚯蚓抽提物对癌细胞的杀伤有一定的选择性 ,受试人癌细胞达到 5 0 %生长抑制率所需作用浓度约 6 0~110mg L ;但是 ,10 0℃煮沸 5min后 ,该抑制活性完全消失 .通过纤维蛋白平板法 ,测得蚯蚓抽提物同时具有纤溶酶和纤溶酶原激活酶的活性 .体外测得丝氨酸蛋白酶抑制剂aprotinin和PMSF能显著抑制蚯蚓抽提物的细胞杀伤活性 .体内实验中 ,蚯蚓抽提物能有效延长荷瘤 (S180 )小鼠的生存时间 ,并使其身体机能得到明显改善 .腹膜内注射剂量为 2 8mg kg和 36mg kg时 ,生命延长率分别为135 3%和 12 3 5 % ,和标准药物 (环磷酰胺 )治疗后结果 (76 5 % )相比 ,有显著性差异  相似文献

In order to assess the response of epigeic earthworms to seasonal changes we monitored the population dynamics of Eisenia fetida (Oligochaeta, Lumbricidae) in a manure heap in the field during a year. Earthworms were hand-sorted from five 0.25 x 0.25 x 0.20 m blocks around the heap in November (autumn) 1999 and in January (winter), April (spring) and August (summer) 2000 to determine earthworm population dynamics. Earthworms of each block were classified into different age classes: mature, preclitellate, juvenile, hatchling and cocoon, and afterwards counted and weighed. Seasonality had a strong effect on the density, biomass and reproductive activity of the population. The population of E. fetida was characterized by a high density of individuals and the predominance of mature individuals throughout the year. Maximum density, mating activity and size of cocoons were achieved in spring, but there were not changes in the number of cocoons per mature earthworm throughout the year. Unexpectedly, the smallest cocoons were produced in winter by the largest individuals. These results suggest that E. fetida is able to allocate resources to growth and/or reproduction in response to environmental fluctuations.  相似文献

There are increasing concerns over the environmental implications involved in battery plant due to the elevated metal concentrations in the surrounding soils. Earthworms are regarded as a model organism for evaluation of terrestrial environmental pollution. The biochemical responses in the earthworms, Eisenia fetida, exposed to polluted soils were investigated. Soil samples at 0, 50, 100, 200, 500 and 1000 m from a battery plant were collected and used to assess the relationship between sampling distance and biochemical responses in E. fetida. Biochemical alterations were carried out on 1, 3, 7 and 14 d of exposure. The metallothionein (MT), malondialdehyde (MDA) and antioxidant enzymes (superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD)) were measured. The results demonstrated that concentrations of heavy metals, especially Pb, decreased sharply with increase in distance, which may be due to the leaching and accumulation of Pb in soil during production processes. The concentrations of MT and MDA increased with exposure (dosage and time), indicating that E. fetida presented dose and time dependence after heavy metal exposure. The activities of SOD and CAT increased after heavy metal induction, while the activity of POD was lower than that of the control group. The SOD, CAT and POD present a cooperative effect for the protection of lipids and enzymes due to the pollution. The soils around the battery plant were polluted with metals giving rise to adverse biological effects.  相似文献

Comparative studies were performed to evaluate composting potential, biomass growth and biology of a non-native (Eisenia fetida) and an endemic (Lampito mauritii) species of earthworm in the semiarid environment of Jodhpur district of Rajasthan in India. Earthworms were reared in a mixed bedding material comprised of biogas slurry, cowdung, wheat straw, leaflitter, sawdust and kitchen waste. The percentage of organic carbon of the culture bedding material declined upto 105 days with E. fetida and 120 with L. mauritii. The percentage of nitrogen, phosphorous and potassium increased as a function of the vermicomposting period. In contrast, C/N and C/P ratios decreased day by day. Both species were effective for decomposition and mineralization of mixed bedding in the semiarid environment. A comparative assessment of biomass growth of E. fetida and L. mauritii was done under controlled laboratory conditions. The optimum temperature, moisture content and pH for E. fetida were 25 degrees C, 70% and 6.5, respectively. However, the optimum temperature, moisture content and pH for growth and development of L. mauritii were 30 degrees C, 60% and 7.5, respectively. The biology and reproductive rates of both species were also studied in the laboratory using mixed bedding. Cocoon production was higher for E. fetida than L. mauritii. The net reproductive rate was 9 per month in the case of E. fetida and 1 per month for L. mauritii. Fertilized eggs of E. fetida and L. mauritii developed into adults within 4 and 5 1/4 months, respectively. These observations indicate E. fetida may be a more efficient breeder than L. mauritii in the desert region of Rajasthan.  相似文献

Ártioposthia triangulata is a terrestrial planarian that feeds on earthworms. Up to 76% of field collected A. triangulata fed on Eisenia fetida earthworms at 10°C in the laboratory. In one experiment, the feeding rate declined from 63% of earthworms attacked to 36% and 34% in the first, second and third feeding opportunities over a week. The average number of E. fetida eaten was 1.4 per planarian per week. The gain in weight of individual A. triangulata was significantly related to the amount of earthworm tissue lost during the feeding process. Over a one week period, the amount of earthworm tissue lost was related to the total amount presented to the planarians but not to their size. Frequency of attack was, however, related to planarian size. The results are discussed in relation to an hypothetical population of earthworms in pasture. It is concluded that the results support the contention that A. triangulata could seriously deplete earthworm populations.  相似文献

In many species of simultaneous hermaphrodites, body size correlates with fecundity, and larger partners are preferred to small ones. Since sperm exchange is usually reciprocal, small individuals may be rejected by larger partners resulting in size-assortative mating. We studied the mating patterns in a natural population of the simultaneous hermaphroditic earthworm Eisenia fetida (Oligochaeta, Lumbricidae). We found that size-assortative mating processes existed, with variance in body weight within pairs lower than between pairs in mating earthworms. This non-random mating pattern probably reveals the existence of mate selection in this species, which lives at elevated densities with high availability of potential mates.  相似文献

Despite their ubiquity and ecological significance in temperate ecosystems, the behavioural ecology of earthworms is not well described. This study examines the mechanisms that govern aggregation behaviour specially the tendency of individuals to leave or join groups in the compost earthworm Eisenia fetida, a species with considerable economic importance, especially in waste management applications. Through behavioural assays combined with mathematical modelling, we provide the first evidence of self-assembled social structures in earthworms and describe key mechanisms involved in cluster formation. We found that the probability of an individual joining a group increased with group size, while the probability of leaving decreased. Moreover, attraction to groups located at a distance was observed, suggesting a role for volatile cues in cluster formation. The size of earthworm clusters appears to be a key factor determining the stability of the group. These findings enhance our understanding of intra-specific interactions in earthworms and have potential implications for extraction and collection of earthworms in vermicomposting processes.  相似文献

The present investigations were conducted to ascertain the influence of Pb2+ and Cd2+ both individually and in combination on selected enzymes of tricarboxylic acid (TCA) cycle. All the enzymes of TCA cycle examined (-ketoglutarate dehydrogenase, isocitrate dehydrogenase, succinate dehydrogenase, malate dehydrogenase) were affected deleteriously by Pb2+ as well as Cd2+ and these metals in combination gave more or less an additive effect.  相似文献

Earthworms, Eisenia fetida, were exposed to soils spiked with As(III) and As(V), to understand the response of earthworms to As in terms of both toxicity and accumulation using toxicokinetics, and to explain As metabolism and bioavailability. As(III) showed higher toxicity than As(V), in all toxicity endpoints of burrowing time, survival, growth, and cocoon production. Bioconcentration occurred at both As(III) and As(V) treatments during 28-d exposure. Worms did not show the elimination of As for consecutive 28 d of exposure to clean soils. Biotransformation of As was characterized using HPLC-ICP-MS and XANES, showing the reduction of As in worms regardless of the As species to which worms were exposed. Metabolism of As in worms that formed As-thiol complex is thought to limit the excretion of As, and thus induce bioconcentration in worms. Uptake rates by one-compartment model indicated that pore water was the bioaccessible pool of As, and directly controlled the uptake of As by worms. The study suggests that higher uptake rate and bioaccumulation of As(III) than of As(V) are among the factors that make As(III) more toxic than As(V).  相似文献

Eisenia fetida were exposed continuously to (60)Co gamma radiation during two generations (F(0) and F(1)). Adult F(0) reproduction capacity (i.e., number of cocoons produced, hatchability and number of F(1) hatchlings) in controls and at five dose rates (0.18, 1.7, 4, 11 and 43 mGy/h) was measured over a 13-week exposure period. Survival, growth and sexual maturation of F(1) hatchlings were observed for 11 weeks. F(1) adults were exposed for a further 13 weeks to determine their reproduction capacity. There was no radiation-induced effect on the cocoon production rate in either F(0) or F(1). For F(0), hatchability of cocoons produced during the first 4 weeks was reduced to 60% at 43 mGy/h (98% in controls), and none of the cocoons produced at 5-13 weeks hatched. At 11 mGy/h the cocoon hatchability was reduced to 25% at 9-13 weeks. In addition, the number of hatchlings per hatched cocoon was reduced at 11 and 43 mGy/h. Correspondingly, at these dose rates, the total number of F(1) hatchlings per adult F(0) was significantly lower than in the control. This number was also reduced at 4 mGy/h, but the effect was of borderline significance. For adult F(1), the hatchability of cocoons at 11 mGy/h was reduced to 45-69% during the 13-week exposure period. The number of hatchlings (F(2)) per cocoon and the total number of F(2) individuals produced was also reduced. However, and in contrast to the results observed for F(0), hatchability increased with time, suggesting a possible acclimatization or adaptation of the F(1) individuals. In conclusion, chronic irradiation reduced the reproduction capacity of E. fetida, but extensive exposure periods (13 weeks) were needed for these effects to be expressed. The lowest dose rates at which an effect was observed were 4 mGy/h in F(0) and 11 mGy/h in F(1).  相似文献

研究了不同阿维菌素B1a浓度对种土壤微生物生长和呼吸强度的影响及对土壤中蚯蚓的急性毒性.结果表明,土壤中阿维菌素B1a浓度在83.3 mg·kg-1以上时对种土壤细菌均表现出明显的抑制作用,但对土壤真菌不表现抑制作用.高浓度阿维菌素B1a对土壤微生物的呼吸强度有抑制作用,并且在不同土壤其作用有差异.采用滤纸接触试验和人工土壤试验测定阿维菌素对蚯蚓的急性毒性(半数致死量,LD50),接触毒性LD50为.63μg·cm-2,土壤法试验测定的LD50在处理后第7 d和第1 d分别为2.13和17.06 mg·kg-1.  相似文献

The enantioselectivities of individual enantiomers of furalaxyl in acute toxicity and bioaccumulation in the earthworm (Eisenia foetida) were studied. The acute toxicity was tested by filter paper contact test. After 48 h of exposure, the calculated LC50 values of the R‐form, rac‐form, and S‐form were 2.27, 2.08, and 1.22 µg cm‐2, respectively. After 72 h of exposure, the calculated LC50 values were 1.90, 1.54, and 1.00 µg cm‐2, respectively. Therefore, the acute toxicity of furalaxyl enantiomers was enantioselective. During the bioaccumulation experiment, the enantiomer fraction of furalaxyl in earthworm tissue was observed to deviate from 0.50 and maintained a range of 0.55–0.60; in other words, the bioaccumulation of furalaxyl was enantioselective in earthworm tissue with a preferential accumulation of S‐furalaxyl. The uptake kinetic of furalaxyl enantiomers fitted the first‐order kinetics well and the calculated kinetic parameters were consistent with the low accumulation efficiency. We studied metallothionein (MT) response in the manure worm Eisenia fetida after exposures to cadmium (Cd), zinc (Zn) or cadmium and zinc spiked media. MT was studied both at the protein level by Dot Immunobinding Assay, (DIA) and at the expression level by Northern blotting. Cd was highly accumulated by worms whereas Zn body concentration was regulated. In addition, Zn would limit Cd accumulation in worms exposed to low Cd concentrations (1 and 8 mg Cd kg(-1) of dry soil). Exposure to a mixture of Cd and Zn at high concentrations increased cytosolic MT levels. This increase would allow worms to regulate body Zn concentrations and also to limit Cd toxicity. Cd exposures increased gene expression of Cd-binding MT isoform (MT 2A) whereas Zn did not. However, when both metals were at high concentrations in the exposure medium, this expression was further increased. Several hypotheses are proposed to explain the results and the best approach to estimate metal exposure of this earthworm species is given. Further experiments have now to be performed to evaluate the usefulness of these MT responses for field contaminated soils toxicity assessment using this earthworm species.  相似文献

The appearance and development of the GABA-immunoreactive nervous elements in the central nervous system of Eisenia fetida were studied by immunocytochemistry. The nervous system originates from the neuroectoderm situated on the ventral side of the embryo. The organization of the circumpharyngeal ring starts earlier than that of the ventral cord. In the elementary ring the first GABA-immunopositive neurons can be observed (E1 stage) around the mouth. Later the cell number gradually increases and parallel to this process the elementary ring is separeted into a superficial and a deeper portion. The brain and the subesophageal ganglion will be organized from the superficial ring, while the nervous elements of the deeper ring will give rise for the first GABA-immunoreactive elements of the stomatogastric nervous system. In the early stages of the embryogenesis the immunoreactive cells of the developing brain appear solitary, while from the stage E4 they gradually are observed in groups. According to their position, these cell groups are similar to those observed in the brain of the adult earthworms. During embryogenesis the level of the ventral cord ganglia depends on their position in the ectodermal germ bands. It means, that the more organized ganglia are near the circumpharyngeal ring, mean while less developed ganglia are situated caudally from them. By the end of the embryogenesis all ganglia of the ventral cord will be equally well organized. The nerve tracts of the ganglia are built up from contra- and ipsilateral by projected fibres. From E3 stage the medial tracts, mean while from the E4 stage the lateral tracts begin to be formed. During the next stages, more and more fibres connect to the both tracts. At hatching, the development of the central nervous system of Eisenia fetida is not completed, the process is continued during the postembryonic development.  相似文献

通过人工污染土壤的方法,设计芘的暴露浓度为0、60、120、240、480、960μg.kg-1.暴露实验进行1、3、7和14d后,分别检测蚯蚓内脏中细胞色素P450含量、谷胱甘肽转移酶(GST)、超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性和丙二醛(MDA)含量.结果表明,在供试浓度范围内,蚯蚓内脏中各生化指标对污染物暴露指示的敏感性存在差异:其中P450含量、GST和SOD活性最为敏感;POD和CAT活性次之;而MDA含量未对低剂量的芘暴露起到明显的指示作用.研究同时发现,低剂量污染物暴露的时间效应要强于剂量效应的影响.因而,在进行生态毒性诊断时,采用多指标和多时段的检测对增强指示的灵敏性和有效性尤为重要.  相似文献

The cytochemical and functional characteristics of chloragocytes of both 'control' and cold-stressed Eisenia fetida were examined. Flow cytometry revealed the heterogeneity of chloragocytes: the first group was characterized by low, the second one by high acid phosphatase (AcP) content. In 'control' animals the former, in cold-stressed ones the latter type were the dominant form. The elevated AcP-activity correlated with the accumulation of autophagic vacuoles (AVs) in chloragocytes. Both AVs and all small chloragosomes showed high AcP activity, while most of the large chloragosomes did not display any. Most 'control' granules (0.75-1.25 μm) contained high amounts of Ca and P, with less and variable quantities of S, Cl, K, Fe and Zn. Small chloragosomes with low Ca and P concentrations were seldom found. In cold-stressed animals the number of small granules (0.25-0.75 μm) increased up to 40% of total population. Their Ca and P contents were significantly lower; S and Fe concentrations were higher than those of large chloragosomes (1.0-1.5 μm). Our results prove that the formation and elemental composition of chloragosomes can be influenced by environmental stressors and suggest that the mature chloragosomes are tertiary lysosomes and their formation is coupled to autophagocytosis.  相似文献
