The organization of the dwarf male of Bonellia viridis was studied by electron microscopy. The epidermis is formed by two types of epithelial cells: the majority are multiciliated cells; highly vacuolated, non-ciliated cells are less abundant. The body wall musculature consists of an outer circular, a diagonal, and a longitudinal layer. As a unique feature in coelomate spiralians it was found that the perikarya of all muscle cells are located internal to the entire contractile muscular layer. The muscles are solitary myocytes embedded in extracellular matrix. Masses of secretory and indifferent cells occur inside the muscles. Two types of secretory cells were distinguished. Both of them apparently undergo holocrine secretion. A complete lining of thin peritoneal cells delimits the body cavity. Also, the gut and sperm sac have a complete peritoneal lining. The coelomic lining of the gut is a single-layered myoepithelium, that of the sperm sac a pseudo-stratified myoepithelium. The vas deferens was seen to be ciliated. The entrance of the sperm sac is formed by a ciliated funnel that leads into the reservoir by means of a thin, ciliated canal. The existence of repeated transverse nerves and of four longitudinal nerve cords is described for the first time.  相似文献

Trunk irrigatory and locomotory movements in the echiuran Bonellia viridis consist of peristaltic waves of constriction. Irrigation is by antero-posterior waves. Locomotion in the burrow may be produced in three different ways: (a) by antikinetic, (b) by synkinetic, and (c) by antikinetic and synkinetic waves in strict alternation. The animal can turn in the burrow and pass out of narrow holes. The animal can move by peristaltic waves also outside its burrow. The versatility of locomotory modes is related to its inhabiting ready-made burrows.  相似文献

An account is given of the various movements of the proboscis of Bonellia viridis. When at rest, the proboscis is coiled up in front of the trunk. The proboscis lobes progress with a velocity of the order of 1 mm/sec by means of powerful cilia situated on the dorsal surface of the leading edge. The lobes passively drag and uncoil the stem of the proboscis which is further uncoiled by muscular contractions taking place along the uncoiled part of the stem. Proboscis retraction takes place by means of contraction of the numerous longitudinal muscle strands of the stem. The terminal lobes can attach to the substrate and pull the trunk by contraction of the proboscis stem. The proboscis often ties itself into a knot which it can untie.  相似文献

Spermatozoa of the echiurans Bonellia viridis and Hamingia arctica show a similar ultrastructure. They are of a modified type. The head consists of a roughly cylindrical nucleus, which has a cover of electron-dense material. The acrosome is very large and consists of an acrosomal vesicle and a rod-shaped perforatorium or acrosomal rod. In close association with the nucleus, one or two mitochondria are found forming an irregular ring around the posterior tip of the nucleus and the centriolar apparatus. There are two centrioles, the proximal one with the conventional triplet microtubular structure. The tail flagellum is about 50 μm long and has the 9+2 axonemal structure. The oblique attachment of the acrosome to the anterior part of the nucleus gives the spermatozoon a bilateral symmetry. However, in the nuclear morphology, the arrangement of electron-dense material around the nucleus, in the mitochondria, and in the attachment of the tail flagellum, the spermatozoon shows asymmetric organization. The sperm structure in bonelliids is unique but its genesis and the morphology of the mitochondrial midpiece support the theory that the echiurans are related to the annelids. 1. The photodynamic activity of bonellin, an integumentary chlorin of Bonellia viridis, is investigated. 2. 10(-6) M bonellin solutions haemolyze erythrocytes only in the presence of light. Previous illumination (tungsten lamp 2000-4000 lux for 1 hr) of the bonellin solutions does not affect the results. Under lowered oxygen tensions the bioactivity is depressed. Benzoquinone and singlet oxygen quenchers delay the photodynamic effect. 3. Bonellin (2 x 10(-6) M) destroys echinoid gamete function, depresses oxygen uptake of spermatozoa, and arrests development of echinoid and Bonellia eggs. These effects are produced only in the presence of light. 4. Copper bonellin is not photodynamically active. The role of copper may be to protect against photosensitization.  相似文献

The Echiura have been placed in close phylogenetic affinity to the Annelida on the basis of numerous homologous characters including the mode of development, the nearly identical formation of a trochophore larva, as well as the development and ultrastructure of chaetae and spermatozoa. Furthermore, phylogenetic analysis of elongation factor-1 gene sequences supports placement of the Echiura within the Annelida. Nevertheless, the Echiura are generally excluded from the Annelida due to their lack of segmentation. However, it must be considered that this lack could represent a secondary condition and that Echiura are derived from formerly segmented ancestors. In the present study, the combination of methods applied reveals several novel aspects of the central nervous system in developmental stages of Bonellia viridis. The most important of these is the metameric organization of the ventral nerve cord. Antibodies against different neurotransmitters label discrete repetitive units of perikarya in the ventral nerve cord. This organisation is additionally supported by the distribution of peripheral nerves as shown by labelling of neurotubules. These nerves are clearly paired and are evenly distributed, corresponding to the serial units of serotoninergic neurons. Different methods of computer-aided three-dimensional reconstruction display the precise spatial distribution of perikarya and peripheral nerves allowing the repetitive units to be discerned on the basis of relative size, position and number of labelled cells. The repetitive units in the nervous system of B. viridis correspond to segmental ganglia of various Annelida and are interpreted as an indication that Echiura are derived from formerly segmented ancestors, thus supporting the systematic inclusion of the Echiura within the Annelida.  相似文献

An account is given of the development of green pigment in the early larval stages of the echiuran worm, Bonellia viridis . The green pigment first appears in scattered epidermal cells of the gastrula, before the development of the trochal rings. It is contained in spherical vacuoles (0.5-1.0 μm in diameter). As the gastrula develops into the trochophore there is an increase in total pigment content, as well as an increase in number of pigment cells and size of pigment vacuoles. The increase in pigment cell number in the trochophore results from the proliferation of the pigment cells of the gastrula. The pigment content increases rapidly before the trochophores hatch but shows little relative increase after hatching whilst the trochophores remain undifferentiated. We suggest that the pigment may have a protective role in the free-living trochophore for camouflage or chemical defence. In trochophores that undergo male differentiation the pigmentation is lost. Since the males do not lead a free-living existence but live as commensals inside the uterus of the female camouflage or chemical defence would be unnecessary. In trochophores undergoing female differentiation there is an increase in pigment content of the preoral lobe which develops into the proboscis. Following the enlargement of the coelom, clusters of coelomic cells accumulate green pigment whilst still in the coelom and then migrate to the integument of the body wall.  相似文献

In rhe echiuran Bonellia viridis the green pigment first develops in the gastrula, before the formation of the trochal rings, and the trochophores acquire a deep green pigmentation before hatching. Eggs that were cultured under continuous illumination (2000–4000 lx) did not develop any green pigment, and although the trochophores hatched normally, they lacked pigment entirely. The unpigmented trochophores that were kept under continuous illumination developed into unpigmented males and females. The far-blue (380–400 nm) and red (620–640 nm) regions were more effective in preventing pigment development than the blue, green and yellow regions of the spectrum. Illumination of the eggs before the gastrula stage did not affect the development of pigment when these were subsequently returned to the dark. When unpigmented trochophores, that had been cultured in the light, were placed in the dark they acquired a slight pigmentation, but this was much less intense than that of trochophores that had been cultured in the dark throughout. Pigmented trochophores were more vulnerable to high light intensities than unpigmented ones.  相似文献

Sperm proteins presumably play critical roles in reproduction, but in many non‐model animals their identities are unknown. A total of 147 sperm proteins from the echiuran worm Urechis unicinctus, the first sperm proteome in the phylum Annelida, are reported. The echiuran sperm proteome can be classified into diverse functional groups: energy metabolism (31%), protein synthesis and degradation (18%), spermatogenesis and sperm motility (12%), signal pathway (11%), ion channel and transport proteins (6%), cytoskeleton (4%), immunity and stress responses (3%), and fertilization (1%). These results will facilitate studies of mechanisms of fertilization in echiurans, as well as comparative studies of reproduction and evolution across lophotrochozoans. The present paper deals with the isolation, and chemical and serological characterization of the O-antigens (lipopolysaccharides, LPS) of the photosynthetic gram-negative bacterium Rhodopseudomonas viridis. The LPS are extractable with hot phenol/water, but unlike the phenol-soluble LPS of the closely related species Rhodopseudomonas palustris, the R. viridis O-antigens are preferentially extracted into the water phase. A mixture of phenol/chloroform/petroleum ether (PCP-method) does not extract the R. viridis LPS. All R. viridis LPS investigated belong to the same chemotype, the polysaccharide moiety of these O-antigens being composed of 3-O-methyl-l-xylose, 3-O-methyl-d-mannose, d-mannose, d-galactose, d-glucose, in addition to 2-keto-3-deoxyoctonate (KDO), glucosamine, 6-deoxyglucosamine (quinovosamine) and galactosamine uronic acid. The R. viridis O-antigens are clearly distinguishable from the l-glycero-d-mannoheptose containing O-antigens of R. palustris by the lack of this sugar (and of any other heptose) in the R. viridis LPS. The lipid moiety (lipid A) of the R. viridis O-antigen can be split off from the LPS by mild acid hydrolysis. Like lipid A from R. palustris, it differs remarkably from the well known lipid A of Enterobacteriaceae, in that d-glucosamine is replaced by a recently identified 2.3-diamino-2.3-dideoxyhexose in the R. viridis and R. palustris lipid A. Unlike enteric lipid A the R. viridis lipid A is phosphate-free and includes as the only fatty acid β-C14OH which is exclusively amide-linked. All R. viridis strains belong to the same serotype so far as investigated, as shown by passive hemagglutination with the isolated O-antigens and rabbit antisera against heat-killed cells.  相似文献

Echiura is traditionally regarded as a small phylum of unsegmented spiralian worms. Molecular analyses, however, provide unquestionable evidence that Echiura are derived annelids that lost segmentation. Like annelids, echiurans possess chaetae, a single ventral pair in all species and one or two additional caudal hemi-circles of chaetae in two subgroups, but their evolutionary origin and affiliation to annelid chaetae are unresolved. Since annelids possess segmental pairs of dorsal (notopodial) and ventral (neuropodial) chaetae that are arranged in a row, the ventral chaetae in Echiura either represent a single or a paired neuropodial group of chaetae, while the caudal circle may represent fused rows of chaetae. In annelids, chaetogenesis is generally restricted to the ventral part of the notopodial chaetal sac and to the dorsal part of the neuropodial chaetal sac. We used the exact position of the chaetal formation site in the echiuran species, Thalassema thalassemum (Pallas, 1766) and Echiurus echiurus (Pallas, 1767), to test different hypotheses of the evolution of echiurid chaetae. As in annelids, a single chaetoblast is responsible for chaetogenesis in both species. Each chaeta of the ventral pair arises from its own chaetal sac and possesses a lateral formation site, evidencing that the pair of ventral chaetae in Echiura is homologous to a pair of neuropodia that fused on the ventral side, while the notopodia were reduced. Both caudal hemi-circles of chaetae in Echiurus echiurus are composed of several individual chaetal sacs, each with its own formative site. This finding argues against a homology of these hemi-circles of chaetae and annelids’ rows of chaetae and leads to the hypothesis that the caudal chaetal rings evolved once within the Echiura by multiplication of ventral chaetae.  相似文献

Twelve new microsatellite loci were isolated from the Green Lizard (Lacerta viridis viridis). Primers for 28 loci were designed and 18 of these loci amplified well for 10 individuals of four populations. Twelve of these loci were further characterized for a population in Hungary. The results document the suitability of these identified loci for the characterization of the genetic diversity of the endangered species L. viridis viridis.  相似文献

Large numbers of swimming and stranding Urechis unicinctus were observed at night during low tide in Sasuhama, Miyagi Prefecture, northeastern Japan, during the periods from January to February in 2012 and 2013. Worms did not drift passively but swam actively, therefore hinting at a certain purpose for such behavior. As trochophore larvae of U. unicinctus were observed to occur simultaneously in the plankton, we infer the possibility that this is an event of reproductive swarming. Anatomical observations of both swimming and stranding U. unicinctus showed that none of the specimens had gametes, which may suggest that these were completely spent after spawning. Urechis unicinctus seemed to begin swimming after dusk and the observed swimming behavior occurred during the evening ebb tide throughout the night low tide during winter time. Stranding U. unicinctus have long been known in Japan and have been attributed to sea storms. The present study shows for the first time the possibility that U. unicinctus swims in order to reproduce at night and that this swimming behavior is closely linked to the stranding of U. unicinctus individuals.  相似文献

Fast atom bombardment (FAB) mass spectrometry was used to identify a new small myotoxin from the venom of the prairie rattlesnake (Crotalus viridis viridis). FAB mass spectrometry and Edman degradation were used to characterize its structure. This toxin is similar to myotoxin I from C. v. concolor, except that it possesses an additional. C-terminal asparaginyl-alanine. At 45 residues it is the longest known myotoxin a homolog. A myotoxin of 43 residues, identical to myotoxin I from C. v. concolor, was also found. To date no other species has been shown to produce more than one length of myotoxin. The present paper documents 42-, 43-, and 45-residue myotoxins from the venom of a single animal.  相似文献

The histones present in mature oocytes and embryos of Urechis caupo and their pattern of synthesis during early development have been characterized. Acid-soluble proteins extracted from mature oocyte germinal vesicles and from embryonic nuclei were analyzed by two-dimensional polyacrylamide gel electrophoresis. Histones are accumulated in the mature oocytes in amounts sufficient to provide for the assembly of chromatin through the 32- to 64-cell stage of embryogenesis. Two H1 histones, which appear to be variants, were found. Germinal vesicles and cleavage-stage nuclei are enriched in H1M (maternal). During late cleavage a faster-migrating H1, H1E (embryonic), appears among the nuclear histones and, as embryogenesis continues, replaces H1M as the predominant H1. No new core histone variants are detected during early development. Examination of [3H]lysine-labeled histones from germinal vesicles and embryonic nuclei reveals stage-specific patterns of histone synthesis. H1M is the major H1 species synthesized in mature oocytes. After fertilization, a switch to the predominant synthesis of H1E occurs. Comparison of the [3H]lysine incorporated into H1E and core histones indicates that H1E synthesis is disproportionately high from midcleavage through the midblastula stage. By the gastrula stage, a balanced synthesis of H1E and each core histone is established. The results indicate that there is noncoordinate regulation of H1 and core histone synthesis during Urechis development.  相似文献

Summary The burrowing behaviour of the toad Bufo viridis was studied in the laboratory on soil containing 11–13% moisture. These toads usually excavate shallow, temporary burrows from which they emerge intermittently. Time spent subsurface increases with rising ambient temperature in the range of 18–30° C. In these conditions, water loss is effectively reduced and a new water balance is achieved. On return to water access, after two months on soil, the toads rapidly regain only that amount of water lost previously. It is concluded that this temperature-sensitive burrowing behaviour is an important factor in these animals' successful survival in semi-arid conditions.  相似文献
